Abstract

1. 血液は、輸送体流体である一方で、唾液は、プロテアーゼ、酵素、および分泌された物質が豊富であり得る消化液であり、尿は、所望されない老廃物から成る排泄液である。
2. 細胞は、長期間にわたって唾液中で無傷で生存しない。細胞が数週間も(またはいくつかの細胞型については数年さえも)血液中で生存することができる一方で、唾液中の細胞生存は、典型的には、1時間未満である。15、45、および90分後に生存する唾液中の細胞の割合は、それぞれ、わずか約66%、33%、および27%であることが報告されている。唾液は、その生理学的性質によって、分析または診断のための全細胞保存に適さない環境を提示する。いくつかの研究は、細胞の95%が60分まで生存できないことを実証している(Baron et al.,1999)。

(m) 疾患または感染症の休眠および/または潜伏ならびに/もしくは隠密形態を示す、1つまたはそれを上回るバイオマーカならびに/もしくは1つまたはそれを上回る放出された因子。例えば、そのような疾患は、LTBIであり得る。加えて、または代替として、そのような因子は、ペプチドおよび/またはサイトカインであり得る。
(n) 随意に、細胞内HIVウイルスを検出することによる、細胞中のHIV感染の存在。

19. 1mlあたりの保存された細胞または所定の細胞型の数は、少なくとも約5,000、または少なくとも約10,000、もしくは少なくとも約12,000個である、先行項目のうちのいずれかに記載の溶液。
20. 細胞は、T細胞である、先行項目のうちのいずれかに記載の溶液。
21. 溶液は、約6.4〜約8.4のpHで緩衝される、少なくとも1つの化学的固定剤と、少なくとも1つのプロテアーゼ阻害剤とを含む、先行項目のうちのいずれかに記載の溶液。
22. さらなる下流分析のため、および/または病状の診断のために、自然に発現された体液中の少なくとも細胞外成分を保存するための、随意の先行項目のうちのいずれかに記載の溶液であって、溶液は、随意に、約6.4〜約8.4のpHで緩衝される、少なくとも1つの化学的固定剤を含む、溶液。
23. 化学的固定剤は、アルデヒドから成る群から選択される、項目21または22に記載の溶液。
24. 化学的固定剤は、パラホルムアルデヒドである、項目23に記載の溶液。

(a) 疾患の退行または攻撃性を識別するマーカ(細胞もしくは分子)を伴う細胞。疾患は、例えば、癌(随意に、限定されないが白血病)であり得る。痰であるか、または痰を含有する流体の場合、疾患は、肺癌、肺癌腫、非小細胞肺癌(NSCLC)、肺炎、もしくは結核のうちのいずれか1つまたはそれを上回るものであり得る。
(b) 出生前細胞。
(c) (例えば、転移性または別様の)循環腫瘍細胞。
(d) 正常な細胞の希少な形態、例えば、以下のうちのいずれか1つまたはそれを上回るもの:

・ TRAILタンパク質(TNF関連アポトーシス誘発リガンド)は、感染症(細菌対ウイルス)を判別するために使用されることができる。これは、抗生物質耐性菌に対抗するために非常に有用である。
・ TRAILタンパク質は、殆どの細胞によって分泌され、リガンドとしてアポトーシスのプロセスを誘発する、サイトカインである。
(g) 自閉症を示す、1つまたはそれを上回るバイオマーカ。例えば、
オキシトシン−親和結合の確立に影響を及ぼす。負の影響として、ASD中のオキシトシンは、5−HTと負に相関する。オキシトシン受容体をコードするOXTR遺伝子の多型は、自閉症、対結合(pair−conding)、社会的行動、情動的影響、およびASD形質と関連付けられる。
