組織源。頭皮、腹部領域の皮膚、結腸、及び顎下腺のサンプルは、脳・人体寄付プログラムのディレクターである、Dr.Thomas G.Beachの好意により、Banner Sun Health Research Institute(BSHRI)から入手した。標準的な臨床評価に加えて、パーキンソン病の診断は、レヴィー小体の存在により、組織ドナーで確認した。同様に、通常の非PD対照対象の状態を、レヴィー小体の欠如により判定した。サンプル(16名の男性、8名の女性;年齢65〜95;15名がPD、9名が非PD)を、死後入手してホルマリンに固定した。通常の個体、及びPDを有する対象からの、ホルマリン固定パラフィン包埋(FFPE)皮質脳ブロックをそれぞれ、Roche Tissue Diagnostics(RTD)内部組織バンク、及びFolio Biosciencesから入手した。FFPE皮膚生検ブロックの小コホートを、Folio Biosciencesの1〜16歳の8名の個体(3名の男性、5名女性)から入手し、凝集aSynに似ていない、pS129−aSyn染色の程度を評価した。36名の対象からの、72個のFFPE皮膚生検ブロックの、より大きなコホートを、神経学のMontreal General Hospital DepartmentでDr.Ron Postumaから入手した。対象が示した症状の臨床検査に基づくと、36名の対象のうち15名がPD、5名が非定型パーキンソン症候群を有すると診断された。残り(16名の対象)は、PDを有しない対照対象であった。2つの皮膚パンチ生検を、臨床訪問中に対象で実施し、FFPEブロックを、ProthenaによりVentana Medical Systemsに移した。他の全ての組織試料は、RTD内部組織バンクから入手した。特に断りのない限り、FFPEの全組織切片の厚さは、4μmであった。
aSyn又はPGP9.5に対する抗体を使用する、パーキンソン病を有する、及び有しないドナーからのFFPE脳及び皮膚切片のDAB染色
抗リン酸化S129α−シヌクレインウサギモノクローナル抗体7E2は、Roche Diagnostics GmbH CPS R&D Early Development&Reagent Designにより生成された。7E2の生成に用いた免疫原は、N末端のシステイン残基を介してKLHに抱合体化したペプチドである。このペプチドの配列(NEAYEMPpSEEGYQD(配列番号59))は、ヒトα−シヌクレインの残基122〜135に対応する。表面プラズモン共鳴(Biacore)実験は、7E2抗体が、Ser129でリン酸化していない同じペプチドと比較して、Ser129でリン酸化したα−シヌクレイン(122〜135、pS129)免疫原ペプチドに対して、高い特異性を有することを示した。7E2抗体は、α−シヌクレイン(122〜135、pS129)ペプチドに対して、速い会合速度、及び遅い線形解離速度を示した。
タンパク質分解に対する、凝集及び非凝集α−シヌクレインの異なる感度に基づき、凝集α−シヌクレインを検出することにより、脳内でのレヴィー小体の同定、及びパーキンソン病の決定的な診断の基礎が提供される。7E2を含むリン酸化S129α−シヌクレインに対して、抗体がレヴィー小体を検出する能力を評価するために、パーキンソン病を有する2名のドナー由来の皮質脳切片を、VENTANA(登録商標)OptiView DAB IHC検出キットを用いて染色した。8つ全ての抗リン酸化S129α−シヌクレイン抗体(81A、#64、MJF−R13(8−8)、5H5、2G11、7E2、3G2、及び11A5)は、レヴィー小体に特徴的なPD脳切片内で、DAB染色を生み出した(試験をした、非限定的な例示的α−シヌクレイン抗体の一覧については、表10を参照のこと)。試験した8つの抗リン酸化α−シヌクレイン抗体クローンのうち、7E2、3G2、11A5、及びMJF−R13(8−8)が、pS129−aSynに対する高い感度を示した(図1A〜1Cを参照)。
