理論に縛られることを望まないが、タンパク質濃度および種々のロタウイルス構造タンパク質の比率は、RLPの集合効率に重要であり得る。それゆえに感染効率および感染時間は、植物中のRLPのタンパク質濃度およびアッセンブリー効率全体に重要であり得る。
ベンサミアナタバコ植物葉でのロタウイルスタンパク質発現およびVLP産生
次の分析を、G9 P[6]ロタウイルス株からのロタウイルスカプシドタンパク質を利用し、ロタウイルス様粒子がベンサミアナタバコ葉細胞の種々の区画で形成されるか否かを評価した。タバコ植物葉でのVP2およびVP6ならびにVP2、VP6、VP7およびVP4の種々の組み合わせの同時発現を試験した。
プラスミド構築
VP2、VP4、VP6およびVP7の植物コドン最適化ロタウイルスcDNAは、Geneart, Germanyから供与された。プラスミドDNAを製造社の指示に従い、DH5−α化学的コンピテント大腸菌細胞(E. cloniTM, Lucigen)に形質転換した。Rainer Fischer(Fraunhofer Institute for Molecular Biology and Applied Ecology, IME, Germany)から供与された新規バイナリーアグロバクテリウムベクターpTRAc(細胞質)、pTRAkc−rbcs1−cTP(葉緑体ターゲティング)およびpTRAkc−ERH(小胞体ターゲティング)を本研究に使用した。さらなるベクター、pTRAkc−A(アポプラスト)は、多重クローニング部位におけるNcoIおよびXhoI部位での制限酵素(RE)消化によるpTRAkc−ERHの修飾に由来した(図3)。これはヒスチジンタグおよびタンパク質をERに保留するKDEL配列を除去する。タンパク質はその代わりにアポプラストにターゲティングされる。
アグロバクテリウム・ツメファシエンスGV3101株はRainer Fischer教授(Fraunhofer Institute for Molecular Biology and Applied Ecology IME, Aachen, Germany)から供与され、先に記載されたとおりエレクトロコンピテントとした(Shen and Forde, 1989)。300ngの単離ロタウイルスpTRA−VP構築物を100μlのエレクトロコンピテントGV3101細胞と0.1cmエレクトロギャップキュベット(BioRadTM)中で混合し、GenePulser(BioRad)中、次の設定で電気穿孔した:1.8kV、25μFおよび200Ω。インキュベーションを、1時間、27℃で900μlのLBで行い、50μg/mlカルベニシリン(carb)、30μg/ml カナマイシン(kan)および50μg/ml リファンピシン(rif)を含むLAプレートに播種した。プレートを27℃で3日間インキュベートした。陽性形質転換体を確認するために、プラスミドDNAを組み換えアグロバクテリウムコロニーから単離し、大腸菌コンピテントDH5−α細胞に戻し形質転換した。次いでこれらを100μg/ml アンピシリン(amp)LA上で選択した。コロニーPCRおよびcDNA上の制限酵素消化を行い、形質転換体の成功を確認した。関連する組み換えアグロバクテリウムのグリセロール原液を製造し、−70℃で貯蔵した。
本研究に使用したアグロバクテリウム・ツメファシエンスLBA 4404(pBIN−NSs)はMarcel Prins(Laboratory of Virology, Wageningen University, Binnenhaven, Netherlands)から得た。これは、トマト黄化壊疽ウイルス(TSWV)で見られるNSサイレンシングサプレッサーを含んだ。グリセロール原液からの組み換えアグロバクテリウム(pTRA−VP)を27℃で一夜、50μg/ml carb、30μg/ml kanおよび50μg/ml rif添加LB中で増殖させた。次いで組み換えアグロバクテリウムおよびLBA4404(pBIN−NSs)を各々誘導培地(LB、10mM 2−(N−モルホリノ)エタンスルホン酸MES、2mM MgSO4、20μM アセトシリンゴン、50μg/ml carb、30μg/ml kanおよび50μg/ml rifおよびpH5.6)に接種した。
