Abstract
Description
遊離の抗癌薬は、細胞全体に拡散し、ある特定の細胞内部位に濃縮されない。さらに、かかる薬物が静脈内に投与される場合、これらは、身体の全組織に全身的に分布する。これらの意図しない分布の部位におけるこれらの薬物の作用は、観察可能な全身性の副作用をもたらす。したがって、薬物を作用が望まれる身体の部位に局在化させることが好ましい。これらの薬剤を薬剤が最も有効である細胞内部位を標的とすることによって、これらの効果を増大させ、毒性を減少させる。
本明細書では、「HPMA」という用語は、化合物N−(2−ヒドロキシプロピル)メタクリルアミドを意味し、以下の構造によって表される親水性ポリマーである:
「抗癌剤」は、癌と戦うのに有用な任意の薬剤を意味する。担体分子および/またはリンカーに直接的にもしくは間接的に結合することができる、任意の抗癌剤を用いることができる。開示された組成物と共に用いることができる抗癌剤の部分的なリストは、例えば、参照により本明細書に組み込まれる特許文献2、ならびに抗癌剤を含む前記文献中で引用された任意の特許公報および特許、または特許出願において見出すことができる。特許文献3、特許文献4、および特許文献5はまた、抗癌化合物に関連する薬剤を記載している。抗癌剤のクラスは、それだけには限らないが、化学療法薬、細胞毒、代謝拮抗剤、アルキル化剤、蛋白リン酸化酵素(プロテインキナーゼ)阻害薬、アントラサイクリン系薬剤、抗生剤、有糸分裂阻害剤(例えば、抗チューブリン薬)、コルチコステロイド、放射性医薬品、およびタンパク質(例えば、サイトカイン、酵素、またはインターフェロン)が含まれる。具体的な例には、それだけには限らないが、ドセタキセル、ゲムシタビン、イマチニブ(Gleevec(登録商標))、5−フルオロウラシル、9−アミノカンプトテシン、アミン修飾ゲルダナマイシン、ドキソルビシン、パクリタキセル(タキソール(登録商標))、プロカルバジン、ヒドロキシ尿素、メソe−クロリン、シスプラチン、Gd(+3)化合物、アスパラギナーゼ、および放射性核種(例えば、I−131、Y−90、In−111、およびTc−99m)が含まれる。当技術分野で既知の多くの抗癌剤があり、多くは開発され続けている。
任意の担体分子を用いることができる。典型的には、担体分子は、ポリマー(高分子)である。典型的には、担体ポリマー(高分子)は、少なくとも約5,000ダルトンの大型の巨大分子である。他の実施形態において、担体分子は、少なくとも約25,000ダルトン、少なくとも約50,000ダルトン、少なくとも約100,000ダルトン、少なくとも約125,000ダルトンまたは少なくとも約150,000ダルトンである。担体分子は、約5,000ダルトンから約25,000ダルトン、または約25,000ダルトンから約100,000ダルトン、または約50,000ダルトンから約130,000ダルトン、または約100,000ダルトンから約170,000ダルトン、または約120,000ダルトンから約200,000ダルトン、または約120,000ダルトンから約1,000,000ダルトンの範囲であってよい。担体分子は、細胞膜を通って抗癌剤を輸送する助けとなる。したがって、抗癌剤が、担体分子に直接的にまたは間接的に結合されるとき、それは、通常、抗癌剤単独よりも多く細胞膜を横断する。担体分子として機能する当技術分野で既知の多数の担体および高分子担体がある。担体分子の例は、「酵素および光によって活性化可能な薬物の同時送達のための薬物送達系」に関する特許文献6;「合成ポリマー薬物」に関する特許文献7;「親水性N,N−ジエチルアクリルアミドコポリマー」に関する特許文献8;「N−置換アクリルアミド、N−置換メタクリルアミドおよびN,N−二置換アクリルアミドベースのコポリマーならびにその製造方法」に関する特許文献9;「可溶性親水性ポリマーおよびそれらを生成する方法」に関する特許文献10;「親水性亜硝酸コポリマー」に関する特許文献11;および「可溶性親水性ポリマーおよびそれらを生成する方法」に関する特許文献12にも記載され、そのそれぞれは、その全体が個別におよび明確に参照により本明細書に組み込まれる。