タンパク質遺伝子産物9.5(PGP9.5)(より記述的には、ユビキチンカルボキシル末端加水分解酵素L1(UCHL1)として知られている)は、ユビキチン抱合体化タンパク質の脱ユビキチン化、及び、遊離モノユビキチンプール20〜24の維持に不可欠なチオエステラーゼである。これは脳内で非常に発現しており、全神経細胞タンパク質の1%〜5%を占めると推定されている。PGP9.5は、ニューロン及び神経内分泌細胞に対するマーカーとして一般に使用されるが、PGP9.5の発現は、これらの細胞型に排他的に制限されているわけではない。PGP9.5に対する3つの異なる抗体、即ち、Cell Marque(商標)のRTDカタログ(P/N 760−4434)で入手可能なポリクローナル抗体、並びに、Abcam製の2つのモノクローナル抗体(ウサギ(EPR4118)及びマウス(13C/I3C4))を、皮膚での神経的形質、ニューロン、又は神経内分泌細胞の免疫組織化学的検出のために評価した。3つの抗体は全て神経細胞と認識できる構造物を特異的に染色可能であった(図10A〜図10Cを参照)。3つの抗体濃度を一続きに滴定した後、EPR4118及び13C/I3C4は0.2μg/mLにて、1.13μg/mLの分配器濃度にて、Cell Marque製のポリクローナル抗体に匹敵する染色強度を有することが示された(図10A〜10Cを参照)。しかし、これらの濃度において、13C/I3C4抗体は、EPR4118又はCell Marque(商標)ポリクローナル抗体と比較して、著しい量の非特異的なバックグラウンド染色を示した(図10B〜図10Aと、図10Cとを比較のこと)。
プロテアーゼ耐性は、α−シヌクレインを含む、多くの凝集タンパク質の特徴であり、α−シヌクレインの病理学的形態の検出は、プロテアーゼ処理により向上する。Roche pREDは、Prothenaと共同して、BSHRI製のPD及び非PDサンプルのコホートにおける、凝集α−シヌクレインに対する高い特異性を有するアッセイを開発した(後に、Targosに委譲された)(表4及び5を参照)。pRED/Prothena aSynアッセイは、リン酸化及び非リン酸化S129−aSynの両方を認識する5C12抗体、VENTANA(登録商標)プロテアーゼ1による非凝集aSynの除去、並びに、高感度VENTANA(登録商標)OptiView DAB IHC Detectionキットを用いる検出に基づく。しかし、少なくとも数週間切断されている皮膚切片を用いるpRED/Prothena aSynアッセイでは、最小のバックグラウンドが観察された一方で、新しく切断されたスライドでは、膠原線維の強力な染色が観察された(図11A及び図11B)。膠原バックグラウンド染色は、プロテアーゼ1を用いると、更なる膠原回復と共に悪化した(図12Aを参照)が、Ultra CC1を用いると減少した(図12Bを参照)。7E2抗体が、同じ検出スキームを用いる同一膠原染色を起こした(データは図示せず)ため、膠原バックグラウンド染色の原因は5C12抗体ではなかった。ultraView DAB IHC Detectionキットは、あらゆる膠原染色を生み出さなかった(データは図示せず)一方で、OptiView HQ Universal Linker及びHRP Multimerを用いるチラミド−TAMRA(VENTANA Purpleキット,DISCOVERY,P/N 760−229)の沈着は発生した(データは図示せず)ため、バックグラウンドは、膠原線維による非凝集DABの非特異的吸着によるものでもなかった。最終的に、OptiView HRP Multimerが、膠原線維染色の原因であると判定した(データは図示せず)。膠原線維の生理学的架橋により、多くの種類の付加物の形成がもたらされる。抗HQ IgGが、そのような付加物の1つ以上と交差反応することが可能である。この仮説は、CC1処理と関連する膠原華僑の逆転後に、膠原バックグラウンド染色が減少することと一致する。
