本発明のポリヌクレオチドは、DNAまたはRNAを含むことができる。ポリヌクレオチドは一本鎖または二本鎖であり得る。遺伝的コードの縮重の結果として、多数の異なるポリヌクレオチドが同じポリペプチドをコードし得ることは、当業者には理解されるであろう。加えて、当業者は、日常的な技術を用いて、本発明のポリペプチドが発現されないいずれかの特定の宿主生物のコドン使用頻度を反映させるために、本発明のポリヌクレオチドによりコードされるポリペプチド配列に影響しないヌクレオチド置換を生じさせることができることが、理解されるべきである。
本発明で用いられるポリヌクレオチドは、コドン最適化することができる。コドン最適化は、国際公開第1999/41397号および同第2001/79518号に以前に記載されている。異なる細胞は、特定のコドンの使用頻度が異なる。このコドンの偏りは、細胞タイプ中の特定のtRNAの相対的存在量の偏りに対応する。配列中のコドンを、対応するtRNAの相対的存在量に合致するように変化させることにより、発現を増大させることが可能である。同様に、特定の細胞タイプ中で対応するtRNAが稀であることが知られているコドンを意図的に選択することにより、発現を低下させることができる。つまり、追加の段階の翻訳制御を行なうことができる。
ベクターは、ある環境から別の環境への物質の移動を可能にするかまたは促進するツールである。本発明に従って、かつ一例として、組み換え核酸技術で用いられるいくつかのベクターは、物質、例えば核酸のセグメント(例えば異種DNAセグメント、例えば異種cDNAセグメント)が標的細胞に移動することを可能にする。ベクターは、非ウイルスベクターまたはウイルスベクターであり得る。組み換え核酸技術で使用されるベクターの例としては、限定するものではないが、プラスミド、mRNA分子(例えば、in vitro転写mRNA)、染色体、人工染色体およびウイルスが挙げられる。ベクターはまた、例えば、裸の核酸(例えば、DNA)であり得る。最も簡潔な形態では、ベクターは、対象となるヌクレオチドそのものであり得る。

好ましくは、本発明で使用されるレトロウイルスベクターはマウス白血病ウイルス(MLV)ベクターである。広宿主性モロニーマウス白血病ウイルス(MLV-A)由来のレトロウイルスベクターは世界中の臨床プロトコールで一般的に使用されている。これらのウイルスは細胞表面リン酸輸送体受容体を侵入に使用し、次いで増殖性細胞の染色体に恒久的にインテグレートされる。その後、遺伝子は細胞の寿命期間にわたって維持される。MLVに基づく構築物上の遺伝子活性は制御が容易であり、かつ長期間にわたって有効であり得る。これらのMLVに基づく系を用いて行われる臨床試験では、その耐容性が良好であり、有害な副作用がないことが示されている。

別の実施形態では、本発明のベクターはアデノウイルスベクターであり得る。アデノウイルスは二本鎖の直線状DNAウイルスであり、RNA中間体を経由しない。50種を超える種々のヒト血清型のアデノウイルスが存在し、遺伝子配列相同性に基づいて6亜群に分類される。アデノウイルスの天然の標的は呼吸上皮および消化管上皮であり、一般的に軽度の症状しか生じさせない。アデノウイルスベクター系では、血清型2型および5型(95%の配列相同性を有する)が最も一般的に使用され、通常それらは若年者の上気道感染症に関連する。
そのゲノムに大きなDNA断片を容易にクローニングすることができ、また、組み換え弱毒化ワクシニア変異体が報告されているので、本発明に従ってポックスウイルスベクターを用いることができる(Meyer, et al., 1991;Smith and Moss, 1983)。
本発明のベクターは、ワクシニアウイルスベクター、例えばMVAまたはNYVACであり得る。最も好ましいのは、ワクシニア株の改変型ウイルスであるアンカラ(MVA)またはその派生株である。ワクシニアベクターの代替物には、アビポックスベクター、例えば鶏痘またはALVACとして公知のカナリア痘およびその派生株が含まれ、それらはヒト細胞に感染し、組み換えタンパク質を発現することができるが、複製することができない。

LNGFRスペーサー介在CD44v6-CAR.28z構築物の作製
