Abstract
Description
本発明の腸内環境改善組成物は、3−ヒドロキシ酪酸(3HB)を有効成分として含有している。
腸内においては、酢酸などの有機酸の増加によりpHが減少し、その結果有害菌の生育が抑制され、アンモニア濃度が減少すると考えられる。
腸内環境を模した培養方法(特開2018−198560号公報)を利用し、所定濃度の3HBを培養系に添加することで、3HBが腸内環境にどのような影響を与えるかを調べる実験を行った。尚、3HBは、pH調整にNaOHを用いて3HBNa(3−ヒドロキシ酪酸ナトリウム)塩として回収したものを使用した。
当該培地の培養開始時のpHは6.5に調整した。
サンプル1〜9における培養30時間後のそれぞれのアンモニア濃度を測定した。アンモニア濃度は、培養30時間後に回収した培養液を使用し、アンモニア試薬AmVer HR TNT(東亜ディーケーケー株式会社)およびDRB200リアクター(HACH製)により常法に従って測定した。3HBNaを添加しないコントロールのサンプルについても同様にアンモニア濃度を測定した。
サンプル1〜9において、培養30時間後に回収した培養液を使用してそれぞれのpH値を測定した。結果を図2に示した。図2には、3HBNaを添加しないコントロールのpH値(6.5)を基準とし、各サンプル1〜9のpH値が変動した値をそれぞれ示した。
サンプル1〜9において、培養30時間後に回収した培養液を使用してそれぞれの有機酸(短鎖脂肪酸:酪酸、酢酸)の濃度を測定した。これら有機酸の濃度は、3HBNaの定量と同様に高速液体クロマトグラフィー(HPLC)により常法に従って測定した。
サンプル1,2において、上述したように3HBNa濃度が0.5重量体積%(3HB濃度換算:0.41重量体積%)である場合に加えて、3HBNa濃度を0.2重量体積%(3HB濃度換算:0.17重量体積%)および0.72重量体積%(3HB濃度換算:0.59重量体積%)とした場合における培養30時間後のそれぞれのアンモニア濃度を測定した。結果を図5に示した。図5には、3HBNaを添加しないコントロールのアンモニア濃度を基準とした場合のサンプル1,2のアンモニア濃度の増減率をそれぞれ示した。
サンプル1,2において、上述したように3HBNa濃度が0.5重量体積%である場合に加えて、3HBNa濃度を0.2重量体積%および0.72重量体積%とした場合における培養30時間後のそれぞれのpH値を測定した。結果を図6に示した。図6には、3HBNaを添加しないコントロールのpH値(6.5)を基準とし、各サンプル1,2のpH値が変動した値をそれぞれ示した。
サンプル1において、上述したように3HBNa濃度が0.5重量体積%である場合に加えて、3HBNa濃度を0.2重量体積%および0.72重量体積%とした場合における培養30時間後のそれぞれの酢酸の濃度を測定した。
4人の健常者の糞便試料(サンプル11〜14)を用いて、0.5重量体積%3HBNa(3HB濃度換算:0.41重量体積%)を添加し、上記特開2018−198560号公報記載の培養方法を用いて21時間の嫌気条件で培養を実施した。尚、培養の詳細については実施例1と同様であるため、詳細な説明は省略する。

- 3−ヒドロキシ酪酸を有効成分として含有し、腸内環境を改善する腸内環境改善組成物。
- 前記3−ヒドロキシ酪酸を0.01〜10重量体積%含有する請求項1に記載の腸内環境改善組成物。
- さらに添加剤を含有する請求項1または2に記載の腸内環境改善組成物。
- 経口摂取される態様としてある請求項1〜3の何れか一項に記載の腸内環境改善組成物。
