前記シュードタイプ化レトロウイルスベクター粒子またはそのウイルス様粒子が、造血細胞膜に結合し、それと融合することが可能な修飾ヒヒ内在性レトロウイルス(BaEV)エンベロープ糖タンパク質を含み、それにより前記活性化NK細胞に生体物質を導入する、方法を提供する。
ヒヒ内在性レトロウイルス(BaEV)エンベロープ糖タンパク質の膜貫通および細胞外ドメインと、マウス白血病ウイルス(murine leukemia virus)(MLV)エンベロープ糖タンパク質の細胞質尾部ドメインとの融合を含むか、もしくはそれにより構成されるキメラエンベロープ糖タンパク質、または
細胞質尾部ドメインが融合阻害Rペプチドを欠く、修飾BaEVエンベロープ糖タンパク質
であってもよい、前記シュードタイプ化レトロウイルスベクター粒子またはそのウイルス様粒子。
ヒヒ内在性レトロウイルス(BaEV)エンベロープ糖タンパク質の膜貫通および細胞外ドメインと、マウス白血病ウイルス(MLV)エンベロープ糖タンパク質の細胞質尾部ドメインとの融合を含むか、もしくはそれにより構成されるキメラエンベロープ糖タンパク質、または
細胞質尾部ドメインが融合阻害Rペプチドを欠く、修飾BaEVエンベロープ糖タンパク質
を含んでもよい。
a)NK細胞を活性化する少なくとも1つのサイトカインを含む第1の組成物と、
b)シュードタイプ化レトロウイルスベクター粒子またはそのウイルス様粒子を含む組成物
とを含み、前記シュードタイプ化レトロウイルスベクター粒子またはそのウイルス様粒子が、造血細胞膜に結合し、それと融合することが可能な、本明細書に開示の修飾ヒヒ内在性レトロウイルス(BaEV)エンベロープ糖タンパク質を含む、組成物の組合せを提供する。
他に定義しない限り、本明細書において使用する技術的および科学的用語は、本発明が属する技術分野の当業者により一般的に理解されるものと同一の意味を有する。
本発明の一実施形態では、トランスジェニック核酸、例えば、キメラ抗原受容体をコードする核酸は、造血細胞膜に結合し、それと融合することが可能な修飾ヒヒ内在性レトロウイルス(BaEV)エンベロープ糖タンパク質によりシュードタイプ化した、レトロウイルスベクター粒子、例えば、レンチウイルスベクター粒子により活性化NK細胞に導入する。
バフィーコートからのNK細胞の単離では、末梢血単核細胞(PBMC)の調製を、Ficoll−Paque PLUS(GE Healthcare)を使用して、標準的な密度勾配遠心分離により実施した。NK cell isolation kit human(Miltenyi Biotec)を適用して、PBMCからNK細胞を、そのまま濃縮(untouched enrichment)した。単離したNK細胞を3つの群に分割し、それらを異なる期間活性化し、GFPを含有するBaEVシュードタイプ化レンチウイルスベクターにより形質導入した。すべての形質導入は、10ng/mlのvectofusin(Miltenyi Biotec社)およびスピノキュレーション(spinoculation)を用いて、製造者のプロトコールに従って実施した。第1の群のNK細胞は、単離直後に(活性化せずに)形質導入した。第2の群のNK細胞は、5%のAB型血清、500U/mlのIL−2および10ng/mlのIL−15を含有するNK MACS medium(Miltenyi Biotec)中で、1日培養し(IL−2+IL−15中で1日活性化)、次いで、形質導入した。第3の群のNK細胞は、5%のAB型血清、500U/mlのIL−2および10ng/mlのIL−15を含有するNK MACS medium(Miltenyi Biotec)中で、2日間培養し(IL−2+IL−15中で2日間活性化)、次いで、形質導入した。各形質導入群の対照として、ウイルスベクターを添加しないことを除いて(非形質導入)形質導入細胞と同一条件下で、NK細胞を培養した。形質導入から3日後、非活性化NK細胞は、わずか2.44%の形質導入を示した(非活性化、図1の左下パネル)。一方、5%のAB型血清、500U/mlのIL−2および10ng/mlのIL−15を含有するNK MACS medium(Miltenyi Biotec)中で、1日および2日間培養したNK細胞群は、14.59%および22.61%のGFP発現をそれぞれ示した(図1、形質導入、中央および右下のパネル)。各群では、非形質導入NK細胞は、GFP発現をまったく示さなかった(図1、上のパネル、非形質導入)。
