A)材料及び方法
菌株及び増殖条件。本試験において使用した株は図15A〜Mに列挙されている。プラスミド精製及びクローニングのために使用する大腸菌Top10、及びコンジュゲーションのために使用する大腸菌Sm10 λ pir、並びにpKNG101を伝播するために使用する大腸菌BW19610[46]をLB寒天プレート上、及びLBブロス中、37℃で常套的に増殖させた。発現ベクターを選択するために、アンピシリンを200μg/ml(エルシニア属)又は100μg/ml(大腸菌)の濃度で使用した。自殺ベクターを選択するために、ストレプトマイシンを100μg/mlの濃度で使用した。アンピシリン非耐性E40−誘導体[13]であるY.エンテロコリチカMRS40[36]及びそれに由来する株をブレインハートインフュージョン(BHI;Difco)上、室温で常套的に増殖させた。全てのY.エンテロコリチカ株にナリジクス酸を添加し(35μg/ml)、全てのY.エンテロコリチカasd株には、100μg/mlのメソ−2,6−ジアミノピメリン酸(mDAP、Sigma Aldrich)をさらに補充した。S.エンテリカSL1344をLB寒天プレート上、及びLBブロス中、37℃で常套的に増殖させた。S.エンテリカにおける発現ベクターを選択するために、アンピシリンを100μg/mlの濃度で使用した。
YopE融合タンパク質の3型分泌に基づくタンパク質送達系
Y.エンテロコリチカT3SSエフェクターYopE(配列番号1)のちょうどN末端は、異種タンパク質を移行させるために十分な分泌シグナルを含有するが[10]、そのシャペロン(SycE)に対するシャペロン結合部位(CBS)は含まれない[63]。YopEのN末端の138アミノ酸(配列番号2)は他の異種T3S基質の移行に関して最良の結果が示されているので[38]、これを、送達するタンパク質と融合するために選択した。これらのYopEのN末端の138アミノ酸はCBSを含有するので、さらに、SycEを同時発現させることに決めた。精製されたY.エンテロコリチカpYV40病原性プラスミドからクローニングされたSycE−YopE1−138断片は、YopEの内因性プロモーター及びシャペロンSycEの内因性プロモーターを含有する(図10)。したがって、SycE及び任意のYopE1−138融合タンパク質は、室温での増殖から37℃への迅速な温度シフトによって誘導される。37℃での培養時間は、細菌内に存在する融合タンパク質量に影響を及ぼす。YopE1−138の3’末端に多重クローニング部位(MCS)を付加し(図10B)、その後にMyc及び6×Hisタグ並びに終止コドンが続いた。
インビトロ分泌アッセイ(図1A参照)では、周囲の液体へのタンパク質分泌を人工的に誘導する。TCAに基づくタンパク質沈殿の後、抗YopE抗体を用いたウェスタンブロット分析を使用して、分泌されたタンパク質量を決定した(図1B)。wt株からは完全長YopEが分泌されたが、ΔHOPEMT asd株からは分泌されなかった。YopE1−138−Myc−His(今後YopE1−138−Mycと称する;配列番号3)が存在すると、より小さなYopEバンドが目に見えるようになった(図1B)。したがって、YopE1−138断片は本明細書に記載の設定において十分に分泌された。真核細胞へのタンパク質移行の均一性を分析するために、HeLa細胞にYopE1−138−Mycをコードする株を感染させ、IFによってmycタグを染色した(図2A及びB)。最初は細菌のみが染色されたが、感染後(p.i.)30分の時点では、細胞の輪郭が目に見え始め、これは感染時間を増大させると増強される(図2B)。この傾向は、HeLa細胞の内部のmycタグ染色強度によく反映される(図2A及びB)。YopE1−138−Mycは、核内以外は[68]細胞のどこででも検出することができる(図2A)。驚くべきことに、この手法は、全てではないが大多数の細胞に同等に到達した。Y.エンテロコリチカは多くの異なる細胞型に感染することが公知であるので[69]、種々の細胞株へのYopE1−138−Myc送達を追跡した。感染させたマウス線維芽細胞、ジャーカット細胞及びHUVECにおいて同じ同種抗MycIF染色が観察された(図11)。さらに、感染多重度を高く又は低く調整することにより、なお大多数の細胞を標的としたまま、送達されるタンパク質量を調節することができる(図2C)。細菌数を少なくすると、送達されるタンパク質を大量に有する少数の細胞がもたらされるのではなく、送達されるタンパク質を少量含有する最多数の細胞がもたらされる(図2C)。
