[00107] ナノ粒子は、MERS CoVウイルスに対する免疫または実質的な免疫が付与される免疫応答を刺激するための免疫原性組成物を調製するために有用である。粘膜免疫と細胞性免疫はどちらも感染および疾患に対する免疫に寄与し得る。上気道において局所的に分泌される抗体は自然感染に対する抵抗性の主要な因子である。分泌性免疫グロブリンA(sIgA)は上気道の防御に関与し、血清IgGは下気道の防御に関与する。感染によって誘導される免疫応答により、同じウイルスまたは抗原性が類似したウイルス株への再感染から保護される。本明細書に開示されているナノ粒子を用いた免疫後に宿主において産生される抗体は、他者に投与し、それにより、当該対象における受動的な投与をもたらすこともできる。
[00112] 本明細書に開示されているナノ粒子は、宿主における高親和性抗MERS−CoV抗体の産生を誘導するものである。これらの高親和性抗MERS−CoV抗体では、従来の方法(例えば、ファージディスプレイ; Zhangら、2014参照))によって産生された抗MERS−Co−V抗体よりも高い親和性が実証される。
MERS CoVスパイクナノ粒子の製造
[00122] ナノ粒子は、Sf9細胞においてバキュロウイルスでMERS CoVスパイクタンパク質(図8および図9)を過剰発現させることによって産生される。精製後、スパイクタンパク質を含有するナノ粒子VLPは主に三量体の形態で回収された(図4および図6)。
MERS CoVスパイクナノ粒子を使用した免疫応答の誘導
[00123] 図5に示されているとおり、VLPをマウスに投与した。45日の期間の最後に、マウスから血液を抽出して免疫応答を評価した。図7には、SARS CoVスパイクナノ粒子VLPまたはMERS CoVスパイクナノ粒子VLPのいずれかを投与したマウスについての中和抗体力価が示されている。アジュバントであるMatrix M1を使用することによって最も高い応答が得られた。
トランス染色体(transchromosomic)ウシにより産生される強力な抗MERS−CoVヒト免疫グロブリンにより、in vivoにおいてMERS−CoVが阻害される
[00124] ワクチン接種後にTcウシから得られた抗MERS CoV hIgG抗体および免疫グロブリンにより、in vitroおよびin vivoにおいてMERS CoVを中和できることを実証するために、3種の実験的なMERS CoVワクチンを3つの別々の群のTcウシに投与して、MERS CoVに対する高力価の中和抗体を誘導した。試験した第1のワクチンは、全死菌(whole killed)Jordan株(クレードA)MERS−CoVビリオン(WKV)ワクチンであり、第2ワクチンはAl−Hasa(クレードB)MERS−CoVスパイクナノ粒子(SN)ワクチンであり、第3のワクチンはAl−Hasa MERS−CoVスパイクタンパク質pDNAワクチンであった。pDNAワクチンは、in vitroアッセイにおいて免疫原性が低いことが見いだされ、さらなる評価は行わなかった(データは示していない)。しかし、WKV(SAB-300と称される)を用いてワクチン接種したTcウシおよびSN(SAB-301と称される)を用いてワクチン接種したTcウシ由来の回復期血清および高度に精製したhIgG免疫グロブリンはどちらも、in vitroにおいてウイルスを交差中和することができ、MERS−CoVのヒトラジB細胞への抗体依存性侵入を誘導せず、また、Ad5−hDPP4を形質導入したMERS−CoVのマウスモデルにおけるウイルス感染(Erasmus株)を急速に低下させた。
動物試験
トランス染色体(Tc)ウシのクローニングおよび抗体産生
