I.定義

− エクソン80スキッピングに続くVII型コラーゲンの合成は、ウエスタンブロット法によって評価することができる(Titeuxら、2010年)。しかし、その開示の全体が参照により組み込まれるWO2013/053819(図1A)に記載されている通り、スキップされたrnRNAは、標的としたエクソン80のサイズが小さいために、見かけの分子量が野生型タンパク質と同様であるタンパク質を合成する。
− αlコラーゲン鎖のホモ三量体への正確な組み立ては、非還元条件下でのウエスタンブロット法によって証明することができる。VII型コラーゲンホモ三量体の正確なタンパク質分解パターンは、ペプシンおよびコラゲナーゼ消化に続いてのウエスタンブロット法によって評価することができる(Titeuxら、2010年)。
− コロイド金移動アッセイ。RDEB細胞は、正常ケラチノサイトおよび線維芽細胞に比べて増強された運動性を示すので、コラーゲンマトリックス上で、処置された細胞の細胞能動性をアッセイすることができる(Chenら、2002年)。

ペプチド核酸(PNA)とは、骨格が、デオキシリボース骨格と構造的に同形である、DNAの類似体であって、ピリミジン塩基またはプリン塩基を結合させた、N−(2−アミノエチル)グリシン単位からなる類似体である。天然のピリミジン塩基およびプリン塩基を含有するPNAは、ワトソン−クリック塩基対合則に従う相補的オリゴマーとハイブリダイズし、塩基対認識の点で、DNAを模倣する(Egholm、Buchardtら、1993年)。PNAのヌクレオシド間連結は、ホスホジエステル結合ではなく、ペプチド結合により形成されることから、アンチセンス適用(下記の構造を参照されたい)に良好に適する。骨格が非荷電である結果として、通常を超える熱安定性を呈示する、PNA/DNA二重鎖またはPNA/RNA二重鎖がもたらされる。PNAは、ヌクレアーゼによっても、プロテアーゼによっても認識されない。PNAサブユニットを含むPNAオリゴマーの非限定的な例を、下記:
アンチセンスオリゴマー化合物はまた、「ロックト核酸」サブユニット(LNA)も含有しうる。「LNA」とは、架橋核酸(BNA)と呼ばれる修飾のクラスのメンバーである。BNAは、リボース環のコンフォメーションを、C30−エンド(ノーザン)糖パッカー(sugar pucker)にロックする共有結合的連結を特徴とする。LNAでは、架橋は、2’−O位と4’−C位との間のメチレンから構成される。LNAは、骨格の事前組織化(preorganization)および塩基スタッキングを増強して、ハイブリダイゼーションおよび熱安定性を増加させる。

トリシクロDNA(tc−DNA)とは、各ヌクレオチドを、シクロプロパン環の導入により修飾して、骨格のコンフォメーション可撓性を制約し、ねじれ角γの骨格形状を最適化した、拘束DNA類似体のクラスである。ホモ塩基性のアデニン含有tc−DNAおよびチミン含有tc−DNAは、相補的なRNAと共に、極めて安定的なA−T塩基対を形成する。トリシクロDNAおよびそれらの合成については、参照によりその全体において本明細書に組み込まれる、PCT公開第WO2010/115993号において記載されている。本開示の化合物には、1または複数のトリシクロDNAサブユニットを組み込むことができ、場合によって、化合物は、トリシクロDNAサブユニットから完全に構成されうる。

