材料及び方法
(i)試薬
LIMP−2ヒツジポリクローナル抗体をペプチド配列CKKLDDFVETGDIRTMVFP(配列番号13)(Mimotopes Pty Ltd.、ビクトリア、オーストラリア)を用いて生成した。この試験に用いた一次抗体は、APPL1(0.4μg/mL、Abcam PLC、Cambridge United Kingdom、カタログ番号ab95195)、APPL2(0.4μg/mL、Abcam、カタログ番号ab95196)、Rab4(1μg/mL、Abcam、カタログ番号ab13252)、TGN46(10μg/mL、Abcam、カタログ番号ab50595)、TfR1(1μg/mL、Abcam、カタログ番号ab108985)、TfR2(1μg/mL、Abcam、カタログ番号ab80194)、Akt(1/1000,Cell Signaling Technology,Inc.、MA、USA)及びホスホ−Akt(Thr308 1/1000、Cell Signaling Technology,Inc.)に対するウサギポリクローナル抗体などであった。ヤギ抗Rab5(1μg/mL、Santa Cruz Biotechnology、CA、USA)、Rab7(1μg/mL、Santa Cruz Biotechnology)及びEEA1(1μg/mL、Santa Cruz Biotechnology)ポリクローナル抗体も使用した。LAMP−1(1μg/mL)マウスモノクローナルBB6は、ウメオ大学、スウェーデンから提供された)。ウエスタンブロット分析用HRP複合二次抗体は、抗ヤギ/ヒツジ(1/2000、Merck Millipore Pty.Ltd.、ビクトリア、オーストラリア)、抗ウサギ(1/2000、Sigma Aldrich Pty.Ltd.、ニューサウスウェールズ、オーストラリア)及び抗マウス(1/2000 Sigma Aldrich Pty.Ltd.)などであった。HRP複合抗GAPDH(1/20000 Sigma Aldrich Pty.Ltd.)。用いた二次及び他の抗体複合蛍光団は、Life Technologies Pty Ltd.、ビクトリア、オーストラリア製Alexa Fluor(登録商標)488(1/250)、Alexa Fluor(登録商標)633(1/250)、トランスフェリン633(1/1000)、ファロイジン488(1/100)、LysoTracker(登録商標)(5μM)などであった。プライマーをGeneworks Pty Ltd.、アデレード、オーストラリアから得た。その配列を支持情報表1に列挙した。
細胞株PNT1a及びPNT2、22RV1及びLNCaP(クローンFCG)を細胞バンクオーストラリア(CellBank Australia)(小児医学研究所(Children’s Medical Research Institute)、ニューサウスウェールズ、オーストラリア)を介して欧州細胞培養コレクション(European Collection of Cell Cultures)から得た。これらの細胞株は、交差汚染又は誤認細胞株のリスト、バージョン6.8(2012年3月9日)にはなかった。PNT1a及びPNT2をサル空胞形成ウイルス40(SV40)不死化細胞株から前もって誘導した。22RV1癌細胞株を異種移植片から前もって誘導した。異種移植片は、去勢によって誘導された親のアンドロゲン依存性異種移植片の退行及び再発後にマウスにおいて連続的に増殖されたものである。この細胞株は、アンドロゲン受容体を発現したが、その増殖は、アンドロゲン刺激に反応しなかった。LNCaPは、前立腺腺癌のリンパ節転移から前もって誘導され、アンドロゲンに反応する。
培地を80〜90%コンフルエントな細胞培養物から吸引し、細胞をPBSで1回洗浄し、次いで800μLの20mM Tris(pH7.0)、500mM塩化ナトリウム及び2%SDS溶液と一緒にインキュベートした。細胞を収集し、65℃に加熱し、1分間超音波処理して抽出物を調製した。次いで、溶解物を25ゲージ針に6回通した。細胞抽出物中の総タンパク質量を製造者の指示に従ってビシンコニン酸アッセイにより定量化した(Micro BCAキット、Pierce、ロックフォード、IL、USA)。Wallac Victor(商標)光学プレートリーダー及びWorkoutソフトウェアv2.0(Perkin−Elmer Pty,Ltd.、ビクトリア、オーストラリア)、5点パラメータ検量線を用いて試料を定量化した。細胞抽出物を−20℃で貯蔵した。
細胞株を80〜90%コンフルエントに培養し(3つ組T75フラスコ)、培地を吸引し、細胞層をPBSで洗浄し、その後、1mLのTRI試薬(登録商標)(Applied Biosystems Pty Ltd.、ビクトリア、オーストラリア)を添加して、収集した。TRI試薬(登録商標)1ミリリットル当たり200マイクロリットルのクロロホルムを添加し、試料を1分間激しく振とうし、室温で3分間インキュベートし、次いで16,000g、4℃で15分間遠心分離した。RNA抽出をRNeasy(登録商標)ミニキット(Qiagen Pty Ltd.、ビクトリア、オーストラリア)を用いて製造者の指示に従って実施した。抽出RNAの濃度をNanoDrop(商標)2000分光光度計(Thermo Fisher Scientific Australia Pty Ltd.、ビクトリア、オーストラリア)を用いて260nmで測定した。260/280nm及び260/230nmの各比を測定して、試料にタンパク質及びDNA混入がないことを確認した。1.6より大きい比は、試料が汚染されていないことを示す。
細胞溶解物10μgを加熱変性し(NuPAGE(登録商標)LDS試料緩衝剤及び還元剤中で100℃で5分間)、次いで前もって成形されたゲルを用いて、XCell SureLock Mini−Cellシステム(Life Technologies Australia Pty Ltd.、ビクトリア、オーストラリア)において120Vで1.5時間電気泳動させた。次いで、タンパク質をポリフッ化ビニリデン膜(Polyscreen(登録商標)、PerkinElmer、ビクトリア、オーストラリア)に35Vで1時間移した。転写膜をTBS−tween(0.1%;ブロック)中の5%(w/v)脱脂乳溶液を用いて室温で1時間ブロックし、一次抗体と一緒に4℃で終夜インキュベートした。膜をTBS−tween(0.1%)で3×5分間洗浄し、次いでブロック溶液で1/2000希釈した適切なセイヨウワサビ−ペルオキシダーゼ複合二次抗体と一緒にインキュベートした。Novex(登録商標)ECL化学発光基質試薬キット(Life Technologies Australia Pty Ltd.、ビクトリア、オーストラリア)を用いて膜を現像し、ImageQuant(商標)LAS4000画像装置、ソフトウェアバージョン1.2.0.101(GE Healthcare Bio−Sciences Pty Ltd.、ニューサウスウェールズ、オーストラリア)を用いてタンパク質を可視化した。3つ組試料を分析し、AlphaViewSA(商標)ソフトウェアv3.0.0.0(ProteinSimple、サンタクララ、カリフォルニア)を用いて基準GAPDH添加コントロールに対して画像を定量化した。Stata/SE v11.2(StataCorp LP、テキサス、米国)を用いたクラスカル ワリス順位和統計分析を実施して、非悪性コントロール細胞株群と癌細胞株群の間の有意性を求めた(95%信頼限界;p<0.05)。
