Abstract
Description
1)本発明の第5の態様のいずれか1つの宿主細胞を、培養培地に接種し、続いて段階的培養及び増殖を行い;
2)宿主細胞を収集し、組み換え細胞を培地中に再懸濁し、そしてメタノールを添加し、発現を誘導し;
3)誘導発現の完結の後、培養上清を収集し、そして精製し、血小板由来増殖因子Bタンパク質を得る、工程を含む。
(1)工程1)における宿主細胞は、モノクローナル細胞系であり;
(2)工程1)に記載される段階的培養及び増殖が二段階培養を意味し、培養温度が28〜30℃であり、そしてOD600が1〜12になるまで、例えば2〜6のOD600になるまで各工程において細胞が培養され;
(3)工程2)での発現を誘導するための温度は、約28℃であり;
(4)工程2)でのメタノールの最終濃度は、0.3〜1.0%(v/v)、例えば0.4〜0.8%(v/v)、例えば0.5%(v/v)であり;
(5)工程2)での発現を誘導するための時間は、48〜96時間、例えば72時間であり;
(6)工程3)での精製工程が、疎水性相互作用クロマトグラフィー、イオン変換クロマトグラフィー及びゲルろ過クロマトグラフィーを連続的に行うことを含む。
前記疎水性相互作用クロマトグラフィーは、以下の工程:
(1)血小板由来増殖因子B又はその変異体を含む培養上清又は細胞溶解物の導電率を、コンディショニング緩衝液で調節する工程、ここで前記コンディショニング緩衝液を加えた後の最終体系が10〜50mMリン酸緩衝液、0.8〜1Mの(NH4)2SO4、pH6.8〜7.5である;
(2)前記カラムクロマトグラフィーを平衡化緩衝液で平衡化する工程、ここで前記平衡化緩衝液の配合が10〜50mMリン酸緩衝液、0.8〜1Mの(NH4)2SO4、pH6.8〜7.5である;
(3)試料をカラム上にロードした後、カラムを平衡化緩衝液で洗浄する工程;
(4)溶出緩衝液で溶出し、目的のタンパク質を回収する工程、ここで前記溶出緩衝液の配合が10〜50mMリン酸緩衝液、30〜50%エチレングリコール、pH6.8〜7.5である;
を含み、
前記イオン交換クロマトグラフィーは、以下の工程:
(1)疎水性相互作用クロマトグラフィーの溶出ピークを平衡化緩衝液で6mS/cm以下の導電率となるように希釈する工程、ここで前記平衡化緩衝液の組成は10〜50mMリン酸緩衝液、pH6.8〜7.5である;
(2)カラムを前記平衡化緩衝液で平衡化する工程;
(3)試料をカラム上にロードした後、カラムを平衡化緩衝液で洗浄する工程;
(4)溶出緩衝液でグラジエントをかけて溶出し、目的のタンパク質を回収する工程、ここで前記溶出緩衝液の配合が10〜50mMリン酸緩衝液、0.8〜1.2mMのNaCl,pH6.8〜7.5である;
を含み、
前記ゲルろ過クロマトグラフィーは、以下の工程:
(1)カラムをリン酸緩衝液で平衡化する工程、ここで前記リン酸緩衝液の配合が10〜50mMリン酸緩衝液、0.1〜0.5mMのNaCl,pH6.8〜7.5である;
(2)前記イオン交換クロマトグラフィーの溶出ピークをロードする工程、ここで各ロード体積は、カラム体積の0.3〜4%以下である;
(3)前記工程(1)におけるリン酸緩衝液を用いてカラムの洗浄を継続し、目的のタンパク質を回収し、そして精製された血小板由来増殖因子B又はその変異体を得る工程、
を含む。

種々のPDGF変異体をコードするDNA配列は、Shanghai Sangon Inc.により合成された。その遺伝子フラグメントを、制限部位XhoI及びEcoRIを介して発現ベクターpMEX9K中にクローン化し(特許ZL02117906.9号を参照のこと)、そしてシーケンシングにより確認した。組換えプラスミドを抽出し、SalI消化により線状化し、そして次に、エレクトロポレーションにより、ピチア・パストリス(Pichia pastoris)発現株GS115コンピテント細胞を形質転換した。酵母形質転換体を、ヒスチジン欠損MDプレートによりスクリーニングし、そして陽性組換え酵母株を、PCRにより同定した。