初代NK細胞を実施例1に記載のように単離することで、純粋なCD3−およびCD56+NK細胞を得た(図2A)。T細胞を同一の血液試料から、MiltenyiのCD4に対するMicroBeadsおよびCD8に対するMicroBeadsを使用して、ユーザーマニュアルに従って単離し、純粋なCD3陽性T細胞をもたらした(図2A)。単離後、NK細胞を2日間、5%のAB型血清、10ng/mLのIL−15および500U/mLのIL−2を含むNK MACS medium中で活性化した。単離後、T細胞を2日間、200U/mLのIL−2およびMiltenyiのT Cell Transact humanを含むTexMACS medium中で、ユーザーマニュアルに従って活性化した。次いで、活性化した初代T細胞およびNK細胞、ならびにNK−92細胞株に、VSV−GエンベロープまたはBaEVのいずれかでシュードタイプ化した、GFP発現をコードするレンチウイルスベクターを含有する上清により形質導入した。VSV−Gシュードタイプ化ウイルス粒子による形質導入から3日後、NK細胞は、高ウイルス力価でも、約2%のGFP発現しか示さず、一方、T細胞は、同一条件下で、約77%の高いGFP発現を示した(図2B)。この知見により、VSV−Gシュードタイプ化レンチウイルスベクターによるNK細胞への形質導入は、非効率的であるが、T細胞を含む他の細胞型には作用することが確認される。驚くべきことに、BaEVシュードタイプ化ウイルスベクターによるNK細胞株NK−92への形質導入から3日後、ほぼ100%の高いGFP発現が観察されたが、VSV−GによるGFP発現は、初代NK細胞と同様に、非効率的であった(図2C)。NK−92細胞は、活性化条件下で、200U/mLのIL−2を含有するNK MACS medium中に維持した。BaEVシュードタイプ化ベクターによる形質導入を適用した場合、NK−92についての知見と一致して、60%の驚くほど高い割合の活性化初代NK細胞がGFPを発現し、T細胞と同等の形質導入率に達した(図2D)。重要なことには、細胞数の経時的上昇を意味する増殖率は、非形質導入NK細胞、およびBaEVシュードタイプ化により形質導入したNK細胞と同等であり、形質導入方法により、NK細胞の細胞死も誘導されず、増殖も影響されないことが示された(図5)。これは、治療により高度の細胞死が引き起こされる、遺伝子修飾のための他の方法、例えば、エレクトロポレーションとは異なる。加えて、遺伝子修飾のための他の方法、例えば、エレクトロポレーションでは、導入遺伝子の一過性発現のみが可能となる。注目すべきことに、BaEVシュードタイプ化レンチウイルスベクターによる形質導入後、導入遺伝子を発現したNK細胞が、常に高い割合で、数週間にわたって安定であった(図6)。
初代NK細胞を実施例1に記載のように単離した。精製NK細胞を2日間、IL−2(500U/ml)およびIL−15(10ng/ml)、またはIL−2(500U/ml)、IL−15(10ng/ml)およびIL−33(30U/ml)のいずれかを含有する、5%のAB型血清を含むNK MACS medium中で活性化した。2日目に、目的の遺伝子を含有する、ヒヒ(BaEV)(図3A)またはVSV−G(図3B)エンベロープによりシュードタイプ化した、レンチウイルスベクターによって細胞を形質導入した。形質導入は、実施例1に記載のように行った。非形質導入細胞は、形質導入細胞と同一条件下で培養した。形質導入後、IL−33を含まない培地中で細胞を培養した。形質導入NK細胞における導入遺伝子発現を、目的の遺伝子の産物に対する特異的モノクローナル抗体により、形質導入後3日目、8日目、および14日目に検出した。