YopE自体は細胞質に局在していたので(図2A)、YopE1−138断片により核融合タンパク質の局在化が妨害されるかどうかを決定することが特に興味深い。したがって、YopE1−138−EGFPのC末端(及びN末端、同様の結果)にSV40 NLSを付加した(それぞれ配列番号39及び配列番号38)。感染させたHeLa細胞において、YopE1−138−EGFP(配列番号37)では弱い細胞質染色が導かれたが、YopE1−138−EGFP−NLSでは、より強力な核内EGFPシグナルが生じた(図3)。これにより、YopE1−138断片がNLSの使用と適合することが示される。mCherryはすでに植物病原体において使用されていたが[70]、これにより、T3SSをコードするヒト又は動物病原性細菌を介したGFP様タンパク質の送達が上首尾であることが示される。これにより、SycE及びYopE1−138依存性戦略が多くの選択されたタンパク質の送達に関して非常に有望であることが検証される。
YopE1−138断片の細菌による送達は大いに役立つものであるが、YopE1−138断片は、融合タンパク質機能及び/又は局在化を妨害する可能性がある。したがって、タンパク質送達後に除去することが最適であると思われる。この目的のために、YopE1−138と融合パートナー(転写制御因子ET1−Myc(配列番号36及び41)[74]及びヒトINK4C(配列番号40及び配列番号43))の間に2つのTEV切断部位(ENLYFQS)[71−73]を導入した。提供される方法の利点を保持するために、別のY.エンテロコリチカ株においてTEVプロテアーゼ(S219V変異体;[75])をYopE1−138(配列番号42)とさらに融合した。HeLa細胞に、両方の株を一度に感染させた。タンパク質の移行画分のみの分析を可能にするために、感染HeLa細胞を、感染の2時間後に(図4)、細菌は溶解させないことが公知であるジギトニンを用いて溶解させた([76];コントロールについては図12を参照されたい)。ウェスタンブロット分析により、細胞に対応する株を感染させた場合にのみ、YopE1−138−2×TEV切断部位−ET1−Myc又はYopE1−138−2×TEV切断部位−Flag−INK4C−Mycが存在することが明らかになった(図4A及びC)。この細胞溶解物を精製TEVプロテアーゼで一晩消化すると、バンドのシフトを観察することができた(図4A及びC)。このバンドは、ET1−Myc(図4C)又はFlag−INK4C(図4A)に対応し、1つのセリンのみの可能性が最も高い、TEV切断部位のN末端残余を有する。細胞にTEVプロテアーゼを送達する株を同時感染させると、同じ切断されたET1−Myc又はFlag−INK4C断片が目に見えるようになり、これにより、T3SSにより送達されたTEVプロテアーゼが機能性であること、及び単一の細胞に両方の菌株が感染したことが示される(図4A及びC)。切断は完全なものではないが、移行したタンパク質の大多数が、感染の2時間後にすでに切断され、さらに、精製TEVプロテアーゼを用いた一晩消化によってより良好な切断率はもたらされなかった(図4B)。報告の通り、TEVプロテアーゼ依存性切断には、融合タンパク質に応じた最適化が必要であり得る[77、78]。したがって、移行後のYopE1−138付属物のTEVプロテアーゼ依存性除去により、初めて、N末端アミノ酸の1つのみのアミノ酸組成の変化で、ほぼ天然の異種タンパク質のT3SSタンパク質送達がもたらされる。
