ペプチド又はポリペプチド
ペプチド又はポリペプチドは、ペプチド結合により結合したアミノ酸残基からなる高分子である。用語ペプチド及びポリペプチドは、ペプチドは通常より短い配列を指す(一般的に、アミノ酸残基50個未満、又は30個未満、又は20個未満)一方、ポリペプチドは通常より長い配列を指すが(一般的に、アミノ酸残基20個、30個、又は50個を超える)、本明細書では交換可能に用いられる。タンパク質という用語は、本明細書では、用語ポリペプチドと交換可能に用いられる。アミノ酸残基は、20種類の天然起源のアミノ酸、及び/又は当業者にとって公知の非天然起源のアミノ酸の中から選択され得る。アミノ酸残基は、天然起源の修飾又は非天然起源の修飾により修飾され得る。天然起源の修飾として、特にセリン、トレオニン、及び/又はチロシン残基のリン酸化、N-結合型及びO-結合型グリコシル化を含むグリコシル化、特にリジン残基のユビキチン化、SUMO化又はユビキチン様タンパク質によるその他の修飾等が挙げられる。

VHH及びVHH含有ポリペプチドの配列。
VHH A12と命名された、配列番号3の配列を備えるVHHは、VHH免疫ライブラリから選択された。VHH免疫ライブラリは、1997年にGhahroudiらが記載するように、依然に構築されていた。アルパカ1頭を、アルツハイマー病患者から単離されたヒト脳海馬抽出物で免疫化した。この抽出物は、異なるタンパク質を含有する。循環性Bリンパ球から抽出されたmRNAを逆転写した。従来型の抗体ではなく、VHHのみをコーディングするcDNAを選択するために、mRNAをFR1及びCH2領域に相補的なプライマーを活用して増幅した。VHH配列を、ファージ外被タンパク質III(pIII)と融合した状態で、ファージミドpHEN1にクローン化し、そして該ファージミドを用いて病原性大腸菌TG1を形質転換した。組換えVHHの発現を、IPTGで誘発した。ファージディスプレー技術によりパニングした後、個々のコロニーを選択、培養し、試験した。特定のVHHを選択したら、細菌培養物について、NcoI及びNotIにより、37℃で3時間、消化工程を実施した。消化生成物をアガロースゲルにロードし、プラスミドpHEN1ベクターからインサートを分離するために、電気泳動を180Vで45分間実施した。VHH配列に対応する400bpの分画を回収し、NcoI及びNotIを用いて消化しておいた別のプラスミドpASK-Iba2とのライゲーションを16℃で、一晩実施した。ライゲーションの場合、プラスミド/インサートの比は1/5であった。ウルトラコンピテントXL2細菌(Agilent technologies社)を、β-メルカプトエタノールを添加して最初に処理し、そして2分毎に軽く旋回させながら、4℃で10分間インキュベートした。DNA(30ng)、1μLを添加し、4℃で30分間インキュベートした。ヒート-パルスを42℃で30秒間実施し、細菌を、次に氷上で2分間インキュベートした。500μLの2YTを添加し、培養物を、37℃で1時間、撹拌しながらインキュベートした。200個の培養物をペトリ皿上に播き、37℃で、一晩インキュベートした。同操作は、細菌が良好に形質転換されるよう管理するために実施した。事前培養物を、2YT + A内、30℃で、一晩インキュベートした。事前培養物、1mLを100mLの2YT + Aを播種するのに用いた。OD550が0.5に等しくなったら、アンヒドロテトラサイクリン(最終濃度:200ng/mL)、10μLを添加し、そして20℃で、一晩振とうさせた。VHHが、細菌周辺細胞質内に発現し、それは、C-末端領域にストレプ-タグを備えていた。タグを活用して、製造業者の推奨に基づき、アフィニティークロマトグラフィーカラム、ストレプ-タクチンセファロース(Iba)を用いて、VHHを細菌抽出物から精製した。サンプルの純度を管理するために、ポリアクリルアミドゲル電気泳動も実施可能であった。タンパク質を、クーマシーブリリアントブルーを用いて、撹拌しながらRTで1時間着色することにより可視化した。次に連続した水浴により脱色を実施した。アルツハイマー病患者から得られた脳抽出物(Sg τ 4697)に対応する1つの免疫ファージディスプレーライブラリ、Intiを用いて選択を実施した。Sg τ 4697スクリーニングにより単離されたクローン、61例の中から、2例のクローン、VHH-A12及びVHH-E8が大腸菌に発現し(図1)、更に特徴づけを行った。