[00125] TcウシをMatsushitaら、PLoS One、2014.9(3):e90383頁、Sanoら、PLoS One、2013.8(10):e78119頁、およびKuroiwaら、Nat Biotechnol、2009.27(2):173〜81頁に記載のとおり製造した。簡単に述べると、この試験において使用するTcウシは、内在性ウシ免疫グロブリン遺伝子の三重ノックアウトについてホモ接合性であり(IGHM-/- IGHML1-/- IGL-/-)、ヒト免疫グロブリン重鎖遺伝子座全体を含有するヒト第14染色体断片と、ヒト免疫グロブリンカッパ軽鎖遺伝子座全体を含有するヒト第2染色体断片とからなるヒト人工染色体(HAC)を有する。
[00126] ワクチン接種用の全不活化(whole inactivated)MERS−CoVビリオンを、Jordan−N3/2012株(GenBank KC776174.1)を感染させたVero CCL−81細胞から培地を収集することによって製造した。4×108PFUのMERS−CoVを、コバルト線源を用いて線量が6MRadになるまで照射した。材料を不活化について安全性試験した。照射通過材料においてMERS−CoVに特異的なシグナルは検出されなかった。
[00127] 精製Al−Hasa株MERS−CoVスパイクタンパク質ナノ粒子を、Colemanら、2014に記載のとおり製造した。簡単に述べると、1ml当たり細胞2〜3×106個のSf9細胞に特定の組換えバキュロウイルスを感染させた。感染したSf9細胞を27±2℃で継続的に撹拌しながらインキュベートし、68〜72hpiにおいて4000×gで15分遠心分離することによって回収した。非イオン性界面活性剤を用いて細胞膜からスパイクタンパク質を抽出し、10,000×gで30分遠心分離することによって不溶性材料を除去した。陰イオン交換クロマトグラフィー、アフィニティクロマトグラフィー、およびサイズ排除クロマトグラフィーの組合せを使用してスパイクタンパク質集合体を精製した。精製中に大多数の界面活性剤を除去し、それにより、スパイクタンパク質三量体に高次タンパク質−タンパク質ミセル状ナノ粒子を形成させた。精製スパイクナノ粒子を0.2ミクロンで濾過し、−80℃で保管した。
[00128] 図10Aに示されているとおり、群1のTcウシ3頭(#2244;#2252および#2254)を、水中油エマルションとしてMontanide ISA25アジュバント(Seppic)、加えてサポニン由来免疫刺激薬Quil A(Accurate Chemicals)と一緒に製剤化した全死菌MERS−CoVウイルス(WKV)を1〜2×108PPF/用量で用いて免疫した。群2のTcウシ2頭(#2178および#2183)を、同様に水中油中水エマルションとしてMontanide ISA206アジュバント、加えてQuil Aと一緒に製剤化した組換えMERS−CoVスパイクナノ粒子(SN)を2mg/用量で用いて免疫した。両群のTcウシを3〜4週間の間隔を空けて5回免疫した。
[00129] 形質導入されたhDPP4 BALB/cマウスにErasmus株MERS−CoVを感染させた。簡単に述べると、BALB/cに陰性対照またはウシから採取した試験免疫グロブリン(SAB-300またはSAB-301)100μgまたは500μgを腹腔内注射した。12時間後、マウスに総体積50μlのMERS−CoV(1×105PFU)を鼻腔内感染させた。ウイルス力価を得るために、肺を取り出してPBS中に入れ、手動ホモジナイザーを使用してホモジナイズした。ベロ81細胞においてウイルスの力価測定を行った。ウイルス力価は、組織1g当たりのMERS−CoVのPFUとして表される。
Tc動物において産生される抗原特異的hIgGの評価
[00130] MERS−CoVスパイクタンパク質に特異的なELISAを実施して、WKVまたはSNを用いた免疫後のTcウシ(図10B)ならびにSAB−300およびSAB−301(図10C)における抗原特異的hIgG応答を評価した。ELISAの結果により、血清ならびにSAB−300およびSAB−301のどちらについても、2回目のワクチン接種(V2)時にロバストなMERS−CoVスパイク特異的hIgG応答が実証され、最後の免疫(V5)まで高力価が維持された(図10)。このデータにより、MERS−CoVタンパク質を標的とする高力価のhIgG抗体がWKVまたはSNを用いて免疫したTcウシで生成されたこと、および、どちらの免疫グロブリンの活性も精製後に保持されることが示される。
