材料及び方法
ダイヤモンドディスクソーを用いて、サンゴ外層を除き、そして所望する寸法の代表的な小さなセクションを、サンゴブロックから切断した。
種々の領域からのサンプル除去が、それらの物理的特徴において変化する材料を提供するかどうか、及びそのような変動がそのような特徴に他の性質を提供するかどうかを決定するために、血液及び他の列挙される流体を、サンゴサンプルに適用し、そしてサンゴサンプル内の流体の吸収を評価した。
実施例1における発見に基づいて、細胞又は組織の増殖又は機能回復を促進するための最適化されたサンゴ系の固体基材を選択するためのスクリーニングプロトコルを確立することができる。図2は、想定されるスクリーニングプロトコル工程の流れ図を提供する。サンゴサンプルを、同定し、単離し、そして所望するサイズ及び形状に機械処理するか、又はブロック下で評価する。次に、サンプルを洗浄し、そして任意には、殺菌し、次に乾燥する。評価されるサンゴサンプルを、真空化で乾燥し、そして/又はこの端部に向かって加熱することができる。
実施例1のプラグにおける血液吸収の表現型変動の結果を評価するために、サンゴプラグを、3回の次亜塩素酸洗浄、過酸化水素処理及び複数回の再蒸留水洗浄を含む標準生成方法を用いて調製した。それらの自発的流体摂取及び全流体摂取値を、実施例2に記載のようにして決定し、そして水が、この場合、評価されるサンプル流体である。75%超の自発的流体摂取値を示すサンプルインプラントをまた、それらの自発的血液摂取容量について調べた。
細胞又は組織の増殖又は機能回復を促進することへの適用のために、固体基材を、流体、例えば、水を吸収するそれらの能力について評価する。次に、少なくとも75%の特異的流体摂取容量値を提供する基材を、所望する組織部位に移植する。例えば、及び1つの実施形態によれば、そのような固体基材を、骨軟骨欠損又は疾患における修復又は再生への適用のために軟骨及び隣接する骨内に移植することができる。
天然の表面は、それらの可変材料組成、表面粗さ及び多孔性のために不均一であり、そして従って、可変撥水性/接着特性を示す。接触角測定は、表面のトポグラフィー及び化学構造を考慮して、粗い表面の湿潤性を特徴づけることができる。
サンゴサンプルを、上記実施例1に記載されるようにして処理した。次に、サンプルを、標準方法に従って、環境走査型電子顕微鏡及び原子間力顕微鏡にかけた。
材料及び方法
ヒドロサンゴ ポリテス・ルテア(Porites lutea)のサンゴサンプルを、実施例1に記載のようにして単離し、そしてプラグを調製した。
実施例2は、少なくとも75%の特異的流体摂取容量値により特徴付けられる固体基材が細胞又は組織の増殖又は機能回復を提供し、そして海洋生物骨格誘導体の所定の単離されたブロックにおいて、所望する特異的流体摂取容量値を示す、それらから単離され得るプラグの数に関して、変動性が存在することを示した。従って、40%未満、又は41%〜74%、又は75%〜99%の特異的流体摂取容量値により特徴づけられるサンプルが、細胞又は組織の増殖又は機能回復を提供する各基材の能力を改善するよう、高められ得るかどうかを決定することが興味の対象であった。
ヒドロサンゴポリテス・ルテア(Porites lutea)のサンゴサンプルを、実施例1に記載のようにして単離し、そしてインプラントを調製する。インプラント20−10を、保持用カセット20−20に配置し、そしてカバー20−70を装置上に配置する。インプラントが十分に乾燥し、そして次に、適用が停止されるまで、負圧を、真空20−30を介して、所定の時間、適用する。次に装置は、乾燥重量を確立するために、各インプラントの重量を個別に計量する。充填を促進する自動サイクルが開始され、そして所望する流体レベルが、その工程の間、維持される。自発的流体摂取を可能にするために、カセット20−20を、流体内のカセットマニピュレーター20−40を通して、個々に上昇させ、そして第1流体レベルに低下し、そしてインプラントマニピュレーター20−60が、自発劇流体摂取値を決定するために、重量決定のための個々のインプラントを指向し/移動し、続いて任意には、乾燥/ブロッティングステーションの先に各インプラントの通過を指向し/移動せしめる。