イタドリ(P. cuspidatum)の根から30種以上の化学化合物が単離されている。これらの成分の中には:スチルベン、アントラキノン、フラボノイド、フェノール、ステロール、精油(essential oils)及びアミノ酸がある。伝統的な民間薬(folk medicine)におけるその使用の要因であり得る、報告された生物学的活性を有するイタドリ根の成分は:レスベラトロール、ポリダチン(polydatin)、エモジン(emodin)、フィスシオン(physcion)、レイン(rhein)並びにアントラグリコシド(anthraglycosides)A及びBを含む。これらの生理活性物質のいくつかは、光増感反応に関与する、照射時に励起三重項状態を引き起こす構造的特徴を所持する。いくつかの実施形態において、照射により生じる励起三重項状態は、酸素及び他の種との光増感反応に関与する。エモジン、フィスシオン及びそれらのそれぞれのグリコシル化された誘導体(derivatives)、アントラグリコシドA及びBは、微生物の光力学的不活性化及び光力学療法の効果を生じさせる。いくつかの実施形態において、単離方法は、結果として生じる抽出物を、光力学療法及び微生物の光力学的不活性化の要因となる光増感分子に関して富化するために開発されている。一つの実施形態において、イタドリ抽出物中の様々な化学物質の相対量は、表1に記載される通りである。

本発明はまた、本発明によるイタドリ抽出物を含む組成物(compositions)又は製剤に関する。概して、本発明の組成物は、細菌、ウイルス、真菌のような微生物の光力学的不活性化(PDI)提供するために有効な本発明によるイタドリ抽出物の有効量;及び一つ又はそれ以上の賦形剤を含む。
本発明のイタドリ抽出物は、改善された経口生物学的利用能(バイオアベイラビリティ)ばかりでなく、改善された細胞有効性、薬物動態特性をも有するという事実が利用され得ることもまた、明確に理解されるべきである。
他の実施形態において、イタドリ抽出物の相対百分率比は、約95%である。他の実施形態において、イタドリ抽出物の相対百分率比は、約100%である。

ジメチルスルホキシド(DMSO、99.7%)はFisher Scientificから購入された。5%水性プロピレングリコール(PG)は、市販のプロピレングリコール(99.5%、Sigma-Aldrich(登録商標))を滅菌脱イオン水で希釈することによって、in vivo試験のための賦形薬として調製された。トルイジンブルーO(TB)は、Sigma-Aldrichから購入され、滅菌脱イオン水で希釈された。Listerine(登録商標) Zero(Johnson & Johnson(登録商標))は、地元の食料品店で購入され、Oro-Clense(Germiphene Corporation, DIN 02209055)は、歯科医院から得られた。クロルヘキシジンジグルコネート(CHX)は、購入され(Sigma-Aldrich)、滅菌水で希釈された。
イタドリの植物根から過剰な土壌及び根毛が取り除かれ、イタドリの植物根は、続いて往復動鋸及び剪定鋏を使用して小片に切断された。根片は、余分な土壌を除去するために洗浄され、Earthwise Chipper Shredder(モデル番号GS70014)を用いて削られたか、又は市販のVita-Mixer Maxi-4000(Vitamix Corporation、モデル番号479044)を使用して粉砕し、粉末にした。処理された根は、抽出前に少なくとも1日間乾燥させられた。
イタドリ植物は、2012年4月にHalifax, NSから収穫され、promix soil(ASB Greenwood)に移植され、2013年2月の処理まで周囲条件下で人工気象室(phytotron)内に保持された。根粉(0.5g)は、200mlエタノールを用いて、溶媒が無色になるまでソックスレー抽出された。エタノール抽出物は減圧下で濃縮され、132mgの粗抽出物を得た。