蛍光低下中和試験−50%低下(FRNT50)。
[00131] 2種の中和アッセイを実施して、in vitroにおけるSAB−300およびSAB−301による中和の潜在性を同定した。ワクチン接種したウシ由来の血清、SAB−300、およびSAB−301を、in vitroにおいてMERS−CoVの感染を阻害するそれらの能力について蛍光低下中和試験(FRNT50)およびウイルス感染中和アッセイを使用してアッセイした。FRNTアッセイでは感染が蛍光を使用して報告され、ウイルス感染中和アッセイでは細胞の生存能力を使用して報告される。
[00134] 抗体が結合したMERS−CoVウイルスにより、通常は感染しない細胞へのビリオンの侵入が可能にならないことを実証するために、一般にはMERS−CoVが感染しない不死化ヒトB細胞株であるラジ細胞を試験して、抗MERS抗体の存在により、ウイルスRNAの転写および生ウイルスの放出が可能になり得るかどうかを調べた。ラジ細胞に、SAB−300およびSAB−301と一緒にプレインキュベートしたMERS−CoVを感染させた(図12B)。ウイルスmRNAのRT−PCRおよびTCID50アッセイを感染の48時間後の培地に対して実施して感染を評価した。TCID50アッセイでは、ラジ細胞から放出されたMERS−CoVを検出することはできず、当該細胞にMERS−CoVが感染しなかったことを示している。逆に、このアッセイにより、同様に処理した、一般にMERS−CoVが感染するベロE6細胞では感染の48時間後に高レベルのウイルスが生じたことが見いだされた(データは示していない)。MERS−CoVをSAB−300およびSAB−301と一緒にプレインキュベートした後に感染させたラジ細胞においてMERS−CoV RNAが検出されるかどうかを決定するために、感染細胞からRNAを抽出し、MERS−CoVの新しく転写されたRNAに特異的なプライマー(リーダープライマー)を用いてTaqmanリアルタイムPCRによって分析した(図12B)。MERS−CoVを単独で感染させたラジ細胞、予めワクチン接種した血漿から精製した非特異的な陰性対照hIgGを感染させたラジ細胞、またはSAB−300もしくはSAB−301を用いて前処理したMERS−CoVを感染させた細胞におけるMERS−CoVの新しく転写されたウイルスRNA(リーダープライマーセット)の有意な検出はなかった。これらのデータにより、SAB−300またはSAB−301によるMERS−CoV感染の抗体依存性感染増強は引き起こされないことが実証される。
[00135] MERS−CoVのマウスモデルにおいてSAB−300およびSAB−301の有効性を試験した。マウスは、MERS−CoV非許容的であるが、MERS−CoV受容体であるヒトジペプチジルペプチダーゼ4(hDPP4)を発現するアデノウイルスを用いて形質導入すると許容的になり、ウイルスが複製される(Zhaoら、Proc Natl Acad Sci U S A、2014. 111(13): p. 4970-5)。Tc抗体の抗ウイルス活性を試験するために、BALB/cマウス(6〜8週)に、PBS75μ中Ad5−hDPP4を2.5×108PFUで鼻腔内に用いて形質導入した。形質導入の5日後に、マウスを単回の100μgまたは500μg用量の対照hIgG、SAB−300またはSAB−301を用いて腹腔内注射によって処置した。12時間後、マウスに総体積50μlのMERS−CoV(1×105PFU)を鼻腔内感染させた。感染の5日の経過にわたって追加的な抗体注射は行わなかった。