Abstract
Description
本願は、2018年1月18日に出願したシンガポール国仮出願第10201800488Wの優先権を主張するものであり、その内容は本記載をもって完全に組み込まれるものとする。
a.(任意で)多能性幹細胞を提供する工程、
b.工程aの細胞を、本願で定義する細胞培養培地に24時間(0日目〜1日目)暴露し、T−ブラキウリ(T−Bra、原条/早期中胚葉マーカー)陽性細胞を得る工程と、
c.工程bの細胞を、本願で定義する細胞培養培地に24時間(1日目〜2日目)暴露する工程と、
d.工程cのマイクロキャリア付着細胞を本願で定義する細胞培養培地に48時間(2日目〜4日目)暴露する工程であって、工程b〜dで中胚葉誘導を実施し、
e.細胞培養培地を除去し、工程dで得られたKDR+PDGFRα−造血前駆細胞を単離する工程。
万能ドナー血液型であるO陰性Rh因子D陰性(O陰性)血液は、緊急時の輸血用において、限りある貴重な赤血球(RBCs)源と考えられている。人口の高齢化および発生し得るウイルスや病原のリスクによって起こり得る未来の供給不足に対する懸念は、入手容易な万能ドナー血液の代替物の開発に対するイニシアチブを加速させた。O陰性hiPSCsを万能ドナー赤血球作製のための出発材料として使用する可能性は、長らく考えられて来たアイデアであり、近年、実行に移された。hiPSCsの無制限の増殖能と、その造血細胞系へと分化する可能性との組み合わせは、これら細胞を、万能赤血球作製のための無限の細胞資源として魅力的なものにした。10個という少なさの、珍しい血液表現型のhiPSCクローンが、繰り返し輸血を必要とする人口の99%をカバーするのに十分であろうと仮定されている。高品質GMPグレードのhiPSCsの作製および貯蔵における近年の発展によって、臨床グレードの万能RBCsの大量誘導が次に起こる主な前進の一歩と考えられる。
振盪条件が分化のばらつきの主たる原因であるという仮説を試験するために、hES−3を初めに静置条件(静置ヒト多能性幹細胞(hPSC)増殖)または振盪条件(振盪ヒト多能性幹細胞増殖)のいずれかで7日間増殖させ、BMP4ベースの分化プロトコルを用いて静置条件下で分化させた。マイクロキャリア上で成長した細胞は、静置培養では大きなクラスターとなり、そして振盪培養では凝集体(直径約400μm)になった(図1A)という事実にもかかわらず、どちらの培養も多能性マーカーの同様の発現を示した(図1B)。しかし、分化の開始に続き、以下の発現には有意な差が見られた:原条/中胚葉マーカーであるT−bra[2日目のT−bra:静置対振盪hPSC増殖培養で32.6±1.2%および0.50±0.02%(p<0.0001)]および造血中胚葉マーカーであるKDR[4日目のKDR:静置対振盪hPSC増殖培養で27.9±3.3%および1.1±0.12%(p=0.001)](図1C)。造血分化を示す鍵となる造血マーカーの上方制御は、振盪hPSC増殖群においては有意に妨げられた。RT−PCR解析は、静置hPSC増殖群の4日間の分化後にCD31(早期血内皮マーカー)、GATA2、GATA1、SCL/Tal−1およびRUNX1の有意な発現増加を示したが、振盪hPSC増殖群(図1D)にはなかった。hPSC−MC凝集体の連続振盪の効果は、続く分化ステージにおいて最も明らかであり(図1DおよびE)、2週間の分化後の振盪hPSC増殖群には、造血前駆体の増殖は見られない(ウェル当たりの総造血前駆体:1.39×105±2.18×104)が、一方、静置hPSC増殖群では、有意な増殖が認められた(ウェル当たりの総造血前駆体数:5.77×106±6.30×105、p=0.0009)。同様に、振盪由来の培養においては、静置hPSC増殖群と比べて、続く赤芽球の作製が非常に減じていた(ウェル当たりの総赤芽球数:4.11±2.4×104対26.2±4.7×106、p=0.005、総CD235a集団:0.51±0.1%対51.8±2.0%、p<0.0001)(図1E)。これらの知見は、初めは、多能性増殖ステージにおけるhES−3−MC凝集体の連続振盪は、KDR+細胞の早期生成における分化プロセスのみならず後の赤血球ステージにも負の影響を与えるように見えた。
振盪マイクロキャリア培養で増殖させた多能性細胞の造血分化の効率に対するCHIRの効果を立証するために、O陰性hiPSC系(初めに振盪条件下のマイクロキャリア上で増殖)の分化を、BMP4/アクチビンAプロトコル(非CHIR仲介)またはCHIR仲介プロトコル(表1、図6の実験計画条件から確立)のいずれかを使用して評価した。BMP4/アクチビンAプロトコルは、非常に少量の原条/中胚葉誘導という結果になり、これは誘導後48時間のT−bra+細胞の低いパーセンテージ(6.66〜8.36%)および誘導後96時間の低い造血中胚葉誘導(KDR+細胞の5.3〜13.84%)(表1)に反映されている。これら条件は、造血転写因子SCLおよびRUNX1の非常に低い誘導という結果ももたらし、ブラスト成長培地(BGM)による2週間の培養後の造血前駆体増殖が無しまたは少量となり、続く赤芽球の増殖は失敗した(表1)。
