Description
実施例1:N−{2−{[2−(ジメチルアミノ)エチル](メチル)アミノ}−6−(2,2,2−トリフルオロエトキシ)−5−{[4−(1−メチル−1H−インドール−3−イル)ピリミジン−2−イル]アミノ}ピリジン−3−イル}アクリルアミド
中間体1c:N2−メチル−N2−[2−(ジメチルアミノ)エチル]−6−(2,2,2−トリフルオロエトキシ)−3−ニトロピリジン−2,5−ジアミンの製造方法については、中国特許出願第CN201410365911.4号の実施例を引用する。
測定にかかる実施例1:SDラット(Sprague Dawleyラット)による薬物吸収実験
静脈投与:上海西普爾−必凱実験動物有限公司(Shanghai Sippr−BK laboratory animal Co.Ltd.)から提供された健康なSDラット(オス・メス各16匹、体重200〜280g)をランダムに4群に分けた。下表に列記した投与量で上記の実施例1、実施例2、比較例1及び比較例2の物質を静脈投与し、投与前と、投与から5min後、15min後、0.5h後、1.0h後、2.0h後、4.0h後、8.0h後、12h後及び24h後にラットの眼球後静脈叢から静脈血0.2mlを採取した。これを分離して血漿を作製し、液体クロマトグラフィー/タンデム質量分析法により血漿中の薬物濃度を測定することで、薬物濃度−時間曲線を取得した。
胃内投与:上海西普爾−必凱実験動物有限公司から提供された健康なSDラット(オス・メス各16匹、体重200〜280g)をランダムに4群に分けた。下表に列記した投与量で上記の実施例1、実施例2、比較例1及び比較例2の物質を胃内投与し、投与前と、投与から0.5h後、1.0h後、2.0h後、4.0h後、6.0h後、8.0h後、10h後、12h後及び24h後にラットの眼球後静脈叢から静脈血0.2mlを採取した。これを分離して血漿を作製し、液体クロマトグラフィー/タンデム質量分析法により血漿中の薬物濃度を測定することで、薬物濃度−時間曲線を取得した。
本発明における実施例2で作製して得た式(I)化合物のメシル酸塩と、比較例2の物質(比較例2の構成は下記の通りであり、製造方法については中国特許出願第CN201410365911.4号の実施例16を参照した)によるヒト肺癌H1975ヌードマウス皮下移植瘤に対する阻害作用を観察した。
Claims (6)
- 式(I)化合物のメシル酸塩。
- 請求項1に記載する式(I)化合物のメシル酸塩の製造方法であって、
溶剤中で式(I)化合物をメシル酸に直接反応させて式(I)化合物のメシル酸塩を得ることを特徴とする製造方法。 - 前記溶剤はアセトンと水の混合溶剤であることを特徴とする請求項2に記載の製造方法。
- 請求項1に記載する式(I)化合物のメシル酸塩と、薬学的に許容可能な担体とを含むことを特徴とする薬物組成物。
- 請求項1に記載する式(I)化合物のメシル酸塩の、癌治療の薬物の製造における使用。
- 請求項4に記載する薬物組成物の、癌治療の薬物の製造における使用。