葉全体または構築物あたり2個の葉片を収穫し、液体窒素中で粉砕した。粉砕した葉物質を、Completeプロテアーゼ阻害剤(無EDTA;Roche)含有無菌PBSに再懸濁した。ついでこれを5分、13000rpmで遠心分離し、ペレット(植物葉物質)を廃棄した。次いで100μlの各構築物を5×SDS−PAGE充填緩衝液と混合し、2分、95℃で沸騰させ、さらなるSDS−PAGEゲルおよびウェスタンブロットの準備が整った。サンプルの残りをさらに使用するまで−20℃で貯蔵した。図4はロタウイルスcDNAについてのクローニングおよび浸潤手順の概要を示す。
アポプラスト構築物であるpTRAkc−Aでさらなる抽出法を行った。アポプラストは植物細胞の原形質膜と細胞壁の間の自由に拡散する間隙である(図5a)。細胞質でのタンパク質発現は輸送配列を有し、これによりアポプラストに標的化され、それゆえにこれらはここに蓄積される。下記抽出法において、各抽出日からの葉全体をCompleteプロテアーゼ阻害剤含有無菌PBSで吸引浸潤または注入浸潤した。吸引浸潤について、個々の植物葉をPBSに懸濁し、吸引タンク中100mbarで10分の吸引下に置いた。次いで葉を巻き、底に孔が開いたスピンカラム(Qiagenスピンカラムに類似)に穏やかに入れた(図5b2)。孔は固形葉物質を通すことなく葉からの流体を容易に通す。スピンカラムを2mlエッペンドルフチューブに入れ、遠心分離を4000rpmで15分行った(図5b3)。濾液を回収し、SDS−PAGEゲルおよびウェスタンブロット分析のためのタンパク質ローディング・ダイを100μlの各濾液サンプルに添加した。
ウェスタンブロットおよびクーマシーブルー染色SDS−PAGEゲルを先に記載のとおり使用した。マウス抗ロタウイルスVP6抗体(US Biologicals)(1:5000)、抗マウスヒスチジンタグ抗体(Sigma(登録商標))(1:2000)、ニワトリ抗VP2およびニワトリ抗VP4血清(1:2000)を使用して、それぞれのタンパク質の各々をウェスタンブロットでプローブした。クーマシーブルー染色SDS−PAGEゲルを使用して、Syngene Gel Documentation Systemを使用したバンドの密度走査によりタンパク質を定量した。
発現タンパク質がRLPに集合しているか否かを決定するために、免疫捕捉(immune-trapped)粒子の透過型電子顕微鏡検査(TEM)を細胞質発現VP6、VP2/6およびVP2/6/4の発現3日目に、全てサイレンシングサプレッサーNss存在下で行った。グロー放電炭素/銅グリッドを20μlのマウス抗ロタウイルスVP6抗体(1:5000)上に5分置き、無菌蒸留水で3回洗浄した。次いでグリッドを10μlのタンパク質抽出物上におき、2分放置し、再び無菌蒸留水で3回洗浄した。最後に、グリッドを20μlの2%酢酸ウラニル上、1分浮遊させて、TEM(Zeiss 912 OMEGA Energy Filter Transmission Electron Microscope, University of Cape Town)下で視察した。
植物タンパク質抽出物を、最初にミラクロスで濾過して、固形植物体を除去した。10〜60%スクロースのスクロース勾配を40mlチューブに各々無菌PBS(pH7.4)に溶解した5mlのスクロースの6層を作ることにより設定した。5〜10ml体積の浄化したタンパク質サンプルを次いで各勾配カラムの頂上に乗せた。
総可溶性タンパク質(TSP)をブラッドフォードアッセイにより決定した。これは、細胞質同時発現VP2/6中の蓄積タンパク質のレベルを比較するために行った。タンパク質IgG(1.43mg/ml原液)を標準として希釈系列で使用した。5μlの標準およびサンプルを各々清潔な乾燥したマイクロタイタープレートに添加した。総可溶性タンパク質試薬AおよびBを製造社の指示に従い添加した(Bio-Rad Dc Protein Assay)。全ての実験を3組行った。吸光度測定値を750nmでマイクロプレートリーダー(Bio-tek PowerWave XS)を使用して記録した。