12μMのCHIRへの一過性暴露による、単一O陰性(O陰性)ヒト人工多能性幹細胞−マイクロキャリア(hiPSC−MC)細胞系(初めに連続振盪下で増殖)の赤血球細胞系への分化の成功に続き、7種(7)の異なるO陰性hiPSC細胞系(BR2、BR7、D5、D9、D11、D12、X13)と、1種(1)の市販のhiPSC細胞系(IMR90)と、hESC系(hES−3)の分化を試験した。これらすべてのhPSC細胞系は、マイクロキャリア連続振盪下で7日間の増殖に成功し(図7)、5.3〜12.5倍の増殖倍率を達成し、平均凝集体径は255〜510μmの範囲であった(図4A)。多能性マーカーの発現は、Oct−4が72.9〜94.6%の範囲、Tra1−60が84.8〜93.5%の範囲、SSEA4が97.2〜99.7%の範囲であった(図4A)。G分染法核型解析を、ここで使用したヒト多能性細胞系の内の8種に対して実施し、全ての核型が正常であることが判明した(図8)。
O陰性赤芽球(D5)を初期ヒトMSCsと共培養することで最終成熟させ、分化赤芽球の機能的特徴づけを実施した。ヘモグロビンサブタイプの発現のRT−PCR評価は、O陰性赤芽球(D5赤血球)が、成人赤芽球と比べて、アルファ(33.13±5.16倍の増加、p=0.0034)、ガンマ(1.31±0.33倍の増加)、エプシロン(279±43.89倍の増加、p=0.0032)ヘモグロビンを発現するが、ベータ(0.009±0.001倍の増加、p<0.0001)ヘモグロビン発現が非常に少ないことを示した。同様に、hES−3由来赤芽球と成人赤芽球との比較は、アルファ、ガンマおよびエプシロンヘモグロビンは高いが、ベータヘモグロビンは低いことを示した(図5A)。免疫ブロット解析は、アルファ、ガンマおよびエプシロンヘモグロビンの発現と、ベータヘモグロビン発現が非常に少量または無しであることを、hES−3およびD5分化赤芽球(図5B)のみならず、他のhPSC系(図11)おいても確認した。興味深いことに、BR7から分化した赤芽球もベータヘモグロビンサブタイプの発現を示した(図11)。ドナー5のヒト人工多能性幹細胞(hiPSC)から分化した3種(3)のO陰性赤芽球細胞系の酸素平衡曲線の解析は、同様の酸素結合親和性を示し(D5赤芽球−1:p50− 14.7±0.8、D5赤芽球−2:p50− 13.2±0.04、D5赤芽球−3:p50− 13.8±0.6)、これらは成人赤血球(p50− 19.6±0.2)とは有意に異なっている(p<0.01)が、臍帯血赤血球と類似すると以前に示されたヒトES細胞系由来の赤芽球(hES−3:p50− 13.4±0.1)とは類似していた(図5C)。O陰性赤芽球は、成熟培地中で初期ヒトMSCsと19日間にわたり共培養することで最終成熟させた。フローサイトメトリー解析によると、赤芽球の39.0±1.0%はCD235a+且つDRAQ5(細胞浸透性核染色剤)陰性であり、除核赤血球細胞であることを示した(図5D)。この結果は、核染色のないCD235a+赤血球を示す、最終成熟赤芽球の免疫蛍光染色(図5E)およびより長期の成熟によるより多くの除核赤血球を示す細胞のギムザ染色(図5F)によってさらに確証された。
hPSCsを赤血球細胞へと分化させるために、数種の胚様体(EB)ベースの分化アプローチが試みられているが、今のところ、スケールアップの可能を示すものはない。ヒト多能性幹細胞−マイクロキャリア(hPSC−MC)凝集体培養物は、心臓前駆細胞分化について、以前にスピナー培養プラットホームおよびバイオリアクターによるスケールアップに成功している。従って、ヒト多能性幹細胞−マイクロキャリア(hPSC−MC)凝集体の大規模赤血球分化プロセスの開発にも使用できる可能性がある。
震盪懸濁培養下の限定細胞外マトリクス(ECM)被覆マイクロキャリアで増殖させた9種のヒト多能性幹細胞(hPSCs)の内の6種が、BMP4、アクチビンAおよびCHIRからなる、初期中胚葉誘導のための最適プロトコルによって、赤血球細胞を効率的に分化できることが過去に報告されている。しかし、懸濁撹拌条件下のhiPSCの増殖以外の残りのステージは、すべて静置条件下で実施された。さらに、造血細胞に運命づけられた中胚葉細胞からの造血前駆体の初期増殖は、全て半固体のメチルセルロースベースの培地で行われ、プロセスの容量のスケールアップが困難である。メチルセルロースベースの増殖プロトコルのスケーラビリティの無さによる限界を解消するために、本願に開示する分化プロセスを開発した。このプロセスでは、hiPSC増殖ステージから赤血球増殖ステージまでを震盪懸濁培養形式で実施することが可能であり、プロセス全体の容量のスケールアップが可能となり、これら培養を制御下のバイオリアクターへと適応させることができる。マイクロキャリア増殖hiPSCsのために最適化された中胚葉誘導条件と、造血細胞誘導条件のために改変されたプロセスとを組み合わせて、赤芽球の震盪懸濁培養分化および増殖を可能にした。

