Abstract
Description
1−1.ジメンヒドリナート(Dimenhydrinate)効果の確認
1−1−1.ジメンヒドリナートの筋芽細胞(myoblast)増殖促進効果の確認
マウス筋芽細胞株C2C12を96−ウェルプレートに1.5X103cells/wellで分注した後、10%牛胎児血清(FBS)を含むDMEM培地で37℃及び5%CO2の条件で、一日、低密度培養した。その後、培地を400nMのH2O2とジメンヒドリナート(OC−501)0nM、250nM、500nM、1μM、2.5μM、5μM、10μM、又は20μMが含まれたDMEM培地でそれぞれ交換した。交換16時間後、ウェルにMTT試薬を処理し、3時間暗いインキュベーター内でインキュベーションした。上澄み液を除去し、100μlのDMSOをウェルに処理した後、OD(optical density)を595nmで測定し、細胞を顕微鏡で確認した。
マウス筋芽細胞株C2C12を12−ウェルプレートに0.7X105cells/wellで分注した後、細胞密度が70〜80%になるように培養した。その後、細胞をPBSで洗浄した後、培地を2%ウマ血清(horse serum)、400μMのH2O2、及びジメンヒドリナート(OC−501)0nM(DMSO)、500nM、5μM、10μM又は20μM含まれたDMEM培地(分化培地)にそれぞれ交換した。前記培地は、一日置きに交換し、且つ5〜7日間分化を誘導した。分化後、細胞を顕微鏡で確認し(4X及び10X倍率)、細胞を破砕して分化マーカーであるMyogenin及びMHC(myosin heavy chain)抗体を用いてウェスタンブロット法を行った。
1−2−1.ハルモルの筋芽細胞増殖促進効果の確認

2−1.細胞毒性の確認
C2C12細胞株を96−ウェルプレートに1.5X103cells/wellで分注した後、ジメンヒドリナート(OC−501)、ハルモル(OC−503)及びパントテン酸カルシウム(OC−504)を組み合わせて処理して(OC−501 5μM+OC−503 250nM、OC−5015μM+OC−503 500nM、OC−501 5μM+OC−504 5μM、OC−501 5μM+OC−504 10μM、OC−501 10μM+OC−503 250nM、OC−501 10μM+OC−503 500nM、OC−501 10μM+OC−504 5μM、OC−501 10μM+OC−504 10μM、OC−503 250nM+OC−504 5μM、OC−503 250nM+OC−504 10μM、OC−503 500nM+OC−504 5μM、OC−503 500nM+OC−504 10μM、OC−501 5μM+OC−503 250nM+OC−504 5μM、OC−501 5μM+OC−503 250nM+OC−504 10μM、OC−501 10μM+OC−503 250nM+OC−504 5μM、OC−501 10μM+OC−503 250nM+OC−504 10μM、OC−501 5μM+OC−503 500nM+OC−504 5μM、OC−501 5μM+OC−503 500nM+OC−504 10μM、OC−501 10μM+OC−503 500nM+OC−504 5μMとOC−501 10μM+OC−503 500nM+OC−504 10μM)、10%FBSが含まれたDMEM培地で培養した。処理24、48及び72時間後、細胞培養液にMTT溶液を処理し、3時間の間、暗いインキュベーターでインキュベーションした。上澄み液を除去した後、DMSOをウェル当り100μlずつ処理した。OD(optical density)を595nmで測定し、細胞毒性を確認した。前記の実験は、3反復行った。

3−1.ジメンヒドリナート及びパントテン酸カルシウムの組み合わせによる筋芽細胞分化促進効果の確認
マウスの前脛骨筋(tibialis anterior muscle)にコブラ毒素(cobratoxin)を刺した後、3日目に中間葉幹細胞(mesenchymal stem cell)を分離し、分離2日後からジメンヒドリナート(OC−501)10μM、及びパントテン酸カルシウム(OC−504)5μMをそれぞれ又は組み合わせて投与しながら分化マーカーであるPAX7(paired box protein−7)及びMYF5(Myogenic factor 5)抗体を用いてウェスタンブロッド法を行い、分化した後、細胞を顕微鏡で確認した(4X及び10X倍率)。
