Abstract
Description
本発明は、HIVファミリーのウイルスに対するワクチンの分野に関するものである。
ヒトHIV感染の約90%は、HIV−1ウイルスによって起こる。ヒト免疫不全ウイルス1型(HIV−1)は、ウイルス複製時に生じる突然変異の蓄積によって起こる、及び組換え事象によっても起こる、顕著な遺伝的変異性によって特徴づけられる。HIV処置の化学療法の長期にわたる失敗は、特に、HIV−1ウイルス株の高い変異原活性によって説明される。異なる経過の抗レトロウイルス療法後に、及び多剤療法(HAART)後であっても、患者から耐性ウイルス変異体が速やかに生じたことがこれまでに示された。これらの耐性ウイルスは、それらのタンパク質コンフォメーション及び構造に特異的変化を有する。通常、HIV−1が現行処置を回避する原因となるそのような突然変異は、処置条件下で選択を受ける結果として、代々のウイルス世代にわたり維持され、蓄積する。

で示されるペプチドを含む免疫原性化合物であって、式(VIb)で示されるペプチドが、CRM197タンパク質から成る担体タンパク質に共有結合的に連結された免疫原性化合物に関する。

− R1は、CRM197の一反応基から成り、但し、R1に結合したNH基は、CRM197のN末端に位置するα−アミノ基又は(ii)CRM197のリシン(K)アミノ酸残基由来の側鎖アミノ基から得られ、
− R2は、式(I)で示されるペプチドから成り、但し、R2に結合した硫黄(S)原子は、式(I)で示されるペプチドのN末端又はC末端に位置するシステイン残基のスルフヒドリル(SH)基から得られる。いくつかの態様では、スルフヒドリル部分は、非天然アミノ酸の部分、又は式(I)で示されるペプチドの末端に存在する任意の他の分子でありうる。
実施例1:免疫原性化合物の調製及びそれらの特性のいくつかの決定
A. 免疫原性化合物の調製
以下の免疫原性化合物又はコンジュゲートを合成した。それらは、架橋剤分子としてMBS又はSMPBのいずれかを使用してKLH及びCRM197から得た。使用されたペプチドは、そのアミノ末端又はカルボキシ末端のいずれかに追加的なシステイン残基を有する、配列番号2から成る3Sペプチドであった。
− CRM197−MBS−Nter(Cys)−3S
− CRM197−SMPB−Nter(Cys)−3S
− CRM197−SMPB−Cter(Cys)−3S
− KLH−MBS−Nter(Cys)−3S
驚くことに、得られた、C末端にCysを有する配列番号2の3Sペプチド、CRM197担体、及びリンカーとしてのSMPB又はMBSに対応する全ての免疫コンジュゲートは、たとえ長時間経った後であっても、加熱しても振盪しても、水にも0.9%NaCl溶液にも不溶性であった。これらの免疫コンジュゲートは、均一で再現性のある物質を得るために適切ではなかったので、これらの化合物を動物において試験しなかった。
A. 材料及び方法
A.1. 実施例2で試験された様々な免疫コンジュゲート化合物は、実施例1に開示されたように調製した。
− Adjuphos(登録商標)2%(リン酸アルミニウムゲル)はBrenntag(Frederikssund, Denmark)から購入した。Adjuphos(登録商標)は、最終濃度を注射1回あたりAl3+イオン300μgの投与に合わせたAl3+イオン最終濃度3mg/mlで使用した。
− Alhydrogel(登録商標)2%(水酸化アルミニウムゲル)は、Brenntag(Frederikssund, Denmark)から購入した。Alhydrogel(登録商標)は、最終濃度を注射1回あたりAl3+イオン300μgの投与に合わせたAl3+イオン最終濃度3mg/mlで使用した。
− 不完全フロイントアジュバント(IFA)は、SIGMA(Missouri, USA)から購入した。IFA50μlと水性免疫コンジュゲート溶液50μlとの混合物を1時間ボルテックス撹拌することによって、IFAを免疫コンジュゲート化合物と共に乳化した。
動物は、Charles River Laboratories(Lyon, France)によって提供された、実験0日目に8週齢のBALB/cJ雌であった。
