A.結合特異性および機能特性
本発明は、マウス15H3抗体ならびにキメラ、ヒト化、およびヒト15H3抗体を提供する。
ヒト化抗体は、非ヒト「ドナー」抗体由来CDRがヒト「アクセプタ」抗体配列に移植されている遺伝子工学によって作られた抗体である(例えば、Queen、US5,530,101および5,585,089;Winter、US5,225,539;Carter、US6,407,213;Adair、US5,859,205;ならびにFoote、US6,881,557を参照のこと)。アクセプタ抗体配列は、例えば、ヒト抗体配列、そのような配列の複合物、ヒト抗体配列の共通配列、または生殖細胞系領域配列であり得る。重鎖に好ましいアクセプタ配列は、生殖細胞系VHエクソンVH1−46および、Jエクソン(JH)には、エクソンJH−4である。軽鎖の場合、好ましいアクセプタ配列は、エクソンVK2−30(文献においてKV2−30とも記載される)、およびJエクソンにはJκ−2である。重鎖に好ましい代わりのアクセプタ配列には、Jエクソン(JH)、JH−1、JH−2、JH−3、JH−4、またはJH−5を伴う生殖細胞系VHエクソンVH1−8またはVH1−3がある。軽鎖に好ましい代わりのアクセプタ配列には、JエクソンJκ−1、Jκ−2、Jκ−3、Jκ−4、またはJκ−5を伴うエクソンVK2−29(文献においてKV2−30とも記載される)がある。したがって、ヒト化抗体は、完全にまたは実質的に非ヒトドナー抗体および可変領域フレームワーク配列に由来するいくつかのまたは全てのCDR、ならびに存在する場合には、完全にまたは実質的にヒト抗体配列に由来する定常領域を有する抗体である。同様に、ヒト化重鎖は、完全にまたは実質的にドナー抗体重鎖、および重鎖可変領域フレームワーク配列に由来する少なくとも1つ、2つ、通常は3つ全てのCDR、ならびに存在する場合には、実質的にヒト重鎖可変領域フレームワークおよび定常領域配列に由来する重鎖定常領域を有する。同様に、ヒト化軽鎖は、完全にまたは実質的にドナー抗体軽鎖、および軽鎖可変領域フレームワーク配列に由来する少なくとも1つ、2つ、通常は3つ全てのCDR、ならびに存在する場合には、実質的にヒト軽鎖可変領域フレームワークおよび定常領域配列に由来する軽鎖定常領域を有する。いくつかの態様では、ヒト化抗体は、ヒト化重鎖とヒト化軽鎖を含む。それぞれのCDRの間で対応する残基の少なくとも60%、85%、90%、95%または100%(Kabatで定義される)が同一である場合、ヒト化またはヒト抗体のCDRは、非ヒト抗体の対応するCDRに実質的に由来するまたは実質的に同一である。いくつかの実施形態では、各CDRにある保存的アミノ酸置換が3個以下である場合、ヒト化抗体またはヒト抗体のCDRは、非ヒト抗体の対応するCDRに実質的に由来するまたは実質的に同一である。Kabatで定義される対応する残基の少なくとも70%、80%、85%、90%、95%または100%が同一である場合、抗体鎖の可変領域フレームワーク配列または抗体鎖の定常領域は、それぞれヒト可変領域フレームワーク配列またはヒト定常領域に実質的に由来する。本発明のいくつかのヒト化抗体において、抗体の重鎖可変フレームワーク領域に少なくとも1つのマウス15H3復帰変異がある。
(1)非共有結合的に抗原に直接結合する、
(2)CDR領域に隣接している、
(3)さもなければCDR領域と相互作用する(例えば、CDR領域から約6Å以内である)、または
(4)重鎖と軽鎖との相互作用を媒介する
ことが合理的に期待されれば、ヒトフレームワークアミノ酸をマウス抗体由来の同等のフレームワークアミノ酸で置換することができる。
