Abstract
Description
α放射体は以前に、腹腔内癌の処置として提案された。2タイプの化学薬品クラス:(1)放射性免疫抱合体及び(2)マイクロ又はナノサイズの特定の懸濁液、が提案された。放射性免疫抱合体の利点は、細胞特異的ターゲッティングの可能性であり、不利益な点は、潜在的な全身的毒性を引き起こす血流内への生成物の大量の漏出である。
よって、腔内癌に対するα粒子放射のための改善された送達系の必要性が存在する。
本発明の一実施形態において、放射性核種は、224Ra、212Bi、212Pb、223Ra、225Ra、225Ac、213Bi、211At、227Thから成る群から選択されるである。
本発明の別の実施形態において、分解性化合物は、CaCO3、PEG修飾CaCO3、タンパク質修飾CaCO3、炭水化物修飾CaCO3、脂質修飾CaCO3、ビタミン修飾CaCO3、有機化合物修飾CaCO3、ポリマー修飾CaCO3、及び/又は無機結晶修飾CaCO3から成る群から選択される。

本発明者らは、狭い範囲のα放射体に基づいて腸毒性のリスクがより低い癌治療を明らかにした。現在の発明は、α放射性核種、及び/又はα放射性娘核種、例えば224Ra、を作り出す放射性核種を含む遅分解性(slowly degradable)ナノ又はマイクロ粒子に基づく。
よって、本発明の1つの目的は、分解性化合物と、α放射性核種、及び/又はα放射性娘核種を作り出す放射性核種とを含む粒子に関する。
本発明の別の態様は、CaCO3、α放射性核種224Ra、並びに220Rn、216Po、及び212Pbから成る群から選択される224Ra放射性子孫核種を含む粒子に関する。220Rnは224Raの娘核種であり、216Poは224Raの放射性孫娘核種であり、及び212Pbは224Raの放射性曾孫娘核種である。
本発明の放射性核種は、任意のα放射性核種、及び/又はα放射性娘核種を作り出す放射性核種であってよい。
例えば腹腔内腔における、局所療法におけるα粒子放射性化合物の主な利点は、90Y、131I、及び32Pのような医療用β放射体からのβ粒子のmm〜cmの範囲と比べて、αの、一般的に0.1mm未満の、より狭い範囲である。
表1は、224Raの主な放射線特性を示す。224Ra及び娘核種の完全な崩壊は、合計4個のα粒子を作り出す。重要な態様は、この核種としての220Rnは、それが潜在的に結晶へと接着するのに化学的に不活性であるので、潜在的に母核種から拡散する場合がある結末である。
よって、一実施形態において、α放射性核種224Raは、娘核種220Rn、放射性孫娘核種216Po、及び放射性曾孫娘核種212Pbを伴う。本発明の粒子に関して、224Raがα放射性核種であるとき、これらはすべて粒子に含まれる。

よって、一実施形態において、「医療用デバイス」という用語は、先のFDA定義のように理解される。
1分岐による224Ra変換あたりの平均。1%を上回る実効存在度が占めたX線又はγのみ。224Ra及び娘核種の完全な崩壊あたり約26.5MeVのαの、0.7MeVのβの総実効エネルギーを意味する。
よって、本発明の一実施形態において、放射性核種は、224Ra、212Bi、212Pb、223Ra、225Ra、225Ac、213Bi、211At、227Thから成る群から選択される。
投与量は、癌型、例えば、疾患がどれくらい攻撃的であるかに依存する。一実施形態において、投与量は、10〜100kBq/kg、例えば20〜50kBq/kgなどである。別の実施形態において、投与量は、10〜1000kBq/kg、例えば25〜300kBq/kgなどである。更なる実施形態において、投与量は、100〜500kBq/kg、例えば150〜300kBq/kgなどである。
例えば、100人の患者分の投与量が1日あたり1バッチで製造される場合、これは、100個の単一の投与バイアル又はすぐに使用できるシリンジに分割される、合計1〜10GBqに調整され得る。本発明の別の実施形態において、医薬組成物は、複数の投与量の工業規模製造に好適な、例えば50MBq〜100GBqの量の放射性核種を用いて調製される。