サルモネラ・エンテリカ由来のSopEは、Cdc42と相互作用し、それによりアクチン細胞骨格リモデリングを促進する、よく特徴付けられたグアニンヌクレオチド交換因子(GEF)である[79]。HeLa細胞へのYopE1−138−Mycの移行の影響はなかったが、移行したYopE1−138−SopE(配列番号5及び135)によりアクチンネットワークの劇的な変化が誘導された(図5A)。別のGEFエフェクタータンパク質、シゲラ・フレックスネリ由来のIpgB1(配列番号4)を用いて同様の結果が得られた。驚くべきことに、アクチン細胞骨格の最初の変化は感染後2分という速さで観察された(図5A)。したがって、T3SS依存性タンパク質送達は、遠心分離によって感染を開始したすぐ後に起こると結論づけることができる。厳密なT3SS依存性輸送を証明するために、真核細胞膜への移行ポアを形成するT3SSタンパク質の1つを欠失させた(YopB、[80]参照)(図12)。
ヒトタンパク質をIII型分泌によって移行できることを示すために、ヒトアポトーシス誘導因子を、Y.エンテロコリチカによる送達のためにYopE1−138と、又は、S.エンテリカによる送達のためにSteA1−20、SteA、SopE1−81又はSopE1−105と融合した。次いで、Bcl−2タンパク質ファミリーのプロアポトーシスメンバーであるヒトBH3相互作用ドメインデスアゴニスト(Bid、配列番号24)の移行をモニターした。Bidは、カスパーゼ−8(CASP8)によって誘導されるミトコンドリア損傷のメディエーターである。CASP8によりBidが切断され、切除されたBid(tBID、配列番号25)はミトコンドリアに移行し、そこでチトクロムC放出を誘発する。後者により、17kDaと12kDaのサブユニットに切断される、カスパーゼ3(CASP3)活性化の固有の様式が導かれる[85]。YopE1−138−Myc又はYopE1−138−Bidを発現するY.エンテロコリチカの1時間にわたる感染では、アポトーシスを誘導することができなかったが、ヒトtBIDの移行により、細胞死がよく特徴付けられたアポトーシス誘導因子スタウロスポリンよりも大きな程度で誘発された(図7A及びC)。予測通り、tBIDの移行により、CASP3 p17サブユニットの産生が、スタウロスポリンを用いた場合より大きな量でまで誘導される(図7A)。移行したタンパク質量と内因性Bidの比較を可能にするために、HeLa細胞をジギトニンを用いて溶解させ、抗Bid抗体を使用したウェスタンブロット法によって分析した(図7B)。T3SSにより送達されたYopE1−138−tBIDは、ほぼHeLa細胞における内因性Bidレベルに達したが、送達されたYopE1−138−Bidはさらに高分量で存在した(2.5倍)(図7B)。HeLa細胞のディーププロテオーム及びトランスクリプトームマッピングにより、単一細胞当たり4.4倍の105コピーのBidが推定された[86]。したがって、3SS依存性ヒトタンパク質送達が細胞当たり105〜106のタンパク質に達すると結論づけることができる。これらの数は、大腸菌T3SSにより移行したナノボディの細胞当たりのコピー数と一致する[19]。感染多重度について及び感染の持続時間について10の係数、抗生物質添加の時点及び感染前の37℃での培養時間について3.2の係数のレベリングを仮定すると、送達されるタンパク質コピー/細胞を数1000コピー/細胞から数106コピー/細胞まで調整することができる。総じて、これらの結果から、移行したtBIDが機能性であり、有意味なレベルで送達されたことが示された。これにより、細胞生物学の中心的側面であるアポトーシスの調節におけるタンパク質の役割を試験するための移行ツールが検証された。
