を含む。適切な濃度は、先に記載されており、好ましくは4mM〜50mM、より好ましくは4mM〜20mMの範囲に含まれる。
細胞含有生体試料、ここでは全血試料を、室温(RT)で最長6日間または7日間インキュベートする試験系を設計した。その試験系では、本発明の添加剤の試料安定化特性を、0日目、3日目、および6/7日目に試料で試験した。試料を、適用できる場合には以下のプロトコルに従って処理した(詳細については、結果部分の具体例も参照されたい)。
1.1.赤血球の溶解
−血液試料2mlを新しい15mlのFalcon管に移す
−5倍緩衝液EL(キアゲン(QIAGEN))を添加する
−試料を反転させる(10×)
−氷上でインキュベートする(10分)
−400×gおよび4℃で10分間、遠心分離処理を施す
−上清を破棄する
−2倍緩衝液EL(キアゲン(QIAGEN))を白血球細胞ペレットに添加する
−わずかにボルテックスすることによって、ペレットを緩衝液EL(キアゲン(QIAGEN))に再溶解させる
−400×gで10分間、遠心分離処理を施す
−上清を破棄する
−FACS Flow(ベクトン ディッキンソン、プリマウス、UK(Becton、Dickinson Plymouth、UK))500μlを白血球細胞ペレットに添加する
−わずかにボルテックスすることによって、ペレットをFACS Flowに再溶解させる
−FACS Flow 1mlを新しいFACS管に移す
−再溶解させたペレット100μlをFACS管に移す
測定を、製造者の指示(FACSCalibur、ベクトン ディッキンソン、プリマウス、UK(Becton、Dickinson Plymouth、UK))に従って実施した。
全血から血漿を分離するために、血液試料を5000rpmで15分間、遠心分離処理し、得られた血漿試料を、16,000×gで4℃において10分間、再び遠心分離処理した。
QIAamp(登録商標)循環NA(Circulating NA)キット(ハンドブックに従う)を使用して、得られた血漿試料から循環細胞外核酸を精製した。簡潔には以下の通りであった。
−試料投入量10ml、
−溶解。プロテイナーゼK1mlおよび緩衝液ACL(キアゲン(QIAGEN))8ml
−結合。緩衝液ACB(キアゲン(QIAGEN))18ml
−洗浄ステップ。変更なし、ハンドブックに従う
−緩衝液AVE(キアゲン(QIAGEN))60μl中で溶出
3に従って得た溶出液を、すべての試料(6/7日目の試料を含む)が精製されるまで−20℃で貯蔵した。後に、同じ条件による溶出液をプールし、以下の通り処理した。
−プライマー濃度を8μmから16μmまで拡大した。
−アニーリング/伸長ステップを、1分から2分まで延長した。
(試料を、1:10希釈した後に増幅した)
引き続き、実施試験の詳細を説明する。実施例で使用した方法の詳細は、Iで既に記載した。
広範なカスパーゼ阻害剤として作用する2つの異なるオリゴペプチド、Q−VD−OPhおよびZ−Val−Ala−Asp(OMe)−FMKを試験した。
溶出した無細胞循環DNAを、チップゲル電気泳動法(その方法の詳細については、先のI、4.1を参照されたい)を使用して、サイズによって分離した。図1aは、得られた結果を示す。DMSO対照およびK2E血液(本発明の教示に従って処理していない)は、同じはしご状のバンドパターンを示す。このパターンは、アポトーシスが生じる試料で発生する。アポトーシス中に、エンドヌクレアーゼがヌクレオソーム間リンカー領域でゲノムDNAを分解し、約180bpまたは180bpの倍数のDNA断片を産生する。したがって、アポトーシスは、明確なはしご状のパターンを示す試料で発生する。さらに、パターン強度(濃い)が明白である。バンドが濃くなるにつれて、多くのゲノムDNAが細胞から放出された。したがって細胞外核酸集団を汚染する。