イタドリ根は、2013年5月に、Wolfville, NSにおいて生長する43の植物から収穫された。根は人工気象室に移植され、そこでは22の標本(specimens)がネイティブな土壌内で増殖させられ(propagated)、21の標本がpromix soil内で増殖させられた。植物標本は、全43の根が収穫された2014年1月から2014年5月までに、人工気象室から集められた。この材料は削られ、組み合わされ、1.5kgは7.5Lのエタノールを用いて室温で抽出された。11週目に、40mLのアリコート(aliquot)が取り出され、真空で濃縮されて497mgの粗抽出物を与えた。
トリプシン大豆寒天(TSA)は、生体外(in vitro)寒天ウェル拡散試験のために使用された。培地(media)は、250mLの脱イオン(DI)水の中に7.5gのトリプシンソイブロス(TSB)を3.75gの寒天と合わせることにより、500mLのエルレンマイヤーフラスコ(三角フラスコ)内で調製された。フラスコ上端を綿栓と箔で覆い、懸濁液は121℃で1時間オートクレーブ処理された。液体寒天のよく混合されたアリコート(20mL)は、ブンゼンバーナの近くで滅菌血清ピペット(25mL)を用いて100×15mmペトリ皿に移され、固化させられた。平均寒天深さは4mmであった。プレートは標識され、Parafilm(登録商標)を用いて密封され、使用する前に1日から2日間4℃で保存された。
無菌技術を使用して、1バイアルのストレプトコッカス・ミュータンス(S. mutans Clarke, ATCC 25175, 指定(designation)NCTC 10449)は、凍結乾燥ペレットの半分を、滅菌ループを使用して2mLのブレインハートインフュージョン(Brain Heart Infusion)培地(BHI、Oxoid)を含む培養管に移し、旋回により穏やかに混合することによって、増殖させられた。チューブは、ゆるく蓋をかぶせられ、37℃のインキュベーター内に24時間にわたって置かれた。翌日、10回の連続希釈が行われ(10−1から10−8)、次いで各希釈物からの0.1mLアリコートが、滅菌ループを使用して未使用の(fresh)BHI寒天プレート(3.8%BHI)の上に広げられ、斜めに蓋をして乾燥させられ、次いで37℃のインキュベーター内に上下逆さにして一晩置かれた。コロニー増殖の純度が確認され、細菌培養管は、遠心分離(5000rpm、5分間)し、上清を慎重に注ぎこぼして廃棄し、5mLの未使用の培地と交換することによって、継代培養された(subcultured)。ストレプトコッカス・ミュータンスの凍結ストックは、ストレプトコッカス・ミュータンス培養物の500μLアリコートを、水中の滅菌70%グリセロール500μLを含む滅菌1.5mLマイクロ遠心チューブに移すことによって、調製された。チューブは、短時間ボルテックスすることによって混合され、続いて−80℃で保存された。
ストレプトコッカス・ミュータンスの接種物は、室温の二次増殖プレートからのコロニーを、5mL滅菌蒸留水を含む滅菌15mLコニカルチューブに移すことによって調製され、内容物は、ボルテックス処理によって十分に混合された。625nmでの吸光度が読み取られ、おおよその濃度がマクファーランド硫酸バリウム標準曲線に従って算出された。次いで、濃度は、マクファーランド基準0.5(およそ1.5×108CFU mL−1)に一致するように調整された。接種物の300μLアリコートは、菌叢(bacterial lawn)の均一な増殖を可能にするために、滅菌綿チップアプリケータ(MedPro,コード018-426)を用いて各TSAプレート上に均一に広げられた。プレートは、斜めに蓋をして約30分間乾燥させられ、試料は、穿孔の中又は紙ディスクの上に加えられた(delivered)。
穿孔ウェル(直径5.5mm、深さ4mm)は、
紙ディスクは、直径6mmのディスクを与える穴開けパンチを用いて、Whatman filter paper 1(GE(登録商標)Healthcare Life Sciences,コード1001 090)から作られた。紙ディスクは、エタノールで殺菌され、次いで使用の前に乾燥させられた。滅菌鉗子を使用して、紙ディスクはBHADのそれに類似したパターンで、接種されたTSA上に静かに置かれた。ひとたび紙ディスクが適切に配置されると、10μLの試料が各ディスクに塗布された。
