Abstract
Description
(1) a) ヒトMHCクラスIb分子、又はペプチド抗原をT細胞に提示しうるポリペプチドであって、ここで、前記ポリペプチドがヒトMHCクラスIb分子の[α]3ドメイン、又は、ヒトMHCクラスIb分子の[α]3ドメインのILT2又はILT4に結合しうる誘導体を含み;

本発明による組成物は、医薬組成物の調製のための公知の標準に従って調製される。
例えば、組成物は、適切に保存及び投与されうるように調製されるが、それは例えば、担体、賦形剤及び/又は安定剤等の医薬的に許容される成分を用いることによる。
当該医薬的に許容される成分は、医薬組成物を患者に投与するときに用いられる量で毒性とはならない。医薬組成物に添加される医薬上許容される成分は、当該組成物中に存在する活性成分の化学的性質、医薬組成物の特定の用途及び投与経路に依存しうる。
一般に、本発明に関連して用いられる医薬的に許容される成分は、当技術分野で利用可能な知識、例えば、Remington's Pharmaceutical Sciences, Ed. AR Gennaro, 20th edition, 2000, Williams & Wilkins, PA, USAから入手可能な知識に従って用いられる。
上記ので特定されたペプチド抗原を含む、本発明で用いうるペプチド抗原は、MHC分子上への提示能以外には特に限定されない。MHC分子上に提示されうるペプチドは、当技術分野において公知の方法で作製しうる(例えば、Rammensee, Bachmann, Emmerich, Bachor, Stevanovic. 本発明の全ての実施形態の好ましい態様では、本発明により用いられるペプチド抗原は、MHCクラスI分子に対して予測される位置に、いかなるアンカー残基又は好ましいアミノ酸残基をも含む。

あるいは、本発明のペプチド抗原の非アンカーアミノ酸残基は、ヒトタンパク質由来のペプチド抗原の対応するアミノ酸残基と同一であってもよく、又は、少なくとも50%、好ましくは少なくとも60%、より好ましくは少なくとも70%、さらにより好ましくは少なくとも80%、さらにより好ましくは少なくとも90%の配列同一性を有してもよい。あるいは、本発明のペプチド抗原の非アンカーアミノ酸残基は、ヒトタンパク質由来のペプチド抗原の対応するアミノ酸残基に関して、保存的置換、好ましくは2以下の保存的置換、より好ましくは1の保存的置換を含んでよい。好ましい態様では、前記ヒトタンパク質は、病理的免疫反応の影響を受ける組織又は細胞で発現するタンパク質である。
本発明のペプチド抗原を含むペプチド抗原の合成方法は、当技術分野で周知である。

全ての工程は滅菌条件下で行われ、保護容器は層流フード下でのみ開放した。特に明記しない限り、細胞は常に350×gで5分間遠心分離した。全ての生細胞を、37℃、5% CO2、及び>95%湿度のインキュベーター中に維持した。37℃に設定した水浴を用いて、細胞に添加する培地、PBS又は他の溶液を予め加温した。細胞計数にはノイバウアーチャンバーを用いた。統計解析にはStudent's T検定を用いて、p値が0.05未満の場合を有意とした。
〔材料及び方法〕
JEG−3は、高レベルのHLA−Gを高レベルに発現するものの、古典的MHCクラスI分子をほとんど発現しない、ヒト絨毛がん細胞株である(Rinke de Wit et. al., J Immunol. 1990 Feb 1;144(3):1080−7)。JEG−3細胞を、10%ウシ胎仔血清、0.5%ピルビン酸ナトリウム溶液(100mM)及び1%ペニシリン(10kU/ml)及びストレプトマイシン(10mg/ml)溶液を含む完全RPMI1640培地(「完全RPMI」)で培養した。3×105個のJEG−3細胞を、12ウェルプレートに1mlの完全RPMIで播種した。