組み換え酵母株の単一クローンを、25mlのBMGY培地のフラスコ中に接種し(BMGY培地は、次の通りにして調製した:10gの酵母抽出物粉末及び20gのトリプトンを計量し、700mlの水に溶解し、121℃で20分間オートクレーブ処理し;室温に冷却し、100mlの1Mリン酸カリウム緩衝液、100mlの10×YNB及び100mlの10×GYを添加し、そして4℃で貯蔵した。ここで、10×YNB(13.4%酵母窒素源ベース)、10×GY(10%グリセロール)、1Mリン酸カリウム緩衝液(132mlの1MのK2HPO4及び868mlの1MのKH2PO4を測定し、リン酸又はKOHにより、pH6.0±0.1に調整し、121℃で30分間オートクレーブ処理し、そして室温で貯蔵した)、酵母抽出物(LP0021)はOXOID Inc.の製品であり、そしてペプトン(211677)は、B&D Inc.の製品である)、220〜250rpm下で28〜30℃で、OD600=2〜6(約16〜18時間)まで培養し、そして増殖した。25mlの酵母培養物を、1LのBMGYを含むフラスコ中に接種し、そして220〜250rpm下で28〜30℃で、OD600=2〜6まで培養し、そして増殖し続けた。酵母を、1500〜3000gで5分間、室温での遠心分離により集めた。上清液を除去し、そして酵母を、1LのBMMY培地により再懸濁し、発現誘導を開始した。誘導温度は28℃であり、そして回転速度は220rPmであった。メタノールを、最終濃度が0.5%になるまで、24時間ごとに添加し、そして誘導時間は72時間であった。誘導の完結の後、組み換えタンパク質を含む上清液を、室温で、7000rpmでの遠心分離により集めた。
ピチア・パストリス(Pichia pastoris)の発現上清液を、遠心分離及びろ過により、適切な緩衝液中に調整し、そして疎水性相互作用クロマトグラフィー(Phenyl Sepharose 6 Fast Flow)、イオン交換クロマトグラフィー(Source 30S)及びゲルろ過クロマトグラフィー(Hiload Superdex 75 prep grade)を連続的に行い、95%以上の純度を有する目的のタンパク質を得た(図5)。クロマトグラフィー媒体は、すべて、GE Amersham Bioscience Inc.からの製品である。
適切な濃度を有する精製タンパク質30μlを、それぞれ、10μlの4×SDS−PAGE緩衝液(20mMのDTTを含む及びそれを含まない)に添加し、100℃で5分間、変性し、そして遠心分離した。次に、30μlの上清液を、SDS−PAGE電気泳動分析のために採取した(分離ゲルは15%である)。電気泳動に続いて、ゲルをクーマシーブリリアントブルーR250により染色した。
試料を、SDS−PAGEにおけるのと同じ手段で調製した。3μlの試料を、SDS−PAGEのために採取した。電気泳動に続いて、タンパク質を、300mAの定電流下で1時間、ニトロセルロースに移し、そして5%脱脂粉乳/TBSTにより室温で1時間ブロックした。一次抗体PDGF−B(F−3)(Santa Cruz Biotechnology, SC-365805)を、1:1000で希釈し、室温で1時間、被覆し、そしてTBSTにより数回、洗浄した。HRP標識された二次抗体(Cell Signaling Technology, #7076)を、1:10000で希釈し、室温で1時間インキュベートし、そしてTBSTにより洗浄した。基質を添加し、そしてLAS400ミニゲルイメージングシステム(GE)により映像化した。
試料調製及びSDS−PAGE工程は、上記と同じであった。電気泳動の完結に続いて、タンパク質を、300mAの定電流下で1時間、CAPSエレクトロブロッティング緩衝液によりPVDF膜に移し、そして0.1%クーマシーブリリアントブルーR250により染色し、この後すぐに、タンパク質バンドが見えるまで、50%メタノールにより十分に脱色した。決定されるタンパク質バンドを切断し、そして測定のために、Chromatography Laboratory of Biomedical Analysis Center, Military Medical Academyに送った。