LNGFRに基づくスペーサーの配列を、シグナルペプチドを除いた、低親和性神経成長因子受容体(LNGFR)の細胞外部分から誘導した(P08138、TNR16_HUMAN)。野生型長型(NWL)設計は、4個のTNFRシステインリッチドメインとセリン/トレオニンリッチストークの両方を含む。野生型短型(NWS)設計は、4個のTNFRシステインリッチドメインのみを含む。突然変異型長型(NML)設計は、4個のTNFRシステインリッチドメイン、セリン/トレオニンリッチストークを含み、かつNGFに対する結合を回避するための4番目のドメイン中の特定の改変を含む(Yan et al, J Biol Chem, 1991, Jun 25;266(18):12099-104)。突然変異型短型(NMS)設計は、4番目のドメイン中の特定の改変を含む4個のTNFRシステインリッチドメインのみを含む。スペーサーはGENEART社により合成され、特異的制限部位(BamH1およびPflMI)が隣接し、それにより、IgG1 CH2CH3スペーサーの代わりに、本発明者らの独自のCD44v6特異的第二世代CAR構築物(図9;配列番号15)へとクローニングすることが可能になった。すべての構築物が、ヒトでの発現に対してコドン最適化されている。すべての構築物を、SFG-RV骨格(一般的に用いられるスプライシングMoMLVベースのレトロウイルスベクター(Riviere et al, PNAS, 1995, Jul 18;92(15):6733-7))中で発現させた。

T細胞を、細胞サイズのCD3/CD28ビーズ(ClinExVivo、Invitrogen社)およびIL-7/IL-15(5ng/mL、Peprotech社)を用いて活性化し、刺激後2日目および3日目での2ラウンドの遠心接種(spinoculation)によりRVを形質導入した。6日目に、ビーズを除去し、T細胞をRPMI1640(Gibco-Brl社)10%FBS(BioWhittaker社)中にてIL-7およびIL-15の存在下で培養した。CH2CH3スペーサー介在CD44v6特異的CAR構築物(CHWおよびCHM)の表面発現はIgG1 CH2CH3スペーサーに対して特異的なmAb(Jackson Laboratories社)を用いて検出し、LNGFRスペーサー介在CD44v6特異的CAR構築物(NWL、NWS、NMLおよびNMS)の表面発現はLNGFR特異的mAb(BD Bioscience社から(Clone: C40-14579)またはMiltenyi社から(Clone: ME20.4))を用いて分析した。活性化後9日目から15日目の間、CH2CH3スペーサー介在CD44v6-CAR.28z T細胞はポリクローナルIgG1 CH2CH3特異的mAbを用いてFACSソーティングし、LNGFRスペーサー介在CD44v6-CAR.28z T細胞はPEコンジュゲート化LNGFR特異的mAbであるC40-14579を用いて染色し、かつ抗PE常磁性ビーズ(Miltenyi社)を用いたカラムによりソーティングした。ソーティング後のT細胞増殖を、増加倍率として表わしている:x日目のT細胞数/ソーティング後のT細胞数。
共培養アッセイでは、CARソーティングされたT細胞を、標的細胞と共に、様々なE:T比で培養した。4日後、生存細胞を、FACSによりカウントおよび分析した。無関係なCAR(CD19)を形質導入されたT細胞を、対照として常に用いた。排除指数を以下のように算出した:1−(CD44v6.CAR28z+ T細胞の存在下での残留標的細胞数)/(CTR.CAR28z+ T細胞の存在下での残留標的細胞数)。CFSE希釈アッセイでは、CARソーティングされたT細胞にCFSEをロードし、照射済み(10,000rad)腫瘍細胞を用いてE:S比=1:5にて、または生物学的に活性な濃度のNGFを用いて刺激した。6日後、T細胞増殖を、CFSE色素を希釈した細胞の割合(%)を分析することにより、FACSによって測定した。
実験プロトコールは、米国動物実験委員会(Institutional Animal Care and Use Committee;IACUC)により承認された。最小残存病変モデル(minimal-residual disease model)に関して、NSGマウス(Jackson社)に1.5×106個のTHP1白血病細胞/マウスをi.v.注入した。3日後、マウスを、5×106個のソーティングされたLNGFRスペーサー介在CD44v6-CAR.28z T細胞、CH2CH3スペーサー介在CD44v6-CAR.28z T細胞または無関係なCAR(CD19)を担持するT細胞を用いてi.v.処置した。