Abstract
Description

本願は、電子方式の配列リストと共に提出されている。本配列リストは、2019年5月27日作成の「CD39−9_ST25」という名称で提供されており、サイズは、72kBである。本配列リストの電子方式の情報は、その全体において参照により本明細書中に組み込まれる。

本開示の抗体の例としては、抗体mAb1〜mAb24の何れか1つのVHドメインと、VLドメインとを含む抗体が挙げられる。抗CD39抗体mAb1〜mAb24のVH及びVL配列は、表A及びBに提供される(実施例13)。

本開示の抗CD39剤を使用して個体(特にヒト個体)を処置する方法も提供される。ある実施形態において、本開示は、ヒト患者への投与のための医薬組成物の調製における、本明細書で説明された抗体又は抗体フラグメントの使用を提供する。典型的には、この個体は、癌又は感染性疾患(例えば、ウイルス感染、細菌感染)に罹患しているか又はこれらのリスクがある。ある実施形態において、この個体は、循環及び/又は組織サンプル(例えば、腫瘍サンプル若しくは腫瘍隣接組織サンプル)において、検出可能な可溶性(細胞外)CD39タンパク質を有する。
a)循環又は腫瘍環境、任意選択的に腫瘍及び/又は隣接組織内において可溶性(細胞外)CD39タンパク質及び/又はCD39発現細胞を検出することと、
b)任意選択により、参照レベル(例えば、健常組織で観察されるレベル、任意選択により健康な個体又は抗CD39抗体から実質的な利益を引き出していない個体に対応するレベル)と比較して増加しているレベルにおいて、可溶性(細胞外)CD39タンパク質及び/又はCD39発現細胞が循環又は腫瘍環境中に含まれていると決定すると、この個体に抗CD39抗体を投与することと
を含む方法を提供する。CD39発現細胞は、腫瘍細胞又は白血球、例えば循環細胞又は腫瘍浸潤細胞、例えばCD4T細胞、CD8T細胞、TReg細胞、B細胞を含み得る。
a)この個体が、任意選択により循環、任意選択により腫瘍及び/又は隣接組織において、検出可能な可溶性(細胞外)CD39を有するかどうかを評価することと、
b)任意選択により、参照レベル(例えば、健康な個体又は本開示の抗CD39抗体から実質的な利益を引き出していない個体に対応する)と比較して増加しているレベルにおいて、可溶性(細胞外)CD39タンパク質を検出すると、この個体に本開示の抗CD39抗体を投与することと
を含む方法を提供する。
a)腫瘍環境中において、任意選択により腫瘍及び/又は隣接組織内においてCD73発現細胞を検出することと、
b)腫瘍環境が、CD73発現細胞を、任意選択により参照レベル(例えば、健常組織で観察されるレベル)と比較して増加しているレベルにおいて含むと決定すると、可溶性ヒトCD39タンパク質に結合し、且つその活性を阻害する本開示の抗体をこの個体に投与することと
を含む方法を提供する。任意選択により、腫瘍環境内のCD73発現細胞を検出することは、個体から、癌組織及び/又は癌の近位若しくは周辺の組織(例えば、癌隣接組織、隣接非腫瘍組織又は正常隣接組織)を含む生物学的サンプルを得ることと、CD73発現細胞のレベルを(例えば、免疫組織化学アッセイを実行することにより)検出することとを含む。CD73発現細胞は、例えば、腫瘍細胞、CD4T細胞、CD8T細胞、B細胞を含み得る。
CD39突然変異体の生成