表1に記載された各組成物を、抗原性ペプチド当量として表現されるとき用量40μgで皮下経路によってマウスに注射した。
ELISAアッセイは、抗3S16Nterペプチドとも呼ばれる配列番号2のペプチドを認識するIgG抗体の測定を行うように計画した。抗3S16Nter IgG抗体力価は、酵素結合免疫吸着検定法(ELISA)によって決定した。表1において報告された、0日目、14日目及び28日目に1回あたり3S16Nterペプチド当量20又は50μgの免疫コンジュゲートをワクチン接種されたラットの血清プールを使用して、異なる96ウェルプレート間の値を基準化した。
抗3S抗体のIgG力価は、材料及び報告の節に記載されたELISAアッセイによって測定した。
アルミニウム塩への抗原吸着は、アジュバント効果に重要であり、ワクチン抗原、特に塩の製剤化は、アルミニウムと抗原との間の潜在的相互作用の重要な要素である。水酸化アルミニウムの表面は、アルミニウムに配位したヒドロキシル基から構成される。リン酸アルミニウムの表面電荷は、ヒドロキシル基とリン酸基との両方から構成される。アルミニウムアジュバントによるタンパク質の吸着は複合過程であり、静電力、疎水力及び他の引力の寄与を伴う。
製剤1:7mMリン酸ナトリウム、135mM塩化ナトリウム、Al3+イオンが1mg/mLの水酸化アルミニウム(pH7)
製剤2:15mMリン酸ナトリウム、135mM塩化ナトリウム、Al3+イオンが1mg/mLの水酸化アルミニウム(pH7)
製剤3:150mM塩化ナトリウム、Al3+イオンが1mg/mLのリン酸アルミニウム(pH5.5)
本実施例は、マウス及びラットにおける3S原薬候補の免疫応答により、ヒトに注射することができる3S原薬の量を説明する。
最初のヒト試験で使用されるべき最大ヒト用量を決定するために、マウスBalB/CByJ(Charles River Laboratories, Lyon, France)及びラットCD(登録商標)IGS(Charles River Laboratories, Lyon, France)において3S原薬又はCRM197−3S16Nterを用いて2種の用量範囲判定研究を行った。被験製剤は、水酸化アルミニウム(1mg/mLのAl3+イオン、150mM塩化ナトリウム及び3.6mMリン酸ナトリウムに入れたものである。記載された実験では、3S原薬の被験ロット中のリン酸ナトリウム濃度が高すぎるので、3S原薬は、3.6mMリン酸ナトリウムで試験した。医薬品、すなわち「VAC−3S」は、3.6mMリン酸ナトリウムの代わりに1mMリン酸ナトリウムで製剤化されている。水酸化アルミニウムへの3S原薬の吸着は、1から3.6mMリン酸塩の間で同等である。そのうえ、免疫原性のブリッジング実験は、3.6mMリン酸ナトリウムで製剤化されたVAC−3Sと3S原薬との間で行う。臨床試験で、投与スケジュールは、1か月間隔で3回のワクチン接種である。マウス及びラットでも、3回のワクチン接種で同じスケジュールを行ったが、間隔は14日であった。3S原薬に対する生物学的応答として抗3S16Nter IgG力価を測定した。そのような抗体は、3SペプチドとヒトCD4+T細胞上のそのレセプターとの間の相互作用を阻害することが知られている。
マウス9匹6群に漸増する用量の3S医薬品をワクチン接種した:体積0.05mLの150mM塩化ナトリウム、3.6mMリン酸ナトリウム、及びアジュバントとしての1mg/mL水酸化アルミニウムに入れて製剤化された0.02、0.2、1、2、及び4μgの3S16Nter−ペプチド当量の3S原薬。マウス6匹の1群にアジュバントのみをワクチン接種した。
ラット6匹5群に漸増する用量の3S医薬品をワクチン接種した:ワクチン接種1回あたり体積0.5mLの150mM塩化ナトリウム、3.6mMリン酸ナトリウム、及びアジュバントとしての1mg/mL水酸化アルミニウムに入れて製剤化された0.02、0.2、2、20及び40μgの3S16Nter−ペプチド当量の原薬。ラット6匹の1群は陰性対照としてワクチン接種しなかった。
