Abstract
Description
上述したように、本発明の態様は、老化に関連する状態について対象を処置する方法を含んでいる。老化に関連する状態は、生物の老化に付随する状態、例えば疾患の状態又は他の好ましくない状態を意味する。老化に関連する状態は、多くの異なる方法で、例えば身体の中心の臓器又は末梢の臓器の老化に関連する損傷として現れる場合があり、このような損傷として、細胞損傷、組織損傷、臓器機能不全、老化に関連する寿命の短縮及び発癌があるが、これらに限定されず、注目する特定の臓器及び組織は、皮膚、ニューロン、筋肉、膵臓、脳、腎臓、肺、胃、小腸、脾臓、心臓、脂肪組織、精巣、卵巣、子宮、肝臓及び骨が含まれるが、これらに限定されない。場合によっては、本方法に従った対象の処置により、中心的臓器、例えば脳、脊髄などの中枢神経系臓器に変化がもたらされ、このような変化は、例えば以下に更に詳細に記載されるように、分子的、構造的及び/又は機能的を含むがこれらに限定されない多くの異なる方法で現れる場合がある。場合によっては、本方法に従った対象の処置により、肝臓、筋肉、心臓、血液などの末梢の臓器に変化がもたらされ、このような変化は、例えば以下に更に詳細に記載されるように多くの異なる方法で現れる場合がある。
本方法、及び若い血漿を含有する本血液製剤は、個体の認知能力の障害のような老化に関連する状態の予防を含む処置の際に使用される。例えば本明細書に開示されている方法によって、血漿を含有する本血液製剤での処置によって恩恵を受ける老化に関連する認知障害を患うか又は発症する危険性がある個体は、約50歳以上、例えば60歳以上、70歳以上、80歳以上、90歳以上であって、通常100 歳以下、つまり約50歳と100 歳との間の年齢、例えば50、55、60、65、70、75、80、85、90、95又は約100 歳であり、自然老化現象に関連した認知障害、例えば軽度認知障害(M.C.I.)を患う個体と、約50歳以上、例えば60歳以上、70歳以上、80歳以上、90歳以上であって、通常100 歳以下、つまり約50歳と90 歳との間の年齢、例えば50、55、60、65、70、75、80、85、90、95又は約100 歳であり、認知障害の症状をまだ示し始めていない個体とを含んでいる。自然老化による認知障害の例として、以下が含まれる。
アルツハイマー病は、b-アミロイドとタウタンパク質から成る神経原線維濃縮体とを更に含んでいる、大脳皮質及び皮質下灰白質内の過剰な数の老人斑に関連した認知機能の進行性の止められない消失である。一般的な形態は60歳を超える人に発症し、年齢が進むにつれてその発生率が増加する。アルツハイマー病は、高齢者の認知症の65%を超える割合を占める。
因子は活発な研究の焦点である。
パーキンソン病(PD)は、緩慢な動作、筋固縮、静止振戦及び姿勢の不安定によって特徴付けられる特発性で、緩徐進行性の中枢神経系の変性疾患である。当初は主に運動障害とみなされていたが、PDは現在、認知、行動、睡眠、自律神経機能及び感覚機能に更に影響を及ぼすと認識されている。最も一般的な認知障害として、注意及び集中、作業記憶、実行機能、言語の生成並びに視空間機能の障害が含まれる。
前頭側頭型認知症(FTD )は、脳の前頭葉の進行性悪化に起因する状態である。変性は、側頭葉に経時的に進む場合がある。FTD は、有病率でアルツハイマー病(AD)に次いで2番目であり、初老期の認知症の症例の20%を占める。症状は、影響を受けた前頭葉及び側頭葉の機能に基づき3つのグループ:一方で嗜眠及び自発性喪失を含み、他方で脱抑制を含む症状を有する行動異常型FTD (bvFTD );明瞭な発音の困難、音韻体系の誤り及び/又は構文誤りによる発語の流暢性の崩壊が観察されるが、言葉の理解は保持されている進行性非流暢性失語(PNFA);及び、患者は正常な音韻論及び構文で流暢なままであるが、呼称及び言葉の理解で困難さが増す意味性認知症(SD)に分類される。全てのFTD 患者に共通する他の認知症状として、実行機能及び集中力の障害が含まれる。知覚、空間能力、記憶及び練習を含む他の認知能力が典型的には損なわれないままである。FTD は、構造的MRI スキャンにおいて明らかになる前頭葉及び/又は前側頭葉の萎縮を観察することにより診断され得る。