CD39突然変異体をPCRにより生成した。増幅した配列をアガロースゲル上で泳動させ、Macherey Nagel PCR Clean−Up Gel Extractionキット(リファレンス740609)を使用して精製した。次いで、各突然変異体に関して生成した精製済PCR産物をClonTech InFusionシステムにより発現ベクターに連結した。突然変異した配列を含むベクターをMiniprepとして調製し、次いで配列決定した。配列決定後、Promega PureYield(商標)Plasmid Midiprep Systemを使用して、突然変異した配列を含むベクターをMidiprepとして調製した。HEK293T細胞をDMEM培地(Invitrogen)中で増殖させ、InvitrogenのLipofectamine 2000を使用してベクターを遺伝子移入し、48時間にわたりCO2インキュベータ中で37℃においてインキュベートした後、導入遺伝子の発現に関して試験した。下記の表に示すように、突然変異体をHek−293T細胞に遺伝子移入した。下記の表1において標的となるアミノ酸突然変異を、配列番号1のナンバリングを使用して示す。
分子生物学
下記のプライマー:TACGACTCACAAGCTTGCCGCCACCATGGAAGATACAAAGGAGTC(配列番号41)(フォワード)及びCCGCCCCGACTCTAGATCACTTGTCATCGTCATCTTTGTAATCGACATAGGTGGAGTGGGAGAG(配列番号42)(リバース)を使用して、ヒトPBMC cDNAからhuCD39タンパク質をクローニングした。次いで、精製済PCR産物を、InFusionクローニングシステムを使用して発現ベクター中にクローニングした。精製ステップのためにタンパク質のC末端部分にM2タグ(FLAGタグ、配列番号44において下線を引いた)を追加しており、(例えば、配列番号44の)CD39細胞外ドメインタンパク質は、何れの実施形態においても、M2タグを欠くように任意選択により指定され得ることが認識されるであろう。
クローニングした配列の検証後、CHO細胞をヌクレオフェクト(nucleofect)し、次いで産生プールをサブクローニングして、huCD39タンパク質を産生する細胞クローンを得た。ローラー中で増殖したhuCD39クローンから上清を回収し、M2クロマトグラフィーカラムを使用して精製し、M2ペプチドを使用して溶出させた。次いで、精製済タンパク質をS200サイズ排除クロマトグラフィーカラム上にロードした。単量体に相当する精製済タンパク質をTBS PH7.5緩衝液で製剤化した。M2タグを有しないCD39−M2細胞外ドメイン組換えタンパク質のアミノ酸配列は、下記のとおりであった。
産生された可溶性CD39タンパク質の酵素活性の抗体による阻害を、存在するATPの量に比例して発光シグナルを生じる試薬の使用によりATP加水分解の評価を可能にするCell Titer Glo(商標)(Promega、リファレンスG7571)を使用して評価した。この方法では、可溶性CD39により媒介されるATP加水分解の阻害を評価し得る。簡潔に説明すると、100μg/ml〜6×10−3μg/mlの抗CD39抗体の用量範囲を37℃で1時間にわたり、方法セクションで説明したアミノ酸配列(配列番号44)を有する400ng/mlの可溶性組換えヒトCD39タンパク質と共にインキュベートした。このプレートに37℃でさらに30分にわたり20μMのATPを添加した後、CTG(Cell Titer Glo)試薬を添加した。暗所での5分間の短いインキュベーション期間後、Enspire(商標)ルミノメーターを使用して放射光を定量した。抗体の存在下での放射光と、ATPのみ(最大光放射)並びにATP及び可溶性CD39タンパク質(最小光放射)との比較により、抗CD39抗体の効力を決定した。