細胞を22mmカバーガラス上で培養し、PBS中の4%(v/v)ホルムアルデヒドを用いて室温で20分間固定し、次いでPBS中の0.1%Triton−X(v/v)で10分間透過処理した。PBS中の5%(w/v)ウシ血清アルブミンと一緒に室温で2時間インキュベートすることによって非特異抗体の反応性をブロックした。次いで、細胞を5%BSA中で一次抗体と一緒に室温で2時間、次いで二次抗体と一緒に室温で1時間インキュベートした。PBSで3回洗浄して結合していない抗体を除去し、カバーガラスをDAPI核染料を含むProLong(登録商標)Gold Antifade試薬(Life Technologies Australia Pty Ltd.、ビクトリア、オーストラリア)中に置いた。共焦点顕微鏡法をZeiss LSM710META NLOレーザー走査顕微鏡及び付属のCarl Zeiss Zen2009ソフトウェアを用いて実施した。370、488、543及び633nmのレーザー線をそれぞれDAPI、Alexa Fluor(登録商標)488、Cy3及びAlexa Fluor(登録商標)633蛍光に利用した。画像をグレースケール16ビットTIFFファイルとしてエクスポートし、Adobe(登録商標)Photoshop(登録商標)CS5(Adobe Systems Inc.、サンホゼ、カリフォルニア、米国)を用いて処理した。
対応ヒト非悪性及び腫よう前立腺組織切片(3μm)をSuperfrost Ultra Plus(登録商標)スライド(Menzel−Glaser)に載せ、50℃で終夜加熱した。次いで、切片をキシレンで脱脂し、エタノール中で再水和させ、PBS中の0.3%H2O2中で室温で15分間インキュベートした。抗原賦活化装置(Decloaking Chamber)(Biocare Medical)中で10mMクエン酸緩衝剤(pH6.5)を用いて125℃で5分間HIERを実施した。スライドをまず(製造者の指示に従って)Invitrogenアビジン/ビオチンキットを用いてブロックし、次いで高湿度チャンバ内の5%ブロッキング血清(SIGMA)中で室温で30分間ブロックした。次いで、切片を高湿度チャンバ内で一次抗体と一緒に4℃で終夜インキュベートし、続いて高湿度チャンバ内で適切なビオチン化二次抗体(1/400;DAKO)と一緒に室温で1時間インキュベートし、次いで高湿度チャンバ内で室温で1時間ストレプトアビジン−セイヨウワサビ過酸化物化し(1/500;DAKO)、最後にDAB/H2O2と一緒にインキュベートした。次いで、組織切片をLillie−Mayerヘマトキシリンで対比染色し、水洗し、再水和し、DPXと一緒にスライドに載せた。Nanozoomer(Hamamatsu)を用いてスライドを走査して画像を得た。
(i)前立腺癌細胞におけるエンドソーム関連遺伝子及びタンパク質発現の増加
エンドソーム及びリソソーム関連遺伝子の発現をコントロール及び前立腺癌細胞におけるqRT−PCRによって定量化し、GAPDH mRNAの発現に対して規格化した。APPL1、APPL2、EEA1、RAB5A、RAB4A及びLIMP2 mRNA量は、非悪性コントロール細胞株よりも前立腺癌で有意に増加した(p<0.05;図1)。各場合において、mRNA発現が約2〜3倍増加した。前立腺癌細胞において検出されたRAB7A又はLAMP1 mRNAの量は非悪性コントロールに比べて有意差がなかった。ウエスタン分析によれば、前立腺癌細胞由来の抽出物におけるAPPL1、APPL2、EEA1、Rab4及びLIMP−2タンパク質量が非悪性コントロール細胞に比べて有意に増加した(p<0.05;図2)。さらに、LIMP−2とRab4の両方で、前立腺癌での増加は、非悪性コントロール細胞の約2〜4倍であった(図2)。非悪性細胞において検出されたRab5A、Rab7及びLAMP−1タンパク質量は前立腺癌細胞に比べて有意差がなかった(図2)。
エンドソーム及びリソソームの分布の代表的な共焦点像(図3)は、非悪性コントロールに比べて前立腺癌において染色が増加し、区画分布が変化した。APPL1陽性エンドソームは、非悪性コントロール細胞の核周囲領域で主に検出されたが、前立腺癌細胞においてはこれらの区画は細胞周辺に向かって分布し、細胞伸長部/仮足において原形質膜近くでより濃縮される傾向にあった。Rab5Aは、非悪性コントロール細胞と前立腺癌細胞の両方においてそのエフェクターEEA1に類似した分布を示した。しかし、非悪性コントロール細胞においてはRab5AとEEA1の両方が核周囲領域で濃縮されたが、前立腺癌細胞においてはこれらのエンドソーム区画は細胞質全体に見られ、一部の区画は細胞伸長部における細胞周辺に向かって位置した。Rab7陽性エンドソームは、主に、非悪性コントロール細胞と前立腺癌細胞の両方の核周囲領域に位置した。非悪性コントロール細胞においては、LIMP−2は、核周囲領域で濃縮され、細胞質の残部において、さらに細胞表面近くで、幾らかの管状及び点状(punctuate)小胞染色があった。それに対して、前立腺癌細胞は、細胞質全体に均一に分布した比較的小さいLIMP−2区画を示した。非悪性コントロール細胞においては、LAMP−1は、核周囲領域で高濃度である区画上で検出されたが、前立腺癌細胞においては、LAMP−1区画は、核周囲領域から離れて分布し、細胞伸長部で高濃度であった。LAMP−1染色と一致して、LysoTracker(商標)陽性酸性区画は、主に非悪性コントロール細胞の核周囲領域において高濃度であったが、前立腺癌細胞においては、これらの区画は、核周囲領域と細胞質伸長部の両方で検出された(図4)。
以前の研究は、前立腺癌細胞におけるトランスフェリン取り込みの増加を報告し、エンドソームタンパク質発現の増加及びエンドソーム分布の変化が認められたことに関連して受容体発現及びトランスフェリンエンドサイトーシスの調査を促した。非悪性コントロール細胞においては、エンドサイトーシスによって取り込まれたトランスフェリンが、5分後に点状細胞内構造に認められ、15及び30分で核周囲領域に認められた(図5)。前立腺癌細胞は、非悪性コントロール細胞株よりも多量のトランスフェリンをエンドサイトーシスによって取り込み、内部に取り込まれたトランスフェリンは、非悪性コントロールに比べて、前立腺癌細胞の核周囲領域に濃縮されず、細胞周辺に多い(図5)。前立腺癌では、非悪性コントロール細胞株に比べてアクチン染色も劇的に減少した(図5)。非悪性コントロール細胞においては、トランスフェリンは、核周囲領域に局在するLIMP−2及びRab7陽性エンドソームにおいてクラスターを形成した(図6)。前立腺癌細胞は、核周囲領域において幾らかのLIMP−2陽性染色を示し、ゴルジマーカーTGN46と幾らか共存したが、トランスフェリンの大部分は、細胞質全体及び細胞伸長部に分布した異なるエンドソーム区画(すなわちAppl1、Rab5A、EEA1)に局在した(図6)。Rab4リサイクリングエンドソーム及びLAMP−1陽性リソソームは、前立腺癌細胞株と非悪性コントロール細胞株で類似したトランスフェリン染色パターンを示した(図6)。トランスフェリン受容体の更なる分析によって、TfR1及びTfR2で遺伝子及びタンパク質の発現が変わりやすいことが明らかになった(図7)。前立腺癌細胞におけるTFR1遺伝子発現は非悪性コントロールに比べて有意に増加したが、前立腺癌細胞株22RV1ではTFR1タンパク質は定性的にしか増加せず、LNCaPでは増加しなかった(図7A)。前立腺癌細胞において検出されたTFR2タンパク質は非悪性コントロールに比べて有意に増加したが、TFR2遺伝子発現の増加はLNCaPでしか認められなかった(図7A)。したがって、前立腺癌におけるTFRタンパク質量は非悪性コントロール細胞に比べて増加したが、22RV1におけるTFR1及びLNCaP細胞におけるTFR2が比較的多かった。
