式(1)で表される化合物は、公知化合物を原料として、以下の製造方法で製造することができる。
式(1)で表される化合物またはその薬学的に許容される塩は、例えば以下の方法に従って製造することができる。
化合物(1−1)は、国際公開第2008/114817号パンフレットに記載の方法に従って製造することができる。具体的には、化合物(1−5)[2,6−ジクロロ−9−(テトラヒドロ−2H−ピラン−2−イル)−9H−プリン(式(1−4))より製造]を塩基の存在下、化合物(1−6)と反応させることにより製造することができる。
式(1)の化合物は、式(1−3)の化合物を酸性条件下で反応させることで製造することができる。用いられる酸は、例えば、塩酸またはトリフルオロ酢酸であってよい。酸の使用量は、例えば、式(1−3)の化合物1モルに対して0.1モル当量から20モル過剰量であってよい。塩酸は水溶液としてか、またはメタノール、1,4−ジオキサン等の有機溶媒中の溶液として用いることができる。工程2で用いられる溶媒としては、例えばテトラヒドロフラン、ジエチルエーテル、1,4−ジオキサン、1,2−ジメトキシエタン等のエーテル系溶媒;ジクロロメタン、クロロホルム、1,2−ジクロロエタン等のハロゲン系溶媒;メタノール、エタノール、イソプロパノール等のアルコール系溶媒、またはこれらの混合物が挙げられる。反応温度は、好ましくは約0℃〜60℃の範囲から選択されるが、これに限定されない。
式(2−1)の化合物は、国際公開第2008/114817号パンフレットに記載の方法に従って製造することができる。式(2−2)の化合物の脱離基LG3としては、ハロゲン原子、アルキルスルホニルオキシ基、アリールスルホニルオキシ基が挙げられるが、これらに限定されない。式(2−2)の化合物は、例えば、トリエチルアミン、ジイソプロピルエチルアミン、ピリジン、ジメチルアミノピリジン、炭酸ナトリウム、炭酸カリウム等の塩基の存在下、式(2−1)の化合物を塩化メタンスルホニル、塩化p−トルエンスルホニル等と反応させることにより、製造することができる。
製造法1の工程1と同様の条件を用い、式(2−2)の化合物と式(2−3)の化合物から式(1−3)の化合物を製造することができる。
式(1−3)の化合物またはその塩は、以下の方法にしたがって製造することもできる。

「ACVT−03」
工程1−油相の調製
実施例3の化合物22mgをスクリューキャップ付きの容器中で秤量し、2.5gのスクアレン油を添加した。実施例3の化合物を、粒子が見えなくなるまで25℃で60分間超音波処理することにより可溶化した。
別の容器に、1X PBS(95g)、ポロキサマー188(0.3g)およびSpan(登録商標)85(トリオレイン酸ソルビタン)(0.25g)を加えた。この溶液を粒子がなくなるまで室温で20分間混合した後、0.2ミクロンのPESまたはPVDFメンブレンフィルターを通して無菌的に濾過した。
工程1および工程2に従って調製した油相および水相を単一の容器中で混合し、ブレンドした。ブレンドは、9〜10,000RPMまでランプアップさせ、混合物の外観が乳白色の連続相となるまで5分間処理することにより行った。
工程3で調製した連続相エマルジョンを、Y型相互作用チャンバーで25,000〜30,000PSIにてマイクロ流動化した。8回通過後、プロセスは完了したと判断された。得られた乳白色の液体を脱パイロゲートガラスバイアルに分注し、使用しない場合は4℃で保存した。
「ACVT−01」
実施例6と同様の方法で調製したが、工程1の成分として実施例3ではなく実施例1の化合物を用いた。
「ACVT−02」
実施例6と同様の方法で調製したが、工程1の成分として実施例3ではなく実施例2の化合物を用いた。
「ME7」
以下の材料を用い、実施例6と同様の方法で調製した:

データは、2〜3回の実験の代表例である。