溶出した無細胞循環DNAも、DNA分解に対して感度が高いリアルタイムPCRアッセイで定量化した(その方法の詳細については、先のI、4.2を参照されたい)。図1b)は、RTにおける7日間の貯蔵期間中の細胞外核酸集団の安定化に対する、試験したカスパーゼ阻害剤の効果、ここではDNAの増加について示す(18S DNA二本鎖アッセイ)。
リアルタイムPCRおよびゲル電気泳動法の結果をまとめると、Q−VD−OPhまたはZ−Val−Ala−Asp(OMe)−FMKの添加により、DNA断片化が阻害され、さらには血漿へのゲノムDNAの放出が低減されることが実証された。したがって、カスパーゼ阻害剤を全血に添加することは、試料、特に細胞外核酸集団を安定化するのに室温でも有効である。したがって、本発明の安定化方法を使用すると、試料の質を危険にさらすことなく、室温でも全血試料を輸送することができる。また、より長い貯蔵期間にわたってゲノムDNAの放出を完全に防止するために、Q−VD−OPhの濃度を増大することもできる。
この実施例では、低濃度のカスパーゼ阻害剤Q−VD−OPhを、グルコースと組み合せて試験し、グルコースを組合せパートナーとして添加して、血液細胞が生き延びるのを支持した(細胞損傷を防止することによって)。21.4mMのグルコースおよび4μM、1μMまたは0μMのQ−VD−OPhを、BDバキュテイナ管に採血した血液10mlに添加し、室温で最長7日間貯蔵した。全血試料を、セクションIに記載の通り処理した。2(血漿の調製)および3(核酸の単離)を参照されたい。
溶出したDNAを、チップゲル電気泳動法(その方法の詳細については、先のI、4.1を参照されたい)を使用して、サイズによって分離した。図2aは、カスパーゼ阻害剤を添加しなかった対照試料と比較して、1μMのカスパーゼ阻害剤でも、7日目のゲノムDNA放出/断片化が著しく低減されたことを示している。この効果は、4μMのカスパーゼ阻害剤を使用すると改善される。したがってカスパーゼ阻害剤は、非常に低濃度でも、特に炭水化物と組み合わせると、血液試料を安定化するのに有効である。
図2bは、21mMのグルコースと組み合わせた試験濃度のカスパーゼ阻害剤Q−VD−OPhの、RTにおける7日間の貯蔵期間中の血漿中ゲノムDNA増加に対する効果を示している(18S DNA二本鎖アッセイ)。Q−VD−OPhをグルコースと組み合せて添加すると、血漿へのゲノムDNAの放出が著しく低減する。図2bは、安定化のために1μMのQ−VD−OPHを全血試料に添加するだけでも、7日間の貯蔵期間中のゲノムDNAはわずかしか増加しなかったことを示している。4μMのQ−VD−OPhを添加すると、血漿へのゲノムDNAの放出は、最大4倍まで阻害される。それとは対照的に、本発明による安定化なしに全血をK2E管中に採血すると、血漿中DNAは約40倍増加する。
驚くべきことに、本発明者らはまた、全血中で高張媒体として作用する試薬を添加することによって、血液細胞を安定化できることを見出した。例えばヒドロキシル化有機化合物(複数可)を全血に添加することによって高張媒体を生成すると、全血に含有されている血液細胞からの水の放出が微量まで抑えられ、細胞収縮によって安定性が増大する。前記細胞収縮は、機械力に対して細胞を安定化すると想定される。
血液細胞の完全性を、FACSを使用して分析した(その方法の詳細については、先のI、1を参照されたい)。図3は、フローサイトメトリーによって測定した血液細胞の完全性を示す。ドットプロットは、3種の異なる細胞集団、顆粒球(1)、単球(2)およびリンパ球(3)を可視化している。プロットの左下領域の雲(4)は、主に赤血球の溶解によって生じた破片を表す。