多くのエピトープの免疫組織化学的検出は、抗原回復手順後に向上する。7E2抗体を用いて、FFPE皮膚切片でのpS129−aSynの検出における、2つの異なる抗原回復処理プロセス:VENTANA(登録商標)ULTRA細胞コンディショニング(ULTRA CC1)バルク溶液及びプロテアーゼの効果を評価した。EPR4118抗体を用いる、PGP9.5染色におけるULTRA CC1又はULTRA CC2溶液による処理の効果もまた評価した。
前のセクションに記載した実験結果により、自動化ホスホS129−α−シヌクレイン、及びPGP9.5銀/黄色二重IHCアッセイに対する、2つの最適化プロトコル(Bumblebeeアッセイプロトコル1及びプロトコル2)がもたらされた。上記セクションで詳述したとおり、プロトコル1は、pS129−aSyn検出のための抗原回復工程を含有していない。プロトコル2は、脱パラフィン処理後、及び7E2抗体インキュベーション前に、サンプルをアルカリホスファターゼ、続いてプロテアーゼで処理したことを除いて、プロトコル1と全ての態様が同一である。Bumblebeeアッセイプロトコル1及び2の実施内、及び実施間の再現性を、PDを有する3名の異なる対象由来の頭皮サンプルで評価した。実施内の再現性に関して、各PD対象からの、5つの連続して切片化したスライドを、「方法」に記載のプロトコル1又は2を用いて染色し、固有のpS129−aSynを有する、及び有しない神経的形質の数を病理学者により数えた。各PD対象における全神経的形質の百分率としての、固有のpS129−aSyn染色を有する皮膚神経的形質の量を、5つの染色切片から平均し、表11に示す。
リン酸化α−シヌクレイン及びPGP9.5銀/黄色二重IHCアッセイ(Bumblebeeアッセイという相性)の最適化の間に、なめらかな固有のリン酸化α−シヌクレイン染色パターンが、実行可能性試験に使用したPD頭皮サンプルの小コホートと排他的に関係したことが観察された。この傾向が、PD及び非PD対象由来の頭皮サンプルのより大きなコホートに拡大されるのか否かを判定するために、pS129−aSyn及びPGP9.5銀/黄色二重IHCアッセイ(Bumblebeeアッセイ)のプロトコル1及びプロトコル2の両方を使用して、15名のPD、及び9名の非PD対象由来の頭皮切片を染色した。更に、10名のPD、及び4名の非PD頭皮サンプルを含有するこのコホートのサブセット由来の切片を、自動化プロテアーゼ耐性aSyn DAB IHCアッセイ(pRED/Prothenaアッセイプロトコル)を使用して染色した。図29に示すように、Bumblebeeアッセイのプロトコル1又はプロトコル2のいずれかを用いる染色の後、PD対象由来の15個のサンプルのうち13個(87%)は、異なる種類のpS129−aSynを示した。9個の非PD頭皮サンプルのうち、Bumblebeeプロトコル1を用いると、5つ(56%)は、固有のpS129−aSyn染色を示した。対照的に、9個の非PD頭皮サンプルは、適度なプロテアーゼ及びホスファターゼ処理を伴うBumblebeeプロトコル2を用いると、固有のpS129−aSyn染色を示さなかった。びまん性顆粒タイプのpS129−aSyn染色は、プロトコル1を用いると観察されたのみで、プロトコル2を用いては観察されなかった。
