Abstract
本发明公开了一种人工抗原递呈细胞及其制备方法与应用。本发明建立了一种对聚合物微球表面拓扑结构形貌控制方法,经过信号分子修饰发挥微球抗原递呈功能,可用于刺激免疫细胞扩增和活化。本发明使用的材料具有生物相容性、可降解性,且这种制备表面粗糙结构的方法操作简便、环保,重现性好。
Description
技术领域
本发明属于细胞治疗及免疫治疗领域,具体涉及一种人工抗原递呈细胞及其制备方法与应用。
背景技术
近年来T细胞免疫治疗研究发展迅速,尤其是基因工程T细胞的出现显著提高了免疫治疗的特异性和杀伤强度,并在急性B淋巴细胞白血病的治疗中首次获得巨大成功(Brentjens et al.,2011)。截至目前,基因改造T细胞免疫治疗已在多种恶性肿瘤中相继开展大量临床试验,使T细胞免疫治疗成为最具价值和发展潜力的新型抗肿瘤疗法(Johnson and June,2017)。为获得足够量T细胞,采用自体DC细胞在体外刺激扩增T细胞已开展大量研究(Wang et al.,2014)。尽管DC细胞表现出了对T细胞激活的重要作用,但体外分离、诱导活化DC细胞操作复杂、培养周期长,原代DC细胞的大规模制备困难;另外DC细胞活化状态不稳定,造成刺激T细胞效果批间差异大;更重要的是DC细胞会通过表达抑制性信号分子防止T细胞过度活化,由于缺乏对这种负调节的控制,会严重影响刺激T细胞的功能和后续治疗效果(Steinman and Banchereau,2007),难以形成商品。人工抗原递呈细胞(artificial antigen presenting cells,aAPCs)的出现,可以实现根据需要向T细胞递呈不同的信号分子,以达到稳定、高效的激活效果。
aAPCs是通过在载体表面整合T细胞活化所必需的信号分子,来模拟抗原递呈过程的一种方法。除信号分子外,载体的形貌,尤其是表面拓扑结构直接影响aAPCs功能。研究显示,载体表面拓扑结构会影响蛋白类信号分子在aAPCs表面的空间结构、生物学活性,以及与T细胞相互作用的接触面积(Fadel et al.,2010)。这些因素直接影响T细胞免疫突触的形成,因此是aAPCs研制中需要重点考虑的设计因素。由于高分子聚合物载体形貌难于控制,所以目前aAPCs拓扑结构的研究主要采用无机碳纳米管和介孔硅材料。但FDA批准的医用高分子聚合物具有生物相容性好,毒副作用小等优点,是aAPCs载体的最优选择。因此开发具有表面拓扑结构的高分子聚合物aAPCs具有重要的临床应用价值。
发明内容
本发明的目的是提供一种人工抗原递呈细胞及其制备方法与应用。
本发明要求保护的人工抗原递呈细胞(aAPCs),由表面具有粗糙拓扑结构的实心聚合物微球和信号分子组成;
所述实心聚合物微球中包载有四氧化三铁纳米粒子。
本发明还要求保护一种表面具有粗糙拓扑结构的实心聚合物微球以及该表面具有粗糙拓扑结构的实心聚合物微球在制备人工抗原递呈细胞中的应用;
所述实心聚合物微球内部包载四氧化三铁纳米粒子。
具体的,所述信号分子为生物素偶联信号分子;
所述实心聚合物微球表面修饰有亲和素;
所述信号分子与所述实心聚合物微球通过生物素-亲和素系统方式结合。
构成所述实心聚合物微球中的聚合物选自PLLA、PDLA、PLA、PLGA或PGA;
所述实心聚合物微球的粒径为2μm-10μm;
所述四氧化三铁纳米粒子的粒径为7nm-30nm;具体为15nm;
所述生物素偶联信号分子为T细胞激活抗体。
更具体的,所述T细胞激活抗体为anti-CD3和anti-CD28抗体;具体为摩尔比为1:1的anti-CD3和anti-CD28抗体。
所述粗糙拓扑结构为纳米级粗糙拓扑结构。
本发明还要求保护一种制备表面具有粗糙拓扑结构的实心聚合物微球的方法,该方法包括如下步骤:将表面光滑且内部包载四氧化三铁纳米粒子的聚合物微球与聚合物溶解试剂进行反应。
具体的,所述聚合物为PLLA,所述PLLA分子量为50000以上。
具体的,所述聚合物溶解试剂选自四氢呋喃、NMP和丙酮中的至少一种;具体选自四氢呋喃或由NMP和丙酮组成的混合液;所述由NMP和丙酮组成的混合液中,NMP和丙酮的体积比为1:0.5-1:2或1:1;
