したがって、本発明の具体的な目的は、ワクチンによる予防および療法において使用するための、そのC末端で抗原と融合した配列番号1の配列のエキソソームアンカータンパク質を含むかもしくはそれからなる融合タンパク質を発現するヌクレオチド配列、または前記ヌクレオチド配列を含むDNA発現ベクターであって、配列番号1が以下の配列:

分子構築物および細胞培養物
すべての分子構築物は、IE−CMVにプロモートされたベクターに基づく。Nefmut(13)、Nefmut−GFP(13)、NefG2A−GFP(23)、wtNef(24)、およびHPV−E7(25)を発現するベクターの構築物については、既に記載されている。293T細胞、マウスのC2C12筋肉細胞、およびHPV−E7発現TC−1腫瘍細胞は、10%の熱不活性化ウシ胎仔血清(FCS)を加えたダルベッコ改変イーグル培地中で成長させた。トランスフェクションアッセイは、リポフェクタミン2000に基づく方法を使用して行い、C2C12細胞の場合には、新たにトリプシン処理した細胞にリポソームを添加することによって改変した。マウスの脾臓細胞とEL−4細胞、すなわち、9,10−ジメチル−1,2−ベンズアントラセンで処理した際にC57 BI/6マウスから最初に得たマウスの胸腺リンパ腫CD4+ T細胞の両方を、10%のFCSを補充したRPMI培地中で培養した。
エキソソームを、分画遠心分離によって、細胞上清から単離した。詳細には、上清を500×gで10分間、遠心分離した。次いで、上清を、10,000×gで30分間の第1の超遠心分離からなる分画遠心分離にかけた。次いで、上清を採取し、0.22μM孔径で濾過し、70,000×gで1時間、超遠心分離した。ペレット化した小胞を1×PBS中に再懸濁し、70,000×gで1時間、再度超遠心分離した。その後、ペレットを、1×PBSの最初の体積の1:100で再懸濁した。接種したマウスの血漿からのエキソソームの回収を、その実行時間が2倍である遠心分離を開始する前に試料を5倍希釈したことを除いて同様の方法で行った。回収したエキソソームの量を、製造業者の推奨に従ってAmplex Redキット(Molecular Probes)によって、アセチルコリンエステラーゼ(AchE)、すなわち、古典的なエキソソームマーカーの活性を測定することによって評価した。AchE活性をmU/mLとして測定し、ここで、1mUは、1ピコモルのアセチルコリンを、pH8.0で37℃にて、1分当たりコリンとアセテートに加水分解する酵素の量として定義される。
トランスフェクトされた細胞培養物由来の蛍光エキソソームは、FACS(Gallios、Beckman Coulter)によって直接検出されるか、または、血漿から単離したエキソソームの場合には、アルデヒド/サルフェートラテックスビーズ(Invitrogen Molecular Probes)に結合した際に分析されるかのいずれかであった。この目標を達成するために、試料を、回転プレート上で、室温にて終夜、5μlのビーズと共にインキュベートし、次いで、洗浄し、1×PBS−2%v/vのホルムアルデヒド中に再懸濁し、FACS分析した。
本明細書に記載した動物を用いるすべての研究は、実験動物保護に関するイタリアのEuropean Directive 86/609/EECで実施されてきたLegislative Decree 116/92に従ったEthical Committee of the Istituto Superiore di Sanita、Rome、Italy(プロトコール番号555/SA/2012)によって承認されている。研究において使用される動物は、本明細書で上述したLegislative Decreeに挿入されたガイドラインに従って収容され処理された。C57 BI/6マウスはCharles River Laboratoriesから購入し、エンドトキシンを含まないQiagen kitで精製したプラスミドDNAをそれぞれの後ろ肢に50μg、10日間隔で2回、i.m.接種した。