ホスホaSyn及びPGP9.5銀/黄色二重IHCアッセイの感度及び特異度を更に評価するために、20名のPD、及び20名の非PD対照対象由来の腹部領域皮膚サンプルを、Bumblebeeアッセイプロトコル1及び2を用いて染色した。分析結果を図31に示す。Bumblebeeアッセイプロトコル1及び2を用いて染色した20個のPD腹部皮膚サンプルのうち、固有のpS129−aSyn染色は、それぞれ19個及び17個のサンプルで観察された。対照的に、20個の非PD腹部皮膚サンプルは、あらゆる固有のpS129−aSynシグナルを示さなかった。図31に示すように、びまん性顆粒pS129−aSyn染色は、Bumblebeeアッセイプロトコル1を用いて染色したPD及び非PD腹部皮膚サンプルの大部分に、並びに、Bumblebeeアッセイプロトコル2を用いて染色した1個のPDサンプルに低濃度で存在した。これらの結果は、20名のPD、及び20名の非PD対象由来の腹部皮膚の拡大コホートにおける、固有のpS129−aSyn染色の存在とPD臨床状態との、高い相関度を示した(ピアソンカイ二乗=Bumblebeeアッセイプロトコル1及び2に対してそれぞれ36.2、及び29.6、両方のプロトコルに対してP値<0.0001)。
皮膚に加えて、結腸及び顎下腺(SMG)は非常に神経支配されており、凝集α−シヌクレインを検出するための生検部位であることができる。剖検の時点で採取した、3名のPD、及び3名の非PD対象由来の、2通りの頭皮、結腸、及びSMGサンプルを、Bumblebeeアッセイプロトコル2を用いて染色した。通常の結腸及びSMGサンプルの以前の研究では、プロトコル1は、プロトコル2を用いるプロテアーゼ及びホスファターゼ処理により除去可能な、重質で固有のリン酸化α−シヌクレイン染色を生成したことが示されたためプロトコル1は用いなかった(図32A及び図32Bを参照)。3つの異なる組織部位における固有のリン酸化α−シヌクレイン染色の、典型的な画像を示す(図33A〜図33Cを参照)。染色したスライドは、3つの独立した読者により盲検で評価された(データは図示せず)。リーダーのうち一人は、他の2名とは異なるスコア付け法を用いたものの、Bumblebeeプロトコル2を用いる固有のリン酸化α−シヌクレイン染色の存在に基づく、非PD対象からの、PDの完全な分離には全面的な同意が存在した。特異度及び感度は共に100%であった。
アッセイ開発及び検証に使用したBSHRI頭皮及び腹部皮膚サンプルは、部検の間に入手した。診断用途のための、リン酸化S129−α−シヌクレイン及びPGP9.5銀/黄色二重IHCアッセイの利用可能性を評価するために、PD症状が診断された、及び診断されていない対象から、臨床訪問の間に入手した皮膚パンチ生検のFFPE切片を、Bumblebeeアッセイプロトコル1及び2の両方を用いて染色した。36名の対象由来の、合計72個のFFPEブロックの組織切片を、各対象から2通りのブロックが利用可能となるように染色した。対象の臨床状態については盲検とした病理学者により、染色したスライドを確認した。サンプルは皮膚の小さなパンチ生検であったため、スライド1枚当たりで入手可能な神経的形質の数は限定的(約5〜10)であった。その結果、銀染色神経的形質のパーセンテージの形態での定量的測定はなされなかった。顆粒でないことがはっきりしており、バックグラウンド銀散粉と区別するには十分なサイズを有する、固有の銀染色が存在する場合、スライドは、リン酸化aSynに対して陽性であるとみなした。2通りのブロックのうちの1つの切片が、リン酸化aSyn染色に対して陽性である場合、対象は全体として陽性であるとみなした。小児皮膚生検サンプルにおけるように、Bumblebeeプロトコル1は、プロトコル2によっては存在しなかった、固有のモルホロジーを有する重質リン酸化aSyn銀染色を生成した(図34を参照)。これにより、プロトコル1を用いる陽性で固有のリン酸化aSyn染色を有するのが、15名のPDの93%、及び16名の対照対象の50%(非定型パーキンソン症候群の5症例を含む、合計36名の対象の69%)がもたらされた。プロトコル2では、15名のPDの60%、及び16名の対照対象の6%(合計36名の対象の31%)が、固有のリン酸化aSynシグナルを示した(表15及び16を参照)。合計36名の対象の、各個体の組織ブロック、及び対応するPD状態の非盲検結果を、表16に示す。合計36名の対象からの結果をカイ二乗分析すると、固有のリン酸化aSynシグナルの存在と、臨床的症状に基づくPD状態の指定とには、非常に優位な相関が示された(ピアソンカイ二乗=10.506、DF=1、P値=0.001)。非定型パーキンソン症候群を有する5名の対象を分析から除外しても、相関は有意なままであった(ピアソンカイ二乗=10.236、DF=1、P値=0.001)。

