Abstract
Description

スペーサーは、その細胞標的化または浸透を改善するために、ステープルペプチドと追加のペプチドとの間に存在してもよい。
「細胞ターゲティングまたはステープルペプチドの浸透を改善するための追加のペプチド」として、Tat、ペネトラチンまたはPep1などの細胞浸透性ペプチドを挙げることができる。
ペプチド模倣大環状分子の形成をもたらすためのさまざまな方法が当該分野で公知である。たとえば、本発明のペプチド模倣大環状分子の製造は、Yuら(Chem. Soc. Rev.、2015、44、91)に記載されている。
(i)保護基を含む複数のペプチドを提供すること(各ペプチドは固体支持体に固定化されている);
(ii)脱保護試薬を固定化ペプチドに曝露して、固定化ペプチドの少なくとも一部から保護基を除去すること;
(iii)脱保護試薬の少なくとも一部を除去すること;
(iv)保護アミノ酸残基を溶媒、好ましくは、DMFに可溶化すること;
(v)カップリング試薬、好ましくは、HATUを使用すること;
(vi)塩基試薬、好ましくは、DIEAを使用すること;
(vii)活性化アミノ酸残基の少なくとも一部が固定化ペプチドに結合して新たに結合したアミノ酸残基を形成するように、保護アミノ酸残基およびカップリング試薬を固定化ペプチドに曝露すること;および
(viii)固定化ペプチドに結合しない活性化アミノ酸残基の少なくとも一部を除去すること;
(ix)ペプチド模倣大環状分子を生成するメタセシス反応のために最終的な直鎖状ポリペプチドをグラブス触媒試薬に曝露すること;
(x)最終的なペプチド模倣大環状分子を最終的脱保護のための開裂剤に曝露すること;
(xi)好ましくは95%を超える純度を得るための、最終的ペプチド模倣大環状分子の沈殿、精製および凍結乾燥。
本発明のもう1つの目的は、少なくとも1つの上記のペプチド模倣大環状分子、および薬学的に許容される担体を含む医薬組成物である。
医薬組成物は、アジュバントまたは追加の成分をさらに含みうる。アジュバントまたは追加の成分は、1つ以上のペプチドの生物学的活性を増強することができる。
医薬組成物は、注射(たとえば、筋肉内注射、皮下注射、静脈内注射、または腹腔内注射)、経口投与、局所投与、経皮投与、鼻腔内投与、または吸入用に製剤化することができる。
薬学的に許容される担体は、当業者に周知であり、たとえば、滅菌食塩水、ラクトース、スクロース、リン酸カルシウム、ゼラチン、デキストリン、寒天、ペクチン、植物油、脱イオン水およびそれらの混合物が挙げられる。
追加の成分および/または活性成分は上記に定義されている。
「薬学的に許容される誘導体」とは、レシピエントへの投与時に(直接的または間接的に)本発明の化合物を提供することができる、本発明の化合物の薬学的に許容される塩、エステル、エステルの塩、プロドラッグまたは他の誘導体を意味する。
いくつかの薬学的に許容される誘導体は、ペプチド模倣大環状分子の水溶解度または能動輸送を増加させる化学基を含む。
本発明はさらに、薬物としての使用のための、本発明による上記のペプチド模倣大環状分子、または医薬組成物もしくは組み合わせ剤に関する。
実施例においてさらに説明されるように、本発明のペプチド模倣大環状分子は、効率的であり、そして「同時」、「予防的」または「治療的」条件におけるそれらの使用に応じた特定の効率さえも有する。
1.1 炭化水素架橋を有するステープルペプチドの合成
IRAK2およびIRAKMステープルペプチドの合成は、手動でSPPS方法論によって行われる。合成スケールは、0.4 mmol/gで装填された40-RAM Amphisphereリンクアミド樹脂を用いて0.1 mmolであり、そして過剰の5当量が、保護されたアミノ酸のために使用される。すべての保護されたアミノ酸ならびにカップリング剤O-(7-アザベンゾトリアゾール-1-yl)-N,N,N',N'-テトラメチルウロニウムヘキサフルオロホスフェート(HATU)を、予めDMFに溶解して、濃度0.5Mの原液を作る。樹脂を最初に6mlのDMF中で15分間ボルテックス撹拌下で膨潤させる。DMFを濾過により除去した後、1分間ボルテックスしながら20%ピペリジン/DMF溶液(6ml)を添加することによりFmoc基の脱保護を行った。これを2回行う。樹脂をDMFで3回洗浄する。