本発明の分解性化合物は、分解され得る任意の化合物であり得る。
分解は、高pH、低pH、プロテアーゼ、酵素、ヌクレアーゼから成る群から選択される任意の経路によって、及び/又は(食作用も含めた)エンドサイトーシスのような細胞プロセスによっておこなわれ得る。

炭酸カルシウム(CC)粒子は、他の塩、タンパク質又はペプチドとの複合物として使用されてもよく、そして、オレイン酸塩及び類似物のような界面活性剤により表面修飾を受けてもよい。
特別な実施形態において、CC粒子は、機能的受容体及び/又はモノクローナル抗体や誘導体を含めた抗原結合基、並びに粒子の受容体又は抗原結合基が個々の標的細胞及び疾患組織に結合することを可能にする、ビタミン及び誘導体で修飾される。これは、粒子の修飾がCCへの化合物の付加と関連することを意味する。これは、様々な方法でおこなうことができ、そして、例えば双極子相互作用、イオン双極子及びイオン誘起双極子力、水素結合形成、ファンデルワールス力、及び力の相対強度などの相互作用による。
粒子は、さまざまな特徴を有し得る。粒子のサイズは、用途及び適用によって異なる可能性がある。結晶タイプは、任意の既知のCC形態であり得、且つ、1nm〜500μmの異なるサイズが、使用され得る。より好ましくは、サイズは、100nm〜50μmの範囲内にあり、更に好ましくは、サイズは、1〜10μmの範囲内にある。
好ましい一実施形態において、サイズは、1〜10μmである。
本発明の一実施形態において、粒子のサイズは、1nm〜500μmである。
放射性核種の添加前の粒子中の炭酸カルシウムの総量は、例えば共沈物が添加されるか否かによって変動する可能性がある。ある実施形態において、炭酸カルシウムの量は、90%超である。範囲は、90〜95%、又は90〜99%であり得る。量はまた、98%超又は99%超でもあり得る。
分解性粒子は、多くの異なる追加化合物を含み得る。これらは、ターゲッティング、安定性、溶解性及び分解速度を含めた様々な目的に役立ち得る。
本発明の一実施形態において、粒子は、モノクローナル抗体、ポリクローナル抗体、放射性免疫抱合体、免疫抱合体、キレート−抗体抱合体、葉酸や葉酸誘導体を含めたビタミン、ペプチド、ミニボディ、及びアフィボディから成る群から選択される1若しくは複数の化合物を含む。
これらは、環状キレート剤であっても、又は直鎖キレート剤であっても、又は分岐キレート剤であってもよい。
骨格窒素に取り付けられた酸性(例えば、カルボキシアルキル)基を有する直鎖又は環状又は分岐ポリアザアルカン骨格を含むポリアミノポリアシッドキレート剤に対して特に言及し得る。
放射性標識手順は、一般に、放射性標識が起こる前にキレート剤が抗体に結合されるのであれば、使用される時間などに関してより簡便になる。抗体に取り付けられるキレート剤を使用した放射性標識抱合体を調製する原理は、例えば、Liu, 2008に大まかに記載されている。
態様は、本発明による粒子を含む組成物に関する。組成物は、224Ra及び/又は放射性子孫核種で標識された単分散又は多分散粒子を含む粒子懸濁液であり得る。
組成物は、水性組成物であることが好ましい。
本発明の更なる態様において、本発明は、本発明による1若しくは複数の粒子、及び希釈剤、担体、界面活性剤、解膠剤、及び/又は賦形剤を含む組成物又は医薬組成物に関する。
よって、本発明の別の実施形態において、医薬組成物は、α放射性核種及び/又はα放射性娘核種を作り出す放射性核種で標識された単分散又は多分散粒子を含む粒子懸濁液である。
本発明の別の実施形態において、医薬組成物は、静脈内、腫瘍内又は腔内注射に好適である。