抗体による、CD39を発現する細胞におけるCD39酵素活性の阻害を、存在するATPの量に比例して発光シグナルを生じる試薬の使用によりATP加水分解の評価を可能にするCell Titer Glo(商標)(Promega、リファレンスG7571)を使用して評価した。そのため、このアッセイを、細胞培養上清中においてCD39により加水分解されるATPの阻害の評価を可能にするように設計した。簡潔に説明すると、5×104個のRamosヒトリンパ腫細胞、5×103個の、ヒトCD39を発現するCHO細胞、カニクイザルCD39を発現するCHO細胞及びマウスCD39を発現するCHO細胞を、30μg/ml〜5×10−4μg/mlの抗CD39抗体と共に37℃で1時間インキュベートした。次いで、細胞を37℃でさらに1時間にわたり20μMのATPと共にインキュベートした。プレートを400gで2分にわたり遠心分離し、細胞上清50μlを発光マイクロプレート(白色ウェル)に移す。この上清にCellTiter−Glo(商標)Reagent(CTG)50μlを添加し、暗所での5分間のインキュベーション後、Enspire(商標)ルミノメーターを使用して放射光を定量した。抗体の存在下での放射光と、ATPのみ(最大光放射)並びにATP及び細胞(最小光放射)との比較により、抗CD39抗体の効力を決定した。
抗ヒトCD39抗体を得るために、上記で説明した組換えヒトCD39−M2細胞外ドメイン組換えタンパク質によりBalb/cマウスに免疫付与した。マウスに、腹腔内で、CD39タンパク質50μg及びComplete Freund Adjuvantの乳濁液による1回の1回目の免疫付与を行い、腹腔内でCD39タンパク質50μg及びIncomplete Freund Adjuvantの乳濁液による2回目の免疫付与を行い、最後に静脈内でCD39タンパク質10μgによる追加免疫を行った。免疫脾臓細胞を追加免疫の3日後にX63.Ag8.653不死化B細胞と融合させ、放射線照射した脾臓細胞の存在下で培養した。ハイブリドーマを半固体メチルセルロース含有培地に播種し、増殖中のクローンを、Clonepix(商標)2装置(Molecular Devices Corp.)を使用して採取した。
抗体がどのように細胞性CD39の酵素(ATPアーゼ)活性を阻害することができるかについての洞察を得るために、本発明者らは、細胞アッセイにおいてCD39のATPアーゼ活性を阻害することが報告されている抗体(国際公開第2009/095478号パンフレットで開示されたBY40)が結合するエピトープを調べた。
細胞アッセイにおいてCD39のATPアーゼ活性を阻害することが報告されている2種の抗体(BY40及びBY12)を、組換え可溶性CD39タンパク質のATPアーゼ活性を阻害することができるかどうかを決定するために評価した。上記で説明したように産生された可溶性CD39タンパク質の酵素活性の抗体による阻害を、Cell Titer Glo(商標)(Promega、リファレンスG7571)を使用して評価した。細胞性CD39タンパク質の酵素活性の抗体による阻害を上記で示したように評価した。
sCD39のATPアーゼ活性を中和する抗体を探すために一連の免疫付与を実行した。抗ヒトCD39抗体を得るために、上記で説明した組換えヒトCD39−M2細胞外ドメイン組換えタンパク質で動物に免疫付与した。まとめると、免疫付与の系列は、様々なプロトコルを含み、且つ様々なマウス株、ラット及びウサギを含む様々な動物におけるものであった。
本発明者らは、BY40様抗体と競合しない抗体を有するために、Q96、N99、E143及びR147領域(突然変異体5により定義される)に結合しない抗CD39抗体を特定しようとした。CD39のATPアーゼ活性をブロックする能力を必要としないそのような抗体は、例えば、細胞性CD39を阻害するBY40抗体又はBY40様抗体の存在下で細胞上の遊離CD39タンパク質を検出して定量するための、BY40結合部位に結合する細胞性CD39を阻害する抗体の薬理学的研究に有用であり得る。
I−394重鎖可変ドメイン配列:
可溶性CD39の抗体により媒介される阻害が可能であることを示す実施例4の結果に基づいて、sCD39のATPアーゼ活性を中和する抗体を探すために、実施例3と異なるプロトコルを使用する異なる免疫付与からの融合を再検討した。