ニューロカーゴ-ニューロヴィータ分子のクローニング及び発現
ニューロカーゴ-ニューロヴィータ遺伝子配列(VHH1:配列番号12、VHH2:配列番号14、VHH3:配列番号16)を、Eurofin MWG Operon社により化学的に合成し、そしてテトラサイクリンプロモーターのコントロール下で、pASK-IBA2プラスミド(IBA、BioTAGnology社、米国)に挿入した。このプラスミドでは、ompAシグナル配列が発現したタンパク質を細胞膜周辺腔内に誘導し、転位プロセス期間中に切り取られる。細胞膜周辺腔では、適切に折り畳まれたジスルフィド結合形成タンパク質が、見出され得る。組換えプラスミドは、XL1-Blue病原性大腸菌(Stratagene社、米国)を形質転換するのに用いた。組換え細菌を、PCRにより同定し、グリセロールストックを作成した。P.Lafaye(2008年)の記載に従い、VHH含有ポリペプチドを発現させ、細菌周辺細胞質から精製した。或いは、タンパク質は、細胞質から直接抽出することも可能である。周辺細胞質抽出物を、マウス高アフィニティー抗ストレプタグmAb、例えばC23.21(P. Lafaye、Institut Pasteur biological collection)等を用いて、イムノアフィニティークロマトグラフィーにより更に精製した。VHH含有ポリペプチドの発現を、mAb C23.21を用いたウェスタンブロッティング法によりモニターした。
VHH A12ニューラルの特徴付け
VHH A12は、下記の特徴を有することが明らかとなった:塩基性のpI(すなわち、pI>8.5;特にVHH単独の場合9.86、及びVHH1ポリペプチドの場合10.36);ウェスタンブロッティング法により、ヒト脳抽出物(すなわち、Sg τ4697)、及びマウス脳抽出物(すなわち、Tg 4510)、及びGFAP(すなわち、グリア線維性アストロサイトタンパク質)に対して無反応性;及び免疫組織化学検査により、Tg 4510マウス脳組織について免疫無反応性。
神経突起の伸長
神経突起伸長アッセイ法は、Prehaudら、2010年に広範に記載されている。ニューロスクリーン細胞(neuroscreen cell)(NS細胞)は、pC12サブクローンである。製造業者の説明書の記載に従い(http://www.cellomics.com/content/menu/Neuroscreen-1_Cells/)、RPMI培地内で細胞を増殖させた。NS細胞を72時間モニターし、time=0において200ng/mlのNGF(1ヒットのみ)、及びニューロカーゴ-ニューロヴィータ(10pg、time=6において、1ヒットのみ)を用いて処理した。神経突起の伸長(NO)は、SH-SY5Yに関する記載と同様に、基本的に認められたが、但し、NOは感染後72時間においてモニターされ、またNS細胞はそのフィーディング培地(Cellomics社、米国)内で常に増殖した点を除く。
スクラッチアッセイ法(軸索再生)
スクラッチ誘発式のアッセイ法では、200mm2のNT2-N細胞(n=8)を、ポリ-D-リジン-ラミニンがコーティングされた細胞+(Sarstedt社、ドイツ)12ウェルプラスチックウェア上に播種し、そしてトリプシン処理後完全に回復させるために、2日間増殖させた。培地を、スクラッチング前4時間において変更し、10pgのニューロカーゴ-ニューロヴィータを、1ウェル毎に添加した。個々の創傷は、注射針(26GX1/2"、12-4.5)で作成した。少なくとも10回のスクラッチングをウェル毎に行った。或いは、ニューロカーゴ-ニューロヴィータを、スクラッチング前1時間において添加した。受傷後6日目に、細胞をPFA (4%)で固定し、クリスタルバイオレット溶液で染色した。細胞をDC 300FXカメラ(対物×20)を備えたLeica DM 5000B顕微鏡を用いて画像化し、そしてImageJ 1.38Xソフトウェア(Wayne Rasband、NIH、米国、http://rsb.info.nih.gov/ij/)及びそのプラグ-インNeuronJを用いて分析した。平均ニューロン再生割合(%)を8回の実験から求めた。
AchR結合及び、フローサイトメトリーによりモニターした競合アッセイ、ニューロカーゴ-ニューロヴィータのNS細胞への進入