個々のインプラントをすべて計量し、そしてカセット内のそれらの位置に戻す。次に、カセット20−20を、個々に上昇させ、そしてインプラントの十分な浸漬けを促進するために、過剰の流体内に、カセットマニピュレーター20−40を通して、第2の有意に高い流体レベルに下げ、そして負圧を、一定の時間、真空を介して再び適用し、各インプラント内に最大の流体摂取を確保する。インプラントマニピュレーター20−60は再び、全流体摂取を提供する、第2重量決定のために個々のインプラントを、個々に指向し/移動せしめる。装置のデータ処理ユニットは、特異的流体摂取値の出力を決定し、そして提供し、任意には、サンプルが示される基準に基づいて選択されるべきであることを特異的に同定する。
材料及び方法
ヒト胚口蓋間葉(HEPM)細胞を、適切な増殖培地において増殖した。
細胞付着が、評価されたすべてのサンプルにおいて、播種の1日後ほどで見出され、ここで最適化された(opt)とは、SEM分析により評価される場合、75%超の特異的流体摂取容量値を有し、そして非最適化された(nonopt)とは、60%未満の特異的流体摂取容量値を有する。
材料及び方法
サンゴサンプルを、実施例1に記載される方法に従って、単離し、そして調製した。サンプルを、[Chemical staining methods used in the identification of carbonate minerals, Tamer AYAN, Mineral Research and Exploration Institute of Turkey http://www.mta.gov.tr/v2.0/eng/dergi_pdf/65/11.pdf]に記載のようなプロトコルに従って染色した。手短に言及すると、ファイグル溶液を、標準方法に従って調製する。次に、サンゴサンプルを、ファイグル溶液により染色し、そしてそれらの特異的流体摂取容量値について前に評価されたサンプルを、それにより染色した。この場合、実施例9において評価されたような2種のサンプル、すなわち95%の特異的流体摂取容量値を有するサンプルR91、及び42%の特異的流体摂取容量値を有するサンプルR48を評価した。

・少なくとも75%の特異的流体摂取容量値により特徴づけられるか、又は、流体と接触するときに60°未満の接触角値を有することにより特徴づけられる、海洋生物骨格誘導体系の固体材料を選択すること

・少なくとも75%の特異的流体摂取容量値により特徴づけられるか、又は、流体と接触するときに60°未満の接触角値を有することにより特徴づけられる、海洋生物骨格誘導体系の固体材料を選択し;
・海洋生物骨格誘導体系の固体材料上の実質的に粗い表面の存在を確立し、ここで、前記実質的に粗い表面は、電子顕微鏡、X線回折又は原子間力顕微鏡を走査することにより決定され;そして
・海洋生物骨格誘導体系の固体材料上での実質的に粗い表面の存在の決定により特徴づけられる、海洋生物骨格誘導体系の固体材料を選択すること
を含んで成る、方法。 - 前記方法が、前記確立工程においてE−SEMによる湿潤性の評価をさらに含む、請求項46に記載の方法。
- 細胞又は組織の増殖又は機能回復を促進するための、最適化された海洋生物骨格誘導体系の固体基材の選択方法であって、
・海洋生物骨格誘導体系の固体材料を単離するか又は調製し;
・前記海洋生物骨格誘導体系の固体材料の特異的流体摂取容量値を確立するか、又は接触角値を確立し、ここで、前記特異的流体摂取容量値は、全流体摂取値により割り算された自発的流体摂取値を確立することにより決定され;
・少なくとも75%の特異的流体摂取容量値により特徴づけられるか、60°未満の接触角値を有することにより特徴づけられる、海洋生物骨格誘導体系の固体材料を選択し;そして
・X線回折又はファイグル染色陽性の使用により、前記海洋生物骨格誘導体系の固体材料の結晶組成又は構造を確立すること