ハンチントン病(HD)は、情緒異常、行動異常及び精神異常の発症と、知的機能又は認知機能の消失と、動作異常(運動障害)とにより特徴付けられる遺伝性の進行性神経変性障害である。HDの古典的な徴候として、顔、腕、脚又は体幹に影響を及ぼす場合がある付随性で急速且つ不規則な痙攣様運動である舞踏病の発症と、思考過程能力及び後天性の知的能力を徐々に消失する認知機能低下とが含まれる。記憶、抽象的思考及び判断の障害;時間、場所又はアイデンティティの不適切な知覚(失見当識);激越の増加;及び人格変化(人格の崩壊)がある場合がある。症状は典型的には30歳代又は40歳代の間に明らかになるが、発病の年齢は不定的であり、幼児期から成人後期(例えば70歳代又は80歳代)の範囲内である。
筋萎縮性側索硬化症(ALS )は、運動ニューロンを攻撃する急速に進行して常に致死的な神経疾患である。筋力低下と前角細胞の萎縮及び機能障害の徴候とは最初、ほとんどの場合手に認められ、足に認められることはより少ない。発病の箇所はランダムであり、進行は非対称である。痙攣がよく見られ、衰弱より先に生じる場合がある。例外的に、患者は30年間生存し、50%が発病後3年以内に死亡し、20%が5年間生存し、10%が10年間生存する。診断上の特徴として、中年期又は成人後期中の発病、及び感覚異常のない進行性で全身性の運動障害が含まれる。この疾患の末期まで、神経伝達速度は正常である。最近の研究は、同様に認知障害の症状、特に即時性の言語記憶、視覚記憶、言語及び実行機能の低下を実証した。
多発性硬化症(MS)は、寛解及び再発性の悪化と共に、中枢神経系の機能障害の様々な症状及び徴候によって特徴付けられる。最も一般的な主症状は、一又は複数の四肢、体幹又は顔の一側における感覚異常;脚又は手の衰弱又は失調;又は、視覚障害、例えば部分盲及び一方の目の痛み(球後視神経炎)、視覚の薄暗さ又は暗点である。一般的な認知障害として、記憶(新しい情報の取得、保持及び検索)、注意及び集中(特に分割的注意)、情報処理、実行機能、視空間機能及び言語流暢性の障害が含まれる。一般的な初期症状は、眼筋麻痺による複視(double vision )(複視(diplopia))、一又は複数の四肢の一時的な衰弱、肢の僅かな硬直又は異常な疲労感、軽微な歩行障害、膀胱制御困難、回転性めまい、及び軽微な情緒障害であり、全ての症状は、散在性の中枢神経系障害を示し、疾患が認識される前の何ヶ月又は何年も前に生じることが多い。過剰な熱が症状及び徴候を際立たせる場合がある。
緑内障は、網膜神経節細胞(RGC )に影響を及ぼす一般的な神経変性疾患である。RGC を含むシナプス及び樹状突起において区画化された変性プログラムが存在することをサポートする証拠がある。最近の証拠は、高齢者の認知障害と緑内障との相関関係を更に示している(Yochim BP等著、「Prevalence of cognitive impairment, depression, and anxiety symptoms among older adults with glaucoma.」、J Glaucoma. 2012;21(4):250-254)。
筋強直性ジストロフィ(DM)は、ジストロフィ性筋力低下及び筋強直症によって特徴付けられる常染色体優性多系統障害である。分子欠損は、染色体19q の筋強直タンパク質キナーゼ遺伝子の3'非翻訳領域の伸長トリヌクレオチド(CTG )反復である。症状はあらゆる年齢で生じる場合があり、臨床重症度の範囲は広い。筋強直症は手の筋肉で顕著であり、軽度な症例でも眼瞼下垂が一般的である。重度な症例では、著しい末梢の筋低下が、多くの場合、白内障、早期の脱毛、斧状顔貌、不整脈、精巣萎縮及び内分泌異常(例えば糖尿病)と共に生じる。精神遅滞は重症先天性の形態で一般的である一方、前頭部及び側頭部の認知機能、特に言語機能及び実行機能の加齢性の低下は、障害のより軽症な成人の形態に観察される。重症の人は、50代前半までに死亡する。
認知症は、日常機能を妨げるのに著しく十分な思考能力及び社会能力に影響を及ぼす症状を有する障害のクラスのことである。上記に記載された老化に関連する障害の後期段階で観察される認知症に加えて、認知症の他の症例として、以下に述べる血管性認知症と、レヴィ小体型認知症とが含まれる。
