Abstract
Description
T細胞レセプター(TCR)はCD4+及びCD8+T細胞により天然に発現される。TCRは、主要組織適合性複合体(MHC)分子(ヒトでは、MHC分子はヒト白血球抗原又はHLAとしても知られる)と複合体化して、抗原提示細胞の表面に提示される短いペプチド抗原を認識するよう設計されている(Davisら(1998), Annu Rev Immunol 16:523-544.)。CD8+T細胞(細胞傷害性T細胞とも呼ばれる)は、MHCクラスIに結合したペプチドを特異的に認識し、概して罹患した細胞の発見及びその破壊の媒介を担っている。CD8+T細胞はガン細胞及びウイルス感染細胞を破壊することができる;しかし、天然のレパートリ中のガン特異的T細胞が発現するTCRの親和性は、代表的には、胸腺選択の結果として低く、このことは、ガン細胞が頻繁に検出及び破壊を免れることを意味する。T細胞によるガン認識の促進を目的とする新規な免疫療法アプローチは、有効な抗ガン治療の開発に関して高度に有望なストラテジを提案する。
第1の観点において、本発明は、HLA-A*02と複合体化したGVYDGREHTV(配列番号1)に対する結合性を有し、TCR α鎖可変ドメイン及び/又はTCR β鎖可変ドメインを含んでなるT細胞レセプター(TCR)であって、
α鎖可変ドメインは、配列番号2のアミノ酸残基1〜113の配列に対して少なくとも90%の同一性を有するアミノ酸配列を含み、及び/又は

本発明のTCRは、β鎖FR2領域及びβ鎖FR3領域を含んでなってもよく、ここで、該FR2領域及び該FR3領域は、それぞれ配列番号14及び15を含んでなり、並びに/又は、1若しくは2以上(例えば1、2又は3つ)の保存的置換及び/若しくは3つまでの寛容される置換を含む。

特定の実施形態において、α鎖CDRには1、2、3、4、5、6、7、8若しくは9つの変異、例えば4、5若しくは9つの変異が存在し、及び/又はβ鎖CDRには、1、2、3、4、5、6、7、8、9つ若しくは10の変異、例えば4、9つ若しくは10の変異が存在する。
2つのアミノ酸配列又は2つの核酸配列のパーセント同一性は、最適比較を目的として配列を整列させ(例えば、最良整列のために第1の配列にギャップを導入することができる)、対応する位置でアミノ酸残基又はヌクレオチドを比較することにより決定する。「最良整列」は、最も高いパーセント同一性をもたらす2つの配列の整列である。パーセント同一性は、比較する配列中の同一アミノ酸残基又はヌクレオチドの数により決定する(すなわち、%同一性 = 同一位置の数/位置の総数×100)。
本発明の特定の可溶性TCRは高収率精製が可能であり得る。高収率は、1%以上、より好ましくは10%以上、又はより高い収率を意味する。
結合親和性(平衡定数KDに反比例する)及び結合半減期(T1/2と表す)は、本願実施例3の表面プラズモン共鳴(BIAcore)及び/又はOctet法を用いて決定することができる。TCRの親和性が二倍になればKDは1/2になると理解される。T1/2はln2/解離速度(koff)として算出される。したがって、T1/2が二倍になればkoffは1/2になる。TCRのKD値及びkoff値は、通常、可溶形態のTCR(すなわち、細胞質ドメイン及び膜貫通ドメインの残基を除去するように短縮された形態のもの)について測定する。好ましくは、所与のTCRの結合親和性又は結合半減期は、同じアッセイプロトコルを用いて数回、例えば3回又は4回以上測定し、その結果の平均をとる。
当業者に自明なように、提供される配列は、TCRの結合特性に実質的に影響することなく、そのC末端及び/又はN末端で1、2、3、4、5又は6以上の残基を欠失させること(短縮化)も可能である。このような特段の意味のないバリアントも全て本発明に包含される。
本発明により提供されるTCRのバリアント、フラグメント及び誘導体もまた本発明の範囲に含まれる。