メラトニン−24時間周期および季節リズム、ならびに免疫応答および神経可塑性の変調で良好に役割を果たすように確立されている。メラトニンは、酵素、すなわち、アセチルセロトニンメチルトランスフェラーゼ(ASMT)を介して、N−アセチルセロトニン、次いで、メラトニンに変換される、5−HTから合成される。プロセスは、日中/光によって阻害される。ASD患者の血漿中の低いレベルのメラトニンは、本ASMT酵素の欠如に起因すると考えられる(エピジェネティック(DNA、タンパク質、およびRNA適用可能分析))。ASMT遺伝子バリアントは、ASDと関連付けられる。同じ遺伝子はまた、437人の対照のいずれにも認められなかったのに対して、398(1.51%)人のASD個体のうちの6人において破壊的コーディング変異を宿している。

24. 少なくとも1つのイオン性界面活性剤または洗剤が、トリトン、トリトン−X、トリトン100、トリトン−X100、マルトシド(例えば、n−ドデシル−β−D−マルトピラノシドおよび/またはDDM)、グリコシド(例えば、ジギトニン)、Tween(例えば、Tween 20および/またはTween 80)、3−[(3−コラミドプロピル)ジメチルアンモニオ]−1−プロパンスルホネート(および/またはCHAPS)、ノニルフェノキシポリエトキシルエタノール(および/またはTergitol型NP−40および/またはNP−40)、前述のうちのいずれかの誘導体のうちのいずれか1つもしくはそれを上回るものとして選択される、項目22または23に記載の組成物。
25. 塩は、塩化ナトリウム(NaCl)、塩化リチウム(LiCl)、塩化カリウム(KCl)から選択される、いずれか1つまたはそれを上回るものである、項目22、23、または24に記載の組成物。
26. 組成物は、249mM、250mM、275mM、300mM、325mM、350mMから選択される、少なくとも1つを上回る、総塩濃度を有する、項目22〜25のいずれかに記載の組成物。
27. イオン性および非イオン性界面活性剤または洗剤は、混合ミセル集団ならびに/もしくは混合ミセルを形成する、項目22〜26のいずれかに記載の組成物。
28. エタノールをさらに含む、先行項目のうちのいずれかに記載の組成物。
29. 組成物は、実質的にエタノールを含まない、項目1〜27のいずれかに記載の組成物。
30. 自然に発現された体液を受容するためのコンテナと、収集されたサンプル中の核酸を抽出して保存するための組成物を含有するチャンバとを備える、自然に発現された体液の収集のための体液サンプル収集デバイスであって、組成物は、先行項目のうちのいずれかに従って規定される通りである、体液サンプル収集デバイス。
31. 自然に発現された体液のサンプルを受容するためのコンテナと、収集されたサンプル中の核酸を抽出して保存するための組成物の第2の体積を含有するチャンバを備える、自然に発現された体液の所定の第1の体積の収集のための、随意に、項目30に記載の体液サンプル収集デバイスであって、第2の体積は、第1の体積を実質的に上回らない、随意に、それより小さい、体液サンプル収集デバイス。
32. 第2の体積が、第1の体積の約100%以下;第1の体積の約90%以下;第1の体積の約80%以下;第1の体積の約70%以下;第1の体積の約60%以下;第1の体積の約50%以下;第1の体積の約40%以下;第1の体積の約30%以下;第1の体積の約20%以下;第1の体積の約90%;第1の体積の約80%;第1の体積の約70%;第1の体積の約60%;第1の体積の約50%;第1の体積の約40%;第1の体積の約30%;第1の体積の約20%;第1の体積の約3の1;第1の体積の約4分の1から選択される少なくとも1つによって規定される、項目31に記載の体液サンプル収集デバイス。
33. 第1の体積は、約0.5ml〜約2.25ml、約0.75ml〜約2.25ml、約1ml〜約2ml、約1ml〜約1.5ml、約1mlのうちの少なくとも1つによって規定される、項目31または32に記載のデバイス。
34. 第1の体積は、約1mlであり、第2の体積は、約250μlである、項目31、32、または33に記載のデバイス。
