Abstract
外気と密閉されたカートリッジを用いて、試薬の混合、攪拌、精製、反応等を行えるようにする。
【解決手段】
外気と密閉されたカートリッジ1の内部には、送液する試薬の部屋38と、送液先の部屋39とが設けられ、それらを送液通路で繋ぐ。カートリッジ本体30には溝が彫られ、その上から弾性体であるメンブレン31を貼り付けることで送液通路36を形成する。メンブレン31に空気圧を与え、送液通路31の容積を変化させることで内部の流体を動かす。各部屋の入り口には弁機能を持たせ、送液通路の変化と合わせて任意の方向に内部の流体を動かす。これにより、外気と密閉されたカートリッジ内部での送液が可能となり、カートリッジ内部で試薬の混合や攪拌、精製、反応等が行える。また、弁の構造をカートリッジ内部に持たせず、カートリッジをセットするホルダ2に持たせることで、カートリッジ自身のコストを抑えることができる。

Description
(2)本発明に係る生化学処理装置は、前記生化学用カートリッジに加えて、次のような構成要素、すなわち、前記カートリッジを保持し、前記メンブレンを前記ポンプ機構として作動させるための空気圧を加える空気圧印加部を有するカートリッジホルダと、空気圧源と接続されて前記カートリッジホルダへの前記空気圧の供給、排気を制御する空気給排機構と、を有する。
Claims (11)
- 送液する試薬を封入する送液元の部屋と、前記試薬の送液先の部屋と、それらをつなぐ送液通路とを備え、これらの部屋と送液通路とがカートリッジ本体に密閉されて設けられており、
前記カートリッジ本体の底面には、前記送液通路が形成され且つ弾性体からなるメンブレンが張り付けられ、
このメンブレンの一部が、前記送液通路の壁面の一面となり、且つ外部から与えられる圧力の変化により往復動作して送液通路の容積を変化させるポンプ機構として構成されていることを特徴とする生化学用カートリッジ。 - 前記部屋は、試薬を封入する部屋と、液体試料を封入する部屋と、前記液体試料と前記試薬とを混合した混合液から標的となる生体物質を抽出・精製するための一連の処理が順次行われる複数の部屋とからなり、これらの部屋のうち関連する部屋同士が前記メンブレン付きの前記送液通路を介して外気と遮られて接続されている請求項1記載の生化学用カートリッジ。
- 前記抽出・精製される生体物質は、核酸であって、前記複数の部屋は、前記核酸を抽出・精製するための一連の処理に加えて、前記核酸を増幅させるために必要な処理を行う部屋を含み、これらの部屋のうち関連する部屋同士が前記メンブレン付きの前記送液通路を介して外気と遮られて接続されている請求項1記載の生化学用カートリッジ。
- 前記送液通路でつながる前記関連する部屋同士の上部が、前記カートリッジ本体の上部に設けた通気溝或いは通気孔により通じ合っている請求項1または2記載の生化学用カートリッジ。
- 前記カートリッジ本体における前記関連する部屋同士の下端には、前記送液通路に通じる送液口が設けられ、前記メンブレンの一部が、弾性変形によって前記送液口を開閉する弁構造を有している請求項1ないし4のいずれか1項記載の生化学用カートリッジ。
- 前記カートリッジ本体は、前記核酸を増幅させるために必要な一連の処理を行う複数の部屋のうち最終段の部屋が、電気泳動DNAシーケンサのキャピラリに接続可能に構成されている請求項3記載の生化学用カートリッジ。
- 前記カートリッジの底面材質が、弾性を有する樹脂もしくはゴム材で成形される請求項1ないし6のいずれか1項記載の生化学用カートリッジ。
- 前記カートリッジ本体は、混合液を攪拌する攪拌部屋を有し、液体の攪拌は、前記送液通路を介して該攪拌部屋につながる送液元の部屋の送液口を閉じて該攪拌部屋だけが前記送液通路に通じる状態にして、該送液通路での前記メンブレンの往復動作で生じさせ、このメンブレンの往復動作で前記攪拌部屋の液体の一部の引き戻りと押し返しを生じさせて行うように設定されている請求項1ないし7のいずれか1項記載の生化学用カートリッジ。
- 送液する試薬を封入する送液元の部屋と、前記試薬の送液先の部屋と、それらをつなぐ送液通路とを備え、これらの部屋と送液通路とがカートリッジ本体に密閉されて設けられており、前記カートリッジ本体の底面には、前記送液通路が形成され且つ弾性体からなるメンブレンが張り付けられ、このメンブレンの一部が、前記送液通路の壁面の一面となり、且つ外部から与えられる圧力の変化により往復動作して送液通路の容積を変化させるポンプ機構として構成されている生化学用カートリッジと、
前記カートリッジを保持し、前記メンブレンを前記ポンプ機構として作動させるための空気圧を加える空気圧印加部を有するカートリッジホルダと、
空気圧源と接続されて前記カートリッジホルダへの前記空気圧の供給、排気を制御する空気給排機構と、を有することを特徴とする生化学処理装置。 - 前記カートリッジホルダは、前記メンブレンの一部を介して前記各部屋の送液口を開閉するエアシリンダ機構を有する請求項9記載の生化学処理装置。
- 前記空気給排機構は、前記メンブレンに対して陽圧或いは陰圧を加えてメンブレンを作動させる請求項9又は10記載の生化学処理装置。
