Abstract
Description
アルカリ水溶液中においてセルロース繊維を膨潤させるステップと、
膨潤したセルロース繊維をアルカリ水溶液から取り出すステップと、
膨潤したセルロース繊維を、金属塩の水溶液およびポリマー溶液と混合することにより、繊維に金属ナノ粒子を含浸させるステップと、
金属ナノ粒子を含浸させた、膨潤したセルロース繊維を溶液から取り出すステップ
とを含む、金属ナノ粒子を含浸させたセルロース繊維を製造する方法
が提供される。
1.1a 銀ナノ粒子が含浸されたセルロース繊維の製造方法
下記は、銀ナノ粒子が含浸されたセルロース繊維を製造するための、研究室における一般的な方法を説明する。製造規模を拡大または縮小するために、量および条件が調整され得ることが理解されるものである。
2. 300 mlの水酸化ナトリウム水溶液(3 M; 新鮮な脱イオン水を用いて調製)を、繊維を含む容器に加える。繊維全体が、水酸化ナトリウム水溶液によって完全に覆われるようにしなければならない。
3. その後、繊維と水酸化ナトリウム溶液とを含む容器密閉し、90℃に設定した油浴または水浴の中に置き、繊維を膨潤させる。
4. 水酸化ナトリウム水溶液中で、およそ30分間、繊維を膨潤させる。
5. 膨潤が完了したら、過剰量の水酸化ナトリウム溶液を容器から捨て、繊維から手動で搾り出す。
6. その後、膨潤段階に用いたのと同じ温度/保圧条件を用いて、高温の新鮮な脱イオン水中で繊維を濯ぎ、続いて、繊維を手動で搾ることにより液体を取り除く。
7. その後、繊維を未使用のガラス容器中に入れ、100 mlの硝酸銀水溶液(30 w/w %; 新鮮な脱イオン水を用いて調製)を加える。
8. その後、新鮮な脱イオン水中15%のポリ(N-ビニルピロリドン)(平均分子量40000 gmol-1)の溶液200mlを、繊維/硝酸銀に加える。硝酸銀およびポリマー溶液は、同時に繊維に加えられてもよいことが理解される。膨潤したセルロース繊維、硝酸銀、およびポリ(N-ビニルピロリドン)を含む容器をその後密閉し、油浴(または水浴)に戻して硝酸銀を還元し、続いて、ポリ(N-ビニルピロリドン)で安定化された銀ナノ粒子をin situで生成させる。油浴または水浴の温度は90℃に設定される。
9. 反応は、およそ90分間進行させる。
10. 反応が完了したら、銀化された繊維を反応溶液から取り出し、手動で搾ることにより、繊維中から残留液体を取り除く。
11. その後、繊維は、一連の洗浄サイクルに供され、繊維を洗浄して過剰な反応物および反応副生成物を取り除く。第一の洗浄では、希クエン酸溶液(0.2 M)が用いられる。その後、繊維は、300 mlの温かい脱イオン水で洗浄した後、冷たい脱イオン水を用いて洗浄する。繊維はその後、繊維を「解膨潤」する手段として、アセトン中で洗浄される。
12. 繊維が洗浄され「解膨潤」されたら、溶媒は、50〜100℃の範囲の高温で蒸発させることにより取り除く。
1.1aの方法が、下記の変更を用いて繰り返された:
ステップ2.において、水溶液は、211gの脱イオン水、36gの水酸化ナトリウム、および53gの炭酸ナトリウム(水酸化ナトリウムのモル数と炭酸ナトリウムのモル数との比は1.8:1である。)から調製される;
ステップ8.において、ポリ(N-ビニルピロリドン)の分子量は40〜80kg/molである。
上述の方法1.1aに従って製造された繊維は、柔らかいドレープと手触りを有する、濃褐色/黒色のものであることが分かった。
上述のように製造された銀ナノ粒子が含浸されたセルロース繊維0.5gは、50 mlの脱イオン水に入れ、37℃でインキュベーションした。その後毎日、2 mlの液体を採取しUV-Visにより分析した。採取した液体は、新鮮な脱イオン水で置き換えた。それぞれの日に測定された液体の銀含量は、表2aに示される。
上述のように製造された銀ナノ粒子が含浸されたセルロース繊維0.5gは、25gのミルク中に入れ、30℃で7日間インキュベーションした。結果は表3に示される。
方法1.1bに従って製造された繊維は、柔らかいドレープと手触りを有する、濃褐色のものであることが分かった。繊維は、方法1.1aを用いて製造された繊維よりも一層柔らかく、分離(separate)しやすいものであった。
上述のように製造された銀ナノ粒子が含浸されたセルロース繊維0.5gは、50 mlの脱イオン水に入れ、37℃でインキュベーションした。その後毎日、2 mlの液体を採取しUV-Visにより分析した。採取した液体は、新鮮な脱イオン水で置き換えた。それぞれの日に測定された液体の銀含量は、表2bに示される。