また図4aに提示した結果は、DHAで処理した試料では、PAXgene(登録商標)DNA採血管で貯蔵した試料よりもゲノムDNAの放出が著しく低減されるので、DHAの添加によって血液試料が安定化されることを示している。さらに、図4aから明らかなように、DHAで安定化された試料は、はしご状の分解パターンを示さず、このことは、アポトーシス、それぞれにDNAの分解が効率的に防止されることを示唆している。
図4bは、RTでの6日間の貯蔵期間にわたる、DNAの増加に対するDHAの効果を示す(18S DNA二本鎖アッセイ)。リボソーム18S DNAのレベルは、貯蔵して3日目まで一定のままであるように見えたため、3×MOPSに溶解したDHAによって最良の結果を得た。
この実施例では、異なる濃度(0.7M、0.5Mおよび0.2M)のDHAの安定化効果を試験した。
図5は、フローサイトメトリーによって測定した血液細胞の完全性を示す。ドットプロットは、3種の異なる細胞集団、顆粒球(1)、単球(2)およびリンパ球(3)を可視化している。プロットの左下領域の雲は、主に赤血球の溶解によって生じた破片を表す。
図6aに提示の結果も、異なる濃度のDHAを添加することによってゲノムDNAの放出およびDNAの分解が効率的に防止されるので、血液試料が安定化されることを示している。
図6bは、RTでの6日間の貯蔵期間中の、DNAの増加に対する異なるDHA濃度の効果を示している(18S DNA二本鎖アッセイ)。図6bに示した通り、全血中0.5MのDHAは、ゲノムDNAの放出を最も効率的に防止する。さらに、短いアンプリコンコピー数と長いアンプリコンコピー数の比は、3日目まで一定のままであり、6日目までにごくわずかに低下する。これらの結果は、全血の安定化に対する高張剤DHAの効果が注目に値することを実証している。
採血管中のEDTAを増大すると、PAXgene(登録商標)DNA採血管について知られている通り、マイクロクロット(microclotting)およびマクロクロット(macroclotting)が阻害される。したがって、より高濃度のEDTAは、血液細胞および血漿中の細胞外核酸の安定化を支持することができる。さらに、先に提示の実験は、カスパーゼ阻害剤、特にQ−VD−OPhおよび浸透活性化合物DHAの、血液細胞損傷に対する阻害作用を示しており、特に、細胞外核酸集団中のゲノムDNA、特に断片化ゲノムDNAの増加が効率的に低減されることを示している。驚くべきことに、試験したカスパーゼ阻害剤も、無細胞(血漿)画分へのゲノムDNAの漏出を防止/阻害した。したがって、これらの試薬の組合せにより、全血中の細胞外核酸、特に細胞外DNAの安定化が改善され、その安定性が少なくとも6日間継続し、さらには血液細胞が効率的に安定化され、それによって、安定化されなければ血漿中の天然細胞外核酸レベルを希釈するはずのゲノムDNAの放出が防止される。
図7aは、フローサイトメトリーによって測定した血液細胞の完全性を示す。ドットプロットは、3種の異なる細胞集団、顆粒球(1)、単球(2)およびリンパ球(3)を可視化している。プロットの左下領域の雲は、主に残存する赤血球の溶解によって生じた破片を表す。
図7bは、EDTA、DHAおよびカスパーゼ阻害剤Q−VD−OPHの組合せの、RTにおける6日間の貯蔵期間中のDNAの増加に対する効果を示す(18S DNA二本鎖アッセイ)。この結果は、EDTA、DHAおよびQ−VD−OPHの組合せによって、血漿中の細胞外DNAが著しく強力に安定化され(測定した18S rDNAのレベルは、6日目まで一定のままである)、3日間の貯蔵期間が経過するまで、血液細胞からのゲノムDNAの放出が強力に防止される(短いアンプリコンコピー数と長いアンプリコンコピー数の比は、一定のままである)ことを示している。血漿中ゲノムDNAがごくわずかに増大したことが、3日間の貯蔵期間と6日間の貯蔵期間との間で明白である。
