Abstract
Description
しかし、この方法は、図12に示すように、タンパク質の単分子の間に塩成分が析出しやすく、純粋なたんぱく質の結晶化が困難である。
しかし、この方法は、図13に示すように、タンパク質の単分子の間に界面活性剤成分が入り込みやすく、純粋なたんぱく質の結晶化が困難である。
しかし、電磁波は、弱めあったり強めあったりする性質があり、また、尖った部分に集中しやすい性質がある。さらに、電場が印加された状態で2個または3個の金属ナノ粒子が近接すると、ホットスポット(ホットスペース)と呼ばれる局所的な高温領域が発生することが知られている。
(1)生体高分子の含有溶液に電磁波を照射しながらナノ構造基板を含侵して生体高分子を濃縮化する濃縮化方法において、当該ナノ構造基板は基材、当該基材に固定された基体群、および当該基体群に析出された金属層群から構成され、当該基体群は離反して当該基材に固定され、当該金属層群の総放射面側における半球体換算の幾何学的な表面積(SA)と総受光面側における幾何学的な裏面積(SB)との比(SA/SB)が1を超えることを特徴とする生体高分子の濃縮化方法。
(4)上記金属層が山谷構造を有していることを特徴とする(1)または(2)に記載の方法。
以下本発明における原理を図面によって説明する。本発明におけるタンパク質の結晶化の原理は、図1の(a)〜(f)のステップで基本的に示される。
(a)に示す図は、過飽和溶液中で遊離したタンパク質を1単位の分子として模式的に図示したものである。隣接する析出層間には隣接距離が近づくとともにタンパク質単分子が連続的により強く変質されていく電場的分極がある。このため、一方の析出層の表面に吸着されたタンパク質分子は隣接距離の狭い谷のほうへ移動する。
(b)に示す図は、電場的分極が最も強い領域にタンパク質分子が位置したときの状態を示すものである。隣接する析出層間に挟まれたタンパク質分子は強力な電場的分極を受けるので、水平方向に強い分極作用を受け変質する。
(c)に示す図は、最も下の領域でタンパク質分子が紙面の奥方向へ連結されていくときの様子を模式的に示すものである。連結した複数のタンパク質分子のクラスターは全体で1単位の分子として作用するようになる。また、紙面の奥方向へ進むにしたがって電場的分極が弱まっていくので、適当な個数のタンパク質分子のクラスターがブラウン運動によって最も下の領域から切断される。
(d)に示す図は、最も下の領域から適当な個数のタンパク質分子のクラスターが切断された直後に、新たなタンパク質分子が落ちてきた場合を示すものである。新たなタンパク質分子の足と切断されたタンパク質分子の足が相互作用によって対をなす。新たなタンパク質分子は、上記の(c)ステップのように適当な個数の後のタンパク質分子のクラスターを構成し、やがて先のタンパク質分子のクラスターと後のタンパク質分子のクラスターが対をなす。なお、新たなタンパク質分子のクラスターの足と切断されたタンパク質分子のクラスターの足が(e)ステップで対をなさない場合は、後述する(f)ステップで先後のタンパク質分子のクラスターが対をなすと考えられる。
(f)に示す図は、析出層の山々に囲まれたたまり場でタンパク質分子のクラスター対と遊離したタンパク質単分子が自律的に再配列し、多数のタンパク質断片を形成した様子を示す模式図である。このタンパク質分子のクラスター対は電場的分極を受けているので変質しやすく、自律的に最適なタンパク質断片の位置へ再配列することができる。また、再配列したタンパク質断片は活性があるので、濃集して、より活性の高い活性サイトを形成することができる効果がある。
(生体高分子の含有溶液)
本発明において、生体高分子には生体高分子の単分子も含む。また、含有溶液には生体高分子のまま単離している場合と生体高分子がイオン的に溶解している場合も含む。すなわち、本発明において、生体高分子を含有する溶液は溶液中に生体高分子が集合体として存在していてもよく、可溶化して単分子として存在していてもよいことを意味する。好ましくは過飽和溶液である。生体高分子の(重量平均)分子量は1,000以上であることが好ましく、1,000以上100万以下がより好ましい。
本発明においては、金属層群の総放射面側からナノ構造基板をみたとき、金属層群が山谷構造を有していることが好ましい。また、基体群が金属微粒子群であることがより好ましい。また、金属層群または金属層群および基体群の複合粒子群がプラズモン特性を示すことがさらに好ましい。また、山谷構造は、金属層群の総放射面側において滑らかな連続膜であることが好ましい。析出された金属層群がなく、隣接する金属微粒子が基材に離れて固定された、基体群だけからなるナノ構造基板の場合、総放射面側における半球体換算の幾何学的な表面積(SA)と総受光面側における幾何学的な裏面積(SB)との比(SA/SB)は1である。