所述表面光滑且内部包载四氧化三铁的纳米粒子聚合物微球与聚合物溶解试剂的用量比为225mg:1mL-5mL;
所述反应步骤中,温度为10-30℃,具体可为25℃;时间为5min-40min,具体为30min;
所述方法还包括:在所述反应完毕后,离心,除去上清液;
具体的,所述离心步骤中,转速为3000rpm-8000rpm;时间为3-5min;
上述表面光滑且内部包载四氧化三铁纳米粒子的聚合物微球可按照各种常规方法制得,如可按照如下方法制得:
将聚合物、四氧化三铁纳米粒子与有机溶剂混合得到溶液a后,滴加到PVA水溶液中,先匀浆搅拌,后磁力搅拌,离心,除上清液即得;
具体的,所述四氧化三铁纳米粒子的粒径为7nm-30nm,具体为15nm;
所述有机溶剂为二氯甲烷;
所述聚合物与四氧化三铁的用量比为50:1-500:1,具体为150:1;
所述聚合物在溶液a中的浓度为10-25mg/mL,具体为15mg/mL;
所述PVA水溶液的浓度为0.1-1g/mL,具体为1g/mL;
所述滴加步骤中,滴加速率为1mL/min-5mL/min,具体为2mL/min;
所述匀浆搅拌步骤中,转速为3600rpm-7200rpm;时间为2-5min;匀浆搅拌的目的是控制粒径;
所述磁力搅拌步骤中,搅拌速度为600-800rpm;时间为8-18h;具体为12h;磁力搅拌的目的是使有机溶剂挥发,固化微球;
所述离心步骤中,离心力为3000rpm-8000rpm;
本发明提供的制备所述人工抗原递呈细胞的方法,包括如下步骤:
1)将所述表面具有粗糙拓扑结构的实心聚合物微球记为微球a;
在所述微球a的表面修饰亲和素,记为微球b;
2)将所述微球b与所述信号分子进行结合,得到所述人工抗原递呈细胞。
上述方法的步骤1)中,修饰方法为物理吸附法。
具体的,所述物理吸附法包括:将所述微球a与亲和素溶液混合进行反应,反应完毕后离心,除去上清液,PBS洗涤而得;
所述反应步骤中,温度为4-37℃;时间为1小时以上,具体为1小时至12小时;
所述离心步骤中,转速为2000rpm-5000rpm;时间为3-5min;
所述亲和素溶液的浓度为0.1-10mg/mL,具体为1mg/mL;溶剂为PBS;
所述微球a与亲和素的用量比为3×106个微球:0.1-10mg;更具体为3×106个微球:1mg。
所述步骤1)在所述PBS洗涤步骤后,还包括离心,除去上清液的步骤;所述离心步骤中,转速为2000rpm-5000rpm,具体为3000rpm;时间为3-5min;
所述步骤2)中,所述微球b与生物素偶联信号分子的用量比为3×106个微球与0.24μg/mL-6μg/mL,具体为3×106个微球与1.2μg/mL;
所述结合步骤中,生物素偶联信号分子为anti-CD3和anti-CD28分子,且两者投入比为1:1;
所述结合步骤中,温度为4-37℃;时间为1小时以上,具体为1小时至12小时;
所述结合在缓冲溶液中进行;所述溶液具体为PBS缓冲液;所述生物素偶联信号分子在所述缓冲溶液中的浓度为0.24μg/mL-6μg/mL;
所述离心步骤中,转速为2000rpm-5000rpm,具体为3000rpm;时间为3-5min。
上述方法中,所用PBS缓冲液的pH值均为7.2-7.4。
另外,上述本发明提供的所述人工抗原递呈细胞在制备免疫细胞扩增和/或激活产品、在制备抗肿瘤免疫治疗产品或在制备自身免疫性疾病治疗产品中的应用,也属于本发明的保护范围。所述免疫细胞具体可为T细胞。
本发明还要求保护一种免疫细胞扩增和/或激活产品、一种抗肿瘤免疫治疗产品及一种自身免疫性疾病治疗产品,其有效成分均为前述本发明提供的人工抗原递呈细胞。上述产品具体可为药物或药物制剂。
所述人工抗原递呈细胞使用时抗体浓度具体可为40ng/mL-1000ng/mL,具体可为200ng/mL。
所述人工抗原递呈细胞与免疫细胞的比例为1:2-1:20。
所述免疫细胞具体可为T细胞;