T細胞の生着を、採血およびFACS分析により週に1回モニタリングした。7週間後、マウスを屠殺し、その肝臓を、THP-1細胞の存在について組織病理学およびFACSにより分析した。十分に確立された疾患モデルに関して、NSGマウスに2×106個のMM1.S骨髄腫細胞/マウスをi.v.注入した。5週間後、マウスを、5×106個のソーティングされたLNGFRスペーサー介在CD44v6-CAR.28z T細胞、CH2CH3スペーサー介在CD44v6-CAR.28z T細胞または無関係なCAR(CD19)を担持するT細胞を用いてi.v.処置した。T細胞生着および骨髄腫進行を、採血およびFACS分析により週に1回モニタリングした(骨髄腫細胞は、異なるヒトCD45/CD3表現型に従って、T細胞とは区別されるであろう)。循環MM1.S細胞が30細胞/μLを超え、かつ/またはマウスが腫瘍に関連する苦痛の明らかな徴候を示した場合(麻痺状態または10%超の体重減少)、マウスを安楽死させた。
FACS分析のために、本発明者らは、ヒトCD44v6、CD4(e-Bioscience社)、CD123、CD19、CD14、CD3、CD8、CD45RA、CD62L、CXCR4、CD127、CD33、CD38、CD45、LNGFR、マウスCD45、7AAD(BD Biosciences社)およびIgG1 CH2CH3(Jackson laboratories社)に対するFITC、PE、PerCP、PE-Cy7、APC、APC-Cy7およびパシフィックブルーコンジュゲート化抗体を用いた。細胞(2×105個)を、4℃にて15分間、抗体と共にインキュベートし、PBS 1%FBSを用いて洗浄した。サンプルをFACS Canto IIフローサイトメーター(BD Biosciences社)にかけ、Flow Joソフトウェア(Tree star Inc)を用いてデータを分析した。相対的蛍光強度(RFI)を、以下の通りに算出した:サンプルの平均蛍光強度/対応するアイソタイプ対照の平均蛍光強度。
本発明者らは、近年、CD28の内因性共刺激ドメイン(endo-costimulatory domain)と組み合わせたCD3ζ鎖に基づくCD44v6特異的CARを構築した(Casucci et al, Blood 2013, Nov 14;122(20):3461-72)。このCARの細胞外スペーサー領域には、IgG1 CH2CH3スペーサーが、CD44v6抗原のよりよい標的化のために、そして形質導入されたT細胞の選択およびin vivo追跡を可能にするために、挿入された。しかしながら、CH2CH3スペーサー介在CARの重大な欠点は、それらがFcγ受容体(FcγR)と相互作用することであり(Hombach et al, Gene Ther 2000, Jun;7(12):1067-75)、これは、潜在的に、これらの受容体を発現する細胞(例えば、単球/マクロファージ)の非特異的な標的化および/または形質導入されたT細胞のin vivoクリアランスをもたらす(図1A)。この問題を回避するために、本発明者らは、元のCH2CH3スペーサーを、低親和性神経成長因子受容体(LNGFR)由来の種々の細胞外ドメインと置き換えた。細胞内シグナル伝達構成要素を欠損したLNGFRの切断型は、T細胞の遺伝子マーキングのために既に臨床的に用いられている(Bonini et al, Nat Med, 2003, Apr;9(4):367-9;Ciceri et al, Lancet Oncol, 2009, May;10(5):489-500)。LNGFRの細胞外部分は、4個のTNFRシステインリッチ領域および1個のセリン/トレオニンリッチストークから構成される(図1B)。まず初めに、本発明者らは、2種類のCD44v6-CAR.28z構築物を作製した:一方はLNGFRの細胞外部分全体によりスペーサー介在されており(LNGFR野生型長型またはNWL)、他方は4個のTNFRシステインリッチ領域のみによりスペーサー介在されている(LNGFR野生型短型またはNWS)。天然リガンドNGFを介するLNGFRスペーサー介在構築物の抗原非依存的活性化の可能性を除外するために、本発明者らは、NGFシグナル伝達を取り消すことが知られている4番目のTNFRシステインリッチドメインの特定の欠失を担持する2種類の追加のCD44v6-CAR.28z構築物を作製し(Yan et al, J Biol Chem, 1991, Jun 25;266(18):12099-104)、それによりLNGFR突然変異型長型アイソフォームまたはNMLおよびLNGFR突然変異型短型アイソフォームまたはNMSをそれぞれ生成した。