i.p.癌に対するα放射性マイクロ粒子の使用は、以前にも示唆された。Archerら(US4970062A)は、212Pbに重きをおいて、α放射体の担体として水酸化第二鉄コロイドを使用することを示唆したが、224Raを含めた他のいくつかの潜在的に有用なα放射体が列挙された。Bloomerら(1981)は、211At標識テルルコロイドを使用することを示唆したが、その一方、Vergoteら(1992)は、211At標識単分散ポリマー粒子を使用することを示唆した。Larsen及びSalberg(US8142758B2)は、223Ra又は224Raを含めた他のα放射体で標識したヒドロキシアパタイト粒子を使用することを示唆した。Archerらのケースに存在する問題は、アルカリ土類、特にラジウムの水酸化物が水への比較的高い溶解性を有するので、ラジウム標識粒子を調整するために良好ではない可能性がある(Kirby et al., 1964)。
本発明の粒子及び組成物は、放射線療法用化合物、及び/又は放射線療法用混合物として使用されてもよい。本発明の粒子の医学的利用としては、(1)腔内療法(2)放射線塞栓療法(3)放射線滑膜切除術におけるヒト又は獣医学への使用が挙げられる。腔内療法としては、例えば腹腔内癌、頭蓋内癌、胸膜癌、膀胱癌、噴門癌、クモ膜下腔の癌の処置を挙げることもできる。粒子が使用され得る腔の例は、腹膜若しくは髄膜上に散った癌及び任意のこれらの腔内の臓器を含めた、頭蓋内腔、胸腔、肺腔、脊椎腔、骨盤腔、心膜、胸膜腔、膀胱腔、又はこれらの組み合わせである。
本発明の一実施形態において、感染症は、細菌感染症及びウイルス感染症から成る群から選択される。
放射線塞栓療法は、臓器、例えば肝臓、の原発性癌又は転移癌の処置を含んでもよく、肝臓又は腫瘍組織によって浸潤された別の固形臓器内の腫瘍に通じる血管に本発明の粒子を投与することによる。
今日、それは、炎症性疾患又はリウマチ様疾患、或いは様々な関節、特に膝、手及び足首の滑膜関節症に使用されるβ粒子放射性化合物に基づいている。分解性である、本明細書中に記載した224Ra−CC粒子は、放射線滑膜切除術において非常に有用である可能性がある。
粒子は、例えば腔内の、局所注射によって投与されるのが好ましい。
物品は、例えば、医療用注射に適合する様々なバッファー、例えば溶解させた塩及び/又はタンパク質及び/又は脂質及び/又は糖、の中に分散させてもよい。

本発明の一実施形態において、癌は、腹腔内癌、頭蓋内癌、胸膜癌、膀胱癌、噴門癌、及びクモ膜下腔の癌から成る群から選択される。
本発明の一実施形態において、癌は、転移癌、肺癌、卵巣癌、結腸直腸癌、胃癌、膵臓癌、乳癌、新生物性髄膜炎、腹膜癌、胸膜浸出液、悪性中皮腫、乳癌、肉腫、神経膠芽腫や星状細胞腫のような脳癌、膀胱癌、及び肝臓癌から成る群から選択される。

本発明の別の態様は、本発明の粒子を調製する方法に関し、該方法は、放射性核種の担体を使用の有無にかかわらず、α放射性核種と生分解性化合物とを互いに接触した状態にさせることを含む。
α放射体を含む溶液又は組成物、すなわち、混合物の状態で子孫212Pbを伴った224Ra溶液又は組成物は、212Pb抗原特異的処置のための放射性免疫抱合体、及び全面的な腔処置のためのα放射体、例えば224Ra−粒子を含む二成分処置システムを作り出すために、粒子を標識する前に、212Pbを錯化するためにキレート−抗体抱合体を用いて前処置される場合がある。
これを使用する望ましい方法は、キレート抱合抗体が入ったバイアルA及びα放射体、例えば娘核種と平衡状態の224Ra、が入ったバイアルB、並びにマイクロ粒子が入ったバイアルCを含むキットによるものであって、それによって、Aの内容物をバイアルBに加えるか又はその逆、そして、その混合物をバイアルCに移す前に数分間〜数時間インキュベートし、シリンジに移す前に数分間〜数時間更にインキュベートし、そして、患者に注射した。

現行の手法及び生成物は、放射性核種に数日の半減期があるので、集約された製造及びエンドユーザーへの出荷を可能にしている。本発明の別の態様は、カルシウムと炭酸塩に徐々に溶解する生分解性粒子の使用であり、これにより、既に体内に大量に存在している生成物を少量取り出す。それはまた、以下の特徴でも注目に値する:α放射体、例えば224Raが炭酸カルシウム粒子の表面に吸収されるとき、より長寿命のβ放射体212Pb(t1/2=10.6時間)に崩壊する前に、超短寿命の216Po(t1/2=0.16秒間)と一緒の短寿命の220Rn(t1/2=56秒間)の顕著な放出が、2つのα粒子を生じる。鉛は、炭酸カルシウムと共に非常に高い沈殿性を有し、そのため、i.p.体液中の212Pbは、体循環内への212Pbの漏出を減少させる粒子への再結合の傾向がある。