本発明者らが使用した理論的根拠は、マウスにおいて抗3S16Nter IgG抗体力価のプラトーを得るために必要なアジュバント加3S原薬の用量が、ヒトにおいて抗3S16Nter IgG抗体力価のプラトーを得るために必要な用量の10分の1に対応するというものである。これは、抗S16Nter IgGのプラトーが、2(0.2μgの10倍)から10μg(1μgの10倍)の間の3S16Nterペプチド当量でヒトにおいて達成されるであろうことを意味する。
実施例5の目的は、アジュバント加3S原薬をワクチン接種されたラットの抗血清が3Sペプチド、すなわち3S16Nterペプチドによって誘導された活性化ヒトCD4+T細胞表面でのNKp44Lの発現を阻害する能力を試験することであった。
細胞:
ヒトCD4+T細胞は、EFS(「Etablissement Francais du Sang」)に注文した白血球−血小板残留物パウチから磁気分離によって得た。
1. パウチを50mL Falconチューブ4本に分配する:4×15mL
2. RPMI1640+glutamax培地(ref 72400-054, GIBCO, Life Technologies, Carlsbad, USA)で50mLにする
3. 50mLチューブ8本中のFicoll(Ref 17-829E, Eurobio, Les Ulis, France))15mL上に希釈血液を分配する
4. 2800rpmで20分間、ブレーキをかけずに室温で遠心分離する
5. 白血球のリングを回収し、50mLチューブ4本に1本あたり最大25mLでそれらを分配する
6. チューブをRPMI1640培地で50mLにする
7. 2000rpmで7分間、室温で遠心分離する
8. 上清を捨てる
9. ペレットをプールし、RPMI1640培地50mLで洗浄し、1500rpmで5分間、室温で遠心分離する
10. トリパンブルーに入れて細胞を計数する
11. 4億個のPBMCを選別する
12. 選別用緩衝液(Mg及びCa不含PBS、0.5%BSA、2mM EDTA)15mL中で細胞を洗浄する。磁気ビーズの貪食を避けるために、その時点から細胞を4℃又は氷中で維持する)
13. 20×106個のPBMCあたり磁気ビーズ10μL=4億個のPBMCについて磁気ビーズ懸濁液200μLを添加する(MACS CD4 microbeads, Miltenyi Biotec, Paris, France)
14. 4℃で30分間インキュベーションする。
15. 選別用緩衝液1mL中に細胞を再懸濁する
16. quadroMACSマグネット(Miltenyi Biotec, Paris, France)に2本のLSカラム(Miltenyi Biotec, Paris, France)を装着する
17. 選別用緩衝液3mLで2本のLSカラムのそれぞれを平衡化する
18. 重力によって細胞懸濁液にカラムを通過させる(各カラムで0.5mL)
19. 選別用緩衝液5mLで各カラムを2回洗浄する
20. マグネットからカラムを取り外す
21. 選別用緩衝液5mLで、最初は重力によって、2回目はピストンを使用して、各カラムを2回溶出する
22. 各カラムから溶出した細胞を完全培地15mL(RPMI1640+glutamax、10%非働化SVF Gibco、可欠アミノ酸100×(ref GIBCO, Life Technologies, Carlsbad, USA)、抗生物質/抗真菌薬(ref 15240-112, GIBCO, Life Technologies, Carlsbad, USA)で洗浄する
23. アセティックブルー(acetic blue)に入れて細胞を計数する
1. 5千万個のCD4+T細胞を、37℃、5%CO2の加湿通気インキュベーター中に立てた75cm2フラスコ(Ref 353136, Falcon, lieu, pays)中の完全培地30mLに入れて培養した。
2. フィトヘマグルチニン(PHA)を最終濃度1μg/mLで添加する:培地1mLあたり1mg/mLを1μL分量。(PHAはThermo Fisher Scientific, Waltham, USA製)
3. PHA活性化の3日後に100UI/mL IL−2(Aldesleukine, lieu, pays)を添加する。