別の1つの観点において、本発明は、本発明の核酸を含んでなるベクターを提供する。好ましくは、ベクターはTCR発現ベクターである。
本発明の可溶性TCRは、抗原提示細胞及び抗原提示細胞を含有する組織への検出可能な標識又は治療用薬剤の送達に有用である。したがって、これらは、(TCRを用いてGVYDGREHTV-HLA-A*02複合体を提示する細胞の存在を検出する診断目的のための)検出可能な標識;治療用薬剤;又はPK調整成分と(共有結合又はその他の様式で)結合していてもよい。
診断目的の検出可能な標識としては、例えば、蛍光標識、放射性標識、酵素、核酸プローブ及び造影剤が挙げられる。
本発明のTCRと結合され得る治療用薬剤としては、免疫調整剤、放射性化合物、酵素(例えば、パーフォリン)又は化学療法剤(例えば、シスプラチン)が挙げられる。毒性効果が確実に所望の位置で発揮されるために、毒物は、緩徐に放出されるようにTCRに連結されたリポソームの内部に存在し得る。このことにより、人体における輸送の間の損傷効果が防止され、該当する抗原提示細胞へのTCRの結合後に、毒物が最大効果を有することが保証される。
・小分子細胞傷害性物質、すなわち、哺乳動物細胞を殺傷する能力を有する分子量700ダルトン未満の化合物。このような化合物はまた、細胞傷害性効果を有することができる毒性金属を含有し得る。更に、これら小分子細胞傷害性物質にはまた、プロドラッグ、すなわち、生理学的条件下で崩壊又は変換して細胞傷害性物質を放出する化合物が含まれると理解される。このような物質の例として、シスプラチン、メイタンシン(maytansine)誘導体、ラケルマイシン(rachelmycin)、カリケアマイシン(calicheamicin)、ドセタキセル、エトポシド、ゲムシタビン、イホスファミド、イリノテカン、メルファラン、ミトキサントロン、ソルフィマーソディウムホトフィリンII(sorfimer sodiumphotofrin II)、テモゾロマイド(temozolmide)、トポテカン、トリメトレキサート(trimetreate)、ラルブーレート(larbourlate)、オーリスタチンE(auristatin E)、ビンクリスチン及びドキソルビシンが挙げられる;
・ペプチド細胞毒素、すなわち、哺乳動物細胞を殺傷する能力を有するタンパク質又はそのフラグメント。例えば、リシン、ジフテリア毒素、シュードモナス細菌外毒素A、DNAアーゼ及びRNAアーゼ;

本発明の物質の投薬量は、治療すべき疾患又は異常、治療すべき個体の年齢及び状態などに依存して広範に変化し得る。抗CD3抗体に結合した本発明の可溶性TCRに適切な用量範囲は、25ng/kg〜50μg/kgであり得る。医師が、使用すべき適切な投薬量を最終的には決定する。
本発明のTCR、医薬組成物、ベクター、核酸及び細胞は、実質的に純粋な形態、例えば、少なくとも80%、少なくとも85%、少なくとも90%、少なくとも91%、少なくとも92%、少なくとも93%、少なくとも94%、少なくとも95%、少なくとも96%、少なくとも97%、少なくとも98%、少なくとも99%又は100%純粋で提供されてもよい。

ガンは、乳ガン、食道ガン、頭部頸部ガン、肺ガン、卵巣ガン又は膀胱ガンであり得る。腫瘍はMAGE A4を発現してもよく、及び/又は充実腫瘍であってもよい。本発明のTCR、TCR-抗CD3融合分子、核酸、医薬組成物又は細胞は、注射により、例えば静脈内注射又は直接腫瘍内注射により投与されてもよい。ヒト対象はHLA-A*02サブタイプであってもよい。
本発明の各々の観点の好適な特徴は、他の観点の各々についても同様である(ただし、必要な変更は加える)。本明細書において言及した先行技術文献は、法が許容する最大範囲まで組み込まれる。
実施例
実施例1−可溶性TCRの発現、リフォールディング及び精製
方法