(実施例1)
アンチセンスオリゴマーのデザインおよび製造
本開示のアンチセンスオリゴマーを、ヒトVII型コラーゲンプレmRNA転写物のエクソン80/イントロン80スプライス接合部にわたる標的領域に結合するようにデザインして、以下のプロトコールを使用して調製した。
脱トリチル化溶液:4:1ジクロロメタン/アセトニトリル中に、10%のシアノ酢酸(w/v);中性化溶液:3:1のジクロロメタン/イソプロパノール中に、5%のジイソプロピルエチルアミン;カップリング溶液:所望の塩基型および連結型を有する、0.18M(または20サブユニットより長くなったオリゴマーには、0.24M)の活性化モルホリノサブユニット、ならびに1,3−ジメチルイミダゾリジノン中に、0.4MのNエチルモルホリン。ジクロロメタン(DCM)を、異なる試薬溶液の洗浄を隔てる、一
過性洗浄液として使用した。
アンチセンスオリゴマーについてのin vitro研究
in vitro実験を実施して、上記で記載した通りにデザインおよび調製されたアンチセンスオリゴマーが、エクソン80を含有するヒトVII型コラーゲンプレmRNAの発現を減少させて、機能的なヒトVII型コラーゲンの発現を増加させる能力について探索した。言及した実験では、正常なヒト皮膚成体線維芽細胞(HDFa細胞;cat#C−013−5C、Cascade Biologics(Life Technologies;グランドアイランド、NY)および正常なヒト上皮成体ケラチノサイト(HEKa細胞;cat#C−005−5C、Cascade(Biologics、Life Technologies;グランドアイランド、NY)の両方を使用した。HDFa細胞またはHEKa細胞に、SGヌクレオフェクター溶液中またはP3ヌクレオフェクター溶液中のそれぞれに、10、3、1、および0.3μMのアンチセンスオリゴマーをヌクレオフェクトし、5%のCO2を伴う37℃で、終夜インキュベートした。全RNAを、細胞から単離し、プライマーである、DEB 79 FWD Set2(5’TAC CAG GAG AGC GTG GTA T 3’;配列番号25)と、DEB 83 REV Set2(5’ GTC CTG GAG GTC CTG TCT 3’;配列番号26)とを使用して、RT−PCRを実施した。エクソン80スキッピングパーセントを決定するように、LabChip Caliperを使用して、増幅された試料を解析した。GraphPad Prismにおける非線形回帰解析を使用して、EC50値を決定した。EC50値は、被験PMOが、50%のエクソンスキッピングを誘導する濃度を表す。

該被験体に、有効量の、請求項1から7のいずれか一項に記載のアンチセンスオリゴマー化合物または請求項8から20のいずれか一項に記載の化合物を投与するステップを含む、方法。 - 表皮水疱症および関連障害が、劣性型栄養障害型表皮水疱症、優性型栄養障害型表皮水疱症、Hallopeau−Siemens型栄養障害型表皮水疱症、非重症型の劣性栄養障害型表皮水疱症、反対型の劣性栄養障害型表皮水疱症、求心型の劣性栄養障害型表皮水疱症、非Hallopeau−Siemens型栄養障害型表皮水疱症を含む、請求項22に記載の方法。

- 前記配列が、前記標的領域内の少なくとも12連続ヌクレオチドに相補的なターゲティング配列を含み、該ターゲティング配列は、配列番号2、3、4、または6から選択される、請求項24に記載のアンチセンスオリゴマー化合物。
- 前記配列が、前記標的領域に対する、少なくとも90%の配列相補性を含み、前記ヌクレオチドまたはヌクレオチド類似体が、配列番号2、3、4、または6から選択される、請求項24に記載のアンチセンスオリゴマー化合物。
- 機能的なヒトVII型コラーゲン発現の増加を必要とする被験体において、機能的なヒトVII型コラーゲン発現を増加させるための方法であって、
ヒトVII型コラーゲンプレmRNA転写物中の標的領域に対するアンチセンスオリゴマーを投与するステップと、
該アンチセンスオリゴマーを、該ヒトVII型コラーゲンプレmRNA転写物中の該標的領域に結合させるステップと、
を含み、機能的なヒトVII型コラーゲンの発現が増加する、方法。 - 前記ヒトVII型コラーゲンプレmRNA転写物中の前記標的領域が、ヒトVII型コラーゲンプレmRNAのエクソン/イントロン接合部にわたっており、該接合部は、エクソン80/イントロン80のスプライス接合部を含む、請求項28に記載の方法。