植物葉細胞区画でのVP6の発現
VP6は、サイレンシングサプレッサーを伴いおよび伴わずに、全細胞区画で発現し、標的化された(図6;(線はVP6を印す、〜42kDa))。細胞質において、タンパク質はタイムトライアルの1日目から発現し、1週間にわたる試験の間細胞質でタンパク質蓄積は増え続けた(図6a)。ERにおいて、タンパク質蓄積は3日目のみに明らかに見られた(図6b)。タンパク質は他のタンパク質より高いバンドサイズ(約11kDa多い)で動いた。これは、おそらくタンパク質のC末端に付加した6−ヒスチジンタグならびに開裂部位(pProExベクター配列と呼ぶ)の結果である。
4種のロタウイルスVPをさらなるベクターであるpTRAc−HTにクローン化した。このベクターは、細胞質を標的とするタンパク質に対する6−ヒスチジンタグを含み、興味あるタンパク質に対する抗体が利用可能ではないとき、抗ヒスチジンタグ抗体の使用により検出を容易にする。我々の場合、VP6のみ市販抗体があり、それゆえに我々は、血清待ちの間全タンパク質の初期検出のためにこの手順を試した。細胞質もVP6発現に十分働き、我々が他のタンパク質を試す動機付けとなった。
VP2およびVP4をベンサミアナタバコ植物葉に浸潤させ、ER、葉緑体、細胞質およびアポプラストを標的化した。我々は、大腸菌で陽性クローンを得ることができなかったため、アポプラストを標的としたVP2のベクターを発現することが不可能であった。しかしながら、タンパク質発現および標的化は他の3区画で成功した(図8a)。ニワトリ抗VP2および抗VP4血清(1:2000)を抽出物のウェスタンブロット分析に使用した。VP2およびVP4バンドは、それぞれ図8aおよび8bに見られるとおり、100kDa印(矢印で示すタンパク質バンド)の直ぐ下に見られた。発現はVP2について細胞質およびERで最良であり、VP4については細胞質およびアポプラストで最良であるように見えた。サイレンシングサプレッサーはタンパク質発現に顕著な効果を有しなかった。ウェスタンブロットに見られるとおり、VP2 ER構築物で発現をわずかに上昇させたのみであり、残りではほとんどなかった。VP4構築物は全てサイレンシングサプレッサー存在下で発現された。
細胞質はロタウイルスカプシドタンパク質発現に最良であり、最高抽出効率を示すように見えた。全てのさらなる発現実験は、それゆえに細胞質を標的化したタンパク質で行った。
VP2/6およびVP2/6/4を、10〜60%スクロースの範囲のスクロース勾配で精製した(図11a)。2mlフラクションを各チューブの底から回収し、マウス抗VP6抗体および/またはニワトリ抗VP2およびVP4血清でプローブして、どのフラクションがタンパク質を含むかを決定した。VP2/6タンパク質は、ブロット上でVP6タンパク質が陽性であるとして、フラクション16および17で見られた(図11b)。ニワトリ抗VP2およびVP4血清を用いたVP2/6/4ブロット分析では、全フラクションを通して陽性結果が示された。これは、おそらくニワトリ血清による高レベルの背景タンパク質検出の結果である。しかしながら、ドットの強度は図11cで見られるとおりフラクション17および18で最高であり、これはこれらのフラクション中の興味あるタンパク質が高濃度であるためであると考えられる。
同時発現VP2/6のウェスタンブロットおよびSDS−PAGEを行い、フラクション13〜20におけるVP2およびVP6タンパク質の存在を確認した。マウス抗VP6抗体でプローブしたVP6タンパク質のウェスタンブロット分析は丁度フラクション16〜20で陽性であった(図12a、一番下の矢印および図12c)。ニワトリ抗VP2血清でプローブしたVP2タンパク質はフラクション17〜20で見られた(図12a、一番上の矢印)。VP2はこれまでの同時発現試験でVP6より発現が少ないことが示されており、これはまたここでVP6と比較してVP2タンパク質バンドの強度が低い図12aにおいても示された。
