発明の概要
本発明は、腫瘍細胞培養物、より特に、腫瘍細胞と相違するマトリクス細胞のマトリクス内に3次元的に配置された腫瘍細胞を含むin vitroの3次元器官型細胞共培養物を提供する。
前記マトリクス細胞は、腫瘍細胞が増殖する器官型環境を形成する。「器官型培養」という用語は、培養中の細胞が、該細胞が由来する器官の生化学的及び生理学的な特性をできる限り近く再現する方法で連合することを示す。「共培養」では、少なくとも相違する2種類の細胞が一緒に培養される。共培養細胞は、相違する供給源に由来してもよいが、in vivoの状態を再現するために一緒に付随していても良い。共培養物は、全ての細胞の種類から生じ、共培養物の中の細胞間の相互作用から生じる特性を有する。
本発明の共培養物は、半透性支持体の1つの表面で調製され、増殖されるが、上述のように、本発明の方法及び共培養物は、外因性の足場又は細胞外マトリクスタンパク質を必要としない。
本発明によると、マトリクス細胞は、腫瘍細胞が最初に均一に播種されるマトリクスを提供する。したがって、本発明によると、腫瘍細胞はマトリクス細胞のマトリクス内に3次元に配置される。前記腫瘍細胞はマトリクス内で培養される。前記腫瘍細胞は、いくつかの実施態様ではマトリクスを通って遊走する場合がある。
一般
本発明の器官型共培養物は、多種多様の細胞、多種多様の器官及び組織から調製できる。工程で用いられる細胞の性質は、必要とされる器官型腫瘍共培養物に依存する。
本発明によると、マトリクス細胞は本発明の腫瘍細胞と相違する細胞を含む。マトリクス細胞は本発明の腫瘍細胞と相違する少なくとも1種類の細胞種を含む。
同様に、前記腫瘍細胞は、いずれの細胞、組織又は器官の種類に属してもよい。特に、前記腫瘍細胞は、前記マトリクス細胞について上記のいずれかの種類に属してもよい。前記腫瘍細胞は、細胞株由来か、若しくは1次がん細胞由来であってもよい。また、前記腫瘍細胞は、がん幹細胞の場合がある。好ましくは、前記腫瘍細胞は、幹細胞、膵臓、血液、頸部、大腸、腸、腎臓、脳、乳腺、卵巣、前立腺、皮膚、肝臓、肺、及び、脾臓の腫瘍から選択されるか、若しくは由来であってもよい。
好ましくは、本発明の方法ステップ(d)における気相に面している半透性支持体の表面上での混合細胞のインキュベーション中に、前記腫瘍細胞は、前記マトリクス細胞によって形成されたマトリクス内に3次元に配置されて増殖する。
都合のよいことには、本発明に従って用いられた培地は、液体の形態、例えば、液体懸濁培地又は液体培養培地の場合がある。好ましくは、本発明による培養培地は、共培養される細胞種全ての増殖を支持する完全増殖培地を含む。最も適切な培地は、培養での細胞が生じる器官(又は器官)に依存して変わってもよい。好ましくは、培地は、器官型増殖に必要な栄養を提供する。脳組織用の適切な液体培地の例が、例えば、Stoppini L.ら(1991)及びMullerら(2001)で記載される。他の器官用の他の適切な培地は発表されるか、若しくはそれらの器官から生じる細胞の培養から得ることができる。用いるべき適切な培養培地は、当業者の知識の範囲内である。用いられる増殖条件及び培地は、通常は、腫瘍細胞が培地組成によって影響されないように、マトリクス細胞によって好ましいものである。
いずれかの適切な支持体は、支持体が半透性であることを条件として、混合された細胞の共培養物を増殖するために用いられてもよい。したがって、前記支持体は浸透性又は半浸透性である。支持体は半透性膜の場合がある。
前記マトリクス細胞及び/又は腫瘍細胞は、遺伝子操作されてもよい。例えば、前記マトリクス細胞及び/又は腫瘍細胞はトランスジェニック、又は、別の方法で遺伝子操作された動物(例えば、非ヒト哺乳類、非ヒト霊長類、げっ歯類又はマウス)由来の場合がある。トランスジェニック又は別の方法で遺伝子操作された動物は組換えゲノムを有する。天然で起こるよりもむしろ、意図的な改変が行われた。
