I.概要
本発明は、部分的には、種々のウイルス性または非ウイルス性標的(例えば、HIV−1 Env、エボラGP、HCV E2タンパク質または結核菌抗原)に対する新規ワクチン免疫原の、ならびに安定性および活性の改善を示すナノ粒子提示免疫原(すなわち、ワクチン組成物)の、本発明者らの開発に基づく。典型的には、本発明のワクチンまたはワクチン組成物は、自己組織化ナノ粒子またはウイルス様粒子(VLP)上に提示された免疫原ポリペプチドまたはタンパク質(例えば、HIV−1 Env由来三量体タンパク質)を含有する。ナノ粒子ワクチンは、本明細書に記載されている1以上の新規構造成分をも含有する。これらの追加的構造成分は、ナノ粒子の表面上の免疫原の提示を促進させるように、および/または提示免疫原の安定性を増強するように、および/または自己組織化タンパク質ワクチンの収量および純度を改善するように機能する。幾つかの実施形態においては、本発明のナノ粒子ワクチンは、ナノ粒子を安定化させるロッキングドメインを含有する。本明細書の実施例に詳細に記載されているとおり、ロッキングドメインはナノ粒子を内部から安定化させ、その結果、免疫原ポリペプチド(例えば、HIV−1 Env由来三量体タンパク質)を提示するナノ粒子は製造、ワクチン製剤化および免疫化において無傷のままとなりうる。このようにして構築された新規ワクチン免疫原は、有意に増強した安定性を有する。また、ロッキングメカニズムはナノ粒子プラットフォームには左右されない。本明細書に例示されているとおり(例えば、HIV−1ワクチン)、それは、異なるナノ粒子、例えば60量体I3−01およびE2pナノ粒子に適用可能であり、SEC、BN−PAGE、DSC、ネガティブ染色EMおよび抗原プロファイリングにより示されるとおり、ほぼ同じ結果をもたらす。

VII.ポリヌクレオチドおよび発現構築物
本発明のワクチン組成物は、典型的には、まず、本明細書に記載されている種々の構造成分の機能的連結コード配列を含有する発現構築物(すなわち、発現ベクター)を作製することにより製造される。したがって、幾つかの関連態様においては、本発明は、本明細書に記載されている新規構造成分(例えば、ロッキングドメインで安定化されたHIV−1 Env三量体提示ナノ粒子)を含有するナノ粒子提示免疫原をコードする実質的に精製されたポリヌクレオチド(DNAまたはRNA)、ならびにそのようなポリヌクレオチドを含有する発現ベクター(例えば、本明細書に例示されているCMVベクター)、およびワクチン免疫原を製造するための宿主細胞(例えば、本明細書に例示されているExpiCHO細胞)を提供する。該ポリヌクレオチドによりコードされる、または該ベクターから発現される融合ポリペプチドも、本発明に含まれる。本明細書に記載されているとおり、そのようなポリペプチドは、表面上に免疫原ポリペプチドまたはタンパク質を提示するナノ粒子ワクチンへと自己組織化する。

以下の実施例は本発明を例示するために記載されており、本発明を限定するものではない。
表面上に提示されたHIV−1 BG505 UFO三量体を含有する60量体E2PおよびI3−01ナノ粒子に関して、種々のロッキングドメイン(LDS)の有用性を検証した。構築物の設計を図6に図示する。LD含有ナノ粒子を100mLのExpiCHO細胞において一過性に発現させ、ついで2G12またはPGT145抗体カラムを使用して精製した。ついでスーパーロース(Superose)6 10/300 GLカラムでのサイズ排除クロマトグラフィー(SEC)により、ナノ粒子サンプルをそれらの収量および純度に関して特徴づけした。SECプロファイルは、ほとんどのLDがHIV−1 BG505 UFO gp140ナノ粒子を種々の度合の収量および純度で安定化させうることを示した。E2pの場合には、LD4およびLD5は、高品質BG505 UFO gp140ナノ粒子を他のLDより高い収量で与え、一方、I3−01の場合には、LD4、LD5およびLD7はナノ粒子の収量および純度の点で他のLDより優れていた。注目すべきことに、E2p−LDxナノ粒子は2G12抗体カラムにより精製され、これは部分的組織化ナノ粒子および単一三量体をも上清から抽出し、一方、I3−01−LDxナノ粒子はPGT145抗体カラムにより精製され、これは完全組織化ナノ粒子に非常に特異的である。したがって、E2p−LDxとI3−01−LDxとの間の純度の差異は、ナノ粒子プラットフォームやLD設計によってではなく、精製に使用された抗体カラムによって生じたものである。