本発明の可溶性TCRのα及びβ細胞外領域をコードするDNA配列を、(Sambrookら,Molecular cloning. Vol. 2(1989) New York: Cold spring harbour laboratory pressに記載のような)標準的方法を用いて、別々にpGMT7ベースの発現プラスミドにクローニングした。この発現プラスミドを別々にE. coli株Rosetta(BL21pLysS)に形質転換し、単一アンピシリン抵抗性コロニーを、TYP(+アンピシリン100μg/ml)培地中で37℃にて、約0.6〜0.8のOD600まで増殖させた後、0.5mM IPTGでタンパク質発現を誘導した。誘導の3時間後に細胞を遠心分離により採集した。BugBusterタンパク質抽出試薬(Merck Millipore)を製造業者の指示に従って用いて細胞ペレットを溶解させた。封入体ペレットを遠心分離により回収した。ペレットをTriton緩衝液(50mM Tris-HCl pH8.1、0.5% Triton-X100、100mM NaCl、10mM NaEDTA)中で2回洗浄し、最後に界面活性剤フリーの緩衝液(50mM Tris-HCl pH8.1、100mM NaCl、10mM NaEDTA)中に再懸濁した。6Mグアニジン-HClで可溶化し、OD280を測定することによって、封入体タンパク質の収量を定量した。次いで、消光係数を用いてタンパク質濃度を算出した。8M尿素で可溶化し、約2μgを還元条件下の4〜20% SDS-PAGEにロードして封入体の純度を測定した。次いで、デンシトメトリーソフトウェア(Chemidoc, Biorad)を用いて純度を推定又は算出した。封入体を、短期保存については+4℃にて、長期保存については−20℃又は−70℃にて保存した。
方法
ImmTACの調製は、TCR β鎖を抗CD3単鎖抗体にリンカーを介して融合させたことを除き、実施例1に記載のとおり行った。加えて、カチオン交換工程を、アニオン交換後の精製の間に行った。この場合、アニオン交換のピーク画分を20mM MES(pH6.5)中20倍希釈し、POROS(登録商標) 50HSカチオン交換カラムに適用した。結合タンパク質を、20mM MES中0〜500mM NaClのグラジエントを用いて溶出させた。ピークImmTAC画分をプールし、50mM Tris pH8.1に調整後、濃縮し、実施例1に記載のようにゲル濾過マトリクスに直接適用した。
当該ペプチド-HLA複合体に対する精製済み可溶性TCR及びImmTAC分子の結合分析を、BIAcore 3000若しくはBIAcore T200装置を用いる表面プラズモン共鳴により又はForteBio Octet装置を用いるバイオレイヤー干渉分光法により行った。当業者に公知の方法を用いて、ビオチン化クラスI HLA-A*02分子を目的のペプチドと共にリフォールドさせて精製した(O'Callaghanら(1999),Anal Biochem 266(1):9-15;Garbocziら(1992),Proc Natl Acad Sci USA 89(8):3429-3433)。全ての測定は、0.005% P20を補充したダルベッコPBS緩衝液中で25℃にて行った。
ビオチン化ペプチド-HLAモノマーをストレプトアビジン結合CM-5センサチップに固定した。平衡結合定数は、約200応答単位(RU)のペプチド-HLA-A*02複合体を被覆したフローセル上に30μl/分の定流量で注入した可溶性TCR/ImmTACの系列希釈物を用いて決定した。平衡応答は、各TCR濃度について、無関係のペプチド-HLAを含むコントロールフローセルでのバルク緩衝液応答を減算して規格化した。KD値は、Prismソフトウェア及びLangmuir結合等温式 結合 = C×Max/(C+KD) (式中、「結合」は注入したTCRの濃度Cでの平衡結合(RU)であり、Maxは最大結合である)を用いる非線形曲線フィッティングにより得た。