ラット(Rattus norvegicus)Crl/CD(SD)の血清を試験した:0日目、14日目及び28日目に、この動物に水酸化アルミニウムアジュバント加3S原薬を1回あたり2μgの3S16Nterペプチド当量でワクチン接種した。49日目にその動物の血清は抗3S16Nter抗体陽性であった。
陽性対照:Charles River Laboratories, Lyon, FranceからのウサギNew Zealand(Oryctolagus cuniculus)から49日目に採取した抗血清を1/50希釈で陽性対照として使用した。このウサギに0日目、14日目及び28日目に水酸化アルミニウムアジュバント加CRM197−(3S16Nter)免疫コンジュゲートをワクチン接種し、49日目にその動物の血清は抗3S16Nter IgG陽性であった。
対照は、3S16Nterペプチド不在下又は存在下で二つ組で試験した。ウェル及びチューブの識別番号を下表2に示す。
1. 被検血清を三つ組で試験する、
2. 2mg/mL 3S16Nterペプチド溶液の1/40希釈液20μL:IVV−B122 25μL+完全RPMI1640培地975μL。したがって、3S16Nterペプチドは最終濃度5μg/mLであった、
3. フラスコからのCD4+T細胞の細胞懸濁液180μL
血清の1/100希釈のために、1/4希釈の血清8μLを添加する。
血清の1/400希釈のために、1/16希釈の血清8μLを添加する。
血清の1/1600希釈のために、1/64希釈の血清8μLを添加する。
ラット血清は、3S16Nterペプチドの存在下で三つ組で試験した。ウェル及びチューブの識別番号を表3に示す。
ウェル1では細胞を染色しなかった;ウェル2では、バックグラウンドを判定するために細胞を抗IgM−PE抗体単独の存在下に置いた;ウェル3では、細胞を抗CD4−APC単独で染色した。
5. マイクロプレートを400gで5分間遠心分離する。
6. 上清を除去する
7. 10μL/ウェルのマウス抗NKp44L IgM7.1+30μL/ウェルのPBS、0.5%BSA(抗体溶液500μL+PBS、0.5%BSA 1500μL)を添加する
8. 4℃で1時間インキュベーションする
9. 150μL/ウェルのPBS、0.5%BSAを添加する
10. マイクロプレートを400gで5分間遠心分離する。
11. 上清を除去する
12. 二次抗体抗マウスIgM−PEをPBS、0.5%BSAで1/25希釈した液50μL/ウェルを添加する。
13. 4℃で30分間インキュベーションする。
14. 150μL/ウェルのPBS、0.5%BSAを添加する。
15. マイクロプレートを400gで5分間遠心分離する。
16. 抗ヒトCD4−APC抗体をPBS、0.5%BSAで1/25希釈した液50μLを添加する。
17. 細胞懸濁液をFACSチューブ中に移す。
18. 室温で15分間インキュベーションする。
19. 各チューブに1×PBS 2mLを添加する
20. 400gで5分間遠心分離する
21. 1×PBS 300μLを添加する
22. 細胞蛍光測定装置でチューブを取得する

データは分析日にソフトウェアGalliosで分析し、印刷する。
結果は、細胞のPEマーカーのX−平均蛍光を報告するものである。
− APC強度/前方側方強度ドットプロットでCD4+T細胞にゲート設定
− SSC強度/前方側方強度ドットプロットでリンパ球にゲート設定
− 血清なし、3S16Nterペプチドなしのウェルでは、活性化CD4+T細胞はNKp44Lを発現しなかった。
− 血清なし、3S16Nterペプチドの存在下のウェルでは、活性化CD4+T細胞はNKp44Lを平均レベル62で発現した。
− 1/50希釈のウサギ抗3S16Nter抗体陰性血清あり、3S16Nterペプチドなしのウェルでは、活性化CD4+T細胞はNKp44Lを発現しなかった。
− 1/50希釈のウサギ抗3S16Nter抗体陰性血清あり、3S16Nterペプチド存在下のウェルでは、活性化CD4+T細胞は平均レベル63のNKp44Lを発現しなかった。
− 1/50希釈のウサギ抗3S16Nter抗体陽性血清あり、3S16Nterペプチドあり又はなしのウェルでは、活性化CD4+T細胞はNKp44Lを発現しなかった。
