前記メープルリーフ型の葉の単一劣性核遺伝子が、キュウリゲノムの第2染色体の16383700−16385719塩基に位置する遺伝子であり、通常の葉型のキュウリの当該遺伝子に比べて、その859番目の塩基AがTに変異することを特徴とするメープルリーフ型キュウリ植物を提供する。
II.同系交配の後代から分離したメープルリーフ型の表現型を有する植物を選抜する。

発明者は、2012年春に北京市海淀区上庄の実験温室に植えた、農家品種唐山秋瓜、白葉三とヨーロッパ鮮食類キュウリ品種によりマルチハイブリダイズされた後代の近交系において、メープルリーフ型の変異体を発見し、CC81221と命名し、その写真を図2、図3に示す。
(2)華南型(刺の少ない種類,掌状五角形の葉)の2つの近交系、それぞれ
(a)短くて太い唐山秋瓜類近交系TA511、
(b)中程度の短さの乾燥地キュウリ(dryland cucumber)類近交系HA411である。
(3)ヨーロッパ鮮食類(棘のない種類、ハート型五角形)の2つの近交系、それぞれ
(a)緑キュウリ類近交系SL311、
(b)白キュウリ類近交系SL322である。
50ウェルのプレートに苗を移植し、F1世代、BC1世代ごとに50粒を播種し、F2世代を100粒播種した。
(1)メープルリーフ型:同系交配
(2)メープルリーフ型×通常の葉→F1→F2
F1×メープルリーフ型
(3)通常の葉×メープルリーフ型→F1→F2
F1×メープルリーフ型
(1)2012年5月に、北京市海淀区上庄の実験温室で、番号CC81221のメープルリーフ型の葉の材料を親の1つとし、通常の葉の異なるタイプのキュウリ近交系をそれぞれもう1つの親として、正交雑交配種と逆交雑交配種(メープルリーフ型を雌親として、通常の葉の近交系と交配することが正交雑であり、メープルリーフ型を雄親として、通常の葉の近交系と交配することが逆交雑である)をそれぞれ収穫し、F1世代を得た。
メープルリーフ型の葉を有するキュウリ植物CC81221又はそれから誘導されるメープルリーフ型の葉を有するキュウリ近交系、F1交配種又はヘテロ接合体を、メープルリーフ型のドナーとすることで、メープルリーフ型の葉を有するキュウリ植物を育種した。よって、本発明の方法により育種されるいずれのメープルリーフ型の葉を有するキュウリ植物、及び番号がCC81221であるメープルリーフ型キュウリ植物またはそれらから誘導されるメープルリーフ型キュウリ植物は、全て本発明の範囲に属する。
戻し交配法を本発明に用い、その主な目的は、原品種の葉形を変更または置き換えるとともに、基本的に原品種の他の全ての望ましい遺伝学的組成、および望ましい生理学的組成と形態学的組成を保留することである。
本発明以外の交配種と比べて、本発明の方法により培養した交配種は、二つのメープルリーフ型の葉を有するキュウリを親として利用して交配することを特徴とする。本試験の目的は、メープルリーフ型の葉を有するキュウリ植物のハイブリッド種子の商業生産を実現することである。具体的には、以下のことを含む。
(一)試験材料:
雄親:M111(Beijing Beinong Sanyi Cucumber Ecological Breeding Science and Technology Centerにより提供されるもの):通常の葉(優性ホモ接合)。
雌親:CC81221(受託番号:CGMCC No.11285)、メープルリーフ型(劣性ホモ接合)。
F1:通常の葉(ヘテロ接合)。
1、葉のDNAの抽出
Aidlab Biotechnologies Co.,Ltdの植物Rapid DNA Extraction Kitで葉のDNAを抽出し、キットの取扱説明書を参照して試験を実施した。
SNPサイト変異に使用されるプライマー配列は下記の通りである:
配列番号1で示される上流プライマー:GTCCAGGTTCTCAATCGAGCT、
配列番号2で示される下流プライマー:GCCACGATATTGAGAGGTTCT。

増幅された産物を配列決定に供し、その結果は図14(雄親)、図15(雌親)、図16(F1)に示すとおりであり、当該SNP分子マーカーがメープルリーフ型の葉の単一劣性核遺伝子を有するキュウリ植物を標識できることが示されている。
Claims (9)
- 1)受託番号がCGMCC No.11285のククミス属植物CC81221である、メープルリーフ型の葉の単一劣性核遺伝子を有するキュウリ変異体植物、または、
2)CC81221から誘導されるメープルリーフ型の葉の単一劣性核遺伝子を有するキュウリ植物であって、
前記メープルリーフ型の葉の単一劣性核遺伝子が、キュウリゲノムの第2染色体の16383700−16385719塩基に位置する遺伝子であり、通常の葉型のキュウリの当該遺伝子に比べて、その859番目の塩基AがTに変異する
ことを特徴とするメープルリーフ型キュウリ植物。 - 植物細胞、種子、葯、花粉、胚珠、葉、胚芽、根、根冠、花、果実、茎、枝、穂木、プロトプラストまたはカルスであることを特徴とする請求項1に記載のメープルリーフ型キュウリ植物の一部。
- 観光ガーデニングにおける請求項1に記載のメープルリーフ型キュウリ植物の応用。
- メープルリーフ型キュウリ植物の品種鑑定における請求項1に記載のメープルリーフ型の用途。
- キュウリゲノムの第2条染色体の16383700−16385719塩基に位置する遺伝子であり、通常の葉型のキュウリの当該遺伝子に比べて、その859番目の塩基AがTに変異することを特徴とするメープルリーフ型の葉の単一劣性核遺伝子。
- 請求項5に記載のメープルリーフ型の葉の単一劣性核遺伝子を含むベクター及び前記ベクターの宿主。
- CC81221、またはそれから誘導されるメープルリーフ型の葉の単一劣性核遺伝子を有するキュウリ植物をメープルリーフ型の遺伝源として、メープルリーフ型の葉を有するキュウリ品種を育種することを特徴とするメープルリーフ型キュウリ植物品種の育種方法。
- 特異的なプライマー対で同定待ちの植物をPCR増幅させ配列決定を行い、配列決定の結果がTTホモ接合のピーク、またはATヘテロ接合のピークを有するものが、メープルリーフ型の葉の単一劣性核遺伝子を有するキュウリ植物であり、
前記特異的なプライマー対の配列が配列番号1で示される上流プライマー:GTCCAGGTTCTCAATCGAGCT、配列番号2で示される下流プライマー: GCCACGATATTGAGAGGTTCTである
ことを特徴とするメープルリーフ型の葉の単一劣性核遺伝子を有するキュウリ植物又はその一部の分子同定方法。 - 配列番号1で示される上流プライマー:GTCCAGGTTCTCAATCGAGCT、
配列番号2で示される下流プライマー:GCCACGATATTGAGAGGTTCT
であることを特徴とするメープルリーフ型の葉の単一劣性核遺伝子を有するキュウリ植物またはその一部を同定するためのプライマー対。