採用したフローサイトメトリー手順は、Prehaudら、2003年に記載されている。要するに、α7 AchR発現HEK 293細胞(Yamauchiら、2011年)、及び親のHEK 293細胞を10%のFBSを加えたDMEM high glucose(Life Technologies社、フランス)内で増殖させる。HEK 293細胞又はAchR発現HEK 293細胞を6ウェルプレートに播種した(1ウェル当たり1.e+06個の細胞)。播種後24時間において、細胞を氷上に配置し、染色バッファーを用いてフローサイトメトリー用に処理する前に、ニューロカーゴ-ニューロヴィータ、20pgで30分間処理した。或いは、競合アッセイの場合、細胞を、狂犬病ウイルス-CVS系統のU.V.不活性化された2.e+07個のPFU(Megretら、2005年の記載に従い)又は16μM αブンガロトキシン(Sigma社、米国)を用いて処理した。ニューロカーゴ-ニューロヴィータ分子の進入をモニターするために同一の実験を実施したが、但し、透過化処理バッファーを用いて、受容体に結合した分子及び細胞質内の分子の両方の検出を可能にした。細胞質の膜受容体に結合したニューロカーゴ-ニューロヴィータ分子を、Cy5結合抗マウス抗体(Jackson Laboratory社、米国)を加えた上記抗ストレプタグmAbを用いて検出した。
ニューロン細胞(NS)成長円錐の運動性測定
NS細胞を、time=0において200ng/mlのNGF(1ヒットのみ)、及びニューロカーゴ-ニューロヴィータ(10pg、time=6において、1ヒットのみ)を用いて分化させた。コントロールとして、細胞を、ニューロカーゴ-ニューロヴィータを発現しない細菌の細菌性周辺細胞質抽出物で処理した。48時間目に、細胞を、PFA固定化し、0.3%のトリトンX100で透過化処理し、そして免疫蛍光検査用にすでに記載された通り(Prehaudら、2010年)処理した。核を、Hoescth 33342、マウス抗β3チューブリン抗体を含むβIIIチューブリン(G7121、Promega社、フランス)、及び抗マウスAlexa 488抗体(Jackson laboratory社、米国)、及びAlexa 546結合ファロイジンを含むFアクチンネットワーク(A22283、Life Technologies社、フランス)を用いて検出した。
細胞遺伝子発現のリアルタイムPCR分析
RT-PCR遺伝子発現分析手順は、Prehaudら、2005年に広範に記載されている。ここでは、アッセイで用いた細胞培養プラスチックに起因して、cDNA分子は、200ng〜1μgの範囲のRNAを用いて作成した。この試験で用いられるプライマーは:TH(HSTH1SG、Qiagen社、ドイツ)、GFAP(F:CTGCTTCTTAACCCCAGTAAGCCA(配列番号39)、R:CAGCAGTGCCCTGAAGATTAGCAG(配列番号40))、AQP4(F:GGTATAGTCAATTCTTATTTGAAT(配列番号35)、R:CTTGAATCTCAATAGGTGCCCTTA(配列番号36))、PYGB(F:TCCTGCTGTGTCCTGAGGTGCATT(配列番号43)、R:GCCCAGATCCAGCATGCAAGGTGC(配列番号44))、NEFH(F:CCCCAGGCGATGGACAATTATGAT(配列番号41)、R:CACTTGGTTTTATTGCACAGAAGC(配列番号42))、CD200R(F:TTAACACTTCATGGCCTGTAAGA(配列番号37)、R:TGTGCCATTGCTCCAGTATTCTTG(配列番号38))、及びドイツ、Qiagen社製(HSSLC2A1-1、HSSLC7A5、HSSLC1A1-1、HSSLC38A5-1、HSSLC16A1、HSSLCO1C1、HSSLCABCB1-1、HSABCG2-1、HSABCC1-1、HSABCC2-1、HSABCC4-1、HSABCC5-1、HSLDLR-1、HSLRP1-1、HSINSR -1、HSLEPR-1、HSPVLAP-1、HSLU-1、HSTFRC-1、HSAGER1、HSSTRA6-1)である。
ヒトBBBのin vitroモデルの構築