を含んで成る、方法。 - 細胞又は組織の増殖又は機能回復を促進するために準最適な海洋生物骨格誘導体基材の固体材料を、最適化された海洋生物骨格誘導体系の固体基材に転換するための方法であって、
a.海洋生物骨格誘導体系の固体材料グループの特異的流体摂取容量値を確立し、ここで前記特異的流体摂取容量値は、前記グループにおける各サンプルについて、全流体摂取値により割り算された自発的流体摂取値を確立することにより決定され;
b.特異的流体摂取容量値により特徴づけられる海洋生物骨格誘導体系の固体材料を選択し;
c.(b)の前記海洋生物骨格誘導体系の固体材料と、両親媒性物質、極性溶媒、カチオン性物質、アニオン性物質、又はそれらの組み合わせとを接触し;
d.(c)で得られた前記海洋生物骨格誘導体系の固体材料において(a)におけるような特異的流体摂取容量を決定し;そして
e.新しく確立された、高められた特異的流体摂取容量値を有する、(d)で得られた海洋生物骨格誘導体系の固体材料を選択すること
を含んで成る、方法。 - 前記高められた特異的流体摂取容量値が、少なくとも5%高められる、請求項49に記載の方法。
- 前記選択された海洋生物骨格誘導体系の固体材料が、75%〜100%の特異的流体摂取容量値により特徴づけられる、請求項50に記載の方法。
- 前記高められた特異的流体摂取容量値が、少なくとも15%高められる、請求項49に記載の方法。
- 前記選択された海洋生物骨格誘導体系の固体材料が、45%〜70%の特異的流体摂取容量値により特徴づけられる、請求項52に記載の方法。
- 前記高められた特異的流体摂取容量値が、少なくとも35%高められる、請求項49に記載の方法。
- 前記選択された海洋生物骨格誘導体系の固体材料が、1%〜40%の特異的流体摂取容量値により特徴づけられる、請求項54に記載の方法。
- 前記固体基材が、サンゴ又はサンゴ系の誘導体から実質的に構成される、請求項49に記載の方法。
- 前記固体基材が、アラゴナイト、カルサイト、ヒドロキシアパタイト又はその組み合わせから実質的に構成される、請求項49に記載の方法。
- (d)における前記特異的流体摂取容量値を確立する前、アラゴナイトである海洋生物骨格誘導体系の固体材料を、ヒドロキシアパタイトに完全に又は部分的に転換するか又は前記海洋生物骨格誘導体系の固体材料をヒドロキシアパタイトに完全に又は部分的に被覆する工程をさらに含んで成る、請求項49に記載の方法。
- (d)における前記特異的流体摂取容量値の確立後に、アラゴナイトである海洋生物骨格誘導体系の固体材料を、ヒドロキシアパタイトに完全に又は部分的に転換する工程をさらに含んで成る、請求項49に記載の方法。
- 前記海洋生物骨格誘導体系の固体材料が、骨充填材、骨ペースト又は骨代替材として使用され得る、請求項59に記載の方法。
- 前記両親媒性材料、極性溶媒、カチオン性材料、又はアニオン性材料が、Tween、プルロニック、エタノール、メチレンブルー、ヒアルロン酸、コンドロイチン硫酸、又はそれらの組み合わせである、請求項59に記載の方法。

- 前記海洋生物骨格誘導体系の固体材料がさらに、骨充填材又は骨代替材を含む、請求項59に記載の方法。
- 前記海洋生物骨格誘導体系の固体材料と流体とを、0.1〜15分間接触し、前記自発的流体摂取値に到達するよう、前記海洋生物骨格誘導体系の固体材料内の前記流体の自発的流体摂取を可能にする工程をさらに含んで成る、請求項59に記載の方法。
- 前記海洋生物骨格誘導体系の固体材料と流体とを、0.05〜24時間接触し、前記自発的流体摂取値に到達するよう、前記海洋生物骨格誘導体系の固体材料内の前記流体の自発的流体摂取を可能にする工程をさらに含んで成る、請求項59に記載の方法。
- 前記海洋生物骨格誘導体系の固体材料と流体とを接触し、そして海洋生物骨格誘導体系の固体材料に負の圧力を適用し、前記全流体摂取値に到達するよう海洋生物骨格誘導体系の固体材料内の前記流体の最大摂取を提供する工程をさらに含んで成る、請求項59に記載の方法。
- 前記特異的流体摂取容量値が、前記海洋生物骨格誘導体系の固体材料における重量変化の関数である、請求項59に記載の方法。
