Claims (21)
- バイオリアクター(2;12;32)と、音響定在波細胞分離器(5;15;35)と、フィルター(7;17;37)とを備える細胞培養用の装置(1;11;31)であって、バイオリアクターの出口(3;13;33)が音響定在波細胞分離器の入口(4;14;34)と流体連通し、音響定在波細胞分離器の培地出口(6;16;36)がフィルター(7;17;37)と流体連通しいる、装置。
- フィルターが、保持液側(20)及び透過液側(21)を有するクロスフローフィルター装置(17)であり、音響定在波細胞分離器(15)の培地出口(16)が保持液側の入口(22)と流体連通し、音響定在波細胞分離器の細胞濃縮液出口(18)及び保持液側の出口(23)がバイオリアクターの入口(19)と流体連通しており、当該装置が、透過液側(21)から透過液(24)を回収するように構成されている、請求項1に記載の装置(11)。
- クロスフローフィルター装置(17)が中空繊維フィルターカートリッジである、請求項2に記載の装置(11)。
- クロスフローフィルター(17)が、0.1〜5μmの公称細孔径を有する精密濾過膜又は10〜500kDのカットオフを有する限外濾過膜を含む、請求項2又は請求項3に記載の装置(11)。
- フィルター(7;17;37)がデプスフィルターである、請求項1に記載の装置(1;11;31)。
- 出口(33)が、バイオリアクター(32)から上清を吸引するように構成された吸引チューブである、請求項1乃至請求項5のいずれか1項に記載の装置(31)。
- 音響定在波細胞分離器(5;15;35)が、2以上の直列に連結された分離器チャンバーを備える、請求項1乃至請求項6のいずれか1項に記載の装置(1;11;31)。
- バイオリアクター(2;12;32)がフレキシブルバッグを備える、請求項1乃至請求項7のいずれか1項に記載の装置(1;11;31)。
- フィルター(7;17;37)と流体連通し、フィルター(7;17;37)から濾液(8;38)又は透過液(24)を受け入れるように配置された1以上の分離カラム(87;97,98)をさらに備える、請求項1乃至請求項8のいずれか1項に記載の装置(1;11;31)。
- 細胞培養の方法であって、
a)請求項1乃至請求項9のいずれか1項に記載の装置(1;11;31)を設けるステップと、
b)バイオリアクター(2;12;32)に培地及び細胞を導入するステップと、
c)バイオリアクター(2;12;32)で細胞を培養するステップと、
d)音響定在波細胞分離器(5;15;35)及びフィルター(7;17;37)を介して、濾液(8;38)又は透過液(24)を引き出すステップと
を含む方法。 - ステップa)が、請求項2又は請求項3に記載の装置(11)を設けることを含み、ステップd)あ、透過液(24)を引き出すこと、及びi)音響定在波細胞分離器(15)からの細胞濃縮液、及びii)クロスフローフィルター装置(17)からバイオリアクター(12)への保持液の両方を再利用することを含む、請求項10に記載の方法。
- ステップa)は、請求項6に記載の装置(31)を設けることを含み、当該方法が、ステップd)の前に、凝集剤又は沈殿剤をバイオリアクター(2;12;32)に添加すること、及び上清及び沈殿を生成できるようにすることからなるステップc’)をさらに含む、請求項10に記載の方法。
- ステップc)の少なくとも一部で、バイオリアクター(2;12;32)の細胞濃度が10×106細胞/ml以上、例えば15×106細胞/ml以上である、請求項10乃至請求項12のいずれか1項に記載の方法。
- ステップc)の少なくとも一部で、バイオリアクター(2;12;32)において細胞で発現されたターゲットタンパク質の濃度が5g/l以上である、請求項10乃至請求項13のいずれか1項に記載の方法。
- バイオリアクター(82;92)と、音響定在波細胞分離器(85;95)と、1以上の分離カラム(87;97,98)とを備える生体分子回収用の装置(81;91)であって、バイオリアクターの出口(83;93)が音響定在波細胞分離器(85;95)の入口(84;94)と流体連通し、音響定在波細胞分離器の培地出口(86;96)が1以上の分離カラム(87;97,98)と流体連通しいる、装置(81;91)。
- 培地出口(86;96)と1以上の分離カラム(87;97,98)との間にフィルターを備えていない、請求項15に記載の装置(81;91)。
- 擬似移動床又は周期的向流プロセスのような連続分離に適した複数の分離カラム(87)を備える、請求項15又は請求項16に記載の装置(81)。
- 1以上の分離カラム(87;97,98)が1以上の吸着流動床カラムである、請求項15乃至請求項17のいずれか1項に記載の装置(81;91)。
- 1以上の分離カラム(87;97,98)が、体積加重平均粒径80μm以上の粒子のような分離マトリックス粒子からなる充填床を含む、請求項15乃至請求項18のいずれか1項に記載の装置(81;91)。
- 分離マトリックス粒子が充填されたガードカラム(97)の入口(99)が音響波細胞分離器(95)の培地出口(96)と流体連通し、ガードカラムの出口(100)が分離マトリックス粒子が充填されたメインカラム(98)の入口(101)と流体連通しいる、請求項19に記載の装置(91)。
- 細胞培養物から生体分子を回収する方法であって、
a)請求項15乃至請求項17のいずれか1項に記載の装置(81;91)を設けるステップと、
b)バイオリアクター(82;92)に培地及び細胞を導入するステップと、
c)細胞培養物を生成するために、バイオリアクター(82;92)で細胞を培養するステップと、
d)音響定在波細胞分離器(85;95)に、細胞培養物の少なくとも一部分を引き出すステップと、
e)細胞除去フラクションを生成するために、音響定在波細胞分離器(85;95)の細胞培養物から細胞を分離するステップと、
f)細胞除去フラクションを1以上の分離カラム(87;97,98)に搬送するステップと
を含む方法。