35. 自然に発現された体液のサンプルを受容するためのコンテナと、収集されたサンプル中の核酸を抽出して保存するための組成物を含有するチャンバとを備える、自然に発現された体液のサンプルの収集のための、随意に、項目30〜34のいずれかに記載の体液サンプル収集デバイスであって、組成物の体積は、約100μl〜約1ml、少なくとも約100μlおよび1ml未満、約250μl〜約750μl、少なくとも約100μl、少なくとも約250μl、少なくとも約500μl未満、1ml未満、約750μl未満、約500μl未満、約100μl、約250μl、約500μl、または約750μlから選択される、少なくとも1つによって規定される、体液サンプル収集デバイス。
36. 自然に発現された体液のサンプル中の核酸を抽出して保存するための項目1〜29のいずれかに記載の組成物のサンプル収集デバイスにおける使用。
37. 家庭で使用するためのサンプル収集デバイスを使用する、ミトコンドリアDNAの抽出および保存のためのプロセスにおけるプロテイナーゼKの使用。
(i)そこから核酸を解放するように細胞および/または細胞小器官を溶解させるための少なくとも1つの細胞ならびに/もしくは細胞小器官溶解剤。好適な溶解剤は、1つまたはそれを上回る界面活性剤、および/もしくは1つまたはそれを上回る洗剤、および/もしくは1つまたはそれを上回る塩を含んでもよい。例示的界面活性剤は、ドデシル硫酸ナトリウム(SDS)であってもよいが、加えて、または代替として、他の界面活性剤が使用されてもよい。例示的洗剤は、トリトン(例えば、トリトン−X、トリトン100、トリトン−X100)、および/またはデオキシコール酸塩(例えば、デオキシコール酸ナトリウム)であってもよいが、加えて、または代替として、他の洗剤が使用されてもよい。例示的塩は、塩化ナトリウム(NaCl)、および/または塩化リチウム(LiCl)、ならびに/もしくは塩化カリウム(KCl)から選択されてもよいが、加えて、または代替として、他の塩が使用されてもよい。付加的界面活性剤および/または洗剤が、以下でさらに説明される。誤解を避けるために、文脈が許可する場合、「界面活性剤」および「洗剤」という用語は、同義的に使用されてもよく、同義的に読み取られることを意図している。また、界面活性剤または洗剤が名前で記述される場合、範囲は、任意の誘導体(例えば、市販の誘導体)を同等に網羅することを意図している。
(a)0%まで(SDSを省略する)を含む、SDSの割合を変動させるステップ。
(b)0%まで(トリトンを省略する)を含む、トリトンの割合を変動させるステップ。
(c)NaClの代用として(NaClを省略する)、もしくは相補体としてNaClと組み合わせてのいずれかで、KClおよび/またはLiCl(緩衝液のイオン性強度を維持する塩)を添加するステップ。
(d)個別および/または総塩濃度を変動させるステップ。
(e)例えば、上記で説明されるような数量で、プロテイナーゼKを添加するステップ。
(f)膜および細胞成分を可溶化することが可能な生物学的洗剤であり、細胞小器官溶解を増加させ得る、0.1〜1%のデオキシコール酸ナトリウムを添加するステップ。デオキシコール酸ナトリウムは、SDSおよび/またはトリトンを代替するために使用されてもよい、もしくは両方と組み合わせて使用されてもよい。SDSおよび/またはトリトンの相対的割合もまた、変動させられてもよい。(例えば、上記で引用される範囲は、唾液と混合させられたときの組成物を参照し得る。濃度は、本明細書に説明される因数R+1に従って、組成物単独中で増加させられてもよい。)
(g)尿素を添加するステップ。尿素は、タンパク質変性を増加させ、細胞溶解を増加させてもよい。SDSおよび/またはトリトンならびに/もしくは他の変性剤の割合は、変動させられてもよい。
(h)SDSおよび/またはトリトンに加えて、もしくはそれの代替物として、1つまたはそれを上回る他のイオン性ならびに/もしくは非イオン性界面活性剤および/または洗剤を使用するステップ。
Claims (20)