進行性核上性麻痺(PSP )は、複雑な目の動き及び思考の問題と共に、歩行及びバランスの制御に関する重大で進行性の問題を引き起こす脳障害である。この疾患の古典的な徴候の内の1つは、目を適切に向ける能力の欠如であり、目の動きを調整する脳の領域の病変のために生じる。一部の個体はこの影響をかすむと表現する。発症した個体は、抑うつ及び無感情並びに進行性の軽度認知症を含む、気分及び行動の変化を示すことが多い。この疾患の長い名前は、疾患がゆっくり始まり悪化し続け(進行性であり)、目の動きを制御する細胞核と称される豆粒大の構造の上方の脳のある部分(核上)を損傷することにより衰弱(麻痺)を引き起こすことを示している。3人の科学者がこの状態をパーキンソン病と区別する論文を発表した1964年に、PSP が別個の障害として最初に表現された。PSP は、この障害を定めた科学者の名前の組合せを反映して、スティール−リチャードソン−オルスゼフスキー症候群と称されることもある。PSP は次第に悪化するが、PSP 自体で死亡する人はいない。
運動失調を呈する人は、動き及びバランスを制御する神経系の部分に変調をきたすため、調整に関する問題を有する。運動失調は、指、手、腕、脚、身体、発語及び目の動きに影響を及ぼし得る。運動失調という言葉は、感染、損傷、他の疾患又は中枢神経系の変性変化と関連付けられ得る協調運動障害の症状を表現するために使用されることが多い。運動失調は更に、National Ataxia Foundationの主たる重点である遺伝性及び散発性の運動失調と称される神経系の特異的な変性疾患のグループを示すために用いられる。
複合神経系萎縮(MSA )は変性神経疾患である。MSA は、脳の特定の領域の神経細胞の変性と関連付けられる。この細胞変性は、動き、バランス、及び膀胱の制御又は血圧の調節のような身体の他の自律神経機能に関する問題を引き起こす。MSA の原因は分かっておらず、特異的な危険因子が識別されていない。症例の約55%が男性で生じ、発症する典型的な年齢は50代後半から60代前半である。MSA は、パーキンソン病と同じ症状の一部を示すことが多い。しかしながら、MSA 患者は、パーキンソン病に使用されるドーパミン剤に反応するにしても、一般にごく僅かしか反応しない。
更に、上記の方法の内の一又は複数を実施するための試薬、デバイス及びキットを提供する。本試薬、本デバイス及び本キットは大幅に異なってもよい。注目する試薬及びデバイスは、血漿を含有する血液製剤をその血液製剤を必要とする対象に輸血するために調製する方法に関して上述した試薬及びデバイス、例えば抗凝血剤、凍結保存料、緩衝液、等張液などを含んでいる。
以下の実施例は、本発明をどのように作製して使用するかについての完全な開示及び説明を当業者に提供するために記載されるのであって、本発明者が自身の発明であるとみなすものの範囲を限定することを意図するものではなく、また、以下の実験が、行われた全ての又は唯一の実験であると表すことを意図するものでもない。使用された数値(例えば、量、温度等)に関して正確性を確保するために努力がなされたが、ある程度の実験誤差及び偏差は考慮に入れられるべきである。別途記載のない限り、部は重量部であり、分子量は重量平均分子量であり、温度は摂氏であり、圧力は大気圧であるか又は大気圧に近い圧力である。
本明細書では、老齢動物を若い血液に曝すことにより、構造レベル、機能レベル及び認知レベルで以前から存在する脳の老化の影響を弱め得ることが示されている。若齢動物及び老齢動物の循環系が結合された異年齢の並体結合体のゲノム全体に亘るマイクロアレイ解析を用いて、老齢マウスの海馬の可塑性の向上を示す発現プロファイルが識別された。更に、成熟ニューロンの樹状突起スパインの密度の構造的向上、及びシナプス可塑性の機能的な改善が、老いた異年齢の並体結合体の海馬で観察された。最後に、若い血液血漿を老齢マウスの静脈内に与えることにより、文脈的恐怖条件付けと空間の学習及び記憶との両方で加齢性の認知障害が改善された。更に、これらのデータは、後年期に若い血液に曝すことにより、老いた脳の加齢性の変化を反転させ得ることを示している。
動物
C57BL/6 の若齢マウス(Jackson Laboratory)及びC57BL/6 の老齢マウス(National Institutes on Aging)を、特定病原体フリーの条件下で12時間の明暗サイクルで収容し、全ての動物の取り扱い及び使用については、the VA Palo Alto Committee on Animal Researchによって承認された組織的ガイドラインに従った。