対照として、本発明者らは、FcγRIを認識することができない突然変異型の元のCH2CH3スペーサー(CHM)を含むCD44v6-CAR.28z構築物も作製した(Hombach et al, Gene Ther 2000, Jun;7(12):1067-75)。意外なことに、FcγRIIおよびFcγRIIIの両方が、この共通セット以外の残基を用いることができ、このことは、この突然変異が結合性を完全に無効にするわけではないことを示唆する(Shields et al, J Biol Chem, 2001, Mar 2;276(9):6591-604;Armour et al, Mol Immunol, 2003, Dec;40(9):585-93)。
種々のLNGFRスペーサー介在CD44v6-CAR.28z構築物を、初代T細胞に形質導入するためにレトロウイルスベクター(RV)へとクローニングした。形質導入のために、T細胞を、それらの機能的表現型をよりよく保存するプロトコールに従って、CD3/CD28ビーズおよびIL-7/IL-15を用いて活性化した(Kaneko et al, Blood, 2009, Jan 29;113(5):1006-15;Bondanza et al, Blood 2011, Jun 16;117(24):6469-78;Cieri et al, Blood, 2013, Jan 24;121(4):573-84)。形質導入後、すべての構築物は、抗LNGFR mAb C40-1457を用いてT細胞表面上で特定することができ(図2A)、このことは、これらが適正にプロセシングされ、細胞膜上に埋め込まれ、かつ、最も重要なことに、抗NGFR mAbにより認識されたことを示す。結果として、種々のLNGFRスペーサー介在CD44v6-CAR.28z T細胞を、免疫磁性ビーズを用いてソーティングすることができた(図2B)。より詳しく調べたところ、本発明者らは、NWLスペーサー介在アイソフォームのみが、別の抗LNGFR mAbであるME20.4に結合することを見出し、これにより、種々の長さのLNGFRスペーサーにより決定づけられるコンホメーション変化が、ME20.4エピトープのアクセス可能性を制御し得ることが示唆された。重要なことに、種々のLNGFRスペーサー介在細胞の増殖動態は、CH2CH3スペーサー介在CD44v6-CAR.28z T細胞のものと似通っており(図2B)、このことは、CAR上に搭載された細胞外LNGFR配列により引き起こされる潜在的な増殖上の有利さを除外する。培養終了時に、結果として生じた集団は、早期分化型T細胞に富んでおり(図2C)、これはIL-7/IL-15の存在下でのビーズ活性化T細胞の機能的分化経路に関する干渉がないことを示す。
元のCH2CH3スペーサーをLNGFRスペーサーと置換した後にCD44v6特異的認識が保存されていることを確認するために、LNGFRスペーサー介在CD44v6-CAR.28z T細胞を、CD44v6発現腫瘍細胞との共培養実験で調べた。CH2CH3スペーサー介在細胞と同様に、LNGFRスペーサー介在CD44v6-CAR.28z T細胞は、CD44v6陽性腫瘍細胞(MM1S細胞株およびTHP-1細胞株)を効率的に排除したが、CD44v6陰性腫瘍細胞(BV173細胞株)は排除しなかった(図3A)。さらに、CD44v6特異的認識は、活発なT細胞増殖を伴い(図3B)、このことは、LNGFRスペーサー介在CD44v6-CAR.28z T細胞の治療可能性の完全な保存を示唆する。したがって、本発明に従うLNGFRスペーサー介在CARは、それらが標的とする特異的抗原を発現する腫瘍モデルに対して、結果として有効となる。