− ハイブリドーマ上清300μlにプロテインAビーズ10μlを添加する
− 最終濃度が1,5μMとなるようにNaClを添加する
− 4℃で3〜4時間にわたりチューブを回転させる
− 1500rpmで1分間遠心分離する
− 上清を除去し、TBS 1mlによる3回の洗浄を実施する
− 3回目の洗浄後にTBSを全て除去する
− クエン酸塩0,1M pH3 50μlを添加し、ホモジナイズし、次いで5分にわたりRTでインキュベートする
− 1500rpmで1分にわたりビーズを遠心分離する
− 溶出液50μlを回収し、TBS 450μlを速やかに添加し、次いで4℃で貯蔵する。
実施例4で示すように、I−394は、CD39の突然変異体19を発現する細胞への結合の完全な喪失を示したが、突然変異体5への結合を喪失していなかった。実施例5のさらなる抗CD39抗体のエピトープを定義するために、これらを、実施例1及び表1で説明されているCD39突然変異体のパネルへの結合の喪失に関して試験した。配列番号1のナンバリングを使用して表1に示す突然変異体をHek−293T細胞に遺伝子移入した。試験抗体の用量範囲(10〜2.5〜0.625〜0.1563〜0.0391〜0.0098〜0.0024〜0.0006μg/ml)を、フローサイトメトリーにより、20種の生成された突然変異体で試験する。
抗体I−394を、ヒトCD39を発現するCHO細胞、カニクイザル(マカカ・ファスキクラリス(macaca fascicularis))CD39を発現するCHO細胞、マウスCD39を発現するCHO細胞及びヒトRamosリンパ腫細胞(ATCC(商標)、リファレンスCRL−1596)への結合に関して2回の反復実験で試験した。細胞を4℃で30分にわたり30μg/ml〜5×10−4μg/mlの様々な濃度の非標識抗CD39と共にインキュベートした。洗浄後、4℃で30分にわたり細胞をヤギ抗マウスH+L標識二次抗体と共にインキュベートした。
上記(方法)で説明した細胞性CD39の酵素活性の阻害に関して使用したアッセイを使用して、CD39発現細胞中におけるCD39のATPアーゼ活性の抗体I−394による阻害を評価した。
上記(方法)で説明した可溶性CD39の酵素活性の阻害に関して使用したアッセイを使用して、可溶性CD39タンパク質のATPアーゼ活性の抗体I−394による阻害を評価した。結果を図6に示す。I−394は、可溶性CD39タンパク質の酵素活性を阻害する。比較すると、抗体BY40は、可溶性CD39タンパク質の酵素活性を阻害しなかった。算出したIC50は、0.003μg/mlである。阻害最大値は、74.9%であった。
抗体I−394を、一晩4℃で500ng/ml又は1μg/mlにおいてPBS 1Xで96ウェルプレートにコーティングした、下記に示すアミノ酸配列を有する組換えヒトCD39アイソフォーム(Rec−huCD39アイソフォーム)への結合に関して試験した。ウェルをTBS Tween 20で洗浄し、TBSブロッキング緩衝液でRTにおいてさらに2時間飽和させた。一次抗体の用量範囲濃度をRTで2時間にわたりTBSブロッキング緩衝液中でインキュベートした。ウェルをTBS Tween 20で洗浄した。二次抗体(TBSブロッキング緩衝液中のGAM−HRP又はGAH−HRP)をRTで1時間にわたりインキュベートし、TMBで明らかにした。光学密度をOD=450でEnspire(商標)により測定した。
ヒトCD39−L1(NTPDアーゼ2又はENTPD2としても既知である):
