Abstract
Description

最近のデータは、腫瘍低酸素が転移性播種を促進する因子の分泌のための強力な推進力であることを示唆している8,9。転移の促進に関与されると思われる分泌された因子の重要な要素は、エキソソームの放出である。増加する証拠は、腫瘍由来のエキソソームにより担持されるプロテオーム及びゲノム情報の豊富な配列が、癌細胞が周囲の間質及び悪性細胞の行動を変える新規メカニズムであることを示差している10。エキソソームは、心血管新生11、免疫抑制12に関連するシグナル伝達プロセスに影響を与え、そして薬剤耐性及び発癌性転移を誘発する13-15.さらに、全身性変化を誘発するエキソソームの能力は、転移性播種を促進すると思われ、これは、患者の死因の大部分を占める16。
細胞株及び細胞培養物
ヒト非小細胞肺癌(NSCLC)細胞株を、American Type Culture Collection (ATCC)から購入した。全ての細胞株は、シュートタンデムリピート(STR)プロファイリングにより確認され、そしてマイコプラズマに対して陰性であることが見出された。全ての細胞は、5%CO2の加湿インキュベーターにおいて37℃で維持された。SKMES1細胞は、10%FBS(Gibco, Thermo Fisher Scientific)及びペニシリン−ストレプトマイシンにより補充されたDMEMにおいて培養された。全ての他の細胞は、10%FBS及びペニシリン−ストレプトマイシンにより補充されたRPMIにおいて培養された。低酸素実験のために、細胞は、2%CO2及び5%CO2の加湿されたインキュベーターにおいて37℃で培養された。

エキソソームは、透過型電子顕微鏡(TEM)を用いて可視化された。3μlのエキソソーム懸濁液は、50〜100μlの2%パラホルムアルデヒドにおいて固定された。次に2μlのマイクロフィルターアリコートは、2つのFormvar−炭素被覆された電子顕微鏡グリッドのそれぞれに移され、そして次に、20分間、覆われた。グリッドは洗浄され、そして50μlのシュウ酸ウラニル溶液(pH7)に、5分間、次に50μlのメチルセルロースUA(それぞれ、4%酢酸ウラニル及び2%メチルセルロースの100μl/900μlの比での混合物)に、氷上で10分間、移された。グリッドは取り外され、乾燥した後、80kVでJEM 1.011透過型電子顕微鏡で観察された。
エキソソーム濃度及びサイズは、45mmのストレッチでNP100ナノポアを用いてTRPS (qNano, Izon Science Ltd)により分析された。エキソソーム濃度及びサイズは、既知の濃度で70nmのカルボキシル化ポリスチレンビーズを用いた多重圧力較正を用いて標準化された。