感染の1日後、3日後および5日後にマウスを安楽死させ、それらの肺を切開した。ウイルス力価を得るために、手動ホモジナイザーを使用して肺をPBS中でホモジナイズし、遠心分離することによって清澄化し、ベロ細胞で力価測定を行った(図13)。無処置群、および陰性対照ヒトIgGを受けた群の肺において感染の1日後および3日後に肺組織1mg当たり約1×106PFUの力価が見いだされ、注射5日後には力価が1×105に降下した。SAB−300を100μgまたは500μg注射したマウスでは、hIgG対照注射群と比較して、それぞれ約50分の1または500分の1のウイルス力価の低下が感染の1日後に見いだされた(図13A)。感染の3日後までに、100μg群についてのウイルス力価の低下は約5000分の1であり、500μgを受けたマウスにおける力価は検出レベル未満であった。感染の5日後には、どちらの処置群においてもウイルス力価は検出レベル未満であった(図13A)。SAB−301抗体についても、感染の1日後の肺力価がSAB−300で処置したマウスと比較してわずかに高かった以外は同様のウイルス力価の低下が見いだされた(図13B)。感染の5日前までに、SAB−301で処置したマウス全てでウイルス力価が検出レベル未満であった(図13B)。これらのデータにより、SAB−300とSAB−301がどちらも単回の予防注射でマウスをMERS−CoV感染から保護できることが実証される。
MERS−CoVスパイクタンパク質(S)ナノ粒子を用いて免疫した動物における抗体のBIACORE解析
[00136] 図10Aおよび実施例3に示されている一連の免疫に従ってMERS−CoV Sナノ粒子ワクチンを用いて免疫したTcウシ2頭(#2178および2183)由来の2種の抗体の結合活性を抗S親和性(Biacore(商標))解析によって試験した。これらの動物の、ワクチン接種3(v3)、ワクチン接種4(v4)、およびワクチン接種5(v5)後の血清中の抗体の結合活性を試験した。簡単に述べると、CM5チップとアミンカップリングしたプロテインA/Gを使用して、ウシTc血清IgGをフローセルに捕捉した。MERSスパイクタンパク質抗原を0nM、20nM、および40nMでフローセルに180秒にわたって注射し、その後600秒にわたって解離させた。1:1フィットモデルを適用した。会合速度および解離速度を時間に応じて測定し、センサーグラムにプロットし、会合速度および解離速度から解離定数を算出した。図14および表1に結果が示されている。

- MERS−CoVナノ粒子を含む免疫原性組成物であって、ナノ粒子がスパイクポリペプチドの三量体を少なくとも1つ含む免疫原性組成物。
- ナノ粒子がスパイクポリペプチドの三量体を少なくとも約5個〜約30個含む、請求項1に記載の組成物。
- ナノ粒子がスパイクポリペプチドの三量体を少なくとも約10個〜約20個含む、請求項2に記載の組成物。
- ナノ粒子の濃度が少なくとも約20μg/mL〜約60μg/mLである、請求項1に記載の組成物。
- ナノ粒子の濃度が少なくとも約30μg/mL〜約50μg/mLである、請求項4に記載の組成物。
- スパイクポリペプチドが配列番号2を含む、請求項1〜3のいずれかに記載の組成物。
- スパイクポリペプチドが配列番号2からなる、請求項1〜3のいずれかに記載の組成物。
- 高親和性抗体の製造方法であって、動物に、MERS−CoVナノ粒子を含む免疫原性組成物であって、ナノ粒子がスパイクポリペプチドの三量体を少なくとも1つ含む免疫原性組成物を投与する工程、動物から血清および/または血漿を採取する工程、ならびに血清および/または血漿から抗体を精製する工程、を含む方法。
- ナノ粒子がスパイクポリペプチドの三量体を少なくとも約5個〜約30個含む、請求項8に記載の組成物。
- ナノ粒子がスパイクポリペプチドの三量体を少なくとも約10個〜約20個含む、請求項9に記載の組成物。