〔材料及び方法〕 1×106個のJEG−3細胞を未処理か、又は、6ウェルプレートに、1mlの完全RPMI1640のSTEAP1ペプチド(「st」、実施例1参照)で添加した。5×105 STEAP1特異的CD8+ T細胞を、5×105 PRAME特異的CD8+ T細胞(エフェクタ)と混合し、これらを未処理のままか又はJEG−3と共培養した。10μg/mlの中和抗ヒトHLA−G抗体(クローン87G、BioLegend ドイツ)を、指示された場所に添加した。翌日、HLA−A2+ UACC−257メラノーマ細胞(標的)を発現するホタルルシフェラーゼを、アキュターゼ溶液(PAA ドイツ)を用いて分離し、STEAP1ペプチド(5μg/ml、「st loaded」)又は同量のDMSO(「unloaded」)を37℃のシェーカー上で4時間洗浄し、次いで、ウェル当たり1×104個のUACC細胞を白色の丸底96ウェルプレートに播種した。次いで、非接着性混合T細胞を収集し、4×104個の初期T細胞(各々2×104個)の同等物及びホタルD−ルシフェリン(PJKドイツ、最終濃度140μg/ml)を添加した。8時間後に標的細胞の生存をルミノメーターで測定した(方法の詳細はBrown et al., J Immunol Methods. 2005 Feb;297(1−2):39−52)。
〔材料及び方法〕
図3に示す実験では、HLA−A2 STEAP1特異的CD8+ T細胞(CD8st)及びPRAME特異的CD8+ T細胞(CD8pr)を混合し、未処理のまま放置するか、又はSTEAP1ペプチドを添加した、又は添加しなかったJEG−3細胞と8時間共培養した(方法は図2を参照のこと)。次いで、懸濁液中のT細胞を収集し、ルシフェラーゼ発現PRAME−ペプチド(濃灰色バー)又はHLA−A2+ UACC−257黒色腫細胞(T細胞は前処理後には計測せず、初期エフェクタ:標的比1:1)を添加したSTEAP1−ペプチド(薄灰色バー)と組合せた。
MHC Ib陽性細胞株に関連して、混合CD8 T細胞クローンを1の同種ペプチドに事前に曝露すると、同種T細胞の細胞毒性ポテンシャルは約50%に低下したが、他方、他のT細胞クローンの細胞毒性活性は約90%にとどまり、これは、HLA−G+ JEG−3細胞単独のペプチド非依存性免疫抑制作用に匹敵した。その結果、このアプローチは、異なる(例えば、ウイルス)抗原に対する望ましい免疫応答を同時に損なうことなく、特異的(自己)免疫関連標的抗原に対して寛容を誘導しうることを示す。JEG−3細胞により示されたMHCパターン及び中和抗体を用いた以前の実験に基づき、これらのペプチド特異的効果はHLA−Gを介して介在されることが理解されるであろう。この実験は、MHC Ib分子上の抗原ペプチドの提示が、同種CD8+ T細胞の細胞溶解能を阻害する可能性を示唆する。
〔MHC Ibペプチド複合体の設計〕
HLA−G等のMHCクラスIb分子は、天然に、1つの複合体中に3つのポリペプチド分子からなる。図4に示すように、これらをリンカーで連結すると、安定性を改善しうる。
あるいは、図5に示すように、全ての構成要素を直線的に表示しうる。

〔材料及び方法〕
可溶性ペプチドMHC Ib構築物が抗原依存的にエフェクタT細胞を除去できるかどうかを検討するため、当該構築物を、IL−4、GM−CSF及びIL−10(DC−10)の存在下で増殖させた樹状細胞に添加した。DC−10−Medium(完全RPMI1640培地、10ng/ml IL−4、10ng/ml IL−10、100ng/ml GM−CSF)中で、健康なドナーから精製した5x106 MACS精製(CD14ビーズ、Miltenyi、ドイツ) CD14+細胞/mlを7日間培養してDC−10を生成した。3日目と5日目に新しい培地を添加した。得られたDC−10細胞は細胞培養皿に付着しなかった。