− それらの表面にNKp44Lを自然には発現せず、
− 3S16Nterペプチドへの曝露に応答してそれらの表面にNKp44Lを発現する能力があり、
− この発現が無関係の抗血清により誘導も阻害もされず、
− NKp44Lの表面が抗3S16Nter IgG陽性血清によって完全に阻害された
ことを示した。
1/50希釈のラット抗3S16Nter抗体陰性血清あり、3S16Nterペプチド存在下のウェルでは、活性化CD4+T細胞は平均蛍光レベル53でNKp44Lを発現した、
− 1/50希釈のラット抗3S16Nter抗体陽性血清あり、3S16Nterペプチド存在下のウェルでは、活性化CD4+T細胞でのNKp44Lの表面発現は完全に阻害された。
− 1/100希釈のラット抗3S16Nter抗体陽性血清あり、3S16Nterペプチド存在下のウェルでは、活性化CD4+T細胞は、平均蛍光レベル14でNKp44Lを発現した。
− 1/400及び1/1600希釈の抗3S16Nter抗体陽性血清あり、3S16Nterペプチド存在下のウェルでは、活性化CD4+T細胞上のNKp44Lの表面発現は阻害されなかった
ことを示す。
ワクチンの調製:
VAC−3Sは、緩衝等張食塩水中に水酸化アルミニウムに吸着した3S原薬を含有する筋肉内注射用無菌懸濁液である。VAC−3Sの製造は、GMPを遵守して行った。
振盪後、ワクチンは、使用準備済みの均一な白色懸濁物である。ワクチンは、三角筋に筋肉内注射することができる。無菌針を備える無菌シリンジを注射に使用する。患者は、各0.5mLの投与3回を、4週間のワクチン接種間隔で受けるべきである。
A. 免疫原性化合物の調製
以下の免疫原性化合物又はコンジュゲートを合成した。それは、CRM197から架橋剤分子としてSMPBを使用して得られた(実施例1に示す)。使用されたペプチドは、架橋剤の化学的カップリングを可能にしてCRM197−SMPB−Nter(Cys)−m3Sをもたらすためにそのアミノ末端に追加的なシステイン残基を有する、配列番号6(NH2−PWNASASNKSLDDIW−COOH)から成る突然変異型3S(m3S)ペプチドであった。
N末端にCys残基、CRM197担体及びリンカーとしてのSMPBを含む、得られた、配列番号7のm3Sペプチドに対応する免疫コンジュゲートは、水又は0.9%NaCl溶液に自然に可溶性であることが見いだされた。そのような免疫コンジュゲートの免疫原性を下記実施例10でさらに研究した。
A. 材料及び方法
A.1. 実施例10で試験された免疫コンジュゲート化合物は、実施例9に開示されたように調製した。
動物は、Charles River Laboratories(Lyon, France)によって提供されたBALB/cByJ雌であり、実験0日目に8週齢であった。
実施例5記載のワクチン調製物は、抗原性ペプチド当量として表現されるとき用量2μgでマウスに筋肉内経路により注射した。
ELISAアッセイは、抗m3Sペプチド抗体とも呼ばれる配列番号6のペプチドを認識するIgG抗体の測定を行うために設計した。
抗3S抗体のIgG力価は、材料及び方法の節に記載されたELISAアッセイによって測定した。

- 請求項1から7のいずれか一項記載の免疫原性化合物を1種又は複数の免疫アジュバント物質と組み合わせて含む組成物。
- 抗原性ペプチド当量で表現されるとき、1投薬単位あたり0.01μg〜200μg、好ましくは1投薬単位あたり0.05μg及び50μg、最も好ましくは1投薬単位あたり0.1μg〜20μgの範囲の量の免疫原性化合物を含む使用準備済みのワクチン組成物を形成するように適応されている、請求項8記載の組成物。
- 免疫アジュバント物質が、水酸化アルミニウム(Al(OH)3)から成る、請求項8及び9のいずれか一項記載の組成物。
- Al3+イオン含量として表現されるとき、0.1mg/mL〜5mg/mL、好ましくは0.05mg/mL〜2mg/mLの範囲、最も好ましくは約1mg/mLの最終濃度の水酸化アルミニウムを含む使用準備済みのワクチン組成物を形成するように適応されている、請求項10記載の組成物。
