Abstract
Description
本出願は米国特許仮出願第61/391,558号(2010年10月8日出願)(参照によりその全体が本明細書に組み込まれる)に基づく優先権を主張する。
癌は、活発に免疫系を抑制することによって免疫監視を回避する。このような免疫抑制に対抗することを目的とする方法の一つは、腫瘍細胞によって特異的に発現または過剰発現される抗原のエピトープを用いるワクチン接種によるものである。例えば、シグナル伝達経路を阻害する、増殖因子を抑制する、および/または免疫応答を誘導するモノクローナル抗体(mAb)は、癌および他の疾病を治療するために臨床導入され、成果を上げてきた。その好適な特性および臨床における成果のため、mAbは、これまでも、また現在も、タンパク質工学において重点的な取り組み課題となっている。これらの取り組みにより、改善された標的化のための二重特異性mAb;向上された腫瘍浸潤性および血液クリアランスのための1本鎖Fab可変領域フラグメント(scFv)、ダイアボディ(diabody)、およびミニボディ(minibody);そして、(変異またはグリコシル化による)免疫刺激の変更または薬物動態学的/薬力学的特性の改善を目的とした改変型Fcが開発されてきた。同様に、mAbを再設計し、送達改善のために小分子の部位特異的コンジュゲートを可能にすること(例えばThioMAB)、または、その抗原に不可逆的に結合させること(例えば無限(infinite)親和性mAb)が行われてきた。また、生体活性ペプチドおよび他の生物製剤(例えばCovXbody)の循環および提示を向上するためのmAbも開発されてきた。ヘテロ多量体scFv、または、アビジンに融合したscFvもしくはmAbも、事前標的化療法および腫瘍イメージングの検出限界向上のために開発されてきた。
抗体結合ペプチドである、C-QFDLSTRRLK-C(cQFD;SEQ ID NO:1)およびC-QYNLSSRALK-C(cQYN;SEQ ID NO:2)は、新規のmAb結合特性を有することが明らかにされた。特に、cQFDおよびcQYN(別名「メディトープ(meditope)」)は、抗EGFR mAbであるセツキシマブのFabフレームワーク領域に結合し、抗原が結合する相補性決定領域(CDR)には結合しないことが明らかにされた。そのFabフレームワーク上の結合領域は、他のフレームワーク結合抗原(例えばスーパー抗原、黄色ブドウ球菌プロテインA(SpA)(Grailleら, 2000)およびペプトストレプトコッカス・マグヌス(Peptostreptococcusmagnus)プロテインL(PpL)(Grailleら, 2001))とは異なっており、これまで知られていなかった。従って、第1の態様は、配列CQFDLSTRRLKC(SEQ ID NO:1)またはCQYNLSSRALKC(SEQ ID NO:2)を有する環状メディトープに結合する、ユニークなマウス-ヒト抗体またはその機能性フラグメントのフレームワーク領域を含有するフレームワーク結合インターフェースである。
治療剤またはイメージング剤を標的組織に選択的に送達させる抗体送達システム、およびそれらの使用法を本明細書で提供する。抗体送達システムは、抗体フレームワーク結合インターフェース(「フレームワーク結合インターフェース」または「結合インターフェース」)が抗体結合分子(または「メディトープ」)に結合して抗体-メディトープ複合体を形成したものを含んでもよい。抗体-メディトープ複合体は、下記に詳述する方法に使用するための1つまたはそれ以上の更なる抗体-メディトープ複合体、治療剤、イメージング剤、またはそれらを組み合わせたものに更にコンジュゲートしていてもよい。
試料と方法
試料。セツキシマブの抗原結合フラグメント[F(ab)'](または「Fab」)を得るため、IgGを固定化パパイン(Pierce社)で消化し、その後、プロテインAによる逆相精製およびSuperdex75カラム(GE Healthcare社)によるサイズ排除クロマトグラフィー(SEC)を行った。セツキシマブの1本鎖結合フラグメント(scFvC225)を、軽鎖および重鎖の間に20アミノ酸長のリンカーを挿入して合成した。scFvC225および可溶性上皮細胞増殖因子受容体ドメインIII(sEGFRdIII)をSf9細胞で発現させ、過去の報告(Donaldsonら, 2009)に従って精製した。