所述人工抗原递呈细胞与T细胞共培养时抗体终浓度为40ng/mL-1000ng/mL,具体可为200ng/mL;
此外,本发明还要求保护一种激活和/或扩增免疫细胞的方法,包括如下步骤:将所述人工抗原递呈细胞和免疫细胞共培养;
上述共培养具体可为在37℃条件下培养。培养完毕后检测T细胞活化因子及扩增倍数。
所述免疫细胞具体可为T细胞;
所述人工抗原递呈细胞与T细胞共培养时抗体终浓度为40ng/mL-1000ng/mL,具体可为200ng/mL;
所述人工抗原递呈细胞与免疫细胞的个数比为1:2-1:20,具体可为1:10,优选1:2。
本发明还要求保护所述人工抗原递呈细胞在激活和/或扩增免疫细胞中的应用。所述免疫细胞具体可为T细胞。
本发明以表面具有拓扑结构聚合物粗糙微球为模板的aAPCs,1)粗糙结构的出现使微球比表面积显著增加、提高了信号分子结合量;2)表面纳米尺度的拓扑结构提高了对蛋白质非特异吸附能力,3)亲和素可以同时偶联4个分子的生物素化信号分子,四聚体形式的信号分子可以提高TCR对其识别亲和力,利于TCR交联向胞内传递信号。以上特点使粗糙aAPCs表现出比光滑aAPCs更显著的T细胞扩增、活化效果。同时借助生物素-亲和素反应系统,可以在微球表面结合任何需要的信号分子。如针对自身免疫疾病,可将生物素偶联的抑制性信号分子结合后,用于抑制过度活化的免疫细胞。因此所制备的PLLA聚合物粗糙微球载体作为免疫细胞调节平台,既可用于T淋巴细胞体外扩增,细胞过继性免疫治疗、抗肿瘤免疫治疗;也在免疫调节治疗方面具有应用价值。本发明的方法操作简便、经济环保,所用材料具有良好的生物相容性、可降解性,具有重要的应用价值。

具体实施方式
下面结合具体实施例对本发明作进一步阐述,但本发明并不限于以下实施例。所述方法如无特别说明均为常规方法。所述原材料如无特别说明均能从公开商业途径获得。
下述实施例中,亲和素购自Thermo公司,产品编号A887;anti-CD3抗体也即biotinanti-CD3购自BD公司,产品编号为553060,anti-CD28抗体也即biotin anti-CD28购自BD公司,产品编号为553296;MojoSortTMMouse CD3 T Cell Isolation Kit购自biolegend公司,产品编号为480024;CBA Mouse IFN-γFlex Set kit购自BD公司,产品编号为558296;粒径为15nm的四氧化三铁购自Ocean Nanotech,产品编号为SOR-15。
下述实施例中所用含Fe3O4的PLLA光滑微球按照如下方法制备而得:
1)称取225mg的PLLA,充分溶于15mL的二氯甲烷中,制成15mg/mL的溶液,加入60μL粒径为15nm的Fe3O4;
2)以2mL/min的速度滴入100mL浓度为1%的PVA水溶液中,采用高速匀浆3,600rpm搅拌乳液5min;
3)置于磁力搅拌器上以600rpm继续搅拌过夜,使二氯甲烷挥发完全。8,000rpm离心5min,去上清,用去离子水洗三次,得内部分布有四氧化三铁的光滑微球(s-MS),平均粒径为10μm。
将步骤2)匀浆转速由3600rpm替换为7200rpm,所得内部分布有四氧化三铁的光滑微球(s-MS)的平均粒径为2μm。
图1为未包载Fe3O4的PLLA(a)与包载Fe3O4(b)的红外光谱图。由图可知,图b中出现Fe3O4的伸缩振动峰,表明Fe3O4已被成功包载。
实施例1、制备PLLA粗糙微球
1)含Fe3O4的PLLA粗糙微球的制备
A、8,000rpm离心5min,收集不同PLLA分子量(Mw:5000,10000,50000)制备的光滑微球,将水分充分去除。
B、向离心管内加入5mL四氢呋喃,用移液器分散均匀,混悬25℃反应30min;8,000rpm离心5min;
C、重复上述溶解过程2次;8,000rpm离心5min,去离子水洗3次,表征制备的微球形貌。
其中分子量5000,10000的PLLA微球被四氢呋喃完全溶解,分子量50000的PLLA微球被部分溶解,并且形成粗糙形貌,平均粒径10μm。因此粗糙微球制备选用PLLA分子量范围在50000以上。
