定義
ある特定の実施形態では、可視染料(例えば、可視スペクトルにおける染料、例えば、緑色染料、例えば、FITC)を、局所的にまたはIVにより投与することができて、神経を、蛍光性シグナルによって見るために使用することができる。ある特定の実施形態では、ペプチドが結合した可視染料が、神経組織に優先的に付着して、染料からの光を、外科医は自分の視界で見ることができる。例えば、製剤は、神経自体に局所的にまたは神経の近くに局所的に適用することができて、例えば、製剤は、末梢神経幹(例えば、外科的露出後、転移性リンパ節または原発性腫瘍に隣接するかまたはその近傍にある)に局所的に適用することができて、製剤の組成物が神経幹から神経組織のさらに小さい枝に拡散することを可能にする。光は、組成物の蛍光剤により放射されて、検出されて表示され得るか(例えば、リアルタイムで)、または十分に明るくて、手術手順中に外科医が補助なしで自分の視力で直接観ることができる。神経組織への局所適用(およびそれに続く神経組織を通る拡散)は、血液タンパク質(例えば、ヘモグロビン)からのバックグラウンドシグナルが、製剤の静脈内投与と比較して減少されるので、より大きいコントラストを提供することができる。
線状NP41(図1A)およびその環状類似体(図1B)を、標準的Fmocベースの固相ペプチド合成(SPSS)プロトコルを使用してクロロトリチル樹脂上で合成した。線状ポリペプチドは、TFA:TIS:EDT:水(85:5:5:5)のカクテルを用いてペプチド−樹脂を開裂/脱保護し、続いて逆相HPLC精製により得た。環状類似体を調製するために、N末端のFmoc基を除去して、次に、完全に保護された線状ポリペプチドを、ヘキサフルオロイソプロパノールを使用する緩和な条件下で樹脂から切断した。頭尾結合環状類似体を、N末端残基とC末端残基を溶液中で連結して所望の環状前駆体を提供する分子内カップリング反応により得た。次に、粗材料を全体的に脱保護して逆相HPLCにより精製した。この合成手法を使用して、所望の51員の大環状のペプチド生成物が容易に得られた。このサイズの環状ペプチドは、合成的に難題であることが、注目されるべきである。しかしながら、ここで使用した手法は、優れた純度(例えば、95%超)および良好な収率(約40%)で環状生成物を提供した。線状および環状両方のペプチドを、システイン残基の遊離チオールをマレイミド−Cy5で改変することにより蛍光で標識した。最終生成物を特徴付けして、LCMSにより図1Aから1Dに示したように確認した。

- 請求項1から22のいずれか一項に記載の組成物を含む製剤を対象に投与し、前記組成物を前記対象の神経組織に選択的に結合させること;
前記対象の組織を励起光に曝露すること;および
前記組成物の前記蛍光剤により放射される光を検出して画像を創り出し、前記画像を表示すること
を含む撮像方法。 - 前記神経組織から放射される光が、周囲の組織から放射される光より強く(例えば、神経対筋肉のシグナル比は少なくとも2である)、その結果、前記神経組織が前記周囲の組織から視覚的に識別可能である、請求項23に記載の撮像方法。