(b)神経的形質に結合可能な一次抗体と、
を含むキット。 - (a)抱合体化した第1のラベルを有する第1の二次抗体であって、前記第1の二次抗体が、前記リン酸化α−シヌクレインを結合可能な前記一次抗体と免疫反応性である第1の二次抗体と、
(b)前記第1の二次抗体の前記第1のラベルと反応したとき、第1の検出可能なシングルを生成するひとまとまりの試薬と、
(c)抱合体化した第2のラベルを有する第2の二次抗体であって、前記第2の二次抗体が、前記神経的形質を結合可能な前記一次抗体と免疫反応性である、第2の二次抗体と、
(d)前記第2の二次抗体の前記第2のラベルと反応したとき、第2の検出可能なシングルを生成するひとまとまりの試薬と、
を更に含み、
前記第1の検出可能なシグナルと前記第2の検出可能なシグナルが異なっている、請求項26に記載のキット。 - 前記第1の検出可能なシグナルが銀染色であり、前記第2の検出可能なシグナルがQM−Dabsyl又はFast−Redである、請求項27に記載のキット。
- 前記神経的形質に結合可能な前記一次抗体が、ユビキチンC末端加水分解酵素L1(PGP9.5、UCHL1、NDGOA;PARK5;PGP95;SPG79;Uch−L1;HEL−117;PGP 9.5;HEL−S−53)、RNA結合fox−1ホモログ3(RBFOX3、FOX3、NEUN、FOX−3、HRNBP3)、微小管関連タンパク質2(MAP2、MAP2A、MAP2B、MAP2C)、160kDaの中神経フィラメント(NEFM、NFM、NEF3、NF−M)、200kDaの重神経フィラメント(NEFH、NFH、CMT2CC)、シナプトフィジン(SYP、MRX96、MRXSYP)、及び、ディスク大型MAGUK足場タンパク質4(DLG4、DLGH4、PSD−95、PSD95、SAP−90、SAP90、SAP90A)からなる群から選択されるタンパク質を結合可能な抗体から選択される、請求項26に記載のキット。
- 対象がパーキンソン病(PD)を有するか否かの判定方法であって、
(a)PDを有することが疑われる対象から、生体サンプルの固定又は凍結切片を調製することと、
(b)リン酸化α−シヌクレインが前記切片の神経的形質内で局在化するか否かを検出することと、
(c)リン酸化α−シヌクレインが神経的形質内で局在化するときに、前記対象をPDと診断することと、
を含む、方法。 - 工程(c)が、
前記切片を、リン酸化α−シヌクレインを結合可能な一次抗体及び前記神経的形質を結合可能な一次抗体と接触させることを更に含む、請求項30に記載の方法。 - 前記神経的形質に結合可能な一次抗体が、ユビキチンC末端加水分解酵素L1(PGP9.5、UCHL1、NDGOA;PARK5;PGP95;SPG79;Uch−L1;HEL−117;PGP 9.5;HEL−S−53)、RNA結合fox−1ホモログ3(RBFOX3、FOX3、NEUN、FOX−3、HRNBP3)、微小管関連タンパク質2(MAP2、MAP2A、MAP2B、MAP2C)、160kDaの中神経フィラメント(NEFM、NFM、NEF3、NF−M)、200kDaの重神経フィラメント(NEFH、NFH、CMT2CC)、シナプトフィジン(SYP、MRX96、MRXSYP)、及び、ディスク大型MAGUK足場タンパク質4(DLG4、DLGH4、PSD−95、PSD95、SAP−90、SAP90、SAP90A)からなる群から選択されるタンパク質を結合可能な抗体から選択される、請求項31に記載の方法。