例えば、かなりの程度の腹腔癌が、腹部の腸の上を覆う組織の大きな脂肪パッドである網内で増殖する傾向があるので(Gerber et al., 2006)、マイクロ粒子から放出される場合、220Rnが高度に親油性であることは、有益なことであり得る。
二官能性懸濁液は、例えば、以下の方法、pH5〜6のバッファー中、224Ra溶液を、1mg/mlのTCMC標識抗体に加え、2分から数時間インキュベートし、その後、その溶液を炭酸カルシウム(CC)粒子が入ったバイアルに加え、そして、数分から数時間インキュベートする、によって作製され得る。前記混合物は、212Pb標識生成物の比放射能の低減を避けるために、できるだけ早く投与されなければならない。これは、
224Ra−CC粒子を伴った混合物に極めて強力なターゲッティング抱合体を与えるために212Pbを加えることもまた、可能であり得る。通常、斯かるシステムにおける224Raと212Pbの間の比は、1:1に近くなり得るが、いくつかの処置状況において、それは、212Pb−抱合体対224Ra粒子の量を10:1又はそれ以上の程度まで高めることが有益にもなり得る。最後の場合では、ターゲッティング抱合体の調製前に追加的な212Pbを加えるか、又は処置混合物の投与前に一部の224Ra−CC粒子を取り出すことが必要とされる。
特別な実施形態において、224Raは、カルシウムと共に共結晶化されて、炭酸塩結晶を形成させ、それによって、224Raは、娘核種の逃避を避けるために、結晶の内側に存在し、表面には存在しない。
しかしながら、いくつかの設定では、放射性核種の部分的な持続放出は、これが、例えば、腹膜の表面、におけるより良好な用量の均一性、及び結晶粒の局所的な集合体による放射線「ホットスポット」の減少に作用し得るので、有益になり得る。
本発明による化合物及び粒子に関連して先に考察したあらゆる特徴及び/又は態様が、本明細書中に記載した方法及び適用に対して例えを用いて適用されることは、理解されるべきである。

溶媒の留去などを含め、濃縮放射性調製物を用いたすべての作業を、グローブボックス内でおこなった。1MのHNO3中の228Thの線源を、市販の供給業者から入手した。Ac−樹脂を、あらかじめパックされたカートリッジの形態でEichrom Technologies LLC(Lisle, IL, USA)から入手した。
放射性サンプルを、Cobra II Autogammaカウンター(Packard Instruments, Downer Grove, IL, USA)又はHidex Automatic Gamma Counterでカウントした(Hidex, Turku, Finland)。228Th線源からの224Raの抽出中、CRC−25R線量キャリブレーター(Capintec Inc., Ramsey, NJ, USA)を使用した。
炭酸カルシウムマイクロ粒子を、自然沈降法によって調製した。0.33MのNa2CO3(Merck, Germany)溶液を、同じ体積の0.33MのCaCl2(Merck, Germany)中に素早く注ぎ入れた。30秒間の激しいボルテックス処理後に、粒子懸濁液を5分間静置した。粒子を、濾紙によって濾別し、約30mlの水で洗浄し、室温にて一晩乾燥させた。濾過及び洗浄を、0.45μmのニトロセルロースフィルター(Whatman)を備えたガラス真空濾過装置(Whatman)でおこなった。乾燥したマイクロ粒子を室温にて保存した。得られたマイクロ粒子は、Countess(商標)Automated Cell Counter(Invitrogen)による分析によって支持される、顕微鏡によって示した1〜10μm以内の直径、及び3〜5μmの中央値を有する形状の球であった。