形質導入後3日目の導入遺伝子発現はそれぞれ、IL−33を用いない場合は、16.62であり、IL−33を用いた場合は、21.27%であった(図3A、左下のパネル)。形質導入後8日目の導入遺伝子発現はそれぞれ、IL−33を用いない場合は、32.59%であり、IL−33を用いた場合は、45.03%であった(図3A、中央のパネル)。形質導入後14日目では、導入遺伝子発現は、IL−33を用いない場合、33.66%であり、IL−33を用いた場合、60.17%であった(図3A、右下のパネル)。この結果は、NK細胞をIL−33により予め刺激すると、形質導入に相加効果があることを示唆する。非形質導入NK細胞は、非常に低いレベルの抗体染色を示した(1%未満、図3A、上のパネル)。VSV−Gエンベロープによりシュードタイプ化したレンチウイルスベクターは、ヒヒシュードタイプ化ベクターと比較して著しく低い形質導入効率を示したが、IL−33の相加効果は、依然として著しい(図3B)。
3人の異なるドナーから、初代NK細胞を実施例1に記載のように単離した。単離後、同一のドナー由来のNK細胞を、2日間、5%のAB型血清、10ng/mlのIL−15および500U/mLのIL−2を含むNK MACS medium中で、20ng/mLのIL−18を添加するか、またはIL−18を添加せずに活性化した。次いで、活性化初代NK細胞を、BaEVによりシュードタイプ化した、GFPをコードするレンチウイルスベクターにより形質導入した。形質導入後、IL−18を含まないが、IL−2およびIL−15を含む培養培地中で、細胞を培養した。形質導入から3日後、NK細胞は、IL−18を用いずに予め活性化した場合、40%の平均GFP発現を示したが、同一のドナー由来のNK細胞は、IL−18を用いて予め活性化した場合、66%の著しく高いGFP発現を示した(図4)。
CARを発現するT細胞は、CD19陽性B細胞白血病の治療のための臨床試験において非常に肯定的な結果をもたらした。CAR NK細胞が、CAR T細胞の代替物であり得るため、本発明者らは、NK細胞形質導入についての一例示的適用のように、CD19に対するCARをNK細胞に形質導入した。初代NK細胞は、4人の異なるドナーから、実施例1に記載のように単離した。単離後、同一のドナー由来のNK細胞を2日間、5%のAB型血清、10ng/mlのIL−15および500U/mLのIL−2を含むNK MACS medium中で活性化した。次いで、活性化初代NK細胞を、CD19−CARをコードするレンチウイルスベクターにより形質導入した。使用した「CD19B」CAR構築物およびVSV−Gによる形質導入後のT細胞におけるその発現は、これまでに記載されている(Schneiderら2017)。NK細胞への形質導入では、BaEVによりシュードタイプ化したレンチウイルスベクターは、この構築物のために生成し、使用した。形質導入から7日後、CAR発現およびNK細胞の細胞毒性を解析した。詳細には、NK細胞を48時間、無血清培養培地中でインキュベーション後、25μg/mLの組換えヒトCD19 Fc(R&D Systems)との30分間の結合により、CD19−CAR発現を判定し、Fcに対するマウスAPCコンジュゲートmAb(Miltenyi Biotec)を使用して、フローサイトメトリーにより検出した。CD19−CARの発現は、平均して、種々のドナー由来のNK細胞間で70%であり、一部のドナーは、形質導入率が90%に達した(図7A)。種々の標的細胞株の標的細胞の殺滅を、フローサイトメトリーをベースとするアッセイにより解析した。標的細胞をCellTrace Violet(Life Technologies)により標識し、1ウェルあたり2×103細胞を、96ウェルの丸底プレート中に播種した。次いで、対照として培地、またはNK細胞を種々のNK対標的比で添加した。24時間後、CellTrace Violet陽性標的細胞を、フローサイトメトリーにより定量化した。NK細胞を含む試料中の生標的細胞とNK細胞を含まない対応する試料との差を、殺滅された標的として定義した。