ビオチン化ペプチド-HLAモノマーを、ストレプトアビジンを予め固定した(SA)ストレプトアビジンバイオセンサ(Pall ForteBio)に1nmで捕捉させた。センサを遊離ビオチン(2μM)で2分間ブロックした。平衡結合定数は、ロードされたバイオセンサを96ウェル又は384ウェルサンプルプレート中で系列希釈した可溶性TCR/ImmTACに浸漬させて決定した。プレート振盪を1000rpmに設定した。低親和性相互作用(μM範囲)については、短い結合時間(約2分間)及び短い解離時間(約2分間)を用いた。結合曲線は、Octetデータ分析ソフトウェア(Pall ForteBio)を用いて、無関係のpHLAをロードした参照バイオセンサの参照データの2倍を減算して処理した。平衡時の応答(nm)を用い、式:応答 = Rmax×conc/(KD+conc)(式中、「応答」は各TCR濃度(conc)での平衡結合(nm)であり、RmaxはpHLA飽和時の最大結合応答である)にフィットさせた定常状態プロットからKD値を推定した。
可溶性天然型TCRは、実施例1に記載の方法に従って調製し、pHLAへの結合は実施例3に従って分析した。α鎖及びβ鎖のアミノ酸配列は図2に示したものに相当した。可溶性ビオチン化HLA-A*02は、MAGE A4ペプチドGVYDGREHTVを用いて調製し、BIAcoreセンサチップに固定した。
結合は種々の濃度で決定し、相互作用のKD値は142μMであると決定した。交差反応性(特異性)は、15の無関係のペプチド-HLA-A*02複合体のパネルに対して、実施例3の平衡BIAcore法を用いて評価した。15の無関係のpHLAは3群に分けてプールし、フローセルあたり約1000RUの各pHLAが得られるように3つのフローセルの1つにロードした。20μLの可溶性野生型TCRを、全てのフローセルに濃度73μMで20μL/分で注入した。いずれの濃度でも有意な結合は検出されなかった。このことは、天然型TCRがGVYDGREHTV-HLA-A*02複合体に関して特異的であることを示す。
このデータは、このTCRが標的に適切な親和性及び特異性で結合し、したがって治療用TCRの有用な出発配列を提供することを示す。
可溶性変異TCR及びImmTAC分子は図2に示す配列に基いて作製した。サンプルは実施例1及び2に記載のように調製し、結合特性を実施例3に従って決定した。
結果
α鎖の19位での単一システイン→バリン点変異(配列番号6)は、親和性にも特異性にも影響することなくリフォールディング及び精製収量を改善することが判明した(KDは145μMと記録され、WTで試験した同じ15の代替ペプチド-HLA複合体のパネルに対する交差反応性は観察されなかった)。
少なくとも1つのCDR領域に、図2に示すCDR配列に比して変異を含むTCR α鎖及び/又はβ鎖を同定した(配列番号4及び5)。これらTCR配列は、GVYDGREHTV-HLA-A*02複合体を特に適切な親和性及び/又は半減期で認識した。幾つかの場合において、TCRの安定性及び/又は収量を改善する更なる変異(α鎖変異K1A(配列番号4の番号付けを参照して)を含む)を同定した。本発明の特定の変異TCR α鎖及びβ鎖の可変領域のアミノ酸配列をそれぞれ図4及び5に示す。下記の表は、示されたα及びβ可変領域を含む可溶性TCR又はImmTAC分子(可溶性TCR-抗CD3融合分子)の結合特性を提供する。

同族GVYDGREHTV-HLA-A*02複合体への結合に加え、本発明のTCRを、MAGE A8及びMAGE B2に由来し、HLA-A*02により提示される類似のペプチドへの結合についても評価した。下記表の数値は、示したα及びβ可変ドメイン配列を含む3つのImmTAC分子についてBiacore結合データを示す。3つ全てのImmTAC分子は、MAGE-A8ペプチドを同族ペプチドに類似するレベルで認識し、MAGE-B2ペプチドをより弱いレベルで認識する。

変異α及びβ可変鎖配列を含み、標的抗原に関して特に高い親和性を有するImmTAC分子を、T細胞活性化の指標としてインターフェロン-γ(IFN-γ)分泌を用いるELISPOTアッセイにより、CD3+T細胞の強力で特異的な再指向化を媒介する能力について試験した。
