Abstract
Description
「ペプチド」という用語は、2〜50のアミノ酸で構成される分子を意味する。
加えて、可変領域は非ヒト抗体のヒト化において用いられてよい。
配列番号14によってコードされるマウス抗AGEκ軽鎖のタンパク質配列は、配列番号18に示される。マウス抗体の可変領域は配列番号21に示され、配列番号18の21〜132位に対応する。抗体軽鎖はあるいは、キメラ抗AGEヒトκ軽鎖である、配列番号15のDNA配列によってコードされてよい。配列番号15によってコードされるキメラ抗AGEヒトκ軽鎖のタンパク質配列は、配列番号19に示される。キメラ抗AGEヒト免疫グロブリンは、21〜132位に配列番号21のマウス可変領域を含む。

実施例1:抗糖化最終産物抗体の投与のin vivo検査
抗糖化最終産物抗体の効果を検討するために、老齢CD1(ICR)マウス(Charles River Laboratories)に抗体を3週間の間、週に1回(1日目、8日目および15日目)、1日2回静脈内注射により投与し、その後10週間の無処置期間が続いた。試験抗体は、キーホールリンペットヘモシアニンと共役結合されたカルボキシメチルリジンに対して産生された市販のマウス抗糖化最終産物抗体であり、このカルボキシメチルリジンモノクローナル抗体(Clone318003)はR&D Systems, Inc.(Minneapolis,MN;カタログ番号MAB3247)から入手可能であった。対照動物において生理食塩水の対照基準を使用した。
細胞のAGE修飾タンパク質のためのモデル基質としてNα,Nα−ビス(カルボキシメチル)−L−リジントリフルオロ酢酸塩(Sigma−Aldrich,St.Louis,MO)を使用して、実施例1で使用した試験抗体の親和性および動態を解析した。ブランクとしてFc1、および試験抗体で固定したFc2(分子量150,000Da)とともにシリーズSセンサチップCM5(GE Healthcare,Pittsburgh,PA)を用いて、ラベルフリー相互作用解析をBIACORE(商標)T200 (GE Healthcare,Pittsburgh,PA)上で行った。泳動緩衝液は、温度25℃で、HBS−EP緩衝液(10mMのHEPES、150mMのNaCl、3mMのEDTAおよび0.05%のP−20、pH7.4)であった。ソフトウェアは、BIACORE(商標)T200評価用ソフト、2.0版であった。二重基準(Fc2−1および緩衝液だけの注入)を解析で使用し、データをLangmuir1:1結合モデルにフィットさせた。
マウス抗AGE抗体とキメラヒト抗AGE抗体を調製した。マウス抗AGE抗体IgG2b重鎖のDNA配列を配列番号12に示す。キメラヒト抗AGE抗体IgG1重鎖のDNA配列を配列番号13に示す。マウス抗AGE抗体κ軽鎖のDNA配列を配列番号14に示す。キメラヒト抗AGE抗体κ軽鎖のDNA配列を配列番号15に示す。これらの遺伝子配列を合成し、哺乳動物高発現ベクター中にクローン化した。配列は、コドン最適化された。完成した構築物は、トランスフェクションに進む前に配列を確認した。
実施例3に記載のマウス(親)抗AGE抗体およびキメラ抗AGE抗体の結合を直接結合ELISAによって検討した。抗カルボキシメチルリジン(CML)抗体(R&D Systems,MAB3247)を対照として使用した。CMLをKLHに共役結合し(CML−KLH)、次いでCMLとCML−KLHの両方をELISAプレート上に終夜被覆した。HRPヤギ抗マウスFcを用いて、対照抗AGE抗体とマウス(親)抗AGE抗体を検出した。HRPヤギ抗ヒトFcを用いて、キメラ抗AGE抗体を検出した。
ヒト化抗体を、親(マウス)抗体配列の抜粋した部分をヒトフレームワーク配列と融合する複数のハイブリッド配列を作成することによって設計した。アクセプターフレームワークを、フレームワークにわたる全体の配列同一性、マッチング界面位置、同様に分類されたCDR正準位置、および除去されるべきN型糖鎖付加部位の存在に基づいて同定した。3つのヒト化軽鎖と3つのヒト化重鎖を、2つの異なる重鎖および軽鎖ヒトアクセプターフレームワークに基づいて設計した。重鎖のアミノ酸配列を配列番号29、31および33に示し、それらは配列番号30、32および34に示すDNA配列によってそれぞれコードされる。軽鎖のアミノ酸配列を配列番号35、37および39に示し、それらは配列番号36、38および40に示すDNA配列によってそれぞれコードされる。ヒト化配列を、抗原結合を保持している可能性が最も高い配列を単離するために目視およびコンピュータモデリングで系統的に分析した。目標は、最終ヒト化抗体におけるヒト配列の量を最大にし、一方で本来の抗体特異性を保持することであった。ヒト化軽鎖および重鎖を組み合わせて、9つの異なる完全ヒト化抗体を作成できる。