セツキシマブ上のメディトープ結合部位を同定するために、Fabフラグメントを作製および精製し、cQFDメディトープと1:10の割合で混合し、市販の因子(factorials)を用いて結晶形成のスクリーニングを行った。20℃で1-3日後に結晶が形成された。これらの結晶の初期回折解析から、ユニット・セルは、既にProtein Data Bankに登録されているセツキシマブFab(1YY8.pdb)(Liら, 2005)と同様であることが明らかとなり、結晶パッキング(例えば、CDRが除去された)から、ペプチドがCDRループに存在しないことが示唆された。しかしながら、分子置換によって構造が解明され、実験で得られたマップの解析から、メディトープと一致するモデリングされていない電子密度が同定された。初期Fo-Fcマップは、Fabフラグメントの中央部分が可能性のある結合部位であることを明示している(図1)。Fabモデルだけを用いる初期ラウンドの精密化の後、メディトープと一致するひと続きのモデリングされていない密度が観察された。メディトープを密度に組み込み、それに伴ってRおよびRFreeは低下した。Phenix(Adamsら, 2002)を用いた精密化の際に、水分子を付加した。回折データおよび精密化統計値を、以下の表3に示す。
試料と方法
上記実施例1に記載するものに加え、以下の 試料および方法を使用した。
メディトープおよび点変異。上記のように、CQFDLSTRRLKC(cQFD;SEQ ID NO:1)およびCQYNLSSRALKC(cQYN;SEQ ID NO:2)の合成、酸化、および精製をシティ・オブ・ホープ 合成化学/高分子化学コア施設で行った。アラニン点変異をcQFDメディトープ中の残基3(Phe3をAlaに)、残基5(Leu5をAlaに)、残基8(Arg8をAlaに)、および残基10(Leu10をAlaに)に作製し、SMT3のC末端にこのペプチドをコードさせ、細菌を用いて生成した(Mossessovaら, 2000)。ユビキチン様プロテアーゼ(Ulp1)をサンプルに添加してペプチドを遊離させた後、表面プラズモン共鳴(SPR)分析を行った。
本明細書で同定されたメディトープおよびFab間の結合部位のインターフェースは、IgGの4つのドメイン全てで形成される(例えば重鎖および軽鎖の可変ドメインおよび定常ドメイン)。PISAサーバーを用いて、cQFDまたはcQYNメディトープ-Fabインターフェースの埋没した表面領域は、それぞれ904(±28)A2および787(±42)A2であり、軽鎖および重鎖に等しく分布していた。図2および4に、メディトープと接触するFabの残基およびループを示す。
メディトープ-Fabインターフェースの位置に基づいて、メディトープがリガンド非結合型構造および/またはリガンド結合型構造に対するIgGドメインの相対配向を妨害するか否かを確認した。まず、両メディトープ複合体の非対称単位セル内の軽鎖および重鎖を比較した後、各鎖をリガンド非結合型構造およびEGFRリガンド結合型構造と比較した。いずれのメディトープに結合した軽鎖の可変ドメインも、本質的にセツキシマブのリガンド非結合型構造と同一であった(r.m.s.d.平均:cQFD,0.231±0.014A;cQYN,0.18±0.01A)。しかしながら、重鎖の可変ドメインは、有意に高い相違性を示した(r.m.s.d.平均:cQFD,0.85±0.01A;cQYN,0.88±0.01A)。留意すべきことに、この相違は主に、フレームワーク・ループ2(残基39-46)の位置に起因するものであり、これは、このループ中の残基を欠失させてr.m.s.d.を再計算すると、かなり低い値となるためである(cQFD,0.18±0.01A;cQYN,0.31±0.01A)(図6)。更に、このループはFab C225-EGFR共結晶構造中では移動され、その相対B-factor値は、これがフレキシブルであることを示している(図6)。最後に、メディトープの存在は、EGFR結合型または非結合型構造関してCDR構造に有意な変化を与えない。EGFRリガンド結合型構造の重鎖CDRループ3中のTyr101のバックボーンは、いずれのメディトープに結合したFab構造と比較しても反転しているが、この反転は、リガンド非結合型セツキシマブFab構造でも観察される(Liら, 2005)。