マウスには、10日間隔で3回、DNAベクターを注入したマウスの血漿から精製した、6mU等量のAchE活性のエキソソームを皮下(s.c.)にも接種し、最後の免疫化の10日後に屠殺した。E7−およびNef特異的CD8+ T細胞免疫応答を検出するために、脾臓細胞を、C57 BI/6マウスのH−2Kb複合体に効率的に結合させるために既に特定されたHPV−E7またはHIV−1 Nef8もしくは9量体ペプチド、すなわち、E7(HPV−16 Gene Bank受託番号AAD33252.1)に対するDLYCYEQL(アミノ酸21〜28)(配列番号3)およびRAHYNIVTF(アミノ酸49〜57)(配列番号4)、ならびにNef(HIV−1、F12株、受託番号EMBL Z11530)に対するTAATNADCA(アミノ酸48〜56)(配列番号5)のいずれか5μg/mlの存在下で、IFN−γ Elispotマイクロウェル(Millipore)中の培養に供した。HPV E6特異的KLPQLCTEL(アミノ酸18〜26)(配列番号6)およびYDFAFRDL(アミノ酸50〜57)(配列番号7)ペプチドに結合するH−2 Kb(HPV−16 Gene Bank受託番号AAD33253.1)を無関連ペプチドとして使用した。o.n.インキュベーションの後、IFN−γ Elispotプレートを発色させ(Mabtech AB)、スポット形成細胞を分析し、Elispotリーダー(A.EL.VIS. Elispot reader and Analysis software GmbH)を使用して計数した。
蛍光顕微鏡による分析のために、接種したマウスの四頭筋由来の7μMの切片をクリオスタット(Leika CM 3050)切片作製によって調製し、スライドに配置した。次いで、切片を、褪色防止用封入剤と一緒に、4’,6’−ジアミジノ−2−フェニルインドール(DAPI、Vector Laboratories)とインキュベートした。最終的に、スライドにカバーガラスを載せ、次いで、Zeiss Axioskop 2 Plus蛍光顕微鏡で観察した。
CD8+ T細胞を、正の免疫磁気選択(Miltenyi Biotec)によって、接種したマウスの脾臓細胞から単離した。これらを、RPMI 10%のFCS中で6時間、カルボキシフルオレセインスクシンイミジルエステル(CFSE、Invitrogen)で以前に標識したEL−4細胞との共培養に供し、E7または無関連ペプチドのいずれかで終夜処理した。共培養は、U底96ウェルプレートにおける200μlのRPMI 20%中で、異なる細胞比(すなわち、20:1から5:1までのエフェクター/標的細胞)で実行した。その後、最終濃度1μg/mlでの7−AADの添加後間もなく、EL−4細胞の死亡率をFACS分析によってスコア化した。
接種したマウス由来の血漿をプールし、1:10から開始する2倍連続希釈物を、抗E7 Abの存在についてアッセイした。エンドポイント希釈は、450nmにおける吸光度がOD0.1未満に相当した。各血漿を三連でアッセイし、吸光度値の平均を最終読み出しとして得た。組換えE7とNefの両方をアッセイのために使用した。タンパク質を、炭酸緩衝液(pH9.4)中4℃で、1ウェル当たり0.25μgの濃度で、Maxisorpのマイクロタイタープレート(NUNC)中に終夜吸着させた。3%のノンファットドライミルク(NFDM)を含有するPBS中、37℃で2時間のブロッキングステップの後、プレートを、37℃で1時間、1%のNFDM−PBS中で連続希釈した血漿100μLと共にインキュベートした。特異的抗原−抗体複合体を、基質としてテトラメチルベンジジンを使用して、ペルオキシダーゼにコンジュゲートしたヤギ抗マウスIgG(GE Healthcare Ltd)によって検出した。室温で30分後、1ウェル当たり1Mの硫酸50μlを添加することによって、酵素反応を停止させた。洗浄ステップは、自動洗浄機中で、0.05%のTween−20を含有するPBSを1ウェル当たり200μl用いて行った。