同年齢の並体結合体及び異年齢の並体結合体からの海馬を解剖して、RNA 全体をTrizol試薬(Invitrogen)を使用して抽出した。cDNA及びcRNAを、製造業者のプロトコルに従ってRNA 増幅キット(Ambion)を使用して連続的に合成して増幅した。その後、cRNAを、製造業者の指示に従ってIllumina beadchip array MouseWG-6 v2.0(Illumina)にハイブリッド形成した。製造業者のガイドラインに従ってIllumina beadstudioデータ解析ソフトウェア(Illumina)によってデータを解析した。Zスコアのデータセットの無監督の階層的クラスタリングのためにCluster 3.0 ソフトウェアを使用した。ヒートマップを生成するために、Java(登録商標) TreeViewソフトウェアを使用した。「stat.stanford.edu/~tibs/SAM/index.htm」の前に「www.」を置くことによりワールド・ワイド・ウェブで見出されるSignificance Analysis of Microarray software(SAM )3.00アルゴリズムに基づく、P<0.01及び絶対のd-score>2 (図2b)又はd-score>1.5 (図2b)夫々でのカットオフをデータセット解析のために適用した。著しく変化したプローブセットを、Ingenuity Pathway Analysis(IPA; Ingenuity Systems, www.ingenuity.com)を使用して統計的に高められた経路に関して解析して、 「godatabase.org/cgi-bin/amigo/go/cgi」の前に「www.」を置くことによりワールド・ワイド・ウェブで見出されるAmiGO (The Gene Ontology Consortium)を使用して生物学的機能に関して分類した。
組織処理及び免疫組織化学法を、標準的な公開された技術(Ruckh et al., Rejuvenation of Regeneration in the Aging Central Nervous System, Stem Cell 10, 96-103 (2012))に従って浮遊性切片で行った。マウスを抱水クロラール(Sigma-Aldrich)を用いて麻酔し、0.9 %の生理食塩水を用いて経心的に灌流し、脳を除去して、リン酸緩衝の4%のパラホルムアルデヒドに48時間固定してから、30%のスクロースを用いて凍結保護した。ウサギ抗Egr-1 (1:500;Santa Cruz)一次抗体、ウサギ抗cFos(1:500;Millipore )一次抗体又はウサギ抗ホスホCREB(1:5000; Oncogene Research Products)一次抗体のいずれかを用いて浮遊性冠状切片(40μm)を一晩培養し、ビオチン化二次抗体とジアミノベンジジン(DAB, Sigma-Aldrich)を有するABC キット(Vector)とを用いて染色を明らかにした。個々の細胞数を定量化し、Egr-1 及びcFos並びにリン酸化されたCREBを、NIH ImageJソフトウェアを使用して平均信号強度として定量化した。
脳を除去した後、10mlの「modified Golgi-Cox staining solution」(Jing, Deqiang and Lee, Francis, Cornell University)に半球を暗所で7〜10日間室温で浸した。脳をdH2O中で30%のスクロースに4℃で72時間導入した。切片(150 μm)をdH2O中で0.3 %のゼラチンで覆われたスライドに取り付けた。短時間乾燥した後、スライドを40%のスクロースに3回浸漬し、暗所で72時間空気乾燥した。スライドをdH2Oに10分間3回ゆるく振り混ぜながら浸し、次に展開液に6分間導入した。その後、スライドをdH2O中で10分間3回洗浄し、等級付けされたエタノールにより脱水し、Histoclearに浸し、その後DPX 封入剤を使用してカバーガラスで封入した。ニューロンを100Xで追跡し、Neurolucida ソフトウェア(v10, MBF Bioscience)を使用してその後の全ての解析を行った。細胞体から10μmの間隔を離して同心球を置くことにより、ショール解析をニューロン毎に行った。樹状突起が各球と交差する回数、及び各球内の樹状突起の全長を定量化した。樹状突起の長さを、x面、y面及びz面における距離に亘って、及び各半径内に含まれるニューロンの複数の樹状突起の分岐に亘って合計した。