本発明において、金属層は液相または気相から析出された金属の層である。いわゆる湿式めっきまたは乾式めっきされた金属の層である。液相が好ましく、水溶液から還元析出された、例えば湿式めっきがより好ましい。湿式めっきには置換めっき、化学めっき、無電解めっき、電気めっきなどさまざまなめっき手法を用いることができる。さらに好ましくは、置換めっきや化学めっきや無電解めっきなどの湿式めっきによる金属層、特に自己触媒的に析出する無電解めっきが好ましい。高さが不ぞろいな金属層からなる複合粒子群が得やすいからである。このような複合粒子群の活性面を生体高分子の吸着・脱離作用に用いることができる。なお、湿式めっきは、析出速度の極端に遅い自己触媒式無電解めっき液または置換めっき液を用いてめっき作業を行うことが特に好ましい。
本発明の濃縮化または結晶化用ナノ構造基板において、微粒子体が離散して基板に固定されることとしたのは、入射する電磁波を基体群に均等に分散させるためである。微粒子体の材質は特に限定されない。金属、セラミック、ガラス、プラスチックなどあらゆる素材を用いることができる。微粒子体は、インクジェットや3Dプリンターによって水平方向に間隔をあけて整列することができる。インクジェット等で小さな半球を形成した場合は、小さな半球体上に大きな半球体が接合された形状を構成する。また、真空蒸着のような乾式めっきによって微粒子体を形成することができる。さらに、可視光に透過性のある基材にV溝やくぼんだ凹みを設けて微粒子体を整列させることができる。
本発明においては、ナノ構造基板に電磁波が照射される。入射した電磁波によって複合粒子から電場的分極が発生し、この電場的分極が生体高分子に作用するからである。本発明のナノ構造基板を用いると、電磁波による分極作用が緩和されているので、生体高分子が変質したり、劣化したりすることが少なくなる。電磁波発生装置の出力は弱めることができる。また、表面積(SA)と裏面積(SB)との比(SA/SB)を1.05以上にすることもできる。
本発明のナノ構造基板において、基材は600nm波長における吸光度が0.01〜1.0の樹脂フィルムまたはガラスであることが好ましい。吸光度0.05以上の樹脂フィルムであることがより好ましい。ガラスよりも樹脂フィルムのほうが便利だからである。例えばポリイミド樹脂、ポリアミド酸樹脂、フルオレン樹脂、ポリシロキサン樹脂、ポリエチレンテレフタレート樹脂、ポリフェニレンエーテル樹脂、エポキシ樹脂、フッ素樹脂、ビニル樹脂、フェノール樹脂等や、イオン交換樹脂などを挙げることができる。この樹脂材料は、単独の樹脂からなるものであっても良く、複数の樹脂を混合して用いたものでも良い。
本発明の結晶化容器は、本発明のナノ構造基板を備えた結晶化容器である。また、本発明の結晶化容器は、容器自体と本発明のナノ構造基板とが物理的または化学的に結合していても、結合していなくともよい。具体的には、例えば、本発明の結晶化容器中に本発明のナノ構造基板の蓋を接着してもよいし、本発明のナノ構造基板を容器中に単に入れるだけでもよい。また、本発明の結晶化容器は、本発明のナノ構造基板を1つのみ有していても、2以上有していてもよい。2個以上有している場合、特に多数個も受けた場合には、サンプルを同時に並行処理することができる。
本発明の生体高分子の濃縮化方法は、濃縮した生体高分子群の溶液を冷暗所で保管する保管工程を更に含むことができる。電磁波照射工程と同時に保管工程を行ってもよい。例えば、電磁波照射を行いながら生体高分子群の溶液を静置して結晶を生成させてもよい。
また、本発明の生体高分子の濃縮化方法における結晶化の形式は、特に制限はなく、公知の方法を用いることができる。特に、生体高分子の結晶化を行う場合には、例えば、ハンギングドロップ蒸気拡散法、シッティングドロップ蒸気拡散法、サンドイッチドロップ蒸気拡散法、ミクロ透析法、自由界面拡散法、保管バッチ法等の方法を好適に用いることができる。その他の生体高分子の結晶化を促進する条件は、日本生化学会編、新生化学実験講座1「蛋白質I−分離・精製・性質−」、高野常弘氏執筆、第14章「結晶化」、および、A. McPherson著、“Preparation and Analysis of Protein Crystals”(John Wiley & Son、 Inc.)などを参照することができる。
透明な半硬化性のポリエステル樹脂フィルム(ガラス転移温度(実測値)140℃、吸収スペクトル曲線は図6の一番下の曲線)上に還元金微粒子群(平均粒径20nm)を自己集合化させ、所定の熱処理をしてこの還元金微粒子群を半分沈めて固定した。これを図2に示す。吸収スペクトル曲線は図6の下から2番目の曲線である。幾何学的な表面積(SA)と裏面積(SB)の比(SA/SB)は1である。このナノ構造基板を用いた以外は、後述する実施例1と同様にしてタンパク質の結晶化実験を行った。7日経過後も結晶が析出しなかった。
次に、60℃の無電解金めっき液(日本エレクトロプレイティング・エンジニヤース株式会社製プレシャスファブACG3000WXの改良浴)にこの透明な基材を15秒間浸漬し、1サイクルとした。この工程を6サイクル繰り返し、金金属層とした。すなわち、固定した金微粒子体上に金粒子を析出させた複合粒子群である。これを図4に示す。図4に示す金属層群の総放射面側における半球体換算の幾何学的な表面積(SA)は、総受光面側における幾何学的な裏面積(SB)よりも大きいことが自明である。
