Abstract
Description
まず、クルクマ・キサントリーザ抽出物を準備する。
本撹拌工程では、前述のクルクマ・キサントリーザ抽出物とアミノ酸類とを縮合剤の存在下において溶媒中で撹拌する。これにより、キサントリゾールアミノ酸エステルを含む反応混合物が得ることができる。
次いで、前述の工程で得られた反応混合物に、エステル系溶媒を加えて混合溶液を調製し、かかる混合溶液を酸性条件下で水抽出する。なお、前述の撹拌工程において、反応混合物を濃縮する工程を経て、粗キサントリゾールアミノ酸エステルの濃縮残渣(濃縮物)を得る場合、濃縮残渣(濃縮物)にエステル系溶媒を加えて混合溶液を調製し、かかる混合溶液を酸性条件下で水抽出する。
最後に、前述の工程で水抽出された抽出物に含まれるキサントリゾールアミノ酸エステルを加水分解して、キサントリゾールを得る。
まず、アミノ酸類としてN,N−ジメチルグリシン(472.30g、4.5774モル)と、縮合剤として1,1’−カルボニルジイミダゾール(CDI)(742.70g、4.5803モル)を、フラスコに仕込んだ。次いで、フラスコに、溶媒としてジクロロメタン(3000.0mL)をさらに加えて、室温で1時間撹拌した。その後、クルクマ・キサントリーザ抽出物(1000.0g、このうち21.2質量%がキサントリゾール)(PT.PHYTOCHEMINDO REKSA社製)を加えて、室温下で一晩撹拌した。翌日、室温下において、反応混合物に10%クエン酸水溶液(1000.0mL)をゆっくりと滴下し、その後30分撹拌した。静置後、有機相を分離した。さらに、分離した有機相を、10%クエン酸水溶液(1000.0mL)を用いて2回洗浄した。その後、洗浄した有機相を減圧下で濃縮して、粗キサントリゾールジメチルグリシンエステルの濃縮残渣(1116.3g)を得た。
本実施例と比較するための参考例として、MIKIO YAMAZAKI et al., “Studies on Pharmacologically Active Principles from Indonesian Crude Drugs.I.Principle Prolonging Pentobarbital−Induced Sleeping Time from Curcuma xanthorrhiza ROXB.”, Chem. Pharm. Bull. 36 (6) 2070−2074 (1988)(以下、引用参考文献という)に開示されている方法を考察した。
Claims (7)
- キサントリゾールを調製する方法であって、
クルクマ・キサントリーザ抽出物とアミノ酸類とを縮合剤の存在下において溶媒中で撹拌して、キサントリゾールアミノ酸エステルを含む反応混合物を得る工程、
前記反応混合物に、エステル系溶媒を加えて混合溶液を調製し、前記混合溶液を酸性条件下で水抽出する工程、および
前記水抽出された抽出物に含まれるキサントリゾールアミノ酸エステルを加水分解して、キサントリゾールを得る工程を含む、キサントリゾールの調製方法。 - 前記酸性条件下で水抽出する工程に使用される酸性水溶液は、クエン酸水溶液、酒石酸水溶液、塩酸水溶液、硫酸水溶液、酢酸水溶液、トリフルオロ酢酸水溶液、およびメタンスルホン酸水溶液からなる群から選択される1つ以上である、請求項1に記載のキサントリゾールの調製方法。
- 前記反応混合物に前記エステル系溶媒を加える前に、前記反応混合物を濃縮する工程をさらに含む、請求項1または2に記載のキサントリゾールの調製方法。
- 前記縮合剤は、1,1’−カルボニルジイミダゾール(CDI)、N,N’−ジシクロヘキシルカルボジイミド(DCC)、N,N’−ジイソプロピルカルボジイミド(DIC)、1−エチル−3−(3−ジメチルアミノプロピル)−カルボジイミド(EDC)、1−エチル−3−(3−ジメチルアミノプロピル)−カルボジイミド塩酸塩(EDC・HCl)、および1−[ビス(ジメチルアミノ)メチレン]−1H−1,2,3−トリアゾロ[4,5−b]ピリジニウム3−オキシドヘキサフルオロホスフェ−ト(HATU)からなる群から選択される1つ以上である、請求項1〜3のいずれか1項に記載のキサントリゾールの調製方法。
- 前記アミノ酸類は、N,N−ジメチルグリシン、N,N−ジエチルグリシン、無水ベタイン、サルコシン、N−メチルイミノ二酢酸、ニトリロ三酢酸、N,N−ジ(2−ヒドロキシエチル)グリシン、イミノ二酢酸、エチレンジアミン四酢酸、1,3−プロパンジアミン−N,N,N’,N’−四酢酸、ジエチレントリアミン五酢酸、N,N−ジメチルアラニンおよびそれらの塩からなる群から選択される1つ以上である、請求項1〜4のいずれか1項に記載のキサントリゾールの調製方法。
- 前記溶媒は、ジクロロメタン、ジエチルエーテル、テトラヒドロフラン、クロロホルム、エチルアセテート、ベンゼン、およびトルエンからなる群から選択される1つ以上である、請求項1〜5のいずれか1項に記載のキサントリゾールの調製方法。
- 前記水抽出された抽出物に含まれるキサントリゾールアミノ酸エステルを加水分解して、キサントリゾールを得る工程は、前記抽出物を濃縮して、濃縮残渣を得て、前記濃縮残渣に水および有機溶媒を加えてアルカリ性条件下で加水分解し、前記加水分解した溶液を濃縮し、その後蒸留することによりキサントリゾールを得る工程である、請求項1〜6のいずれか1項に記載のキサントリゾールの調製方法。
