Abstract
【解決手段】基材表面に分子量が10000〜150000の非架橋の温度応答性高分子が0.02〜0.3分子鎖/nm2の割合で固定化されている温度応答性細胞培養基材を作製し、温度応答性細胞培養基材を利用して、各組織から得られた細胞を効率良く培養でき、この培養方法を利用すれば、温度を変えるだけで損傷なく、効率良く細胞シートを剥離することができる細胞を培養する方法。

Claims (9)
- 基材表面に分子量が10000〜150000の非架橋の温度応答性高分子が0.02〜0.3分子鎖/nm2の割合で固定化されている、温度応答性細胞培養基材。
- 基材表面に0〜80℃の温度範囲内で水和力が変化する温度応答性高分子が固定化された請求項1記載の温度応答性細胞培養基材。
- 温度応答性高分子が、ポリ−N−置換アクリルアミド誘導体、ポリ−N−置換メタアクリルアミド誘導体、ポリアクリレート誘導体、ポリメタクリレート誘導体の単独、もしくはこれらの二つ以上の共重合体からなる、請求項1、2のいずれか一項記載の温度応答性細胞培養基材。
- 固定化した温度応答性高分子鎖の末端に官能基を有する、請求項1〜3のいずれか1項記載の温度応答性細胞培養基材。
- 官能基がジチオエステル基、水酸基、カルボキシル基、アミノ基のいずれか1種もしくは2種以上が混合されたものである、請求項4記載の温度応答性細胞培養基材。
- 温度応答性高分子の基材表面への固定化がリビングラジカル重合法によるものである、請求項1〜5のいずれか一項記載の温度応答性細胞培養基材の製造方法。
- 重合法が可逆的付加−開裂連鎖移動型ラジカル重合法である、請求項6記載の温度応答性細胞培養基材の製造方法。
- 基材表面に固定化されたアゾ系重合開始剤を利用することを特徴とする、請求項6、7のいずれか一項記載の温度応答性細胞培養基材の製造方法。
- 固定化した温度応答性高分子鎖の末端のジチオエステル基をチオール基に変換し、さらにそのチオール基を水酸基、カルボキシル基、アミノ基へ変換する、請求項6〜8のいずれか一項記載の温度応答性細胞培養基材の製造方法。