また、本発明の共培養物は抗がん剤をスクリーニングする方法で用いられてもよい。したがって、本発明は試験薬剤と本発明の共培養物とを接触することを含む方法を提供し、ここで、共培養物の腫瘍細胞の成長、増殖、生存能力、生存、又は、遊走における試験薬剤の阻害効果は前記試験薬剤が候補抗がん剤であるということを示す。
ヒト膠芽腫腫瘍細胞株、LN−18(ATCC #CRL2610)が培養され、緑色蛍光タンパク質(GFP)を発現するプラスミド、又は、赤色蛍光タンパク質(RFP)を発現するプラスミドのいずれかがトランスフェクションされた。細胞は、ウェル1つあたり1x105個の細胞で6ウェルプレート上で培養され、配置され、一晩増殖され、75%までのコンフルエントであることが確認された。前記細胞は、製造者のプロトコールに従って、ジーンジュース(Gene Juice)(ノバジェン、カタログ番号# 70967−EA)を用いてpEGFP−N3(BDバイオサイエンス、カタログ番号# 6080−1)がトランスフェクションされた。トランスフェクション後24時間で、前記細胞は、ジェネティシン(GIBCO、カタログ番号#10131−019)で選択され、選択された細胞の集団が実験で用いられた。選択された細胞の集団は、トランスフェクションされた細胞の集団を維持するための臨時の選択で延長された時間にわたって培養された。腫瘍細胞による蛍光タンパク質の発現は、増殖及び形態学的変化に対して前記腫瘍細胞を観察可能にする。トランスフェクションされた腫瘍細胞(LN−18)は、フラスコからそれらを除去するためにトリプシン/EDTA(シグマ#T4299)で処理され、培養培地に再懸濁された。生存能力のある細胞は、トリパンブルー排除を用いて計測された。懸濁液は1000rpm/3分で遠心分離され、結果として生じる細胞ペレットが50,000細胞/μlで再懸濁された。
器官型培養物は、単離したラット大脳皮質から解離細胞を調製することによって作製された。P0ウィスターラットが屠殺され、脳が除去された。大脳皮質はペトリ皿で解剖され、小片に切断され、冷EBSS(GIBCO、カタログ番号# 24010)に入れられた。その後、組織片は非常に穏やかに粉砕された。結果として生じる細胞懸濁液が、50mLのファルコンチューブに取り付けられている100μmのメッシュフィルターによって濾過された。前記懸濁液は1000rpm/3分で遠心分離された。生存能力のある細胞は、トリパンブルー排除を用いて計測され、細胞ペレットが50,000細胞/μlで再懸濁された。
解離したラットの脳の大脳皮質は、気液界面の半多孔性膜上で再凝集できるようにされた(図1の右手部分を参照せよ。)。ラット胚から新たに解離された脳細胞が、培地上に配置された親水性のPFTE膜上に配置され、20日間増殖できるようにされた。細胞塊の横断面が、神経細胞(ベータ−チューブリン)及び活性化したアストロサイト(グリア細胞繊維性酸性タンパク質、GFAP)に対するマーカーを用いる免疫蛍光のために処理された。図1bで示されるように、染色は、神経細胞及びアストロサイトの両方がかかる3D培養で存在すること、及び、生理学的な構造が保存されることを示した(図1d、e、fも参照せよ。)。その後、コリン作動性神経細胞及び神経末端は、コリンアセチルトランスフェラーゼに対する抗体を用いて可視化された(ChAT;図1c)。予測通り、ChAT染色は、全脳の共培養物の至る所に分布され、前記神経末端で特に豊富であった。接続を確立している成熟神経細胞の存在が、電気生理学的パラメーターの測定によってさらに検討された。脳の共培養物の播種後4週間まで、自発的及び誘発の電気活性が、実験開始後14日間最適活性で観察された(図1d)。かかる電気信号の伝播は、一群の相互接続した神経細胞に特有である。