所望の量のCaCO3粒子を、エッペンドルフチューブに移し、1mlの水中に懸濁した。粒子懸濁液を、超音波浴中で10〜15分間、超音波処理し、続いて4つ洗浄ステップ:1mlの水で最初の2回、その後、1mlの0.1M Na2SO4(Alfa Aesar, Germany)で2回、を実施した。粒子を、遠心分離によって洗浄液と分離した。洗浄後、粒子を、0.5%のウシ血清アルブミン(15mgの粒子あたり0.1ml)を補ったDPBS(Gibco, Life Technologies, Carlsbad, CA, USA)中に懸濁し、HulaMixer(Invitrogen, Life Technologies, Carlsbad, CA, USA)により室温にて30分間インキュベートした。混合プログラムは以下の通りであった:回転の軌道幅は14rpmであり、反転幅は20°であり、及び振動幅は3°であった。1mgの粒子あたり3μgのSO4(0.3%)に相当する0.1M Na2SO4溶液の体積を、粒子懸濁液に加えた。更に、224Ra溶液を、粒子懸濁液が入ったチューブに移し、直後に、1mgの粒子あたり3μgのBa(0.3%)に相当する0.07M BaCl2・2H2O(Merck, Germany)溶液を加えた。異なる溶液を添加する間には、粒子懸濁液を、ボルテックスミキサーにより十分に混合した。添加されるべき放射性溶液及び/又はBaCl2・2H2O溶液の体積が10μlを超える場合、それを段階的に(一度に5〜10μl、間には十分にボルテックス処理しながら)加えた。総放射性標識体積は、15mgの粒子あたり0.1mlの溶液と等しかった、すなわち、SO4溶液を加える前に取り出される上清の体積を、添加される他の溶液の体積に応じて調整した。放射性標識溶液中の粒子を、先に記載したのと同じ混合プログラムを用いて、HulaMixerにより最短1時間30分間、室温にてインキュベートした。最後に、粒子を、スクロースバッファーで1〜3回洗浄した。スクロースバッファーは、94mg/mlのスクロース(Sigma Ultra, St. Louis, MO, USA)と2.1mg/mlのNa2SO4を含んでいた。標識効率を、HPGe検出器を用いて粒子と(単数若しくは複数の)洗浄液を計測することによって決定した。
実施例4に記載のとおり調製した放射性標識マイクロ粒子のインビトロにおける安定性を、2つの異なる溶液中で試験した。粒子を、室温にて1〜1.4mlのスクロースバッファー中、又は37℃にて0.5mlのウシ胎仔血清中のいずれかでインキュベートした。様々な時点で、懸濁液を遠心分離し、上清及びペレット化した粒子中の放射能を計測した。その後、安定性研究が後の時点まで続けられる場合には、粒子ペレットを、スクロースバッファー又はウシ胎仔血清のいずれかの新しいアリコート中に再懸濁し、更にインキュベートした。
CaCO3マイクロ粒子を、放射性溶液を加えないことを除いて、放射性標識手順について記載したように調製した。代わりに、放射性標識に関して同じ条件下で調製した「低線量」マイクロ粒子上に吸収された212Pbの量を計測するために、粒子を、450μlのウシ胎仔血清及び50μlの224Ra溶液の(あらかじめ37℃に加熱した)混合物中でインキュベートした。粒子懸濁液を、800rpmの回転速度で37℃にてインキュベートした。10分後に、粒子懸濁液を、スピンダウンし、250μlの上清をエッペンドルフチューブに移し、そして、放射能を計測した。その後、粒子を、上清中に再懸濁し、そして、試験を1時間及び24時間後の測定に対応して延長した。表3には、試験の結果を示す。
背景:腔内使用に関して、224Ra標識炭酸カルシウム粒子の有用性を評価するために、粒子懸濁液を、マウスの腹腔内に注射し、その後の224Raの生体分布を計測した。