IRAK2およびIRAKMラクタムペプチドの合成は、手動でSPPS方法論によって行われる。合成スケールは、0.4 mmol/gで装填された40-RAM Amphisphereリンクアミド樹脂を用いて0.1 mmolであり、そして過剰の5当量が保護されたアミノ酸のために使用される。すべての保護されたアミノ酸ならびにカップリング剤O-(7-アザベンゾトリアゾール-1-yl)-N,N,N',N'-テトラメチルウロニウムヘキサフルオロホスフェート(HATU)を、予めDMFに溶解して、濃度0.5Mの原液を作る。樹脂を最初に6mlのDMF中で15分間ボルテックス撹拌下で膨潤させる。DMFを濾過により除去した後、1分間ボルテックスしながら20%ピペリジン/DMF溶液(6ml)を添加することによりFmoc基の脱保護を行った。これを2回行う。樹脂をDMFで3回洗浄する。
所望のアミノ酸(1ml、0.5mmol)、N,N-ジイソプロピルエチルアミン(DIEA)(0.164ml、1mmol)およびHATUカップリング剤(1ml、0.5mmol)を連続して加え、ボルテックス撹拌下で5分間撹拌する。2回のカップリングの間に、この操作を、DMF洗浄により2回行う。アミノ酸fmoc-L-Lys(アリルオキシカルボニル)-OHおよびFmoc-L-Asp(Oall)-OHに関しては、1時間の攪拌時間により単純なカップリングを行う。Fmocのこの脱保護およびアミノ酸のカップリングを、予想される線状配列が得られるまで繰り返す。線状ペプチドが合成されると、30分間のボルテックスを2回行いながら、DCE(6ml)中のテトラキス(トリフェニルホスフィン)パラジウム(0)(0.04 mmol)およびフェニルアラニン(2 mmol)を使用して、アリルオキシカルボニルとアリルの脱保護を開始する。この反応を、各脱保護反応の間にDCM洗浄により2回行う。脱保護後、ラクタムは、N,N-ジイソプロピルエチルアミン(DIEA)(0.100ml、0.6mmol)およびHATUカップリング剤(0.6 ml、0.3 mmol)で実現される。
架橋化合物の精製は、逆相C18カラム(Waters)での高速液体クロマトグラフィー(HPLC)(PLC2250、Gilson)によって達成され、純粋な化合物が得られた。純粋な生成物の化学組成は、LC/MS質量分析(Waters 2790 HPLCシステムと接続されたMicromass ZQ)およびアミノ酸分析(Beckman System Gold High Performance Liquid Chromatograph)によって確認された。
2.1. 小胞体ストレス(ERストレス)の活性化
細胞内では、大部分の分泌タンパク質および膜タンパク質は、小胞体内で合成され、そこで輸送される前に折り畳まれ、組み立てられる。特定の条件下では、異常なコンホメーションのタンパク質が小胞体(ER)に蓄積し、ストレス(ERストレス)およびUPR(展開タンパク質応答)を誘導する。ERストレスはまた、NF-κB転写因子および炎症遺伝子(炎症性サイトカイン)の転写をさまざまな方法で活性化することによって炎症反応に寄与する。UPR応答は、標的遺伝子の転写の活性化および翻訳の徹底的な阻害を包含し、これは折りたたみおよび分解能力を増大させ、そして小胞体における新しいタンパク質の到達を制限する。研究は、IRAK2分子が、小胞体ストレスの誘導(Benosmabら、PLoS One 2013)と、IRAK4と直接相互作用することによるTLR(Toll様受容体)後期が仲介する抗ウイルス反応の両方に不可欠であること(カワゴエら、Nature Immunol 2008)を示す。
PMA(ホルボール-12-ミリステート-13-アセテート)による単球THP-1の刺激は、マクロファージにおけるそれらの分化を誘導する。これは、特定のTLR(2および4)の発現の誘導、IRAKファミリーのメンバー、NF-kB経路の活性化および炎症性サイトカインIL-1βの分泌と関連している。干渉IRAK1、IRAK2およびIRAK4に対するRNAを使用して、以前の研究は、mRNAレベルおよびタンパク質レベルの両方で、IL-1βの発現の減少を示した(Tiwariら、Journal of Immunology 2011)。