cQFDメディトープ-Fab複合体の構造、並びに、cQYNおよびcQFDの配列同一性に基づき、いくつかの点変異をcQFDメディトープ中に作製し(Phe3→Ala、Leu5→Ala、Arg8→Ala、およびLeu10→Ala)、メディトープの全体的な結合親和性に対するこれらの残基の役割をキャラクタライズした。結合親和性を評価するために、標準的なアミン化学を用いてFabフラグメントをCM5チップに結合させた。次に、合成cQFDおよびcQYNメディトープのセツキシマブFabに対する親和性を測定した。cQFDメディトープは950±30nMの親和性でFabに結合し、cQYNメディトープの結合親和性は3.5±0.1μΜであった(n=3)。結合反応速度も測定した(図3)。二分子相互作用とした場合の結合定数は、cQFDおよびcQYNでそれぞれ4.2(±0.1)x104M-1s-1および1.8(±0.1)x104M-1s-1であった。解離定数は、cQFDおよびcQYNでそれぞれ2.5(±0.1)x10-2s-1および8.6(±0.1)x10-2s-1であった。それらの測定値に基づくKD値(cQFDで430(±30)nM、およびcQYNで3.5(±0.1)μΜ)は、平衡測定とほぼ一致する。
試料と方法
上記実施例1および2に記載するものに加え、以下の試料および方法を使用した。
試薬。セツキシマブの1本鎖結合フラグメント(scFvC225)を、軽鎖および重鎖の間に20アミノ酸長のリンカーを挿入して合成した。ScFvC225および可溶性上皮細胞増殖因子受容体ドメインIII(sEGFRdIII)をSf9細胞中で発現させ、過去の報告(Donaldsonら, 2009)に従って精製した。
回折データは、これらのペプチドがワクチンとして有効であるという仮説と矛盾する。特に、原子モデルは、メディトープがCDRに直接結合しておらず、そのため、抗原エピトープを模倣しないことを示している。しかしながら、cQYNメディトープを接種したマウスから採取した血清は、細胞増殖を阻害した(Riemerら, 2005)。従って、メディトープが抗原結合を遮蔽するか否かを試験するために、セツキシマブFabをEGFRドメインIIIおよびcQFDと共にインキュベートし、SEC分析カラムに施与した。13.9mLにピークが観察され、このピークの非還元SDS-PAGEにより、3つの成分全ての存在が確認された(図4B)。個々の成分は15.2mL(Fab C225)、15.6mL(sEGFRdIII)、および16.3mL(SMT-CQFDLSTRRLKC;SEQ ID NO:1)に溶出された。
FabはメディトープおよびEGFRドメインIIIへ同時に結合できるが、メディトープが完全長IgGとしてのセツキシマブのEGFR発現細胞への結合に影響を与えるか否かを確認した。これを試験するために、FACS分析を用いて、メディトープ濃度の関数として、IgGのMDA MB-468細胞(EGFRを過剰発現する)への結合を追跡した。細胞を、漸増濃度のcQFDメディトープ存在下でセツキシマブと共にインキュベートした。60μMを超えるメディトープ濃度でも、セツキシマブの細胞への結合に有意な変化は見られなかった(図5)。この知見は上記のSEC分析と一致しており、メディトープが抗原結合のアロステリック調節剤として作用しないことを示している。
メディトープを、マウスキメラ化またはマウス化ヒトmAbの軽鎖または重鎖N末端に、フレキシブルなリンカーを介して連結することができる(図11)。mAbIgGのN末端は抗原結合部位と並列しており、フレキシブル・リンカーを介するN末端からの伸長によって抗原結合は立体的に妨害される。リンカー内に腫瘍特異的プロテアーゼ部位(例えばMMP9、MMP14、前立腺特異的抗原(PSA)セリンプロテアーゼ、または他の好適な部位)をコードさせることにより、分子内「遮蔽された」IgG構築物の立体的な制約が腫瘍部位切断され、抗体結合が可能となる。この設計原理では、分子内「遮蔽された」IgGの正常組織への結合が阻害され、オフターゲット結合による有害な副作用が回避される。セツキシマブ上のアビジン-ペプチド遮蔽のオフレート測定を図12に示す。この図は、多価メディトープが1価メディトープより高い親和性で結合するが、遮蔽はしないことを示している。
同じ抗原(例えばEGFR)上の独自のエピトープを認識する複数のモノクローナル(mAb)を組み合わせることにより、細胞死が促進され、腫瘍増殖が阻害されることが明らかになっている。この細胞死促進の正確なメカニズムは議論されているところであるが(免疫応答vs受容体の抑制的制御vsリガンド拮抗の促進)、過去の研究から、いずれのmAbも、細胞死を促進するためには多価(例えば完全長IgGまたはF(ab)'2)である必要があることが示唆されている。そのように、多価メディトープを、後述のように第2の抗体として代用してもよい。従って、本明細書に記載するメディトープをスキャホールドに連結させ、選択性および結合親和性を向上するための多価メディトープ作製してもよい。