Nefmut/E7発現ベクターの接種によって誘導された抗腫瘍活性を、2×105個のTC−1細胞を以前にチャレンジしたマウスにおいて評価した。DNA接種は、腫瘍細胞のチャレンジの4および11日後に、上記報告したプロトコールに従って、かつ触診できる腫瘍を発生したマウスにおいてのみ実施した。腫瘍成長は、目視検査、触診、および(長さ×幅2)/2として計算した腫瘍小結節の直径の測定によってモニタリングした。観察時間の終了時に、腫瘍を外植し、秤量した。
適当な場合に、データは、平均+標準偏差(SD)として表す。一部の例では、対応のあるStudentのt検定を使用し、ノンパラメトリックWilcoxon順位和検定を使用して確認した。p<0.05を有意とみなした。
DNAをトランスフェクトされたマウス筋肉細胞により放出された操作されたエキソソームの検出
筋肉細胞は、in vivoでの投与の際の異所性DNAの効率的かつ安定な発現に対する理想的な標的を表す。目的は、in vivoでNefmut系DNAベクターを発現させ、接種したDNAを発現する細胞によって構成的に放出されるエキソソームを操作することであった。これらの内因性エキソソームは、抗原特異的CTL免疫応答の誘導に関して、組織培養で生成されるものと少なくとも類似する特徴を示すことが期待される。
次に、Nefmut−誘導体を発現するDNAベクターの接種によって生じた操作されたエキソソームにアップローディングされた抗原の免疫原性を評価した。この目的のために、C57 BI/6マウス(1群当たり6頭)に、Nefmut/E7もしくはE7単独のいずれかを発現するベクター50μg、または空ベクターを、各後ろ肢に、i.m.接種した。とりわけ、E7単独を発現するベクターの注入後の免疫応答の分析は、CD8+ T細胞の免疫原性に関して、Nefmut融合の利益を評価する助けとなった。10日後に、接種を繰り返し、さらに10日後、マウスを屠殺し、無関連、NefまたはE7特異的H−2 Kb9量体の存在下で、IFN−γ Elispotマイクロウェル中で、脾臓細胞をo.n.培養した。無関連ペプチドとの培養で観察されたCD8+ T細胞の活性化レベルは、バックグラウンドレベルのままであり、ペプチドの非存在下で培養した脾臓細胞で検出されたものと同様であった(図示せず)。一方、細胞活性化は、E7またはNef9量体を接種した後に、Nefmut/E7発現ベクターを接種したマウス由来の脾臓細胞中で、はっきりと検出可能であった(図4A)。逆に、CD8+ T細胞応答は、いかなるペプチドが使用されても、E7発現ベクター、空ベクターのどちらを受けるマウス由来の脾臓細胞の培養においても検出されなかった。
最後に、抗腫瘍作用に関してNefmut/E7発現ベクターの注入によって惹起されるCD8+ T細胞免疫応答の効力を評価した。この目標を達成するために、2×105個のTC−1細胞をs.c.接種したC57 BI/6マウスに関する治療的免疫化アッセイを設定した。次いで、触診によって検出可能な、すなわち、約2mmの直径の腫瘍塊を発生するマウスに、細胞移植の4日後と11日後の両方に、後ろ肢当たり50μgの、空ベクター、Nefmut(各群当たり4頭のマウス)またはNefmut/E7(6頭のマウス)のいずれかを発現するベクターを接種した。対照として、4頭の腫瘍を移植したマウスに、ビヒクルのみを注入した。21日目に、E7特異的CD8+ T細胞免疫応答の誘導を評価するために供したNefmutまたはNefmut/E7発現ベクターを注入したマウスに関して、後眼窩採血を行った(図7A)。腫瘍の成長を30日にわたってモニタリングし、その後、マウスを屠殺し、腫瘍を外植し、秤量した。図7Bは、腫瘍重量の評価によって確認されるように、対照DNAベクターの注入は移植された腫瘍細胞の成長に影響を及ぼさなかったが、Nefmut/E7ベクターを接種したマウスでは、これらの増殖が著しく損なわれ、腫瘍細胞は、3頭のマウスにおいてさらに明らかに取り除かれたことをはっきりと示す(図7C)。