- i.前記切片を、抱合体化した第1のラベルを有する第1の二次抗体であって、前記第1の二次抗体が、リン酸化α−シヌクレインを結合可能な前記一次抗体と免疫反応性である第1の二次抗体と接触させることと、
ii.前記切片を、抱合体化した第2のラベルを有する第2の二次抗体であって、前記第2の二次抗体が、前記神経的形質を結合可能な前記一次抗体と免疫反応性である、第2の二次抗体と接触させることと、
を更に含む、請求項31に記載の方法。 - i.前記切片を、前記第1の二次抗体の前記第1のラベルと反応性のひとまとまりの試薬と接触させて、前記サンプルのリン酸化α−シヌクレインの近くで、第1の検出可能なシグナルを生成することと、
ii.前記切片を、前記第2の二次抗体の前記第2のラベルと反応性のひとまとまりの試薬と反応させて、前記サンプルの前記神経的形質の近くで、第2の検出可能なシグナルを生成することと、
を更に含む、請求項33に記載の方法。 - 前記切片を前記リン酸化α−シヌクレインを結合可能な一次抗体と接触させた後、及び、前記切片を前記神経的形質を結合可能な前記一次抗体と接触させる前に、前記サンプル中で免疫複合体を変性させることを更に含む、請求項34に記載の方法。
- 前記切片が、一次抗体と接触する前に、少なくとも1つのプロテアーゼと接触する、請求項30に記載の方法。
- 前記切片を少なくとも1つのホスファターゼと接触させることを更に含む、請求項36に記載の方法。
- 前記リン酸化α−シヌクレインを結合可能な一次抗体及び前記神経的形質を結合可能な前記一次抗体が、同一の宿主種に由来する、請求項31に記載の方法。
- 前記第1の検出可能なシグナルと前記第2の検出可能なシグナルが異なっている、請求項34に記載の方法。
- 前記第1の検出可能なシグナルが銀染色であり、前記第2の検出可能なシグナルがQM−Dabsyl又はFast−Redである、請求項39に記載の方法。
- 対象におけるPDの診断方法であって、
(a)PDを有することが疑われる対象由来の少なくとも1つの神経的形質を含む生体サンプルを得ることと、
(b)前記サンプルを抗PGP.5抗体と接触させ、リン酸化α−シヌクレインとPGP9.5との同時局在化を判定することにより、リン酸化α−シヌクレインが前記サンプルの前記神経的形質により局在化するか否かを検出することと、
(c)前記神経的形質において、リン酸化α−シヌクレインとPGP9.5との同時局在化があることが断定的に判定されたとき、前記対象をPDと診断することと、
を含む、方法。 - 対象におけるPDの診断及び治療方法であって、
(a)PDを有することが疑われる対象由来の少なくとも1つの神経的形質を含む生体サンプルを得ることと、
(b)前記サンプルを抗PGP.5抗体と接触させ、リン酸化α−シヌクレインとPGP9.5との同時局在化を判定することにより、リン酸化α−シヌクレインが前記サンプルの前記神経的形質により局在化するか否かを検出することと、
(c)前記神経的形質において、リン酸化α−シヌクレインとPGP9.5との同時局在化があることが断定的に判定されたとき、前記対象をPDと診断することと、
(d)PDを有すると診断された前記対象において、PDを治療する治療法を投与することと、
を含む、方法。 - PDと診断された対象の治療方法であって、前記対象に、α−シヌクレイン抗体の効果的な治療方式を投与することを含み、リン酸化α−シヌクレインを結合可能な抗体、及び、神経的形質を結合可能な抗体が、前記対象由来の皮膚サンプルの神経的形質内で同時局在化することが示されている、方法。
- PDを有すると判定された対象の治療方法であって、前記対象に、α−シヌクレイン抗体の効果的な治療方式を投与することを含み、前記対象が、請求項1〜25、又は30〜41のいずれか一項に記載の方法により、PDを有すると判定された方法。