2.1 金属ナノ粒子が含浸されたセルロース繊維を用いて不織布を製造する方法
1. 金属ナノ粒子が含浸されたセルロース繊維を、50 mm〜100 mmの短い長さに切り分ける。
2. 親水性の、吸収性および/またはゲル状の繊維を、銀含有セルロース繊維と同じ長さに切り分ける。
3. その後、2つの種類の繊維を、当技術分野において知られている繊維をブレンドするための任意の技法を用いて、所望の割合でブレンドする。好ましくは、ブレンドされた繊維は、カーディング技法を用いて製造され、これにより、繊維は、開放構造を有し、配向性を有し、密接にブレンドされる。カーディングは、繊維の一体性および布帛の強度を保持しつつ、可能な限り銀を最も均一に分布させるのに必要なだけ多くの回数、実施される。これが最終的には、最終製品である被覆材からの金属の放出を促進し、創傷接触領域の全体に亘って、創傷からの液体の取り込みまでも確保する。
4. ブレンドされた繊維は、カーディングされた後、交差するように折り重ねられて、不織繊維の網状布に作製され、これは、この段階で、創傷被覆材を製造するのに用いることができる。この網状布を引き続いてニードルパンチングするのがより好ましいが、これは、繊維を互いに絡ませ、網状布に剛性、一体性、および強度を与えるものである。
5. その後、網状布は、創傷被覆材として用いられ得る適切な大きさの布片、例えば10cm x 10cmに切り分けることができる。
3.1 被覆材の組成
銀ナノ粒子が含浸されたセルロース繊維は、実施例1に説明されるようにして製造された。その後、繊維は、他の繊維とブレンドされ、実施例2に説明される方法を用いて創傷被覆材に製造され、被覆材A、B、およびCを得た。被覆材AおよびBの組成は表2aに示される。被覆材Cの組成は表2bに示される。
被覆材Aの一般的な物理的・機械的特定は、標準的な技術を用いて測定され、比較例である被覆材D(標準的なCESをベースとする被覆材、CESとTENCEL(登録商標)とを含む)の特性と比較した。
被覆材Aからの銀ナノ粒子の放出が試験された。0.5gの創傷被覆材を、25 mlの脱イオン水または0.9%塩化ナトリウム溶液のいずれかに入れ、37℃で保管した。定期的に、少量の液体サンプルを採取し、紫外線分光測定法(UV-vis)を用いて、金属ナノ粒子により示される「表面プラズモン共鳴」を利用して試験した。採取した液体の部分は、新鮮な脱イオン水または塩化ナトリウム溶液で置き換えた。銀含量は、4日間に亘ってモニターした。
被覆材Aの抗菌特性が調べられた。寒天プレートにPseudomonas aeruginosaを播種した。その後、同一の大きさおよび形状を有する被覆材(または対照材)を4片、寒天プレートに置き、37℃で48時間インキュベーションした。未処理のガーゼを陽性対照として用いた。酢酸に浸した布帛を陰性対照として用いた。
被覆材は、(布帛から100%放出されると想定して)ミルクの重量に基づき100 ppm の濃度の銀をシミュレーションするのに適切な重量で、25gのミルクに入れた。ミルク サンプルは、ビンに密閉し、37℃で7日間インキュベーションした。結果は、下記の表4に示す。
表5は、比較例である被覆材E(標準的なアルギナートをベースとする被覆材)と比較した、被覆材Bの一般的な物理的・機械的試験を示す。
実験は、被覆材Aについてと同様に実施された(上述の実施例3.5を参照)。結果は、下記の表6に示される。
表7は、比較例である被覆材F(CMCをベースとする被覆材)と比較した、被覆材Cの一般的な物理的・機械的試験を示す。
被覆材Cについて、より定型的な抗菌試験が実施され、標準試験法BS EN 16756 (開発(development)における試験法)ならびにGallant-BehmらとThomasおよびMcCubbinとに従う方法を用いて、Staphylococcus aureus (ATCC6538)とPseudomonas aeruginosa (ATCC9027)とを24時間ごとに7日間繰り返し播種および計数した。

Claims (32)
- アルカリ水溶液中においてセルロース繊維を膨潤させるステップと、
膨潤したセルロース繊維をアルカリ水溶液から取り出すステップと、
膨潤したセルロース繊維を、金属塩の水溶液およびポリマー溶液と混合することにより、繊維に金属ナノ粒子を含浸させるステップと、
金属ナノ粒子を含浸させた、膨潤したセルロース繊維を溶液から取り出すステップ