实施例2、制备人工抗原递呈细胞
1)含Fe3O4的PLLA粗糙微球的制备:
A、8,000rpm离心5min,收集含Fe3O4的PLLA光滑微球,将水分充分去除。
B、向离心管内加入5mL四氢呋喃,用移液器分散均匀,混悬25℃反应30min;8,000rpm离心5min;
C、重复上述溶解过程2次;8,000rpm离心5min,去离子水洗3次,得含Fe3O4的PLLA粗糙微球(c-MS),平均粒径10μm。
选用粒径为2μm的内部分布有四氧化三铁的光滑微球(s-MS),其它同本实施例,制备出表面出现孔洞、表面糙程度更深、平均粒径在2μm的PLLA粗糙微球。
2)物理吸附法在微球表面修饰亲和素:
A、取3×106个微球,用PBS洗两遍:5,000rpm离心5min;
B、配置含0.1mg/mL,1mg/mL和10mg/mL的亲和素蛋白PBS溶液,将微球重悬于1mL亲和素溶液中,4℃混悬过夜;
C、将反应过夜的悬液离心,用PBS洗涤微球两遍:3,000rpm离心5min;得表面修饰亲和素的微球。用MicroBCA检测微球表面结合亲和素水平,最终选用1mg/mL亲和素蛋白修饰微球用于抗体结合。
3)微球表面结合生物素偶联抗体:
A、取亲和素修饰的微球用1mLPBS重悬于1.5mL EP管中;
B、微球用PBS洗两遍:3,000rpm离心5min;
C、按1:1比例分别加入生物素偶联的anti-CD3和anti-CD28抗体,抗体加入终浓度分别为0.24μg/mL,1.2μg/mL和6μg/mL,将EP管置于混合器中4℃混匀反应过夜,用PBS洗微球两遍:3000rpm离心5min,用100μL完全培养基重悬得aAPCs。
实验证明,用FITC标记的生物素,其它同本实施例,可同样结合在微球表面,因此PLLA粗糙微球可作为结合平台,可根据需要结合其他生物素化信号分子。
实施例3、粗糙PLLA微球制备的aAPCs用于T细胞激活和扩增
1)用CD3阴性试剂盒磁珠分选C57/B6小鼠脾脏中T细胞;
2)将aAPCs与小鼠原代T细胞按照1:2,1:10和1:20的个数比例混合,在含有10%FBS、1%双抗、1%非必需氨基酸和IL2(300U/mL)的RPMI1640培养基中共培养,37℃,5%CO2培养箱中培养至5天,用CCK-8试剂盒检测T细胞增殖,CBA法检测T细胞培养上清分泌IFN-γ的水平。
实施例证明,aAPCs与小鼠原代T细胞的个数比例为1:2时激活效果最好,并且aAPC与T细胞共培养时抗体终浓度为200ng/mL时刺激T细胞扩增、活化效果优于40ng/mL和1000ng/mL。
粗糙微球形貌见图2,显示微球表面形成缝隙,粗糙度增加。图3为含Fe3O4的PLLA粗糙微球在磁极中分离的照片,含Fe3O4的PLLA粗糙微球可以在刺激中成功分离。图4结果显示,相同条件下,与光滑微球相比,粗糙微球可以提高对亲和素蛋白的吸附水平。
图5为CCK-8检测不同条件下T细胞扩增情况。图6为CBA法检不同条件下T细胞分泌IFN-γ情况。由图5和图6结果可知,相同制备条件下,粗糙微球制备的aAPCs刺激T细胞扩增和激活的效果都好于光滑微球制备的aAPCs。
可见,本发明提供的亲和素修饰的PLLA聚合物粗糙微球,可通过生物素-亲和素反应体系,与不同的生物素偶联信号分子相结合,以用于对免疫细胞扩增和激活。
Claims (16)
1.一种人工抗原递呈细胞,由表面具有粗糙拓扑结构的实心聚合物微球和信号分子组成;
所述实心聚合物微球内部包载四氧化三铁纳米粒子;
所述信号分子为生物素偶联信号分子;
所述实心聚合物微球表面修饰有亲和素;
所述信号分子与所述实心聚合物微球通过生物素-亲和素系统方式结合;
构成所述实心聚合物微球中的聚合物为PLLA,分子量为50000以上;
所述表面具有粗糙拓扑结构的实心聚合物微球的制备方法,包括如下步骤:
将表面光滑且内部包载四氧化三铁纳米粒子的聚合物微球与聚合物溶解试剂进行反应;