マウスの前脛骨筋(tibialis anterior muscle)にコブラ毒素(cobratoxin)を刺した後、3日目から3日間ジメンヒドリナート(OC−501)及びパントテン酸カルシウム(OC−504)を、それぞれ30mpk(mg/kg)ずつ、及び15又は30mpk(mg/kg)ずつ併用投与した後、その次の日、マウスを犠牲にして筋肉の再生の程度を確認した。
マウスの前脛骨筋にコブラ毒素ではない心臓毒素(cardiotoxin)を投与した後、ジメンヒドリナート(OC−501)及びパントテン酸カルシウム(OC−504)を、それぞれ30mpk(mg/kg)又はこれらを15又は30mpk(mg/kg)ずつ組み合わせて併用投与した後、筋繊維を確認した。
大腸癌細胞CT26を移植したマウスに抗癌剤である5−FU(5−fluorouracil)を投与して癌関連サルコペニアを誘発させた。また、大腸癌細胞移植後からジメンヒドリナート(OC−501)及びパントテン酸カルシウム(OC−504)をそれぞれ単独又は併用処理し、1週間ごとにMRIを測定してOC−501、504単独群と併用群での筋肉と脂肪の変化を測定した(図19)。
Claims (21)
- ジメンヒドリナート(Dimenhydrinate)、ハルモル(Harmol)又はパントテン酸カルシウム(calcium pantothenate)を有効成分として含有する筋肉疾患の予防又は治療用薬学組成物。
- ジメンヒドリナート及びハルモル、ジメンヒドリナート及びパントテン酸カルシウム、ハルモル及びパントテン酸カルシウム、又はジメンヒドリナート、ハルモル及びパントテン酸カルシウムを有効成分として含むことを特徴とする請求項1に記載の筋肉疾患の予防又は治療用薬学組成物。
- 筋肉疾患は、筋機能低下、筋肉消耗又は筋肉退化による筋肉疾患であることを特徴とする請求項1に記載の筋肉疾患の予防又は治療用薬学組成物。
- 筋肉疾患は、アトニー症(atony)、筋萎縮症(muscular atrophy)、筋ジストロフィー(muscular dystrophy)、筋無力症(myasthenia)、悪液質(cachexia)及びサルコペニア(sarcopenia)からなる群から選択されるいずれか一つ以上であることを特徴とする請求項1に記載の筋肉疾患の予防又は治療用薬学組成物。
- サルコペニアは、老化や癌によるものであることを特徴とする請求項7に記載の筋肉疾患の予防又は治療用薬学組成物。
- ビタミンC、ビタミンB1、ビタミンB2、ビタミンB6、ビタミンH、ビタミンPP、もしくはプロ−ビタミンB5、又はこれらの混合物から選択された水溶性ビタミンをさらに含むことを特徴とする請求項1に記載の筋肉疾患の予防又は治療用薬学組成物。
- ビタミンA、ビタミンD、ビタミンE、ビタミンK1、もしくはカロテン、又はこれらの混合物から選択された脂溶性ビタミンをさらに含むことを特徴とする請求項1に記載の筋肉疾患の予防又は治療用薬学組成物。
- ジメンヒドリナート、ハルモル又はパントテン酸カルシウムを有効成分として含むことを特徴とする抗癌剤投与による筋肉及び脂肪減少の予防又は治療用薬学組成物。
- ビタミンC、ビタミンB1、ビタミンB2、ビタミンB6、ビタミンH、ビタミンPP、プロ−ビタミンB5、ビタミンA、ビタミンD、ビタミンE、ビタミンK1、もしくはカロテン、又はこれらの混合物から選択されたビタミンをさらに含むことを特徴とする請求項11に記載の抗癌剤投与による筋肉及び脂肪減少の予防又は治療用薬学組成物。
- ジメンヒドリナート、ハルモル又はパントテン酸カルシウムを含むことを特徴とする筋肉疾患の予防又は改善用食品組成物。
- ジメンヒドリナート及びハルモル、ジメンヒドリナート及びパントテン酸カルシウム、ハルモル及びパントテン酸カルシウム、又はジメンヒドリナート、ハルモル及びパントテン酸カルシウムを含むことを特徴とする請求項13に記載の筋肉疾患の予防又は改善用食品組成物。
- 筋肉疾患は、筋機能低下、筋肉消耗又は筋肉退化による筋肉疾患であることを特徴とする請求項13に記載の筋肉疾患の予防又は改善用食品組成物。
- 筋肉疾患は、アトニー症、筋萎縮症、筋ジストロフィー、筋無力症、悪液質及びサルコペニアからなる群から選択されるいずれか一つ以上であることを特徴とする請求項13に記載の筋肉疾患の予防又は改善用食品組成物。