ATPは、炎症誘発活性を有するが、ATPのCD39により媒介される異化により、樹状細胞(DC)活性化を損ない得、次いで腫瘍抗原に対するより広範な適応免疫反応が変化すると考えられる。抗CD39抗体を使用するCD39遮断が、ATPの存在下での樹状細胞(DC)活性化のCD39により媒介される変化を克服し得たかどうかを評価するために、本発明者らは、ATPの存在下で抗CD39抗体と共に単球由来のDC(moDC)をインキュベートした。
T細胞増殖アッセイ
健常ドナーからの末梢血をEFSから入手し、単核細胞をFicoll(商標)勾配上で単離した。リンパ球を52%のPercoll(商標)勾配上で細胞ペレットの回収によりさらに濃縮し、Cell Trace色素(Thermofisher)により、製造業者から提供されるTDSに従って染色した。染色細胞の5×104〜1×105個を96丸底プレート中に分配し、抗huCD73抗体(国際公開第2016/131950号パンフレットに記載の抗体6E1)及び/又は抗huCD39 Ab(本明細書に記載のI−394)と共に37℃において1時間インキュベートし、抗CD3/抗CD28コーティングビーズ(ビーズ:細胞=1:4;Life Technologies)の添加により3〜5日間活性化させた。T細胞増殖の阻害をATP(200μM)の添加により達成した。T細胞増殖及びAMPの免疫抑制効果をブロックするAbの能力を、フローサイトメトリーにより、増殖T細胞サブセットにおける色素希釈を定量することにより評価した。
抗体を、腫瘍環境中で見出され得る条件を表すことを意図する、添加されたATPの存在下でCD4又はCD8T細胞増殖を回復させる能力に関して検査した。抗CD73及びCD39のそれぞれを抗CD73又は抗CD39抗体の他方の3つの異なる用量における用量範囲において検査した。抗CD39抗体I−394は、CD4又はCD8T細胞増殖の回復において抗CD73抗体の効果を強力に増強し、その結果、低濃度の抗CD73抗体(例えば、0.01μg/ml未満、0.001μg/ml未満及びさらに0.001μg/ml未満)であっても、抗CD39抗体と組み合わせて使用した場合、CD4又はCD8T細胞増殖を強力に増強させた。さらに、抗CD73を用いず単独の用量範囲で検査した場合、抗CD39抗体I−394は、0.1μg/ml及び1μg/mlの濃度においてCD4又はCD8T細胞増殖の顕著な増強をもたらした。図12Aは、抗CD39抗体I−394の3つの異なる用量、0.01μg/ml、0.1μg/ml及び1μg/mlの何れかでのCD4T細胞増殖に対する抗CD73抗体6E1の用量範囲を示す。可溶性及び/又は単量体ヒトCD39を中和することができる抗CD39抗体は、CD4T細胞増殖の回復において抗CD73抗体での効果の強力な増強を示す。この効果は、抗CD73抗体が、抗CD73抗体での処理の過程中に腫瘍組織において観察され得る濃度範囲に対応する準最適活性である濃度において特に強力であった。0.01μg/mlの濃度において、抗CD39抗体は、抗CD73抗体の効力のおよそ1−logの増加を提供し、0.1μg/mlの濃度において、抗CD39抗体は、抗CD73抗体の効力のおよそ4−logの増加を提供した。従って、抗CD39抗体は、特に腫瘍組織中において、例えばCD73発現細胞を有する腫瘍中で抗CD73抗体の活性を増強させるために有用であり得る。さらに、検査した抗CD73抗体(可溶性CD73タンパク質を中和することができる)は、CD4T細胞増殖を回復させる高い能力を有した一方、他の抗体は、より低い効力を有し(例えば、酵素阻害アッセイにおいて、T細胞増殖アッセイ又は他の適切なアッセイにおいて評価)、抗sCD39抗体との組合せからより一層利益を受け得る。図12Bは、CD8T細胞増殖に対する抗CD73抗体の用量範囲を示す。ここでも、抗CD39抗体は、CD8T細胞増殖の回復において抗CD73抗体との強力な相乗及び/又は相加効果を示す。この効果は、抗CD73抗体が、抗CD73抗体での処理の過程中に腫瘍組織において観察され得る濃度範囲に対応する準最適活性である濃度において特に強力であった。