本項は、ヒト人工多能性幹細胞(hiPSC)から分化した赤芽球および臍帯血由来(CB)赤芽球から非アポトーシス性除核赤血球を高パーセンテージで誘導するための方法について記載する。文献に記載の従来のアプローチ、即ち、(EPO、インスリン、ホロトランスフェリンに加えて)10〜15%のヒト血清/血漿を含む培地での赤芽球の培養を使用した際の除核効率は、臍帯血由来赤芽球(<40%の除核赤血球)およびhiPSC赤芽球(<10%の除核赤血球)では通常は非常に低い。臍帯血由来/hiPSC赤芽球を、連続振盪下のマイクロキャリアで培養したOP9マウス間質細胞系(ATCC:CR−2749)と共培養する方法を試験した。このような懸濁撹拌共培養システムにおいては、非アポトーシス性(アネキシンV陰性)除核赤芽球(DRAQ5陰性)のパーセンテージが、hiPSC赤芽球では6.3±0.3%(従来法)から59.3±1.5%に増加し、臍帯血由来分化赤芽球では44.6±0.8%(従来法)から84.5±0.5%に増加した。従来のアプローチと同様に細胞を最終成熟させると、非常に高いパーセンテージのアポトーシス性細胞が検出された。OP9との共培養は、アポトーシス性細胞の割合を有意に減少させることが判明した。論理に縛られるものではないが、実験データは、OP9によるこの抗アポトーシス性効果は、細胞−細胞接触によるものというよりは、パラクリン効果による可能性を示した。さらに、OP9との共培養のプロセスに振盪を含めることは、除核効率を有意に改善することが示された。
O陰性Rh因子D陰性(O陰性)ヒト人工多能性幹細胞(hiPSCs)の分化は、輸血用途に有用な万能ドナー赤血球(RBCs)を作製し得る。赤血球の産生について報告されているアプローチの中では、異種物不含および条件限定で開発された胚様体(EB)仲介分化アプローチが、将来の臨床展開に最も有望である。しかし、強制的な凝集などによる胚様体作製のための従来のアプローチも、大規模の懸濁培養での成功は示されていない。3次元(3D)−凝集体としての、または限定細胞外マトリックス(ECM)被覆マイクロキャリア(MCs)上でのhiPSCの培養は、ヒト多能性幹細胞(hPSC)および胚様体の懸濁培養による増殖をスケールアップする手段となる可能性がある。hPSC−MC凝集体はBMP4ベースのプロトコルで分化させたときに、造血前駆体および赤芽球に分化し得ることがすでに示されている。しかし、連続振盪条件下で初めに増殖させた複数のhPSC細胞系を分化させる試みの繰り返しは、赤血球分化のばらつきを示した。論理に縛られるものではないが、振盪せん断応力が、SMAD7の発現を誘導し得ると仮定した。SMAD7は、中胚葉分化の初期ステージでBMP4によって活性化されるTGF−βシグナル伝達経路の成分であるSMAD1、5および8の全てのリン酸化に対して阻害効果を示すことが知られている。よって、振盪培養におけるBMP4シグナル伝達の阻害は、不十分な中胚葉誘導および分化結果のばらつきの理由である可能性がある。振盪下で初めに増殖させたヒト多能性幹細胞−マイクロキャリア(hPSC−MC)凝集体の赤血球分化のための最適プロトコルをここに示し、これは万能赤血球の大規模産生のための基礎またはプロセスとなる。
O陰性ヒト人工多能性幹細胞(hiPSC)系(D5、D9、D11、D12、X13、BR2、BR7)は、(シンガポール国立大学の倫理委員会の承認のもとで)同意を得たヒトドナーのフィンガープリック(finger−prick)血液に由来のCD71+赤芽球から、センダイウイルスによる4種の再プログラミング因子の形質転換によって再プログラミング化したものである。
簡単に説明すると、Accutase(米国、ThermoFisher Scientific)による酵素処理後の単層培養由来のヒト多能性幹細胞の100万個の単細胞を、5mlのmTeSR1培地、10μMのROCK阻害因子 Y27632(STEMCELLTechnologies)および細胞外マトリックス被覆ポリスチレンマイクロキャリアであるiPS−Spheres(シンガポール国、Brilliant Research)を含む、6穴の超低接着表面(ULA)プレート(米国、Corning)の各ウェルに植え付け、製造社の指示書に従って培養した。ヒト多能性幹細胞−マイクロキャリア(HPSC−MC)凝集体は、実験で使用する前に、110rpmの振盪プラットホームによる連続振盪下、または毎日の培地交換を行いながらの静置条件下で、7日間培養した。
BMP4をベースとするプロトコルを用いたhPSC−MC凝集体の中胚葉誘導は既に報告されている。100万個のHPSC−MC凝集体を、50ng/mlのBMP4および50ng/mlのVEGF165を含むStemlineII造血幹細胞増殖培地(米国、Sigma−Aldrich)中、静置条件下で培養した。48時間後に培地の半分を除去し、50ng/mlのBMP4、50ng/mlのVEGF165および20ng/mlのbFGF(全てのサイトカインはSTEMCELL Technologies製)を含むStemlineII培地で交換した。HPSC−MC凝集体は、造血前駆体分化に使用する前に、さらに48時間培養した。