それらに、約5mgのマイクロ粒子に結合させた11〜18kBqの224Raを含有する0.4mlの粒子懸濁液を腹腔内注射によって投与した。マウスを屠殺し、そして、注射後20時間(n=2)、4日(n=3)、及び7日(n=3)の放射能測定のために、様々な組織を採取した。対照として、約12kBqの224Raを伴った0.25mlの0.9% NaCl溶液を各マウスへと腹腔内に投与することによって、遊離224Ra(溶解したRaCl2)を用いた生体分布実験を実施した。224RaCl2溶液は、5.5のpHを有した。比較のために、放射性標識マイクロ粒子を用いた生体分布試験に関して、3匹のマウスから成る群を、注射後の同時点にて屠殺した(図1A)。
背景:224Ra標識炭酸カルシウムマイクロ粒子の処置活性を試験するために、腹腔内微小転移のヌードマウス腫瘍モデルを使用した。
結論として、224Ra標識炭酸カルシウムマイクロ粒子は、腹腔内腫瘍に罹患しているマウスにおいて強力且つ一貫した抗腫瘍活性を示した。
背景:ヒト卵巣癌は、腹腔内腹水につながることが多い。ヒト卵巣癌細胞株ES−2は、ヌードマウスにおいて進行性腫瘍細胞増殖及び癌性腹水を生じる。
いくつかの態様において、224Ra標識炭酸カルシウム粒子を細胞特異的放射性医薬品と組み合わせることが有益になり得る。これは、炭酸カルシウム粒子と接触させる前に、娘核種と平衡状態にある224Ra溶液を、212Pb結合キレート抱合体と組み合わせたときに得られる。
212Pb標識抗体及び224Ra−CaCO3マイクロ粒子から成る放射線療法用混合物を調製した。212Pbで抗体を標識するために、抗体セツキシマブを、最初に、キレート剤、TCMCに結合させた。
水溶液(例えば、0.5Mの酢酸アンモニウム、pH5〜6)中の224Raの溶液が入ったバイアル(A)を、212Pbを作り出すために1日以上崩壊させる。TCMC−抗体抱合体若しくは同様のキレート抱合抗体の水溶液(B)と、乾燥又は水を含む炭酸カルシウムマイクロ粒子が入ったバイアル(C)。バイアルAとBの内容物を、該バイアルの一方の中で一緒に混合し、1分〜4時間インキュベートし、その後、バイアルCと混合し、1分〜4時間インキュベートした。それぞれのインキュベーションステップ後に、品質管理をおこなっても又はおこなわなくてもよい。最後に、A、B及びCを組み合わせた混合物をシリンジに流し込み、そして、患者に投与する。
224Ra及びその娘核種212Pbの標識効率と経時的な保持を、1〜10μmの好ましいサイズ範囲内の2つの異なるサイズの放射性標識CaCO3マイクロ粒子に対して調査した。粒子の数加重メジアン径が、単一粒子の光検知によって1.1及び8.9μmであることがわかり、こうして、粒子の好ましいサイズ範囲の両極を表した。最も大きい粒子を、実施例3に記載のとおり調製し、そして、より小さい粒子を、PlasmaChem GmbH, Berlin, Germanyから購入した。その粒子を、超音波処置ステップ及びDPBS中の0.5% BSAの中でのインキュベーションを省略したことを除いて、実施例4に記載のとおり224Raを用いて標識した。標識手順が終了した後に、粒子及び洗浄溶液を、Hidexガンマカウンターで1分間計測した。224Ra崩壊が非常に小さいガンマ線放出をもたらすので、224Raの放射能の測定を、その崩壊生成物212Pbのガンマ線放出の計測によって主に実施する。224Raのγ放射能、並びに娘核種212Pbのγ及びエックス線放射能を含む、65〜345keVからのカウントを選択し、更なる計算のためにまとめた。224Raと212Pbの間の平衡状態を確立するために、サンプルを、それらを再計測する前に、標識の後に最低でも24時間崩壊させた。標識効率を、結合放射能として測定した:マイクロ粒子に加えた全放射能に対する、標識手順後に粒子に結合している放射能のパーセンテージ。
保持実験(図5)からの結果は、224Raと娘核種212Pbが、スクロースバッファー中、インビトロにおいて数日間、CaCO3粒子上で十分に保持されたことを示し、そしてそれは、放射性標識粒子のための関連医薬担体になり得る。平均した保持放射能は、両方の粒度に関して、全時点において95%超である。これは、生成物が、集約した製造及び遠方のエンドユーザーへの出荷を可能にする数日の有効期間を有し得ることを示唆している。