この細菌ツールの興味深い特徴は、生きている動物における潜在的使用である。胎児性/胚性状態にあるゼブラフィッシュは透明性を維持することができ、それにより、蛍光染色及び顕微鏡観察が可能になる[54、88、89]。いくつかのゼブラフィッシュアポトーシス誘導因子が詳細に記載されており、z−BIMが最も強力である[90]。したがって、z−BIMを我々の系にクローニングすることに決めた。ヒトBIMに対する相同性が弱いとしても、ヒト上皮細胞におけるYopE1−138−z−BIM(配列番号21)のアポトーシス誘導の力価をアッセイした。YopE1−138−z−BIMを移行させる株を1時間にわたって感染させたHeLa細胞では、細胞死の明らかな徴候が示された。次いで、受精後2日(dpf)ゼブラフィッシュ胚を用い、局在感染モデルを使用して、後脳への細菌のマイクロインジェクションによってインビボ実験を実施した[54]。5.5時間にわたる感染後、フィッシュを固定し、透過処理し、CASP3 p17の存在について染色した。YopE1−138−Mycを発現する株を感染させると、後脳領域では細菌が目に見えたが(染色「b」、図8A I)、細菌の周囲でのアポトーシスの誘導は検出されなかった(染色「c」、図8A I)。対照的に、YopE1−138−z−BIMを送達する株を感染させると、細菌周囲の領域において、切断されたCASP3の存在の強力な増加が観察された(図8A II)。最大値z投影に対する自動画像解析により、YopE1−138−z−BIMを移行させる細菌により、近くの細胞のアポトーシスがコントロール細菌によるものよりもはるかに多く誘導されることが確認される(図8B)。これにより、z−BIMが、細菌による移行に際して、ゼブラフィッシュにおいて機能性であることが示される。これらの結果から、生きている動物における真核生物タンパク質送達のためのT3SSの使用がさらに検証される。
リン酸化は、生物学的プロセスを活性化するか又は不活化することが可能であり、したがって、試験シグナル伝達事象の適切な標的となる、広く拡散した翻訳後修飾である[91、92]。これにもかかわらず、現在アポトーシスにおけるリン酸化のシステムレベルの分析は利用可能になっていない。HeLa細胞に送達されたヒトtBidの影響を分析するために、LC−MS/MSによる、標識を用いないリン酸化プロテオミクス手法を使用した。3つの独立した実験において、細胞を無処理のままにしたか、又は、ΔHOPEMT asd+YopE1−138−Myc又はΔHOPEMT asd+YopE1−138−tBidを30分にわたって感染させた。細胞を溶解させ、その後、酵素により消化し、リンペプチドを濃縮し、個々のリンペプチドを数量化及び同定した。ΔHOPEMT asd+YopE1−138−Mycを感染させた細胞とΔHOPEMT asd+YopE1−138−tBidを感染させた細胞を比較し、それにより、363のtBid依存性リン酸化事象を同定することが可能になった。tBid送達に際して、286のリンペプチドではリン酸化の増加が示され、77ではリン酸化の減少が示され、これは、243の異なるタンパク質に対応し、これをtBidリン酸化プロテオームとして定義した。STRINGデータベースを使用して、tBidリン酸化プロテオームのタンパク質間相互作用ネットワークを創出した[93](図9A)。さらに、ミトコンドリアでのアポトーシスに関連することが公知の27のタンパク質をネットワークに追加し、中心的なクラスターを構築した。興味深いことに、tBidリン酸化プロテオームからの数種のタンパク質のみがこの中心的なクラスターとつながり、これにより、多くのタンパク質が、これまでアポトーシスタンパク質と直接関連づけられていなかったリン酸化の変化を受けることが示される。tBidリン酸化プロテオームによりカバーされる生物学的機能の特徴を明らかにするために、Database for Annotation,Visualization,and Integrated Discovery(DAVID、http://david.abcc.ncifcrf.gov/)[94、95]データベースの機能性アノテーションツールを使用した遺伝子オントロジー分析を実施した。同定された生物学的機能により、多様な細胞プロセスがtBidの影響を受けることが示される。