非悪性コントロール細胞において検出されたAktタンパク質総量は、前立腺癌細胞において検出されたものと同様であった(図8)。しかし、前立腺癌株におけるリン酸化Akt量は異なり、22RV1はリン酸化Akt量が著しく減少したのに対して、LNCaPはリン酸化Akt量が増加した(図8)。より重要なことには、トランスフェリンの添加後、非悪性コントロール細胞におけるリン酸化Akt量は有意に増加したが、どの癌細胞においてもリン酸化Akt量が変化しなかった(図8B)。興味深いことに、(トランスフェリンで処理されなかった)LNCaP細胞において認められたリン酸化Akt量の増加は、この細胞株において検出されたTfR2量と相関した(図7A及び図8)。
免疫組織化学を利用して、前立腺癌患者組織試料におけるLAMP−1及びAPPL1の分布を調べた。リソソームマーカーLAMP−1は、一部の患者試料において幾らかの腫よう特異的染色を示したが(図9)、以前の研究と一致して、結果が変動し、一部の患者試料は、ほとんど又は全くLAMP−1染色を示さなかった(データ示さず)。しかし、APPL1は、癌周縁部を特異的に描出し、腫りゅう内の染色が劇的に増加した(図9)。このAPPL1染色パターンは、調べた6個の異なる患者試料の各々で一致した。
前立腺癌は、男性において最も頻繁に診断された癌のひとつであり、世界的な、特に米国及びオーストラレーシア人において癌関連死の一主要原因である。前立腺特異抗原は、依然として前立腺癌を検出するのに一般に使用される試験であるが、誤診及び予後予測の点で重大な問題がある。前立腺癌抗原3(PCA3:prostate cancer antigen 3)、コレステロール硫酸の分析、及びタンパク質プロテインキナーゼC−ζ(PRKCZ(protein kinase C−zeta))−PrCに翻訳される遺伝子プロテインキナーゼC−ゼータの新規配列を含めて、幾つかの有望な補助試験が最近開発された。しかし、これらのバイオマーカーは、適切な治療的介入を可能にする前立腺癌の早期の正確な検出のための有用な方法を与えない幾つかの欠点がある。
材料及び方法
(i)抗体試薬
この試験で使用した一次抗体は、hK2(1μg/mL、Abcam PLC、ケンブリッジ、英国、カタログ番号ab40948)、hK3(1μg/mL、Abcam、カタログ番号ab40949)、hK4(1μg/mL、Abcam、カタログ番号ab40950)、hK15(1μg/mL、Abcam、カタログ番号ab40961)、酸性セラミダーゼ(1μg/mL、Abcam、カタログ番号ab74469)に対するウサギポリクローナル抗体などであった。前立腺酸性ホスファターゼ(1μg/mL、Abcam、カタログ番号ab75704)、カテプシンB(0.25μg/mL、Abcam、カタログ番号ab58802)、カテプシンD(5μg/mL、Abcam、カタログ番号ab6313)に対するマウスモノクローナル抗体。αグルコシダーゼ(1μg/mL)、βグルコシダーゼ(1μg/mL)及びαガラクトシダーゼA(1μg/mL)に対するヒツジポリクローナル抗体は、リソソーム疾患研究ユニット(Lysosomal Diseases Research Unit)(女性及び小児病院(Women’s and Children’s Hospital)、SA Pathology Services、アデレード、南オーストラリア)の好意で提供された。LAMP−1(1μg/mL)マウスモノクローナルBB6は、ウメオ大学、ウメオ、スウェーデンの好意で提供された。ウエスタンブロット法用セイヨウワサビ−ペルオキシダーゼ(HRP:horseradish−peroxidase)複合二次抗体は、抗ヤギ/ヒツジ免疫グロブリン(1/2000、Merck Millipore Pty.Ltd.、ビクトリア、オーストラリア)、抗ウサギ免疫グロブリン(1/2000、Sigma Aldrich Pty.Ltd.、ニューサウスウェールズ、オーストラリア)、抗マウス免疫グロブリン(1/2000 Sigma Aldrich Pty.Ltd.)、HRP複合抗GAPDH(1/20000 Sigma Aldrich Pty.Ltd.)などであった。
細胞株PNT1a及びPNT2、22RV1及びLNCaPクローンFCGを細胞バンクオーストラリア(CellBank Australia)(小児医学研究所(Children’s Medical Research Institute)、ニューサウスウェールズ、オーストラリア)を介して欧州細胞培養コレクション(European Collection of Cell Cultures)から得た。細胞株RWPE−1、CaHPV10及びDU−145を、Cryosite(Cryosite Ltd.、ニューサウスウェールズ、オーストラリア)を介してアメリカ培養細胞系統保存機関(American Tissue Culture Collection)から入手した。これらの細胞株は、交差汚染又は誤認細胞株のリスト、バージョン6.8(2012年3月9日)にはない。細胞株をATCC及びECCCによって推奨された培地中で維持した。PNT1a、PNT2及び22RV1細胞株を、10%ウシ胎仔血清(FCS;fetal calf serum、In Vitro Technologies Pty Ltd.、ビクトリア、オーストラリア)及び2mM L−グルタミン(Sigma Aldrich Pty Ltd.、ニューサウスウェールズ、オーストラリア)を補充したロズウェルパーク記念研究所(RPMI:Roswell Park Memorial Institute)1640培地(Gibco(登録商標)、Life Technologies Australia Pty Ltd.、ビクトリア、オーストラリア)中で培養した。RWPE−1及びCaHPV10細胞株を、供給されたヒト組換え上皮成長因子1−53(EGF1−53)及びウシ下垂体抽出物(BPE:bovine pituitary extract)を補充した、L−グルタミンを含む角化細胞無血清培地(K−SFM:Keratinocyte Serum−Free Media)(Gibco(登録商標))中で培養した。DU−145細胞株を最小必須培地(MEM:minimum essential medium)(Gibco(登録商標))中で培養し、2mM L−グルタミン及び10%FCSを補充した。LNCaPを、2mM L−グルタミン、10%FCS、10mM HEPES及び1mMピルビン酸ナトリウムを補充したRPMI−1640培地中で培養した。細胞を加湿恒温器中で5%CO2と一緒に37℃でインキュベートした。

