Abstract
Description
1.グリセロール及びソルビトールを混合する。
2.研磨剤を添加する。
3.KNO3、TiO2、サッカリン、スクラロース、フッ化ナトリウム、及び水酸化ナトリウムを添加する。
4.増粘シリカを添加する。
5.界面活性剤を添加する。
6.ガムを添加する。
7.香味オイルを添加する。
試料の調製
本研究において使用された手順は、ADA及びISO 11609により推奨される、歯磨き剤の相対的な象牙質研磨性を測定するためのHefferren研磨性試験であった。
前処理を実行した後、「サンドイッチデザイン」を使用して、試験を実施した(150g、1500ストローク)。歯磨き剤スラリー(25g/40ml水)でブラッシングする前後に、各歯のセットをADA参照物質(Ca2P2O710g/0.5%のCMC 50ml)でブラッシングした。各製品が、各歯のセットにおいて評価されるように、この手順を複数回繰り返した。処理デザインは、処理が別の処理に一貫して続かないように変更したラテン方格(Latin Square)デザインであった。
1mlの試料を取り、重さを量り(0.01g)、「Ultima Gold」シンチレーションカクテル4.5 mlに添加した。試料をよく混合し、放射線測定のため、すぐにシンチレーションカウンターに置いた。カウント後、1分あたりの正味のカウント(counts per minute:CPM)値を、試料の重量で割ることにより、スラリー当たりの正味のCPM/グラムを計算した。その後、各試験スラリーについて、ADA参照物質の前後の正味のCPM/gを計算し、平均して、試験物質についてのRDA(相対的象牙質摩耗性)の計算に使用した。ADA参照物質の値を100として定め、試験物質に対するその割合を計算した。
PCR法
試料の調製
ウシの永久中切歯を切り取り、約10×10mmの唇側のエナメル質の試料を得た。その後、エナメル質の表面のみが露出するように、エナメル質の試料を常温重合メタクリレート樹脂に埋め込んだ。その後、宝石加工ホイール(lapidary wheel)上で、エナメル質の表面を滑らかにし、艶出しして、軽くエッチングしてステインの蓄積及び付着を促進させた。試料を(37℃のインキュベーター中の)回転ロッド上に置き、空気と、トリプチケースソイブロス、茶、コーヒー、ムチン、FeCl3、及びマイクロコッカス・ルテウス(Micrococcus luteus)BA13からなる溶液とに、試料を交互に接触させた。着色ブロスを交換し、試料を7日間毎日洗った。12日後、暗く着色されたペリクル膜がエナメル質の表面に見られた。その後、試料を洗い、空気乾燥させ、使用するまで冷蔵した。同時に調製した試料を使用して、全ての製品を試験した。
in vitroのステイン量を、分光光度計(Minolta CM2600d.)を使用して、L*a*b*スケールのL値のみを使用して、光度測定的に評価した。スコアリングした試料の領域は10×10mmのエナメル質の中心の1/4インチ径の円であった。30〜42のスコアを有する試料(30はより暗く着色されている)を使用した。これらのスコアに基づいて、試料を、各群が同一の平均基準スコアを有する、それぞれ16個の試料を有する群に分けた。
その後、試料を、柔らかいナイロン繊維(Oral-B 40)の歯ブラシを備えた機械であるV-8 交叉ブラッシング機に取り付けた。エナメル質の表面へのブラッシング力を150gに調節した。歯磨き剤を、歯磨き剤25gと脱イオン水40mlを混合することにより調製したスラリーとして使用した。ADA摩耗参照物質(Ca2P2O7)を、物質10gを0.5%のCMC溶液50ml中に混合することにより調製した。試料を800ストローク(4 1/2分)でブラッシングした。機械的変動を最小化するために、1つの群につき1つの試料を8個のブラッシングヘッドのそれぞれでブラッシングした。4つの試料をブラッシングするのに使用した後、新しいスラリーを調製した。ブラッシング後、試料を洗い、拭いて乾燥させ、前記のようにステインを再度スコアリングした。
ブラッシング前後のステインのスコアの差を決定し、参照群についての平均及び標準誤差を計算した。参照物質群についての洗浄率を100として定めた。参照群の平均減少量を100で割って、試験における個々の試験の減少量に乗じられる定数値を得た。その後、各試料の個々の洗浄率を計算した(減少量×定数)。その後、各群(N=16)について、平均及びSEMを、個々の洗浄率を使用して計算した。洗浄率が大きくなればなるほど、本試験において除去された着色ペリクルの量は多くなる。
REA法
試料の調製
ADAにより概説される制御された条件下で、8個のヒトのエナメル質の試料に中性子衝撃を与えて、試料中に放射性リン(32P)を形成させた。V-8交叉ブラッシング機に取り付けられるように、試料をメチルメタクリレートに埋め込んだ。試料を、0.5%のCMCグリセリン溶液50ml中のADA参照物質10gからなるスラリーを使用して、5000ストロークでブラッシングして、前処理を実行した。使用したブラシは、ブラシ張力150gの、ADAにより指定されたものであった。
前処理を実行した後、「サンドイッチデザイン」で、上記パラメーター(150g、5000ストローク)を使用して、試験を実施した。試験製品(製品 25g/水 40ml)でブラッシングする前後に、各歯のセットをADA参照物質(Ca2P2O710g/0.5%のCMC 50ml)でブラッシングした。各製品が各歯のセットにおいて評価されるように、この手順を複数回繰り返した。処理デザインは、処理が別の処理に一貫して続かないように変更したラテン方格デザインであった。

CEIデータ
次に、上記のPCR及びRDA試験で得られた値を使用して、以下の洗浄効率指標(CEI)値(CEIは、(PCR-50+RDA)/RDAである)を得た。
Claims (10)
- 研磨剤としての球状の溶融酸化アルミニウム粒子と、経口的に許容できる担体とを含む歯磨き剤組成物。
- 球状の溶融酸化アルミニウムの少なくとも90%が、球状である、請求項1に記載の組成物。
- 球状の溶融酸化アルミニウム研磨剤のメジアン粒径が、約1〜約15ミクロン(最大スパンは約2.5である)の範囲である、請求項1又は2に記載の組成物。
- 球状の溶融酸化アルミニウム研磨剤が、組成物総量の約0.01重量%〜約5.0重量%の量で存在する、請求項1〜3のいずれか一項に記載の組成物。
- 補足的な研磨剤を含む、請求項1〜4のいずれか一項に記載の組成物。
- 球状の溶融酸化アルミニウムが、歯磨き剤における唯一の研磨剤である、請求項1〜4のいずれか一項に記載の組成物。
- 水溶性縮合リン酸塩を含む、請求項1〜6のいずれか一項に記載の組成物。
- 縮合リン酸塩が、水溶性アルカリ金属トリポリリン酸塩である、請求項7に記載の組成物。
- 脱感作剤を含む、請求項1〜8のいずれか一項に記載の組成物。
- フッ化物イオン源を含む、請求項1〜9のいずれか一項に記載の組成物。