50μlの試料及び陽性対照IFN−ωを、3μlの10×糖タンパク質変性緩衝液(NEB PNGアーゼF酵素を含む)及び15μlの水中に添加し、そして100℃で10分間、加熱変性した。冷却の後、3μlのNP−40、3μlのG7緩衝液(NEB PNGアーゼF酵素を含む)及び2μlのペプチドN−グリコシダーゼF(PNGase F)(New England Biotech Inc. (NEB)の製品)を添加し、そして100℃で加熱し、酵素を不活性化し、そしてSDS−PAGE電気泳動を行った。電気泳動の完結に続いて、染色を、糖タンパク質の染色キット(Themo Scientific, # 24562)を用いて実施した。最初に、ゲルを、100mlの50%メタノール中に添加し、そして30分間、固定し;ゲルを3%酢酸により数度、洗浄し、25mlの酸化溶液に移し、そして15分間、軽く振盪し;ゲルを3%酢酸により数度、洗浄し、25mlの糖タンパク質染色試薬に移し、そして15分間、軽く振盪し;その後、ゲルを25mlの還元溶液に移し、そして5分間、軽く振盪し;次に、ゲルを3%酢酸により洗浄し、そして脱イオン水によりすすいだ。
BALB/3T3細胞(Beijing Xiehe Cell Resource Centerから購入された)を、10%FBSを含むDMEM完全培地(Life Technology)において、37℃及び5%二酸化炭素の条件下で培養した。消化及び収集の後、細胞を、1mlの完全ブイヨン当たり5.0×104個の細胞を含む細胞懸濁液として調製し、96ウェル細胞培養プレート中に接種し(ウェル当たり100μl)、そして続いて、37℃及び5%二酸化炭素の条件下で培養した。24時間後、培地を、維持培地(0.4%FBSを含むDMEM)、続いて、37℃及び5%二酸化炭素の条件下で培養した。24時間の培養に基づいて、培養培地を捨て、そして予備勾酸希釈PDGF−BB溶液(ウェル当たり100μl)を添加した。細胞を、タンパク質の作用下で、さらに64〜72時間、培養し、そして次の通りに、WST−1方法により細胞増殖についてアッセイした:10μlのWST−1溶液(Roche, 11644807001)を、各ウェル中に添加し、37℃及び5%二酸化炭素の条件下で3時間、培養し、そして次に、マイクロプレートリーダー(参照の波長:630nm)を用いて、450nmの波長で吸光度を測定した。実験データを、4パラメーター回帰計算法により処理した。2つのタンパク質のEC50値を、それぞれ計算した。実験を3度、反復した。2つのグループ間の異なった統計学的分析を、t−検定により実施した。
本発明者は、最初に、タンパク質分解が多様なPDGF−Bモノマーの形成の原因であると疑った。還元SDS−PAGE電気泳動により、PDGF−Bの異なったモノマーを分離し、そして5バンドを、クーマシーブリリアントブルー染色を介して検出した(図3A)。5タンパク質バンドを、N末端アミノ酸配列についてのシーケンシングにかけ、そしてその結果は、第1、第2及び第3バンドにおけるN末端での最初の5個のアミノ酸残基がすべてTIAEPであり、これはPDGF−BThr6の正しいN末端配列に対応し、ところが第4及び第5バンドにおけるN末端での最初の5個のアミノ酸残基がTNANFであったことを示した。タンパク質配列のアラインメントは、第4及び第5タンパク質フラグメントが、Arg32−Thr33でのタンパク質分解性切断により生成された切断タンパク質であったことを決定した(図A)。