細胞溶解物10マイクログラムを加熱変性し(NuPAGE(登録商標)LDS試料緩衝剤及び還元剤中で100℃で5分間)、次いで前もって成形されたゲルを用いて、XCell SureLock Mini−Cellシステム(Life Technologies Australia Pty Ltd.、ビクトリア、オーストラリア)において120Vで1.5時間電気泳動させた。次いで、タンパク質をポリフッ化ビニリデン膜(Polyscreen(登録商標)、PerkinElmer、ビクトリア、オーストラリア)に移した。転写膜を0.1%(v/v)TBS−tween中の5%(w/v)脱脂乳溶液を用いて室温で1時間ブロックし、一次抗体と一緒に4℃で終夜インキュベートした。膜を0.1%(v/v)TBS−tweenで3×5分間洗浄し、次いでブロック溶液で1/2000希釈した適切なHRP複合二次抗体と一緒にインキュベートした。Novex(登録商標)ECL化学発光基質試薬キット(Life Technologies Australia Pty Ltd.、ビクトリア、オーストラリア)を用いて膜を現像し、ImageQuant(商標)LAS4000画像装置(GE Healthcare Bio−Sciences Pty Ltd.、ニューサウスウェールズ、オーストラリア)を用いてタンパク質を可視化した。3つ組試料を分析し、AlphaViewSA(商標)ソフトウェアv3.0.0.0(ProteinSimple、サンタクララ、カリフォルニア)を用いて基準GAPDH添加コントロールに対して画像を定量化した。クラスカル ワリス順位和統計分析をStata/SE v11.2(StataCorp LP、テキサス、米国)を用いて実施して、非悪性コントロール細胞株群と癌細胞株群の間の有意性を求めた(95%信頼限界;p<0.05)。
(i)前立腺癌細胞抽出物及び培地における現在の前立腺癌バイオマーカーの検出
ウエスタンブロット法を使用して、非悪性前立腺細胞株(RWPE−1、PNT1a及びPNT2)及び前立腺癌細胞株(22RV1、LNCaP、CaHPV10及びDU−145;図10)由来の細胞溶解物及び培地における前立腺酸性ホスファターゼ(PAP:prostatic acid phosphatase)、前立腺特異抗原(hK3)及び他のカリクレイン(hK2、hK4及びhK15)の量を規定した。PAPの細胞内量は、前立腺癌細胞株22RV1及びLNCaPにおいて非悪性コントロール細胞株よりも高いが、タンパク質はCaHPV10及びDU−145前立腺癌細胞株においてほとんど又は全く検出されなかった。同様に、hK2、hK3及びhK4は、LNCaPにおける細胞内タンパク質量が非悪性コントロール細胞株に比べて増加したが、他の前立腺癌細胞株においては最小量が検出された。細胞内hK15量は、すべての前立腺癌細胞株並びに非悪性コントロール細胞株RWPE−1及びPNT2で類似していたが、PNT1aにおいて検出された量は少なかった。PAP、hK3、hK2及びhK4は、LNCaP細胞から分泌され、量は、他の前立腺癌及び非悪性コントロール細胞株に比べて増加した。特に、22RV1及びLNCaPからのhK15の分泌は、他の前立腺癌及び非悪性コントロール細胞株に比べて増加した。これらの知見は、前立腺癌細胞株のタンパク質含有量及びPAP/カリクレイン分泌が可変であり、これらのマーカーが幾つかの前立腺癌細胞株と非悪性コントロール細胞株を識別できないことを示した。
ウエスタンブロット法を使用して、非悪性コントロール及び前立腺癌細胞株由来の細胞溶解物及び培地における異なるリソソームタンパク質量を規定した(図11、図12)。細胞内αグルコシダーゼ(αGluc)、βグルコシダーゼ(βGluc)、αガラクトシダーゼA(αGal A)、LAMP−1及びカテプシンD(26kDa)の量は、細胞株間で変動し、非悪性コントロールを前立腺癌細胞株から区別することができなかった。細胞抽出物におけるプロカテプシンD量は、非悪性コントロール、CaHPV10及びDU−145癌細胞株で類似していたが、22RV1及びLNCaP前立腺癌細胞株の細胞抽出物において検出可能なタンパク質はなかった。βグルコシダーゼ、αガラクトシダーゼ、プロカテプシンB及びDの分泌は、RWPE−1において他の非悪性細胞株PNT1a及びPNT2よりも増加した。RWPE−1から分泌されたαガラクトシダーゼの量は、CaHPV10前立腺癌細胞株と類似していた。LAMP−1及びカテプシンD(26kDa)分泌量は、非悪性コントロール及び前立腺癌細胞株の各々で類似していた。22RV1及びDU−145前立腺癌細胞株は、αグルコシダーゼ及びβグルコシダーゼ分泌が他の前立腺癌及び非悪性コントロール細胞株よりも増加した。他の前立腺癌及び非悪性コントロール細胞株に比べて限られた量のカテプシンB(22kDa)が22RV1前立腺癌細胞株において検出された。プロカテプシンB量は、LNCaPにおいて他の前立腺癌細胞株よりも増加した。RWPE−1は、プロカテプシンB量がPNT1a及びPNT2非悪性コントロール細胞株よりも増加した。カテプシンB(22kDa)の分泌は、CaHPV10、DU−145及びLNCaP前立腺癌細胞株が、非悪性コントロール細胞株及び22RV1前立腺癌細胞株よりも多かった。最小量の酸性セラミダーゼが非悪性コントロール細胞株並びに前立腺癌細胞株22RV1、CaHPV10及びDU−145において検出された。それに対して、多量の酸性セラミダーゼがLNCaP癌細胞株由来の細胞抽出物において検出された。酸性セラミダーゼ(10〜15kDa)は非悪性コントロール細胞株から分泌されなかったが、22RV1、DU−145及びLNCaP前立腺癌細胞株からはCaHPV10及び非悪性コントロール細胞株よりも多量に分泌された。これらの知見は、非悪性コントロール及び前立腺癌細胞株でのリソソームタンパク質の細胞内含有量及び分泌が可変であり、非悪性コントロール細胞株と前立腺癌細胞株を識別するカテプシンB及び酸性セラミダーゼの可能性を示した。したがって、酸性セラミダーゼ及びカテプシンBの分泌は、非悪性細胞株と癌細胞株を識別するある程度の能力をもたらし得る。
前立腺癌細胞においてエンドソーム生物学の変化が観察されたことは、特異的エンドソーム遺伝子及びタンパク質の発現の増加が、前立腺癌の発生及び又は進行に決定的に影響し得ることを示唆した。したがって、エンドソーム生合成の変化は、前立腺癌の診断及び予知に使用することができるバイオマーカーの研究に新しい道をもたらし得る。しかし、インビトロ観察と患者データを相関させて、新規変化が生物学的に関連することを確立することが重要であった。前立腺癌マイクロアレイデータベースにおけるエンドソーム関連遺伝子発現が変化することを検証することは、このプロセスにおける重要な第一ステップと考えられた。
(i)前立腺癌におけるリソソーム関連遺伝子の発現は、TFEB発現とは無関係に変化する。
エンドソーム及びリソソーム生合成に影響するLIMP2などのリソソーム関連遺伝子の転写は、転写因子EB(TFEB:transcription factor EB)によって調節される。したがって、TFEB遺伝子発現は、生体内でエンドソーム及びリソソーム生物学が変化した証拠となり得る。TFEBの発現は、前立腺癌においては非悪性コントロール前立腺組織に比べて変化せず(図13)、一方、LIMP2の発現は、前立腺癌においては非悪性組織よりも有意に増加した(P≦0.0001)。逆に、やはりTFEBの標的であるLAMP1は、前立腺癌における発現が非悪性コントロール組織に比べて有意に減少した(P≦0.01)。TFEBによって調節される他のリソソーム遺伝子を分析して、その発現がこの転写因子とは無関係に変化するかどうか判定した(図14)。カテプシンB(CTSB)は、テイラーコホートを用いた前立腺癌組織における発現が非悪性コントロール組織に比べて有意に減少し(P≦0.0001)、一方、αグルコシダーゼ(GAA)は、前立腺癌における発現が非悪性コントロール組織に比べて有意に増加した(P≦0.05)。酸性セラミダーゼ、βグルコシダーゼ(GBA)及びαガラクトシダーゼA(GLA)の発現は、テイラーコホートにおける前立腺癌組織において非悪性コントロール組織に比べて有意に変化しなかった。