K2EDTA、Q−VD−OPhおよびDHAの組合せは、全血中の遊離循環DNAおよび血液細胞の完全性に対して注目すべき安定化効果を示したので、遊離循環RNAに対するこれらの薬剤の安定化能も分析した。血漿において一定レベルの遊離循環RNAを保存するために(採血時に提示のものとして)、安定化試薬(複数可)は、RNAが分解しないように保護し、RNAが崩壊血液細胞から放出されないように防止するだけでなく、その代謝経路を阻害するべきであり、それぞれに代謝経路の変化が細胞外DNA血漿レベルに影響を及ぼさない効果を有するべきであり、それぞれの作用をそれぞれに低減すべきである。したがって、実験5を反復し、mRNAレベルをリアルタイムRT−PCRによって測定した。
2種の異なる濃度のDMAAを、K2EDTAと共に試験し、EDTA単独と比較した(K2E BD、K2EDTA18mg)。
図11は、血漿中ゲノムDNAの増大に対する、DMAAの試験濃度の効果を示している。DMAAの添加は、血漿へのゲノムDNAの放出を著しく低減する。多量のDMAAを全血に添加するほど、放出されるDNAが減少する。1.5%DMAAを全血試料に添加すると、6日間の貯蔵期間中、無細胞DNAの増加はわずかしか観測されなかった。さらに、1.5%DMAAを添加すると、0.75%の場合よりも効率的に全血試料中の無細胞DNAレベルが安定化され、測定した短い18S DNAコピー数と長い18S DNAコピー数の比は、0日目から6日目まで減少するので、DMAA濃度が1.5%よりも高いと、より効率的な安定化効果を得ることができる。

まず、全血試料10mlを、BDバキュテイナK2E−EDTA(4.45mMのEDTA=参照)に収集した。次に、以下の溶液2mlを添加した(示した濃度は、安定化された血液溶液中の最終濃度を表す)。各条件を、異なるドナーからの6つの管で試験した。

5.EDTA50mg、1μMのOPH、5%DMAA、
6.EDTA50mg、1μMのOPH、10%DMAA、
7.EDTA50mg、2μMのOPH、5%DMAA、
8.EDTA50mg、2μMのOPHおよび10%DMAA。
まず、全血試料10mlを、BDバキュテイナK2E−EDTA(4.45mMのEDTA=参照)に収集した。後に、以下の溶液2mlを添加した(示した濃度は、安定化された血液溶液中の最終濃度を表す)。各条件を、6つの管、したがって6人の異なるドナーで試験した。

5.0.01MのDHA、3%DMAA、1μMのOPH、14mMのEDTA;
6.1μMのOPH、14mMのEDTA、0.01MのDHA、0.01Mのマルチトール;
7.1μMのOPH、14mMのEDTA、0.01MのDHA、0.05Mのイノシトール;
8.1μMのOPH、14mMのEDTA、0.01MのDHA、0.05Mのイノシトール、0.01Mのマルチトール。
6人の異なるドナーから、全血試料をBDバキュテイナK2Eに収集し、次に全血10ml当たり以下の安定化溶液2mlを添加した(示した濃度は、安定化された血液溶液中の最終濃度を表す)。
1.2μMのOPH、14mMのEDTA、5%DMAA、
2.1μMのOPH、14mMのEDTA、3%DMAA、
3.1μMのOPH、14mMのEDTA、0.01MのDHA、3%DMAA。