FITC標識した2価メディトープの合成を、(化合物2)を用い、「クリック」ケミストリーに基づいて行った(図13)。テンプレート4および5(図13)を使用することにより、それぞれから2価および3価メディトープの形成が可能となる。この合成はすばらしい進歩である。なぜなら、この発見は多価メディトープ調製のために開発された化学を表すものであり、最適な結合のために種々の長さのポリエチレングリコール(PEG)(または他の)リンカーの検討に重点的に取り組むことができる。合成法はまた、放射性核種イメージングのためのDOTA導入にも適している。例えば30AのPEG2価アームが、FITC標識された2価メディトープ(すなわち化合物13)の合成で導入された(図13)。IgG内でのCDR領域間の距離は〜130Aである。このように、PEGリンカーの長さは体系的に多様であり、この方法は確かに最適である。市販のPEGの末端から末端までの距離は90Aであり(Pierce社)、IgGの距離より長い。
それぞれの多価メディトープのキャラクタリゼーションをSPRおよびITCによって行い、多価スキャホールドへのコンジュゲーションがメディトープ-IgG相互作用に影響しないことを確認した。しかながらこれらの測定は、IgGが腫瘍表面に結合しないため、多価性の測定における有効性に限界がある。これに代わって、FACS分析および細胞生存率アッセイを使用して、多価メディトープの影響をEGFRを過剰発現する細胞上で直接定量してもよい。
セツキシマブと併用した場合の単量体メディトープまたは2価メディトープ-Fcの細胞死誘導に対する効果を、MTTアッセイを用いて評価した。4000個のMDA-MB-468細胞を80μlの培養液中、96ウェルプレートの各ウェルに播種した。10μlの1μMセツキシマブを10μlの0.1、1、または10μMのメディトープまたはメディトープ-Fcと共に添加し、最終濃度を0.1μMセツキシマブ、および、0.01、0.1、および1μMメディトープまたはメディトープ-Fcとする。また、コントロールとして各成分を単独でPBSと共に添加した。48時間のインキュベーション後、10μlのMTT試薬を添加し、更に4時間インキュベートした。その後、培養液上清を除去し、100μlのMTT結晶溶解液を添加し、プレートを630nmで測定した。メディトープまたはメディトープ-Fc単独では細胞増殖は有意に変更されなかったが、メディトープ-Fcだけがセツキシマブとの併用で細胞増殖を阻害した(単量体メディトープは阻害しなかった)(図28A)。
メディトープ部位の内表面に並ぶ残基を体系的または無作為的に変更し(縮重ライブラリーおよび選択)、メディトープまたはメディトープ類似体の特異性を向上および/または変更することができる(改変方法については、例えばSheedyら 2007およびAkamatsuら 2007(参照により本明細書に組み込まれる)参照)。これらの位置を非天然アミノ酸、天然アミノ酸、またはその両方で置換し、メディトープ相互作用の親和性を向上することができる。近年、二重特異性抗体を作製するための非天然アミノ酸の抗体への導入が報告された(Hutchinsら 2001)。
ある態様では、メディトープ結合部位を既存の抗体および全ての今後開発される抗体、例えばIgG1-4(λおよびκアイソフォーム)、IgE、IgA1、IgA2、IgD、およびIgM上に、他のIgG(例えばヒト、マウス、またはニワトリ)の残基の変異を介して作製することができる。
メディトープの組成を変更して、pHの関数として結合親和性に影響を与えることができる。図27に示すように、3つの異なるメディトープ改変体の結合親和性を、バッファーpHの関数として測定した。QYDメディトープ改変体は、低pHで著しい親和性の低下を示す。QYN改変体中のアスパラギン酸をアスパラギンに置換すると、pH依存性はフラットとなる。同様に、QFD改変体の親和性は、pHが高くなるにつれ、わずかに上昇する。総合すると、これらの実験は、メディトープ-mAb相互作用の親和性はpHに合わせて調整できることを示している。

- 1つまたはそれ以上の治療物質、イメージング物質、またはそれらを組み合わせたものと共にメディトープを含有する組成物。