8〜9週齢のCB6F1マウスを0日目および14日目に、アジュバントとして実施例6、7および8(それぞれ「ACVT−03」、「ACVT−01」および「ACVT−02」)を添加した10μgのHSV2 gDt抗原(アミノ酸25−281)でi.m.にて免疫した(1群4匹のマウス)。実施例6、7および8は、マウス1匹当たりに送達されるTLR7Lの用量が20μg(100μl中)になるように投与した。比較のために、ある群のマウスを、AddaVax(InVivogen)(ワクチンFLUADに用いられているNovartis社の商業的に許諾されたスクアレンエマルジョンMF59と組成が同等である市販の水中油型スクアレンエマルジョン)および20μgのCpG−Cオリゴデオキシヌクレオチド2395(InVivogen)中50%v:vで製剤化したgDt抗原で免疫した。このCpG+AddaVax組成物は、マウスにおいて強いT細胞応答を誘導することが観察されており、陽性対照として用いた。単に可溶性TLR7Lをエマルジョンと混合する場合と、実施例6〜8の方法により調製したようにTLR7Lをエマルジョンの油滴内に送達し統合した場合のアジュバント活性の比較として、1つの群のマウスを、実施例2の化合物と単に混合した参考例10(「ACVT」)で免疫した。28日目に、マウスを出血させて脾臓を採取した。
脾細胞を、ELISPOTプレートにてgDtペプチド(gDt配列全体にまたがる11で重複する15マー)で24時間刺激した。スポット形成細胞をスキャンし、CTL ImmunoSpot Analyzer(Cellular Technologies Limited)を用いて分析した。図1では、実施例6〜8の3つすべてが、CpG/AddaVax製剤よりも高いIFN−γ応答を誘導した。図2では、各群の4匹のマウスの間で同様に脾細胞をプールし、抗CD4磁気ビーズ(Miltenyi)と結合させ、MACS(磁気活性化細胞ソーティング)を介してCD4 T細胞を除去して脾細胞集団(<1.0%CD4+であり、したがって主にCD8 T細胞を含有)が得られた。これらの細胞集団は、CD8特異的T細胞応答を検出するために、ELISPOTアッセイでも用いた。TLR7L−スクアレンコンジュゲートを含む製剤のうちの2剤である実施例6および8(それぞれ「ACVT−03」および「ACVT−02」)は、CpG/AddaVax製剤またはTLR7L+参考例10の混合物よりも高いCD8 T細胞応答を誘導した。
脾細胞を各群内の4匹のマウスの間で同等にプールし、gDtペプチドプール+GolgiPlug(BD Biosciences)で6時間刺激し、次いでラット抗マウスCD3e-BV421、ラット抗マウスCD4 PerCP-Cy5.5、ラット抗マウスCD8a APC-H7(BD Biosciences)およびLive/Dead染色ブルー試薬(Invitrogen)を用いて細胞表面マーカーについて染色した後、2%Cytofix(BD Biosciences)で固定した。細胞をBD Perm Wash 透過性バッファー(BD Biosciences)に再懸濁し、室温で30分間インキュベートした後、IFN-γ-APC、IL-2-PE、GM-CSFおよびTNF-αAlexa488(BD Biosciences)を含むカクテルで染色した。FACS DIVA Softwareを利用してBD Biosciences LSR2サイトメーターで取得(10万イベント/サンプル)を実施した。FLOWJOソフトウェア(TreeStar、Menlo Park、CA)を使用して、サブセットおよびブールゲーティング分析(多機能性T細胞について)を行った。図3において、データを、CD4−またはCD8−ゲートT細胞集団内の%サイトカイン陽性サブセットとして報告する。実施例6〜8の3つすべてが、多機能性CD4 T細胞の等価または優れた誘導を示した。実施例7よりも低いレベルであるが、実施例8のアジュバント調製物は、IFN-γ+ TNF-α+サブセットだけでなく、より高いパーセンテージのIFN-γ+ TNF-α+ IL-2+トリプル陽性CD4 T細胞を誘導した。図4は、CD3+ CD8+ T細胞集団でゲートした場合、実施例7はCD8+サイトカイン発現細胞の最大レベルの応答を誘導するが、大部分がIL−2+のみであるのに対し、実施例8は検出可能なIFN-γ+ IL-2+およびGM-CSF+ IFN-γ+サブセットを誘導することを示している。