細胞培養

Claims (22)
- 対象における癌の攻撃性の決定方法であって、前記対象のエキソソームサンプル中の1又は複数のマーカーの発現レベルを決定する工程を含み、ここで前記マーカーは、表1及び/又は表2に列挙されるそれらのタンパク質のうち1つ以上を含み、及び前記1又は複数のマーカーの発現レベルが、前記癌の攻撃性のレベルを示すか、又はそれと相関する、方法。
- 対象における癌の予後の決定方法であって、前記対象のエキソソームサンプル中の1又は複数のマーカーの発現レベルを決定する工程を含み、ここで前記マーカーは、表1及び/又は表2に列挙されるそれらのタンパク質のうち1つ以上を含み、及び前記1又は複数のマーカーの発現レベルが、前記癌についてのより好ましくないか又はより好ましい予後を示すか又はそれと相関する、方法。
- 1又は複数のマーカーの比較的低減された発現レベルが、より好ましい予後及び/又は低い攻撃性の癌を示すか、又はそれと相関し;及び/又は1又は複数のマーカーの比較的増加した発現レベルが、あまり好ましくない予後及び/又は高い攻撃性の癌を示すか、又はそれと相関する、請求項1又は2に記載の方法。
- (i)高い攻撃性の癌又は低い攻撃性の癌;及び/又は(ii)それほど好ましくない予後又はより好ましい予後を有するとして前記対象を診断するさらなる工程を含む、請求項1〜3のいずれか1項に記載の方法。
- 前記癌の予後又は攻撃性が、前記対象における癌の転移の可能性を決定するために、少なくとも部分的に使用される、請求項1〜4のいずれか1項に記載の方法。
- 前記1又は複数のマーカーの比較的低減された発現レベルが、前記癌の転移の低減された可能性を示すか又はそれと相関し;及び/又は前記1又は複数のマーカーの比較的増加した発現レベルが、前記癌の転移の増加した可能性を示すか又はそれと相関する、請求項5に記載の方法。
- 対象における抗癌治療に対する癌の応答性の予測方法であって、対象のエキソソームサンプル中の1又は複数のマーカーの発現レベルを決定する工程を含み、ここで前記マーカーは、表1及び/又は表2に列挙されるそれらのタンパク質のうち1つ以上を含み、及び前記1又は複数のマーカーの改変された又は調節された発現レベルが、抗癌治療に対する癌の比較的増加した又は低減された応答性を示すか又はそれと相関する、方法。
- 対象における癌を治療するさらなる工程を含む、請求項1〜7の何れか1項に記載の方法。
- 対象における癌の治療方法であって、前記対象のエキソソームサンプル中の1又は複数のマーカーの発現レベルを決定する工程を含み、ここで前記マーカーは、表1及び/又は表2に列挙されるそれらのタンパク質のうち1つ以上を含み、及び行われた決定に基づいて、抗癌治療を開始するか、継続するか、修正するか、又は中止することを含む方法。
- 前記抗癌治療が、1又は複数のマーカーの発現及び/又は活性を低減する、治療的有効量の抗癌剤の前記対象への投与を含む、請求項7〜9の何れか1項に記載の方法。
- 前記抗癌治療が、前記癌の転移を予防するか、又は阻害する、治療的有効量の抗癌剤の前記対象への投与を含む、請求項7〜10の何れか1項に記載の方法。
- 前記抗癌剤が、抗体又は小分子である、請求項10又は11に記載の方法。
- 前記対象からエキソソームサンプルを得る工程をさらに含む、請求項1〜12の何れか1項に記載の方法。
- 前記エキソソームサンプル中の1又は複数のマーカーの発現レベルと、それぞれ1又は複数のマーカーの参照エキソソーム発現レベルとを比較する工程をさらに含む、請求項1〜13のいずれか1項に記載の方法。
- 前記癌が肺癌であるか、又はそれを含む、請求項1〜14の何れか1項に記載の方法。
- 前記肺癌が、非小細胞癌であるか、又はそれを含む、請求項15に記載の方法。
- 対象における癌の治療への使用のための薬剤の同定又は製造方法であって、
(a)表1及び/又は表2に列挙されるマーカーを発現する細胞と、候補薬剤とを接触すること;及び
(b)前記候補薬剤が前記マーカーの発現及び/又は活性を調節するか否かを決定する工程を含む、方法。 - 前記候補薬剤が、前記マーカーの発現及び/又は活性を、少なくとも部分的に、低減するか、排除するか、抑制するか又は阻害する、請求項17に記載の方法。
- 請求項10〜16の何れか1項に記載の方法に従って使用するための、請求項17又は18に記載の方法により製造された薬剤。
- 前記1又は複数のマーカーが、ガレクチン−3−結合タンパク質、移行性小胞体ATPアーゼ、中性α−グルコシダーゼAB、60kDa熱ショックタンパク質、リシルオキシダーゼホモログ2、テネイシンC、脂肪酸シンターゼ、アグリン、アスパルチルアミノペプチダーゼ、プロテアソームサブユニットα1型、プロテアソームサブユニットα2型、プロテアソームサブユニットα3型、プロテアソームサブユニットα4型、プロテアソームサブユニットα5型、プロテアソームサブユニットα6型、プロテアソームサブユニットβ1型、プロテアソームサブユニットβ2型、プロテアソームサブユニットβ3型、プロテアソームサブユニットβ4型、プロテアソームサブユニットβ5型、プロテアソームサブユニットβ6型、プロテアソームサブユニットβ7型、プロテアソームサブユニットβ8型、トロンボスポンジン−1、潜在的トランスフォーミング増殖因子β結合タンパク質3、及びそれらの何れかの組合せから成る群から選択される、請求項1〜18の何れか1項に記載の方法、又は請求項19に記載の薬剤。
- 前記1又は複数のマーカーが、ガレクチン−3−結合タンパク質、移行性小胞体ATPアーゼ、テネイシンC、プロテアソームサブユニットα2型、トロンボスポンジン−1及びそれらの何れかの組合せから成る群から選択される、請求項20に記載の方法又は薬剤。
- 前記1又は複数のマーカーが、ガレクチン−3−結合タンパク質、移行性小胞体ATPアーゼ、テネイシンC、プロテアソームサブユニットα2型、中性αグルコシダーゼAB及びそれらのいずれかの組合せから成る群から選択される、請求項20に記載の方法又は薬剤。