背景:ヒト卵巣癌は、腹腔内腹水につながることが多い。ヒト卵巣癌細胞株ES−2は、ヌードマウスにおいて進行性腫瘍細胞増殖及び癌性腹水を生じる。
背景:悪性腹水は、腹腔内の進行癌において一般的であり、患者の平均寿命の短縮とクオリティオブライフに関係しているので、改善された処置戦略の医療的必要性が存在する。224Ra−CaCO3マイクロ粒子の抗腹水活性を、細胞接種後の様々な日数に対する処置を施すことによって、ヌードマウスのES−2腹水モデルにおいて評価した。
背景:最大腫瘍縮小手術を用いて処置した、卵巣癌に罹患している患者は、腹腔内腫瘍細胞及び微小転移が残存するため再発する傾向がある。患者の残存腹腔内疾患の状態を模倣するために、1・105細胞の腹水を生じる細胞株ES−2をヌードマウスに接種し、そして、細胞の1時間後に224Ra標識CaCO3マイクロ粒子を用いた処置をおこなった。
24時間の間に非放射性粒子に吸収された224Ra標識CaCO3マイクロ粒子から放出された212Pbの量を測定するために、透析手順を使用した。CaCO3マイクロ粒子を、超音波処理ステップとDPBS中の0.5% BSA中でのインキュベーションを省略したことを除いて、実施例4に記載のとおり224Raを用いて標識した。加えて、非放射性粒子を、放射性溶液を加えるステップを除いて、全く同じ方法で調製した。非放射性粒子(50mg)を、コニカル15ml遠心分離チューブ内で14mlの全体積までDPBS(pH7)で希釈した。DPBS中の約5mgの放射性粒子(0.15〜0.40ml)を、透析デバイス(Slide-A-Lyzer MINI Dialysis Devices, 20 kDa MWCO, ThermoFisher)に加え、そして、透析バッファーとして機能する非放射性粒子を含むチューブ内に配置した。挿入された透析デバイスを伴ったチューブを、振盪しながら室温にて24時間インキュベートした。インキュベーション後に、15mlチューブをスピンダウンし、そして、上清を取り出した。透析デバイス内の放射能(AD)、透析バッファーの上清(AS)、及び15mlチューブからのペレット化した粒子(AP)を、Hidexガンマカウンターで計測した。24時間の間に放出された212Pbのパーセンテージを、次のようにして概算した:
この実施例では、実施例7からのデータを、様々な組織の1グラムあたりのBqとして再び提示する。注射した用量を、1マウスあたり10kBqに標準化した。マウスを屠殺した約2.5〜3時間後のサンプルの測定値を、212Pbの量を概算するのに使用したが、それに対し、マウスの屠殺の3〜4日後のサンプルの再測定値を、224Raの量を決定するのに使用した。224Raに関するデータを、屠殺の時間でのサンプル中の放射能を示すように崩壊補正した。
この実施例では、実施例9からのデータを、マウスの追跡期間を延長し、そして、Capintec CRC-25R投与量キャリブレーターの再較正後に用量レベルを調整した。マウスに与える用量を、35kBq/kg、100kBq/kg、及び165kBq/kgに調整した。

Claims (19)
- 以下の;

を含む粒子。 - 前記CaCO3が、CaCO3、PEG修飾CaCO3、タンパク質修飾CaCO3、炭水化物修飾CaCO3、脂質修飾CaCO3、ビタミン修飾CaCO3、有機化合物修飾CaCO3、ポリマー修飾CaCO3、及び/又は無機結晶修飾CaCO3から成る群から選択される、請求項1に記載の粒子。
- 前記粒子のサイズが、1nm〜500μmである、請求項1〜2のいずれか1項に記載の粒子。
- 前記粒子が、モノクローナル抗体、ポリクローナル抗体、放射性免疫抱合体、免疫抱合体、キレート−抗体抱合体、葉酸及び葉酸誘導体を含めたビタミン、ペプチド、ミニボディ、並びにアフィボディから成る群から選択される1若しくは複数の化合物を更に含む、請求項1〜3のいずれか1項に記載の粒子。