in vitroでの効率的かつ特異的な認識が実証された後、LNGFRスペーサー介在CD44v6-CAR.28z T細胞を、最初は最小残存病変モデルで、次いで十分に確立された疾患(WED)モデルで、in vivoでの抗腫瘍活性に関して調べた。最初のモデルでは、NSGマウスにTHP-1白血病細胞を注入し、3日後にCH2CH3スペーサー介在細胞または種々のLNGFRスペーサー介在CD44v6-CAR.28z T細胞を用いて処置した。種々のLNGFRスペーサー介在CD44v6-CAR.28z T細胞は、CH2CH3スペーサー介在CD44v6-CAR.28z T細胞よりもよく増殖し(図6A)、かつ持続した(図6B)。したがって、LNGFRスペーサー介在CD44v6-CAR.28z T細胞は、CH2CH3スペーサー介在CD44v6-CAR.28z T細胞を注入されたマウスと比較して、THP1浸潤肝臓重量のより良好な標準化により実証される通り、優れた抗腫瘍作用を媒介するようである(図6C)。2番目の十分に確立された疾患モデルでは、NSGマウスにCD44v6発現MM1.S骨髄腫細胞を注入し、5週間後に、腫瘍が骨髄に定着してから、CH2CH3スペーサー介在細胞または様々なLNGFRスペーサー介在CD44v6-CAR.28z T細胞を用いて処置した。NMLアイソフォームを担持するCD44v6-CAR.28z T細胞もまた含められた。このより厳格なモデルでは、CH2CH3スペーサー介在CD44v6-CAR.28z T細胞がほとんど生着せず、かついかなる顕著な抗腫瘍作用も媒介しなかった一方で、様々なLNGFRスペーサー介在CD44v6-CAR.28z T細胞は増殖し(図7A)、持続して著しい抗腫瘍活性を生じた(図7B)。
LNGFRスペーサー介在CD19-CAR.28z構築物およびCEA-CAR.28z構築物の作製
実施例1に記載されたものと類似の戦略を用いて、CD19特異的CAR構築物およびCEA特異的CAR構築物を作製した(図22)。以下の構築物が作製された:

共培養アッセイでは、CARソーティングされたT細胞を、標的細胞と共に、1:10のE:T比で培養した。4日後、生存細胞を、FACSによりカウントおよび分析した。排除指数を以下のように算出した:1−(CD44v6-4GS2.CAR28z+ T細胞、CD19.CAR28z+T細胞およびCEA.CAR28z+ T細胞の存在下での残留標的細胞数)/(CTR.CAR28z+ T細胞の存在下での残留標的細胞数)。24時間のインキュベーション後に共培養物の上清を回収し、サイトカイン産生(IFNy、IL-2およびTNFα)に関して、CBAアッセイ(BD Biolegend社)を用いて分析した。
最小残存病変モデルに関して、NSGマウス(Jackson社)に1.5×106個のALL-CM白血病細胞/マウスをi.v.注入した。3日後、マウスを、5×106個のソーティングされたLNGFRスペーサー介在(NWL、NMS)CD19-CAR.28zまたはCD44v6-4GS2.CAR.28z T細胞を用いてi.v.処置した。T細胞の生着を、採血およびFACS分析により週に1回モニタリングした。7週間後、マウスを屠殺し、その骨髄(BM)を、抗hCD45 mAbおよび抗hCD19 mAbを用いて、ALL-CM細胞の存在についてFACSにより分析した。
種々のLNGFRスペーサー介在CAR.28z構築物を、初代T細胞に形質導入するためにレトロウイルスベクター(RV)へとクローニングした。形質導入のために、T細胞を、それらの機能的表現型をよりよく保存するプロトコールに従って、CD3/CD28ビーズおよびIL-7/IL-15を用いて活性化した(Kaneko et al, Blood, 2009, Jan 29;113(5):1006-15;Bondanza et al, Blood 2011, Jun 16;117(24):6469-78;Cieri et al, Blood, 2013, Jan 24;121(4):573-84)。形質導入後、免疫磁性ビーズを用いてT細胞をソーティングすることができ(図23)、このことは、CD44v6抗原を標的とするCARを用いて示された通り、LNGFR由来スペーサーが、CD19抗原およびCEA抗原に特異的な2種類の別のCARの場合でも、適正にプロセシングされ、かつ細胞膜上に埋め込まれたことを示している。