総可溶性タンパク質(TSP)を、VP2/6タンパク質の相対量を決定するために同時発現VP2/6フラクションで決定した(図13)。タンパク質濃度は、IgG標準を使用して、フラクション17および18でそれぞれ0.538mg/mlおよび1.012mg/mlであると計算された(図13a)。これらのフラクションのVP2/6に対応するタンパク質バンドをSyngene Gel Documentation System上の密度走査で計算し、それぞれ約0.108mg/mlおよび0.202mg/mlであることが判明した。
精製VP2/6フラクションを一緒にプールし、スクロースを除去するために高塩PBSで透析して、透過電子顕微鏡で観察した。TEMを行い、純度を決定し、RLPが精製手順後も無傷のままであることを確認した。図15に見られるとおり、主として宿主細胞の生成物から成るほとんどの背景物質(図10b、cおよびd)を除去し、RLPを残した。ほとんどのRLPは無傷のままであったが、幾分かはおそらくEMグリッド上の条件による変形の結果、形を失っているように見えた。
この予備的分析は、宿主発現系例としてのベンサミアナタバコ葉中のロタウイルス構造タンパク質VP2(配列番号1)、VP4(配列番号2)、VP6(配列番号3)およびVP7(配列番号4)の発現に焦点を当てた。ここで選択したロタウイルス株は、南アフリカおよび他のサハラ以南地域で優勢に伝播しているG9 P[6]株である。この株を標的とするRLPワクチンは、アフリカのサハラ以南での疾患の負荷を軽減するのに役立つはずである。

遺伝子構築物の集合およびアグロバクテリウム形質転換
プラスミド1710、1713、1730および1734を含む全プラスミドを、エレクトロポレーション(Mattanovich et al., 1989, Nucleic Acid Res. 17:6747)によりアグロバクテリウム・ツメファシエンス(AGL1;ATCC, Manassas, VA 20108, USA)を形質転換するのに使用し、あるいは、CaCl2調製コンピテント細胞を使用したヒートショック(XU et al., 2008, Plant Methods 4)に使用し得る。作製したアグロバクテリウム・ツメファシエンス株中のプラスミドの完全性を制限酵素マッピングにより確認した。あるバイナリープラスミドで形質転換したアグロバクテリウム・ツメファシエンス株をAGL1/“プラスミド番号”と命名する。例えば、構築物番号1710で形質転換したアグロバクテリウム・ツメファシエンス株を“AGL1/1710”と名づける。
ベンサミアナタバコ植物を、市販ピートモス培養土を満たしたフラット中、種子から生育させた。植物を、16/8光周期および25℃日中/20℃夜間の温度管理下、温室で生育させた。播種3週間後、個々の小植物を選択し、鉢に移植し、同じ環境条件下、さらに3週間温室で生育させた。
タンパク質を、ブレンダーで3体積の抽出緩衝液(TNC:10mM Tris pH7.4、140mM NaCl、10mM CaCl2)を用いる機械的抽出により凍結バイオマスから抽出した。スラリーを大孔ナイロンフィルターで濾過して、大きな残骸を除き、5000gで5分、4℃で遠心分離した。上清を回収し、再び5000gで30分(4℃)で遠心分離して、さらに残骸を除去した。上清をデプスフィルタで濾過し、限外濾過し、濾液を75000gで20分(4℃)、遠心分離してロタウイルス様粒子を濃縮した。粒子含有ペレットを1/12体積のTNCに再懸濁し、不溶物を5000gで5分、遠心分離して除去した。上清をミラクロスで濾過し、イオジキサノール密度勾配に充填した。
タンパク質濃度をBCAタンパク質アッセイ(Pierce Biochemicals, Rockport IL)により決定した。タンパク質をSDS−PAGEで還元または非還元条件下に分離し、クーマシーブルーで染色した。染色されたゲルを走査し、濃度測定分析をImageJ Software(NIH)で行った。