所述聚合物溶解试剂为四氢呋喃;
所述人工抗原递呈细胞的制备方法,包括如下步骤:
1)将所述表面具有粗糙拓扑结构的实心聚合物微球记为微球a;
在所述微球a的表面修饰亲和素,记为微球b;
2)将所述微球b与所述信号分子进行结合,得到所述人工抗原递呈细胞。
2.根据权利要求1所述的人工抗原递呈细胞,其特征在于:所述实心聚合物微球的粒径为2μm-10μm;
所述四氧化三铁纳米粒子的粒径为7nm-30nm;
所述信号分子为T细胞激活抗体;
所述粗糙拓扑结构为纳米级粗糙拓扑结构。
3.一种表面具有粗糙拓扑结构的实心聚合物微球,
所述表面具有粗糙拓扑结构的实心聚合物微球的制备方法,包括如下步骤:
将表面光滑且内部包载四氧化三铁纳米粒子的聚合物微球与聚合物溶解试剂进行反应;
所述聚合物溶解试剂为四氢呋喃;
构成所述实心聚合物微球中的聚合物为PLLA,分子量为50000以上;
所述实心聚合物微球内部包载四氧化三铁纳米粒子。
4.根据权利要求3所述的实心聚合物微球,其特征在于:所述实心聚合物微球的粒径为2μm-10μm;
所述四氧化三铁纳米粒子的粒径为7nm-30nm;
所述粗糙拓扑结构为纳米级粗糙拓扑结构。
5.权利要求3或4所述的表面具有粗糙拓扑结构的实心聚合物微球在制备人工抗原递呈细胞中的应用。
6.一种制备权利要求3或4所述表面具有粗糙拓扑结构的实心聚合物微球的方法,包括如下步骤:
将表面光滑且内部包载四氧化三铁纳米粒子的聚合物微球与聚合物溶解试剂进行反应;
所述聚合物溶解试剂为四氢呋喃。
7.根据权利要求6所述的方法,其特征在于:所述表面光滑且内部包载四氧化三铁纳米粒子的聚合物微球与聚合物溶解试剂的用量比为225mg:1mL-5mL;
所述反应步骤中,温度为10-30℃;时间为5min-40min;
所述方法还包括:在所述反应完毕后,离心,除去上清液。
8.一种制备权利要求1或2所述人工抗原递呈细胞的方法,包括如下步骤:
1)将所述表面具有粗糙拓扑结构的实心聚合物微球记为微球a;
在所述微球a的表面修饰亲和素,记为微球b;
2)将所述微球b与所述信号分子进行结合,得到所述人工抗原递呈细胞。
9.根据权利要求8所述的方法,其特征在于:所述步骤1)中,修饰方法为物理吸附法。
10.根据权利要求9所述的方法,其特征在于:所述物理吸附法包括:将所述微球a与亲和素溶液混合进行反应,反应完毕后离心,除去上清液,洗涤而得;
所述反应步骤中,温度为4-37℃;时间为1小时以上;
所述离心步骤中,转速为2000rpm-5000rpm;时间为3-5min;
所述亲和素溶液的浓度为0.1-10mg/mL;溶剂为PBS;
所述洗涤步骤中,洗涤剂为PBS缓冲液;
所述微球a与亲和素的用量比为3×106个微球:0.1-10mg;
所述步骤2)中,所述微球b与信号分子的用量比为3×106个微球与0.24μg/mL-6μg/mL;
所述结合步骤中,温度为4-37℃;时间为1小时以上;
所述结合在缓冲溶液中进行;所述信号分子在所述缓冲溶液中的浓度为0.24μg/mL-6μg/mL;
所述离心步骤中,转速为2000rpm-5000rpm;时间为3-5min。
11.根据权利要求10所述的方法,其特征在于:所述反应步骤中,时间为1小时至12小时;
所述微球a与亲和素的用量比为3个微球:0.1-10ng;
所述步骤2)中,所述微球b与信号分子的用量比为3×106个微球与1.2μg/mL;
所述结合步骤中,时间为1小时至12小时;
所述结合步骤中的缓冲溶液为PBS缓冲液。
12.权利要求1或2所述人工抗原递呈细胞在制备免疫细胞扩增和/或激活产品、在制备抗肿瘤免疫治疗产品或在制备自身免疫性疾病治疗产品中的应用。
13.一种免疫细胞扩增和/或激活产品,其有效成分为权利要求1或2所述人工抗原递呈细胞。
14.根据权利要求13所述的产品,其特征在于:所述免疫细胞为T细胞。
15.一种抗肿瘤免疫治疗产品,其有效成分为权利要求1或2所述人工抗原递呈细胞。
16.一种自身免疫性疾病治疗产品,其有效成分为权利要求1或2所述人工抗原递呈细胞。