元のCH2CH3スペーサーをLNGFRスペーサーと置換した後にCD19およびCEA特異的認識が保存されていることを確認するために、LNGFRスペーサー介在CD19-CAR.28z T細胞およびCEA-CAR.28z T細胞を、抗原発現腫瘍細胞との共培養実験で調べた。CH2CH3スペーサー介在細胞と同様に、LNGFRスペーサー介在CD19-CAR.28z T細胞、CEA-CAR.28z T細胞およびCD44v6-4GS2.CAR.28z T細胞は、CD19+、CEA+およびCD44v6+腫瘍細胞をそれぞれ効率的に排除し、抗原陰性腫瘍細胞は容認(spare)した(図24A)。特に、LNGFRスペーサー介在CD19-CAR.28z CAR T細胞はCD19+のALL-CM細胞およびBV-173細胞を特異的に排除したが、CD19-のHL-60細胞およびBXPC3細胞は排除しなかった(図24A)。同様に、LNGFRスペーサー介在CEA-CAR.28z T細胞はCEA+のBXPC3細胞を特異的に排除したが、CEA-のHL-60細胞、ALL-CM細胞およびBV-173細胞は排除せず(図24A)、またCD44v6-4GS2.CAR.28z T細胞はCD44v6+のBXPC3細胞を特異的に排除したが、CD44v6-のALL-CM細胞、BV173細胞およびHL-60細胞は排除しなかった(図24A)。同等の結果が、抗原特異的サイトカイン放出(IFNy、IL2およびTNFα)を評価した場合に得られた(図24B)。
FcRγとの相互作用により媒介される非特異的認識の欠如を実証するために、LNGFRスペーサー介在CD19-CAR.28z T細胞、CEA-CAR.28z T細胞およびCD44v6-4GS2.CAR.28z T細胞を、FcγR+、CD19-、CEA-のHL-60細胞と共培養した。この系では、CH2CH3スペーサー介在CD19-CAR.28z T細胞およびCEA-CAR.28z T細胞のみがHL-60標的細胞を排除することができ、つまり、LNGFRに基づくスペーサーの使用が、望ましくない生来型の免疫応答の活性化を回避することを確認する。
in vitroでの効率的かつ特異的な認識が実証された後、LNGFRスペーサー介在CD19-CAR.28z T細胞を、最小残存病変モデルで、in vivoでの抗腫瘍活性に関して調べた(challenge)。NSGマウスにALL-CM白血病細胞を注入し、3日後に種々のLNGFRスペーサー介在(NWLおよびNMS)CD19-CAR.28z T細胞を用いて処置した。この場合、ALL-CM白血病細胞はCD44v6抗原を発現しないので、LNGFRスペーサー介在(NWLおよびNMS)CD44v6-4GS2.CAR.28z T細胞を陰性対照として用いた(図25)。CD44v6-CAR.28z T細胞を注入されたマウスと比較して、より低い濃度のALL-CM細胞が骨髄に浸潤することにより実証される通り、両方のLNGFRスペーサー介在CD19-CAR.28z T細胞が抗腫瘍作用を媒介するようである(図25)。

- 共刺激ドメインが、CD28、CD137(4-1BB)、CD134(OX40)、DaplO、CD27、CD2、CD5、ICAM-1、LFA-1、Lck、TNFR-I、TNFR-II、Fas、CD30、CD40およびそれらの組合せのいずれか1以上に由来する共刺激性ドメインを含む、請求項13または19に記載のCAR。
- 細胞内シグナル伝達ドメインが、ヒトCD3ゼータ鎖、FcyRIII、FcsRI、Fc受容体の細胞質テイル、細胞質受容体を担う免疫受容体チロシンベース活性化モチーフ(ITAM)、およびそれらの組合せのいずれか1以上の細胞内シグナル伝達ドメインを含む、請求項13〜20のいずれか1項に記載のCAR。
- 抗原特異的標的化ドメインがCD44v6を標的とし、膜貫通ドメインがCD28の膜貫通ドメインを含み、細胞内シグナル伝達ドメインがヒトCD3ゼータ鎖の細胞内シグナル伝達ドメインを含み、かつ共刺激ドメインがCD28内因性共刺激性ドメインを含む、請求項13〜21のいずれか1項に記載のCAR。
- 請求項1〜22のいずれか1項に記載のCARをコードするポリヌクレオチド。
- 請求項23に記載のポリヌクレオチドを含むベクター。
- ベクターがウイルスベクターである、請求項24に記載のベクター。
- 請求項1〜22のいずれか1項に記載のCAR、請求項23に記載のポリヌクレオチド、または請求項24もしくは25に記載のベクターを含む細胞。
- 細胞がT細胞である、請求項26に記載の細胞。
- 請求項26または27に記載の細胞を含む医薬組成物。
- 癌の治療での使用のための、請求項13〜22のいずれか1項に記載のCAR、請求項23に記載のポリヌクレオチド、請求項24もしくは25に記載のベクター、または請求項26もしくは27に記載の細胞。
- CD44を発現する腫瘍の治療での使用のための、請求項22に記載のCAR、該CARをコードするポリヌクレオチド、該ポリヌクレオチドを含むベクター、または該CAR、ポリヌクレオチドもしくはベクターを含む宿主細胞。
