Abstract
Description
本件は、2015年6月24日出願のGB1511125.5の優先権及びその権益を主張し、同出願の内容は、ここにその全体が参照により援用される。
(i)目標の場所において、成分を保持している非晶質の金属有機構造体を提供する工程と、
(ii)非晶質の金属有機構造体からの成分の放出を可能にし、それによって目標の場所において成分を提供する工程と
を含む方法が提供される。
本発明は、成分を保持する非晶質の金属有機構造体を提供する。非晶質の金属有機構造体は、成分を保持する結晶質の金属有機構造体から取得される又は取得可能である。したがって、結晶質の金属有機構造体の物理的及び化学的特色並びに非晶質化方法によって、非晶質の金属有機構造体の物理的及び化学的特色が決まる。
非晶質の金属有機構造体は、成分を保持する。成分は、構造体から放出可能である。成分は、一般に、被包し、その後放出することが望ましい又は有益である、有用な成分である。非晶質の金属有機構造体は、成分を保持し、目標の場所に送達して必要に応じて放出するのを可能にする送達システムとして有用となりうる。
非晶質の金属有機構造体は、結晶質の金属有機構造体から調製することができる。結晶質の金属有機構造体を非晶質化する方法は、当技術分野でよく知られている。
本発明は、成分を保持する非晶質の金属有機構造体を含む医薬組成物を提供する。ここで、成分は、医学的処置の方法において使用する活性薬剤である。非晶質の金属有機構造体は、場合により、薬学的に許容される1種又は複数の含有物と共に提供される。
本発明は、非晶質の金属有機構造体から成分を放出する方法を提供する。本発明者らは、成分の放出が、たとえば、結晶質の金属有機構造体からの成分の放出と比べて、制御され、持続し、比較的緩徐であることを見出した。
本発明の一態様では、処置の方法において使用するための非晶質の金属有機構造体が提供され、非晶質の金属有機構造体は、成分を保持し、成分は、医学的処置の方法において使用する活性薬剤である。同様に、本発明の医薬組成物も、処置の方法において使用するために提供される。
本発明の更なる態様では、目標の場所に成分を送達するための非晶質の金属有機構造体の使用が提供される。成分は、金属有機構造体によって保持され、金属有機構造体が目標の場所に提供されたなら、成分は、構造体から放出されうる。
(i)目標の場所において、成分を保持している非晶質の金属有機構造体を提供する工程と、
(ii)非晶質の金属有機構造体からの成分の放出を可能にして、それによって目標の場所において成分を提供する工程と
を含む方法が提供される。
上述の実施形態の両立できるありとあらゆる組合せを、ありとあらゆる組合せが個別かつ明示的に列挙されたかのように、本明細書において明示的に開示する。
計測装置
PXRDデータは、Bragg-Brentanoの幾何学的配置において、Cu Kα1用の主Geモノクロメーター及びSol-X固体状態検出器を備えたD8 Bruker回折計で収集した。収集条件は、2〜70°の2θ、ステップサイズ0.02°、15秒/ステップ、発散スリット0.2mm、受光スリット0.2mmとした。
ジルコニウム系MOFであるUiO-66(UiO=University of Oslo)[Zr6O4(OH)4(BDC)6](BDC=1,4-ベンゼンジカルボキシレート)を、成分を目標の場所に送達するための典型的な金属有機構造体として使用した。
化合物を細胞膜に不透過性にするその親水性の性質を考慮して、カルセインをモデル活性薬剤として選択した。加えて、カルセインは、蛍光分子であるので、共焦点顕微鏡法によって容易に追跡することができる。その自己消光特性のために、高い局所濃度のカルセイン(たとえば、金属有機構造体に吸着されているとき)は、検出することができない。したがって、カルセインは、検出されるとき、一般に、構造体からの薬剤の放出と関連付けられる。
カルセインローディングした結晶質のUiO-66(cal@UiO-66、0.2g)を、8mmのステンレス鋼ボールと共に、テンレス鋼広口瓶に入れた。次いで、Retsch MM200ミルを使用して広口瓶を20Hzで30分間振動させ、カルセインローディングした非晶質のUiO-66(cal@aUiO-66)を得た。
結晶質のUiO-66、カルセインローディングした結晶質のUiO-66(cal@UiO-66)、及びカルセインローディングした非晶質のUiO-66(cal@aUiO-66)を、粉末X線回折(PXRD)によって分析した。