NefmutのC末端に融合した抗原を発現するDNAベクターの接種に基づく免疫化戦略は、様々なさらなるウイルス抗原に対しても首尾よく適用された(表1を参照のこと)。詳細には、Nefmutと融合したこのような抗原を発現するベクターを、本明細書の上記のスケジュールに従い、C57 BI/6マウスまたはBalb/cマウスのいずれかに注入した。最後の接種の10から15日後に、表1に列挙したペプチドを使用して、注入したマウス由来の脾臓細胞を用いて、IFNγ ELISPOTアッセイを行った。
材料および方法
分子構築物
活性化rHER2/neuの細胞外ドメイン(ECD)をコードするDNAを、N202.1A細胞、すなわち、rHER−2/neuにトランスジェニックであるFVBマウスに由来する細胞株から抽出した全RNAに関して行ったRT−PCRによって回収した(27)。それぞれの5’末端にNhe IおよびEco RI制限部位を含む以下のプライマーを使用した:フォワード(シグナルペプチドに対してすぐ下流)
293T細胞、MCF−7細胞、マウスのC2C12筋肉細胞(すべて、American Type Culture Collectionから入手した)、およびTC−1細胞(28)は、10%の熱不活性化ウシ胎仔血清(FCS)を加えたダルベッコ改変イーグル培地中で成長させた。トランスフェクションアッセイは、リポフェクタミン2000に基づく方法(Invitrogen、Thermo Fisher Scientific)によって行い、C2C12細胞の場合には、新たにトリプシン処理した細胞にリポソームを添加することによって改変した。HLA−A.02 B−LCL(29)、マウスの脾臓細胞、およびCD8+ Tリンパ球を、10%のFCSを加えたRPMI培地中で培養した。ヒトの初代骨格筋細胞(SKMC)はLonzaから入手し、推奨される培地で培養した。
エキソソームを、トランスフェクションの48から72時間後に、293T細胞の上清から出発して、以前に記載されたように(30)、分画遠心分離によって単離した。回収されたエキソソームの量は、製造業者の推奨に従ってAmplex Redキット(Molecular Probes、Thermo Fisher)によって、アセチルコリンエステラーゼ(AchE、すなわち、古典的なエキソソームマーカー)(31)の活性を測定することによって評価した。
細胞溶解物とエキソソームの両方のウエスタンブロット分析は、記載されているように(13)行った。フィルターを、1:1000に希釈したヒツジの抗Nef抗血清ARP444(MRC)、1:250に希釈したSigmaからの抗β−アクチンAC−74 mAb、および1:100に希釈したSanta Cruzからの抗Alix H−270ポリクローナルAbを使用して明らかにした。
本明細書に記載した動物を用いるすべての研究は、実験動物保護に関するイタリアのEuropean Directive 86/609/EECで実施されてきたLegislative Decree 116/92に従ったEthical Committee of the ISS(プロトコール番号107/2016−PR)によって承認されている。本発明者らの研究において使用される動物は、本明細書で上述したLegislative Decreeに挿入されたガイドラインに従って収容され処理された。ISSの動物施設(32)で産まれて飼育された129Sv−NeuTトランスジェニックマウスのコロニーを使用した。これらのマウスでは、活性化したrHER−2/neu遺伝子が、MMLV LTRによってプロモートされ、処女雌は、15〜20週齢で、触診可能となる乳癌を自発的に発生させる。rHER2/neu導入遺伝子の存在は、通常、記載されているように(32)、PCRによってチェックされた。マウスに、エンドトキシンを含まないQiagenのキットを用いて精製したプラスミドDNAを各四頭筋当たり50μg、15週齢と17週齢において、2回、i.m.接種した。乳腺を、腫瘍モニタリングの間、1週間に一度、検査した。直径30mmを超える腫瘍を保有するマウスを安楽死させた。