U底96ウェルマイクロタイタープレートを、150mM NaCl含有10mM PBS pH7.4(リン酸緩衝化食塩水)で1:100000希釈したマウスモノクローナル抗VP4(Koki Taniguchi教授より恵与)で16〜18時間、4℃で被覆した。インキュベート後、プレートを1M NaCl含有0.1%Tween−20含有10mM PBS pH7.4で3回洗浄し、150mM NaCl含有0.1%Tween−20含有10mM PBS pH7.4中5%BSAで1時間、37℃でブロッキングした。ブロッキング工程後、プレートを10mM PBS pH7.4、1M NaCl含有0.1%Tween−20で3回洗浄した。サンプルを添加し、プレートを1時間、37℃でインキュベートした。次いでプレートを1M NaCl、1mM CaCl2、0.5mM MgCl2含有0.1%Tween−20含有10mM PBS pH7.4で3回洗浄した。残りの全ての洗浄工程について、洗浄緩衝液は同じままであり、3回目の洗浄の間、プレートを洗浄溶液の完全除去前に10分、室温でインキュベートした。150mM NaCl、1mM CaCl2、0.5mM MgCl2含有0.1%Tween−20含有10mM PBS pH7.4中3%BSAで1:10000希釈したロタウイルスに対して惹起したウサギポリクローナル抗体(Koki Taniguchi教授より恵与)を添加し、プレートを1時間、37℃でインキュベートした。次いでプレートを3回洗浄し、150mM NaCl、1mM CaCl2、0.5mM MgCl2含有0.1%Tween−20含有10mM PBS pH7.4中3%BSAで1:5000希釈した西洋ワサビペルオキシダーゼ結合ヤギ抗ウサギ抗体(111-035-144, Jackson Immunoresearch, West Grove, PA)を添加し、プレートを1時間、37℃でインキュベートした。プレートを3回洗浄した。最終洗浄後、プレートをSureBlue TMBペルオキシダーゼ基質(KPL, Gaithersburg, MD)と20分、室温でインキュベートした。1N HCl添加により反応を停止させ、A450値をMultiskan Ascentプレートリーダー(Thermo Scientific, Waltham, MA)で読んだ。
VP2およびVP6を含むロタウイルス様粒子をベンサミアナタバコ中の一過性発現により産生した。植物を、1:1比のAGL1/1710とAGL1/1713の混合物を含むアグロバクテリアの接種によりアグロインフィルトレーションし、収穫前7日間インキュベートした。ロタウイルス様粒子を、材料および方法の章に記載した方法を使用してバイオマスから精製した。イオジキサノール密度勾配上での浄化した抽出物の遠心分離後、チューブの底からの最初の10フラクションをクーマシー染色SDS−PAGEにより分析した。図45Aに示すとおり、ロタウイルス抗原(VP2およびVP6)は、ロタウイルス様粒子が見られることが予測される濃度であるイオジキサノールの濃度が約35%である、密度勾配のフラクション2および3で主に見られた。これらのフラクションへの植物タンパク質の極少量の混入が見られた。抗ロタウイルス過免疫ウサギ血清およびポリクローナルウサギ抗VP2抗体を用いたフラクションのウェスタンブロット分析は、密度勾配フラクション中のVP2およびVP6の同一性を確認した(図45Bおよび45C)。フラクション2および3を貯留し、イオジキサノールを高速遠心分離により除去し、再懸濁し、精製粒子を低温電子顕微鏡分析に出して(NanoImaging Services Inc., La Jolla, CA)、ロタウイルス粒子を模倣する粒子へのVP2およびVP6の集合を確認した。図49(左パネル)に示すとおり、VP2/VP6粒子の極低温EM画像は、ロタウイルス様粒子への抗原の正しい集合を確認した。

b) 植物、植物の一部または植物細胞を、第一、第二および第三核酸の一過性発現を可能にする条件下でインキュベートして、RLPを産生する
ことを含む、方法。 - 植物内で活性であり、第四ロタウイルス構造タンパク質をコードする第四ヌクレオチド配列に作動可能に連結する第四調節領域を含む第四核酸を工程a)において植物、植物の一部または植物細胞に導入し、工程b)において植物、植物の一部または植物細胞をインキュベートしたとき発現させる、請求項1に記載の方法。
- 第一ロタウイルス構造タンパク質がVP2であり、第二ロタウイルス構造タンパク質がVP6であり、第三ロタウイルス構造タンパク質がVP4である、請求項1に記載の方法。