試料は、以前に記述されたBHAD又はPDADの方法に従って、0.5マクファーランドバリウム標準接種物(barium standard inoculum)(1.5×108CFU mL−1)をそれぞれ有する、一双に(in duplicate)(暗プレート及び明プレート(dark and light plates)))調製されたTSAプレートに送達された。試料は:3mg mL−1クロルヘキシジン、脱イオン水、5%PG、5%プロピレングリコール中の3mg mL−1の本開示のイタドリ抽出物、及び5%プロピレングリコール中の本開示の11mg mL−1イタドリ抽出物、を含んでいた。試料を有するプレートは室温で30分間インキュベートされ、透過プロジェクタ(transparency projector)(Bell & Howell,モデル番号310 LA)を用い、プロジェクタのベースから16.2cmの距離での、15分間にわたる明プレートの照射が続き、100Jcm−2の光線量を得た。この照射期間中、暗プレートは、同等の時間にわたって、光から保護された状態に保たれた。明又は暗の処置に続いて、プレートは37℃で一晩インキュベートされ、翌朝分析された。
本開示のイタドリ抽出物の濃度ベースのアッセイは、DMSO中0.1、1、10、100mg/mLの抽出物を使用して実行された。5mg/mLのクロルヘキシジン及びDMSOのみの対照が含まれていた。TSAプレートは、1.0マクファーランド標準(3.0×108CFU mL−1)を用いて、BHAD又はPDADの方法に従って、一双に(暗プレート及び明プレート)調製された。明プレートは、光反応器(Luzchem Research Ltd.モデル番号LZC-4V)内で、16個のクールホワイト電球(Osram Sylvania Inc.コードF8T5-CW)を用いて1時間にわたって照射され、35Jcm−2の光線量を得た。この照射期間中、暗プレートは、同等の時間にわたって、光から保護された状態に保たれた。明又は暗の処置に続いて、プレートは37℃で一晩インキュベートされ、翌朝分析された。
市販のリステリン、Oro-Clense、及び1%TBは、DMSO中の10mg mL−1及び100mg mL−1イタドリ抽出物と一緒に、濃度ベースのアッセイ研究において記述された寒天拡散分析に付された。明プレートに対して放出された総光線量は36Jcm−2であった。
最小阻害濃度(MIC)及び最小殺菌濃度(MBC)は、96ウェルマイクロタイタープレート内で増殖する懸濁培養物に対して、マイクロウェル希釈法(Sahin, Fら、J Ethnopharmacol 2003, 87, 61-65、Methods for Antimicrobial Susceptibility Testing of Anaerobic Bacteria; 承認された標準、,第6版。Wayne, Pennsylvania: National Committee for Clinical Laboratory Standards; 2004)に従って決定された。接種懸濁液は、100μLの希釈された懸濁液(アッセイにおける最終的な濃度5×105CFU mL−1)及び水中の最大で5%のプロピレングリコール100μLの希釈された抽出物を組み合せることによって、二次増殖プレートから調製され、アッセイにおいて0.01mg mL−1と50mg mL−1の間の最終的な抽出物濃度を得た。2つのマイクロタイタープレートが並行して同じように調製され、一つは、37℃で30分間のプレインキュベーションの後に、光反応器内で電球を用いて、1時間にわたって35Jcm−2の光処置を受けた。その後、光処置プレート及び暗プレートの両方は、37℃で16時間にわたってインキュベートされ、次いで、CFU mL−1は、TSA寒天プレートを使用したMiles及びMisraの液滴計数技術(Miles, A. A,ら、J Hyg(Lond)1938, 38, 732‐749)に従って、各ウェルについて定量された。