LD含有HIV−1 BG505 gp140ナノ粒子の組織化(集合、assembly)を更に検証するために、まず、ブルー・ネイティブ・ポリアクリルアミドゲル電気泳動(BN−PAGE)によりE2p−LDxおよびI3−01−LDxナノ粒子を分析した。結果を図7に示す。ナノ粒子は大きなサイズおよび高い分子量を有するため、予想通り、BN−PAGEは低分子量バンドを何ら示さず、全てのサンプルがレーン最上部のウェル内に捕捉されることを証明した。選択されたE2p−LDxおよびI3−01−LDx構築物をネガティブ染色電子顕微鏡法(EM)により分析し、これは、良好に形成されたナノ粒子を示した。EM生イメージにおいては、より低いストリンジェンシーの抗体カラムを使用したため、E2p−LDxナノ粒子は単一三量体または部分的組織化ナノ粒子と時々混合していた。それでも、BN−PAGEおよびネガティブ染色EM分析は、ロッキングドメインが、天然様HIV−1 gp140三量体を提示するための2つの大きなナノ粒子プラットフォームを有効に安定化させうることを示した。
2つの構築物を第1世代ワクチン構築物として選択した。それぞれはBG505 UFO.664 gp140、10アミノ酸GSリンカー(I3−01専用)、ナノ粒子サブユニット(E2pまたはI3−01のいずれか)、5アミノ酸GSリンカー、自己組織化に際してナノ粒子バックボーンを安定化させるロッキングドメイン(LD)、およびB細胞発生をbNAbへと導く強力な濾胞性Tヘルパー(Tfh)応答を誘導する汎反応性T細胞エピトープ(PADREエピトープ)をコードしている。哺乳類発現に一般に使用されるCMVベクターに基づいてDNAプラスミドを設計し、この場合、「抗原」遺伝子をベクター内に挿入した(図2)。これらの2つのナノ粒子構築物は「UFO−E2p−LD4−PADRE」および「UFO−10GS−I3−01−LD7−PADRE」と称される。ロッキングメカニズムが図2の右隅に図示されている。簡潔に説明すると、二量体LDタンパク質のN末端はナノ粒子サブユニットのC末端に融合している。自己組織化中に、2つのナノ粒子サブユニットが一緒になって二量体界面を形成すると、それらはそれらの結合LDを接近させて、第2の遥かに強力な界面をナノ粒子殻の直下に形成し、これはその安定性を有意に増強する。
ワクチンは精製ナノ粒子タンパク質であり、これは液相中でアジュバントと混合される。該タンパク質物質は一過性ExpiCHO細胞または安定化CHO−S細胞のいずれかにおいてGMP施設で製造されうる。製造プロセスは、3つの工程、すなわち、CHO細胞における発現、PGT145アフィニティーカラムによる精製および品質管理(QC)を含む。
CHO/ExpiCHO製造ナノ粒子タンパク質の品質は、(1)収量および純度に関してはSuperose 6 10/300 GLカラムおよびブルー・ネイティブ・ポリアクリルアミドゲル電気泳動(BN−PAGE)により、(2)熱安定性に関してはMicroCal VP−キャピラリー熱量計(Malvern)での示差走査熱量測定(DSC)により、(3)抗原性に関してはOctet RED96、定量バイオセンサーおよびbNAbと非NAbとのパネルを使用するバイオレイヤー(bio−layer)干渉法(BLI)により、ならびに(4)ナノ粒子組織化および構造的完全性に関してはネガティブ染色電子顕微鏡法(EM)により評価されうる。QCプロセスは、選択されたナノ粒子構築物に関して本発明者らの研究室において検証されている(後記を参照されたい)。注目すべきことに、QC工程(1)〜(2)は、タンパク質の発現、製造および精製の後、任意のGMP施設の現場で容易に行われうる。
