候補制御領域を同定するために、全ゲノム高分解能DNAメチル化マップを関心対象(神経節)の細胞タイプにおいて生成した。候補エンハンサーは、細胞タイプ特異的DNA低メチル化の存在に基づいて選択した。そのように選択されたエレメントは、マウス網膜内でのインビボ高スループットレポーターアッセイを使用して発現についてスクリーニングした。その後、神経節細胞特異的配列エレメントを合成し、最小プロモーター配列
の前でクローニングした。
AAV投与のために、麻酔下の動物の眼に先の尖った30ゲージのニードルによって水晶体に近い強膜内に穿刺した。Hamiltonシリンジによって網膜下に2μLのAAV粒子懸濁液を注射した。3週間後、単離された網膜を30分間にわたりPBS中の4%のPFA中で固定し、4℃でPBS中での洗浄工程を続けた。全網膜は、室温で1時間にわたり10%の標準ロバ血清(NDS)、1%のBSA、PBS中の0.5%のTriton X−100により処置した。PBS中の3%のNDS、1%のBSA、0.5%のTriton X−100中のモノクローナルのラット抗GFP抗体(Molecular Probes社;1:500)およびポリクローナルのウサギ抗マウス椎体アレスチン抗体(Millipore社:1:200)を用いる処置を室温で5日間にわたり実施した。二次ロバ抗ラットAlexa Fluor−488抗体(Molecular Probes社;1:200)、抗ウサギAlexa Fluor−633およびHoechstを用いた処置は、2時間にわたり実施した。切片を洗浄し、スライドガラス上にProLong Gold退色防止試薬(Molecular Probes社)とともに載せ、Zeiss LSM 700 AxioイメージャーZ2レーザー走査型共焦点顕微鏡(Carl Zeiss社)を使用して撮影した。
Claims (12)
- 配列番号1の核酸配列を含むかもしくはそれからなるか、または配列番号1の前記配列と少なくとも80%の同一性を有する少なくとも400bpの核酸配列からなる単離された核酸分子であって、方向選択性網膜神経節における遺伝子の特異的発現を、前記遺伝子をコードする核酸配列が前記単離された核酸分子に機能的に連結される場合にもたらす、単離された核酸分子。
- 最小プロモーター、例えば配列番号2の前記最小プロモーターをさらに含む、請求項1に記載の単離された核酸分子。
- ストリンジェントな条件下において、請求項1または2に記載の単離された核酸分子にハイブリダイズする配列を含む単離された核酸分子。
- 特定の細胞内での遺伝子発現を促進するエレメントとして、請求項1または2に記載の単離された核酸を含む発現カセットであって、前記単離された核酸は、方向選択性網膜神経節において特異的に発現される遺伝子をコードする少なくとも核酸配列に機能的に連結される、発現カセット。
- 請求項4に記載の発現カセットを含むベクター。
- 前記ベクターは、ウイルスベクターである、請求項5に記載のベクター。
- 方向選択性網膜神経節細胞内で遺伝子を発現させるための、請求項1もしくは2に記載の核酸、請求項4に記載の発現カセットまたは請求項5に記載のベクターの使用。
- 方向選択性網膜神経節内で遺伝子を発現させる方法であって、単離された細胞、細胞系または細胞集団を請求項4に記載の発現カセットでトランスフェクトする工程を含み、前記発現される遺伝子は、前記細胞が方向選択性網膜神経節であるか、または前記細胞が方向選択性網膜神経節を含む場合、前記単離された細胞、前記細胞系または前記細胞集団によって特異的に発現される、方法。
- 請求項4に記載の発現カセットまたは請求項5に記載のベクターを含む単離された細胞。
- 前記発現カセットまたはベクターは、前記細胞のゲノム内に安定的に組み込まれる、請求項9に記載の細胞。
- 前記遺伝子の産物は、光感受性分子、例えばハロロドプシンまたはチャネルロドプシンである、請求項1もしくは2に記載の単離された核酸分子、請求項4に記載の発現カセット、請求項5に記載のベクター、請求項7に記載の使用、請求項8に記載の方法または請求項9に記載の細胞。
- 請求項1または2に記載の単離された核酸分子を含む、方向選択性網膜神経節細胞内で遺伝子を発現させるためのキット。