- 病状を診断および/または監視する方法であって、
自然に発現された体液のサンプルを取得するステップと、
前記サンプルを前記サンプル中の細胞および/または細胞外成分を安定させるための保存溶液と接触させるステップと、
前記病状を識別、診断、もしくは監視するために、前記サンプルからの安定化された細胞および/または細胞外成分を分析するステップと、
を含む、方法。 - 前記自然に発現された体液は、口腔液、唾液、痰、尿、肺吸引物の流体から選択される、1つまたはそれを上回るものを含む、請求項1に記載の方法。
- 前記サンプルを前記保存溶液と接触させるステップに先立って、前記サンプルを前処理剤と接触させるステップをさらに含み、前記前処理剤は、病状を示す1つまたはそれを上回る因子の放出を刺激するように構成される、請求項1に記載の方法。
- 前記方法はさらに、少なくとも特定の細胞外成分から少なくとも特定の細胞を分離するステップを含む、請求項1に記載の方法。
- 前記分析するステップは、フローサイトメトリを含む、請求項1に記載の方法。
- 前記分析するステップは、蛍光活性化細胞分類(FACS)を含む、請求項1に記載の方法。
- 前記分析するステップは、免疫組織化学を含む、請求項1に記載の方法。
- 前記分析するステップは、分子分析を含む、請求項1に記載の方法。
- 前記分析するステップは、前記サンプル中の検出可能な抗体の存在に先立ってHIVウイルス感染を識別するステップを含む、請求項1に記載の方法。
- 前記分析するステップは、T細胞、随意に、CD4+T細胞内のHIVウイルスを識別するステップを含む、請求項1に記載の方法。
- 前記分析するステップは、随意に、HIV感染および/またはAIDSを示す、免疫不全症を識別するために、CD4+T細胞の量を査定するステップを含む、請求項1に記載の方法。
- 前記分析するステップは、(i)COPDを示す、および/または(ii)ACOSを示す、ならびに/もしくは(iii)COPDのACOS型と非ACOS型とを区別するためのバイオマーカを識別するステップを含む、請求項1に記載の方法。
- 前記分析するステップは、電解質測定、脂質測定、ビタミン測定、肝炎検出、鉄欠乏プロファイル測定、肝機能検査、腎臓プロファイル測定、糖尿病スクリーニング検査、血液像測定、甲状腺プロファイル測定のうちの少なくとも1つを行うステップを含む、請求項1に記載の方法。
- 前記保存溶液は、血液に対して高張性である、請求項1に記載の方法。
- 前記サンプルを保存溶液と接触させるステップの後に、前記保存溶液と混合させられた前記サンプルの組み合わせは、略等張性であり、張度は、血液に対して規定される、請求項1に記載の方法。
- さらなる下流分析のため、および/または病状の診断のために、自然に発現された体液中の細胞ならびに/もしくは細胞外成分を保存するための溶液であって、血液に対して高張性である溶液。
- 前記自然に発現された体液は、口腔液、唾液、痰、尿、肺吸引物の流体から選択される、1つまたはそれを上回るものを含む、請求項1に記載の溶液。
- 流体サンプル中の細胞および/または細胞外成分の下流分析のための流体サンプルであって、前記流体サンプルは、保存溶液と混合させられる自然に発現された低張性体液を含み、前記サンプル流体は、略等張性であり、張度は、血液に対して規定される、流体サンプル。
- 自然に発現された体液のサンプルを収集するためのサンプル収集デバイスであって、前記サンプル収集デバイスは、前記自然に発現された体液中の細胞および/または細胞外成分を保存するための溶液を含有し、もしくは含み、前記溶液は、血液に対して高張性である、サンプル収集デバイス。
- 流体サンプルを含有する、サンプル収集デバイスであって、前記流体サンプルは、(ii)保存溶液と混合させられる(i)自然に発現された低張性体液を含み、前記サンプル流体は、略等張性であり、張度は、血液に対して規定される、サンプル収集デバイス。