初期のBMP4、アクチビンAおよびCHIR−99021の用量および暴露期間について、下記応答因子に対する効果を評価するために、分解能IVの一部実施法による実験計画(DoE)をMODDEソフトウエア(ドイツ国、Sartorius Stedim Biotech)を用いて実施した。応答因子:中胚葉誘導の4日後のKDR+細胞の誘導、または分化から2週間後に生成された総造血前駆細胞数。
MODDE(登録商標)ソフトウエアによって作製された、実験計画(DoE)の結果の解析および選択した条件の評価は、初めに連続振盪下で増殖させたヒト多能性幹細胞−マイクロキャリア(hPSC−MC)凝集体の造血中胚葉誘導のための最適なプロトコルの開発を可能にした。3mlのStemline(登録商標)II培地中の約200万から400万個のhPSC−MC凝集体を次のように分化させた。1日目:30ng/mlのBMP4、50ng/mlのVEGF165、12μMのCHIR−99021(米国、Selleck Chemicals)、50ng/mlアクチビンA、2日目:30ng/mlのBMP4、50ng/mlのVEGF165、50ng/mlのアクチビンA、3日目:30ng/mlのBMP4、50ng/mlのVEGF165および20ng/mlのbFGF。分化後4日目には、TrypLETM Express(ThermoFisher Scientific)処理によって誘導し、40μmのセルストレイナー(ドイツ国、Greiner Bio−one)によって濾した単細胞を、本願に記載のように、造血前駆体および赤芽球の増殖ならびに赤芽球の最終成熟に使用した。造血細胞および赤芽球の分化は、フローサイトメトリー、定量的リアルタイムPCR、免疫ブロッティングおよび顕微鏡画像によってモニタリングした。分化赤芽球の酸素結合親和性は、Hemoxアナライザー(米国、TCS Scientific Corp)で測定した。
100万個のHPSC−MC凝集体を、50ng/mlのBMP4および50ng/mlのVEGF165含有StemlineII造血幹細胞増殖培地(米国、Sigma−Aldrich)で培養した。48時間後に、培地の半分を除去し、50ng/mlのBMP4、50ng/mlのVEGF165および20ng/mlのbFGF(全てのサイトカインがSTEMCELL Technologies製)含有StemlineII培地に交換した。HPSC−MC凝集体は、造血前駆体増殖に使用する前にさらに48時間培養した。
HPSC−MC凝集体は、TrypLEによる処理によって単細胞に解離させた。37℃で5分のExpress(ThermoFisher Scientific)による処理の後、40μmのセルストレイナー(Greiner Bio-one)を通し、細胞のペレット化のために1300rpmで3分遠心分離し、リン酸緩衝化生理食塩水(PBS)で一度すすいでから、StemlineII培地に再懸濁した。100μlのStemlineII培地中の20万個の細胞を、6穴の超低接着表面プレートに移し、本願に記載のように、ブラスト成長培地(BGM)中、静置条件下で2週間培養した。
造血前駆体を、50ng/mlのSCF(STEMCELL Technologies)および3U/mlのEPO(米国、Peprotech)を含有する等量のStemlineII培地とさらに7日間混合した。次に細胞をPBSで10倍に希釈し、3000rpmで10分の遠心分離によってペレット化した。赤芽球細胞を、6穴の超低接着表面プレートに静置条件下で、3mlのStemlineII培地に2.5×105細胞/mlの濃度で植え付けた。培地は、1×血清代替物3(Sigma−Aldrich)、0.3%v/vのEx−CYTE成長増強培地添加剤(米国、Merck)、100ng/mのSCF、3U/mlのEPO、1μMのヒドロコルチゾン(Sigma−Aldrich)、200μg/mlのホロトランスフェリン(MP Biomedicals)および1×ペニシリン−ストレプトマイシン(ThermoFisher Scientific)を含有した。細胞は4日ごとに新鮮培地に再懸濁され、細胞濃度が2×106細胞/mlを超えると、2.5×105細胞/mlの濃度で再度植え付けられた。積算増殖倍率は、実験期間にわたる継代の間に達成された増殖倍率を乗算することで計算した。
初期BMP4、アクチビンAおよびCHIR−99021の用量および暴露期間の下記応答因子に対する効果を評価するために、分解能IVの一部実施法による実験計画(DoE)を、MODDEソフトウエア(ドイツ国、Sartorius Stedim Biotech)を用いて実施した。応答因子:中胚葉誘導の4日後のKDR+細胞の誘導、または分化から2週間後に作製された総造血前駆細胞数。MODDEソフトウエアは、試験因子の種々の組み合わせのいろいろな実験条件を作製し、単一因子または2因子の相互作用が応答因子に対して統計的に意味のある効果(陽性または陰性)を発揮するか否かを同定することを可能にする。R2スコア>0.5の計算モデルは、有意な適合を示すモデルであり、Q2スコア>0.1は有意なモデルであることを示すが、Q2>0.5は、未来予測精度の概算に良いモデルであることを示し、モデル有効性スコアが<0.25であることは、モデルの問題及び再現性(全体のばらつきと比べた複製間のばらつき)が統計的に有意であることを示し、0.5超であるべきである。