タンパク質はニワトリ卵白リゾチームを用いた。タンパク質の濃度は15mg/mL、沈殿剤としてNaCl 0.5/Mの溶液を調製した。この溶液の過飽和度は1.25であり、過飽和でありながら自発的に結晶化が起こらない準安定状態の溶液である。
偏光子によってキセノンランプの光を直線偏光にした以外は、実施例1と同様にしてタンパク質の結晶化実験を行った。実施例1と比較すると、約4倍の結晶が出現した。この結果から、図4に示すナノ構造基板の電場的分極によって、金表面に吸着・整列したタンパク質が濃縮化され、結晶核の形成が広範囲にわたって同時進行していったことがわかる。
金めっき工程を9サイクル繰り返して金金属層とした以外は、実施例1と同様にした。これを図5に示す。吸収スペクトル曲線は図6の一番上の曲線である。このナノ構造基板を用いた以外は実施例1と同様にして結晶化実験を行った。幾何学的な表面積(SA)と裏面積(SB)の比(SA/SB)が1を超えていることは自明である。実験開始から1日後に観察すると、図9に示すように、細かい結晶が多数出現した。
無電解銀めっきとした以外は、実施例1と同様にして銀金属層を形成した。このナノ構造基板を用いた以外は実施例1と同様にして4ウェルの同時結晶化実験を行った。実験開始から1日後に観察すると、4ウェル中1ウェルで細かい結晶が多数出現した。
膜タンパク質として、高度好塩菌ハロバクテリウム・サリナルムを培養し、19mg/mLまで濃縮した可溶化バクテリオロドプシンを得た。この溶液を含水率40%w/wのモノオレイン脂質と混合し、立方相を形成した。塩溶液としてpH=5.5、および3モルNa/リン酸緩衝液を用いて塩濃度を2.0Mとした。
7日後に、カットオフフィルターを介したキセノンランプの光を1時間照射した以外は、実施例1と同様にして4ウェルの同時結晶化実験を行った。14日後に観察したところ、4ウェル中2ウェルで結晶が出現した。さらに、28日後では4ウェル中3ウェルで膜タンパク質の結晶が出現した。
金めっき工程を4サイクル繰り返して金金属層とした以外は実施例1と同様にした。これを図3に示す。吸収スペクトル曲線は図6の下から3番目の曲線である。幾何学的な表面積(SA)と裏面積(SB)の比(SA/SB)が1を超えていることは自明である。7日後に観察したところ、結晶が出現していた。
Claims (12)
- 生体高分子の含有溶液に電磁波を照射しながらナノ構造基板を含侵して生体高分子を濃縮化する濃縮化方法において、当該ナノ構造基板は基材、当該基材に固定された基体群、および当該基体群に析出された金属層群から構成され、当該基体群は離反して当該基材に固定され、当該金属層群の総放射面側における半球体換算の幾何学的な表面積(SA)と総受光面側における幾何学的な裏面積(SB)との比(SA/SB)が1を超えることを特徴とする生体高分子の濃縮化方法。
- 生体高分子の含有溶液に電磁波を照射しながらナノ構造基板を含侵して生体高分子を結晶化する結晶化方法において、当該ナノ構造基板は基材、当該基材に固定された基体群、および当該基体群に析出された金属層群から構成され、当該基体群は離反して当該基材に固定され、当該金属層群の総放射面側における半球体換算の幾何学的な表面積(SA)と総受光面側における幾何学的な裏面積(SB)との比(SA/SB)が1を超えることを特徴とする生体高分子の結晶化方法。
- 上記金属層が山谷構造を有していることを特徴とする請求項1または請求項2に記載した生体高分子の濃縮化方法または結晶化方法。
- 上記基体群が金属微粒子群であることを特徴とする請求項1または請求項2に記載した生体高分子の濃縮化方法または結晶化方法。
- 上記金属層群が還元析出された金属または合金から構成されていることを特徴とする請求項1または請求項2に記載した生体高分子の濃縮化方法または結晶化方法。
- 上記金属層群または上記金属層群および上記基体群がプラズモン特性を示すことを特徴とする請求項1または請求項2に記載した生体高分子の濃縮化方法または結晶化方法。
- 上記生体高分子が膜タンパク質であることを特徴とする請求項1または請求項2に記載した生体高分子の濃縮化方法または結晶化方法。
- 電磁波が照射される生体高分子の濃縮化または結晶化用ナノ構造基板において、当該ナノ構造基板は基材、当該基材に固定された基体群、および当該基体群に析出された金属層群から構成され、当該基体群は離反して当該基材に固定され、当該金属層群の総放射面側における半球体換算の幾何学的な表面積(SA)と総受光面側における幾何学的な裏面積(SB)との比(SA/SB)が1を超えることを特徴とする生体高分子の濃縮化または結晶化用ナノ構造基板。
- 上記金属層が山谷構造を有していることを特徴とする請求項8に記載の生体高分子の濃縮化または結晶化用ナノ構造基板。