クロマチン再構成及び転写の調節に関与する多くのタンパク質がリン酸化の変化を受ける(すなわち、CBX3、CBX5、TRIM28、HDAC1)。例えば、HDAC1は、転写の調節において役割を果たすヒストンデアセチラーゼである。HDAC1は、同じくアポトーシスに関与するタンパク質であるNF−kBの転写活性を調節することが可能であることが示されている。さらに、以前示されているRNAプロセシングに関与するタンパク質のクラスターがアポトーシスの調節において重要な役割を果たすことを同定した[96]。例えば、HNRPKは、DNA損傷に対するp53/TP53応答を媒介し、アポトーシスの誘導に必要である[97]。さらに、タンパク質翻訳に関与するタンパク質のリン酸化も影響を受けた。アポトーシス細胞において全体的なタンパク質合成が減少するという観察に沿って、いくつかの真核生物開始因子(すなわち、EIF4E2、EIF4B、EIF3A、EIF4G2)がリン酸化の変化を受ける。興味深いことに、細胞骨格リモデリングに関与する多くのタンパク質のリン酸化(例えば、PXN、MAP1B9)がtBid送達に際して変更される。これは、tBid送達に際して細胞の形態が劇的に変化するという観察と一致する(図9B)。細胞の縮小及び接触の喪失は、ZO2及びパキシリンのような接着関連タンパク質のリン酸化が観察されたという事実により反映される。同様に、核の縮小には、ラミンA/C及びラミンB1のような層状タンパク質のリン酸化が伴う。総じて、tBID送達により、ミトコンドリア完全性の破綻によっても示される迅速なアポトーシス応答が誘導される(図9B)。我々は、tBidにより誘導されるアポトーシスが、多様な細胞プロセスに関与する数百のリン酸化事象に影響を及ぼすことを示した。同定されたタンパク質の多くがアポトーシスに関係しているが、アポトーシス誘導に際してリン酸化されることが分かっているのはほんのわずかである。したがって、リン酸化プロテオミクス手法により、アポトーシスに関するさらなる試験のための有用なリソースがもたらされる。

Claims (34)
- エルシニア属、大腸菌属、サルモネラ属、及びシュードモナス属からなる群から選択される組換えグラム陰性菌株であって、
5’から3’の方向に、
プロモーター、
前記プロモーターに作動可能に連結した、細菌T3SSエフェクタータンパク質由来の送達シグナルをコードする第1のDNA配列、及び
前記第1のDNA配列の3’末端とインフレームで融合した、アポトーシス又はアポトーシス調節に関与するタンパク質からなる群から選択される異種タンパク質をコードする第2のDNA配列
を含むベクターを用いて形質転換された、組換えグラム陰性菌株。 - ベクターがプロテアーゼ切断部位をコードする第3のDNA配列を含み、第3のDNA配列が前記第1のDNA配列の3’末端と前記第2のDNA配列の5’末端の間に位置する、請求項1に記載の組換えグラム陰性菌株。
- 少なくとも1種のT3SSエフェクタータンパク質の産生に関して欠損を有する、請求項1又は2に記載の組換えグラム陰性菌株。
- エルシニア属及びサルモネラ属からなる群から選択される、請求項1から3の何れか一項に記載の組換えグラム陰性菌株。
- 組換えグラム陰性菌株がエルシニア属株であり、第1のDNA配列によりコードされる細菌T3SSエフェクタータンパク質由来の送達シグナルがYopEエフェクタータンパク質若しくはそのN末端断片を含むか、又は組換えグラム陰性菌株がサルモネラ属株であり、第1のDNA配列によりコードされる細菌T3SSエフェクタータンパク質由来の送達シグナルがSopEエフェクタータンパク質若しくはSteAエフェクタータンパク質若しくはそのN末端断片を含む、請求項1から4の何れか一項に記載の組換えグラム陰性菌株。