本明細書に記載される抗体の重鎖および軽鎖可変領域は、ヒト定常領域の少なくとも一部に連結できる。定常領域の選択は、一部において、抗体依存性細胞媒介細胞毒性、抗体依存性細胞貪食作用および/または補体依存性細胞傷害が望ましいかどうかによって決まる。例えば、ヒト同位体IgG1とIgG3は強力な補体依存性細胞傷害を、ヒトアイソタイプIgG2は弱い補体依存性細胞傷害を有し、ヒトIgG4は補体依存性細胞傷害がない。ヒトIgG1およびIgG3は、ヒトIgG2およびIgG4より強力な細胞媒介エフェクター機能も誘導する。軽鎖定常領域は、ラムダまたはカッパであり得る。抗体は、2本の軽鎖および2本の重鎖を含有する四量体として、別々の重鎖、軽鎖として、Fab、Fab’、F(ab’)2およびFvとして、または重鎖と軽鎖可変ドメインがスペーサによって連結されている一本鎖抗体として発現させることができる。
キメラまたはヒト化抗体は、通常は組換え発現によって生成される。通常、組換えポリヌクレオチド構築物は、天然に付随のまたは異種のプロモータ領域を含めた、抗体鎖のコード配列に作動可能に連結された発現制御配列を含む。好ましくは、発現制御配列は、真核生物宿主細胞を形質転換またはそれにトランスフェクトできるベクター内にある真核生物プロモータ系である。ベクターを適当な宿主に一旦組み込んだら、宿主は、ヌクレオチド配列の高レベル発現、ならびに交差反応抗体の採集および精製に適切な条件で維持される。
本発明は、上記の重鎖および軽鎖のいずれかをコードする核酸をさらに提供する。通常、核酸は、重鎖および軽鎖に融合されたシグナルペプチドもコードする。核酸のコード配列は、例えばプロモータ、エンハンサ、リボゾーム結合部位、転写終了シグナルなどの調節配列と作動可能に連結されて、コード配列の発現を確実にすることができる。重鎖および軽鎖をコードする核酸は、単離された形態で存在することができ、または1つもしくは複数のベクターにクローニングされることができる。核酸は、例えば、固体の状態の合成または重なり合うオリゴヌクレオチドのPCRによって合成することができる。重鎖および軽鎖をコードする核酸は1つの連続する核酸として、例えば、発現ベクター内に接合することができ、または別個である、例えば、各々の発現ベクターにクローニングすることができる。典型的な核酸は、配列番号37(マウス重鎖)、配列番号38(マウス軽鎖)、配列番号39(HA)、配列番号40(HB)、配列番号41(HL)、配列番号42(HT)、配列番号43(HN)、配列番号44(HU)、配列番号45(LA)、配列番号46(LC)、配列番号47(LF)、配列番号48(LG)、配列番号49(マウス重鎖シグナルペプチド)、配列番号50(ヒト重鎖シグナルペプチド)、配列番号51(マウス軽鎖シグナルペプチド)、および配列番号52(ヒト軽鎖シグナルペプチド)に示される。核酸は、単離された形態で提供され得る。単離された核酸分子は、その自然環境の成分から同定、分離および/もしくは回収されたものまたは天然に存在しないものである。
抗αvβ6抗体を、細胞障害性部分(薬学的に適合するその塩を含める)に結合体化して、抗体薬物結合体(ADC)を形成することができる。抗体に結合体化するのに特に適切な部分は、細胞毒性薬剤(例えば、化学療法剤)、プロドラッグ変換酵素、放射性同位体もしくは化合物、または毒素である(これらの部分は併せて治療剤と称される)。例えば、抗αvβ6抗体は、化学療法剤または毒素(例えば、細胞増殖抑止剤または細胞破壊剤、例えばアブリン、リシンA、シュードモナス外毒素、またはジフテリア毒素)などの細胞毒性薬剤に結合体化することができる。細胞毒性薬剤の有用な種類の例には、例えば、DNA副溝結合剤、DNAアルキル化剤、およびチューブリン阻害剤がある。典型的な細胞毒性薬剤には、例えば、オーリスタチン、カンプトセシン、デュオカルマイシン、エトポシド、マイタンシノイド(例えば、DM1、DM2、DM3、DM4)、タキサン、ベンゾジアゼピン含有化合物(例えば、ピロロ[1,4]ベンゾジアゼピン、インドリノベンゾジアゼピン、およびオキサゾリジノベンゾジアゼピン)を含めたベンゾジアゼピンならびにビンカアルカロイドがある。タンパク質、特に抗体に治療剤を結合体化する技術は周知である。(例えば、Alleyら、Current Opinion in Chemical Biology 2010年14巻:1〜9頁;Senter、Cancer J.、2008年、14巻(3号):154〜169頁を参照のこと)。