28日目に末端血液サンプルから血清を単離し、ELISAによりHSV−gDt特異的免疫グロブリンについて分析した。HSV2−gDt抗原を用いてELISAプレートをコーティングし、個々のマウス由来の血清試料(1:50から開始する6つの1:3連続希釈物)とインキュベートしてgDt特異的IgGに結合させた。次いで、第2のステップとしてIgG、IgG1およびIgG2cに特異的なビオチン化検出抗体、次いでHRP−ストレプトアビジン、基質を使用して、ELISAプレートリーダーにて450nmで読み取りを行った。実施例7および8は、CpG/AddaVax群と同等のレベルの抗gDt IgG力価を誘導した(図5)。実施例7および8のいずれも、CpG/AddaVaxよりも高いIgG2c:IgG1サブタイプの比率を誘導し、強いTh1応答の誘導を示した。
8〜9週齢のBALB/cマウスを0日目および14日目に、実施例9でアジュバント添加した10μgのCMV pp65抗原を用い、i.m.にて免疫した(1群あたり5匹のマウス)。実施例9は、マウス1匹あたりに送達されるTLR7Lの用量が10μg(100μl中)になるよう投与した。比較のため、1つの群のマウスを、MF59と組成が同等である市販の水中油型スクアレンエマルジョンであるAddaVax(InVivogen)(20μgのCpG−Cオリゴデオキシヌクレオチド2395または20μgのCpG−Bオリゴデオキシヌクレオチド1826(どちらもInVivogen)を含むおよび含まない)中に50%v:vで処方したpp65抗原で免疫した。これらのCpG + AddaVax組成物は、マウスにおいて強いT細胞応答を誘導することが既に観察されており、陽性対照として用いた。単に可溶性TLR7Lをエマルジョンと混合する場合と、実施例9の方法により調整したようにTLR7Lをエマルジョンの油滴内に送達し統合した場合のアジュバント活性の比較として、1つの群のマウスを、実施例2のTLR7Lと単に混合した参考例11(「ME0」)で免疫した。21日目に、マウスを出血させて脾臓を採取した。
脾細胞をELISPOTプレートにてpp65ペプチド(pp65配列全体にまたがる11で重複する15マー)で24時間刺激した。スポット形成細胞をスキャンし、CTL ImmunoSpot Analyzer(Cellular Technologies Limited)を用いて分析した。図6では、実施例9(「ME7」)が、CpG/AddaVax製剤よりも高いIFN−γ応答を誘導した。さらに、可溶性TLR7L+参考例11の単純な混合物は、T細胞誘導応答が弱かった。各群の5匹のマウスの間で同様に脾細胞をプールし、抗CD4磁気ビーズ(Miltenyi)と結合させ、MACS(磁気活性化細胞ソーティング)を介してCD4 T細胞を除去して脾細胞集団(<1.0%CD4+であり、したがって主にCD8 T細胞を含有)を得た。これらの細胞集団は、CD8特異的T細胞応答(図7)を検出するために、ELISPOTアッセイでも用いた。実施例9は、CpG/AddaVax製剤と同等でTLR7L+参考例11の混合物よりも高いCD8 T細胞応答を誘導した。
脾細胞を各群内の5匹のマウスの間で同様にプールし、pp65ペプチドプール+Brefeldin A(BD Biosciences)で6時間刺激し、次いでラット抗マウスCD3e-BV510、ラット抗マウスCD4 APC-Cy7、ラット抗マウスCD8a BV711(BD Biosciences)およびLive/Dead染色ブルー試薬(Invitrogen)を用いて細胞表面マーカーについて染色した後、2%Cytofix(BD Biosciences)で固定した。細胞をBD Perm Wash 透過性バッファー(BD Biosciences)に再懸濁し、室温で30分間インキュベートした後、IFN-γ-BV605、IL-2-PerCP-Cy5.5およびTNF-α-BV421 (BD Biosciences)を含むカクテルで染色した。FACS DIVA Softwareを利用してBD Biosciences LSR2サイトメーターで取得(10万イベント/サンプル)を実施した。