次いで、1ml当たり4x105個のDC−10細胞を、STEAP1ペプチド(dtGsteap、配列は実施例4参照)、Melan A/MART−1ペプチド(ELAGIGILTV、dtGmelA)、又は対照上清を含む単鎖ジスルフィドトラップペプチドHLA−G構築物用のpCDNA3.1発現ベクターを用いて、Lipofectionで一過性トランスフェクションされたCHO細胞(1x106/ml)由来の同量の細胞培養5日目の上清と4時間混合した。次いで、DC−10をPBSで3回洗浄し、5hAB血清+ IL−2(106 DC−10/ml)を含む50μlのRPMI 1640培地に再懸濁した。次いで、5×104ペプチド−MHC Ib添加DC−10細胞を、STEAP1(CD8st)又はPRAME (CD8pr)のいずれかを認識する、HLA−A2制限抗原特異的CD8+ T細胞と16時間、1:1の比で組合せた。次に、製造業者の指示に従いCellEvent Caspase−3/7 Green (5μM、Life Technologies)、並びにヒトCD4(クローンEDU−2)及びCD8(クローンRPA−T8)に特異的な抗体で、細胞を染色した(実施例2を参照)。CD8+ CD4 Caspase−3/7細胞をフローサイトメトリーで定量した。
〔結果〕
図6に示すように、2つの独立した実験にて、STEAP1特異的T細胞は、同種ペプチドを提示する単鎖MHC Ib構築物を添加したDC−10細胞と組み合わされると、16時間以内にほぼ完全に消失した。同条件は対照ペプチド(CD8pr)に特異的なT細胞の生存に悪影響を及ぼさなかった。対照ペプチド(dtGmelA)を含む対照は、STEAP1特異的CD8+ T細胞の生存をわずかに低下させた。このことは、可溶性MHC Ib分子が特異的ペプチドと結合すると、提示されたペプチドに特異的なエフェクタT細胞を選択的に除去し、すなわち、特定抗原に対する免疫応答を選択的に調節しうることを示す。
図7Aに示す実験では、5×106末梢血単核細胞(PBMC)を、2人の独立した健常ドナーから採取し、Melan−Aペプチド又はSTEAP1ペプチドのいずれかを添加した1×106照射JEG−3細胞の存在下で、5%ヒト血清型AB血清、5ng/ml TGF−β1、20ng/ml IL−2(Treg培地)を含む2mlのRPMI1640培地中で14日間共培養した(上記のように添加)。3日目に新鮮な培地を添加した。7日目に培地を交換し、PBMCを新たに照射し、ペプチド添加した1×106個のJEG−3細胞に移した。Treg拡張ビーズ(Miltenyi Biotec、抗CD3/CD28)を、陽性対照として製造業者の指示に従って用いた。得られた細胞を、ヒトCD4(クローンEDU−2、Immunotools)及びCD25(Miltenyi 120−001−311)に対する抗体、並びにHLA−A2 STEAP1デキストラマー(STEAP1 dex、Immudex Denmark、全ての希釈物1:100)で氷上で30分間染色した。CD4+ CD25高Treg細胞中のSTEAP1特異的T細胞の頻度を、フローサイトメトリーにより定量した(Shevach et al., 2002, Nat. Rev. Immunol. 2:389)。STEAP1特異的CD4+ CD25高Treg細胞は、PBMCを単独(ctrl)又は対照ペプチド(melA)の存在下で培養した場合には検出されなかったが、PBMCを同種抗原を提示するJEG−3細胞と共培養した場合、有意な集団が繰り返し観察され、陽性対照設定(aCD3/28)の範囲はより低くなった。