- 1個以上のロタウイルス構造タンパク質がVP2、VP4、VP6およびVP7から成る群から選択される、請求項30〜32のいずれかに記載の方法。
- 付加的ヌクレオチド配列を植物、植物の一部または植物細胞の中で発現させ、該付加的ヌクレオチド配列はサイレンシングのサプレッサーをコードし、該付加的ヌクレオチド配列は植物で活性な調節領域と作動可能に連結する、請求項30〜33のいずれかに記載の方法。
- 1個以上のロタウイルス構造タンパク質がロタウイルス株G9 P[6]由来である、請求項30〜34のいずれかに記載の方法。
- ヌクレオチド配列がさらに区画ターゲティング配列に作動可能に連結する、請求項30〜35のいずれかに記載の方法。
- 区画ターゲティング配列が1個以上のロタウイルス構造タンパク質を植物細胞の小胞体(ER)、葉緑体、色素体またはアポプラストに指向させる、請求項28、29および36のいずれかに記載の方法。
- 区画ターゲティング配列がアポプラストシグナルペプチドまたは色素体シグナルペプチドをコードする、請求項37に記載の方法。
- c) 植物、植物の一部または植物細胞を収穫し、
d) RLPを植物、植物の一部または植物細胞から精製する(ここで、RLPのサイズが70〜100nm範囲である)
工程をさらに含む、請求項30〜38のいずれかに記載の方法。 - 植物、植物の一部または植物細胞中でロタウイルス様粒子(RLP)を製造する方法であって:
a) 植物内で活性であり、1個以上のロタウイルス構造タンパク質をコードするヌクレオチド配列に作動可能に連結した調節領域を含む核酸を含む植物、植物の一部または植物細胞を提供し;
b) 植物、植物の一部または植物細胞を、該核酸の一過性発現を可能にする条件下でインキュベートして、RLPを産生する
ことを含む、方法。 - 植物、植物の一部または植物細胞中でロタウイルス様粒子(RLP)を製造する方法であって:
a) 植物内で活性であり、第一ロタウイルス構造タンパク質をコードする第一ヌクレオチド配列に作動可能に連結する第一調節領域を含む第一核酸、植物内で活性であり、第二ロタウイルス構造タンパク質をコードする第二ヌクレオチド配列に作動可能に連結する第二調節領域を含む第二核酸および植物内で活性であり、第三ロタウイルス構造タンパク質をコードする第三ヌクレオチド配列に作動可能に連結する第三調節領域を含む第三核酸を植物、植物の一部または植物細胞を提供し;そして
b) 植物、植物の一部または植物細胞を、第一、第二および第三核酸の一過性発現を可能にする条件下でインキュベートして、RLPを産生する
ことを含む、方法。 - 植物、植物の一部または植物細胞が工程a)において植物内で活性であり、第四ロタウイルス構造タンパク質をコードする第四ヌクレオチド配列に作動可能に連結する第四調節領域を含む第四核酸を伴い提供され、第四ロタウイルス構造タンパク質は工程b)において植物、植物の一部または植物細胞をインキュベートしたときに発現される、請求項41に記載の方法。
- RLPが少なくともVP4ロタウイルス構造タンパク質を含む、請求項30〜42のいずれかに記載の方法により製造されたRLP。
- 対象に免疫応答を誘発するための有効量の請求項14、15、16および43のいずれかに記載のRLPおよび薬学的に許容される担体を含む、組成物。
- 請求項44の組成物を投与することを含む、対象におけるロタウイルス感染に対する免疫を誘発する方法。
- 組成物を対象に経口、皮内、鼻腔内、筋肉内、腹腔内、静脈内または皮下により投与する、請求項45に記載の方法。
- 請求項1〜13および17〜42のいずれかに記載の方法により製造されたRLPを含む、植物体。
- 植物、植物の一部または植物細胞でロタウイルス構造タンパク質を製造する方法であって、
a) 植物内で活性であり、1個以上のロタウイルス構造タンパク質をコードするヌクレオチド配列に作動可能に連結する調節領域を含む核酸を植物、植物の一部または植物細胞に導入し;
b) 植物、植物の一部または植物細胞を該核酸の一過性発現を可能にする条件下でインキュベートして、1個以上のロタウイルス構造タンパク質を産生する
ことを含む、方法。 - ヌクレオチド配列がさらに区画ターゲティング配列に作動可能に連結する、請求項48に記載の方法。
