Abstract
Description
本発明の他の目的は、このような新規化合物を有効成分として含有する糖尿病及びその合併症予防または治療用医薬組成物または健康機能食品を提供することである。

済州(チェジュ)西帰浦(ソギポ)市安徳面西広里で採集された丸葉留紅草51gを水を溶媒として50℃で熱水抽出或は水、アルコール、有機溶媒及びそれらの混合溶液で構成された群から選択される溶媒を利用して、室温で音波抽出機で2時間毎に15分ずつ2〜3日抽出した。抽出後、抽出物を真空回転濃縮器で常温で減圧濃縮した後、抽出された残渣を真空凍結乾燥器で乾燥した後、乾燥された丸葉留紅草抽出物を得た。前記濃縮液を水に懸濁して活性炭が装填されたカラムクロマトグラフィに通過させて、丸葉留紅草の有効成分を吸着させて、活性炭が装填されたカラムを蒸留水で洗浄して非吸着成分を除去した。前記非吸着成分が除去された活性炭装填カラムに10〜50(v/v)エタノールなどの有機溶媒を連続的または段階的に濃度を高めながら供給して活性炭に吸着された丸葉留紅草の有効成分を溶出させて、分離及び精製した後、丸葉留紅草抽出物を得た。前記分離及び精製された丸葉留紅草抽出物5gに水を加えて、丸葉留紅草抽出物の濃度が20mg/mLになるように溶解させた。この水抽出物を蒸留水に懸濁した後、ノルマルヘキサン(n-hexane)、エチルアセテート(EtOAc)及びブタノール(BuOH)で順次分画して、ノルマルヘキサン分画、エチルアセテート分画、ブタノール分画及び水分画を各々得た。

実施例1から分離した新規化合物は微黄色の粉末でLC−MSにより分子イオンm/z1059.4、916.4(1052.4−145)、786.9(916.4−136)、514.8(786.9−136)、378.9(514.8−136)、232.9(378.9−146)を得ることができて(図2)、これは分子式C49H88O24と一致する。
実施例1で製造された丸葉留紅草から分離した新規化合物の血管内皮細胞成長因子(VEGF)の抑制能を確認するため、ヒト網膜色素上皮細胞株であるARPE19細胞株に前記新規化合物を処理して、糖尿病性網膜症などの糖尿病合併症を誘発する血管内皮細胞成長因子(VEGF)タンパク質の発現を確認した。
その結果、図5に示したように、新規化合物KRIBB−BH−Pを処理した実験群でVEGF発現が減少することを確認した。
丸葉留紅草から分離した新規化合物KRIBB−BH−Pのインスリン分泌促進能をマウス膵臓β細胞株であるMin6細胞株において確認した。
糖尿患者の場合には白内障が早く起きて、悪化も速く、視力が急速に低下する。糖尿病にかかると、水晶体が混濁する現象が促進される。そこで、レンズ培養実験を介して、糖尿病性白内障生成抑制効能を調べた。
6−1:血糖分析
正常マウスと実際糖尿病マウスに丸葉留紅草由来新規化合物を処理することによって、糖尿指標の血糖、糖化血色素及び尿蛋白の変化を確認した。
マウスを各群当たり5匹にして、正常マウス対照群、糖尿病マウス対照群、糖尿病マウス薬物処理(グリメピリド、アカボス)処理群、丸葉留紅草由来新規化合物と薬物の併用処理群に分けて、丸葉留紅草由来新規化合物投与群の場合は各々0.1mg/kg、1mg/kg、10mg/kgの濃度で各群に経口投与して12週間飼育した。各群のマウスは2週毎に血糖の変化を測定し、さらに、6週毎に尿を採取して、尿蛋白濃度を測定し、12週間飼育した後、血液を採取して糖化血色素の濃度を測定した。
経口糖負荷検査は5週齢のマウス(Male C57BL/6J、19g、コアテク、平沢(ピョンテク))を購入して、一定温度(25℃)と湿度(50%)下1週間適応させた後、実験に用いた。マウスを各群当たり5匹にし、対照群では一般飼料を餌にする一般食餌群、高脂肪飼料を餌にする高脂肪食餌群に分けて、2週間飼育した。高脂肪食餌飼育2週目に実験動物を16時間絶食させた後、空腹時の血糖を測定し、試料は経口で24時間、16時間、8時間、4時間、2時間、1時間前投与し、以降30分毎に血糖を測定した。
糖尿病治療を受ける患者に発生する合併症を低減するためには、血糖数値を適切な水準に維持することが重要である。一時点で測定する血糖数値は、種々の要因によって変動が生じうることから、長期間の血糖調節推移を把握する目的で最も広く用いられる検査が、糖化血色素(HbA1c)である。糖化血色素は、赤血球に正常に存在する血色素に糖が結合した形態で血糖が高く維持された場合に糖化血色素数値も高くなる。糖化血色素は、2〜4ヶ月の間の平均血糖数値を反映するので、長期間の血糖調節程度を把握するのに役立つ。
糖尿病患者の場合、腎臓と心臓合併症が伴う場合が多い。糖尿病は体の老廃物をろ過して出す腎臓を損う。クレアチニン検査は血液中にあるクレアチニンの濃度を測定して腎臓の機能を評価することができる。クレアチニンはタンパク質が筋肉から使用された後、作られるクレアチン脱水物であって、極少量だけが血液に存在する。腎臓の機能が下がると、クレアチニンが排泄されない。各群の糖尿病マウスに12週間経口投与したマウスの尿と血液を採取してELISAキット(Exocell、America)を利用して測定した。
7−1:アラニントランスアミナーゼ濃度分析
実施例5で使用したものと同一な6週齢のマウスと6週齢の糖尿病マウスを一定温度(25℃)と湿度(50%)下1週間適応させた後、肝毒性検査に用いた。
丸葉留紅草由来新規化合物を投与した群の肝機能異常有無を確認するため、前記実施例から採取された血液を利用してビリルビン数値をELISAキット(Cusabio biotech、Japan)で測定した。ビリルビン数値の分析結果は図13に示したように、丸葉留紅草由来新規化合物を投与した群のビリルビン数値は正常範囲の中に含まれる数値であった。したがって、本発明の組成物は肝機能の異常を誘発させない安全な組成物であることがわかる。
実施例7の条件で飼育した糖尿病マウス群の空腹血液検査または糖負荷検査を通じてインシュリン感受性指数を計算した。