イタドリ抽出物及び光によるヒトの歯上のバイオフィルム形成阻害は、文献(Cho, Y. S.ら、Biotechnol Bioprocess Eng 2010, 15, 359-364)から適応されたプロトコールに従って評価された。簡潔に述べると、ヒトの歯は、ストレプトコッカス・ミュータンスを接種された(24ウェルマイクロタイタープレート内の)BHIブロス中に37℃で4日間にわたって、キャンドルジャー(5%CO2)内で保持された。2−4日目に1日3回3分間にわたって、歯は取り出され、生理食塩水(対照)又はイタドリ抽出物(5mg mL−1)のいずれかの中に置かれた。この短いインキュベーション期間に続いて、試料の半分は100Jcm−2の光を用いて照射され、残りの半分は暗条件下に保たれ、次に明試料と暗試料の両方が接種されたBHIブロス溶液に戻された。5日目に、試料は画像化のために調製され、6日目に、歯の表面上のバイオフィルム形成が走査型電子顕微鏡(SEM)によって調べられた(Somayaji, K.ら、Iran Endod J 2010, 5, 53-58)。
暗条件下及び光が媒介するイタドリ抽出物の抗菌作用は、トリプシン大豆寒天(TSA)拡散試験(表10;図7)を使用して調査された。0.01%から10%までの、DMSO中の抽出物の増加する濃度は、300μLの黄色ブドウ球菌又はストレプトコッカス・ミュータンス接種物(3×108CFU mL−1)に対して試験された。30分間のインキュベーション期間の後に、1枚のプレートは光を用いて1時間にわたって照射され、35Jcm−2の総光線量を得たが、同時に、暗プレートは光から保護された。阻害ゾーンは更なる16時間のインキュベーション期間の後に測定され、これらの抽出物の強度及び条件に対する細菌の感受性(bacterial sensitivity)の程度を示した。最も低い濃度、0.01%及び0.1%において、イタドリ抽出物は、暗条件下において黄色ブドウ球菌又はストレプトコッカス・ミュータンスに対して識別可能な抗菌作用を全く示さなかった。しかしながら、イタドリ抽出物の光活性化は、試験した両方の細菌種に対して、それぞれ2つの濃度について8mm及び12mmの阻害ゾーンを生じさせた。1%のイタドリ抽出物強度は、両細菌に対して9mmの暗条件下での阻害ゾーン(dark inhibition zones)を与え、10%は黄色ブドウ球菌及びストレプトコッカス・ミュータンスに対してそれぞれ16mm及び15mmの暗条件下での阻害ゾーンを与えた。1%及び10%イタドリ抽出物溶液の光活性化は、両方の種の細菌に対してそれぞれ15mm及び18mmの阻害ゾーンを生じさせた。注目すべきことに、最高濃度のイタドリ抽出物によって生成された阻害ゾーンは、市販の強度の40倍の広範スペクトル抗生物質クロルヘキシジンによって生成されたものと同じであった。市販のクロルヘキシジン製品Oro-Clenseに対して試験された場合、光活性化された1%イタドリ抽出物は同等に有効であり、10%イタドリ抽出物はクロルヘキシジンよりも顕著に活性が高かった(表11;図8)。比較すると、Listerine Zeroはこのアッセイにおいて抗菌効果を有していなかった。これらの結果は、広範スペクトル抗生物質として作用する光反応性イタドリ抽出物の多能性(versatility)を強調している。重要なことに、微生物の光力学的不活性化は、即時的かつ無差別に作用し、従来の抗生物質に疑念を抱かせる抵抗の脅威を除く。

注目すべきことに、イタドリ抽出物の光増感力(photosensitizing power)は、抽出物のうちの何れか一つの特定可能な成分のそれよりもはるかに大きな、微生物の光力学的不活性化効果を引き起こした。アントラキノン又はそのグリコシル化誘導体は、抽出物の光増感効果の要因であり得る。分画によって単離された場合には、エモジンが最も大きな光力学的効果を生じさせ、フィスシオンが続いた。それらのグリコシドである、アントラキノンB及びAは、それぞれ不活性であった。アントラキノン類の市販の試料は、エモジン>フィスシオン>レインの順で微生物の光力学的不活性化を生じさせた。いくつかのイタドリ抽出物において、エモジン及びフィスシオンの割合は、それぞれ0.51‐0.65重量%及び0.24‐0.27重量%の間であった。ストレプトコッカス・ミュータンス(又は黄色ブドウ球菌)が塗られたTSAプレートに送達されたイタドリ抽出物(40μg)の寒天拡散試験において、微生物の光力学的不活性化阻害ゾーンは、それらの最も幅広い直径において実に35‐37mm、平均で24‐28mmの大きさであり、抽出物が光処置に予め曝露された場合に弱められなかった(図9;表12)。