本実施例では、α鎖可変領域の配列は配列番号20〜24から選択し、β鎖可変領域の配列は配列番号26〜29から選択した。可変ドメイン配列は、それぞれのα又はβ細胞外定常ドメイン配列に融合され、非天然型ジスルフィド結合を含んでいた。各場合において、β鎖はリンカーを介して抗CD3 scFvに融合した;リンカーは配列番号30〜37から選択した。試験したImmTAC分子の完全配列は、次の表に記載する配列番号により示す:
アッセイは、ヒトIFN-γ ELISPOTキット(BD Biosciences)を用いて行った。標的細胞は、アッセイ培地(10%熱不活化FBS及び1%ペニシリン-ストレプトマイシン-L-グルタミンを含有するRPMI 1640)に1×106/mlの密度で調製し、50,000細胞/ウェルにて50μlの容量でプレートした。新鮮なドナー血から単離した末梢血単核細胞(PBMC)をエフェクター細胞として用い、10,000〜50,000細胞/ウェルにて50μlの容量でプレートした(各実験に用いた細胞の正確な数はドナーに依存し、当該アッセイに関して適切な範囲内の応答を生じるように調整され得る)。予想される臨床上妥当な範囲にわたる種々の濃度のImmTACを用い、ウェルに50μlの容量で加えた。
図7及び8(上パネル)に示すデータは、それぞれImmTAC分子1〜2及びImmTAC分子3〜5が標的抗原を発現するガン細胞(NCI-H1703−ヒト肺ガン細胞株)に対する強力な(すなわち、100pM以下のEC50で)T細胞再指向化を媒介可能であることを示す。臨床的に妥当な濃度範囲(≦1nM)で抗原陰性ガン細胞(ImmTAC分子1〜2についてはNCI-H441ヒト乳頭腺ガン細胞株;ImmTAC分子3〜5についてはCAMA-1ヒト乳ガン細胞株)に対するT細胞活性化は検出されなかった。このことは、応答が特異的であることを証明する。
正常な健常ヒト組織に由来する細胞を標的細胞として用いて、ImmTAC分子を特異性について試験した。図7の下パネルは、ImmTAC分子1〜2が臨床的に妥当な濃度で、ヒト皮膚脈管構造細胞に対して極僅かな反応性を有することを示す。同様に、図8の下パネルは、ImmTAC分子3〜5が臨床的に妥当な濃度で、ヒト皮膚脈管構造細胞及びヒト腎細胞に対して極僅かな反応性を有することを示す。
ImmTAC分子1及び4を、10より多い正常細胞タイプの拡大パネルに対する反応性について、より細かいImmTAC濃度範囲(0.01nM、0.1nM、0.2nM、0.3nM、0.5nM及び1nM)で、上記と同じELISPOT法を用いて更に試験した。図10は、骨格筋、心臓及び腎臓細胞から得られた代表的データを示す。各場合において、抗原陽性細胞(NCI-H1703)及び抗原陰性細胞(NCI-H441)をコントロールとして含ませた。データは、臨床上妥当な濃度範囲(≦1nM)で、抗原陽性細胞に比して、正常細胞に対する反応性が無視し得るものであることを証明する。
このデータは、これらImmTAC分子が高レベルに強力で特異的であり、したがって治療用途に特に適切であることを証明する。
本発明のImmTAC分子が再指向化T細胞による抗原陽性腫瘍細胞の強力な殺傷を媒介する能力を、IncuCyteプラットフォーム(Essen BioScience)を用いて調べた。このアッセイは、カスパーゼ-3/7(アポトーシスのマーカー)放出の顕微鏡観察によるリアルタイム検出を可能とする。
方法
アッセイは、CellPlayer 96ウェルカスパーゼ-3/7アポトーシスアッセイキット(Essen BioScience,Cat. No.4440)を用いて製造業者のプロトコルに従って行った。簡潔には、標的細胞(NCI-H1703−抗原+ve HLA A*02+ve;NCI-H441−抗原-ve HLA A*02+ve)を5000細胞/ウェルでプレートし、一晩インキュベートして接着させた。ImmTAC溶液を0.5nM〜0.01nMの濃度で調製し、各濃度の25μlを該当するウェルに加えた。エフェクター細胞を10:1のエフェクター:標的細胞の比で用いた(50000細胞/ウェル)。ImmTACを含まないコントロールサンプルも調製した。NucViewアッセイ試薬を30μMで作製し、25μlを全てのウェルに加え、最終容量を150μlとした(最終濃度5μMを得た)。プレートをIncuCyte装置内に置き、3日間にわたって2時間ごとに撮像した(1画像/ウェル)。各画像内のアポトーシス細胞の数を決定し、1mm2あたりのアポトーシス細胞として記録した。アッセイは3つ組で行った。