配列番号6及び7のVH及びVLアミノ酸配列をそれぞれ有する親抗体I−394を、ヒトサブグループIGHV1−3由来の重鎖フレームワーク(FR1、FR2、FR3)をIGHJ1*01(FR4)と一緒にVH中に導入し、且つヒトサブグループIGKV4−1由来の軽鎖フレームワーク(FR1、FR2、FR3)をIGKJ4*01(FR4)と一緒にVL中に導入することにより改変した。
I−394−LP(親Fab軽鎖):
mAb1〜24を、種々のアッセイによりCD39の結合及び阻害に関して、例えば特に高レベルのCD39を発現することが見出されたRamos腫瘍細胞系上に存在するCD39への結合、組換え産生sCD39の酵素活性の阻害並びにヒトCD39を発現するCHO細胞及びRamos腫瘍細胞からの細胞培養上清中で排出されるsCD39タンパク質の酵素活性の阻害に関してさらに評価した。
全てL234A/L235E/G237A/A330S/P331S置換を有するヒトIgG1アイソタイプとして産生された抗体mAb1〜24及び従来技術の抗CD39抗体BY40をおよそ7mg/mLの濃度における下記の参照処方物中の安定性に関して検査した:pH6.0;ヒスチジン緩衝液(10mM);スクロース(200mM);NaCl(50mM);ポリソルベート80(PS80)(0.2g/L)。処方物の安定性を2つの貯蔵条件(+5℃±3℃及び+40±3℃においてモニタリングした。それぞれの試験のため、3つの時点:T0、T15D(15日間)及びT1M(1ヶ月)を実施した。凍結融解(F/T)及び熱シフト安定性アッセイ(TSSA)をフォーマット比較のために実行した。F/Tサイクルを実施するため、サンプルを−20℃において少なくとも2時間凍結させ、室温において少なくとも1時間融解させ、このF/Tサイクルを3回繰り返し、サンプルを最後の凍結/融解サイクルの24時間後に検査した。それぞれの時点において、下記の検査を実施した。
・粒子状物質(MFI)
・目視調査(外観)
・不純物(SE−HPLC)
・濁度(400nm)
・タンパク質濃度(280nm)(Nanodrop、Thermo Fisher Scientific Inc.を用いて実施)

- ヒトCD39ポリペプチドに結合し、且つ可溶性細胞外ドメインヒトCD39ポリペプチドのATPアーゼ活性を阻害することができる抗体又は抗体フラグメントであって、配列番号31のアミノ酸配列を含む重鎖可変領域(VH)と、配列番号36又は37のアミノ酸配列を含む軽鎖可変領域(VL)とを含む抗体又は抗体フラグメント。

- 前記抗体は、外因的に添加されたATPの存在下で前記CD39タンパク質のATPアーゼ活性を阻害し、任意選択により、外因的に添加されたATPは、20μMの濃度で提供される、請求項1〜18の何れか一項に記載の抗体又は抗体フラグメント。
- 前記抗体は、細胞の表面におけるヒトCD39タンパク質に結合することができ、且つ細胞の表面における前記CD39タンパク質のATPアーゼ活性を阻害することができる、請求項1〜19の何れか一項に記載の抗体又は抗体フラグメント。
- 前記抗体は、ヒトCD39発現腫瘍細胞からの細胞培養上清中で排出されるsCD39タンパク質のATPアーゼ活性の50%超、任意選択により70%超、任意選択により80%超の低下を生じさせることができる、請求項1〜20の何れか一項に記載の抗体又は抗体フラグメント。
- 前記抗体は、CD39発現細胞による前記細胞外ATPアーゼ活性の少なくとも80%の低下を生じさせることができる、請求項1〜21の何れか一項に記載の抗体又は抗体フラグメント。