ヒト脳微小血管内皮細胞株(hCMEC/D3)の不死化を、Wekslerら、2005年により実施した。hCMEC/D3を、ウシ胎仔血清(5%、Eurobio社、フランス)、ヒドロコルチゾン(1.4μM、Sigma社、米国)、bFGF(1ng/mL、Sigma社、米国)、アスコルビン酸(5μg/mL、Sigma社、米国)、化学的に定義された脂質(1/100、Life technologies社、フランス)、HEPES(10mM、Life technologies社、フランス)、ペニシリン-ストレプトマイシン(Life technologies社、フランス)が補充されたEBM-2(Lonza社、スイス)中で増殖させた。細胞を、ラット-テールコラーゲンI(150μg/mL、R&D systems社、英国)でコーティングされた培養フラスコ(コーニング社、米国)上でルーチン的に培養し、1週間に2回スプリットした(トリプシン処理による)。分化した単層を、ラット-テールコラーゲンI-(150μg/mL、R&D systems社、英国)、及びフィブロネクチン(10μg/mL、Sigma社、米国)でコーティングされたポリエステルトランスウェル(直径12mm、ポアサイズ0.4μm、コーニング社、米国)上で得た。細胞を細胞25,000個/cm2の密度で播種し、5日間培養した。培養培地を、分化3日目に変更した。5日目に、in vitro内皮性血液脳関門を、デバイスの下部コンパートメントに播種されたミニブレイン細胞と共に培養した(すなわち、ミニブレイン-BBB)、或いは培地単独中に置かれた(BBB)。従って、ミニブレイン-BBBモデルは、内皮細胞に付加して、ニューロン、アストロサイト、及びミクログリア細胞を含む一方、BBBモデルは、内皮細胞のみを含む。
ナノボディーのミニブレイン-BBB横断及びニューロカーゴ-ニューロヴィータの横断及び標的化
1.5%のミクログリア細胞を含有するミニブレイン-BBBデバイスを播種した。播種後1日目に、Alexa 488結合抗GFAP VHH(VHHE9、Liら、2012年)を、上側チャンバーにアプライした(ナノボディー、10pg)。3及び24時間後、下部チャンバー内の細胞をPFA固定化し、0.3%のトリトン X100を用いて透過化処理した。Cellinsight(Cellomics社、米国)を用いて、写真をランダムに撮影した(>500)。蛍光焦点をモニターした。
ルシファーイエローによる限定的な傍細胞透過性の測定
hCMEC/D3細胞の限定的な傍細胞透過性を、その低透過性を非透過性蛍光マーカーであるルシファーイエロー(LY)(Sigma Aldrich社、L0259)と対比することにより評価した。要するに、フィルター上で培養した6日後に、hCMEC/D3単層を、10mMのhepes(Life technologies社、15630-080)、及び1mMのピルビン酸ナトリウム(Life technologies社、11360))が補充された輸送培地(HBSS CaMg(Gibco社、14025-100)、1.5mLをウェル毎に含有する12-ウェルプレートに移した(反管腔側のコンパートメント)。次に50μMのLYを含有する輸送培地、0.5mLを管腔コンパートメントに添加した。インキュベーションを、37℃、5% CO2、95%湿度で実施した。15、25、及び45分後、インサートを、あらかじめ輸送培地、1.5mLを充填した新規ウェルに移した。45分後、各時点において、両方のコンパートメントから一定分量を採取し、蛍光分光光度計(Tecan Infinite F500)を用いてLYの濃度を測定した。
Wekslerら(2005年)がすでに記載しているように、HCMEC/D3のTEERを、Endogro-MV培地(SCME004、メルクミリポア社、フランス)内で培養してから5、6、及び7日間後に測定した。
SK-N-SH D細胞株
SK-N-SH D細胞株は、ATCC HTB-11 SK-N-SHヒト神経芽細胞腫細胞株のサブ-クローンである。β3-チューブリン及びアクチン-F陽性細胞であるニューロン様の細胞として、自然に完全分化し得るサブ-クローンとして、分化培地内で単離された。この細胞株は、CNCM(Collection Nationale de Culture de Microorganismes、Institut Pasteur、Paris、フランス)に2015年9月4日付けで、番号CNCM I-5010として預託された。