最終成熟および除核は、赤芽球を初期ヒト間葉幹細胞(MSCs)と3週間共培養することで誘導した。10%のFCS(Life Technologies)を添加したアルファ最小必須培地(α−MEM、ThermoFisher Scientific)で初めに培養した間葉幹細胞を、6穴プレートにウェル当たり1×105細胞で植え付けた。イスコーブ変法ダルベッコ培地(IMDM、ThermoFisher Scientific)と、8%のヒト血清(Sigma−Aldrich)、1×脂質ミックス(Peprotech)、6U/mlのEPO、50ng/mlのSCF、1000μg/mlのホロトランスフェリンおよび1×ペニシリン−ストレプトマイシンとを含む除核培地で、1×106細胞/mlに再懸濁した赤芽球を、あらかじめ植え付けておいた間葉幹細胞と共培養した。細胞は、2〜3週間の期間にわたり、毎週、新鮮培地への交換と同時に、植え付けたばかりの間葉幹細胞(MSCs、ウェル当たり1×105細胞)に移した。
細胞サンプルをトリゾール試薬(ThermoFisher Scientific)中で溶解し、RNA抽出まで−80℃で保存した。DNAse処理によるRNA抽出は、Direct−zol RNA抽出キット(Zymo Research)を製造社の説明書に従って使用して実施した。RNAサンプルは、NanoDrop UV−Vis分光光度計(ThermoFisher Scientific)を用いてOD260nmを測定することで定量した。
解析用のタンパク質サンプルは、凍結細胞ペレットの2×Laemmliバッファー(Bio−Rad)による溶解後に抽出し、BCAタンパク質アッセイキット(ThermoFisher Scientific)を用いて定量した。50μgの細胞溶解物をSDS−PAGEゲルにロードし、二フッ化ポリビニリデンメンブラン(Bio−Rad)に移した。5%のスキムミルクにより室温で2時間、メンブランをブロックし、一次抗体[1:800希釈のベータグロブリン(Santa Cruz、SC−21757)、1:400希釈のガンマグロブリン(Santa Cruz、SC−21756)、1:2000希釈のアルファグロブリン(Santa Cruz、SC−31110)、1:400希釈のエプシロングロブリン(Abcam、ab156041)および1:2000希釈のアクチン(Santa Cruz、SC−1615)]と室温で1.5時間インキュベートした。次にブロットを、希釈率1:10000の西洋ワサビペルオキシダーゼ(HRP)標識抗マウスIgG(Jackson ImmunoResearch)、HRP標識抗ウサギIgG(Jackson ImmunoResearch)、またはHRP標識抗ヤギIgG(Jackson ImmunoResearch)とインキュベートした。免疫複合体は、SuperSignal West Dura Extended Duration Substrate(Thermo Scientific)を用いて検出し、CLXposureフィルム(Thermo Scientific)上に補足した。バンド強度はImageJ ソフトウエア(https://imagej.nih.gov/ij/)を用いて測定し、ロード対照(アクチン)に対して正規化し、総ヘモグロビンバンドに対するパーセンテージとして報告した。
キャピラリーウエスタンブロットは、完全自動化システムであるPeggy Sue(米国、Proteinsimple,R&D)を用いて実施した。タンパク質の分離、検出および定量は、サイズ排除(12〜230kDa)に基づき、製造社のプロトコル(www.proteinsimple.com)に従って実施した。細胞溶解物は、1×フェニルメチルスルホニルフルオリド(PMSF)、1×ホスファターゼ阻害剤および1×プロテアーゼ阻害剤(BioSpes)を含む1×細胞溶解バッファー(Cell Signaling Technology)を用いて調製した。1mg/mlの総タンパク質サンプル(DTTおよびSDSで95℃で変性)をキャピラリー電気泳動で分離し、ブロッキングバッファーでブロックし、1次ウサギモノクローナル抗体[1:100希釈のブラキウリ(D2Z3J)、TCF1/TCF7(C63D9)、TCF3/TCFF7L1(D15G11)、Lef1(C12A5)、SMAD1(D59D7)、1:50希釈のpSMAD1/5 Ser 463/465(41D10)、1:5000希釈のGAPDH(D16H11)](全てCell Signaling Technology製)[1:50希釈のSMAD7(Sigma−Aldrich)]、次に西洋ワサビペルオキシダーゼ標識二次抗体でプローブ化し、ルミノール/ペルオキシダーゼ基質で検出した。化学発光シグナルの同定および定量は、Compass ソフトウエア(Proteinsimple)で行い、これは化学発光シグナルをウエスタンブロット像に変換する。