APPL1遺伝子発現は、テイラーコホート由来の前立腺癌組織において非悪性コントロール組織に比べて有意に増加した(P≦0.05;図15)。しかし、APPL2、RAB5A、EEA1、RAB4A及びRAB7Aの発現の増加がインビトロで認められたが、原発性前立腺癌におけるこれらのエンドソーム関連遺伝子の発現は非悪性前立腺組織に比べて有意差がなかった(図15)。
エンドソーム及びリソソーム関連mRNA転写物の発現も、原発性癌及び非悪性組織に加えて前立腺上皮内腫よう(PIN)及び転移性癌組織の分析が可能なトムリンスコホートから分析した。TFEBなどの幾つかの遺伝子の発現データは、テイラーらによって使用された市販マイクロアレイに比べて、用いたマイクロアレイ技術が特注であり、プローブセットがより限定されるために、利用できなかった。トムリンスコホートにおけるLIMP2及びLAMP1の遺伝子発現は、非悪性コントロール組織と原発性前立腺癌組織で有意な変化がなかった(図16)。TFEBによって調節される他の遺伝子は、前立腺癌組織において非悪性コントロール組織に比べて発現に差があり、カテプシンBは、原発性癌組織において非悪性組織よりも有意に減少したが(P≦0.05)、トムリンスマイクロアレイ由来の非悪性組織と原発性癌組織ではカテプシンD、酸性セラミダーゼ、αグルコシダーゼ又はαガラクトシダーゼAの発現に有意な変化はなかった(図16)。このコホートにおけるβグルコシダーゼは、発現データを利用できなかった。
幾つかのリソソーム及びエンドソーム遺伝子の発現は、PIN、原発性癌及び転移性癌進行を通して変わるので、遺伝子発現の定量化は、前立腺癌の予知のための貴重なツールであり得る。リソソーム遺伝子の予知可能性を評価するために、本発明者らは、遺伝子発現に基づくK平均クラスタリングを利用して、グリンスキーコホート由来の患者を2つの群に分類した。グリンスキーコホートマイクロアレイは、生検時に検出される初期PSAレベルのデータ、及び治療後の再発についての情報を提供する。高又は低カテプシンB発現のクラスタリングによって、カテプシンB発現の少ない患者は、生化学的再発(BCR:biochemical recurrence)のリスクが有意に増加したことが明らかになった。(図18)。LIMP2、LAMP1、カテプシンD、酸性セラミダーゼ又はαガラクトシダーゼA遺伝子の高又は低発現によるグループ化は、患者を予後群に有意に層別化しなかったが、αガラクトシダーゼAでは傾向が認められた(P=0.077;図18)。テイラー及びトムリンスコホートにおいて認められた初期エンドソーム関連遺伝子の発現の増加から、本発明者らは、患者を予後群に層別化するこれらの初期エンドソーム関連遺伝子の可能性を分析した(図19)。しかし、K平均クラスタリングによって個々の初期エンドソーム遺伝子の発現に基づいて患者を2つの群に分類しても、患者は予後群に有意に区分されなかった。
診断時に7.9ng/mLを超えるPSAタンパク質を発現する患者は、BCRのリスクが低発現群に比べて有意に増加したが(HR2.422、P=0.0087;図20)、低PSAを発現するBCRの患者の一部(34%)は診断及び予後のアッセイの改善から利点を得るであろう。PSA≦7.8ng/mLの患者を高遺伝子発現群と低遺伝子発現群に層別化すると、高LIMP2発現の傾向は、BCRの増加を招くことが明らかになった(図20B)。LAMP1の発現は、予後値を示すようには見えなかった。カテプシンB発現によるクラスタリングは、低発現群の患者が高発現群に比べてBCRのリスクが有意であることを示した(HR0.2752、P=0.019;図20B)。αガラクトシダーゼAの低発現によっても、BCRのリスクがより高い群に患者を層別化することができた(HR0.3859、P=0.0396;図20B)。
エンドソーム及びリソソーム生合成は、通常、「CLEAR配列」を含むリソソーム遺伝子の転写因子の制御を介して、転写因子「EB」(TFEB)によって調節される。TFEBの標的遺伝子としては、前立腺癌細胞株において発現が異なるLIMP2及びLAMP1が挙げられる。本発明者らは、これらのマイクロアレイ由来の前立腺癌組織におけるLIMP2遺伝子発現の増加及びLAMP1遺伝子発現の減少も認めたが、TFEB発現に変化はなく、TFEBの正常な作用が前立腺癌において変化し得ることを示唆している。
結果
(i)細胞外エンドソームタンパク質は、非悪性細胞株と前立腺癌細胞株を区別する(図2)。前立腺癌由来の培地において検出されたAPPL1及びAPPL2の量は、非悪性コントロール細胞株に比べて有意に増加した(P≦0.01)。前立腺癌細胞株由来の培地において検出されたRab5Aの量も、非悪性コントロールに比べて有意に増加した(約20%;P≦0.05)。EEA1は、LNCaP及び22RV1前立腺癌細胞株由来の培地において検出されたが、非悪性コントロール細胞株PNT1a又はPNT2由来の培地においては検出されなかった(P≦0.01)。前立腺癌由来の培地において検出されたRab4の量は、非悪性コントロール細胞株に比べて有意に増加したが(P≦0.05)、後者、すなわちPNT1a及びPNT2細胞株において検出された量は変わりやすかった。前立腺癌由来の培地において検出されたRab7の量は、非悪性コントロール細胞株に比べて有意に減少した(P≦0.05)。
現在、前立腺特異抗原(PSA)は診断マーカーとして使用されるが、これは信頼性に問題があり、予想される癌発生過程を更に決定するためには侵襲性の生検を依然として必要とする。したがって、前立腺癌の正確な検出及び予知を可能にし、無用な侵襲性の費用のかかる手順を不要にする新しいバイオマーカーが緊急に必要である。本発明者らは、前立腺癌細胞株由来の培地において検出された初期エンドソームマーカーの量の有意な増加を認めた。特に、APPL1、APPL2、Rab5A及びEEA1並びにリサイクリングエンドソームタンパク質Rab4を含めた初期エンドソームに関連した小胞機構は、前立腺癌細胞株由来の培地において著しく増加したが、後期エンドソームタンパク質Rab7は、前立腺癌細胞において非悪性コントロールよりも有意に減少した。
材料
抗体及び蛍光プローブ
下表1に共焦点顕微鏡法分析に用いた蛍光複合物抗体の諸性質を詳述する。同じ宿主種の2種の一次抗体を用いて二重標識する場合、Zenon(登録商標)IgG標識化キット(Life Technologies Australia Pty Ltd.、ビクトリア、オーストラリア)を使用して、一次抗体を直接標識した。標識化プロトコルを製造者の指示に従って実施した。ウエスタンブロット法及び免疫蛍光に用いた一次抗体及び二次複合物の具体的濃度を下表に詳述する。
細胞株