イタドリ抽出物及び光による微生物の光力学的不活化が口腔内の細菌負荷を効果的に減少させることができるかどうかを決定するために、オスのCD−1マウスモデルが利用された。ストレプトコッカス・ミュータンスの生存率に対する、5分間の30Jcm−2の光による光増感の効果が、処置の直後に測定された(図5)。マウスは以下の処置の1つを受けた:(i)賦形薬(ビヒクル)、暗;(ii)賦形薬、光;(iii)3mg mL−1抽出物、暗;(iv)3mg mL−1抽出物、光;(v)11mg mL−1抽出物、暗;又は(vi)11mg mL−1抽出、光。群(i)及び群(ii)は対照群であり、これらの2つの群の間に有意差はなかった(歯肉についてp=0.16、舌については0.25)。光の非存在下でのイタドリ抽出物処置による細菌生存率の低下があったが、これらの減少は、最高濃度の抽出物において自然対数で約2(2log10)のみであった。イタドリ抽出物が光と併せて使用された場合、3mg mL−1で更に自然対数で1‐2(1-2log10)の殺滅が達成され、一方で全てのストレプトコッカス・ミュータンスの全破壊は11mg mL−1で起こった(p<0.001)。11mg mL−1抽出物を用いた暗処置と光処置との間の差異は、自然対数でおよそ5(5log10)(p<0.001)であった。これらの傾向は、両方の処置領域に当てはまる。重要なことに、潰瘍形成又は炎症の肉眼で見える形跡はなかった(図10)。組織学的評価に関しては、最高濃度のイタドリ抽出物及び光を用いた場合でさえ、炎症性浸潤の形跡はなかった(図11)。
およそ10週齢で体重25‐30gの、30匹のオスCD−1マウス(Charles River Laboratories International Ltd.,サンコンスタン、カナダ)が、この研究において使用された。マウスは、カナダ動物飼養協議会(Canadian Council of Animal Care)によって定められた規則に従って飼育された。マウスは、PDI処置に先立って、以下のレジメンに従って1週間にわたって接種された。1日目に、マウスは、通常の水の代わりに1%蔗糖(Sigma-Aldrich)水を受け入れた。2‐7日目に、マウスは、1×106CFU mL−1のストレプトコッカス・ミュータンス接種物とともに1%蔗糖水溶液を受け入れた。8日目の処置に続いて、全てのマウスは、新鮮な削りくずと清浄な水とともに滅菌ケージの中に置かれた。PDI処置日に、マウスは、舌の前部背側領域及び上顎切歯の上の歯肉領域に塗布された2×109CFU mL−1のストレプトコッカス・ミュータンスを含有する10μLの水を用いて、更に接種された。前述の場所は、本研究についての処置領域であった。
ケタミン及びキシラジン(各50mg/kgを包含する混合物)の0.1mLのIM(筋肉内)注射を使用して麻酔された。ひとたび標本が完全に麻酔されると、それは仰臥位の特別な台に位置させられた。口腔へのアクセスは、口を開くための弾性バンドを使用して達成され(バンドは、切歯の両方の組の周りに配置された)、舌及び上顎切歯を囲む上部歯肉領域を露出させた。最初に、2×109CFU mL−1のストレプトコッカス・ミュータンスを含有する10μLの水、続いて10μLの試料(ビヒクル、ビヒクル中の3mg mL−1又は11mg mL−1の抽出物、)が、上部歯肉領域に加えられた(delivered)。このプロセスは、舌の前部背側領域に対して繰り返された。これらの処置領域は、5分間にわたる30Jcm−2のLED光を用いて同時に照射されるか、又は5分間にわたって暗条件下に保持された。細菌スワブは、無菌歯内ペーパーポイント(Dia Dent、サイズ40)を使用して各処置領域から採取された。2つのペーパーポイントは、60秒間にわたって各々の特定の領域に塗り付けられ、その後に、更なる分析のために0.5mLの滅菌リンゲル‐ペプトンを含むスクリューキャップバイアルの中に置かれた。次いで標本は、弾性バンドから解放され、麻酔から回復するために清潔なケージの中に置かれた。細菌試料は、3時間以内に処理された。