フローサイトメトリー用のサンプルは、4%パラホルムアルデヒド(eBioscience)で30分間固定し、1%ウシ血清アルブミン(BSA)を含むPBS中、4℃で保存した。多能性の測定には、サンプルを次の希釈率の1次抗体と室温(25℃)で20分インキュベートした:1:100のOct4(米国、R&D Systems)、1:50のTra1−60(Millipore)、1:100のSSEA4(米国、BioLegend)。FACSバッファー(PBS+1%BSA)による洗浄に続き、1:500希釈のウサギ抗マウスIgG−FITC複合体(DAKO)を用いて1次抗体を検出した。FACSバッファーによる洗浄および40μmの篩によるろ過に続き、サンプルをNovoCyteフローサイトメーター(米国、ACEA Biosciences Inc.)に流し、FlowJoソフトウエアを用いて解析した。
100μlのPBS中の細胞サンプルを、Cytospin 4 cytocentrifuge(Thermofisher Scientific)を用いて、500rpm、3分でスライド上に広げた。細胞を100%のメタノール(Sigma−Aldrich)で5分間固定し、蒸留水ですすぎ、室温で保存した。スライドをギムザ染色で20分染色し、PBS(pH7.2)ですすぎ、VECTASHIELD HardSet Antifade封入基剤(VECTOR Laboratories)およびカバーガラスで封入する前に風乾させた。スライドはAxiovert 200M倒立顕微鏡(Zeiss)を用いて画像化した。
hPSC分化赤芽球の酸素結合および解離平衡曲線を作製するために、HemoxアナライザーモデルB装置(TCS Scientific Corp)を用いた。HEMOX溶液(TCS Scientific Corp)中の約1×107個の赤芽球細胞を酸素飽和条件下、圧縮空気を用いて流し(run)、次に窒素ガスを用いて脱酸素条件に付した。Hemox解析ソフトウエアを用いて、p50値(50%のオキシヘモグロビン飽和レベルをもたらす酸素圧)を計算した。成人末梢血(ドナー由来)を対照として流した。全てのサンプルを2回測定した。
本実験に使用したヒト多能性幹細胞系をG分染法核型解析(シンガポール国、KKH女性および小児病院、病理学および臨床検査医学部)に回し、そこで典型的には20個の分裂中期について、全体的な染色体異常および異数性を評価した。
統計解析はGraphPad Prism 6(GraphPad Software Inc.)で実施した。スチューデントの独立2群検定を等分散の2群を比較するために使用し、分散が等しくないと考えられるときには、Mann−Whitney検定を使用した。P値が0.05未満(p<0.05)のときを有意とした。
培養OP9(ATCC:CR−2749)を、20%のFCS(Gibco)および1×ペニシリン/ストレプトマイシンを添加したアルファ−MEM培地(Thermofisher)を用い、市販の組織培養フラスコ中で単層培養した。単一細胞をTrypLETM、37℃で5分の酵素処理により誘導した。20万個の細胞を、5mlのアルファMEM−20%FCS培地および20mgのSolohill(登録商標)Plastic Plusマイクロキャリア(Pall Corporation)を含む、6穴の超低接着表面プレート(Corning)の各ウェルに植え付けた。細胞をマイクロキャリアに付着させるために、ロッキングプラットホームを用いて、プレートを37℃、75rpmで振盪させた。OP9−MCs培養物は、赤芽球との共培養に使用する前に、2日間連続振盪下で培養した。
ヒト人工多能性幹細胞(hiPSC)赤芽球の誘導および培養は、PCT出願明細書の段落[0203]に記載されている。
生/死亡細胞および有核細胞の混合物を不織布(NWF)フィルタ(シンガポール国、Antoshin)に通すことで、除核赤血球(RBCs)を分離した。簡単に説明すると、不織布(NWF)フィルタを10mlのリン酸緩衝化生理食塩水(PBS)ですすいだ。10mlのリン酸緩衝化生理食塩水中の濃度107細胞/mlの細胞を、30mlのシリンジを低速で用いてゆっくりと不織布フィルタに通し、ろ過液を回収した。続いて不織布フィルタを10mlのリン酸緩衝化生理食塩水ですすぎ、ろ過液と同じチューブに回収した。ろ過された細胞を、1500rpm、室温で5分の遠心で落とした。回収した赤血球を、アデニン(Sigma−Aldrich)を含むクエン酸リン酸デキストロース溶液に再懸濁し、さらなる分析のために4℃で保存した。
フローサイトメトリー