細胞外核酸は、試料中にしばしば非常に少量で含まれる。したがって、安定化された試料中の細胞外核酸を効率的に保存するだけでなく、さらにその後、安定化された試料から細胞外核酸を高収率で単離することもできる安定化手順を得ることが重要である。実施例14は、安定化された試料から細胞外核酸をより高収率で単離することができるという点で、本発明の安定化方法が、従来技術の安定化方法よりも優れていることを実証している。本発明の安定化方法は、有利には検出限界を低減し、したがって細胞外核酸集団内の希少な標的核酸も確実に測定することができる。
1.無細胞RNA BCT(Streck社、カタログ番号218976。安定剤としてホルムアルデヒド放出薬を含む)
2.BDバキュテイナK2E(BD、カタログ番号367525。EDTAを含む)=参照
3.安定化用QGN(5%DMAA、14mMのEDTA、2μMのOPH(カスパーゼ阻害剤))
全血試料をBDバキュテイナ2KE管に収集した。後に、以下の安定化溶液2mlを添加した(示した濃度は、安定化された血液溶液中の最終濃度を表す)。次にHIVおよびHCVを、104IU/mlで全血試料に添加した。
1.5%DMAA、EDTA50mg、1μMのOPH、0.05Mイノシトール、
2.5%DMAA、EDTA50mg、1μMのOPH、0.01Mマルチトール、
3.5%DMAA、EDTA50mg、1μMのOPH、0.05Mイノシトール、0.01Mのマルチトール、
4.2%イノシトール、4%ソルビトール。
2人の異なるドナーからの血液を、10mlのK2EDTA管(BD)に収集した。それぞれに収集した血液4.5mlを、それぞれK2EDTA34.2mg、キノリン−Val−Asp−CH2−OPH(カスパーゼ阻害剤)溶液(DMSO388μlに1mgを溶解した)1.2mlおよびN,Nジメチルプロパンアミド0.15gもしくは0.3gもしくは0.45g、またはDMAA0.3mlを含有する安定化溶液0.9ml(安定化溶液1ml当たり)と混合した。それによって、以下の通り、血液/安定化混合物中、それぞれ以下の最終濃度を得た。K2EDTA5.7mg、1μMのキノリン−Val−Asp−CH2−OPH(カスパーゼ阻害剤)および2.5%、5%もしくは7.5%(w/v)のN,Nジメチルプロパンアミド、または5%(v/v)DMAA。
Claims (21)
- 試料をカスパーゼ阻害剤と接触させることによって、細胞含有試料に含まれている細胞外核酸集団を安定化する方法。
- 前記試料を、
a)少なくとも1種のカスパーゼ阻害剤;
b)任意選択により、前記試料に含まれている前記細胞を安定化する少なくとも1種の高張剤;および
c)任意選択により、少なくとも1種の式1の化合物
と接触させるステップを含む、請求項1に記載の方法。 - 前記試料を、抗凝固剤とさらに接触させる、請求項1または2に記載の方法。
- 安定化に起因して、前記試料に含有されている細胞からのゲノムDNAの、前記試料の無細胞部分への放出が低減され、かつ/または前記試料中に存在する核酸の分解が低減される、請求項1から3の1項または複数項に記載の方法。
- a)前記カスパーゼ阻害剤が、以下の特徴:
i)汎カスパーゼ阻害剤(Pancaspase)であること;および/もしくは
ii)Q−VD−OPhおよびZ−Val−Ala−Asp(OMe)−FMKからなる群から選択されること、
の1つもしくは複数を有し、
ならびに/または
b)前記高張剤が、以下の特徴:
i)非荷電であること;
ii)細胞収縮を誘発することによって、前記試料に含まれている前記細胞を安定化すること;
iii)細胞不透過性であること;
iv)水溶性であること;
v)ヒドロキシル化有機化合物であること;
vi)ポリオールであること;
vii)ヒドロキシカルボニル化合物であること;
viii)炭水化物もしくは糖アルコールであること;および/もしくは
ix)ジヒドロキシアセトンであること、