マウス15H3抗体および15H3抗体のキメラ形態、ヒト化形態、またはヒト形態と同様に、αvβ6の細胞外ドメインに結合する他の抗体は、本発明のいくつかの方法、特に膀胱がんの処置法に使用することができる。好ましくは、そのような実施形態では、抗体は細胞毒性薬剤に結合体化されている。抗αvβ6抗体の一群は公知であり、例えば、米国特許出願公開第20100330103号および第20110294982号ならびに米国特許第7,465,449号、第7,943,742号および第6,692,741号を参照のこと、その各々は、全ての目的のためにその全体が参照により本明細書に組み込まれる。
本発明の抗体を、単独でまたは抗αvβ6抗体薬物結合体として使用して、がんを処置することができる。そのようながんのいくつかは、タンパク質(例えば、実証された抗体の1つを使用するイムノアッセイによる)またはmRNAレベルのいずれかで測定される検出可能なレベルのαvβ6を示す。そのようながんのいくつかは、同じ型の、好ましくは同じ患者の非がん性組織と比較して、αvβ6レベルの上昇を示す。より高いまたはより低いレベルを処置することもできるが、処置可能ながん細胞におけるαvβ6の典型的なレベルは、細胞当たり1000〜200,000個のαvβ6分子である。一般に、固形腫瘍の処置には、より高いコピー数、例えば、細胞当たり少なくとも約15、000個のαvβ6分子が好ましい。任意選択で、処置を実施する前にがんのαvβ6レベルが測定される。
抗αvβ6抗体を使用して、臨床診断もしくは処置に関連してまたは研究においてαvβ6を検出することができる。がんにおけるαvβ6の発現は、がんが本発明の抗体を用いて処置可能であることの指標を与える。抗体は、αvβ6を持つ細胞および様々な刺激に対するその応答を検出する実験室研究用の研究試薬として販売することもできる。そのような使用において、モノクローナル抗体は、蛍光分子、スピン標識分子、酵素またはラジオアイソタイプで標識することができ、αvβ6のアッセイを実施するのに必要な試薬全てを含むキットの形態で提供することができる。本明細書に記載される抗体を使用して、αvβ6タンパク質の発現を検出し、がんが抗αvβ6 ADCで処置可能かどうか決定することができる。抗体を使用して、例えば、親和性クロマトグラフィーによって、αvβ6を精製することもできる。
以下の実施例において記載される細胞系は、アメリカンタイプカルチャーコレクション(ATCC)、国立がん研究所(NCI)またはDeutsche Sammlung von Mikroorganismen und Zellkulturen GmbH、Braunschweig、ドイツ(DMSZ)に指定された条件に従って培養で維持された。Detroit562細胞系、HPAFII細胞系、およびBxPC3細胞系は、ATCCから入手した。FreeStyle(商標)293−F(Invitrogen Corp)ヒト上皮腎臓細胞および対応するトランスフェクタントは、製造者によって記載の通り維持された。細胞培養試薬は、Invitrogen Corp.(Carlsbad、CA.)、Molecular Devices(Sunnydale、CA)および他の供給元から入手した。二次抗体試薬は、Jackson ImmunoResearch Laboratories(West Grove、PA)から購入した。組換えヒトαvβ6は社内で調製した。組換えαvβ3およびαvβ8は、R&D Systems(Minneapolis、MN)から購入した。FreeStyle(商標)293−F細胞は、内在性インテグリンαvを発現し、その細胞にヒト、カニクイザルまたはマウスインテグリンβ6をコードする完全長cDNAを安定にトランスフェクトして、それぞれ、HEK293F:huβ6、HEK293F:cynoβ6およびHEK293F:muβ6細胞系を生成した。空のベクター(HEK293F:ベクター)をトランスフェクトしたHEK293F細胞を陰性対照として使用した。内在性マウスインテグリンαvを発現するマウス3T3およびFDC−P1細胞にヒトおよびマウスインテグリンβ6の完全長cDNAクローンをトランスフェクトして、それぞれ、3T3:huβ6およびFDC−P1:muβ6を生成した。