細胞懸濁液を40μMでろ過することで得た未固定の単一細胞をリン酸緩衝化生理食塩水に再懸濁し、フローサイトメトリーに使用した。簡単に説明すると、(96穴のv底プレートの)ウェル当たり約100,000細胞を1500rpm、3分で落とした。細胞ペレットを、1:100希釈のアネキシンV−FITC標識抗体(E−bioscience)(アポトーシス評価用)、または1:100希釈のCD235a−FITC標識抗体(E−Bioscience)を含む100μlの1×アネキシンV結合バッファー(Thermofisher Scientific)に再懸濁し、暗環境下、25℃で20分間静置した。細胞を1500rpm、3分で落とし、その後、200μlの1×アネキシンV結合バッファーで洗浄した。細胞を最終的に、1:5000に希釈したDRAQ5(E−bioscience)を含有する200μlの1×結合バッファーに再懸濁した。細胞はNovocyteフローサイトメーターで評価し、488nmおよび647nmで検出した。
サイトスピン遠心分離機を用いて、350g、5分で約50,000〜100,000細胞を顕微鏡スライド上に落とした。細胞を100%のメタノールで5分間固定し、風乾した。固定した細胞を、pH7.2のリン酸緩衝化生理食塩水バッファー(Sigma)で1:10に希釈したギムザ染色液(Sigma)で、25℃、15分間染色した。染色された細胞をpH7.2のリン酸緩衝化生理食塩水バッファーですすぎ、その後、明視野顕微鏡(Zeiss Axiovert)で可視化/イメージ化した。
ヒト人工多能性幹細胞−マイクロキャリア(hiPSC−MC)凝集体の造血中胚葉誘導
ヒト人工多能性幹細胞−マイクロキャリア(hiPSC−MC)凝集体(1×106細胞/mlまたは1e6細胞/ml)を、StemlineII造血幹細胞増殖培地(SL2)を加えた中胚葉誘導培地(サイトカインはすべてStemcell Technologies製)に移した。移した先は、75rpmで連続振盪下の6穴超低接着表面プレート(5ml)または36rpmで連続振盪下の125mlのスピナーフラスコ(50ml)のいずれかであった。毎日の培地交換は次の内容とした。0日目:SL2+30ng/mlのBMP4+50ng/mlのVEGF−165+40ng/mlのアクチビンA+12〜15μMのCHIR−99021、1日目:SL2+30ng/mlのBMP4+50ng/mlのVEGF−165+40ng/mlのアクチビンA、2日目:SL2+20ng/mlのBMP4+30ng/mlのVEGF165+5ng/mlのアクチビンA+10ng/mlのbFGF+20ng/mlのSCF+0.4ng/mlのβ−エストラジオール。サンプルを1日目および3日目に回収し、それぞれT−ブラキウリ(T−Bra)およびKDR/PDGFRα細胞のそれぞれのフローサイトメトリー解析に付した。
分化から3日目に、hiPSC−MC凝集体に対してTrypLETM Express(ThermoFisher Scientific)による処理を37℃で5分行って単一細胞を得、40μMのセルストレイナー(ドイツ国、Greiner Bio−one)で濾した。6穴の超低接着表面プレート(5ml)、50mlの振盪フラスコ(10ml)または125mlのスピナーフラスコ(50ml)のいずれかの造血細胞誘導培地に、1.25×105から2.5×105細胞/mlの濃度で細胞を植え付けた。(断りがない限り)完全な培地交換を次の内容で行った。3日目:SL2+20ng/mlのBMP4+30ng/mlのVEGF−165+10ng/mlのbFGF+30ng/mlのSCF+10ng/mlのIGF2+10ng/mlのTPO+5U/mlのヘパリン+50μMのIBMX+0.4ng/mlのβ−エストラジオール、5日目:SL2+120ng/mlのBMP4+180ng/mlのVEGF−165+60ng/mlのbFGF+180ng/mlのSCF+60ng/mlのIGF2+60ng/mlのTPO+30Uのヘパリン+300μMのIBMX+2.4ng/mlのβ−エストラジオールを1:6までつぎ足し、7日目:SL2+20ng/mlのBMP4+30ng/mlのVEGF+10ng/mlのbFGF+30ng/mlのSCF+10ng/mlのIGF2+10ng/mlのTPO+5U/mlのヘパリン+50μMのIBMX+0.4ng/mlのβ−エストラジオール+1μMのStem Regenin1(SR1)(StemcellTech)、9日目:SL2+20ng/mlのBMP4+30ng/mlのVEGF165+10ng/mlのbFGF+30ng/mlのSCF+10ng/mlのIGF2+10ng/mlのTPO+5U/mlのヘパリン+50μMのIBMX+0.4ng/mlのβ−エストラジオールを1:2までつぎ足し。