生理的条件に似せるために、37℃のインキュベーターにおいて、オービタル撹拌しながら、pH7.4のリン酸緩衝食塩水(PBS、10mM)を使用して、カルセイン放出実験を行った。カルセインローディングした非晶質及び結晶質のUiO-66(3mg)を、合計体積10mLのPBSと共に、透析バッグ(MWCO 3,500、分子量カットオフDa、Medicell International社)に入れた。異なる時点で、1mLのPBSを採取し、1mLの新たなPBSと入れ替えた。紫外可視分光光度計を498nmで使用して、薬物の放出量を測定した。カルセイン放出の補正濃度は、式[2]によって示される。
10%(v/v)のウシ胎児血清(FBS)、2mMのL-グルタミン、100単位/mLのペニシリン、及び100μg/mLのストレプトマイシンを補充したグルコース高含有(4,500mg/L)ダルベッコ変法イーグル培地(DMEM)において、37℃、5%CO2で、HeLa細胞を維持した。細胞は、1週間に3回、2.8×104細胞/cm2の密度で(75〜80%の集密度で)継代した。
3-(4,5-ジメチルチアゾール-2-イル)-5-(3-カルボキシメトキシフェニル)-2-(4-スルホフェニル)-2H-テトラゾリウム(MTS)(Promega社、英国)還元検定を使用して、空の結晶質UiO-66の細胞毒性活性を調査した。実験前日、HeLa細胞を、96ウェルプレートに、ウェルあたり5×103細胞の密度で接種した。処理前に、細胞をPBSで2回洗浄した。
細胞浸透検定については、HeLa細胞を、NUNC(商標)画像処理用4ウェルプレートに、1.11×105細胞/mLの密度で接種し、DMEM中にて37℃、5%CO2で24時間インキュベートした。次いで、細胞をPBSで2回洗浄し、0.25mg/mLのカルセインローディングした結晶質のUiO-66(cal@UiO-66)又はカルセインローディングした非晶質のUiO-66(cal@aUiO-66)のいずれかと共に、24時間及び48時間インキュベートした。ローディング材料は、培地に十分に分散させてから、細胞を含有するウェルプレートに加えた。未処理細胞及び遊離カルセインを対照として含めた(0.013mg/mL)。Hoechst 33342(H33342)及びヨウ化プロピジウム(PI)(各5μg/mL)を、それぞれ、核の染色用及び生存度対照として使用した。
一連の別の非晶質構造体を調製した。そうした構造体には、それぞれが異なるジカルボキシレート有機配位子を有する、等細網状Zr系ファミリーのMOFが含まれた。異なる6種の配位子L1〜L6を使用しており、これらを以下に示す。
Zr-L2〜Zr-L4は、必要なリンカーL2、L3、又はL4(2.70mmol、1当量)、及びZrCl4(0.629g、2.70mmol、1当量)を、250mLの試薬びんに加えることにより合成した。DMF(60mL)及び塩酸(0.24mL)を加え、混合物を10分間音波処理した後、混合物を24時間120℃のオーブンに入れた。24時間後、びんを取り出し、室温に冷ました。生成物を遠心分離によって集め、DMF(30mL)及びアセトン(2×30mL)で洗浄した。試料を真空デシケーターに入れて乾燥させた。
上記実験において調製した金属有機構造体を、次いで、カルセイン、抗がん薬、又はsiRNAにローディングした。構造体にローディングするモデル成分は、カルセイン、並びにジクロロ酢酸(DCA)、α-シアノ-4-ヒドロキシケイ皮酸(α-CHC)、及び5-フルオロウラシル(5-FU)から選択される3種の抗がん薬の1種とした。
機械的及び温度非晶質化手順を使用して、成分を保持する非晶質の金属有機構造体を生成した。
典型的な実験では、ローディングした金属有機構造体(0.1g)を、8mmのステンレス鋼ボールと共に、ステンレス鋼広口瓶に入れた。次いで、Retsch MM200ミルを使用して広口瓶を20Hzで30分間振動させて、ローディングした非晶質の金属有機構造体を得た。
典型的な実験では、(ローディングしたNU-1000を除き、)40mgのローディングした金属有機構造体を、固体を湿らすのに十分な少量の水と共に、ガラスバイアルに入れた。次いで、試料を180℃で5時間加熱した。
薬物送達実験は、生理的条件に似せるために、37℃のインキュベーターにおいて、pH7.4のリン酸緩衝食塩水(PBS、10mM)を媒質として使用して、オービタル撹拌しながら実施した。
siRNA送達実験は、37℃のインキュベーターにおいて、オービタル撹拌しながら実施した。
HeLa細胞を前に記載したとおりに調製した。