舌及び歯肉組織標本(上顎領域の切歯を含む)は、固定剤(ブアン溶液)中で、4℃で2日間にわたって冷蔵保存された。歯を含む歯肉試料は、慎重に溶液を取り除き、新鮮な0.5mLブアン溶液で置換し、バイアルを室温で24時間放置することによって脱石灰化のために準備された。溶液は取り除かれ、試料は低速の流水中で洗浄された(3x10分)。次いで試料は、水(pH7.36)中の1mLの10%EDTAを含む滅菌24ウェルマイクロプレートに慎重に移された。10%EDTA溶液は2日ごとに15日の期間にわたって交換され、試料は、少なくとも24時間にわたって70%エタノール中に置かれた。舌組織は、2日間のブアン溶液中での保存の後に70%エタノールで1回洗浄され、少なくとも24時間にわたって70%エタノール中で保持された。試料は、100%エタノールで脱水され、トルエン中で清澄化され、ヘマトキシリン‐エオシン(H&E)染色による組織学的評価のためにパラフィンに包埋された。
試料は、以下のように脱水された:
組織試料は85%エタノール中で2時間浮かされ(suspended)、次いで溶媒が取り除かれた。次いで組織試料は95%エタノール中に1時間浮かされ、次いで溶媒が取り除かれた。次いで組織試料は100%エタノール中に0.5時間浮かされ、次いで溶媒取り除かれた。次いで組織試料はトルエン中に1時間浮かされ、溶媒が取り除かれ、組織試料はトルエンに再び浮かされた。1時間後、溶媒が取り除かれ、組織試料は60℃の熱い流動パラフィンに20分間真空下で浮かされ、次いでパラフィンが取り除かれた。パラフィン浮遊及び除去は、更に2回繰り返された。最後に、試料はパラフィンに包埋され、室温で放置されて硬化した。パラフィン包埋組織は、木製ブロックに載せられ、ミクロトーム(American Optical Company 820)を使用して6μmの厚さで切片化され、標識ポリ‐L‐リシン被覆スライドに移された。H&E染色のために、キシレンの2回の交換によってパラフィンが最初に溶解除去された。アルコールの2回の交換はキシレンを取り除き、スライドは、細胞及び組織要素を再水和する(rehydrate)ために水で徹底的にすすがれた。スライドは、ヘマトキシリン染料に浸漬され、水道水ですすがれ、70%エタノール中で脱染された。エオシンのアルコール溶液が塗布され、スライドは、水の全ての痕跡を除去するために数回のアルコール交換ですすがれた。組織を清澄化するためにキシレンの2回の交換が使用され、次いでポリスチレンマウント剤(Permoun(登録商標))が塗布され、ガラスカバースリップで覆われた。スライドは、顕微鏡分析に先立って少なくとも24時間にわたって乾燥させられた。
HL‐60細胞培養
HL−60ヒト前骨髄球性白血病細胞(ATCC CCL-240)は、20%FBS(PAA Laboratories, A15-701)を補充したRPMI 1640(Mediatech Media MT-10−040-CV)中で、37℃、5%CO2条件下で培養され、標準的な無菌手順に従って週に3‐4回継代された。培養は、25cm2組織培養フラスコ内の200,000細胞mL−1で開始され、増殖が800,000細胞mL−1に達したときに、長期間の高細胞密度に関連する老化を回避するために継代培養された。完全培地は、RPMI1640(160mL)及びFBS(40mL、予備秤量(プレアリコート)され、熱不活性化されている)を、250mL Millipore vacuum stericup(0.22μm)内で合わせること及びろ過することにより、必要に応じて200mL分ずつ調製された。