飽和結合アッセイ
以下の抗原を発現している細胞系を使用して飽和結合研究を行った:293F:huβ6;293F:cynoβ6;FDCP1:muβ6およびラットNBT−II。抗原を発現している細胞0.25〜0.5×106個を、96ウェルv底プレートに、1ウェル毎に分注した。m15H3、h15H3(HBLC)およびh15H3(HTLC)を、同等のレベルのビオチンで直接標識し、0.1pM〜150nMの範囲の濃度で、細胞に添加した。細胞を、氷上で30分間インキュベートし、FACS緩衝液(PBS、2%ウシ胎仔血清、1mM MgCl2、1mM CaCl2、0.02%NaN3)で3回洗浄し、FACS緩衝液中でPE結合体化ストレプトアビジン(0.5μg/mL)と共に氷上で45分間インキュベートし、3回洗浄し、FACS緩衝液に再懸濁した。結合は、Becton Dickinson Biosciences FACSCalibur(San Jose、CA)を使用して検出した。GraphPad Software(LaJolla、CA)を使用して、見かけのKdを算出した。
96ウェルMaxisorbプレート(Nunc)を、PBSに希釈した1μg/mL組換えヒトαvβ6、αvβ3およびαvβ8で4℃で終夜コーティングした。0.05%Tween20を含むPBS(PBS−T)でプレートを洗浄した。洗浄緩衝液を取り除き、プレートをTBSブロッキング緩衝液(1mM MnCl2、0.05%Tween20、1%BSAを加えたTBS)中で、室温で30分間ブロッキングした。プレートを洗浄し、次いで4.6ng/mL〜10μg/mLの範囲の濃度でTBSブロッキング緩衝液に希釈したビオチン化抗体(m15H3またはh15H3)と共に1時間インキュベートした。プレートを洗浄し、1μg/mL HRP−ストレプトアビジンと共に1時間インキュベートし、洗浄し、次いでTMB基質と共に10分間インキュベートした。1M H2S04で反応を停止させた。450nmでの吸光度を、Fusion HTプレートリーダー(Perkin Elmer、Waltham、MA)を使用して読み取った。
293F:huβおよびHEK293F:cynoβ6細胞系を使用して競合結合アッセイを行った。抗原を発現している細胞1×105個を、氷上で96ウェルv底プレートの各ウェルに分注した。細胞を、2nM AlexaFluor−647標識マウス15H3および濃度を増加させた(0.024nM〜100nM)未標識のヒト化15H3構築物と共に1時間インキュベートした。細胞をペレット化し、PBSで3回洗浄した。細胞をペレット化し、PBS/BSA 125μLに再懸濁した。蛍光をフローサイトメトリによって分析し、飽和蛍光シグナルのパーセントを使用して、結合している標識マウス15H3のパーセントを決定し、続いて様々な傾きを有するシグモイド用量反応曲線にデータをフィッティングすることによってEC50を推定した。
一次抗体としてマウスαvβ6 mAb、および製造業者(DAKO A/S、Glostrup、デンマーク)によって記載の通りDAKO QiFiKitフローサイトメトリ間接アッセイを使用して、細胞表面のαvβ6コピー数の定量化を決定し、Becton Dickinson FACScanフローサイトメーターを用いて評価した。
腫瘍細胞を、抗αvβ6抗体薬物結合体と共に37℃で96時間インキュベートした。結合しない(h00)ADCを、陰性対照として使用した。CellTiter−Glo(登録商標)発光アッセイ(Promega Corporation、Madison、WI)を使用して細胞生存率を決定し、Fusion HTプレートリーダー(Perkin Elmer、Waltham、MA)で結果を測定した。媒体処理した細胞(対照=100%)と比較して生存率を50%低下させるのに必要な化合物の濃度であるIC50として、結果を記録する。