- 前記修飾糖部分が、ペプチド核酸(PNA)サブユニット、ロックト核酸(LNA)サブユニット、2’O,4’C−エチレン架橋核酸(ENA)サブユニット、トリシクロDNA(tc−DNA)サブユニット、2’O−メチルサブユニット、2’O−メトキシエチルサブユニット、2’−フルオロサブユニット、2’−O−[2−(N−メチルカルバモイル)エチル]サブユニット、またはモルホリノサブユニットのうちの少なくとも1つを含む、請求項31に記載の方法。
- 前記ターゲティング配列が、配列番号2、3、4、もしくは6から選択される配列を含むか、配列番号2、3、4、もしくは6から選択されるか、配列番号2、3、4、もしくは6から選択される配列のうちの少なくとも12連続ヌクレオチドの断片であるか、または配列番号2、3、4、もしくは6から選択される配列に対する、少なくとも90%の配列同一性を有する改変体であり、配列中、Xが、ウラシル(U)またはチミン(T)から選択される、請求項28に記載の方法。
- 前記ターゲティング配列が、配列番号2、3、4、または6から選択される、請求項28に記載の方法。
- 前記アンチセンスオリゴマーによって、機能的なヒトVII型コラーゲンの前記発現が約5%、6%、7%、8%、9%、10%、11%、12%、13%、14%、15%、16%、17%、18%、19%、20%、25%、30%、35%、40%、45%、50%、55%、60%、65%、70%、75%、80%、85%、90%、95%、または100%増加する、請求項28に記載の方法。
- 機能的なヒトVII型コラーゲンの発現を増加させるための方法であって、
ヒトVII型コラーゲンプレmRNA転写物中の標的領域に結合するアンチセンスオリゴマーを投与するステップと、機能的なヒトVII型コラーゲンの翻訳を増加させるステップとを含む方法。 - 機能的なヒトVII型コラーゲン発現が、約5%、6%、7%、8%、9%、10%、11%、12%、13%、14%、15%、16%、17%、18%、19%、20%、25%、30%、35%、40%、45%、50%、55%、60%、65%、70%、75%、80%、85%、90%、95%、または100%増加する、請求項37に記載の方法。
- 前記アンチセンスオリゴマーが、ターゲティング配列を含み、該ターゲティング配列が、配列番号2、3、4、もしくは6から選択される配列を含むか、配列番号2、3、4、もしくは6から選択されるか、配列番号2、3、4、もしくは6から選択される配列のうちの少なくとも12連続ヌクレオチドの断片であるか、または配列番号2、3、4、もしくは6から選択される配列に対する、少なくとも90%の配列同一性を有する改変体であり、配列中、Xが、ウラシル(U)またはチミン(T)から選択され、Iがイノシンである、請求項37に記載の方法。

ヒトVII型コラーゲンプレmRNA転写物中の標的領域に結合するアンチセンスオリゴマーを投与するステップと、機能的なヒトVII型コラーゲンの翻訳を増加させるステップとを含む方法。 - 栄養障害型表皮水疱症および関連障害を処置するための医薬であって、

ヒトVII型コラーゲンプレmRNA転写物中の標的領域に結合するアンチセンスオリゴマーを投与するステップと、機能的なヒトVII型コラーゲンの翻訳を増加させるステップとを含む方法。 - 栄養障害型表皮水疱症もしくはその関連障害を有する被験体において、係留線維における機能的なヒトVII型コラーゲンの蓄積を増加させる、栄養障害型表皮水疱症もしくはその関連障害を有する被験体において係留線維の蓄積を増加させる、または栄養障害型表皮水疱症もしくはその関連障害を有する被験体において、係留線維における機能的なヒトVII型コラーゲンの蓄積、および係留線維の蓄積を増加させるための方法であって、
ヒトVII型コラーゲンプレmRNA転写物中の標的領域に結合する、有効量のアンチセンスオリゴマーを投与するステップと、
機能的なヒトVII型コラーゲンの翻訳を増加させるステップと
を含み、1)該被験体における、係留線維における機能的なヒトVII型コラーゲンタンパク質の蓄積が増加する、2)該被験体における係留線維の蓄積が増加する、または3)該被験体における、係留線維における機能的なヒトVII型コラーゲンタンパク質および係留線維の蓄積が増加する、方法。