実験は、96ウェルマイクロタイタープレート(Corning Costar, Acton, MA)内で三重に(in triplicate)実行され、周辺に沿った外側ウェルは、試料ウェルからの蒸発を最小化するために、2.68mMの塩化カリウム、1.47mMの第一リン酸カリウム、0.137mMの塩化ナトリウム、8.10mMの第二リン酸ナトリウムを有する200μLのpH7.4リン酸緩衝生理食塩水(PBS)を含んでいた。対数期で増殖しているHL−60細胞(およそ8×105細胞)は、50μLのアリコートで温かい培地(25μL)を含む内側ウェルに移され、平衡させるために37℃、5%CO2水ジャケットインキュベーター(Thermo Electron Corp., Forma Series II, Model 3110, HEPA Class 100)内に1時間置かれた。ルテニウム化合物はPBSで連続希釈され、予め温められ、その後に適切な希釈液の25μLのアリコートが細胞に加えられ、1時間又は16時間の薬物対光の間隔にわたって37℃、5%CO2の下でインキュベートされた。処置されていないマイクロプレートは暗いインキュベーター内に維持され、一方でPDT処置されたマイクロプレートは、190W BenQ MS510オーバーヘッドフロジェクタを使用して、光(400‐700nm、27.8mWcm−2)を用いて照射された。照射時間は1時間であり、およそ〜100Jcm−2の光線量を得た。暗条件マイクロプレート及びPDT処理されたマイクロプレートの両方は更に48時間インキュベートされ、その時点において、予め温められた、アラマーブルー試薬(Life Technologies DAL 1025)の10μLのアリコートが、全ての試料ウェルに加えられ、37℃、5%CO2下で15‐16時間にわたってインキュベートされた。細胞生存率は、アラマーブルー酸化還元指示薬の、生細胞によって代謝的に蛍光色素に変換される能力に基づいて決定された。蛍光は、励起フィルターを530±25nmに設定し、発光フィルターを620±40nmに設定して、Cytofluor 4000蛍光マイクロプレートリーダーで定量化された。細胞傷害性及び光細胞傷害性(photocytotoxicity)のEC50値は、等式2(数3)に従い、Graph Pad Prism 6.0を使用して用量応答曲線のシグモイドフィットから計算された。ここで、yi及びyfは、初期蛍光シグナル強度及び最終蛍光シグナル強度である。対数増殖期及び同じ継代数で増殖する細胞に関して、EC50値は、マイクロモル以下のレジメン(submicromolar regime)で、±25%以内に再現可能であった;10μM未満、±10%;10μM以上、±5%。
製剤化された抽出物は有効であり、(示されるように)いくつかの製剤は、製剤化されていない抽出物よりもより有効ある。

接着性CCD-1064SK正常皮膚線維芽細胞(ATCC CRL-2076)は、10%FBS(PAA Laboratories, A15-701)を補充したイスコフ改変ダルベッコ培地(IMDM)中で培養され、37℃、5%CO2条件下でインキュベートされ、標準的な無菌手順に従って週に2〜3回継代された。CCD-1064SK細胞は、75cm2組織培養フラスコ内の200,000細胞mL−1で開始され、増殖が550,000細胞mL−1に達したときに、古い培地を取り除き、ダルベッコリン酸緩衝生理食塩水(DPBS1X、Mediatech, 21-031-CV)で一度細胞単層をすすぎ、続いて細胞単層をトリプシン−EDTA溶液(0.25%w/vトリプシン/0.53mM EDTA, ATCC 30-2101)で解離させること(dissociation)によって継代培養された。細胞の適切なアリコートが新しい細胞容器に移されることを可能にするために、完全増殖培地(Complete growth medium)が細胞懸濁液に加えられた。完全増殖培地は、IMDM(225mL)及びFBS(25mL、予備秤量(プレアリコート)され、熱不活性化されている)を、250mL Millipore vacuum stericup(0.22μm)内で合わせること及びろ過することにより、必要に応じて250mL分ずつ調製された。
Claims (28)
- イタドリ(Polygonum cuspidatum)の抽出物及び賦形剤を含む組成物。