実施例1及び2で説明されたように調製されたトランスフェクションされた腫瘍細胞及びラット初代細胞は50,000細胞/μlで細胞の懸濁液を形成するために一緒に混合された。5μLのこの混合された細胞懸濁液が、6ウェルプレート(NUNC)における1mLの大脳皮質培地(10% Hams F12、20% FCS、5%ウマ血清、10mM Hepes、2mM グルタミン含有DMEM)中に配置された0.4ミクロンのミリポアカルチャーインサート(Millipore culture insert)(ミリポア、カタログ番号# PICM0350)の上部に添加された。その後、プレートは37°C、5%CO2でインキュベーションされた(図1a)。このインキュベーションの最初の数時間に、細胞は、半透膜支持体を介する毛細管現象により5μlの細胞懸濁液から液体を除去することによって高密度培養物(denser culture)を形成するために圧縮される(図1a)。
最適化は、それよりも、過度の低濃度によって細胞は腫瘍様の凝集を形成することができず、過度に迅速に凝集を形成し、過度な高濃度によって細胞の遊走過程が観察されない状態であることが望ましい。前記細胞は適切な濃度及び割合で混合されるとき、前記腫瘍は3−4日間にわたって形成され、ラット脳マトリクスの崩壊をもたらさない。
1つの実施態様では、最適濃度5%ないし95%の腫瘍対ラット細胞を用いて器官型共培養物内でのLN−18腫瘍細胞の増殖が、13日間にわたって顕微鏡的にモニターされた(図3)。in vitroでの3日目(DIV)までに、前記細胞は初代ラット組織で腫瘍様の凝集を形成した。これらの腫瘍様の凝集はin vitroでの13日(DIV)後にまだ存在した。
類似の効果が膠芽腫細胞株GL15、A172(ATCC#CRL1620)及びU87(ATCC#HTB−14)で見られた。それぞれの細胞株はeGFPで標識され、3D器官型共培養物を形成するためにラット脳細胞と混合された。eGFPシグナルによって示されるように、全ての脳由来の腫瘍細胞株は増殖し、LN18細胞と非常に類似の手段で共培養物内で腫瘍細胞の集団を形成した(図3b、c及びd)。
実験は、器官型共培養物中に腫瘍様の塊が細胞のクローン性増殖から発達したか、細胞マトリクスを介する細胞の遊走から発達したかを見るために実施された。ヒト脳腫瘍細胞株LN−18は、eGFP又はRFPのいずれかを細胞に導入するためにトランスフェクションされた。LN18 RFP発現細胞及びLN18 eGFP発現細胞が一緒に混合され、その後、解離されたラット大脳皮質細胞とともに95%ラット脳細胞に対して5% LN−18の割合として混合され、以前に記載されたように小型の3D器官型共培養物を形成するために多孔性膜上に配置された。これらの培養物は14日間にわたってモニターされた。共培養物に取り込まれた後24時間で、前記細胞は緑色及び赤色の細胞の均質の混合物として出現する。取込み後48時間と早期に、いくつかの細胞集団が形成され始めた。取込み後4日目で、細胞は、混合された赤色及び緑色の細胞の凝集を明瞭に形成した。赤色及び緑色の細胞が共局在するという事実は、単一の腫瘍細胞のクローン性増殖よりもむしろ腫瘍細胞同士の活性型クラスタリングを示す。したがって、前記細胞は腫瘍様の凝集を形成するために遊走するように見えた。その効果は細胞のクローン性増殖に起因する場合に、観察された赤色及び緑色の混合された腫瘍よりもむしろ個別の赤色及び緑色の腫瘍の発達が期待される。これは、前記細胞マトリクスが、in vivoの状況をより厳密に表す方法で腫瘍細胞間の相互作用を可能とする環境を提供するという証拠である。
ヒト卵巣がん細胞、SKOV3(ATCC# HTB77)は、ラット脳細胞マトリクスで腫瘍を形成しなかった(図4a)。これは、実施例4の膠芽腫細胞として記載されたようなラット脳細胞と、トランスフェクションされたSKOV3−GFP細胞とを混合することによって試験された。また、前記SKOV3細胞は疑似卵巣組織で凝集しないが、該細胞は両方のマトリクスで増殖する。
腫瘍幹細胞は生検材料から単離され(W.Grayからの贈り物)、前記細胞はフラスコで培養され、その後、前記細胞はレンチウイルスベクター、rHIV PPT−PGK−GFP−WPRE(VSVg)(Genethon)で形質導入(transduced)された。10μlのベクター(〜20x106の感染性粒子)が2:1のMOIを与えるために50μlの懸濁された幹細胞(〜10x106細胞)に添加され、その後、前記細胞は1週間培養され、GFPの発現がモニターされた。これらの形質導入された腫瘍幹細胞は、実施例4に記載されたように、器官型共培養物を作製するために腫瘍細胞株の代わりに用いられた。培養期間中、器官型共培養物では、前記細胞は分化し、突起を出した(図5)。