- 前記抗体は、1μg/ml以下の、CD39のATPアーゼ活性の阻害に関するEC50によって特徴付けられ、CD39の酵素活性の阻害は、ATPを定量することにより、Ramos細胞中におけるATPアーゼ活性の中和を評価することによって決定される、請求項1〜22の何れか一項に記載の抗体又は抗体フラグメント。
- 前記抗体は、Ramos細胞への結合に関して、2μg/ml以下、任意選択により1μg/ml以下、0.5μg/ml以下、0.1μg/ml以下の、フローサイトメトリーによって決定されるEC50によって特徴付けられる、請求項1〜23の何れか一項に記載の抗体又は抗体フラグメント。
- 前記抗体は、配列番号1のCD39ポリペプチドへの結合に関して、配列番号31及び36のそれぞれの重鎖及び軽鎖可変領域アミノ酸配列を含む抗体と競合する、請求項1〜24の何れか一項に記載の抗体又は抗体フラグメント。
- 前記抗体は、ATPの存在下で樹状細胞の活性化を増加させることができる、請求項1〜25の何れか一項に記載の抗体又は抗体フラグメント。
- 前記抗体は、単球由来の樹状細胞が前記抗体及びATPと共にインビトロでインキュベートされる場合、前記moDC中における活性化の細胞表面マーカーの発現の増加を生じさせることができる、請求項1〜26の何れか一項に記載の抗体又は抗体フラグメント。
- 外因的に添加されたATPは、0.125mM、0.25mM又は0.5mMで提供される、請求項26又は27に記載の抗体又は抗体フラグメント。
- 活性化の細胞表面マーカーの発現の増加は、24時間にわたってATPの存在下でmoDCをインキュベートし、且つフローサイトメトリーによってmoDC上でCD80、CD83及び/又はHLA−DRの細胞表面発現を分析することによって評価される、請求項26〜28の何れか一項に記載の抗体又は抗体フラグメント。
- 細胞表面マーカーの発現の前記増加は、陰性対照(例えば、培地)と比較して少なくとも40%、50%、75%又は80%である、請求項27〜29の何れか一項に記載の抗体又は抗体フラグメント。
- 前記抗体は、ATPの存在下において、CD39を発現するDC細胞と共にT細胞がインビトロで共培養された場合にT細胞増殖を増加させることができる、請求項1〜30の何れか一項に記載の抗体。
- 前記抗体は、前記抗体と、配列番号1のアミノ酸配列を含む野生型CD39ポリペプチドとの間の結合と比較して、それぞれの場合に突然変異R138R、M139A及びE142K(配列番号1を基準にする)を含む突然変異体CD39ポリペプチドへの減少した結合を有する、請求項1〜31の何れか一項に記載の抗体又は抗体フラグメント。
- 前記抗体は、裸の抗体、任意選択により細胞傷害剤に結合されていない抗体である、請求項1〜32の何れか一項に記載の抗体又は抗体フラグメント。
- 前記抗体は、抗体フラグメント、任意選択によりFcドメインを欠く抗体フラグメントである、請求項1〜33の何れか一項に記載の抗体又は抗体フラグメント。
- 前記抗体は、ヒトFcドメインを有する抗体であり、前記ヒトFcドメインは、前記FcドメインとヒトFcγ受容体との間の結合を減少させるように改変されている、請求項1〜34の何れか一項に記載の抗体。
- 前記抗体は、ヒトCD16、CD32a、CD32b及び/又はCD64ポリペプチドへの結合を実質的に欠く、請求項1〜35の何れか一項に記載の抗体又は抗体フラグメント。
- 前記抗体は、Fcドメインを含み、任意選択により、前記Fcドメインは、野生型Fcドメインと比較して、ヒトFcγ受容体CD16A、CD16B、CD32A、CD32B及び/又はCD64の1つ若しくは複数又は全てへの減少又は消失した結合を有するアミノ酸改変を含む、請求項1〜36の何れか一項に記載の抗体又は抗体フラグメント。