細胞株PNT1a及びPNT2、22RV1及びLNCaPクローンFCGを細胞バンクオーストラリア(CellBank Australia)(小児医学研究所(Children’s Medical Research Institute)、ニューサウスウェールズ、オーストラリア)を介して欧州細胞培養コレクション(European Collection of Cell Cultures)から得た。細胞株RWPE1、CaHPV10及びDU145を、Cryosite(Cryosite Ltd.、ニューサウスウェールズ、オーストラリア)を介してアメリカ培養細胞系統保存機関(American Tissue Culture Collection)から入手した。これらの細胞株は、交差汚染又は誤認細胞株のリスト(List of Cross−Contaminated or Misidentified Cell Lines)バージョン6.8(2012年3月9日)にはない。
PNT1a、PNT2及び22RV1細胞株を、10%ウシ胎仔血清(FCS)(In Vitro Technologies Pty Ltd.、ビクトリア、オーストラリア)及び2mM L−グルタミン(Sigma Aldrich Pty Ltd.、ニューサウスウェールズ、オーストラリア)を補充したロズウェルパーク記念研究所(RPMI)1640培地(Gibco(登録商標)、Life Technologies)中で維持した。RWPE1及びCaHPV10細胞株を、供給されたヒト組換え上皮成長因子1 53(EGF1 53)及びウシ下垂体抽出物(BPE)を補充した、L−グルタミンを含む角化細胞無血清培地(K−SFM)(Gibco(登録商標))中で維持した。DU145細胞株を最小必須培地(MEM)(Gibco(登録商標))中で維持し、2mM L−グルタミン及び10%FCSを補充した。癌細胞株LNCaPを、2mM L−グルタミン、10%FCS、10mM HEPES(Life Technologies)及び1mMピルビン酸ナトリウム(Sigma Aldrich)を補充したRPMI1640培地中で維持した。Sanyo MCO−17AI CO2恒温器(Sanyo Electric Biomedical Co.,Ltd.、大阪、日本)中で加湿恒温器中で37℃で5%CO2と一緒に細胞をインキュベートした。
免疫ブロッティングのための細胞抽出物調製
細胞株をT75フラスコ中に播種し、37℃及び5%CO2でインキュベートした。80〜90%コンフルエントで、培地を吸引し、細胞をdPBSで1回洗浄し、細胞を無血清培地と一緒に48時間インキュベートした。
ウエスタンブロット法に関して上述したように細胞株を75cm2フラスコ中で3つ組で血清枯渇状態で48時間培養した。培地を吸引し、フラスコをdPBSで洗浄した。1mL TRI試薬(登録商標)(Applied Biosystems Pty Ltd.、ビクトリア、オーストラリア)を各T75フラスコに添加し、ゴム製の細胞かき取り器を用いてかき取り、押し上げ蓋の付いた1.5mL管に移した。更に必要になるまで試料を−80℃で貯蔵した。
細胞培養物を通常通りに継代し、22mm「Number 1」カバーガラス(Menzel−Glaser、Thermo Fisher Scientific Australia Pty Ltd.、ビクトリア、オーストラリア)上に低密度で播種し、培地中で37℃、5%CO2雰囲気で48時間インキュベートした。48時間後、培地を吸引し、4%ホルムアルデヒド(v/v)PBS溶液を用いて細胞を室温で20分間固定した。固定細胞をPBS中の0.1%Triton−X(v/v)で10分間更に透過処理した。
SDS−PAGE及び転写
細胞溶解物由来の総タンパク10μgを、試料緩衝剤(NuPAGE(登録商標)LDS試料緩衝剤(4x)、Life Technologies)及び50mMジチオトレイトールを含む還元剤(NuPAGE(登録商標)試料還元剤(10x))中で100℃で5分間加熱変性した。SDS−PAGE泳動用緩衝液(Life Technologies)で緩衝されたXCell SureLock(商標)Mini−Cell電気泳動システムにおいて10%15ウェル又は12%17ウェルSDS−PAGEプレキャストゲルを用いて調製試料を120V(固定、電流は可変)で1.5時間分離した。電気泳動後、分離したタンパク質をポリフッ化ビニリデン(PVDF:polyvinylidene difluoride)膜(Polyscreen(登録商標)、Perkin−Elmer)に35Vで1時間移した。
膜をTBS−tween(0.1%)中の5%w/v脱脂乳溶液を用いて室温で1時間ブロックし、5%脱脂乳で希釈した適切な抗体(表2.2)と一緒に静かに撹拌しながら4℃で終夜インキュベートした。一次抗体と一緒にインキュベーション後、膜をTBS−tweenで3回室温で5分間洗浄した。5%脱脂乳溶液で1/2000希釈したセイヨウワサビ−ペルオキシダーゼ複合二次抗体(表2.2)をPVDF膜と一緒に静かに撹拌しながら室温で1時間インキュベートした。
一次抗体検出中に、PVDF膜を剥離し、総タンパク質量の添加コントロールとして用いたGAPDHを検出した。Milli−Q H2O中に0.02M Tris、0.5M NaCl、2%SDS及び100mMβメルカプトエタノールを含む剥離緩衝剤でPVDF膜を2回洗浄した。膜を緩衝剤と一緒に55℃で30分間激しく撹拌し、さらにTBS−tweenと一緒に室温で15、20及び30分間の3回激しく撹拌して洗浄した。PVDF膜をTBS−tween中の5%脱脂乳を用いて室温で1時間ブロックし、HRP複合αGAPDH抗体の1:20,000希釈を用いてGAPDHを検出した。
RNA抽出
TRI試薬(登録商標)を用いて凍結させた細胞を解凍し、室温で5分間インキュベートした。細胞収集に用いたTRI試薬(登録商標)各1ミリリットルにクロロホルム200μLを添加した。試料を1分間激しく振とうし、室温で3分間静置した。次いで、試料を16,000g、4℃で15分間遠心分離した。
qRT−PCR用の相補DNA(cDNA)をHigh Capacity RNA−to−cDNAキット(Life Technologies)を用いて製造者の指示に従って調製した。RNA2μgを含む反応物20μLの場合、主混合緩衝剤10μL及び逆転写酵素1μLを添加し、RNaseを含まないH2Oで体積を20μLにした。DNA Engine(登録商標)ペルチェサーマルサイクラー(Bio−Rad Laboratories Pty,Ltd.、ニューサウスウェールズ、オーストラリア)を用いて増幅を行った。反応混合物を37℃で1時間、続いて95℃で5分間インキュベートし、次いで4℃に冷却した。cDNA試料を必要になるまで−20℃で貯蔵した。
下表2.4に詳述したプライマー配列を文献、Harvard PrimerBankから得た、又はNCBI Primer BLASTを用いて設計した。Harvard PrimerBankから選択されたプライマーは、最終アンプリコンサイズが150塩基対未満であり、その融解温度が約60℃であり、可能であれば、エキソン−エキソン接合部にわたる範囲であるという判定基準に基づいて選択された。Primer−BLASTを用いて設計されたプライマーもこれらの判定基準に基づいた。オリゴヌクレオチドをGeneWorks Pty Ltd、南オーストラリアから購入した。