簡便で頑強な製造プロセスならびに優れた生化学的および生物物理学的特性に加えて、更なるインビトロおよびインビボ評価は、ロッキングメカニズムを有するこれらのUFO gp140ナノ粒子が遥かに最も有望なHIV−1ワクチン候補となりうることを示した。
本発明者らはフィロウイルスに対するナノ粒子ワクチンを開発した。エボラウイルス(EBOV)およびマールブルグウイルスのようなフィロウイルスはヒトおよび非ヒト霊長類(NHP)において致命的な出血熱を引き起こしうる。フィロウイルスはヒトの疾患の突発を過去に引き起こしているが、エボラウイルスは2013〜2016年の史上最大の突発の唯一の原因であり、これはアフリカの9カ国に広がり、28,600の症例および11,325人の死亡をもたらした。現在、コンゴ民主共和国(DRC)でエボラ出血熱の突発が続いており、600人以上の命を奪っている。フィロウイルス糖タンパク質(GP)は、付着および膜融合を開始させることにより細胞侵入をもたらし、ワクチン設計の主要標的である。GPは、ヒト生存者および免疫化動物から単離された中和抗体により認識されうる(Saphireら,Cell 174(4):938−952,2018を参照されたい)。
同じ設計戦略に基づいて、本発明者らはアレナウイルスに対するナノ粒子ワクチンを開発した。アレナウイルスもヒトにおいて重度の出血熱を引き起こすことが公知である。ラッサマンマレナウイルス(LASV)はラッサ熱の原因因子であり、西アフリカにおける重大な公衆衛生問題である。LASV糖タンパク質複合体(GPC)は、受容体結合サブユニットGP1と膜貫通型融合媒介性サブユニットGP2とをそれぞれが含有するヘテロ二量体の三量体である。HIV−1 gp160およびエボラGPのような他のクラスI融合タンパク質と同様に、LASV GPCは細胞侵入をもたらし、中和抗体応答のための主要標的となる。現在、LASV感染を予防するために利用可能なワクチンは存在しない。最近、ヒト中和抗体と複合体形成したLASV GPCの結晶構造が報告された(Hastieら,356(6341):923−928,2017を参照されたい)。
本発明者らは更に、呼吸器合胞体ウイルス(RSV)に対するナノ粒子ワクチンを開発した。ヒト呼吸器合胞体ウイルス(hRSV)感染は乳児、幼児および高齢者における細気管支炎および入院の主要原因である。融合糖タンパク質(F)は細胞侵入をもたらし、ワクチン設計の主要標的である。Fは、感染ドナーから単離された中和抗体により認識されうる。HIV−1 gp160、エボラGPおよびラッサGPCと同様に、hRSV FはクラスI膜貫通表面タンパク質であり、N末端にシグナルペプチド(アミノ酸1〜25)を有し、C末端付近に膜アンカーを有する。Fは最初に不活性前駆体タンパク質F0として合成され、これはフューリンまたはフューリン様細胞エンドプロテアーゼによるトランスゴルジ複合体における切断による活性化に際してホモ三量体へと組織化する。該切断はNH2−F2−F1−COOHの形態の2つのジスルフィド結合サブユニットを与える。F1のN末端は、該切断の結果として、融合ペプチド(アミノ酸137〜154)を含有し、これは、宿主細胞膜内に直接挿入されて融合を誘発する疎水性ペプチドである。F1サブユニットは、ヘプタッド反復(7アミノ酸繰り返し)(HR)の、2つの領域をも含有し、これらは融合中に結合し、ウイルス膜と細胞膜とを接近させる。中和抗体と複合体形成した融合前Fの結晶構造は原子分解能で決定されている(McLellanら,340(6136):1113−1117,2013を参照されたい)。