抗αvβ6抗体の抗体薬物結合体は、US20050238649に記載の通り調製した。薬物リンカーvcMMAEとmcMMAFの両方については、US20050238649に記載されており、全ての目的のために参照により本明細書に組み込まれる。
ヌード(nu/nu)マウス(動物7〜10匹/群)に培養で増殖させた腫瘍細胞を移植した。25%マトリゲル中の5×105個の細胞を使用してDetroit562(ATCC)を移植し、25%マトリゲル中の1×106個を使用してHPAFII(ATCC)を移植した。腫瘍が100mm3に達したら(q4d×4回の腹腔内注射)、ヒト化αvβ6 ADCまたは結合しない対照ADCを用いる投薬を開始した。ノギスを使用して腫瘍体積をモニタリングし、腫瘍体積がおよそ800mm3に達したら、動物を安楽死させた。1匹または複数の動物を安楽死させるまで、腫瘍体積中央値プロットを各群に対して続けた。動物の手順の全ては、実験動物ケア評価認証協会によって公認された施設において動物飼育使用委員会によって承認されたプロトコルの下で実施した。
Multi−Tumor Microarrays(TMA)(cat#MC5001)は、US Biomax Inc.から購入した。全ての試料をBond−Max(商標)自動染色機(Leica)で処理した。
Bond(商標)Dewax溶液(Leica、cat#AR9222)を72℃で使用して、マルチ腫瘍アレイブロックMC5001のパラフィン切片を保持するスライドを脱パラフィンし、再水和した。EDTAに基づくBond(商標)Epitope Retrieval Solution2(Leica、cat#AR9640)を使用して、抗原回復を95〜100℃で20分間実施した。スライドは、protein block(DAKO cat#X0909)で30分間処理した後、5μg/mL一次抗体マウス抗インテグリンβ6クローン437216(RnD MAB#41551)と共に25℃で45分間インキュベートした。バックグラウンド染色の陰性対照として、アイソタイプ適合性マウスIgG2b(Zymed #02−6300)を5μg/mLで使用した。自動化IHC染色のために、アルカリホスファターゼ−Fast Redに基づく検出キット(Bond(商標)Polymer AP Red検出キット;Leica、cat#DS9305)を使用した。発色現像の後、切片をヘマトキシリンで対比染色し、カバーガラスを被せた。病理学者によってスライドを評価およびスコア付けし、Olympus BX41顕微鏡を使用して画像を撮像した。
凍結切片を保持するスライドを、アセトン中で、−20℃で10分間インキュベートして、組織を固定した。PeroxAbolish(Biocare cat#PXA96)過酸化物ブロッキング試薬を25℃で15分間適用して、内在性ペルオキシダーゼを消失させた。スライドを、アビジンブロック(Vector cat#SP−2001)で、続けてビオチンブロック(Vector cat#SP−2001)で、25℃で各15分間順次インキュベートした。スライドを、10μg/mLヒト化抗インテグリンβ6クローン15H3一次抗体と共に60分間インキュベートした。50mM MnClを含有するBond(商標)Primary Antibody Diluent(Leica cat#AR9352)に一次および二次抗体を調製した。バックグラウンド染色の陰性対照として、10μg/mLヒトIgG(Ancell cat#295−010)を使用した。一次抗体インキュベーションの後に、ビオチン化ヤギ抗ヒト二次抗体(Jackson cat#109−066−098)を5μg/mLで使用し、25℃で30分間インキュベートした。ビオチン化二次抗体の検出のために、VECTASTAIN Elite ABC試薬(Vector cat#PK−7100)を25℃で30分間適用した。Bond(商標)Polymer Refine検出キット(Leica cat#DS9800)を使用して、DAB発色現像した。切片をヘマトキシリンで対比染色し、カバーガラスを被せた。病理学者によってスライドを評価およびスコア付けし、Olympus BX41顕微鏡を使用して画像を撮像した。