5μLのPower SYBR(登録商標)Green PCR Master Mix(Life Technologies)、各0.5μLの10nM順方向及び逆方向プライマー、DEPC処理H2Oで1:25希釈されたcDNA試料2μLを含み、DEPC処理H2O2μLで10μLにした、反応混合物10μLを準備した。反応物を3つ組で96ウェルプレート(Life Technologies)上で培養した。各プレートは、反応効率を制御するために、標的遺伝子と内在性遺伝子の検量線のための(LNCaP細胞株由来の)基準cDNA試料の段階希釈物を含んだ。
すべての標的のサイクリング条件は、酵素を活性化し、cDNAを変性させるために50℃で2分間、95℃で10分間、続いて95℃で15秒の変性ステップと60℃で60秒の伸長及び信号取得ステップを40サイクルであった。各実行後に溶融曲線を作成して、プライマー二量体又は生成物及び他の混入物がないことを確認した。
免疫蛍光標識化
固定細胞を5%(w/v)ウシ血清アルブミン(BSA:bovine serum albumin)PBS溶液中でゆっくり撹拌しながら室温で2時間インキュベートすることによって非特異抗体反応性を低下させた。次いで、細胞を、5%BSAで希釈した適切な濃度の一次抗体(表2.3)と一緒に室温で2時間インキュベートし、続いて二次抗体と室温で1時間インキュベートした。結合していない抗体をPBSで3回洗浄して除去し、DAPI核染色を含むProLong(登録商標)Gold Antifade試薬(Life Technologies)と一緒にカバーガラスを1mm顕微鏡スライド上に載せた。
共焦点顕微鏡法をZeiss LSM710META NLOレーザー走査顕微鏡(南オーストラリア大学(University of South Australia)、オーストラリア)及び付属のCarl Zeiss Zen2009ソフトウェアを用いて実施した。370、488、543及び633nmのレーザー線をDAPI、Alexa Fluor(登録商標)488、Cy3及びAlexa Fluor(登録商標)633蛍光に利用した。レーザー強度を蛍光標的に応じて5%から25%に設定した。蛍光団間の信号のクロスオーバーを除去するために、各チャネル間のギャップ20nmで発光蛍光をフィルターにかけた。各チャネルを4ライン平均の面順次走査によってピクセル滞留時間3.15μsでキャプチャーした。各レーザーのピンホール直径を1エアリー単位に設定した。細胞を絞り1.4の63x油浸対物レンズで観察し、2x光学ズームを用いた。画像を分解能1024px2、16ビットグレースケール深さでキャプチャーした。画像キャプチャー分解能1024ピクセル及びレンズ/ズーム比に基づいて、各画素は、寸法0.066μMに相当する。少なくとも4個の細胞を各カバーガラス上で無作為に選択し、代表的画像を作図のために処理した。画像をグレースケール16ビットTIFFファイルとしてエクスポートし、Adobe(登録商標)Photoshop(登録商標)CS5(Adobe Systems Inc.、サンホゼ、カリフォルニア、米国)を用いて更に処理した。
LysoTracker(登録商標)陽性小胞を生細胞共焦点顕微鏡法によって可視化して、固定細胞において生じ得るLysoTracker(登録商標)の漏出を防止した。細胞をDKSH疎水性チャンバスライド(DKSHオーストラリアPty Ltd.、ビクトリア、オーストラリア)中で標準状態で培養した。続いて、LysoTracker(登録商標)を培地で1/200希釈し、細胞に添加した。37℃で5分間インキュベーション後、細胞培養物を可視化した。
細胞を本明細書に詳述した通常状態でインキュベートした。48時間後、トランスフェリン633複合物の1/1000希釈物を含む新しい培地で培地を置換した。細胞をトランスフェリン633培地と一緒に20分間インキュベートした。続いて、培地を吸引し、dPBSで短時間洗浄し、固定し、透過処理した。一次及び二次抗体による標識化は、本明細書に詳述したように進行した。
テイラーコホートは、メモリアル スローン ケタリングがんセンター(MSKCC)において前立腺全摘出術による治療を受けた患者からなり、150個の前立腺癌及び29個の非悪性組織のプロファイリングをAffymetrix Human Exon1.0STアレイを用いて実施した。データをcBio Cancer Genomics Pathway Portalから得た。チャンドラン(Chandran)コホートは、18個の正常組織、65個の一次組織、25個の転移性組織及び63個の正常な隣接非悪性組織で構成される。これらの組織の発現プロファイリングをAffymetrix U95Av2ヒト遺伝子アレイを用いて行った。データをNCBI GEO(受託番号GSE6919)から取得した。トムリンスによるコホートをNCBI GEO(受託番号GSE6099)から取得した。トムリンスによるコホートは、32個の一次組織試料、20個の転移性組織試料、13個の前立腺上皮内腫よう(PIN)組織試料及び27個の正常組織試料で構成される。チナイヤンヒト20K Hs6アレイを用いて前もって試料の分析を行った。MSKCCからの追加のコホートであるグリンスキーコホートは、79個の悪性前立腺組織で構成され、前立腺癌の生化学的再発(BCR)を評価するのに使用される106か月までの臨床経過観察データを含む。これは、PSAレベル≧0.2ng/mLで規定される29名のBCR患者及び50名の悪化のない患者を含む。
15個のアミノ酸の長さを有するエピトープを、ヒトAPPL1又はRAB7のタンパク質配列に対する抗体の合成後の検出に適した強い抗原性を有すると予測されたこれらの特異的配列から選択した。

(i)癌進行中のエンドソーム関連遺伝子発現の変化
エンドソーム関連mRNA転写物の発現を、原発性癌及び非悪性組織に加えて前立腺上皮内腫よう(PIN)及び転移性癌組織の分析が可能なトムリンスコホートから分析した。
エンドソーム関連遺伝子の発現の予知可能性を評価するために、本発明者らは、遺伝子発現に基づくK平均クラスタリングを利用して、グリンスキーコホート由来の患者を2つの群に分類した。グリンスキーコホートマイクロアレイは、生検時に検出される初期PSAレベルのデータ、及び治療後の再発についての情報を提供する。
図27は、≦7.8ng/mL PSAを発現する癌患者のリソソーム遺伝子発現のカプラン マイヤー生存分析及び多変量解析を示す図である。リソソーム関連遺伝子発現のカプラン マイヤー分析は、IGF2R、SORT1又はPDCD6IP発現に基づいて≦7.8ng/mL PSAを発現する患者における有意な予知可能性を示した。PSA≦7.8ng/mLの予後良好亜群から、M6PR、IGF2R、SORT1、STEAP4、MYO1B、PDCD6IP、SDC1及びSDCBPの遺伝子発現に基づくK平均クラスタリングによって患者を更に2つの群に層別化した(高発現−黒線、低発現−灰色線)。統計解析をゲーハン−ブレスロー−ウィルコクソン検定によって実施した。BCR:生化学的再発、HR:危険率、CI:信頼区間。
テイラーコホート由来の前立腺癌組織におけるシンタキシン7及びシンタキシン12mRNA転写物の発現を非悪性組織と比較して分析した。
シンタキシン7及び12遺伝子発現の発現の予知可能性を評価するために、本発明者らは、遺伝子発現に基づくK平均クラスタリングを利用して、グリンスキーコホート由来の患者を2つの群に分類した。