アディポネクチンは脂肪細胞から分泌され血液内に非常に豊富に存在し、肥満又は第2型糖尿病患者において減少していることが一貫して証明されている。また、低アディポネクチン血症は、動脈硬化疾患の新たな危険因子として注目されていて、糖尿病患者らの中でも特に、大血管合併症を伴っている患者においてアディポネクチン濃度が減少していた。即ち、アディポネクチンは抗肥満作用、抗糖尿作用、抗動脈硬化作用及び活性酸素生成の抑制作用などがある。実施例7の条件で飼育した糖尿病マウス群の血中アディポネクチンの濃度をELISAキット(R&D、Japan)で測定した。
適当な血糖数値を維持することに中心的な役割をするものに、糖を除去するインシュリンだけではなく糖を増やすグルカゴンというホルモンもある。
インクレティンにはGIP(glucose dependent insulinotropic peptide)とGLP−1(glucagon like peptide)の2種類があって、二つのホルモンすべてが小腸の上皮細胞層内の内分泌細胞(endocrine cell)で分泌され、小腸内血糖が増加し、膵臓β細胞を刺激してインシュリン分泌を促進し、α細胞からのグルカゴン分泌を抑制する。
αグルコシダーゼ阻害剤は、腸粘膜に分布しているαグルコシダーゼの機能を阻害し、それによって増加した食後に血糖を減少させる。すなわち、αグルコシダーゼは多糖を単糖に分解する役割を有し、小腸での単糖の吸収を促進する。ここで、新規化合物はそのような役割を阻害し、そして単糖の吸収を抑制し、消化及び摂取した食物の炭水化物の吸収を遅らせ、食後の血糖及び血中のインシュリンの上昇を減少させ、糖尿病の治療効果を示す。αグルコシダーゼ阻害剤は、高インシュリン血症、又は低血糖症を誘導しないが、小腸におけるインシュリン分泌を促進し、グルカゴン分泌を阻害するグルカゴン様ペプチド−1の分泌を促進する。
腫瘍壊死因子−α(TNF−α)は脂肪細胞から主に発現され、このサイトカインの上昇レベルは肥満及びインシュリン抵抗性と関連している。脂肪組織は腫瘍壊死因子−α、レジスチンとインターロイキン−6(IL−6)のようなサイトカインを生成するが、これらサイトカインはインシュリンの作用を抑制することが確認された。肥満人において交感神経活性が増加するがこれは脂肪分解を増加させ、筋肉の血流を減少させて(ブドウ糖の運搬)インシュリン作用に直接的な影響を与える。腫瘍壊死因子−αは血糖を高め、糖尿病を誘発させたり、血管に流れて炎症形成を防ぐ、コレステロールが蓄積することを抑制するアディポネクチンの分泌を抑制し血管内皮細胞のNF−kB信号伝達を阻害し、大食細胞の貪食作用の活性を抑制する。
実施例7の条件で飼育した糖尿病マウス群から摘出した眼球から水晶体を分離してウェルプレートに移した後LAS3000イメージ分析装備で写真を撮った。混濁度は撮った写真をLAS3000イメージ分析プログラムを用いて分析して水晶体の混濁度を測定した。

(a)留紅草属を水、アルコール、有機溶媒及びこれらの混合物からなる群より選択される溶媒で抽出して、留紅草属抽出物を製造する工程;
(b)前記留紅草属抽出物に水を加えて懸濁し、ノルマルヘキサン、エチルアセテート及びブタノールで順次分画して水分画を得る工程;及び
(c)前記水分画を分離及び精製して化学式2で表わされる化合物を取得する工程;
- 前記留紅草属は丸葉留紅草であることを特徴とする請求項6に記載の製造方法。

- 留紅草の植物培養または組織培養によって得られた抽出物と前記抽出物から得た化合物を追加に添加することを特徴とする請求項16に記載の糖尿病または糖尿病合併症の予防用健康機能食品。
