Abstract
Description
Claims (30)
- 生物学的混合物を処理しかつ前記生物学的混合物の構成成分を選択的に濃縮するための遠心分離機であって、前記構成成分は、異なる比重を有し、および前記遠心分離機によって生じた遠心力場において層状にでき、前記遠心分離機は:
a. 処理アセンブリであって:
i. 前記生物学的混合物を受け入れるように配置された内側チャンバーであって、および前記内側チャンバーは、前記内側チャンバーが周りで回転するように配置された中心縦軸を有し、前記内側チャンバーは、側壁とベースとを有するテーパ付きチューブ状部材を含み、前記テーパ付きチューブ状部材は、そこを通って半径方向に延在しかつ前記中心縦軸に沿って第1の位置に配置された少なくとも1つの押出孔を有する、内側チャンバー;
ii. 前記内側チャンバーから生物学的混合物を受け入れるように配置された外側チャンバーであって、前記外側チャンバーは、前記内側チャンバーの前記中心縦軸と同軸上におよび前記内側チャンバーの周りに配置され、前記外側チャンバーは、側壁および皿状部を有し、かつ前記中心縦軸の周りで回転するように配置されている、外側チャンバー;
iii. 前記内側チャンバーの一部分と前記外側チャンバーとの間に位置決めされた少なくとも1つの切断要素であって、前記内側および外側チャンバーの前記回転に対して静止したままであるように配置されている、少なくとも1つの切断要素
を含む、処理アセンブリ;および
b. 前記中心縦軸の周りで前記内側および外側チャンバーを回転させる回転源
を含む、遠心分離機。 - 前記少なくとも1つの切断要素が、前記内側チャンバーに隣接しかつ前記内側チャンバーと前記外側チャンバーとの間に配置された静止要素を通って半径方向に延在する少なくとも1つの出口孔を含む、請求項1に記載の遠心分離機。
- 前記少なくとも1つの切断要素が、前記内側チャンバーの一部分の周りに配置されたスリーブに出口開口部を含み、前記少なくとも1つの押出孔は、前記内側チャンバーの前記回転によって前記出口開口部と半径方向に位置合わせされるように配置されており、および前記中心縦軸の周りでの前記内側および外側チャンバーの前記回転は、遠心力場を生じて、前記内側チャンバー内の前記生物学的混合物が前記内側チャンバーから前記外側チャンバーへ自動的に排出されるようにし、前記自動的な排出は、前記押出孔が前記スリーブの前記少なくとも1つの出口孔と半径方向に位置が合わせられると、前記内側チャンバーの前記少なくとも1つの押出孔を通して行われて、少量ずつに分けられた生物学的混合物を生じ、かつ前記少量ずつに分けられた生物学的混合物を前記外側チャンバーに導入し、するとすぐに、前記外側チャンバーの前記回転によって、前記少量ずつに分けられた生物学的混合物を前記外側チャンバー内で、少なくとも2つの同心の層状の構成成分層へと層状にする、請求項1に記載の遠心分離機。
- 前記処理アセンブリが、さらに:
iv. 前記少なくとも1つの切断要素と前記外側チャンバーとの間にさらに置かれたスクリーンであって、一連の開口部を提供するスクリーン
を含む、請求項1に記載の遠心分離機。 - 前記スクリーンが、約0.04インチ〜約0.002インチの範囲の複数の開口部を含む、請求項4に記載の遠心分離機。
- 前記側壁に隣接して前記外側チャンバー内に配置されたトラップをさらに含む、請求項1に記載の遠心分離機。
- 前記トラップが、前記側壁と前記皿状部の一部分との間に配置されている、請求項6に記載の遠心分離機。
- 前記処理アセンブリが、さらに、前記内側および外側チャンバーが内部に配置される、静止したハウジングを含む、請求項1に記載の遠心分離機。
- 前記遠心分離機が、前記生物学的混合物が内側チャンバー内に配置されているときに、前記内側チャンバーを第1の回転速度で回転させ、および前記生物学的混合物が外側チャンバー内に配置されているときに、前記外側チャンバーを第2の回転速度で回転させるように配置されている、請求項1に記載の遠心分離機。
- 前記遠心分離機が、長さ約1mm未満の断片を有する、少量ずつに分けられた生物学的混合物を生成するように配置されている、請求項3に記載の遠心分離機。
- 前記回転源によって前記処理アセンブリを選択的に係合できるようにして、その処理アセンブリの回転を生じさせる、解放可能な結合部をさらに含む、請求項1に記載の遠心分離機。
- 前記皿状部が、前記皿状部から前記外側チャンバーの前記内部まで延在するウェッジを含み、および前記皿状部を第1の領域および第2の領域に分割する前記中心縦軸から第1の半径方向距離にウェッジ端部を有する、請求項1に記載の遠心分離機。
- 前記外側チャンバーの前記側壁が屈曲部を含み、前記屈曲部は、前記中心縦軸から第2の半径方向距離にあり、前記第2の半径方向距離は前記第1の半径方向距離以下である、請求項12に記載の遠心分離機。
- 生物学的混合物を処理しかつ前記生物学的混合物の構成成分を選択的に濃縮するための遠心分離機であって、前記構成成分は、異なる比重を有し、および前記遠心分離機によって生じた遠心力場において層状にでき、前記遠心分離機は:
a. 処理アセンブリであって:
i. 前記生物学的混合物を受け入れるように配置された回転可能なチャンバーであって、および前記回転可能なチャンバーは、前記チャンバーが周りで回転するように配置された中心縦軸を有し、前記チャンバーは、側壁と、ベースと、回転可能な管と、回転可能なサイジング処理用らせん体とを有するテーパ付きチューブ状部材を含み、前記サイジング処理用らせん体は、前記回転可能な管内の前記内側チャンバーへと延在する、チャンバーを含み、
ii. 前記生物学的混合物は、前記回転可能な管の前記内部を通って前記チャンバーへ導入され、および前記らせん体の回転に曝されて、前記生物学的混合物のサイジング処理を行うようにされた、処理アセンブリ;および
b. 前記内側および外側チャンバーに結合された回転源
を含む、遠心分離機。 - 前記チャンバーの前記回転によって、前記サイジング処理された生物学的混合物を、前記チャンバー内で、少なくとも2つの同心の層状の構成成分層へと層状にする、請求項14に記載の遠心分離機。
- 生物学的混合物の少なくとも1つの構成成分を選択的に濃縮するための遠心分離機であって、前記構成成分は、異なる比重を有し、および前記遠心分離機によって生じた遠心力場内で層状にでき、前記遠心分離機は、前記生物学的混合物を受け入れるように配置された内側チャンバーを含み、かつ前記遠心分離機は、前記内側チャンバーが周りで回転するように配置された中心縦軸を有し、前記内側チャンバーは、テーパ付き内表面を有する側壁と、ベースと、環状スクリーンと、前記縦軸から第2の半径方向距離に入口を有する第1のポートとを含み、前記環状スクリーンは、内表面を有し、かつ前記中心縦軸から第1の半径方向距離に配置され、前記環状スクリーンは前記ベースから突出している、遠心分離機。
- 前記ベースが、さらに、前記ベースから前記チャンバーの前記内部へ延在しかつ前記中心縦軸から第3の半径方向距離にウェッジ端部を有するウェッジ要素を含む、請求項16に記載の遠心分離機。
- 前記内側チャンバーがさらにトラップを含み、前記トラップは、前記側壁の前記内表面に隣接して前記内側チャンバーに配置され、および、前記内側チャンバーは、さらに、前記縦軸から第4の半径方向距離に入口を有する第2のポートを含み、および前記第2のポート用の前記入口は、前記トラップ内に配置されている、請求項17に記載の遠心分離機。
- 前記第4の半径方向距離が前記第3の半径方向距離を上回り、前記第3の半径方向距離は前記第2の半径方向距離を上回り、および前記第2の半径方向距離は前記第1の半径方向距離を上回る、請求項18に記載の遠心分離機。
- 前記内側チャンバーが、さらに、前記スクリーンの前記内表面の周りを効果的に転がるように配置された少なくとも1つのローラを含み、前記生物学的混合物の少なくとも一部分を、前記スクリーンを通ってかつ前記中心縦軸から離れおよび前記テーパ付き側壁の方へ推進させるようにする、請求項16に記載の遠心分離機。
- 前記内側チャンバーが、さらに、前記中心縦軸に対して平行に延在する少なくとも1つのローラアクスルを含み、前記少なくとも1つのローラは、前記少なくとも1つのローラアクスルの周りで回転して前記スクリーンの前記内表面の周りを転がるように配置されている、請求項19に記載の遠心分離機。
- 前記少なくとも1つのローラアクスルが、前記内側チャンバーに対して静止して保持される、請求項20に記載の遠心分離機。
- 前記内側チャンバーの前記回転によって、前記生物学的混合物の前記構成成分が、前記構成成分の異なる比重に応じて、同心の層状の構成成分層へと層状にするようにし、第1の構成成分層が、前記側壁の前記内表面の円周の一部分に接して配置されるようにする、請求項16に記載の遠心分離機。
- 前記環状スクリーンが、ワイヤーメッシュスクリーン、ポリマーメッシュスクリーン、有孔金属管および有孔ポリマー管からなる群から選択される、穴のあいた材料である、請求項16に記載の遠心分離機。
- さらに、前記トラップから前記構成成分層の少なくとも1つの少なくとも一部分を選択的に排出する少なくとも1つのポートおよび弁を含む、請求項16に記載の遠心分離機。
- 前記スクリーンが、約0.04インチ〜約0.002インチの範囲の複数の開口部を含む、請求項16に記載の遠心分離機。
- 生物学的混合物を処理しかつ前記生物学的混合物の構成成分を選択的に濃縮する方法であって、前記構成成分は、異なる比重を有し、および遠心力場において層状にでき、前記方法は:
a. 前記生物学的混合物を遠心分離機の内側チャンバーへ導入するステップであって、前記内側チャンバーは、テーパ付き内表面を備える側壁と、ベースと、内表面を有する環状スクリーンと、および第1のポートとを含む、ステップ;
b. 前記内側チャンバーを軸の周りで回転させて、前記生物学的混合物の少なくとも一部分が前記スクリーンを通過するようにし、前記生物学的混合物のサイジング処理を行うステップ;および
c. 前記チャンバーを軸の周りで回転させて、前記サイジング処理された生物学的混合物を、前記チャンバー内で少なくとも2つの同心の層状の構成成分層へと層状にするようにするステップ
を含む、方法。 - 前記生物学的混合物が、脂肪組織および水ベースの液体を含む、請求項27に記載の方法。
- 前記少なくとも2つの同心の層状の構成成分層の一方が、多能性細胞を含む、請求項27に記載の方法。
- さらに、前記遠心分離機から前記多能性細胞を採取するステップを含む、請求項29に記載の方法。