とを含む、金属ナノ粒子を含浸させたセルロース繊維を製造する方法。 - 前記金属が、銀、銅、亜鉛、セレン、金、コバルト、ニッケル、ジルコニウム、モリブデン、ガリウム、もしくは鉄、またはこれらの任意の組み合わせである、請求項1に記載の方法。
- 前記金属が銀である、請求項1または2に記載の方法。
- 前記アルカリ水溶液が、1族元素の水酸化物、1族元素の炭酸塩、1族元素の炭酸水素塩、水酸化テトラアルキルアンモニウム、モノアミンもしくはジアミン、およびN-メチルモルホリンオキシド(NMMO)からなる群より選択される化合物、またはその混合物と、LiClとから構成される溶液である、請求項1〜3のいずれか1項に記載の方法。
- 前記アルカリ水溶液が、1族元素の水酸化物および1族元素の炭酸塩を含む、請求項4に記載の方法。
- セルロース繊維をアルカリ水溶液中で20℃〜120℃の温度でインキュベーションするステップをさらに含む、請求項1〜5のいずれか1項に記載の方法。
- 膨潤したセルロース繊維をアルカリ水溶液から取り出した後であって、かつ膨潤したセルロース繊維を金属塩の水溶液およびポリマー溶液と混合する前に、膨潤したセルロース繊維を洗浄するステップをさらに含む、請求項1〜6のいずれか1項に記載の方法。
- 前記ポリマーが、ポリアミド、ポリイミド、ポリエチレンイミン、ポリビニルアルコール、ペクチン、アルブミン、ゼラチン、カラギーナン、ゴム、セルロースもしくはその誘導体、ポリ(N-ビニルピロリドン)、ポリ(N-ビニルカプロラクトン)、またはこれらの混合物である、請求項1〜7のいずれか1項に記載の方法。
- 前記ポリマーがポリ(N-ビニルピロリドン)である、請求項8に記載の方法。
- 金属塩の水溶液、ポリマー溶液、および膨潤したセルロース繊維を20℃〜120℃の温度でインキュベーションするステップをさらに含む、請求項1〜9のいずれか1項に記載の方法。
- 金属ナノ粒子を含浸させた、膨潤したセルロース繊維を溶液から取り出した後、金属ナノ粒子を含浸させた、膨潤したセルロース繊維を洗浄するステップをさらに含む、請求項1〜10のいずれか1項に記載の方法。
- 前記繊維を有機溶媒で洗浄することにより、前記繊維を収縮させる、請求項11に記載の方法。
- 金属ナノ粒子を含浸させたセルロース繊維を乾燥させるステップをさらに含む、請求項1〜12のいずれか1項に記載の方法。
- 請求項1〜13のいずれか1項に記載の方法に製造され得る、金属ナノ粒子を含浸させたセルロース繊維。
- 金属ナノ粒子が1.5 w/w %超の濃度で含浸される、請求項14に記載のセルロース繊維。
- 金属ナノ粒子の平均粒径が5〜50 nmである、請求項14または15に記載のセルロース繊維。
- 金属酸化物および/または金属炭酸塩がさらに含浸される、請求項14〜16のいずれか1項に記載のセルロース繊維。
- 前記金属が銀である、請求項14〜17のいずれか1項に記載のセルロース繊維。
- ポリマーが更に含浸される、請求項14〜18のいずれか1項に記載のセルロース繊維。
- 前記ポリマーがポリ(N-ビニルピロリドン)である、請求項19に記載のセルロース繊維。

- 前記セルロース繊維がリヨセル繊維である、請求項1〜13のいずれか1項に記載の方法、または請求項14〜22のいずれか1項に記載のセルロース繊維。

アルギン酸、改質セルロース、改質キトサン、グアーガム、カラギーナン、ペクチン、デンプン、ポリアクリラートもしくはそのコポリマー、ポリエチレンオキシドもしくはポリアクリルアミド、またはこれらの混合物をベースとするゲル状繊維、および/あるいは
ポリエステル、ポリエチレン、ポリプロピレン、ポリアミド、セルロース、熱可塑性二成分系繊維、ガラス繊維、またはこれらの混合物をベースとする非ゲル状繊維
である、請求項24に記載の吸収材。 - 2〜30 w/w %の金属ナノ粒子を含浸させたセルロース繊維と、5〜30 w/w %のリヨセル繊維と、40〜90 w/w %のCES繊維とのブレンドを含む、請求項24または25に記載の吸収材。
- 2〜20 w/w %の金属ナノ粒子を含浸させたセルロース繊維と、10〜30 w/w %のリヨセル繊維と、50〜90 w/w %のCMC繊維とのブレンドを含む、請求項24または25に記載の吸収材。
- ブレンド繊維の総重量に基づき0.2 w/w %超の金属を含む、請求項24〜27のいずれか1項に記載の吸収材。
- 前記吸収材が、15グラム超の液体/吸収材1グラムの吸収性を有する、請求項24〜28のいずれか1項に記載の吸収材。