マウスDC(mDC)は、野生型C57BL/6マウス由来の骨髄細胞を、マウスGM−CSF遺伝子をトランスフェクトしたAg8653骨髄腫細胞株由来の10% GM−CSF上清を添加した完全RPMI−1640中で7日間培養して作製した(詳細なプロトコル: Lutz et al., J Immunol Methods 1999, 223(1):77−92)。500μlの完全RPMIの4×105mDCを、模擬トランスフェクト細胞(CHO)由来、又は、一本鎖オボアルブミンペプチド(SIINFEKL)、マウスH−2Kb α1ドメイン及びマウスH−2Kb α2ドメイン、並びにヒトHLA−Gα3ドメイン(H2Kb、配列は実施例4のdtH2KbGova)又はヒトHLA−A2 α1及びヒトHLA−A2 α2ドメイン(A2G)をコードするpCDNA3.1ベクターでトランスフェクトされたCHO細胞由来の500μlの「第5日CHO上清」と4時間混合した。上清中の各構築物の存在をウエスタンブロット法により確認した。予備的な結果では、精製した構築物でも誘導が可能であることを示唆する。ここでは、cOmplete His−Tag精製樹脂(Sigma Aldrich)を用いて、ペプチド添加MHC構築物を精製して構築物を結合した後、PBS (3回)及び第Xa因子プロテアーゼ消化(1U/100μl、20℃、6時間、Qiagen)で洗浄して構築物を放出した。その後、第Xa因子は、第Xa因子除去樹脂(Qiagen、全て製造業者の指示に従う)を用いて除去しうる。配列は実施例4に記載されている。その後、mDCを次にPBSで洗浄した。

- a) ヒトMHCクラスIb分子、又はペプチド抗原をT細胞に提示しうるポリペプチドであって、ここで、前記ポリペプチドがヒトMHCクラスIb分子の[α]3ドメイン、又は、ヒトMHCクラスIb分子の[α]3ドメインのILT2又はILT4に結合しうる誘導体を含み;
b) 前記a)のMHCクラスIb分子又はポリペプチドにより提示されるペプチド抗原;

- 前記方法が、感染症又は悪性疾患の治療又は予防の方法である、請求項50〜66のいずれか一項に記載の組合せ。
- 前記疾患ががんであり、ペプチド抗原が腫瘍抗原である、請求項67に記載の組合せ。
- 前記がんが、メラノーマ、腎がん、卵巣がん、結腸直腸がん、乳がん、胃がん、膵管腺がん、前立腺がん、B細胞リンパ腫、T細胞リンパ腫、及び肺がんからなる群より選択される、請求項68に記載の組合せ。
- 前記組合せが1の医薬組成物中に存在する、請求項50〜69のいずれか一項に記載の組合せ。
- 前記抗原性タンパク質又はペプチド抗原に対する前記免疫応答が、前記抗原性タンパク質又はペプチド抗原に特異的である、請求項50〜70のいずれか一項に記載の組合せ。
- ヒト被験体に対するがんの治療方法で用いられる、請求項50〜62のいずれか一項に記載のMHCクラスIb分子とその受容体との結合を遮断しうる薬剤であって、前記方法が、前記がんの細胞からがん抗原の放出をもたらす治療を含む、薬剤。
- 前記がん抗原の放出をもたらす治療が、化学療法又は放射線療法である、請求項72に記載の薬剤。
- ペプチド抗原に対する免疫学的寛容を誘導する方法が、さらに、ペプチド薬物の治療を含み、前記ペプチド抗原が:1)前記ペプチド薬物と同一であるか、又は2)前記ペプチド薬物の断片であるか、又は3)前記ペプチド薬物に対する免疫学的寛容を誘導しうる前記ペプチド薬物の断片の誘導体である、請求項41に記載の医薬組成物又は組換えポリペプチド。
- ペプチド抗原に対する免疫学的寛容を誘導する方法が、さらに、タンパク質薬物の治療を含み、前記ペプチド抗原が1)前記タンパク質薬物の断片であるか、又は2)前記タンパク質薬物に対する免疫学的寛容を誘導しうる前記タンパク質薬物の断片の誘導体である、請求項41に記載の医薬組成物又は組換えポリペプチド。
- 前記ペプチド薬物は、前記ペプチド薬物そのものの形態で投与される、請求項74に記載の医薬組成物又は組換えポリペプチド。
- 前記タンパク質薬物は、タンパク質薬物そのものの形態で投与される、請求項75に記載の医薬組成物又は組換えポリペプチド。
- 前記ペプチド薬物は、前記ペプチド薬物をコードする遺伝子による遺伝子治療により投与される、請求項74に記載の医薬組成物又は組換えポリペプチド。
- 前記タンパク質薬物は、前記タンパク質薬物をコードする遺伝子による遺伝子治療により投与される、請求項75に記載の医薬組成物又は組換えポリペプチド。