1.マウス抗体の結合
マウスαvβ6モノクローナル抗体15H3抗体および選択されたそのヒト化バージョンのKDを、ヒト、カニクイザル、ラットおよびマウスに対して決定した。ラットを除外して、遺伝子工学によって作られた細胞系(293F:huβ6、293F:cynoβ6またはFDC−P1:muβ6)を使用して、インテグリンβ6オルトログに対する競合および飽和結合研究を行った。遺伝子工学によって作られた細胞系は内在性αvを発現し、このαvは組換えβ6鎖と一組になって、内在性αvと組換えβ6からなるヘテロ二量体受容体が生成される。ラットαvβ6に対する親和性は、ラットNBT−II膀胱癌細胞を使用して決定され、この細胞は内在性αvβ6を発現する。
およそ5×106個の3T3:huβ6トランスフェクタントの腹腔内注射によりBALB/cマウスを3回免疫した。融合の3日前に、マウスは、静脈内(6μg)および腹膜内(30μg)に投与される精製組換えヒトαvβ6の最後の注射を受けた。ポリエチレングリコールを使用して、P3X63Ag8.653骨髄腫細胞に脾臓およびリンパ節から採取したリンパ球を融合した。融合した細胞を、ハイブリドーマ増殖培地(4mM グルタミン、10%Fetal Clone I、10%Cloning Factorおよびペニシリン/ストレプトマイシンを含有するIMDM)中で終夜回復させた。回復の後に、細胞を遠沈し、次いで、半固体培地に播いた。半固体培地は、ハイブリドーマ選択用のHATおよびIgG産生用のCloneDetectをプラスしたハイブリドーマ増殖培地で補充されたCloneMatrix培地からなった。ハイブリドーマを、37℃で10日間インキュベートした。10日目に、ClonePixFL(Molecular Devices)を使用してIgGを生成しているハイブリドーマクローンを採集し、HTをプラスしたIgGを枯渇させたハイブリドーマ増殖培地を含有する96ウェルプレートに移した。ハイブリドーマ培養上清を293F:huβ6トランスフェクタントに対してスクリーニングし、検出にAlexifluor−647標識二次抗体を使用して陽性クローンを同定した。プレートを、FMAT8200(Applied Biosystems)で読み取った。293F:huβ6および293F:cynoβ6に結合したが293F:ベクターには結合しなかったハイブリドーマを直接コンジュゲーション用として展開した。直接結合体化した抗体パネルを、結合および細胞毒性アッセイで試験した。m15H3は、複数のαvβ6陽性腫瘍細胞系に対してADCとして細胞毒性活性を示し、ヒトおよびカニクイザル形態抗原に対して同等の親和性を有していたので、リード抗体として選択した。抗体が293F:huβ6トランスフェクタントに結合するがαvβ5陽性親系統(293F:ベクター)に結合しないことが示されたFMATおよびFACS結合研究において、マウス15H3およびヒト化15H3の特異性を確認した。抗体が組換えヒトαvβ6には結合するがαvβ3またはαvβ8には結合しなかったELISAにより、m15H3およびh15H3の結合特異性も確認した。
この実施例において、ヒト化の出発点またはドナー抗体は、マウス15H3抗体である。重鎖に対するVH1−46およびJH4ならびに軽鎖に対するVK2−30およびJk2によって提供されるゲノム配列を使用した。
ホルマリン固定パラフィン包埋組織(FFPE)を使用する様々な腫瘍型の免疫組織化学的分析に、抗B6クローン437216(R&D systems)を使用した。
ADCのin vitro抗腫瘍活性
細胞毒性アッセイを使用して、抗αvβ6 ADCのin vitro抗腫瘍活性を測定した。定量的FACS分析により、様々な細胞系においてαvβ6発現の調査を実施した。下の表を参照すると、マウス15H3 ADC(vcMMAEまたはmcMMAF(US20050238649に記載される小分子および/またはリンカーの両方)と結合体化されているマウス15H3抗体)は、αvβ6細胞を殺すのに有効であった。