上述したように細胞外電気生理処置を行った(Rosenzweig, E.S. & Barnes, C.A., (2003) Impact of aging on hippocampal function: plasticity, network dynamics, and cognition. Progress Neurobiol. 69(3), 143-179)。老齢の並体結合体から鋭い海馬スライス(400 μmの厚さ)を調製した。スライスを、人工脳脊髄液(ACSF; in mM: NaCl 124.0; KCl 2.5; KH2PO4 1.2; CaCl2 2.4; MgSO4 1.3; NaHCO3 26.0;グルコース10.0)に保持し、5%CO2 /95%O2で連続的に酸素添加した。Axopatch-2B 増幅器及びpClamp 10.2 ソフトウェア(Axon Instruments)を用いて記録した。水浸のスライスを、酸素添加されたACSFを用いて2ml/分の流速でリザーバから重力供給によって連続的に灌流した。ACSFが充填されたガラス微小電極(抵抗:4〜8MΩ)を用いて電場電位(集合スパイク)を記録した。二相電流パルス(一相で0.2 msの継続時間、合計で0.4 ms)を、同心バイポーラ刺激電極(FHC, Inc.)を通して10秒間隔で与えた。この頻度の刺激ではシナプスの明らかな低下又は促進が観察されなかった。歯状回の集合スパイクを記録するために、記録電極を、歯状回の背側部の側面又は内面に置いた。貫通線維路の線維を刺激するために、海馬裂の真上に刺激電極を置いた。信号を1kHzでフィルタし、10 kHzでデジタル化した。2列の100 パルス(0.4 msのパルス継続時間、100 Hz)からなる強縮刺激を、5秒の列間の間隔で与えた。集合スパイクの振幅を、陰性波の初位相から測定した。5回までの連続した追跡を測定毎に平均した。刺激強度を最大の30%以下になるように調節して、入出力曲線を生成することによりシナプス伝達を評価した。基底振幅に関する高頻度な刺激の後、集合スパイクの振幅の平均パーセントの変化としてLTP を計算した。
パラダイムは、既に公開されている技術(Alberini, C.M., Transcription factors in long-term memory and synaptic plasticity. Physiol. Rev. 89(1), 121-145 (2009))に従った。マウスは、海馬に依存する文脈的恐怖条件付けに関するテストを可能にする嫌悪刺激(軽度なフットショック;無条件刺激(US))と環境的文脈(恐怖条件付けチャンバ)とを関連付けることを学習した。扁桃体に依存する手がかりの恐怖条件付けを更に評価するために、軽度なフットショックを光及び音の手がかり(条件刺激(CS))と対にした。条件付け恐怖を、フリージング行動として表示した。具体的な訓練パラメータは、音の継続時間30秒;レベル70dB,2kHz ;ショックの継続時間2秒;強度0.6 mAである。1日目、各マウスを、恐怖条件付けチャンバに置いて2分間探検させた後、2秒のフットショック(0.6 mA)で終わる30秒の音(70dB)を付与した。2分後、2回目のCS−US対を与えた。2日目、全く同一の文脈を含むが、CS又はフットショックを与えない恐怖条件付けチャンバに各マウスをまず置いた。フリージングを1〜3分間解析した。1時間後、異なる臭気、洗浄液、床の質感、チャンバの壁及び形状を含む新たな文脈にマウスを置いた。動物を、CSに再度曝す前に2分間探検させた。フリージングを1〜3分間解析した。FreezeScanビデオ追跡システム及びソフトウェア(Cleversys, Inc)を使用してフリージングを測定した。