の1つもしくは複数、好ましくは2つ以上を有し、
ならびに/または
c)前記式1の化合物が、以下の特徴:
i)R1、R2およびR3が、1〜5個の炭素原子を含むこと;
ii)R1、R2およびR3が、1個または2個の炭素原子を含むこと;
iii)R4が、酸素であること;
iv)Ν,Ν−ジアルキル−カルボン酸アミドであること;
v)Ν,Ν−ジメチルアセトアミド、N,N−ジエチルアセトアミド、Ν,Ν−ジメチルホルムアミドおよびΝ,Ν−ジエチルホルムアミドからなる群から選択されること;および/もしくは
vi)N,N−ジメチルプロパンアミドであること、
の1つもしくは複数を有する、
請求項1から4の1項または複数項に記載の方法。 - 前記試料を、少なくとも1種の抗凝固剤、好ましくはEDTAなどのキレート剤とさらに接触させる、請求項1から5の1項または複数項に記載の方法。
- 前記試料を、前記カスパーゼ阻害剤、前記高張剤、式1の前記化合物および/または前記抗凝固剤と接触させた後に得られる混合物が、以下の特徴:
a)前記カスパーゼ阻害剤を、少なくとも0.01μM、少なくとも0.05μM、少なくとも0.1μM、少なくとも0.5μM、少なくとも1μM、少なくとも2.5μMもしくは少なくとも3.5μMから選択される濃度で含むこと;
b)前記カスパーゼ阻害剤を、0.01μM〜100μM、0.05μM〜100μM、0.1μM〜50μM、1μM〜40μM、1μM〜30μMもしくは2.5μM〜25μMから選択される濃度範囲で含むこと;
c)前記高張剤を、少なくとも0.05M、少なくとも0.1M、好ましくは少なくとも0.25M、より好ましくは少なくとも0.5Mの濃度で含むこと;
d)前記高張剤を、0.05M〜2M、0.1〜1.5M、0.15M〜0.8M、0.2M〜0.7Mもしくは0.1M〜0.6Mから選択される濃度範囲で含むこと;
e)式1の前記化合物を、少なくとも0.1%、少なくとも0.5%、少なくとも1%、少なくとも0.75%、少なくとも1%、少なくとも1.25%もしくは少なくとも1.5%の濃度で含むこと;
f)式1の前記化合物を、0.1%〜50%、0.5%〜25%、0.75%〜20%、1%〜15%、もしくは1%〜10%から選択される濃度範囲で含むこと;および/または
g)前記抗凝固剤、好ましくはキレート剤を、0.05mM〜100mM、0.05mM〜50mM、0.1mM〜30mM、1mM〜20mMもしくは2mM〜15mMから選択される濃度範囲で含むこと、
の1つまたは複数を有する、請求項1から6の1項または複数項に記載の方法。 - 前記試料を安定化するために、
a)カスパーゼ阻害剤としての少なくとも1種の汎カスパーゼ阻害剤、
b)少なくとも1種の高張剤、好ましくはヒドロキシル化有機化合物、および
c)任意選択により、少なくとも1種の式1の化合物、好ましくはN,N−ジアルキル−カルボン酸アミド、および
d)任意選択により、抗凝固剤、好ましくはキレート剤、より好ましくはEDTA
と接触させ、
a)〜d)の前記化合物が安定化組成物に含まれる、請求項1から7の1項または複数項に記載の方法。 - 前記試料が、以下の特徴:
a)細胞外核酸を含むこと;
b)全血、血液、血漿、血清、リンパ液、尿、髄液、脳脊髄液、腹水、乳、糞便、気管支洗浄液、唾液、羊水、精液/精漿、スワブ/スメア、体液、身体分泌物、鼻分泌物、膣分泌物、創傷分泌物および排泄物に由来する試料、ならびに細胞培養上清からなる群から選択されること;
c)細胞が欠乏しているもしくは細胞を含有する体液であること;
d)全血、血漿および/もしくは血清から選択されること;ならびに/または
e)全血であること、