「リンカー」は、薬物または標的指向化リガンドを担体分子から空間的に隔てる基を意味する。リンカーは、それだけには限らないが、ポリ(エチレングリコール)、アミノ酸、ポリ(アミノ酸)(例えば、ペプチドもしくはオリゴペプチド)、またはポリペプチド(例えば、タンパク質)などの任意の種類の実体であってよく、その一方の端は担体分子との共有結合を形成することができ、他方の端は薬物または標的指向化リガンドとの共有結合を形成することができる。リンカーは、Gly−Phe−Leu−Gly(配列記号1)などの、リソソーム分解され易い特定の配列を有する短いペプチドを含むこともできる。他の例には、前立腺癌の場合、前立腺細胞に、および配列特異的タンパク質分解能力を有する前立腺特異抗原(PSA)に標的指向化した連結が含まれる。例えば、PSAは、His−Ser−Ser−Lys−Leu−Gln(配列記号5)およびグルタリル−4−ヒドロキシプロリル−Ala−Ser−シクロヘキサグリシル−Gln−Ser−Leu(配列記号6)を加水分解する。
「標的指向化リガンド」という用語は、所望の活性のために特定の部位に本発明の化合物を送達するのに役立つ分子を意味し、すなわち、それは、化合物を局在化させる。局在化は、分子の決定因子の特異的認識、標的指向化薬剤またはコンジュゲートの分子サイズ、イオン相互作用、疎水的相互作用などによって媒介される。特定の組織または領域に薬剤を標的指向化する他の機序は、当業者に知られている。標的指向化リガンドには、例えば、標的指向化した細胞表面上で分子に結合する分子が含まれる。模範的な標的指向化リガンドには、抗体、抗体断片、低分子の有機分子、ペプチド、ペプトイド、タンパク質、ポリペプチド、オリゴ糖、トランスフェリン、HS−糖タンパク質、凝固因子、血清タンパク質、β−糖タンパク質、G−CSF、GM−CSF、M−CSF、EPOなどが含まれる。本発明の模範的な実施形態では、標的指向化系には、RGDfK、EPPT1ペプチド、または葉酸などの標的指向化リガンドを担体分子、リンカー、または抗癌剤に共有結合することが含まれる。
本明細書に記載した化合物は、抗癌剤単独によって用いられるものと典型的には異なる機序を用いて細胞による抗癌剤の取り込みを可能にさせることを特徴とし得る。取り込みの効率および/または特異性が担体分子によって増加されるか否かを決定する多くの方法がある。効率および/または特異性の典型的な増加は、少なくとも2倍、5倍、10倍、25倍、50倍、100倍、500倍、1000倍、5,000倍または10,000倍以上となり得る。
本発明の化合物は、当技術分野で知られている技法を用いて調製することができる。記載されているように、化合物を生成するために用いられる4つまでの構成要素があり、すなわち、抗癌剤、担体分子、任意選択の標的指向化リガンドおよび任意選択のリンカー分子である。具体的な実施形態では、本発明の化合物には、担体分子、抗癌剤、標的指向化リガンド、および少なくとも1種のリンカー分子が含まれる。前述した構成要素のいずれかは、任意の可能な組み合わせまたは順序で互いに反応させて本発明の化合物を生成することができる。構成要素の2つを一緒に結合して(すなわち、反応させて)、新規な反応生成物もしくは中間体を生成し、次いで、中間体を次の成分と化学的に結合することが好ましい場合もある。例えば、抗癌剤は、抗癌/担体分子を生成するために担体分子と反応させることができる。同様に、抗癌剤は、抗癌/リンカー分子を生成するためにリンカー分子と反応させることができる、あるいは、リンカー分子は、リンカー/担体分子を生成するために担体分子と反応させることができる。その場合は、これらの中間体のそれぞれは、個別の成分と反応させることができる(例えば、担体/リンカー/抗癌剤を生成するための担体分子との抗癌/リンカーの反応)、あるいは、中間体のそれぞれは、化合物を生成するために互いに反応させることができる(例えば、抗癌/担体分子との抗癌/リンカー分子の反応)。
開示される化合物は、抗癌剤の細胞への標的指向化した送達のために用いることができる。本明細書に開示されている化合物は、薬学的に許容される形態でおよび抗癌剤もしくは類似の化合物の送達を必要とする対象に有効量で投与することができる。対象は、例えば、マウス、ラット、ウサギ ハムスター、イヌ、ネコ、ブタ、雌ウシ、ヒツジ、ヤギ、ウマ、またはサル、ゴリラ、オランウータン、チンパンジーもしくはヒトなどの霊長類などの哺乳動物とすることができる。
化合物の投与のための用量範囲は、送達が行われる所望の効果を十分にもたらす大きさのものである。用量は、望ましくない交差反応、アナフィラキシー反応などの有害な副作用を引き起こすほどの大きさであるべきではない。一般に、用量は、患者における年齢、状態、性別および疾患の程度によって変わり、当業者によって決定することができる。用量は、任意の使用禁忌の事象において個別の医師によって調整することができる。用量は、1日または数日間、1日1回または複数回の用量投与で約1mg/kgから30mg/kgまで変わり得る。