- メディトープは配列CQFDLSTRRLKC(SEQ ID NO:1)またはCQYNLSSRALKC(SEQ ID NO:2)を含有する、請求項15記載の組成物。

- 1つまたはそれ以上の置換は、セツキシマブ軽鎖のS9I、S10L、K39R、P40T、P40S、G41N、K42G、A43S、F83I、F83V、T85N、T85D、もしくはQ100A;または重鎖のA36SもしくはV89Iである、請求項20記載の方法。
- 置換によって、治療用ヒト抗体またはその機能性フラグメントの結合および/または治療効果を向上させる多価メディトープ連結体の結合が可能となる、請求項20記載の方法。
- フレームワークへの結合は、メディトープとフレームワークとの間に少なくとも1つのジスルフィド結合または少なくとも1つの非天然アミノ酸との結合を伴う、請求項20記載の方法。

結合したメディトープまたはメディトープ改変体を、モノクローナル抗体またはその機能性フラグメントを含有するかまたは含有する疑いのある溶液と接触させて、モノクローナル抗体またはその機能性フラグメントをメディトープに結合させる

- 疾病または病態の治療、イメージング、または診断の方法であって、治療的有効量の医薬組成物を被験体に投与することを含み、医薬組成物は、抗体-メディトープ複合体;モノクローナル抗体またはその機能性フラグメントと組み合わせた多価連結剤;またはそれらを組み合わせたものを含有する、上記方法。
- 抗体-メディトープ複合体;モノクローナル抗体またはその機能性フラグメントと組み合わせた多価連結剤;またはそれらを組み合わせたものは、疾病もしくは病態の診断もしくはイメージングのための1つもしくはそれ以上の診断剤もしくはイメージング剤、疾病を治療するための1つもしくはそれ以上の治療剤、またはそれらを組み合わせたものにコンジュゲートされている、請求項31記載の方法。
- 治療剤は、1つまたはそれ以上の小分子、化学療法剤、治療用抗体もしくは機能性フラグメント、毒物、放射性同位体、酵素、ヌクレアーゼ、ホルモン、免疫調節剤、オリゴヌクレオチド、ナノ粒子、RNAi分子、キレート剤、ホウ素化合物、光反応性物質、色素、またはそれらを組み合わせたものである、請求項32記載の方法。

- 抗体-メディトープ複合体;モノクローナル抗体もしくはその機能性フラグメントと組み合わせた多価連結剤;またはそれらを組み合わせたものは、治療用抗体単独での治療、診断、またはイメージングの有効性を向上する、請求項31記載の方法。

- 多価連結剤は、治療的有効量のセツキシマブを被験体に投与した後、または治療的有効量のセツキシマブを被験体に投与するのと同時に投与される、請求項31記載の方法。
- 腫瘍バイオマーカーを過剰発現する細胞を標的化する方法であって、治療的有効量のメディトープ使用可能モノクローナル抗体またはその機能性フラグメントを、メディトープ、メディトープ改変体、または多価連結剤と組み合わせて、バイオマーカーを過剰発現する、または過剰発現する疑いのある細胞集団に投与することを含む、上記方法。
- 腫瘍バイオマーカーはEGFRであり、メディトープ使用可能モノクローナル抗体はセツキシマブである、請求項38記載の方法。
- メディトープ改変体の結合親和性を改変する方法であって、メディトープ改変体を1つまたはそれ以上の位置で改変して、低pHレベルでのメディトープの結合親和性を増加または低下させることを含む、上記方法。
- リソソーム薬剤送達を行うために、メディトープ改変体は低pHレベルにおいて低下した結合親和性を有する、請求項40記載の方法。
- 腫瘍細胞への特異的結合を行うために、メディトープ改変体は低pHおよび低酸素環境において増加した結合親和性を有する、請求項40記載の方法。
- メディトープまたはメディトープと同様のフレームワーク結合機能性を有する小分子をスクリーニングする方法であって、
推定されるメディトープまたは小分子のライブラリーをメディトープ使用可能抗体と接触させ;
推定されるメディトープまたは小分子がメディトープ使用可能抗体にフレームワーク結合インターフェースで結合するか否かを測定し;
1つまたはそれ以上の候補となるメディトープまたはメディトープと同様のフレームワーク結合機能性を有する小分子を同定し;
1つまたはそれ以上の候補の結合親和性を測定し;そして、
結合解離定数が少なくとも0.70μMである場合に、1つまたはそれ以上の候補をメディトープまたはメディトープと同様のフレームワーク結合機能性を有する小分子であると同定する
ことを含む、上記方法。