- α−シヌクレイン抗体の前記効果的な治療方式が、α−シヌクレインの残基1〜20、α−シヌクレインの残基1〜10、α−シヌクレインの残基4〜15、α−シヌクレインの残基91〜99、α−シヌクレインの残基117〜123、α−シヌクレインの残基118〜126内で結合するモノクローナル抗体;プラシネズマブ(PRX002);抗体クローン1H7(ATCC寄託番号PTA−8220)のCDRを有するヒト化抗体;抗体クローン9E4(ATCC寄託番号PTA−8221)のCDR、抗体クローンNI−202.21D11のCDR、及び抗体クローンNI−202.12F4のCDRを有するヒト化抗体、例えば、米国特許第8,092,801号、同第8,609,820号、同第8,790,644号、同第8,940,276号、同第9,580,493号に開示されているα−シヌクレイン抗体などからなる群から選択されるα−シヌクレイン抗体を含む、請求項43又は請求項44に記載の方法。
- 前記リン酸化α−シヌクレインを結合可能な抗体が、Roche製のモノクローナル抗体クローン7E2、Roche製のモノクローナル抗体クローン3G2、Abcam製のモノクローナル抗体クローンMJF−R13(8−8)(P/N ab168381)、Abcam製のモノクローナル抗体P−syn/81A(P/N ab184674)、WAKO製のモノクローナル抗体クローンpSyn#64(P/N 015−25191)、Abcam製のモノクローナル抗体クローンLB509(P/N ab27766)、Prothena製のモノクローナル抗体クローン5C12(ATCC(登録商標)番号PTA−9197)、及び、Prothena製のモノクローナル抗体クローン11A5(ATCC(登録商標)番号PTA−8222)のうちの1つである、請求項1〜25、若しくは30〜31のいずれか一項に記載の方法、又は、請求項26〜29のいずれか一項に記載のキット。
- 前記神経的形質を結合可能な前記一次抗体が、Abcam製のモノクローナル抗体クローンEPR4118(P/N ab108986)、Abcam製のモノクローナル抗体クローン13C/I3C4(P/N ab8189)、又は、Cell Marque(商標)製の、RTD P/N 760−4434とのポリクローナル抗体のうちの1つである、請求項1〜25、若しくは30〜31のいずれか一項に記載の方法、又は、請求項26〜29のいずれか一項に記載のキット。
- リン酸化α−シヌクレインの検出方法であって、
(a)生体サンプルを、リン酸化α−シヌクレインを結合可能な一次抗体と接触させることと、
(b)前記リン酸化α−シヌクレインを結合可能な一次抗体を検出することと、
を含む、方法。 - 前記サンプルが、前記リン酸化α−シヌクレインを結合可能な一次抗体と接触する前に、少なくとも1つのプロテアーゼと接触する、請求項48に記載の方法。
- 前記サンプルを少なくとも1つのホスファターゼと接触させることを更に含む、請求項49に記載の方法。
- 前記検出することが組織化学的分析を含む、請求項48に記載の方法。
- 前記リン酸化α−シヌクレインを結合可能な一次抗体が、残基S129においてリン酸化したα−シヌクレインを検出する、請求項48に記載の方法。
- 前記リン酸化α−シヌクレインを結合可能な一次抗体が、7E2抗体クローン、又は3G2抗体クローンである、請求項52に記載の方法。
- 前記サンプルが固定されている、請求項48に記載の方法。
- 前記サンプルが、ホルマリン固定・パラフィン包埋(FFPE)サンプルである、請求項54に記載の方法。
- 前記サンプルが凍結サンプルである、請求項48に記載の方法。
- 工程(a)が、前記サンプルを、抱合体化した第1のラベルを有する第1の二次抗体と接触させることを更に含み、前記第1の二次抗体が、前記リン酸化α−シヌクレインを結合可能な一次抗体と免疫反応性である、請求項48に記載の方法。
- 前記サンプルを、前記第1の二次抗体の前記第1のラベルと反応性のひとまとまりの試薬と接触させて、前記サンプルのリン酸化α−シヌクレインの近くで、第1の検出可能なシグナルを生成することを更に含む、請求項57に記載の方法。