- 組換えグラム陰性菌株がエルシニア属株であり、前記エルシニア属株が野生型であるか若しくは少なくとも1種のT3SSエフェクタータンパク質の産生に関して欠損を有し、細菌T3SSエフェクタータンパク質由来の送達シグナルがY.エンテロコリチカ(Y.enterocolitica)YopEエフェクタータンパク質のN末端の138アミノ酸を含むか、又は、組換えグラム陰性菌株がサルモネラ属株であり、前記サルモネラ属株が野生型であるか若しくは少なくとも1種のT3SSエフェクタータンパク質の産生に関して欠損を有し、細菌T3SSエフェクタータンパク質由来の送達シグナルがS.エンテリカ(S.enterica)SteAエフェクタータンパク質若しくはS.エンテリカ(S.enterica)SopEエフェクタータンパク質のN末端の81アミノ酸若しくは105アミノ酸を含む、請求項1から4の何れか一項に記載の組換えグラム陰性菌株。
- 標識分子又は標識分子のアクセプター部位をコードするさらなるDNA配列を第2のDNA配列の5’末端又は3’末端と融合させた、請求項1から6の何れか一項に記載の組換えグラム陰性菌株。
- 核局在化シグナルをコードするさらなるDNA配列を第2のDNA配列の5’末端又は3’末端と融合させた、請求項1から7の何れか一項に記載の組換えグラム陰性菌株。
- 増殖に必須のアミノ酸の産生に関して欠損を有する、請求項1から8の何れか一項に記載の組換えグラム陰性菌株。
- 真核細胞表面又は細胞外マトリックスに結合する接着タンパク質の産生に関して欠損を有する、請求項1から9の何れか一項に記載の組換えグラム陰性菌株。
- 発現ベクターが、第1のDNA配列の3’末端及び/又は第2のDNA配列の5’末端若しくは3’末端に多重クローニング部位を含む、請求項1から10の何れか一項に記載の組換えグラム陰性菌株。
- 5’から3’の方向に、
プロモーター、
前記プロモーターに作動可能に連結した、細菌T3SSエフェクタータンパク質由来の送達シグナルをコードする第1のDNA配列、
前記第1のDNA配列の3’末端とインフレームで融合した、アポトーシス又はアポトーシス調節に関与する異種タンパク質をコードする第2のDNA配列、及び代替として
前記第1のDNA配列の3’末端と前記第2のDNA配列の5’末端の間に位置する、プロテアーゼ切断部位をコードする第3のDNA配列

- 細菌T3SSエフェクタータンパク質が、IpgB1、SopE、SopB、SptP、SteA、SifB、OspF、IpgD、YopH、YopO、YopP、YopE、及びYopTからなる群から選択される、請求項1から4及び請求項7から11の何れか一項に記載の組換えグラム陰性菌株又は請求項12に記載のベクター。
- 細菌T3SSエフェクタータンパク質が、SopE、SteA、及びYopEからなる群から選択される、請求項1から4及び請求項7から11の何れか一項に記載の組換えグラム陰性菌株又は請求項12に記載のベクター。
- 細菌T3SSエフェクタータンパク質がSteA又はYopEである、請求項1から4及び請求項7から11の何れか一項に記載の組換えグラム陰性菌株又は請求項12に記載のベクター。
- 異種タンパク質が、アポトーシス又はアポトーシス調節に関与するタンパク質、細胞周期制御因子、アンキリンリピートタンパク質、細胞シグナル伝達タンパク質、レポータータンパク質、転写因子、プロテアーゼ、低分子GTPアーゼ、GPCR関連タンパク質、ナノボディ融合構築物及びナノボディ、細菌T3SSエフェクター、細菌T4SSエフェクター、及びウイルスタンパク質からなる群から選択される、請求項15から17の何れか一項に記載の組換えグラム陰性菌株又はベクター。
- 異種タンパク質が、アポトーシス又はアポトーシス調節に関与するタンパク質、細胞周期制御因子、アンキリンリピートタンパク質、レポータータンパク質、低分子GTPアーゼ、GPCR関連タンパク質、ナノボディ融合構築物、細菌T3SSエフェクター、細菌T4SSエフェクター、及びウイルスタンパク質からなる群から選択される、請求項13から17の何れか一項に記載の組換えグラム陰性菌株又はベクター。
- 異種タンパク質が、アポトーシス又はアポトーシス調節に関与するタンパク質、細胞周期制御因子、及びアンキリンリピートタンパク質からなる群から選択される、請求項13から17の何れか一項に記載の組換えグラム陰性菌株又はベクター。