本発明者らはまた、コロナウイルス(CoV)に対するナノ粒子ワクチンを開発した。CoVは、プラス鎖RNAゲノムを有するエンベロープウイルスである。2002年のアジアにおける重症急性呼吸器症候群(SARS)の突発は新たなコロナウイルスの発見につながり、それは後にSARS−CoVと命名された。SARS−CoVは突発中に8000人以上に感染し、致死率は約10%であった。2012年に、コロナウイルスの新種である中東呼吸器症候群コロナウイルス(MERS−CoV)が特定され、それ以来、該ウイルスは27か国で2000人以上に感染し、致死率は約35%であった。どちらのコロナウイルスにおいても、ウイルスゲノムはスパイク(S)、エンベロープ(E)、膜(M)およびヌクレオカプシド(N)構造タンパク質をコードし、そのうちのSタンパク質はS1サブユニットにおける受容体結合ドメイン(RBD)を介した宿主受容体への結合ならびにそれに続く膜融合およびウイルス侵入をもたらす。RBDはコアサブドメインおよび受容体結合モチーフ(RBM)を含有する。コアサブドメインはそれらの2つのコロナウイルス間で非常に類似しているが、それらのRBMは、異なる受容体特異性を示す。すなわち、SARS−CoVはアンジオテンシン変換酵素2(ACE2)を認識し、一方、MERS−CoVはジペプチジルペプチダーゼ4(DPP4)に結合する。どちらのコロナウイルスの場合も、Sタンパク質が感染をもたらし、したがって、ワクチン設計の主要標的となる。
本発明者らは更に、HCV E2コアナノ粒子ワクチンの開発において、同じワクチン設計戦略を適用した。C型肝炎ウイルス(HCV)はフラビウイルス科ヘパシウイルス属の小さな包膜一本鎖プラス鎖RNAウイルスである。世界人口の1〜2%が感染しているHCVは重大な健康問題であり、年間約50万人の死亡および毎年推定150〜200万人の新たに感染を引き起こしている。HCVは高い遺伝的多様性を有し、7個の主要遺伝子型および86個の亜型に分類されうる。HCVは迅速な突然変異を受けることがあり、これは感染個体内にウイルス準種を生じさせて、宿主免疫系から逃れさせる。しかし、急性感染患者の20〜30%における自発的ウイルスクリアランスは、HCV感染が、ワクチン接種により誘導される有効な免疫応答で予防可能であることを示唆している。E1およびE2エンベロープ糖タンパク質は、宿主肝細胞内へのウイルス侵入をもたらすHCVエンベロープ上のヘテロ二量体を形成する。E2は、受容体結合タンパク質として、宿主細胞受容体CD81およびSR−B1と直接相互作用し、主にCD81相互作用を遮断することによりHCVを中和する中和抗体の主要標的である。構造解析を容易にするために、高可変領域1(HVR1)ならびに可変領域2および3(VR2/3)(E2コアと称される)にトランケーションを含有するE2構築物を開発した。広域中和抗体AR3Cと複合体形成した分離株H77(遺伝子型1a)由来E2コアの結晶構造(Kongら,Science 342(6162):1090−1094,2013を参照されたい)、および中和抗体2A12に結合した分離株J6(遺伝子型2a)由来トランケート化E2の結晶構造(Khanら,Nature,509(7500):381−384,2014)は、HCVエンベロープ糖タンパク質の免疫認識に対する最初の洞察を提供した。しかし、現在、HCV感染を予防するために利用可能な認可されたワクチンは存在しない。
本発明者らはまた、本明細書に記載されているワクチン設計戦略をジカウイルス(ZIKV)DIIIナノ粒子ワクチンの開発のために適用した。ZIKVはフラビウイルス科のプラス鎖RNAウイルスであり、デング熱(DENV)、西ナイルウイルス(WNV)、日本脳炎ウイルス(JEV)および黄熱病ウイルス(YFV)をも含む。ZIKVゲノムは3つの構造タンパク質(キャプシド、prMおよびエンベロープ)および7つの非構造タンパク質(NS1、NS2a/b、NS3、NS4a/bおよびNS5)をコードする。2015年および2016年のZIKVの突発は世界的な公衆衛生上の危機を引き起こしている。ZIKVは、主にヤブカ(Aedes)属の蚊により伝染し、成人におけるギラン・バレー症候群(GBS)および新生児における小頭症を引き起こしうる。エンベロープ(E)に基づく幾つかのワクチン候補がマウスおよびNHPにおいてZIKV感染を予防することが報告されている。しかし、既存の抗フラビウイルス免疫による、ZIKVの感染および発病の抗体依存性感染増強(ADE)は、現在のワクチン戦略に関する安全性の懸念を招いている。ZIKV感染を予防するのに利用可能な、より有効で安全な他のワクチンは存在しない。