6穴の超低接着表面プレート(5ml)、50mlの振盪フラスコ(10ml)または125mlのスピナーフラスコ(50〜100ml)のいずれかに、1.25×105〜2.5×105細胞/mlの細胞を赤血球誘導培地に植え付けた。(断りがない限り)完全な培地交換を次の内容で行った。11日目:SL2+6.7ng/mlのBMP4+30ng/mlのSCF+50μMのIBMX+1μMのヒドロコルチゾン(Sigma−Aldrich)+16.7ng/mlのFlt3L+6.7ng/mlのIL3+4U/mlのEPO(Peprotech)、13日目:SL2+40.2ng/mlのBMP4+180ng/mlのSCF+300μMのIBMX+6μMのヒドロコルチゾン(Sigma−Aldrich)+100.2ng/mlのFlt3L+40.2ng/mlのIL3+24U/mlのEPO+3μMのプルリポチンを1:6までつぎ足し、15日目から先:SL2+1×血清代替物3(Sigma−Aldrich)+0.3%のv/vのExCyte試薬(Millipore)+1μMのヒドロコルチゾン+100ng/mlのSCF+4U/mlのEPO+10ng/mlのIL3+0.2mg/mlのホロトランスフェリン(MP Biomedicals)+1×ペニシリンおよびストレプトマイシン。15日目から先は、完全な培地交換を3日に一度行った。高い細胞密度の培養には、細胞密度が5×106細胞/ml(5e6細胞/ml)を超えたら、完全な培地交換を毎日実施した。高い積算増殖倍率のためには、細胞密度が5×106細胞/ml(5e6細胞/ml)を超えたら、常に細胞を1×106細胞/ml(1e6細胞/ml)に戻すように植え付けた。積算増殖倍率は、実施した実験の期間にわたり、継代間で達成された増殖倍率を乗算することで計算した。

- 多能性幹細胞を造血前駆細胞に分化させるための方法であって、前記方法が懸濁撹拌下で実施され、中胚葉誘導のステージにおいて、GSK−3阻害因子またはWnt経路活性化因子を添加する、方法。
- 前記多能性幹細胞を造血前駆細胞に分化させるための方法は、以下のステージを含む、請求項1に記載の方法。
a.中胚葉誘導ステージ、
b.造血細胞誘導ステージ、
c.赤芽球誘導ステージ、および
d.赤芽球成熟ステージ。 - 多能性幹細胞増殖ステージにおいて、前記多能性幹細胞を1.5×105〜4×106細胞/mlの濃度に増殖させる、請求項2に記載の方法。

- 前記塩基性線維芽細胞増殖因子(bFGF)またはその変異体が、bFGFの熱安定性キメラ変異体または安定キメラ線維芽細胞増殖因子(FGF)である、請求項29に記載の細胞培養培地。
- 前記サイトカインが幹細胞因子(SCF)である、請求項29〜32のいずれか一項に記載の細胞培養培地。
- 前記血管内皮増殖因子がVEGF165である、請求項29〜33のいずれか一項に記載の細胞培養培地。
- 細胞培養培地がBMP4、アクチビンA、bFGF、βエストラジオール、SCFおよびVEGF165を含む、請求項29〜34のいずれか一項に記載の細胞培養培地。
- 前記BMP4が18ng/ml〜27ng/mlの間の濃度で存在し、前記アクチビンAが3ng/ml〜7ng/mlの間の濃度で存在し、前記bFGFが5ng/ml〜14g/mlの間の濃度で存在し、前記βエストラジオールが0.2ng/ml〜0.8ng/mlの間の濃度で存在し、前記SCFが26ng/ml〜36g/mlの間の濃度で存在し、前記VEGF165が32〜38ng/mlの間の濃度で存在する、請求項29〜35のいずれか一項に記載の細胞培養培地。
- 多能性幹細胞を造血前駆細胞に分化させるための方法であって、懸濁撹拌下で実施され、
a.請求項15〜31のいずれかで定義した細胞培養培地に多能性幹細胞を24時間(0日目〜1日目に)暴露し、T−ブラキウリ(T−Bra、原条/早期中胚葉マーカー)陽性細胞を得る工程と、
b.工程aの細胞を、請求項48〜57のいずれかに記載の細胞培養培地に24時間(1日目〜2日目に)暴露する工程と、
c.工程cのマイクロキャリア付着細胞を請求項58〜76のいずれかに記載の細胞培養培地に48時間(2日目〜4日目に)暴露し、工程aおよび工程bで中胚葉誘導を誘導する工程と、
d.細胞培養培地を除去し、工程cで得られたKDR+PDGFRα−造血前駆細胞を単離する工程と
を含む、方法。 - 多能性幹細胞を造血前駆細胞に分化させるための方法であって、懸濁撹拌下で実施され、
(a)懸濁撹拌に付した培養細胞から単離した多能性幹細胞に対して中胚葉誘導を実施し、KDR+PDGFRα−造血前駆細胞を得る工程と、
(b)工程(a)で単離した細胞に対して造血細胞誘導を実施し、CD34/CD43/CD45造血前駆細胞を得る工程と、
(c)工程(b)で単離した細胞に対して赤芽球増殖を誘導し、CD235a+CD71+赤芽球細胞を得る工程と、
(d)工程(c)で単離した細胞に対して赤芽球成熟を誘導し、CD235a+DRAQ5陰性除核赤芽球細胞を得る工程と、
(e)細胞培養培地を除去し、工程(d)で得られたCD235a+DRAQ5陰性除核赤芽球細胞を単離する工程と
を含む、方法。 - 前記多能性幹細胞がマイクロキャリアに付着している、請求項37または38に記載の方法。
- 前記多能性幹細胞が人工多能性幹細胞である、請求項1〜15のいずれか一項に記載の方法、請求項16〜36のいずれか一項に記載の細胞培養培地、または請求項37〜39のいずれか一項に記載の方法。
- マイクロキャリアと、請求項16〜36のいずれか一項に記載の細胞培養培地とを含むキット。