化合物のいずれかは、1種または複数の薬学的に許容される担体と併用して医薬組成物に治療的に用いることができる。
単一剤形を生成するために担体材料と組み合わせることができる有効成分の量は、治療される宿主および特定の投与方法に応じて変わる。
HPMAコモノマー薬物コンジュゲートを、異なるモル比でHPMAモノマーおよび活性化したMA−GFLG−薬物(配列記号1)コモノマーの重合によって合成した。

HPMAコポリマー−薬物(薬物はゲムシタビンおよびドセタキセルを表す)コンジュゲートを、スキーム5に示す通り開始剤としてN,N’−アゾビスイソブチロニトリル(AIBN、4.4mg)を用いて、50℃で24時間アセトン(0.72ml)/DMSO(0.08ml)中のコモノマーHPMA(86.7mg、0.603mmol)およびMA−GFLG−薬物(配列記号1)(5mg、0.004mmol、薬物がドセタキセルである場合)の遊離基沈殿共重合によってコモノマーから合成した。コモノマーの供給組成を、MA−GFLG−薬物(配列記号1)0、2、および10モル%をそれぞれ含むように変化させた。コモノマー:開始剤:溶媒の比を、12.5:0.6:86.9重量%で一定に保った。典型的には、AIBNおよびHPMAをアセトンに溶解し、少量のDMSO中のMA−GFLG−薬物(配列記号1)の溶液と混合した。混合物を窒素でアンプルに封入し、50℃で24時間撹拌しながら放置して重合させた。沈殿させたポリマーをメタノールに溶解し、20×体積のエーテルに再沈殿した。低分子量の未反応のモノマーおよび他の不純物を蒸留水に再溶解することによってポリマーコンジュゲートから分離し、蒸留水に対して透析して塩を除去し、続いて、凍結乾燥して純粋な生成物を得た。

表1に列挙する通り、一連のポリマー薬物コンジュゲートの合成に成功した。合成されたポリマー−薬物コンジュゲートの重量平均分子量(Mw)および多分散性を、高速タンパク質液体クロマトグラフィー(FPLC)系(Amersham Biosciences)のSuperose12 HR 10/30カラム(Amersham Biosciences)を用いた分子ふるいクロマトグラフィーによって推定した。1mg/mlにおけるサンプルを、溶離溶媒としてPBSを用いて流速0.4ml/分で溶離した。ポリマーの数平均分子量(Mn)、重量平均分子量(Mw)および多分散性(n=Mw/Mn))を、既知の分子量のポリHPMA画分を用いた検量線から推定した。薬物含有量をアミノ酸分析(Commonwealth Biotechnologies Inc、Richmond、VA)を用いて得た。結果を表1に示した。ポリマーの全体的なサイズ分布は、類似の系に関する文献で報告された立証済みの値と一致した。
1)癌細胞株増殖
HPMA−薬物コンジュゲートの効果を決定する癌細胞株を以下の供給源から得た。すなわち、米国アメリカン・タイプ・カルチャー・コレクション(ATCC)(Manassas、VA)から得られたヒトMDA−MB−231(乳癌)、HCT116(結腸癌)およびPANC−1(膵臓癌)、米国国立癌研究所(Bethesda、MD)から得られたUMRC2(腎臓癌)である。細胞を10%FBS、P/Sおよび10mM HEPESを補充したダルベッコ改変イーグル培地(「DMEM」、Invitrogen)中で維持した。すべての細胞を37℃で加湿した5%CO2下でインキュベートした。
ヒト癌細胞系に対するHPMA−薬物コンジュゲートの増殖阻害アッセイを、スルホロ−ダミンB(「SRB」)法を用いて行った(例えば、非特許文献9参照)。簡単に述べると、対数増殖期の癌細胞を2〜3×103細胞/ウェルの密度で96穴プレートに播種し、翌日HPMAコポリマー−薬物コンジュゲートで処理した。各処理をトリプリケートウェルで行った。水を対照として用いた。細胞をHPMAコポリマー−薬物コンジュゲートで加湿した5%CO2雰囲気中で96時間37℃でインキュベートした。96時間インキュベートしてから、細胞を、10%トリクロロ酢酸(「TCA」)で固定し、4℃で1時間インキュベートし、水道水で3回洗浄した。続いて、細胞を、1%酢酸中の0.4%スルホロ−ダミンBで30分間染色し、1%酢酸で3回洗浄し、再び風乾した。10mMトリス溶液中で5分撹拌してから、各ウェルの530nmにおける吸光度を、Benchmark Plus Microplate reader(Bio−Rad Laboratories、Hercules、CA)を用いて測定した。吸光度の値が、HPMAコポリマー−薬物コンジュゲートで処理後の生細胞測定数を直接与える。