の1つまたは複数を有する、請求項1から8の1項または複数項に記載の方法。 - 前記細胞外核酸集団の安定化が、冷蔵されずに、好ましくは室温で、
a)少なくとも2日;
b)少なくとも3日;
c)少なくとも1日〜3日;
d)少なくとも1日〜6日;および/または
e)少なくとも1日〜7日、
から選択される期間にわたって達成可能である、請求項1から9の1項または複数項に記載の方法。 - a)前記1つまたは複数の安定化剤、および任意選択によりさらなる添加剤が、安定化組成物に含まれ、前記安定化組成物と、特定体積の前記細胞含有試料の体積比が、10:1〜1:20、5:1〜1:15、1:1〜1:10および1:2〜1:5から選択され;
b)前記安定化された試料に、核酸の分析方法および/または検出方法を適用し;
c)前記安定化された試料から、細胞外核酸を単離し;
d)前記安定化された試料から、細胞外核酸を単離し、前記単離された核酸を、分析かつ/または検出し;
e)前記安定化された試料に含まれている細胞を除去し;
f)前記安定化された試料に含まれている細胞を除去した後、単離、分析および/または検出ステップを実施し;
g)請求項10に記載の安定化期間が経過した後に、核酸単離ステップを実施し;
h)(i)前記安定化された試料、(ii)細胞が除去された、前記安定化された試料、および/もしくは(iii)前記試料から除去された細胞を貯蔵し;
i)前記安定化された試料から除去された細胞を廃棄し;かつ/または
j)前記安定化された試料から除去された細胞から、核酸を単離する、
請求項1から10の1項または複数項に記載の方法。 - 血液試料をカスパーゼ阻害剤および抗凝固剤と接触させることを含む、前記血液試料に含まれている細胞外核酸集団を安定化するための方法であって、安定化に起因して、前記血液試料に含有されている細胞からのゲノムDNAの、前記血液試料の無細胞部分への放出が低減され、かつ前記試料中に存在する核酸の分解が低減される、請求項1から11の1項または複数項に記載の方法。
- 生体試料、好ましくは血液試料から細胞外核酸を単離する方法であって、
a)請求項1から12の1項または複数項に記載の方法に従って、前記試料中の前記細胞外核酸集団を安定化するステップと;
b)細胞外核酸を単離するステップと、
を含む、方法。 - 以下のステップ:
i)任意選択により、ステップa)とステップb)との間で、前記細胞含有試料から細胞を除去するステップ;
ii)請求項11に定義のステップb)〜j)の1つもしくは複数を実施するステップ;ならびに/または
iii)請求項13に記載のステップb)で、抽出、固相抽出、核酸結合性固相を使用する単離方法、シリカ材料を使用する単離方法、アニオン交換基を含む固相の使用に基づく単離方法、磁気粒子に基づく精製、フェノール−クロロホルム抽出、アルコールおよび/もしくはカオトロピック剤(複数可)に基づく核酸単離方法、クロマトグラフィー、アニオン交換クロマトグラフィー、アニオン交換粒子に基づく単離、電気泳動法、濾過、沈殿、標的核酸に特異的な単離方法、ならびにその組合せを含む群から選択される単離方法を使用して、前記試料から細胞外核酸を単離するステップ、
の1つまたは複数を含む、請求項13に記載の方法。 - 前記単離された核酸を、さらなるステップc)で、処理かつ/または分析し、好ましくは、
i)修飾し;
ii)少なくとも1種の酵素と接触させ;
iii)増幅し;
iv)逆転写し;
v)クローン化し;
vi)配列決定し;
vii)プローブと接触させ;
viii)検出し;
ix)定量化し;かつ/または
ix)同定する、
請求項13または14に記載の方法。 - a)前記試料の無細胞部分から単離され、かつ/または請求項13に記載のステップb)で単離した後に得られる前記細胞外核酸集団が、以下の特徴:
i)単離された前記核酸全体に、一部として含まれること;
ii)主にDNAを含むこと;
iii)主にRNAを含むこと;
iv)細胞外循環核酸を含むこと;
v)疾患関連核酸を含むこと;
vi)腫瘍関連もしくは腫瘍由来の核酸を含むこと;
vii)炎症関連核酸を含むこと;
viii)胎児核酸を含むこと;
ix)ウイルス核酸を含むこと;
x)病原体核酸を含むこと;
xi)哺乳動物の細胞外核酸を含むこと;および/もしくは
xii)DNAとRNAとの混合物であること、
の1つもしくは複数を有し、
ならびに/または
b)ステップc)で、分析され、かつ/またはさらに処理され、好ましくは検出される前記細胞外核酸が、以下の特徴:
i)DNAであること;
ii)RNAであること;
iii)細胞外循環核酸であること;
iv)疾患関連核酸を含むこと;
v)腫瘍関連または腫瘍由来の核酸を含むこと;
vi)炎症関連核酸を含むこと;
vii)胎児核酸であること;
viii)ウイルス核酸であること;
ix)病原体核酸であること;
x)哺乳動物の細胞外核酸であること;および/もしくは
xi)DNAとRNAとの混合物であること、
の1つもしくは複数を有する、
請求項13から15の1項または複数項に記載の方法。 - 生体試料、好ましくは血液試料中の細胞外核酸集団を安定化する組成物であって、
a)少なくとも1種のカスパーゼ阻害剤
を含み、
以下のb)、c)およびd)
b)前記試料に含まれている細胞を安定化する少なくとも1種の高張剤、好ましくは少なくとも1種のヒドロキシル化有機化合物;
c)少なくとも1種の式1の化合物
d)少なくとも1種の抗凝固剤、好ましくはキレート剤、
から選択される少なくとも1種のさらなる化合物を含む、組成物。 - 少なくとも1種のカスパーゼ阻害剤および少なくとも1種の抗凝固剤を含む、請求項17に記載の組成物。
- 以下の特徴:
a)前記試料に含有されている細胞からのゲノムDNAの、前記試料の無細胞部分への放出を低減できること;
b)前記試料中に存在する核酸、特にゲノムDNAの分解を低減できること;
c)前記カスパーゼ阻害剤が、請求項5に記載の特徴の1つもしくは複数を有すること;
d)請求項5に記載の少なくとも1種の高張剤を含むこと;
e)請求項5に記載の少なくとも1種の式1の化合物を含むこと;
f)生体試料、好ましくは血液、血漿もしくは血清と混合した後に得られる混合物が、前記カスパーゼ阻害剤、前記高張剤、式1の前記化合物および/もしくは前記キレート剤を、請求項7に記載の濃度で含むこと;
g)固体形態で提供されること;
h)液体形態で提供されること;ならびに/または
i)前記試料に含有されている前記細胞外核酸集団を、室温で少なくとも3日間、好ましくは少なくとも6日間にわたって安定化できること、
の1つまたは複数を有する、請求項17または18に記載の組成物。 - 前記安定化組成物が、生体試料との混合物として提供され、前記試料が、以下の特徴:
a)細胞外核酸を含むこと;
b)全血、血漿、血清、リンパ液、尿、髄液、脳脊髄液、腹水、乳、糞便、気管支洗浄液、唾液、羊水、精液/精漿、スワブ/スメア、体液、身体分泌物、鼻分泌物、膣分泌物、創傷分泌物および排泄物、ならびに細胞培養上清からなる群から選択されること;
c)細胞が欠乏しているもしくは細胞を含有する体液であること;
d)全血、血漿および/もしくは血清から選択されること;ならびに/または
e)全血であること、
の1つまたは複数を有する、請求項17から19の1項または複数項に記載の組成物。 - 前記安定化組成物と、特定体積の前記細胞含有試料の体積比が、10:1〜1:20、5:1〜1:15、1:1〜1:10および1:2〜1:5から選択される、請求項20に記載の組成物。