K562細胞は、CD19を発現しないが(図7B)、それらが、NK細胞の天然の細胞毒性に対する感受性を有するため、NK細胞の一般的細胞毒性能を試験するための標準的な標的であると考えることができる。予想どおり、CAR NK細胞および非形質導入NK細胞はともに、まったく同様に用量依存的に、K562細胞を非常に効率的に殺滅し、CD19非依存性の天然細胞毒性が、形質導入により影響されないことを立証した(図7C)。K562細胞とは対照的に、RS4;11細胞は、CD19を発現するが(図7B)、それらは、NK細胞の天然細胞毒性に対して完全に非感受性である。結果的に、非形質導入NK細胞は、RS4;11細胞を殺滅することが不可能であり、一方、CD19−CAR NK細胞は、K562細胞と同等に効率的にRS4;11を殺滅し、使用したCD19 CARの高い機能性および特異性を実証した(図7B)。Raji細胞は、CD19陽性であるが(図7B)、RS4;11とは異なり、それらは、活性化NK細胞の天然細胞毒性に対して感受性であるが、K562細胞ほどではない。したがって、非形質導入NK細胞は、既に、用量依存的にRaji細胞を殺滅可能であったが、それにもかかわらず、CD19 CAR NK細胞によるRaji細胞の殺滅は、著しくより高く、CD−19特異的殺滅の相加効果を示唆していた。結論として、BaEVシュードタイプ化レンチウイルスベクターによるNK細胞への形質導入は、白血病標的細胞のCD19特異的殺滅が向上したCD19 CAR発現NK細胞を効率的に生成することを可能とした。
NK細胞は、IL−2およびIL−15を含むNK MACS medium(Miltenyi Biotec GmbH)中で2日間、増殖することにより活性化した。ナイーブおよび活性化細胞をともに、RIPA溶解バッファー中に溶解し、ASCT2に対する抗体を使用してウエスタンブロット解析に供した。この結果は、ヒヒエンベロープ、ASCT2の受容体が、NK細胞活性化後のみに発現するが(図8)、ナイーブNK細胞では発現しないことを示し、ヒヒシュードタイプ化レンチウイルスベクターにより形質導入するために、NK細胞を活性化する必要があることを示唆した。
NK細胞を増殖色素で標識し、広範に洗浄し、次いで、GFPを含有するヒヒシュードタイプ化レンチウイルスベクターにより形質導入した。増殖開始および形質導入から4日後、細胞増殖および形質導入をフローサイトメトリーにより判定した。結果は、静止NK細胞ではなく、増殖性NK細胞のみが、ヒヒシュードタイプ化レンチウイルスベクターにより形質導入可能であることを示唆する(図9A)。GFP形質導入NK細胞を抗CD56抗体で染色し、3つのNK細胞群(増殖GFP+、増殖GFP−、静止状態)にゲーティングしてCD56発現のレベルを判定した(図9B)。増殖し形質導入したNK細胞のすべてが、CD56brightであり、一方、形質導入せず静止状態のNK細胞が、CD56dimであり、CD56dimNKサブセットは、増殖性でなく、形質導入不可能であることを示唆した。次に、本発明者らは、他のNK細胞受容体を、増殖し形質導入した、増殖性であるが形質導入していない、および静止状態のNK細胞のサブセット間でさらに比較した(図10)。NK細胞を増殖色素で標識し、広範に洗浄し、次いで、GFPを含有するヒヒシュードタイプ化レンチウイルスベクターにより形質導入した。形質導入から4日後、種々のNK細胞受容体に対するモノクローナル抗体で細胞を染色し、フローサイトメトリーを使用して発現を判定した。一般的に公知のNK細胞マーカーの種々のサブセット間での発現を示す(図10)。NKp46、DNAM−1、CD94およびNKG2Cの発現は、すべての異なるNK細胞サブセット間で同等であった。増殖性細胞の画分においては、GFP+とGFP−との間の異なるNK細胞マーカーの発現の差は、顕著ではなかった。しかし、いくつかのマーカーの発現によれば、静止NK細胞は、増殖性NK細胞よりも表現型が顕著に異なった。増殖性細胞は、よりCD56強陽性(bright)であり、CD16発現がより低く、活性化受容体NKp30、NKp44およびNKG2Dの発現がより高く、ならびにTRAILおよびNKG2Aの発現がより高い傾向を有した(図10)。