パラダイムは、既に記載されているプロトコル(Jones, M.W. et al. A requirement for the immediate early gene Zif268 in the expression of late LTP and long-term memories. Nat. Neurosci. 4(3), 289-296 (2001))に従った。プラットフォームを含むゴールのアーム位置は訓練段階及びテスト段階を通して一定のままであるが、スタートのアームは各試験中に変わっている。訓練段階の1日目、マウスを15の試験に関して訓練し、試験は目視可能なプラットフォーム及び隠したプラットフォームに関して交互に行う。テスト段階の2日目、マウスを、隠したプラットフォームについて15の試験に関してテストする。正しくないアームへのエントリをエラーとして採点し、エラーを訓練ブロック(3回の連続した試験)について平均する。
200 〜300 の若齢(3ヶ月の)マウス又は老齢(18ヶ月の)マウスから、屠殺の際の心臓内出血によって、プールするマウス血漿を収集した。EDTAを用い、その後遠心分離により収集された血液から血漿を調製した。アリコートを、使用まで−80℃で保管した。EDTAと3.5kDa以下の平均分子量のタンパク質とを除去するために、血漿を与える前、3.5kDa分子量排除膜を通して血漿をPBS 中で透析した。若齢マウス又は老齢マウスから分離された血漿(1回の注射当たり100 μl)を用いて、若齢成体マウスを尾静脈への静脈注射により24日間に亘って8回全身的に処置した。
データを平均±SEM として表現する。Prism 5.0 ソフトウェア(GraphPad Software)を用いて統計解析を行った。2つのグループの平均を、両側不対Student のt検定と比較した。複数のグループからの平均の相互比較、又は1つの対照グループとの比較を、1-way ANOVA 検定及びボンフェローニ事後検定を用いて解析した。行われた組織構造実験、電気生理実験及び行動実験は全て、無作為な盲検法で行った。
ヒト及びマウスでは、海馬は、老化作用を特に受けやすい脳の領域であり、空間的及び突発的な認知機能の障害をもたらす形態変化及び可塑性の低下を示す(Andrews-Hanna, J.R. et al. Disruption of large-scale brain systems in advanced aging. Progress Neurobiol. 69(3), 143-179 (2003))。従って、本発明者らは、異年齢の並体結合(図1a)及び血液血漿の投与(図3a)の組合せを用いて分子レベル、構造レベル、機能レベル及び認知レベルでマウスの老化した海馬の範囲内に存在する若い血液に曝すことがもたらす潜在的な利点について研究した。
認知に関する若返り効果を調節する際の若い血漿にある可溶性因子の必要性を評価するために、本発明者らは、若い血漿又は熱変性血漿の尾静脈注射によって老齢マウス(18ヶ月)を処置した。血液循環にある負因子を弱めることにより認知機能改善を生じさせ得る可能性を評価するために、PBS を対照として注射した。若齢(8週間の)C57Bl/6Jマウスから血漿を収集して、3.5kDa分子量排除膜を用いた透析の前にプールした。血漿の一部を95℃で2〜3分間の変性によって熱変性した。マウスに数週間に亘って8回注射し、海馬に依存する記憶を評価するために恐怖条件付けパラダイムを行った。光及び音と対の軽度なフットショックをマウスに与えた訓練の1日目の後、マウスに同一の文脈を行って、フリージング行動を評価した。その後、ケージ環境を変えて、海馬に依存しない(手がかりの)記憶を評価するために、フリージング行動を測定した。図9に実証されているように、若い血漿の変性は、老齢マウスの認知能力に対する若い血漿のプラス効果を消失させた(図9)。
若い血漿の付与を最適化するために、認知テスト及び組織学的解析の前に2ヶ月のマウスからの血漿を18ヶ月のマウスに1週間に1回のみ3週間与える(1回の注射当たり150 μl)実験を行った(図8)。このような研究は完全な盲検法で行った。図10に示されているように、本発明者らは、文脈的恐怖条件付けパラダイムのフリージングの増加、RAWMのより優れた動作、及び、若い血漿で処置したマウスの多くのBrdU+ニューロンが生理食塩水で処置した同年齢の対照と比較して2倍であることを観察した。
アルツハイマー病をモデル化したマウスに若い健康な循環環境を与えることにより、このようなモデルに関連していると知られ、疾患のある人間の患者に同様に生じると知られている神経異常を低下させることが、本明細書に示されている。