- 前記賦形剤は、研磨剤、界面活性剤、結着剤、湿潤剤、香味剤、甘味剤、着色剤、保存剤及び水から成る群から選択される、請求項1に記載の組成物。
- 前記賦形剤は、水、シリカ、ソルビトール、グリセリン、キシリトール、ココ硫酸塩、デシルグルコシド、香味剤、キサンタンガム、カラゲナン及びグルタミン酸から成る群から選択される、請求項1に記載の組成物。
- 当該組成物は、溶液又は懸濁液として製剤化されている、請求項1に記載の組成物。
- 当該組成物は、ペーストとして製剤化されている、請求項1に記載の組成物。
- 当該組成物は、ゲルとして製剤化されている、請求項1に記載の組成物。
- 当該組成物は、泡状物として製剤化されている、請求項1に記載の組成物。
- 当該組成物内のイタドリ(Polygonum cuspidatum)抽出物の百分率比は、0.01%と20%の間である、請求項1に記載の組成物。
- 当該組成物内のイタドリ(Polygonum cuspidatum)抽出物の百分率比は、約0.1%である、請求項1に記載の組成物。
- 当該組成物内のイタドリ(Polygonum cuspidatum)抽出物の百分率比は、約1%である、請求項1に記載の組成物。
- 当該組成物内のイタドリ(Polygonum cuspidatum)抽出物の百分率比は、0.1%と10%の間であり、20%と45%の間の研磨剤、1%と2%の間の界面活性剤、0.5%と4%の間の結着剤、10%と30%の間の湿潤剤、1%と5%の間の香味剤、甘味剤及び着色剤、0.05%と0.5%の間の保存剤、並びに100%への残部の水、を更に含む、請求項1に記載の組成物。
- 当該組成物内のイタドリ(Polygonum cuspidatum)抽出物の百分率比は、0.1%と10%の間であり、10%と40%の間のシリカ、0%と5%の間のソルビトール、0%と5%の間のグリセリン、0%と1.5%の間のキシリトール、0%と2.5%の間のココ硫酸ナトリウム、0%と1.5%の間の香味剤、0%と1.5%の間のキサンタンガム、0%と1.5%の間のカラゲナン、並びに100%への残部の水、を更に含む、請求項1に記載の組成物。
- 当該組成物内のイタドリ(Polygonum cuspidatum)抽出物の百分率比は、0.1%と10%の間であり、約25%のシリカ、約1%のソルビトール、約1%のグリセリン、約0.5%のキシリトール、約1%のココ硫酸ナトリウム、約0.25%の香味剤、約0.5%のキサンタンガム、約0.5%のカラゲナン、及び100%への残部の水、を更に含む、請求項1に記載の組成物。
- 当該組成物内のイタドリ(Polygonum cuspidatum)抽出物の百分率比は、約1%であり、約76.65%の水、約10%のグリセリン、約8%のソルビトール、約1%のキシリトール、約0.35%の天然香料、及び約3%のラウロイルグルタミン酸ナトリウムを更に含む、請求項1に記載の組成物。
- 当該組成物内のイタドリ(Polygonum cuspidatum)抽出物の百分率比は、約0.1%であり、約79.55%の水、約10%のグリセリン、約8%のソルビトール、約1%のキシリトール、約0.35%の天然香料、及び約1%のココ硫酸ナトリウムを更に含む、請求項1に記載の組成物。
- 微生物を殺すか又は不活性化する方法であって、
前記微生物を、イタドリ(Polygonum cuspidatum)の抽出物及び賦形剤を含む組成物と接触させるステップ、及び
光源を用いて、前記微生物に照射するステップ、

前記癌細胞、癌腫又は腫瘍を、イタドリ(Polygonum cuspidatum)の抽出物及び賦形剤を含む組成物と接触させるステップ、及び
光源を用いて、前記癌細胞、癌腫又は腫瘍に照射するステップ、
を含む、方法。 - 当該方法は、酸素の存在下で実行される、請求項23に記載の方法。
- 良性の口腔又は咽頭の腫瘍を処置する方法であって、
前記口腔又は咽頭の腫瘍を、イタドリ(Polygonum cuspidatum)の抽出物及び賦形剤を含む組成物と接触させるステップ、及び
光源を用いて、前記口腔又は咽頭の腫瘍に照射するステップ、
を含む、方法。 - 当該方法は、酸素の存在下で実行される、請求項25に記載の方法。