炎症は、感染および/または組織損傷に反応して主に先天性免疫細胞によって採用される複雑な生理学的状態である。適応は、過剰な炎症を調節し、エンドトキシンショックに対する宿主の防御として機能するために存在する。そのような保護メカニズムの典型的な例の1つはエンドトキシン耐性である。低濃度のエンドトキシン(LPS)に曝露された細胞または生物が、エンドトキシンによる新たな刺激に反応することができない一過性の低応答状態に入るという現象;言い換えれば、それらはエンドトキシンに対する一種の「耐性」を発症する。この現象は、単球およびマクロファージに対してインビトロで、ならびに動物モデルおよびヒトにおいてインビボの両方で観察されている(Biswas SKら、Trends in Immunology、2009)。耐性条件下では、IRAK-M(またはIRAK-3)は単球で過剰発現し(Escollら、BBRC、2003)、Toll様受容体依存性MyD88(TLR2、TLR4、TLR7/8、およびTLR9)の活性化を負に調節する。LPSに対する寛容性のインビトロTHP-1モデルにおける低応答は、LPSの2回の連続刺激後のTNFαおよびIL-6の発現低下、低濃度での第1の用量、続いて18時間後の強い用量によって特徴付けられる。天然のミドソーム阻害剤IRAK-Mの作用を模倣したステープルペプチド、THP1ステープルIRAK-2ペプチドへの単球の曝露は、LPS耐性の条件を再現すると予想される。
先の実施例は、ステープルペプチドが天然のミドソーム阻害剤IRAK-Mの作用を模倣することを示す。実際に、LPS刺激の6時間後のステープルペプチドIRAK2-S1およびIRAKM-S1へのTHP1単球の曝露は、LPS耐性条件、すなわちIL-6およびTNFαサイトカイン産生の阻害を再現したことが実証されている。本実施例は、異なる実験設定がより良い有効性を明らかにすることができるかどうかを研究する。本実施例は、異なる実験設定が、よりよい有効性を明らかにすることができるかどうかを研究する。LPS刺激の3つの異なる条件を比較した:単球のLPS刺激とステープルペプチドの付加が同時に行われる条件(同時条件:A)、ステープルペプチドがLPS刺激の6時間前(予防条件:B)または6時間後(治療条件:C)に添加される状態(図6)。
・「同時」条件(7A):試験した2つのペプチドについて、TNFαおよびIL-6発現の用量反応阻害が観察された。ペプチドIRAK2-S1およびIRAKM-S1は、10および20μMでTNFαおよびIL-6サイトカインの分泌を有意に阻害し、20μMでIRAKM-S1に対してより大きな効果を有し、これは、それぞれ最大95%および90%でTNFαおよびIL-6の発現を阻害する。しかしながら、この濃度では、ペプチドMock-S対照は、15〜20%の範囲でTNFαおよびIL-6の非特異的阻害を誘導する(図7A)。IRAKM-S1は、単球による炎症性サイトカイン産生を減少させるために、IRAK2-S1よりも効率的であるように思われる。
3つの異なる条件下(同時、治療的、および予防的処置)で、新しいステープルペプチドIRAK2-JMV6649およびIRAKM-JMV6650の、天然配列であるIRAK2 54-71およびIRAKM 66-83に対するマッチングについてのインビトロ有効性を評価し、IRAK2-S1およびIRAK-M-S1と比較した。試験した全ての条件について、THP-1単球を、10%ウシ胎児血清、1%ペニシリン/ストレプトマイシンおよびL-グルタミンを添加したRPMI中で培養した(300,000細胞/ウェル)。THP-1を0.1μg/mLのLPSで24時間処理し、ステープルペプチドIRAK2-S1、IRAK2-JMV6649、IRAKM-S1およびIRAKM-JMV6650を10〜20 μMで使用し、「同時」条件については同時に、「予防的」条件についてはLPSの6時間前に、「治療的」条件についてはLPSから6時間後に添加した。ELISA(eBioscience)を用いて、単球培養上清中のIL-6およびTNFαの定量を実施した。