5人のドナー由来のNK細胞を使用して、371種の表面マーカーについて、フローサイトメトリーにより解析した。静止NK細胞と増殖性NK細胞のマーカープロファイル間の差を解析し、これをさらに、GFP発現集団とGFP陰性集団とに分けた。スクリーニングから、GFPを発現しない静止NK細胞と、形質導入することができGFPを発現する増殖性NK細胞との間で最良に識別されたマーカーを判定した。全体的に、静止NK細胞との直接比較では、増殖性およびGFP陽性のNK細胞は、32種のマーカーをより高いレベルで、5種のマーカーをより低いレベルで発現した(図11A)。これらのマーカーのうち、一部は、発現レベルが異なっただけでなく、特異的マーカーを発現するNK細胞の存在頻度も異なった。最も明白であったことは、増殖性およびGFP陽性のNK細胞間の、CX3CR1の発現の非存在およびTRAILを強力に発現する細胞の存在頻度の高さであった(図11B)。
増殖および形質導入と最良に相関した表面分子から、さらなる調査のためにCX3CR1を選択して、このマーカーが、増殖および形質導入の種々の能力を有するNK細胞サブセット間で直接識別可能かどうかを試験した。種々のNK細胞サブセットは、形質導入および培養の前に、MCASソーティングを使用して、それらのCX3CR1発現に基づいて分離した。ソーティングなしでは、種々のドナー由来の血液由来NK細胞間のCX3CR1の発現は、86%であり、すべてのNK細胞のうちの約14%の小画分のみが、CX3CR1を発現しなかったことを意味した(図5A)。ソーティング後では、CX3CR1negNK細胞の平均存在頻度は、CX3CR1の濃縮により2%に低下させることができ、一方、マーカーの枯渇により91%のCX3CR1negNK細胞の平均存在頻度がもたらされた(図12A)。別々にソートしたNK細胞画分の培養および形質導入の直後、本発明者らは、導入遺伝子発現および増殖率に関して有意差を観察した(図12B、12C)。非ソート画分間では、増殖率およびGFP発現は、それぞれ28%および21%であった。同時に、限界容量のCX3CR1陰性細胞を有するCX3CR1濃縮画分は、たった11%の増殖細胞および7%のGFP陽性細胞しか含有しなかった。印象的なことには、CX3CR1negNK細胞の高い開始存在頻度を有するCX3CR1枯渇画分間で、既に81%が増殖し、69%がGFP陽性であった。CX3CR1濃縮画分においても、増殖およびGFP発現は、残存CX3CR1neg細胞において、主に観察された(図12C)。まとめると、実験により、BaEVシュードタイプ化LVにより効率的に形質導入することができ、高度に増殖性のNK細胞のサブセットを同定した。このNK細胞サブセットは、CX3CR1発現の非存在により明らかに定義することができ、この特性は、形質導入可能なNK細胞の予備濃縮に使用することができる。
新たなNK細胞を末梢血単核細胞(PBMC)から単離し、IL−2/IL−15またはIL−2/IL−15/IL−1β(IL−1ベータ)で3日間刺激した。活性化後2日目に、感染多重度(MOI)3でリンパ球に形質導入した。翌日、細胞を洗浄し、IL−2/IL−15の存在下で、さらに7日間、増殖を継続させた。遺伝子修飾NK細胞の存在頻度を検出するために、CARのscFvドメインを特異的に標的とするビオチン化抗イディオタイプ抗体で細胞を標識した。ビオチンコンジュゲートの検出は、抗ビオチンPE 2次mAb標識化を使用して実施した。陽性細胞の割合は、各ドットプロットにより示し、IL−1ベータを含まずIL−2/IL−15で刺激したものと比較して、IL−2、IL−15およびIL−1ベータで刺激した新たなNK細胞により、優れた形質導入率をもたらすことを示す(68%対52%)。結果は、3つの独立した実験を代表する。

Claims (15)
- シュードタイプ化レトロウイルスベクター粒子またはそのウイルス様粒子により活性化NK細胞に生体物質を導入するためのin−vitroでの方法であって、
a)NK細胞を活性化するステップと、