無処置の血漿のタンパク質及びタンパク質の分子のサブセットを包含する若い血漿の定められた画分が、シナプス可塑性遺伝子ネットワークを活性化するのに十分であることが本明細書には示されている。更に、分子の大きさが非常に異なるタンパク質又は分子が脳と伝達して、海馬の遺伝子発現の変化を引き起こし得る。2〜3ヶ月のマウスから採取されプールされたマウスの血漿は、定められた分子量(3.5kDa、25 kDa、50 kDa)のカットオフを有する透析膜を使用して、血漿の成分を含む分子量におおよそ基づく3.5kDaより大きい画分、25 kDaより大きい画分及び50 kDaより大きい画分にサイズによって分離した。3.5kDaより大きい画分を使用して、タンパク質G親和性沈降によりIgG 免疫グロブリンを消失させることにより、追加の画分を生成した。各画分を、18ヶ月のマウス(画分当たりn=4〜5)に2週間に亘って7回静脈内に注射し(1回の注射当たり125 μl)、対照としてリン酸緩衝の生理食塩水(PBS )を注射した。処置の終わりに、脳を解剖し、海馬のRNA を抽出してアフィメトリクス遺伝子アレイを使用して解析した。PBS で処置されたマウスと比較すると、数百の遺伝子が、血漿画分のいずれかで処置されたマウスの脳で著しく変化した(図13)。ソフトウェアツールのIngenuity Pathway Analysis(登録商標)(IPA )を用いたバイオインフォマティックス解析は、シナプス可塑性並びに学習及び記憶に関する複数のネットワーク(例えば、長期増強、神経炎の分岐、行動)が、25 kDaより大きい画分で著しく高められたが、他の画分では高められなかったか、又はそれ程高められなかったことを明らかにした。これらのネットワーク中の顕著な遺伝子は、リーリン、神経栄養チロシンキナーゼ3受容体及びエフリン受容体4A(表1)を含む学習及び記憶における既知のプレーヤを含んでいる。
老齢マウスに若い血漿を与えることにより、血液の全身的な変化がもたらされ、処置の生物全体に亘る効果を奏していることが、本明細書に示されている。具体的には、12ヶ月の老齢マウスに、2ヶ月のマウスからの血漿を1週間に2回、合計4週間注射した(1つのグループ当たりn=13〜14のマウス)。各注射には、150 μlのリン酸緩衝の生理食塩水(PBS )又は血漿が含まれており、従って、1回の注射当たりマウスの体重の略5%を占める。最後の注射から4日後、全てのマウスから採血し、抗体に基づくマイクロアレイ又はLuminex (登録商標)に基づく定量分析を用いて、200 より多い成長因子のレベル及び他の細胞間情報伝達タンパク質を測定した。マイクロアレイを用いて測定されたトップ6つのタンパク質の無監督の完全な連結クラスタリングは、PBS 又は血漿が処置されたマウスから血液を略完全に分離し(図14a )、若い血漿で処置された老齢マウスの血液及び全身的な環境が大幅に変化していることを実証している。増加した因子の多くは、組織再生の際に公知の機能を有している。血漿が処置された老齢マウスからの血液で増加した因子の例として、インターロイキン22(IL−22)及び白血病抑制因子(LIF )が含まれる(図14b 、14c )。独立して測定されて、マイクロアレイからの結果が確認されたこれらの因子は、複数の組織に有益な効果をもたらすことが示された。例えば、LIF は心筋梗塞の後の心臓機能及び再生を改善し(Zouein et al., Eur Cytokine Netw. 24:11-9, (2013))、骨格筋の再生をサポートし(Hunt et al. Histochem Cell Biol. 139:13-34, (2013))、視神経の再生及び軸索の再生を促進する(Fischer D, Leibinger M., Prog Retin Eye Res. 31:688-701, 2012)。同様に、IL−22は、皮膚、膵臓、肝臓及び消化管を含む複数の組織で有益な効果をもたらすことが実証された(Sabat et al. Nat Rev Drug Discov. 13:21-38, (2014))。更に、これらの研究結果は、若い血漿の処置が血液中の細胞間情報伝達タンパク質の生物全体に亘る変化をもたらし、これらのタンパク質が複数の組織に多面的な有益な効果をもたらすことを示している。