「同時」条件:20μMで、IRAKM-JMV6649は、ペプチドIRAKM-S1と比較して、IL-6の分泌を70%以上減少させるが、TNFα発現に対する相加的な阻害効果を示さなかった(図8A)。JMV6650修飾によるIRAK2ペプチドのこれ以上の有効性は、観察されなかった(図8B)。
「予防的」条件:IRAKM-JMV6649ペプチドは、10μMで60%および20μMで80%のさらなる阻害により、IRAKM-S1ペプチドよりもIL-6発現の阻害を誘導する。TNFα発現の阻害は、修飾ペプチドIRAKM-JMV6649の方が大きく、10 μMで20%を達成する(図9A)。JMV6650修飾によるIRAK2ペプチドのこれ以上の有効性は観察されなかった(データは示さず)。
Claims (16)
- 少なくとも1つの大環状分子形成リンカー、および
(i)ヒト配列IRAK2 54-71(配列番号:1)に対して約50%、60%、70%、80%、90%、または95%の配列同一性および5-6、9-11、13-15位、好ましくは、14-15位のアミノ酸と100%の同一性を有するアミノ酸配列;または
(ii)ヒト配列IRAKM 66-83(配列番号:2)に対して約50%、60%、70%、80%、90%、または95%の配列同一性および5-6、9-11、13-14位のアミノ酸と100%の同一性を有するアミノ酸配列;
からなる群から選択されるアミノ酸配列を含む、3〜80個のアミノ酸からなるペプチド模倣大環状分子であって、
α-ヘリックス、ならびにiおよびi+3位置、またはiおよびi+4位置、またはiおよびi+7位置において大環状分子形成リンカーによって架橋された少なくとも2つの天然または非天然アミノ酸を含む、ペプチド模倣大環状分子。 - 大環状分子形成リンカーが、任意に置換された、飽和または不飽和の炭化水素鎖を含む、請求項1に記載のペプチド模倣大環状分子。
- 大環状分子形成リンカーが、アルキレン、アルケニレン、アルキニレン、およびそれらから誘導されるもの、好ましくは、アルキレンからなる群から選択される炭化水素鎖を含む、請求項2に記載のペプチド模倣大環状分子。
- 該炭化水素鎖の-CH2-基が、アミド官能基-NH-CO-で置き換えられている、請求項2に記載のペプチド模倣大環状分子。
- 大環状分子形成リンカーによって架橋された2つの天然または非天然アミノ酸が、iおよびi+4位置にある、前記請求項のいずれか1つに記載のペプチド模倣大環状分子。

- 請求項1〜9のいずれか1つに記載のペプチド模倣大環状分子と、請求項11に記載の少なくとも1つの追加の成分および/または活性物質とを含む組み合わせ剤。
- 薬物としての使用のための、請求項1〜9のいずれか1つに記載のペプチド模倣大環状分子、または請求項10もしくは11に記載の医薬組成物、または請求項12に記載の組み合わせ剤。
- 炎症、特に急性または慢性の炎症の予防または治療における使用のための、としての使用のための、請求項1〜9のいずれか1つに記載のペプチド模倣大環状分子、または請求項10もしくは11に記載の医薬組成物、または請求項12に記載の組み合わせ剤。
- 感染症または自己免疫疾患、特に、結核、敗血症、髄膜炎、関節リウマチ、多発性硬化症、クローン病、2型糖尿病、がん、心血管疾患、または細菌,ウイルス,寄生虫または真菌などの病原微生物によって引き起こされる病気などの急性および慢性炎症性疾患の予防または治療における使用のための、請求項1〜9のいずれか1つに記載のペプチド模倣大環状分子、または請求項10もしくは11に記載の医薬組成物、または請求項12に記載の組み合わせ剤。
- ペプチド模倣大環状分子が、炎症性疾患の診断前、診断中および/または診断後に投与される、請求項13〜15のいずれか1つに記載の使用のための、請求項1〜9のいずれか1つに記載のペプチド模倣大環状分子、または請求項10もしくは11に記載の医薬組成物、または請求項12に記載の組み合わせ剤。