本発明者らは更に、熱帯熱マラリア原虫[プラスモジウム・ファルシパルム(Plasmodium falciparum)](マラリア)および結核菌[マイコバクテリウム・ツベルクローシス(Mycobacterium tuberculosis)](TB)のような細胞内病原体、ならびにプロタンパク質転換酵素サブチリシン/ケキシン9型(PCSK9)のようなヒトタンパク質へと、本発明者らのワクチン研究を拡張した。本発明者らの主な仮説は、ナノ粒子の提示が細胞内病原体に対する免疫応答を増強し、ヒトタンパク質に対する免疫寛容を破壊するというものである。抗体およびT細胞の応答は主な免疫学的測定値として測定される。
結核(TB)は、主に肺に感染するマイコバクテリウム・ツベルクローシス(Mycobacterium tuberculosis)細菌により引き起こされる潜在的に重篤な感染症である。TBは世界人口の25%に感染しており、2017年には最大1000万の症例を引き起こして、130万人の死亡をもたらした。カルメット・ゲラン桿菌(Bacille Calmette−Guerin)(BCG)ワクチンが、TBがよく見られる国々で広く使用されているが、それはTBに対する完全な防御をもたらさない。結核菌に対する幾つかの新規TBワクチン候補が開発されており、現在、ヒト臨床試験に付されている(Khoshnoodら,Int.J.Biol.Macromol.120:180−188,2018を参照されたい)。
2種類のリポタンパク質、すなわち、低密度リポタンパク質(またはLDL)および高密度リポタンパク質(またはHDL)が、細胞から、そして細胞へと、コレステロールを運搬する。LDLは動脈内の脂肪蓄積に寄与するため、LDLコレステロールは「悪玉」コレステロールと見なされる。血中のLDLコレステロールレベルの上昇は、西欧諸国における早死の一般的原因である心血管疾患のリスクの増加に関連づけられている。プロタンパク質転換酵素スブチリシン・ケキシン9型(PCSK9)は、LDL受容体(LDL−R)の分解を制御することにより、血清LDLコレステロール(LDL−C)を調節する。PCSK9は74kDaのセリンプロテアーゼであり、以下の3つのドメインを含有する:N末端プロドメイン、スブチリシン様触媒ドメインおよびC末端システイン/ヒスチジンリッチドメイン(CTD)(図16A)。PCSK9は自己触媒的切断を受けるが、14kDaのプロドメインは触媒ドメインに非共有結合したままであり、プロテアーゼを不活性にする。機能喪失型PCSK9突然変異はLDLコレステロール濃度の低下および心疾患のリスク低下に関連していることが判明している。これらの機能喪失型突然変異は有害な副作用を引き起こさないため、ワクチン誘導性抗体によるPCSK9阻害は、LDLコレステロール濃度を低下させるための魅力的な治療戦略となる。種々のリガンドと複合体形成した完全長PCSK9およびトランケート化PCSK9の結晶構造が解明されており、ワクチン設計のための合理的な基礎を提供している(図16A)。
Claims (40)
- 自己組織化ナノ粒子の表面上に提示されたポリペプチド免疫原を含むワクチン組成物であって、ロッキングドメインがナノ粒子の内部に包埋されており、自己組織化ナノ粒子のサブユニットに連結されており、ロッキングドメインが、界面における非共有相互作用により溶液中の近くのナノ粒子サブユニットに結合した別のロッキングドメインと二量体を自然に形成しうるタンパク質サブユニットである、ワクチン組成物。
- ロッキングドメインが、別の同一タンパク質ドメインとホモ二量体を形成するタンパク質ドメインである、請求項1記載のワクチン組成物。
- ロッキングドメインのサブユニットが、配列番号1〜9のいずれか1つに示されているアミノ酸配列、その保存的修飾変異体または実質的に同一の配列を含む、請求項2記載のワクチン組成物。
- ロッキングドメインがナノ粒子のサブユニットに共有結合している、請求項1記載のワクチン組成物。