器官型共培養物は、マトリクス及び腫瘍細胞間の相互作用と、関連のある代謝経路との複合研究を可能にする免疫組織学的解析のために容易に調製される。前記器官型共培養物は採取され、4%パラホルムアルデヒド(シグマ#P6148)で固定され、その後、0.001%Tween(シグマ#P5927)のトリス緩衝生理食塩水(TBS)(TBS_T)で3回洗浄された。非特異的結合部位は、5%正常ロバ血清で30分間インキュベーションすることによってブロッキングされ、その後、TBS−Tで5分間3回洗浄された。マウスで作製された1次抗体GFAP(シグマ#G3893)1:200がTBS−Tで希釈され、器官型共培養物に添加され、4°Cで一晩インキュベーションされた。TBS−Tで5分間3回洗浄後、2次抗体アレクサフルオロヤギ抗マウス568(赤色)(インビトロジェン# A11031)@ 1in 500が暗所にて室温で1時間添加された。器官型共培養物はTBS−Tで5分間3回洗浄され、その後、封入液(シグマ#F4680)とともに顕微鏡スライドガラスに配置された。カバーガラスが最上部に配置され、マニキュア液で密封された。スライドはイメージングされるまで4°Cで暗所にて保存された。調製されたスライドは、付属のベクトンディッキンソン社製CARV共焦点細胞イメージングシステムを有するオリンパス社製1X70型顕微鏡でイメージングされた。
多形性膠芽腫(GBM)の典型的な特徴は、脳腫瘍の周辺での反応性アストログリオーシスの存在である。実施例4に記載されたように調製された本発明の培養物がヒト腫瘍のこの特性を再現できるかを決定するために、活性型アストロサイトのマーカーであるグリア細胞繊維性酸性タンパク質(GFAP)の発現がGFAP抗体を用いてGBM/脳の共培養物で評価された。
5−アミノレブリン酸(5−ALA)は脳神経外科手術中に腫瘍組織を可視化するために用いることができる。このガイド方法の手術中の使用が脳腫瘍を患っている患者での腫瘍残存量を低下し、無進行生存を延ばす場合がある。実施例4で調製されたようにGBM(LN18−eGFP)/脳の共培養物は、ヒト腫瘍でのように、5−ALAが腫瘍細胞を特異的に染色できるかどうかを見るために5−ALAで染色された。
悪性神経膠腫の周縁組織は代謝的に極めて活性が高く、高レベルの乳糖を産生し、腫瘍境界周辺の正常組織の細胞外空間を酸性化する。この特徴が本発明の腫瘍共培養物によって再現されるかどうかを確認するために、器官型GBM/脳共培養物が、pH色素染色で干渉されないように、eGFPを発現していないトランスフェクションされていないLN18腫瘍細胞が用いられる以外は、上記(例えば、実施例4)のように調製された。培養物は3日間インキュベーションされた。その後、挿入物は培養培地から除去され、滅菌ハンクス平衡塩液(HBSS)を含むウェルに配置された。最初の15分間の洗浄後、HBSS及び1μlの10mM BCECF酸(2’,7’−ビス−(2−カルボキシエチル)−5−(及び−6)−カルボキシフルオレセイン、混合異性体)、蛍光放射がpH感受性である色素を含むウェルに前記挿入物は移された。514nmで励起されたとき、その蛍光極大は塩基性環境で630nm(緑色染色)であり、酸性のとき570nm(赤色染色)であり、すなわち、細胞のより酸性条件は赤色を示す一方、中性のpHでは緑色を示す。培養物は30分間インキュベーションされ、その後、赤色(蛍光波長610)及び緑色(蛍光波長525)のスペクトルでのイメージングの前にHBSSで2回洗浄された。
器官型共培養物は、抗腫瘍化合物に対する腫瘍細胞の応答の解析のために用いられた。膠芽腫の標準的なケアは、放射線治療の間及び後のテモゾロマイド(TMZ)化学療法を含む。ここで、本発明者らは、LN−18−GFP細胞の標準2D培養物と比較して、器官型共培養物でのTMZ(シグマ#T2577)治療の有効性を試験した。
