Abstract
Description
(i)抗体の一部が、ZNRF3の細胞外ドメインと結合し、抗体のその他の部分が、R−スポンジンの共受容体の細胞外ドメインと結合する抗体、または
(ii)抗体の一部が、RNF43の細胞外ドメインと結合し、抗体のその他の部分が、R−スポンジンの共受容体の細胞外ドメインと結合する抗体
に関する。
(i)抗体の一方の部分が、ZNRF3の細胞外ドメインと結合し、抗体のもう一方の部分が、R−スポンジンの共受容体の細胞外ドメインと結合する抗体、または
(ii)抗体の一方の部分が、RNF43の細胞外ドメインと結合し、抗体のもう一方の部分が、R−スポンジンの共受容体の細胞外ドメインと結合する抗体
に関する。
Claims (19)
- 膜貫通E3ユビキチンリガーゼの細胞外ドメインと特異的に結合する単離抗体またはその抗原結合断片であって、
(a)膜貫通E3ユビキチンリガーゼは、ZNRF3およびRNF43からなる群から選択され、
(b)抗体の、膜貫通E3ユビキチンリガーゼの細胞外ドメインとの結合が、真核細胞においてWntシグナル伝達を増大し、ここで、膜貫通E3ユビキチンリガーゼが、真核細胞の表面上にある、単離抗体またはその抗原結合断片。 - 増大したWntシグナル伝達が、インビトロアッセイにおいて検出することができる、請求項1に記載の単離抗体または抗原結合断片。
- 少なくとも1種の遺伝子改変されたポリペプチドを含む、請求項1に記載の単離抗体または抗原結合断片。
- 抗体または抗原結合断片のアミノ末端からカルボキシル末端に、3つの領域:

- Wntシグナル伝達の増大から利益を受ける疾患またはその他の適応症が、粘膜炎短腸症候群、胃腸管粘膜における細菌移行、腸内毒素原性または腸疾患性感染性下痢、セリアック病、非熱帯性スプルー、ラクトース不耐症、食事曝露が粘膜絨毛の鈍麻および吸収不良を引き起こすその他の状態、萎縮性胃炎、骨粗しょう症、骨折、代謝性疾患、糖尿病、神経変性疾患、メラノーマならびに組織再生、組織修復または創傷治癒を必要とする状態からなる群から選択される、請求項10に記載の単離抗体または抗原結合断片。
- 真核細胞においてWntシグナル伝達を増大するよう、別の細胞表面タンパク質とさらに特異的に結合し、その他の細胞表面タンパク質が、LGR4、LGR5およびLGR6からなる群から選択される、請求項1に記載の単離抗体または抗原結合断片。
- 薬学的に許容される担体中の、請求項1に記載の単離抗体または抗原結合断片。
- 対象においてWntシグナル伝達の低減から利益を受ける疾患またはその他の適応症の治療において医薬として使用するための、膜貫通E3ユビキチンリガーゼの細胞外ドメインと特異的に結合する単離抗体またはその抗原結合断片であって、
(a)膜貫通E3ユビキチンリガーゼが、ZNRF3およびRNF43からなる群から選択され、
(b)抗体または抗原結合断片の、膜貫通E3ユビキチンリガーゼの細胞外ドメインとの結合が、対象においてR−スポンジン刺激性Wntシグナル伝達を遮断する、単離抗体またはその抗原結合断片。 - 疾患またはその他の適応症が、がん、硬結性骨化症、特発性肺繊維症および心肥大からなる群から選択される、請求項14に記載の単離抗体または抗原結合断片。
- 対象においてWntシグナル伝達の低減から利益を受ける疾患またはその他の適応症の治療において医薬として使用するための、薬学的に許容される担体中の膜貫通E3ユビキチンリガーゼの可溶性細胞外ドメインであって、
(a)膜貫通E3ユビキチンリガーゼが、ZNRF3およびRNF43からなる群から選択され、
(b)対象において、膜貫通E3ユビキチンリガーゼの可溶性細胞外ドメインが、R−スポンジンと特異的に結合して、R−スポンジン刺激性Wntシグナル伝達を遮断する、可溶性細胞外ドメイン。 - 疾患またはその他の適応症が、がん、硬結性骨化症、特発性肺繊維症および心肥大からなる群から選択される、請求項16に記載の可溶性細胞外ドメイン。
- II型糖尿病の治療における医薬として使用するための、
(a)ジペプチジルペプチダーゼ−4(DPP−4)阻害剤の対象への投与の前に、または
(b)DPP−4阻害剤の対象への投与と一緒に、または
(c)DPP−4阻害剤の対象への投与の前、次いで、DPP−4阻害剤の対象への投与と一緒に
II型糖尿病を有する対象に投与される、請求項1に記載の単離抗体または抗原結合断片。 - DPP−4阻害剤がビルダグリプチン(Galvus(登録商標))である、請求項18に記載の単離抗体または抗原結合断片。