b)前記シュードタイプ化レトロウイルスベクター粒子またはそのウイルス様粒子を前記活性化NK細胞に添加するステップと
を含み、前記シュードタイプ化レトロウイルスベクター粒子またはそのウイルス様粒子が、造血細胞膜に結合し、それと融合することが可能な修飾ヒヒ内在性レトロウイルス(BaEV)エンベロープ糖タンパク質を含み、それにより前記活性化NK細胞に生体物質を導入する、方法。 - NK細胞の前記活性化の前に、NK細胞が、CX3CR1陽性およびCX3CR1陰性NK細胞を含む試料から、CX3CR1陰性NK細胞について濃縮される、請求項1に記載の方法。
- 前記シュードタイプ化レトロウイルスベクター粒子またはそのウイルス様粒子が、少なくとも
ヒヒ内在性レトロウイルス(BaEV)エンベロープ糖タンパク質の膜貫通および細胞外ドメインと、マウス白血病ウイルス(MLV)エンベロープ糖タンパク質の細胞質尾部ドメインとの融合を含むか、もしくはそれにより構成されるキメラエンベロープ糖タンパク質、または
細胞質尾部ドメインが融合阻害Rペプチドを欠く、修飾BaEVエンベロープ糖タンパク質
を含む、請求項1または2に記載の方法。 - 前記NK細胞の前記活性化が、少なくとも1つのサイトカイン、またはフィーダー細胞もしくはフィーダー細胞の膜粒子、あるいはNK細胞活性化試薬を前記NK細胞に添加することにより達成される、請求項1〜3のいずれか一項に記載の方法。
- 前記少なくとも1つのサイトカインが、IL−2および/またはIL−15である、請求項4に記載の方法。
- NK細胞の前記活性化が、NK細胞を活性化する少なくとも1つのサイトカインと、IL−1ファミリーサイトカインとを含む、サイトカインの組合せを添加することにより達成され、それによりNK細胞の短期的活性化をもたらす、請求項1〜5のいずれか一項に記載の方法。
- サイトカインの前記組合せが、IL2および/またはIL−15と、IL−1ファミリーサイトカインである、請求項6に記載の方法。
- 前記IL−1ファミリーサイトカインが、IL−18、IL−33またはIL−1ベータである、請求項7に記載の方法。
- 前記シュードタイプ化レトロウイルスベクター粒子によるNK細胞の形質導入効率が、少なくとも50%である、請求項1〜8のいずれかに記載の方法。
- 前記生体物質が、前記レトロウイルスベクター粒子により前記NK細胞に形質導入される場合、1つまたは複数の核酸である、請求項1〜9のいずれかに記載の方法。
- 前記レトロウイルスベクター粒子またはそのウイルス様粒子が、レンチウイルスベクター粒子またはそのウイルス様粒子である、請求項1〜10のいずれかに記載の方法。
- 閉鎖系の自動化プロセスにおいて実施される、請求項1〜11のいずれかに記載の方法。
- 活性化NK細胞に生体物質をin−vivoで導入することによる、障害を患うヒトの治療における使用のための、シュードタイプ化レトロウイルスベクター粒子またはそのウイルス様粒子であって、前記シュードタイプ化レトロウイルスベクター粒子またはそのウイルス様粒子が、造血細胞膜に結合し、それと融合することが可能な修飾ヒヒ内在性レトロウイルス(BaEV)エンベロープ糖タンパク質を含み、それにより前記活性化NK細胞に生体物質を導入する、シュードタイプ化レトロウイルスベクター粒子またはそのウイルス様粒子。
- 活性化NK細胞に生体物質をin−vivoで導入することによる、障害を患うヒトの治療における使用のための、組成物の組合せであって、
a)NK細胞を活性化する少なくとも1つのサイトカインを含む第1の組成物と、
b)シュードタイプ化レトロウイルスベクター粒子またはそのウイルス様粒子を含む組成物
とを含み、前記シュードタイプ化レトロウイルスベクター粒子またはそのウイルス様粒子が、造血細胞膜に結合し、それと融合することが可能な修飾ヒヒ内在性レトロウイルス(BaEV)エンベロープ糖タンパク質を含む、組成物の組合せ。 - 前記第1の組成物が、NK細胞を活性化する少なくとも1つのサイトカインと、IL−1ファミリーサイトカインとを含む、サイトカインの組合せを含む、請求項14に記載の組成物の組合せ。