若いヒト血漿で処置されたマウスが脳の神経活動の増加を示し、臍帯からのヒト血漿がニューロンを活性化する際に最も強力であり、若いヒト血漿を用いたより長期間の処置が老齢マウスの認知を改善していることが、本明細書に示されている。本発明者らは、若齢マウスの血漿が、学習及び記憶に関与する脳の領域である海馬のシナプス可塑性をある程度高めることにより、老齢マウスの認知機能を高めるのに十分であることを既に実証している(Villeda et al. Young blood reverses age-related impairments in cognitive function and synaptic plasticity in mice. Nat Med 20, 659-663 (2014))。老いた脳を若返らせるために回復剤としての若い血漿の翻訳有用性を実証するために、本発明者らは、ヒト血液血漿が免疫拒絶反応の悪影響無しに注射され許容され得るマウスモデルを開発しようと努めた。本発明者らは、抗体産生及び補体活性化が激しく損なわれているとすると、機能的免疫系を欠いているNOD/SCID(NSG) マウス(Shultz et al. Human lymphoid and myeloid cell development in NOD/LtSz-scid IL2R gamma null mice engrafted with mobilized human hemopoietic stem cells. J Immunol 174, 6477-6489 (2005))がこのために追跡可能なモデルになると仮定した。老化の文脈の可溶性血漿因子を研究するためのモデルとしての有用性を実証するために、本発明者らは、脳の老化の重要な局面を再現するためのNSG モデルの能力をまず評価した。図15に示されているように、本発明者らは、NSG マウスが、海馬のCD68染色によって反映されているように、海馬歯状回における新生児のニューロンの数の加齢性の欠損を示し、小神経膠細胞症の増加を更に示していることを見出している。本発明者らは更に、歯状回のcfos陽性細胞の数の年齢依存の低下を見出し、これはシナプス可塑性の一因となる前初期遺伝子ファミリの代わりのニューロンであることを見出した。
軽度なアルツハイマー病から中程度のアルツハイマー病の人間の患者に、(30歳より若い)若齢の血液ドナーからの200 mlのヒト血漿を静脈内で注入した。この手順を、例えば1週間に1回、4週間繰返し、この間、介護者が患者の一般的な機能及び日常生活の活動を記録する。処置が終わった後、患者の脳を、機能的MRI を使用して静止状態の脳の活性に関して走査し、患者の認知機能を、一連の神経生理学的テストで評価する。常に、患者、介護者、及び処置又はテストを与える医師は、患者が若い血液血漿を受けたか又は対照として生理食塩水を受けたか分かっていない。次に、処置後に得られた測定結果を、処置が始められる前に患者から得られた同様の測定結果と比較する。若い血漿を受けた患者の一般的な機能及び日常生活の活動が改善されたことが観察されている。
有効量の若い血漿製剤を対象に与えて、老化に関連する状態について前記対象を処置することを特徴とする方法。
若い血漿製剤を与えた後、前記対象の認知能力の低下率の減少を検出することを特徴とする付記1乃至12のいずれかに記載の方法。
前記血液製剤を与えた後、前記対象の認知能力の安定化又は改善を検出することを特徴とする付記1乃至12のいずれかに記載の方法。

Claims (15)
- 対象の老化に関連する状態を処置する方法であって、
有効量の若い血漿製剤を対象に与えて、老化に関連する状態について前記対象を処置することを特徴とする方法。 - 前記若い血漿製剤は全血ではないことを特徴とする請求項1に記載の方法。
- 前記若い血漿製剤は赤血球及び/又は白血球を欠いていることを特徴とする請求項1又は2に記載の方法。
- 前記若い血漿製剤は無細胞であることを特徴とする請求項1乃至3のいずれかに記載の方法。
- 前記若い血漿製剤は、所定の閾値未満の平均分子量のタンパク質を欠いていることを特徴とする請求項1乃至4のいずれかに記載の方法。
- 前記若い血漿製剤は、40歳以下のドナーから得られていることを特徴とする請求項1乃至5のいずれかに記載の方法。
- 前記若い血漿製剤は、新生児の臍帯から得られていることを特徴とする請求項6に記載の方法。