パクリタキセル(タキソール、シグマ#T7402)は、単離された2D培養物で膠芽腫細胞株に対して毒性があると以前に示されたが、臨床試験で用いられるとき、パクリタキセルは効果がないことが証明された。実施例14でのように同一の環境条件を用いて、本発明者らは、器官型培養(3D)及び腫瘍細胞のみの単層培養でタキソールの細胞毒性を比較した。
また、本発明者らは、シトシンアラビノシド(Ara−C)、2D培養での膠芽腫細胞株の増殖を阻害し、臨床試験で失敗したことが知られるさらなる化合物を試験した。図8aで示されるように、Ara−Cは2D培養で抗増殖活性を示した(AraCについて100nMで増殖阻害)。GBM/脳共培養物で用いられるとき、1μMのAra−Cは腫瘍細胞の増殖を減少するために必要とされる。投与量は2D培養で必要な投与量を10倍上回る(図8b)。10μMで、GBM細胞は効果的に殺された。
その後、類似の測定が2D又は脳細胞マトリクスを用いる共培養物(3D)で増殖する3つの他の膠芽腫細胞株(GL−15、A172及びU87)を用いて実施され、タキソール又はテモゾロマイドで処理された。増殖阻害IC50は各細胞株で各化合物について決定され、その結果が表1にまとめられている。全体として、従来の2D培養と比較して、共培養物で増殖する細胞で6倍から25倍までのタキソールの活性の低下が観察された。類似のことがLN−18細胞で観察され、テモゾロマイドの有効性は脳共培養物で2.5倍から12倍まで増加された。したがって、本発明の共培養物は、さまざまな膠芽腫細胞株についての2D培養よりもより近く臨床的状況を反映する。
化学療法が、確立された腫瘍を有する患者に施され、効果が乏しい。テモゾロマイド又はタキソールが、本発明の共培養物で増殖する確立された膠芽腫で腫瘍退縮をもたらすことができるかどうかを検討するため、腫瘍細胞集団が既に存在するとき、本発明者らは、(上記のように)播種するGBM/脳共培養時又は3日後のいずれかに処理を開始した(図9)。上記で観察されたように、タキソール処理は実験の開始時に開始され、最初の3日間の腫瘍細胞の増殖率は低下した。25nMのタキソールで、細胞増殖は停止され、以前に観察されたように、100nMのタキソールが腫瘍集団の縮小を達成するために必要とされた。

Claims (36)
- in vitroにおける3次元器官型細胞共培養物であって、前記共培養物は、腫瘍細胞と相違するマトリクス細胞のマトリクス内に3次元的に配置された腫瘍細胞を含み、基底膜を含まないことを特徴とする、共培養物。
- 請求項1に記載の共培養物であって、該共培養物はさらに半透性支持体及び液体培養培地を含み、
a)前記共培養物は気相に面する支持体の第1表面に配置され、
b)前記支持体の第2表面は前記液体培養培地に面し、及び、接触し、並びに、
c)前記液体培養培地は表面張力及び/又は毛細管現象の効力によって前記支持体及び前記培養物に接触して保持されることを特徴とする、共培養物。 - 請求項1又は2に記載の共培養物であって、前記腫瘍細胞は、少なくとも5個の細胞の凝集物の形態で存在することを特徴とする、共培養物。
- 3次元器官型細胞共培養物を調製する方法であって、
a)腫瘍細胞を含む第1細胞懸濁液及び前記腫瘍細胞と相違するマトリクス細胞を含む第2細胞懸濁液を提供するステップと、
b)液体懸濁液培地に懸濁された混合細胞を提供するために前記第1及び第2の細胞懸濁液を混ぜ合わせるステップと、
c)前記混合細胞を濃縮(concentration)及び/又は圧縮(compaction)させるステップと、