膵臓がん(HPAFII、BxPC−3)ならびに頭頸部がん(Detroit562)モデルを使用して、選択したヒト化αvβ6 ADC(抗体当たり平均4薬物)の抗腫瘍活性をin vivoで(図10〜12)実証した。vcMMAEに結合体化されているαvβ6 ADCは、未処置および対照ADCと比較して有意な腫瘍遅延または腫瘍寛解を示した。h15H3−vcMMAE(4)とは、抗体当たり平均4個のvcMMAE薬物リンカー分子を有するマウス親抗体のヒト化形態の抗体薬物結合体のことを指す。h00−vcMMAE(4)とは、抗体当たり平均4個のvcMMAE薬物リンカー分子を有する、結合しない対照抗体の抗体薬物結合体のことを指す。h15H3−1はHBLC構築物であり、h15H3−2はHLLC構築物であり、h15H3−3はHTLC構築物である。
Claims (32)
- 配列番号1に示される配列を含む重鎖可変領域および配列番号2に示される配列を含む軽鎖可変領域を含む、インテグリンαvβ6に特異的に結合する単離されたマウス15H3モノクローナル抗体、ならびにそのキメラ形態またはヒト化形態。
- 配列番号3(CDR1)、配列番号4(CDR2)、および配列番号5(CDR3)に示される重鎖相補性決定領域(CDR)配列、ならびに配列番号6(CDR4)、配列番号7(CDR5)、および配列番号8(CDR6)に示される軽鎖CDR配列を含み、かつ各CDRに0、1、2または3個の保存的アミノ酸置換を有する、インテグリンαvβ6に特異的に結合する単離されたモノクローナル抗体。
- インテグリンαvβ6に特異的に結合するヒト化モノクローナル抗体であって、前記抗体は、
HA(配列番号9)と少なくとも90%同一のアミノ酸配列を有する重鎖可変領域であって、ただし、重鎖可変フレームワーク領域に少なくとも1つのマウス15H3復帰変異があり、かつ前記抗体が配列番号3(CDR1)、配列番号4(CDR2)、および配列番号5(CDR3)に示される重鎖相補性決定領域(CDR)配列を含み、かつ各CDRに0、1、2もしくは3個の保存的アミノ酸置換を有することを条件とする、重鎖可変領域を含むか、または
LA(配列番号10)と少なくとも90%同一のアミノ酸配列を有する軽鎖可変領域であって、ただし、前記抗体が配列番号6(CDR1)、配列番号7(CDR2)、および配列番号8(CDR3)に示される軽鎖相補性決定領域(CDR)配列を含み、かつ各CDRに0、1、2もしくは3個の保存的アミノ酸置換を有することを条件とする、軽鎖可変領域を含む、

LA(配列番号10)と少なくとも90%同一のアミノ酸配列を有する軽鎖可変領域であって、ただし、前記抗体が配列番号6(CDR1)、配列番号7(CDR2)、および配列番号8(CDR3)に示される軽鎖相補性決定領域(CDR)配列を含み、かつ各CDRに0、1、2または3個の保存的アミノ酸置換を有することを条件とする、軽鎖可変領域を含む、請求項2に記載の抗体。 - 配列番号5に示される重鎖CDR3、配列番号8に示される軽鎖CDR3を有し、かつ残りのCDRに0、1、2または3個の保存的アミノ酸置換を有する、請求項2、3または5に記載の抗体。
- 前記抗体がヒト化抗体であり、かつ前記重鎖可変フレームワーク領域に少なくとも1つのマウス15H3復帰変異がある、請求項5または6に記載の抗体。

- 抗原結合断片である、上述の請求項のいずれかに記載の抗体。
- 前記重鎖可変領域がヒト重鎖定常領域に融合され、かつ前記軽鎖可変領域がヒト軽鎖定常領域に融合されている、上述の請求項のいずれか1項に記載の抗体。
- 前記重鎖定常領域が、Fcガンマ受容体に対する結合が天然のヒト定常領域と比較して減少した天然のヒト定常領域の変異体形態である、請求項17に記載の抗体。

- がんを有する患者を処置する方法であって、前記患者に、上述の請求項のいずれか1項に記載の抗体の有効なレジメンを施すことを含み、前記抗体は、キメラ抗体、ヒト抗体またはヒト化抗体である、方法。