FLOWJOソフトウェア(TreeStar、Menlo Park、CA)を使用して、サブセットおよびブールゲーティング分析(多機能性T細胞について)を行った。図8において、データを、CD4−またはCD8−ゲートT細胞集団内の%サイトカイン陽性サブセットとして報告する。実施例9は、CpG/AddaVaxまたはTLR7L/参考例11製剤と比較して優れた多機能性CD4 T細胞の誘導を示した。図9において、CD3+ CD8+ T細胞集団でゲートした場合、実施例9はCpG/AddaVaxと同程度の多機能性CD8+サイトカイン発現細胞の応答を誘導する。
21日目に末端血液サンプルから血清を単離し、ELISAによりHSV−gDt特異的免疫グロブリンについて分析した。HSV2−gDt抗原を用いてELISAプレートをコーティングし、個々のマウス由来の血清試料(1:50から開始する6つの1:3連続希釈物)とインキュベートしてgDt特異的IgGに結合させた。次いで、第2のステップとしてマウスIgGに特異的なビオチン化検出抗体、次いでHRP−ストレプトアビジン、基質を使用して、ELISAプレートリーダーにて450nmで読み取りを行った。CpG−C/AddaVax、参考例11および実施例9はいずれも、最高レベルの全gDt特異的IgG力価を誘導した(図10)。可溶性TLR7L+参考例11の混合物は、統合された実施例9配合物と比較して準最適であった。血清試料もサブタイプIgG1およびIgG2aに特異的な二次抗体で分析した(図11)。強いTh1応答を示す高いIgG2a:IgG1比が、CpG/AddaVaxおよび実施例9によって達成されたが、TLR7L+AddaVaxでは達成されなかった。
ナイーブBALB/c脾細胞を用いて2種類の標的細胞集団を作製した。一方の集団には10μMのCFSE色素と2μg/mlのpp65重複ペプチドをロードし、他方の集団には1μMのCFSE色素と2μg/mlのEBV gp350重複ペプチド(無関係の抗原由来)をロードしてpp65特異的効果について正規化した。AddaVax中50%v:vで製剤化した20μgのCpGまたは実施例9のいずれかをアジュバント添加した10μgのCMV-pp65 Agで2回(0、14日目)免疫したマウスに、両方の標的集団を同等の割合で28日目に尾静脈内経路で注射した。投与したマウスを18〜24時間後に屠殺し、脾細胞を単離し、フローサイトメトリーでCFSEシグナル(明るい及び淡いCFSEシグナルを用いて標的集団を区別する)を分析した。式:%特異的死滅=(1−%CFSEhi細胞/%CFSElo細胞)×100によってgp350特異的標的死滅を対照として、pp65特異的標的死滅を正規化した。図12は、実施例9でアジュバント添加したpp65で免疫したマウスは、CpG + AddaVaxで免疫したマウスに匹敵する、pp65をロードした標的細胞の細胞傷害性を示したことを示す。
Claims (24)
- 式(1):
L1およびL2は、独立してアルキレンであり、
R1は、水素原子またはアルキルであり、
R2は、置換されていてもよいアルキルであり、
R3は、水素原子、ハロゲン原子、アルキルまたはアルコキシであり、
X1は単結合、酸素原子、硫黄原子、SO、SO2、NR4またはCONR4であり、
R4は水素原子またはアルキルであり、
Aは、単環の芳香族炭素環、または、1〜4個の窒素原子、1個の酸素原子および1個の硫黄原子から選択される、1〜4個のヘテロ原子を含む5もしくは6員の芳香族ヘテロ環であり、
mは0または1であり、
で示される、化合物またはその薬学的に許容される塩。 - L1は、C1−4アルキレンであり、
L2は、C1−4アルキレンであり、
R1は、水素原子またはC1−4アルキルであり、
R2は、置換されていてもよいC1−6アルキルであって、該アルキルが置換されている場合には、ヒドロキシ、ハロゲン原子、C1−6アルキル、C1−6ハロアルキル、C1−6アルコキシ、C1−6ハロアルコキシ、1もしくは2個の同一もしくは異なるC1−6アルキルで置換されていてもよいアミノ、およびカルボキシから選択される同一もしくは異なっていてもよい1〜4個の置換基で置換されており、