Claims (18)
- ラクダ重鎖抗体のVHHを含む又はそれからなるポリペプチであって、前記VHHが、
i)配列番号3の配列を有し、
ii)配列番号3の配列と少なくとも90%の同一性を有する、i)のVHHの配列変異体であり、又は
iii) i)若しくはii)のVHHの一部分であり、
血液脳関門を横断して透過可能であり、8.5に等しい若しくはそれ以上、好ましくは9に等しい若しくはそれ以上の等電点を有するポリペプチド。 - 前記一部分の配列の長さが、VHHフラグメントの配列の長さの少なくとも90%である、請求項1に記載のポリペプチド。
- 配列番号3の配列を備えるVHHフラグメントのCDR領域を含む、請求項1又は2に記載のポリペプチド。
- VHH含有ポリペプチドであり、前記VHH又はその一部分が、追加のペプチドとインフレームで融合している、請求項1から3のいずれか一項に記載のポリペプチド。
- ニューロン細胞標的ペプチドを、好ましくはNH2-末端部に含む、請求項4に記載のポリペプチド。
- 前記ニューロン細胞標的ペプチドは、配列番号1、及び配列番号32〜配列番号34からなる群の中から選択された配列からなる又はそれを含む、請求項5に記載のポリペプチド。
- 二量体化能力、特にホモ二量体化能力を有し、好ましくは、ホモ二量体化は、ジスルフィド架橋により達成される、請求項1から6のいずれか一項に記載のポリペプチド。
- タギングペプチド又はアフィニティーペプチドを含む、請求項4から7のいずれか一項に記載のポリペプチド。
- インフレームで融合したエフェクターポリペプチド、特に細胞生存率、細胞周期の状態、アポトーシス、細胞生存率、細胞死、有糸分裂、細胞分化、細胞のサイズ又は形態、細胞増殖、神経突起伸長、細胞間の結合、細胞シグナル経路の活性化状態、シグナル伝達、細胞の移動性、細胞の運動性、疾患関連の状態、傷害後又は病変後に関連する状態、診断関連の状態に対する効果を有するエフェクターポリペプチドを含む、請求項4から8のいずれか一項に記載のポリペプチド。
- 前記エフェクターポリペプチドが、ニューロンの生存性及び/又は保護及び/又は運動性に対する効果を有し、並びに/或いは神経突起伸長を促進する、狂犬病ウイルスのGタンパク質に由来するペプチド、特に配列番号7の配列を有するペプチドからなる又はそれを含む、請求項9に記載のポリペプチド。
- ペプチドスペーサーが、前記VHHと前記エフェクターポリペプチドとの間に挿入されている、請求項9又は10に記載のポリペプチド。
- 配列番号3等のVHH配列、配列番号1等のニューロン細胞標的ペプチド、配列番号7等のニューロヴィータ配列、及び任意選択で配列番号5等のタグ配列を含み、好ましくは、配列番号11及び配列番号45からなる群より選択される配列を有する、請求項1から11のいずれか一項に記載のポリペプチド。
- 請求項1から12のいずれか一項に記載のポリペプチドをコードするポリヌクレオチド、特に配列番号4の配列、配列番号2の配列、配列番号8の配列、及び任意選択で配列番号6の配列を含むポリヌクレオチド、特に配列番号4、配列番号12、配列番号14、配列番号16、及び配列番号46の群の中から選択される配列からなるポリヌクレオチド。
- 請求項13に記載のポリヌクレオチドを含有するベクター。
- 細菌中での前記ポリヌクレオチドによりコードされるポリペプチドの発現に適する、好ましくは、前記ポリヌクレオチドによりコードされるポリペプチドが、前記細菌の周辺細胞質内において回収されるのを可能にする、請求項14に記載のベクター。
- 細胞又は細胞株、特に、請求項14若しくは15に記載のベクター及び/又は請求項13に記載のポリヌクレオチドを含有する細菌細胞。
- in vivo投与に適する生理学的に許容される媒体と会合した、請求項1から12のいずれか一項に記載のポリペプチド、請求項13に記載のポリヌクレオチド、請求項14又は15に記載のベクター、請求項16に記載の細胞又は細胞株を含む組成物。
- 神経系疾患、特に神経変性疾患、脳卒中、外傷、病変、又は傷害の治療において、薬剤として使用するための、請求項1から12のいずれか一項に記載のポリペプチド、請求項13に記載のポリヌクレオチド、請求項14若しくは15に記載のベクター、請求項16に記載の細胞若しくは細胞株、又は請求項17に記載の組成物。