接種したマウス由来の血漿を、1:20に希釈し、HER2/neu発現ベクターを2日前にトランスフェクトされた293T細胞における抗HER2/neu抗体の存在について試験した。4℃での2時間のインキュベーション後、細胞を洗浄し、FITCにコンジュゲートした抗マウスIgGと共にインキュベートし、1時間後にFACS分析を行った。陽性対照として、1:20に希釈した抗HER2/neu mAbクローン7.16.4(Sigma)を使用した。
HER2/neu特異的CD8+ T細胞免疫応答とNef特異的CD8+ T細胞免疫応答の両方を検出するために、脾臓細胞を、129SvトランスジェニックマウスのH−2Kb複合体に結合するHER2/neuまたはHIV−1 Nef9量体ペプチド、すなわち、それぞれ、ILHDGAYSL(アミノ酸436〜444)(33)およびTAATNADCA(アミノ酸48〜56)(34)のいずれか5μg/mlの存在下で、IFN−γ Elispotマイクロウェル(Millipore)中に置いた。H−2Kbに結合する異種ペプチド(35)を対照として使用した。o.n.接種後に、IFN−γ Elispotプレートを発色し(Mabtech)、スポット形成単位(SFU)を計数した。
合計106個のSKMCに、Nefmut系ベクターまたは対照ベクターのいずれか10μgをトランスフェクトした。48時間後、細胞を、1:5の細胞比で、iDCとの共培養に供し、一部の例では、エキソソーム生合成の阻害剤GW4869とスピロエポキシド2μMの存在下で行った(36〜41)。終夜のインキュベーション後、iDCを単離し、24時間のLPS処理によって成熟させた。その後、iDCを洗浄し、1:10の細胞比で、自己末梢血リンパ球(PBL)との共培養に供した。1週間後、刺激手順を繰り返し、さらに1週間後、CTLアッセイのためにCD8+ T細胞を回収した。
マウス細胞を用いたCTLアッセイは、CD8+ T細胞を正の免疫磁気選択(Miltenyi)によって脾臓細胞から単離することによって実施した。これらを、RPMI 10%のFCS中で6時間、製造業者の推奨に従って、カルボキシフルオレセインスクシンイミジルエステル(CFSE、Invitrogen、Thermo Fisher)で以前に標識したTC−1細胞との共培養に供し、HER2/neu、Nef、または無関連ペプチドのいずれかで終夜処理した。共培養は、U底96ウェルプレートにおける200μLのRPMI 20%中で、10:1のエフェクター/標的細胞比で実行した。その後、最終濃度1μg/mlでの7−AADの添加後間もなく、TC−1細胞の死亡率をFACS分析によってスコア化した。ヒト細胞におけるCTLアッセイを、標的細胞としてMCF−7またはB−LCLを使用したことを除いて、同様の方法で行った。
GFPまたはNefmut/GFPのいずれかを発現するベクターを用いて2日前にトランスフェクトされたiDCとSKMCとを含む終夜にわたる共培養を、GW4869とスピロエポキシドの存在下またはそれら両方の不在下で、1:5の細胞比で行った。その後、細胞を、最初に、4℃で1時間、抗CD45(すなわち、iDCのマーカー)で染色し、次いで、Alexa−Fluor 610にコンジュゲートした二次Abで染色した。最後に、共培養物を4’,6’ジアミノ−2−フェニルインドール(DAPI、Vector Laboratories)で標識し、緩衝したホルムアルデヒド(2%v/v)中で固定した。位相差と蛍光画像をOlympus IX−81デバイスで記録した。
適当な場合に、データは、平均+標準偏差(SD)として表す。一部の例では、対応のあるStudentのt検定を使用し、ノンパラメトリックWilcoxon順位和検定を使用して確認した。p<0.05を有意とみなした。
rHER2/neuの細胞外ドメインは、Nefmutと融合した際にエキソソームに効率的にアップローディングされる