d)気相に面する半透性支持体の第1表面でステップ(c)から得られた前記混合細胞をインキュベーションするステップであって、前記インキュベーション中に前記支持体の第2表面が液体培養培地に面し、及び、接触し、並びに、前記液体は表面張力及び/又は毛細管現象の効力によって前記支持体及び前記混合細胞に接触して保持されるステップとを含むことを特徴とする、方法。 - 請求項4に記載の方法であって、前記ステップ(d)のインキュベーション中に、前記腫瘍細胞がマトリクス細胞によって形成されるマトリクス内に配置され、3次元的に増殖する(grow)ことを特徴とする、方法。

- 請求項1ないし3若しくは17ないし18のいずれかに記載の共培養物又は請求項4ないし18のいずれかに記載の方法であって、前記マトリクス細胞は単一の細胞種からなることを特徴とする、共培養物又は方法。

- 請求項1ないし3若しくは17ないし22のいずれかに記載の共培養物又は請求項4ないし22のいずれかに記載の方法であって、前記腫瘍細胞及び/又はマトリクス細胞はヒト由来であることを特徴とする、共培養物又は方法。
- 請求項1ないし3若しくは17ないし23のいずれかに記載の共培養物又は請求項4ないし23のいずれかに記載の方法であって、前記マトリクス細胞は、幹細胞か、1次細胞か、間質細胞か、又は、生検、死体の組織、細胞株由来の哺乳類若しくはヒト組織、中枢神経系、脳、骨髄、血液、脾臓、網膜、胸腺、心臓、乳腺、肝臓、膵臓、甲状腺、骨格筋、腎臓、肺、腸、卵巣、膀胱、精巣、子宮、若しくは、結合組織から選択される供給源に由来の細胞かを含むことを特徴とする、共培養物又は方法。
- 請求項1ないし3若しくは17ないし24のいずれかに記載の共培養物又は請求項4ないし24のいずれかに記載の方法であって、前記腫瘍細胞はがん幹細胞であることを特徴とする、共培養物又は方法。
- 請求項1ないし3若しくは17ないし25のいずれかに記載の共培養物又は請求項4ないし25のいずれかに記載の方法であって、前記腫瘍細胞は、幹細胞、膵臓、血液、頸部、大腸、腸、腎臓、脳、乳腺、卵巣、前立腺、皮膚、肝臓、肺又は脾臓由来であることを特徴とする、共培養物又は方法。
- 請求項2、3若しくは17ないし26のいずれかに記載の共培養物又は請求項4ないし26のいずれかに記載の方法であって、前記支持体は親水性PTFE、PET又は酸化アルミニウムを含むことを特徴とする、共培養物又は方法。
- 請求項2、3若しくは17ないし27のいずれかに記載の共培養物又は請求項4ないし27のいずれかに記載の方法であって、前記支持体は光学的に透明であることを特徴とする、共培養物又は方法。
- 請求項2、3若しくは17ないし28のいずれかに記載の共培養物又は請求項4ないし28のいずれかに記載の方法であって、前記支持体は、前記共培養物の前記支持体への接着を促進する材料で被覆されていることを特徴とする、共培養物又は方法。
- 請求項2、3若しくは17ないし29のいずれかに記載の共培養物又は請求項4ないし29のいずれかに記載の方法であって、前記支持体を被覆する前記材料は、コラーゲン、ラミニン又はフィブロネクチンより選択されることを特徴とする、共培養物又は方法。
- 請求項4ないし30のいずれかに記載の方法によって得られることを特徴とする、in vitroでの3次元器官型細胞共培養物。
- 抗がん剤をスクリーニングするための方法であって、該方法は請求項1ないし3又は17ないし31のいずれかに記載の共培養物を試験物質に接触させることを含み、前記共培養物の前記腫瘍細胞の増殖(growth)、増殖(proliferation)、生存能力又は遊走に対する前記試験物質の阻害効果が抗がん剤候補であることを示すことを特徴とする、方法。
- 請求項1ないし3又は17ないし31のいずれかに記載の共培養物の、抗がん剤をスクリーニングするための方法における使用。
- 請求項1ないし3又は17ないし31のいずれかに記載の共培養物の、がんバイオマーカー及び/又はがん薬剤標的を同定及び/又は検証する方法における使用。
- 請求項1ないし3又は17ないし31のいずれかに記載の1つ又は2つ以上の共培養物を含むことを特徴とする、アッセイキット。
- 請求項1ないし3又は17ないし31のいずれかに記載の1つ又は2つ以上の共培養物を含むことを特徴とする、化合物をスクリーニングするための装置。