金属有機構造体が細胞活力に及ぼす影響を、MTS還元検定又はアネキシンV検定のいずれかを使用して代謝活性を測定することにより明らかにした。
HeLa細胞を、NUNC(商標)画像処理用4ウェルプレートに、1.11×105細胞/mLの密度で接種し、10%(v/v)のウシ胎児血清(FBS)、2mMのL-グルタミン、100単位/mLのペニシリン、及び100μg/mLのストレプトマイシンで補充したDMEMにおいて、37℃、5%CO2で24時間インキュベートした。
HEK-293細胞を、24ウェルCorning細胞培養皿に、1.6×105細胞/mLの密度で接種し、10%(v/v)のウシ胎児血清(FBS)、2mMのL-グルタミン、100単位/mLのペニシリン、100μg/mLのストレプトマイシン、0.1mg/mLのハイグロマイシンB、及び0.015mg/mLのブラスチシジンで補充したDMEMにおいて、37℃、5%CO2で24時間インキュベートした。次いで、細胞をPBSで2回洗浄し、種々の条件のsiRNAと共にインキュベートした。陰性対照は、DMEM培地と共にインキュベートした。

Claims (23)
- 場合により薬学的に許容される1種又は複数の含有物と共に提供される、医学的処置の方法において使用する活性薬剤である成分を保持する非晶質の金属有機構造体を含む医薬組成物。
- 医学的処置の方法において使用するための、非晶質の金属有機構造体、又は前記非晶質の金属有機構造体を含む医薬組成物であって、前記非晶質の金属有機構造体は成分を保持し、前記成分は医学的処置の方法において使用する活性薬剤である、非晶質の金属有機構造体、又は前記非晶質の金属有機構造体を含む医薬組成物。
- 医学的処置の方法において使用するための、非晶質の金属有機構造体、又は前記非晶質の金属有機構造体を含む医薬組成物であって、前記非晶質の金属有機構造体は成分を保持し、前記成分は医学的処置の方法において使用する活性薬剤であり、前記処置の方法は前記非晶質の金属有機構造体からの前記成分の持続放出を含む、非晶質の金属有機構造体、又は前記非晶質の金属有機構造体を含む医薬組成物。
- 成分を目標の場所に送達する方法であって、
(i)目標の場所において、成分を保持している非晶質の金属有機構造体を提供する工程と、
(ii)前記非晶質の金属有機構造体からの前記成分の放出を可能にし、それによって前記目標の場所において前記成分を提供する工程と
を含む方法。 - 成分を保持する非晶質の金属有機構造体であって、前記成分が分子状ヨウ素(I2)でない成分、たとえば、分子量が少なくとも100である、成分を保持する非晶質の金属有機構造体。
- 前記金属有機構造体がジルコニウム含有金属有機構造体である、請求項1から5のいずれか一項に記載の医薬組成物、非晶質の金属有機構造体、又は方法。
- 前記ジルコニウム含有金属有機構造体がベンゼンジカルボキシレート配位子を有する、請求項6に記載の医薬組成物、非晶質の金属有機構造体、又は方法。
- 前記ベンゼンジカルボキシレート配位子が1,4-ベンゼンジカルボキシレート配位子である、請求項7に記載の医薬組成物、非晶質の金属有機構造体、又は方法。
- 前記金属有機構造体がUiO-66、NU-1000、又はNU-901、たとえばUiO-66である、請求項1から6のいずれか一項に記載の医薬組成物、非晶質の金属有機構造体、又は方法。
- 前記金属有機構造体がビスマス含有金属有機構造体である、請求項1から5のいずれか一項に記載の医薬組成物、非晶質の金属有機構造体、又は方法。
- 前記金属有機構造体がCAU-7である、請求項10に記載の医薬組成物、非晶質の金属有機構造体、又は方法。
- 前記成分が3個以上の原子を有する、請求項1から11のいずれか一項に記載の医薬組成物、非晶質の金属有機構造体、又は方法。
- 前記成分が1個又は複数の炭素原子を有する、請求項1から12のいずれか一項に記載の医薬組成物、非晶質の金属有機構造体、又は方法。
- 前記成分が、少なくとも255の分子量を有する、請求項1から13のいずれか一項に記載の医薬組成物、非晶質の金属有機構造体、又は方法。
- 前記非晶質の金属有機構造体が、前記成分を保持する結晶質の金属有機構造体を非晶質化することにより取得される又は取得可能である、請求項1から14のいずれか一項に記載の医薬組成物、非晶質の金属有機構造体、又は方法。
- 前記非晶質化が、前記成分を保持する結晶質の金属有機構造体のボールミル粉砕である、請求項15に記載の医薬組成物、非晶質の金属有機構造体、又は方法。
- 前記目標の場所が、細胞内、たとえばex vivoの細胞内にある、請求項4に記載の方法。