%生存=生細胞数[試験]/生細胞数[対照]×100
IC50値を非線形回帰分析によって算出した。
動物モデルにおける腫瘍増殖阻害を観察するために、ヌードマウス異種移植モデルを後述の通りHPMAに結合させたドセタキセルまたはゲムシタビンを利用して行った。
Claims (15)
- 担体分子、
前記担体分子に共有結合したリンカー分子、および
前記リンカー分子に共有結合した抗癌剤を含む化合物であって、
前記担体分子は、ポリ(N−2−ヒドロキシプロピルメタクリルアミド)(ポリHPMA)を含み、
前記抗癌剤はドセタキセルであり、
前記担体分子に共有結合している標的指向化リガンドを含む、
ことを特徴とする、化合物。 - 前記担体分子に前記標的指向化リガンドが、第2のリンカー分子を介して共有結合していることを特徴とする請求項1に記載の化合物。
- 前記リンカー分子および第2のリンカー分子が、それぞれ、アミノ酸またはオリゴペプチドを含むことを特徴とする請求項1または2に記載の化合物。
- 前記リンカー分子が、リソソームにより分解されるアミノ酸配列およびオリゴペプチドを含むことを特徴とする請求項1〜3のいずれか一項に記載の化合物。
- 請求項3または4に記載の化合物であって、
前記アミノ酸またはオリゴペプチドが、
Gly−Phe−Leu−Gly(配列記号1)、
Gly−Ileu−Phe、Gly−Val−Phe、Gly−Gly−Phe、Gly−Gly−Phe−Phe(配列記号2)、
Gly−Ileu−Tyr、Phe、Gly、Gly−Gly、Ala、Ser、Gly−Phe、Gly−Leu Phe、Gly−Phe−Phe、Gly−D−Phe−Phe、Ala−Gly−Val−Phe(配列記号3)、
Gly−Gly−Val−Phe(配列記号4)、
Gly−Phe−Tyr、Gly−B−Ala−Tyr、Gly−Lue、Gly−Phe−Gly、His−Ser−Ser−Lys−Leu−Gln(配列記号5)、および
グルタリル−4−ヒドロキシプロリル−Ala−Ser−シクロヘキサグリシル−Gln−Ser−Leu(配列記号6)からなる群から選択されることを特徴とする請求項4に記載の化合物。 - 前記リンカー分子が、Gly−Phe−Leu−Gly(GFLG(配列記号1))オリゴペプチドであることを特徴とする請求項1〜5のいずれか一項に記載の化合物。
- 前記担体分子が、10,000ダルトンから250,000ダルトンの分子量を有することを特徴とする請求項1〜6のいずれか一項に記載の化合物。
- 前記担体分子が、100,000ダルトンから170,000ダルトンの分子量を有することを特徴とする請求項1〜7のいずれか一項に記載の化合物。
- 前記第2のリンカー分子が、アミノ酸またはオリゴペプチドであることを特徴とする請求項2〜8のいずれか一項に記載の化合物。
- 前記第2のリンカー分子が、オリゴペプチドGly−Gly(GG)を含むことを特徴とする請求項2〜8のいずれか一項に記載の化合物。
- 前記標的指向化リガンドが、RGDfK、EPPT1、および葉酸からなる群から選択されることを特徴とする請求項1〜10のいずれか一項に記載の化合物。
- 前記リンカー分子が、GFLG(配列記号1)オリゴペプチドであり、前記第2のリンカー分子が、GGオリゴペプチドであることを特徴とする請求項2〜7、9および11のいずれか一項に記載の化合物。
- 請求項1〜12のいずれか一項に記載の化合物と、薬学的に許容される担体又は希釈剤を含む医薬組成物。
- 癌の治療の医薬品の製造において、請求項1〜12のいずれか一項に記載の化合物を使用する方法。
- 前記癌が、B細胞リンパ腫、T細胞リンパ腫、{きんじょうそく にくしゅ}菌状息肉症、ホジキン病、骨髄性白血病、膀胱癌、脳癌、神経系癌、頭頚部癌、頭部および頚部の有棘細胞癌、腎臓癌、肺癌、神経芽細胞腫/グリア芽腫、卵巣癌、膵癌、前立腺癌、皮膚癌、肝臓癌、黒色腫、口、咽頭、喉頭また肺の有棘細胞癌、結腸癌、子宮頚癌、子宮頚癌、乳癌、上皮癌、頭部および頚部癌、大腸癌、造血性癌;精巣癌;結腸および直腸癌、前立腺癌、膵癌、結腸直腸癌、星状細胞腫、多発性骨髄腫、鼻癌、咽頭癌、胃癌、細胞腫、有棘細胞癌、腺癌、肉腫、神経膠腫、芽細胞腫、形質細胞腫、組織球腫、腺腫、低酸素性腫瘍、骨髄腫、小細胞肺癌、非小細胞肺癌、腎癌、尿生殖器癌、肺癌、食道癌、膵癌であることを特徴とする請求項14に記載の使用する方法。