前立腺癌の存在の診断は、本明細書に記載のマーカーのいずれかについての1つ以上のmRNAの1つ以上のmRNA発現レベル、本明細書に記載のマーカータンパク質のレベル、本明細書に記載のマーカータンパク質の分泌、本明細書に記載の体液中のマーカータンパク質の存在に基づいて、又は本明細書に記載のマーカータンパク質のいずれかの組織若しくは生検試料上の免疫組織学に基づいて、成すことができる。
臨床病期T1〜T2a、グリーソンスコア2〜6、PSA<10ng/mL、平均余命<10年の患者の治療は、監視療法を含む。
臨床病期T2b〜T2c、グリーソンスコア7、PSA10〜20ng/mL、平均余命<10年の患者の治療は、監視療法を含む。
臨床ステージT3a、グリーソンスコア8〜10、PSA>20ng/mL
治療選択肢としては、放射線療法(線量78〜80+Gy)と3D−CRT/IMRTプラス2〜3年の長期ネオアジュバント/同時/アジュバントADT、又は放射線療法(線量78〜80+Gy)と3D−CRT/IMRTと毎日のIGRTプラス4〜6か月の短期ネオアジュバント/同時/アジュバントADTを伴う若しくは伴わない密封小線源治療(推奨線量率:ヨウ素125の場合145Gy、及びパラジウム103の場合125Gy)、又は修復のない選択患者の場合のRPプラスPLNDが挙げられる。

- 対象における前立腺癌を検出する方法であって、エンドソーム関連マーカー及び/又はリソソーム関連マーカーから選択されたマーカーを前記対象から検出し、それによって前記対象における前立腺癌を検出するステップを含む方法。
- 前記選択されたマーカーが、初期エンドソームマーカー、後期エンドソームマーカー、エンドソーム生合成に関連するマーカー、エンドソーム輸送に関連するマーカー、及びエンドソーム再利用に関連するマーカーの1種以上を含む、請求項1に記載の方法。

- 前記マーカーのレベルが、再発率の増加、及び/又は生存率の低下を示す、請求項30に記載の方法。
- 前記マーカーが、LIMP2、カテプシンB、カテプシンD、αガラクトシダーゼ、RAB5A、EEA1、RAB7A、M6PR、IGFR2、SORT1、MYO1B、PDCD6IP、SDC1、STX12、FGF2、FGF3、及び/又は前記のうち1種をコードするmRNA、前記のうち1種の断片、前記のうち1種の誘導体、及び前記のうち1種の加工体の1種以上を含む、請求項40に記載の方法。
- 前記マーカーが、増加した若しくはより高いLIMP2、減少した若しくはより低いカテプシンB、減少した若しくはより低いカテプシンD、減少した若しくはより低いαガラクトシダーゼ、増加した若しくはより高いRAB5A、増加した若しくはより高いEEA1、減少した若しくはより低いRAB7A、減少した若しくはより低いM6PR、増加した若しくはより高いIGFR2、増加した若しくはより高いSORT1、増加した若しくはより高いMYO1B、減少した若しくはより低いPDCD6IP、減少した若しくはより低いSTX12、減少した若しくはより低いFGF2、減少した若しくはより低いFGF3、及び/又は前記のうち1種をコードするmRNA、前記のうち1種の断片、前記のうち1種の誘導体、及び前記のうち1種の加工体の1つ以上を含む、請求項40に記載の方法。
- 前記方法が、さらに、前記対象におけるPSA発現レベルを確認するステップ、及び前記対象における前記PSA発現レベルに基づいて前記マーカーの発現を層別化するステップを含む、請求項40から43のいずれか一項に記載の方法。
- 前記PSAが、低リスクの前立腺癌を示すレベルである、請求項43に記載の方法。
- 前記PSAレベルが10ng/ml未満である、請求項44に記載の方法。
- 前記PSAが7.8ng/ml以下のレベルである、請求項44又は45に記載の方法。
- 前記マーカーが、LIMP2、カテプシンB、αガラクトシダーゼ、RAB5A、EEA1、M6PR、IGFR2、SORT1、MYO1B、PDCD6IP、SDC1、STX12、FGF2、FGF3、及び/又は前記のうち1種をコードするmRNA、前記のうち1種の断片、前記のうち1種の誘導体、及び前記のうち1種の加工体の1種以上を含む、請求項44から46のいずれか一項に記載の方法。
- 前記マーカーが、増加した若しくはより高いLIMP2、減少した若しくはより低いカテプシンB、減少した若しくはより低いαガラクトシダーゼ、増加した若しくはより高いRAB5A、増加した若しくはより高いEEA1、減少した若しくはより低いM6PR、増加した若しくはより高いIGFR2、増加した若しくはより高いSORT1、増加した若しくはより高いMYO1B、減少した若しくはより低いPDCD6IP、減少した若しくはより低いSDC1、減少した若しくはより低いSTX12、減少した若しくはより低いFGF2、減少した若しくはより低いFGF3、及び/又は前記のうち1種をコードするmRNA、前記のうち1種の断片、前記のうち1種の誘導体、及び前記のうち1種の加工体の1つ以上を含む、請求項47に記載の方法。
- 請求項1から48のいずれか一項に記載の方法を実施するためのキット。
- 対象における前立腺癌を治療する方法であって、
エンドソーム関連マーカー及び/又はリソソーム関連マーカーから選択されたマーカーを前記対象から検出するステップ、並びに
検出された前記選択されたマーカーの存在、レベル、分泌及び分布の1つ以上に基づいて前記対象を治療するステップ
を含む方法。 - ASNDHDAAINRYSRLSKKRENDKVKYEVTEDVYT(配列番号1)、DEVASDPLYVPDPDPTKFPVNRNLTRKAGYLNARNKT(配列番号2)、SEGQFVVLSS SQSEESDLGEG GKKRESEA(配列番号3)の1つ以上のアミノ酸配列、前記アミノ酸配列のいずれかの抗原性断片、及び/又は前記アミノ酸配列のいずれか若しくはその抗原性断片の変異体を含むポリペプチドに対する単離又は精製抗体。

- PNTFKTLDSWRDEFLIQASPRDPENFPFVVLGNKI(配列番号4)、DPENFPFVVLGNKIDLENRQVATKRAQAWCYSKNN(配列番号5)、ALKQETEVELYNEFPEPIKLDKNDRAKASAESCSC(配列番号6)の1つ以上を含むヒトRAB7タンパク質中のアミノ酸配列におけるエピトープ、及び/又は前記タンパク質のホモログ、オルソログ若しくはパラログの等価な領域に結合した単離及び/又は精製抗体。
- APPL1タンパク質又はその断片を検出する方法であって、請求項51又は52に記載の抗体を使用して前記APPL1タンパク質又はその断片を検出するステップを含む方法。
- RAB7タンパク質又はその断片を検出する方法であって、請求項53又は54に記載の抗体を使用して前記RAB7タンパク質又はその断片を検出するステップを含む方法。
- 対象における前立腺癌を検出する方法であって、請求項51から54のいずれか一項に記載の抗体を使用して、APPL1若しくはRAB7タンパク質、及び/又はその断片、誘導体若しくは加工体を前記対象から検出するステップを含む方法。
- 前立腺癌の診断及び/又は予知のための選択されたマーカーを特定する方法であって、
エンドソーム関連マーカー及び/又はリソソーム関連マーカーから選択されたマーカーを特定するステップ、並びに
前立腺癌を診断及び/又は予知する前記選択されたマーカーの能力を判定するステップ
を含み、
それによって前記マーカーを前立腺癌の診断及び/又は予知のための選択されたマーカーであると確認する方法。