さらに、本発明者は、上記推論を確認したく、そしてピチア・パストリス(Pichia pastoris)におけるPDGF−Bの発現が均一であることを予測した。最初に、本発明者は、可能性あるO−グリコシル化部位を決定したいと考えた。PDGF−BThr6タンパク質配列のグリコシル化部位の予測を、オンラインウェブサイトCBS(www.CBS.dtu.dk)を用いて実施した(11)。結果は、位置6、101及び109でのThrが可能性あるO−グリコシル化修飾部位であることを示した(図4)。N−グリコシル化と比較すれば、O−グリコシル化部位についての明確なモチーフは存在せず、結果的に、その予測はまた、比較的困難である。しかしながら、位置6、101及び109での予測される可能なグリコシル化部位は、本発明者の結果と一致し、すなわちPDGF−BThr6のタンパク質のC末端でグリコシル化修飾(Thr101、Thr109)が存在するはずであり、なぜならば、それらは抗体への結合を妨げたからであり;Thr33の前にグリコシル化修飾が存在すべきであり、これは、消化されたPDGF−Bについてわずか1つ(バンド4)のグリコシル化修飾された変異体が存在したが、しかし消化されていない、グリコシル化されたPDGF−Bモノマーについて2つのバンド(バンド1、2)が存在した事実を説明している(図3B;表1)。予測される結果を確認するために、本発明者は、PDGF−BThr6の2つの変異体PDGF−M1及びPDGF−M2を構成した。C末端での2つのグリコシル化部位を、PDGF−M1において突然変異誘導し、そしてすべての3種の可能性あるグリコシル化部位を、PDGF−M2において突然変異誘導した(変異のパターンについては、図6Aを参照のこと)。また、プロテアーゼ切断部位Arg32−Thr33を除去するために、本発明者は、アミノ酸の進化的選択を考慮して、Arg32をProに変異させた。本発明者は、成熟PDGF−Bタンパク質がPDGF−Aと60%のアミノ酸配列相同性を有し、そして両者は構造及び機能に関して、高い類似性を有し、ところがPDGF−Aタンパク質のその対応する位置におけるアミノ酸がProであることに注目した。

グリコシル化部位Thr6−Ala、Thr101−Ala及びThr109−Alaの変異及びArg32−Pro KEX切断部位の変異がPDGF−BBの生物学的活性に影響を及ぼすかどうかを分析するために、本発明者は、WST−1方法を用いて、Balb/c 3T3細胞に対するPDGF−BThr6及びPDGF−M2の増殖活性を決定した。結果は、PDGF−BTh6のEC50が5.434±0.6475ng/mlであり、ところがPDGF−M2のEC50が3.492±0.4078ng/mlであることを示した。t−検定は、構築の後のタンパク質活性が、構築の前よりも高い(0.0117のP値を有する)ことを示した(図7)。
PDGF−M2の発現レベルを増強するために、本発明者は、オンラインツールJava(登録商標) Condon Adaptation Toolを用いて、タンパク質発現の間、ピチア・パストリス(Pichia pastoris)のコドン選択に従って、PDGF−M2(PDGF−IM−P)のコドン最適化を実施した。最適化されるDNAコード配列は次の通りである:

方法
組み換えPDGF−B野生型及びPDGF−M2変異体を、37℃で30分間、DTT(2.5mM)により還元し、そして緩衝液A(0.1%蟻酸を含む水溶液)により希釈し、続いて液体クロマトグラフィー及び質量分析(LC/MS)分析を実施した。タンパク質を、EASY−nLC システム (Thermo Fisher Scientific)を用いて、Easy−噴霧カラム(15 cm × 75 μm ID、 3−μm C18粒子)上で分離し、300nl/分の流速で、緩衝液Bの線形勾配(メタノール中、0.1%蟻酸の溶液を含む;0−90%、20分)により溶出した。高分解能スペクトルが、60,000の解像度、m/z350−1600の条件下で、Q Exactive Mass Spectrometer (Thermo Fisher Scientific)を用いて得られ、そしてXtractソフトウェア(Thermo Scientific)を用いて、デコンボリューションした。
高解像度LC/MSによるPDGF−B野生型及びPDGF−M2変異体の分析は、野生型PDGFに関して、6個までの炭水化物残基を含む異なったイソフォームが検出され、ここで3個の炭水化物残基を含むイソフォームの含有率が最高であったことを示した。また、グリコシル化は、PDGF−M2(M2)変異体についてはほとんど検出され得なかった(図9に示されるように)。