図11に示すデータは、ImmTAC再指向化T細胞による腫瘍細胞のリアルタイム殺傷を示す。ImmTAC1及びImmTAC4の結果を示す。両ImmTAC分子とも、0.01nMの濃度で、抗原陽性腫瘍細胞の再指向化T細胞による殺傷を示す。最高濃度(0.5nM)でさえ、抗原陰性細胞の殺傷は観察されない。
これらデータは、ImmTAC1及びimmTAC4が抗原陽性腫瘍細胞の再指向化T細胞による強力な殺傷を媒介することを確証するものである。

β鎖可変ドメインが配列番号25〜29又は65〜81のアミノ酸配列のいずれか1つを含む、請求項1〜12のいずれか1項に記載のTCR。 - α鎖可変ドメイン及びβ鎖可変ドメインが下記:
- α-βヘテロダイマーであり、α鎖TRAC定常ドメイン配列及びβ鎖TRBC1又はTRBC2定常ドメイン配列を有する請求項1〜14のいずれか1項に記載のTCR。
- α鎖及びβ鎖の定常ドメイン配列が、TRACのエキソン2のCys4とTRBC1又はTRBC2のエキソン2のCys2との間の天然型ジスルフィド結合を欠失させるように、短縮化又は置換により改変されている、請求項15に記載のTCR。
- α鎖及び/又はβ鎖の定常ドメイン配列がTRACのThr 48及びTRBC1又はTRBC2のSer 57からシステイン残基への置換により改変されており、前記システイン同士がTCRのα定常ドメインとβ定常ドメインとの間の非天然型ジスルフィド結合を形成している、請求項15又は16に記載のTCR。
- Vα-L-Vβ型、Vβ-L-Vα型、Vα-Cα-L-Vβ型、Vα-L-Vβ-Cβ型(ここで、Vα及びVβはそれぞれTCR α及びβ可変領域であり、Cα及びCβはそれぞれTCR α及びβ定常領域であり、Lはリンカー配列である)の単鎖形式である請求項1〜17のいずれか1項に記載のTCR。
- 検出可能な標識、治療用薬剤又はPK調整成分と組み合わされている請求項1〜18のいずれか1項に記載のTCR。
- 抗CD3抗体がTCR α鎖又はβ鎖のC末端又はN末端に共有結合的に連結されている請求項19に記載のTCR。
- 抗CD3抗体がTCR β鎖のC末端又はN末端にリンカー配列を介して共有結合的に連結されている請求項20に記載のTCR。
- リンカー配列がGGGGS(配列番号30)、GGGSG(配列番号31)、GGSGG(配列番号32)、GSGGG(配列番号33)、GSGGGP(配列番号34)、GGEPS(配列番号35)、GGEGGGP(配列番号36)及びGGEGGGSEGGGS(配列番号37)からなる群より選択される、請求項21に記載のTCR。
- α鎖可変ドメインが配列番号16〜24又は46〜64から選択されるアミノ酸配列を含み、
β鎖可変ドメインが配列番号25〜29又は65〜81から選択されるアミノ酸配列を含み、
抗CD3抗体が、TCR β鎖のN末端又はC末端に、配列番号30〜37から選択されるリンカー配列を介して共有結合的に連結されている、TCR-抗CD3融合分子。 - 配列番号38〜41から選択されるα鎖アミノ酸配列又は配列番号38〜41に記載のアミノ酸配列に対して少なくとも90%の同一性、例えば少なくとも90%、91%、92%、93%、94%、95%、96%、97%、98%、99%若しくは100%の同一性を有するα鎖アミノ酸配列と、
配列番号42〜45から選択されるβ鎖アミノ酸配列又は配列番号42〜45に記載のアミノ酸配列に対して少なくとも90%の同一性、例えば少なくとも90%、91%、92%、93%、94%、95%、96%、97%、98%、99%若しくは100%の同一性を有するβ鎖アミノ酸配列と