R3は、水素原子、ハロゲン原子、C1−4アルキルまたはC1−4アルコキシであり、
X1は、単結合、酸素原子、硫黄原子、SO、SO2、NR4またはCONR4であり、
R4は、水素原子またはC1−4アルキルである、請求項1記載の化合物またはその薬学上許容される塩。 - R2が、ヒドロキシ、ハロゲン原子、C1−6アルコキシ、C1−6ハロアルコキシから選択される、同一もしくは異なる1〜3の基で置換されていてもよいC1−6アルキルである、請求項1または2に記載の式(1)で示される化合物またはその薬学上許容される塩。
- Aが、ベンゼン環またはピリジン環である、請求項1〜3のいずれかに記載の式(1)で示される化合物またはその薬学上許容される塩。
- Aがベンゼン環である、請求項4に記載の式(1)で示される化合物またはその薬学上許容される塩。
- L2がメチレンである、請求項1〜5のいずれかに記載の式(1)で示される化合物またはその薬学上許容される塩。
- R2が、C1−4アルキル、C1−4ヒドロキシ−アルキルまたはC1−4アルコキシ−C1−4アルキルである、請求項1〜6のいずれかに記載の式(1)で示される化合物またはその薬学上許容される塩。
- L1は、C1−3アルキレンであり、R1は、水素原子またはC1−3アルキルである、請求項1〜7のいずれかに記載の式(1)で示される化合物またはその薬学上許容される塩。
- R1が水素原子またはメチルである、請求項1〜9のいずれかに記載の式(1)で示される化合物またはその薬学上許容される塩。
- L1は、メチレンであり、
L2は、メチレンであり、
R1は、水素原子またはメチルであり、
R2は、C1−6アルキル、C1−3アルコキシ−C2−4アルキル、または1〜4個のヒドロキシ基で置換されたC2−6アルキルであり、ここで2以上のヒドロキシ基は異なる炭素原子に結合しており、
R3は、水素原子、C1−3アルキル、C1−3アルコキシまたはハロゲン原子であり、
X1は、単結合、酸素原子、NR4またはCONR4であり、
R4は、水素原子またはC1−3アルキルであり、
Aは、ベンゼン環またはピリジン環であり、
請求項1に記載の式(1)で示される化合物またはその薬学上許容される塩。 - 請求項1〜11のいずれかに記載の式(1)で示される化合物またはその薬学上許容される塩を含む医薬組成物。
- 医薬組成物が、油成分としてのスクアレン、Span(登録商標)85(トリオレイン酸ソルビタン)およびポロキサマー188を含んでなる水中油型エマルジョンである、請求項12に記載の医薬組成物。
- 医薬組成物が、油成分としてのスクアレン、L−α−ホスファチジルコリンおよびポロキサマー188を含んでなる水中油型エマルジョンである、請求項12に記載の医薬組成物。
- 水中油型エマルジョンが含んでなる液滴の平均粒径が10〜1000nm±10nmである、請求項13または14に記載の医薬組成物。
- 更に抗原を含む、請求項12〜15のいずれかに記載の医薬組成物。
- 抗原が病原体由来抗原または腫瘍抗原である、請求項16に記載の医薬組成物。
- 抗原がペプチドまたはタンパク質である、請求項16または17に記載の医薬組成物。
- 請求項1〜11のいずれかに記載の化合物またはその薬学上許容される塩を含む、ワクチンアジュバント。
- 治療において使用するための、請求項1〜11のいずれかに記載の式(1)で示される化合物またはその薬学上許容される塩。
- ワクチンアジュバントとして使用するための、請求項1〜11のいずれかに記載の式(1)で示される化合物またはその薬学上許容される塩。
- 癌の治療または予防において使用するための、請求項1〜11のいずれかに記載の式(1)で示される化合物またはその薬学上許容される塩。
- 抗原の免疫賦活活性を向上させる方法であって、それを必要とする哺乳動物に有効量の請求項1〜11のいずれかに記載の化合物またはその薬学上許容される塩を投与することを含む、方法。
- ワクチンアジュバントの製造のための、請求項1〜11のいずれかに記載の化合物またはその薬学上許容される塩の使用。