- アポトーシス又はアポトーシス調節に関与するタンパク質が、BH3−onlyタンパク質、カスパーゼ、及びアポトーシスのデスレセプター制御の細胞内シグナル伝達タンパク質からなる群から選択される、請求項1から11の何れか一項に記載の組換えグラム陰性菌株又は請求項12に記載のベクター。
- ベクターが、互いとは独立して前記第1のDNA配列の3’末端とインフレームで融合した同一の異種タンパク質又は2種の異なる異種タンパク質をコードする2つの第2のDNA配列を含む、請求項1から21の何れか一項に記載の組換えグラム陰性菌株又はベクター。
- 異種タンパク質の両方がアポトーシス又はアポトーシス調節に関与するタンパク質であり、一方のタンパク質がプロアポトーシスタンパク質であり、他方のタンパク質がアポトーシス防止経路の阻害剤であるか、又は、一方のタンパク質がプロアポトーシスタンパク質であり、他方のタンパク質が生存促進性シグナル伝達又は経路の阻害剤である、請求項22に記載の組換えグラム陰性菌株又はベクター。
- 第1のDNA配列によりコードされる細菌T3SSエフェクタータンパク質由来の送達シグナルが細菌T3SSエフェクタータンパク質又はそのN末端断片を含み、そのN末端断片が細菌T3SSエフェクタータンパク質の少なくとも最初の10アミノ酸を含む、請求項1から23の何れか一項に記載の組換えグラム陰性菌株又はベクター。
- 第1のDNA配列によりコードされる細菌T3SSエフェクタータンパク質由来の送達シグナルが細菌T3SSエフェクタータンパク質又はそのN末端断片を含み、細菌T3SSエフェクタータンパク質又はそのN末端断片がシャペロン結合部位を含む、請求項1から23の何れか一項に記載の組換えグラム陰性菌株又はベクター。
- プロモーターが前記グラム陰性菌株において機能するものである、請求項1から23の何れか一項に記載の組換えグラム陰性菌株又はベクター。
- 真核細胞に異種タンパク質を送達するための方法であって、
i)請求項1から11又は請求項13から26の何れか一項に記載のグラム陰性菌株を培養する工程と、
ii)真核細胞とi)のグラム陰性菌株を接触させる工程であり、細菌T3SSエフェクタータンパク質由来の送達シグナルと異種タンパク質とを含む融合タンパク質がグラム陰性菌株により発現され、真核細胞内に移行する工程と
を含む方法。 - iii)融合タンパク質を切断し、したがって、異種タンパク質を細菌T3SSエフェクタータンパク質由来の送達シグナルから切断する工程をさらに含む、請求項27に記載の方法。
- 融合タンパク質がインビトロ又はインビボにおいて真核細胞内に移行する、請求項27又は28に記載の方法。
- それぞれが細菌T3SSエフェクタータンパク質由来の送達シグナルと異種タンパク質とを含む少なくとも2つの融合タンパク質がグラム陰性菌株により発現され、真核細胞内に移行する、請求項27から29の何れか一項に記載の方法。
- 異種タンパク質を精製する方法であって、
請求項1から11又は請求項13から26の何れか一項に記載のグラム陰性菌株を培養し、したがって、細菌T3SSエフェクタータンパク質由来の送達シグナルと異種タンパク質とを含む融合タンパク質が発現され、培養物の上清中に分泌される工程
を含む方法。 - 異種タンパク質を医薬として又はワクチンとして対象に送達することに使用するための、請求項1から11又は請求項13から26の何れか一項に記載の組換えグラム陰性菌株。
- 移行した異種タンパク質(一又は複数)によって誘発される細胞経路又は事象に対する阻害剤のハイスループットスクリーニングのための、請求項1から11又は請求項13から26の何れか一項に記載の組換えグラム陰性菌株の使用。
- グラム陰性菌株の発現ベクターの第2のDNA配列によりコードされる異種タンパク質がヒトタンパク質又はマウスタンパク質であり、グラム陰性菌株によって発現されるヒトタンパク質又はマウスタンパク質それぞれのアミノ酸配列が異なる、請求項1から11又は請求項13から26の何れか一項に記載のグラム陰性菌株のライブラリー。