- 改変されたヒトIgG1 Fcドメインを含み、前記改変されたヒトIgG1 Fcドメインは、Kabat残基N297においてN結合グリコシル化を含み、且つKabat残基234及び235、任意選択によりさらにKabat残基331、任意選択によりKabat残基234、235、237並びにKabat残基330及び/又は331においてアミノ酸置換を含み、任意選択により、前記Fcドメインは、L234A/L235E/P331S置換、L234F/L235E/P331S置換、L234A/L235E/G237A/P331S置換又はL234A/L235E/G237A/A330S/P331S置換を含む、請求項1〜38の何れか一項に記載の抗体又は抗体フラグメント。
- 請求項1〜39の何れか一項に記載の抗体と、薬学的に許容可能な担体とを含む医薬組成物。
- 請求項1〜39の何れか一項に記載の抗体又は抗体フラグメントを含み、任意選択により請求項1〜39の何れか一項に記載の抗体又は抗体フラグメントを特異的に認識する標識された二次抗体又は抗体フラグメントをさらに含むキット。
- 請求項1〜39の何れか一項に記載の抗体又は抗体フラグメントの重鎖及び/又は軽鎖をコードする核酸。

- 前記個体は、癌を有する、請求項45に記載の方法。
- 癌を有する対象においてT細胞活性、NK細胞活性及び/若しくはB細胞活性を増加させ、且つ/又は癌を有する対象においてT細胞活性、NK細胞活性及び/若しくはB細胞活性のアデノシンにより媒介される阻害を軽減するための方法であって、請求項1〜39の何れか一項に記載の抗体若しくは抗体フラグメント又は請求項40に記載の組成物の有効量を前記対象に投与することを含む方法。
- 癌を有する対象において樹状細胞の活性化を増加させ、且つ/又は癌を有する対象においてDC細胞のATPにより媒介される活性化を回復させるための方法であって、請求項1〜39の何れか一項に記載の抗体若しくは抗体フラグメント又は請求項40に記載の組成物の有効量を前記対象に投与することを含む方法。
- 前記個体は、検出可能な可溶性CD39タンパク質を有する、請求項44〜48の何れか一項に記載の方法。
- 前記個体は、循環、腫瘍組織及び/又は腫瘍隣接組織において、検出可能な可溶性CD39タンパク質を有する、請求項49に記載の方法。
- 癌の処置又は予防を、それを必要とする個体において行うための方法であって、
a)循環及び/又は腫瘍環境において可溶性CD39タンパク質を検出することと、
b)任意選択により、参照レベルと比較して増加しているレベルにおいて、可溶性CD39タンパク質が循環及び/又は腫瘍環境中に含まれていると決定すると、請求項1〜39の何れか一項に記載の抗体若しくは抗体フラグメント又は請求項40に記載の組成物を前記個体に投与することと
を含む方法。 - 可溶性CD39タンパク質を検出することは、前記個体から生物学的サンプルを得ることと、前記サンプルを、可溶性CD39タンパク質に結合する抗体と接触させることと、可溶性CD39タンパク質に結合された抗体を検出することとを含む、請求項51に記載の方法。
- 抗CD39抗体は、可溶性CD39タンパク質の酵素活性の中和に関する少なくともEC50に対応する血液(血清)及び/又は腫瘍組織中における濃度を達成するのに有効な量で少なくとも1回投与される、請求項44〜52の何れか一項に記載の方法。
- 可溶性CD39タンパク質の酵素活性の中和は、前記CD39タンパク質が抗CD39抗体又は抗体フラグメントと共にインキュベートされるときに加水分解されたATPの減少を定量することにより、溶液中におけるCD39細胞外ドメインタンパク質のATPアーゼ活性の中和を評価することによって決定される、請求項44〜53の何れか一項に記載の方法。
- 前記腫瘍又は癌は、固形腫瘍である、請求項44〜51の何れか一項に記載の方法。
- 前記腫瘍又は癌は、頭頸部扁平上皮癌、膀胱癌、卵巣癌、結腸直腸癌、メラノーマ、胃癌、食道癌又は乳癌である、請求項55に記載の方法。