次に、研究の目的は、HER2−ECD特異的CTL活性が検出可能な抗腫瘍活性と結び付くかどうかを評価することであった。未だ触診可能な病変を有さない15週齢の129Sv−Neu Tトランスジェニックマウスに、ビヒクルまたはNefmutおよびNefmut/HER2−ECDを発現するDNAベクターのいずれかを注入した。注入を2週間後に繰り返し、触診可能な腫瘍の外観を毎週モニタリングした。図12に報告したように、Nefmut/HER2−ECDを発現するDNAの注入は、腫瘍発生の著しい遅延と関連した。モニタリングは、倫理的理由で必要な最初の屠殺時に停止した。

Claims (12)
- CTL免疫応答を誘導することによるワクチンによる予防および療法において使用するための、そのC末端で免疫原性抗原と融合した配列番号1の配列のエキソソームアンカータンパク質を含むかもしくはそれからなる融合タンパク質を発現するヌクレオチド配列、または前記ヌクレオチド配列を含むDNA発現ベクターであって、配列番号1が以下の配列:
- 筋肉内投与によって投与される、請求項1に従って使用するための、請求項1に記載のヌクレオチド配列またはDNA発現ベクター。
- 前記抗原が、E6およびE7などのヒトパピローマウイルス抗原、GagおよびTatなどのHIV抗原、VP24、VP40、NP、およびGPなどのエボラウイルス抗原、NS3などのウエストナイルウイルス抗原、CoreなどのHBV抗原、Core、NS3、E1およびE2などのHCV抗原、GPおよびNPなどのクリミアコンゴウイルス抗原、NPおよびM1などのインフルエンザAウイルス抗原、MAGE−A3およびMART−1などのヒトメラノーマ抗原、Her2/Neu、Hox B7などのヒト腫瘍関連抗原からなる群より選択される、請求項1から2のいずれか一項に従って使用するための、請求項1から2のいずれか一項に記載のヌクレオチド配列またはDNA発現ベクター。
- 配列番号1の配列のエキソソームアンカータンパク質を発現するヌクレオチド配列が、以下の配列番号2のヌクレオチド配列:
- HBV、HCVおよびHIVなどの慢性感染疾患、結核およびマラリア、インフルエンザ、ウエストナイル、クリミアコンゴ出血熱およびエボラ疾患などの急性感染疾患、乳房、肺、前立腺または膀胱腫瘍などの腫瘍からなる群より選択される疾患の予防および処置のために、請求項1から4のいずれか一項に従って使用するための、請求項1から4のいずれか一項に記載のヌクレオチド配列またはDNA発現ベクター。
- そのC末端で抗原と融合した配列番号1の配列のエキソソームアンカータンパク質を含むかもしくはそれからなる融合タンパク質を発現するヌクレオチド配列、または前記ヌクレオチド配列を含むDNA発現ベクターを、1つまたは複数の薬学的に許容される賦形剤および/またはアジュバントと共に含むかまたはそれらからなる、免疫原性である医薬組成物であって、配列番号1が以下の配列:
- 前記抗原が、E6およびE7などのヒトパピローマウイルス抗原、GagおよびTatなどのHIV抗原、VP24、VP40、NP、およびGPなどのエボラウイルス抗原、NS3などのウエストナイルウイルス抗原、CoreなどのHBV抗原、Core、NS3、E1およびE2などのHCV抗原、NPおよびGPなどのクリミアコンゴウイルス抗原、NPおよびM1などのインフルエンザAウイルス抗原、MAGE−A3およびMART−1などのヒトメラノーマ抗原、Her2/Neu、Hox B7などのヒト腫瘍関連抗原からなる群より選択される、請求項6に記載の医薬組成物。

- 前記アジュバントが、CD8+ T細胞応答のアジュバントである、請求項6から9のいずれか一項に記載の医薬組成物。
- ワクチンによる予防および療法において使用するための、請求項6から10のいずれか一項に記載の医薬組成物。
- HBV、HCVおよびHIVなどの慢性感染疾患、結核およびマラリア、インフルエンザ、ウエストナイル、クリミアコンゴ出血熱およびエボラ疾患などの急性感染疾患、乳房、肺、前立腺または膀胱腫瘍などの腫瘍からなる群より選択される疾患の予防および処置のために、請求項11に従って使用するための、請求項11に記載の医薬組成物。