を含んでなる、請求項23に記載のTCR-抗CD3融合分子。 - (a)配列番号38に相当するα鎖アミノ酸配列及び配列番号42に相当するβ鎖アミノ酸配列;
(b)配列番号38に相当するα鎖アミノ酸配列及び配列番号43に相当するβ鎖アミノ酸配列;
(c)配列番号39に相当するα鎖アミノ酸配列及び配列番号44に相当するβ鎖アミノ酸配列;
(d)配列番号40に相当するα鎖アミノ酸配列及び配列番号45に相当するβ鎖アミノ酸配列;又は
(e)配列番号41に相当するα鎖アミノ酸配列及び配列番号45に相当するβ鎖アミノ酸配列
を含んでなる請求項24に記載のTCR-抗CD3融合分子。 - 請求項1〜25のいずれか1項に記載のTCR α鎖及び/又はTCR β鎖をコードする核酸。
- 請求項26に記載の核酸を含んでなる発現ベクター。
- (a)請求項1〜25のいずれか1項に記載のTCR α鎖及びβ鎖を単一のオープンリーディングフレーム若しくは2つの別個のオープンリーディングフレームにコードする請求項27に記載の発現ベクター;又は
(b)請求項1〜25のいずれか1項に記載のTCR α鎖をコードする核酸を含む第1の発現ベクターと、請求項1〜25のいずれか1項に記載のTCR β鎖をコードする核酸を含む第2の発現ベクターと
を有する細胞。 - 請求項1〜22のいずれか1項に記載のTCRを提示する天然に存在しない及び/又は単離された及び/又は操作された細胞、特にT細胞。
- 請求項1〜22のいずれか1項に記載のTCR、請求項23〜25のいずれか1項に記載のTCR-抗CD3融合分子、請求項26に記載の核酸及び/又は請求項28若しくは29に記載の細胞を、1又は2以上の医薬的に許容され得る担体又は賦形剤と共に含んでなる医薬組成物。
- 医薬に用いるため、好ましくはヒト対象における医薬に用いるための、請求項1〜22のいずれか1項に記載のTCR、請求項23〜25のいずれか1項に記載のTCR-抗CD3融合分子、請求項26に記載の核酸、請求項30に記載の医薬組成物及び/又は請求項28若しくは29に記載の細胞。
- ガン又は腫瘍の治療法に用いるため、好ましくはヒト対象におけるガン又は腫瘍の治療法に用いるための、請求項1〜22のいずれか1項に記載のTCR、請求項23〜25のいずれか1項に記載のTCR-抗CD3融合分子、請求項26に記載の核酸、請求項30に記載の医薬組成物及び/又は請求項28若しくは29に記載の細胞。
- ヒト対象がMAGE A4を発現する腫瘍を有する、請求項32に記載の使用のためのTCR、TCR-抗CD3融合分子、核酸、医薬組成物及び/又は細胞。
- 腫瘍が充実腫瘍である、請求項32又は33に記載の使用のためのTCR、TCR-抗CD3融合分子、核酸、医薬組成物及び/又は細胞。
- ヒト対象がHLA-A*02サブタイプである、請求項32〜34のいずれか1項に記載の使用のためのTCR、TCR-抗CD3融合分子、核酸、医薬組成物及び/又は細胞。
- 注射、例えば静脈内注射又は腫瘍内への直接注射により投与される、請求項32〜35のいずれか1項に記載の使用のためのTCR、TCR-抗CD3融合分子、核酸、医薬組成物及び/又は細胞。
- 医薬的に有効な投薬量の請求項30に記載の医薬組成物をヒト対象に投与することを含んでなる、ヒト対象を治療する方法。
- 抗新生物剤を別々に、組合せで又は逐次に投与することを更に含んでなる請求項37に記載の方法。
- 請求項1〜22のいずれか1項に記載のTCR又は請求項23〜25のいずれか1項に記載のTCR-抗CD3融合分子を含んでなる、ヒト対象への投与用の注射可能製剤。
- a)請求項28に記載の細胞を、TCR鎖の発現に最適な条件下に維持し、b)TCR鎖を単離することを含んでなる、請求項1〜22のいずれか1項に記載のTCR又は請求項23〜25のいずれか1項に記載のTCR-抗CD3融合分子を製造する方法。