カルボキシメチルリジン修飾を示すタンパク質またはペプチドに結合する、抗AGE抗体。 - 抗AGE抗体であって、配列番号22、配列番号23、配列番号24、配列番号25、配列番号26、配列番号27、および配列番号28からなる群から選択されるアミノ酸配列と少なくとも90%の配列同一性、好ましくは少なくとも95%の配列同一性、より好ましくは少なくとも98%の配列同一性を有する少なくとも1つのアミノ酸配列を含むタンパク質またはペプチドを含み、
カルボキシメチルリジン修飾を示すタンパク質またはペプチドに結合する、抗AGE抗体。 - 抗AGE抗体であって、
重鎖、および
軽鎖
を含み、
前記重鎖が配列番号1、配列番号17、配列番号29、配列番号31、および配列番号33からなる群から選択されるアミノ酸配列と少なくとも90%の配列同一性、好ましくは少なくとも95%の配列同一性、より好ましくは少なくとも98%の配列同一性を有するアミノ酸配列を含み、または

カルボキシメチルリジン修飾を示すタンパク質またはペプチドに結合し、ならびに
マウス、ラット、ヤギ、ヒツジ、ウシ、ウマ、イヌおよびネコからなる群から選択される種に対して実質的に非免疫原性である、抗AGE抗体。 - 抗体複合体であって、
配列番号1、配列番号3、配列番号17、配列番号19、配列番号29、配列番号31、配列番号33、配列番号35、配列番号37、および配列番号39からなる群から選択されるアミノ酸配列と少なくとも90%の配列同一性、好ましくは少なくとも95%の配列同一性、より好ましくは少なくとも98%の配列同一性を有する少なくとも1つのアミノ酸配列を含むタンパク質またはペプチドを含む抗AGE抗体フラグメントと、
前記抗AGE抗体フラグメントに共役結合される、AGE修飾細胞の破壊を引き起こす薬剤と

- 部分的に機能する細胞または非機能細胞を死滅させることにおける使用のための請求項1〜7のいずれかに記載の抗AGE抗体。
- 部分的に機能する細胞または非機能細胞を死滅させることによって対象中のサルコペニアを処置することにおける使用のための請求項1〜8のいずれかに記載の抗AGE抗体。
- 部分的に機能する細胞または非機能細胞を死滅させることによって対象中の組織または臓器の再生を促進することにおける使用のための請求項1〜9のいずれかに記載の抗AGE抗体。
- 部分的に機能する細胞または非機能細胞を死滅させることによって対象中の再生過程を促進するもしくは老化の影響を克服することにおける使用のための請求項1〜10のいずれかに記載の抗AGE抗体。
- 部分的に機能する細胞または非機能細胞を死滅させることによって対象中のアテローム動脈硬化症を処置することにおける使用のための請求項1〜7のいずれかに記載の抗AGE抗体。
- 対象中の白内障の発症を防止するもしくは遅延させることにおける使用のための請求項1〜7のいずれかに記載の抗AGE抗体。
- 対象中の脂肪組織の喪失の開始を防止するもしくは遅延させることにおける使用のための請求項1〜7のいずれかに記載の抗AGE抗体。
- 対象中の脊柱前弯・後弯症の発症を防止するもしくは遅延させることにおける使用のための請求項1〜7のいずれかに記載の抗AGE抗体。
- 対象中の炎症または自己免疫不全を処置することにおける使用のための請求項1〜7のいずれかに記載の抗AGE抗体。
- 対象中の神経変性障害を処置することにおける使用のための請求項1〜7のいずれかに記載の抗AGE抗体。
- 対象中の癌を処置することにおける使用のための請求項1〜7のいずれかに記載の抗AGE抗体。
- 老化細胞を死滅させるための薬物の製造のための請求項1〜18のいずれかに記載の抗AGE抗体の使用。
- サルコペニアを処置するため薬物の製造のための請求項1〜18のいずれかに記載の抗AGE抗体の使用。
- 前記対象がAD、PD、レビー小体病、MS、プリオン病およびALSからなる群から選択される神経変性障害を有する、請求項1〜20のいずれかに記載の抗AGE抗体。
- 前記処置することが前記対象の中枢神経系に前記抗AGE抗体を投与することを含む、請求項1〜21のいずれかに記載の抗AGE抗体。
- 前記対象がALSまたはMDを有し、および
前記処置することが前記対象の筋肉に前記抗AGE抗体を投与することを含む、
請求項1〜21のいずれかに記載の抗AGE抗体。 - 前記抗体がヒト、マウス、ラット、ヤギ、ヒツジ、ウシ、ウマ、イヌおよびネコからなる群から選択される種に対して実質的に非免疫原性である、請求項1〜23のいずれかに記載の抗AGE抗体。

- 前記抗体が最大限で9×10−3/秒の解離速度(kd)を有する、請求項1〜27のいずれかに記載の抗AGE抗体。
- 前記抗体がAGE修飾細胞の破壊を引き起こす薬剤に共役結合される、請求項1〜28のいずれかに記載の抗AGE抗体。
- 前記薬剤が毒物、細胞傷害性薬剤、磁性ナノ粒子、および磁気スピン渦ディスクからなる群から選択される、請求項1〜29のいずれかに記載の抗AGE抗体。
