(2)Fc結合性タンパク質が、
(i)配列番号1に記載のアミノ酸配列のうち、少なくとも17番目のグリシンから192番目のグルタミンまでのアミノ酸を含むタンパク質、または
(ii)配列番号1に記載のアミノ酸配列のうち、少なくとも17番目のグリシンから192番目のグルタミンまでのアミノ酸を含み、かつ前記アミノ酸のうちの一つ以上が他のアミノ酸に置換、挿入または欠失したタンパク質である、
(1)に記載の方法。
(i)配列番号1に記載のアミノ酸配列のうち少なくとも17番目のグリシンから192番目のグルタミンまでのアミノ酸を含むタンパク質や、
(ii)配列番号1に記載のアミノ酸配列のうち少なくとも17番目のグリシンから192番目のグルタミンまでのアミノ酸において特定位置におけるアミノ酸置換が生じたタンパク質、
があげられる。
を発現し得る条件であれば特に限定はないが、培養温度は10から40℃が好ましく、特に好ましい温度は20から33℃である。pHは6から8が好ましい。培養時間は任意に設定できるが、通常は数時間から200時間の間に設定される。
Fc結合性タンパク質を発現可能な大腸菌(特開2016−23152)を、細胞密度が136に達した時、培養温度を25℃に下げ、IPTGを終濃度で0.83mMとなるように培養液に添加することでFc結合性タンパク質の生産誘導を行なった。
Fc結合性タンパク質を発現可能な大腸菌を、細胞密度が100に達した時、すなわち培養開始から18.8時間後に培養温度を25℃に下げ、IPTGを終濃度で0.83mMとなるように培養液に添加した以外は実施例1と同様な方法でFc結合性タンパク質の生産誘導を行なった。
Fc結合性タンパク質を発現可能な大腸菌を、細胞密度が117に達した時、すなわち培養開始から24.2時間後に培養温度を25℃に下げ、IPTGを終濃度で0.83mMとなるように培養液に添加した以外は実施例1と同様な方法でFc結合性タンパク質の生産誘導を行なった。
その結果、培養開始から58時間培養を行なったところ、培養液の濁度は207に達した。この時、供給した炭素源、窒素源、金属塩の混合液量は601gであった。また、培養終了後(培養開始から58時間)の培養液に含まれるFc結合性タンパク質の生産量はHPLCを用いた定量結果より、培養液1Lあたり3.2gであった。
Fc結合性タンパク質を発現可能な大腸菌を、細胞密度が157に達した時、すなわち培養開始から28.9時間後に培養温度を25℃に下げ、IPTGを終濃度で0.83mMとなるように培養液に添加した以外は実施例1と同様な方法でFc結合性タンパク質の生産誘導を行なった。
Claims (2)
- Fc結合性タンパク質をコードするポリヌクレオチドを含む発現ベクターで形質転換して得られた大腸菌を使用してFc結合性タンパク質を生産する方法であって、細胞密度(濁度、OD600nm、600nmにおける吸光度)が120から160の状態で誘導することによりFc結合性タンパク質の生産を開始する製造方法。
- Fc結合性タンパク質が、
(i)配列番号1に記載のアミノ酸配列のうち、少なくとも17番目のグリシンから192番目のグルタミンまでのアミノ酸を含むタンパク質、または
(ii)配列番号1に記載のアミノ酸配列のうち、少なくとも17番目のグリシンから192番目のグルタミンまでのアミノ酸を含み、かつ前記アミノ酸のうちの一つ以上が他のアミノ酸に置換、挿入または欠失したタンパク質である、
